<B>Verfahren</B> zur Herstellung eines Morphinan-Abkömmlings Es wurde gefunden, dass das bisher unbe kannte 2,17-Dimethyl-3-oxy-morphinan eine ausgesprochene hustenlindernde Wirkung hat, ohne dabei gleichzeitig analgetisch zu wirken. Diese Eigenschaft kommt in gleicher Weise der racemischen, (-)- und (+)-Form zu.
Gemäss der vorliegenden Erfindung ge winnt man 2,17-Dimethyl-3-oxy-morphinan, indem man ein 3-Oxy-N-methyl-morphinan mit Formalin oder einer Formalin abgeben den Verbindung und einem sekundären ali- phatischen Amin umsetzt (Mannich-Reaktion) und das entstehende 2-Dialkylaminomethyl-3- oxy-N-methyl-morphinan einer katalytischen Hydrierung unterwirft. Dabei wird das se kundäre Amin hydrierend abgespalten.
Man erhält racemisches, (+)- oder (-)-2,17-Di- methyl-3-oxy-morphinan, je nachdem man von racem., (-f-)- oder (-)-3-Oxy-N-methyl-mor- phinan ausgeht. Diese Basen können mit Säuren in wasserlösliche Salze übergeführt werden.
Für die Umsetzung nach Mannich wird zweckmässig die 3-Oxy-N-methyl-morphinan- base in einem Lösungsmittel, z. B. Methyl-, Äthyl- oder Butylalkohol, gelöst und zusam men mit einem sekundären Amin, z. B. Di- äthylamin oder Dibutylamin, in Gegenwart von Formalin oder Paraformaldehyd in an sich bekannter Weise längere Zeit unter Rück flusskühlung gekocht.
Die entstehenden 2-Di- alkylaminomethyl-3-oxy-N-methyl-morphinane sind starke Basen, die mit Säuren wasserlös- liehe Salze bilden. Zur Überführung in 2,17- Dimethyl-3-oxy-morphinan werden sie zweck mässig als Basen in einem Lösungsmittel, wie Methanol oder Äthanol, bei erhöhtem Druck und erhöhter Temperatur in. Gegenwart von Hydrierkatalysatoren hydriert. Bei etwa 50 Atü und 150 C ist die hydrierende Ab spaltung des sekundären Amins innert kur zer Zeit beendigt.
Die 2,17-Dimethyl-3-oxy- morphinane werden durch Einengen gewon nen Sie bilden als Basen farblose Kristalle, die in Wasser schwer, in den gebräuchlichen organischen Lösungsmitteln leicht löslich sind. Mit Säuren, z. B. Salzsäure, Bromwasserstoff säure, Schwefelsäure, Phosphorsäure, Wein säure, bilden sie wasseilösliche Salze. Sie sol len als hustenlindernde Mittel Verwendung finden.
<I>Beispiel 1</I> Man kocht ein Gemisch von 257 Gewichts teilen rac. 3-Oxy-N-methyl-morphinan mit 80 Gewichtsteilen Diäthylamin und 50 Gewichts teilen Paraformaldehyd in 1000 Raumteilen Äthylalkohol während 10 Stunden am Rück fluss. Nach dem Abdestillieren des Alkohols und des überschüssigen Diäthylamins wird mit 500 Gewichtsteilen Wasser versetzt und mit Salzsäure bei schwach kongosaurer Reak tion in Lösung gebracht.
Die klare Lösung wird mit .Ammoniak versetzt, bis keine Trü bung mehr entsteht, und die ausfallende Base in Benzol aufgenommen. Der nach dem Ver jagen des Benzols verbleibende Rückstand (340-345 Gewichtsteile) wird: in 700 Raum teilen absolutem Alkohol gelöst, mit etwa 220 Raumteilen 48o/oiger Bromwasserstoffsäure schwach kongosauer gestellt und mit Äther bis zur Trübung versetzt, worauf bald Kri stallisation einsetzt. Der Kristallbrei wird nach gründlichem Kühlen geputscht.
Durch Trocknen gewinnt man so praktisch quanti tativ rac. 2-Diäthylaminomethyl-3-oxy-N-me- thyl-morphinan-dihydrobroinid vom Schmelz punkt 252-253 C.
504 Gewichtsteile dieser Verbindung wer den in 600 Raumteilen Wasser gelöst und dar aus die Base mit Ammoniak abgeschieden. Sie ;wird in Äther aufgenommen, der Äther ab- destilliert, der Rückstand (342 Gewichtsteile) in 1600 Raumteilen Methanol gelöst und in Gegenwart von Palladitunkohle bei 50 Atü und 150 C hydrierend gespalten.
Die vom Katalysator getrennte Lösung wird im Teil- vakuum. eingeengt. Der Rückstand wird in 2000 Raumteilen Benzol unter Erwärmen ge löst. Beim Abkühlen kristallisieren grosse Kristalle des rae. 2,17-Dimethyl-3-oxy-morphi- nans, die ein Mol Kristallbenzol enthalten.
Sie werden geputscht und getrocknet. Schmelz- punkt unter vorherigem Sintern 89-92 C.
Diese Kristalle werden in Wasser mit ver dünnter Salzsäure in. Lösung gebracht, die Lösung zur Entfernung des Kristallbenzols eingeengt und hierauf das rae. 2,17-Dimethyl- 3-oxy-morphinan-hydrochlorid kristallisieren gelassen.
Es kristallisiert als Monohydrat, das nach dem Trocknen an der Luft bei etwa 40 C unter Aufschäumen bei 149-150 C schmilzt. Die daraus hergestellte Base schmilzt bei 183 bis 184 C.
Das rae. 2,17-Dimethyl-3-oxy-mor- phinan-hydrochlorid ist in Wasser mit prak- tisch neutraler Reaktion ziemlich leicht, in Alkohol leicht, in Äther umlöslich.
Beispiel <I>2</I> Kocht man 257 Gewichtsteile rac. 3-Oxy- N-met4yl-morphinan mit 180 Gewichtsteilen Dibutylamin und 180 Gewichtsteilen Para- formaldehyd in 2000 Raumteilen Alkohol 12 Stunden am Rückfluss, so gewinnt man bei der Aufarbeitung nach Beispiel 1 das rae. 2-Dibutylaminomethyl-3-oxy-N-methyl-mor- phinan als klares,
in. verdünnten Säuren lös liches Öl.
Es wird ohne Reinigung der hydrierenden Spaltung unterworfen. Dabei gewinnt man nach der Arbeitsweise des Beispiels 1 neben Dibutylamin, das wieder verwendet werden kann, das rae. .2,17-Dimethyl-3-oxy-morphi- nan, das die in Beispiel 1 beschriebenen Ei genschaften aufweist.
<I>Beispiel 3</I> Behandelt man (-)-3-Oxy-N-methyl-mor- phinan nach Beispiel 1, so erhält man das (-) - 2,17 -Dimethyl - 3 - oxy-morphinan-hydro- chlorid als Monohydrat. Es schmilzt bei 171 bis 173 C, die daraus hergestellte Base bei 203-204 C; [a] D = -36,3 für c = 3,0 in H20.
Genau dieselben Eigenschaften, nur ent gegengesetzte Drehung, zeigt das in gleicher Weise aus (-f-) -3-Oxy-N-methyl-morphinan er hältliche (-f-) -2,17-Dimethyl-3-oxy-mor phinan.

According to the present invention, 2,17-dimethyl-3-oxy-morphinan is obtained by reacting a 3-oxy-N-methyl-morphinan with formalin or a formalin-releasing compound and a secondary aliphatic amine (Mannich- Reaction) and the resulting 2-dialkylaminomethyl-3-oxy-N-methyl-morphinane is subjected to catalytic hydrogenation. The secondary amine is split off by hydrogenation.
Racemic, (+) - or (-) - 2,17-dimethyl-3-oxy-morphinan, depending on whether one of racem., (-F -) - or (-) - 3-oxy-N -methyl-morphinane runs out. These bases can be converted into water-soluble salts with acids.
For the Mannich reaction, the 3-oxy-N-methyl-morphinan base is expediently in a solvent, e.g. B. methyl, ethyl or butyl alcohol, dissolved and together men with a secondary amine, z. B. diethylamine or dibutylamine, boiled in the presence of formalin or paraformaldehyde in a conventional manner under reflux for a long time.
The resulting 2-dialkylaminomethyl-3-oxy-N-methyl-morphinans are strong bases which form water-soluble salts with acids. To convert them into 2,17-dimethyl-3-oxy-morphinane, they are conveniently hydrogenated as bases in a solvent, such as methanol or ethanol, at elevated pressure and elevated temperature in the presence of hydrogenation catalysts. At about 50 atmospheres and 150 ° C., the hydrogenative cleavage of the secondary amine is completed within a short time.
The 2,17-dimethyl-3-oxymorphinanes are obtained by concentration. As bases they form colorless crystals which are difficult to dissolve in water and easily soluble in common organic solvents. With acids, e.g. B. hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, tartaric acid, they form water-soluble salts. They should be used as antitussive agents.
<I> Example 1 </I> A mixture of 257 parts by weight of rac is boiled. 3-Oxy-N-methyl-morphinan with 80 parts by weight of diethylamine and 50 parts by weight of paraformaldehyde in 1000 parts by volume of ethyl alcohol for 10 hours at reflux. After the alcohol and the excess diethylamine have been distilled off, 500 parts by weight of water are added and hydrochloric acid is dissolved in a weakly Congo acidic reaction.
Ammonia is added to the clear solution until there is no longer any cloudiness, and the base which precipitates out is taken up in benzene. The residue (340-345 parts by weight) remaining after the benzene has been hunted down is dissolved in 700 parts by volume of absolute alcohol, made weakly Congo acidic with about 220 parts by volume of 48% hydrobromic acid and ether is added until it becomes cloudy, whereupon crystallization soon sets in.
