A process for preparing 4 3> s, 7> 9 (11), 22-ergostapentaen-3-ol acetate. The present invention relates to a process for the preparation of 43,5,7,9 (II), zz-ergo-5tapenta.en-3-ol acetate, which is a known compound.
In the. chemistry of steroids, which have a double bond in position 9-11 become. increasingly important for the synthesis of cortisone and analogues. The compound obtained by the process of the present invention is particularly useful for this purpose, since it can be prepared from a relatively abundant raw material, such as ergo sterol. , and. clu does.
that its side chain at position 17 can be transformed into a side chain identical to that possessed by cortisone. This transformation can be carried out by methods such as cew-, described by Bergmann and Stevens (Journal of Organs Chemistry, 13, 1948).
According to the invention, the acetate of 43,5,7,9 (II), zz-ergostapentaen-3-al, of formula
EMI0001.0027
by reacting d3,5,7, zz-ergostatetraen-3-ol acetate with mercuric acetate in the presence of acetic acid.
Preferably, the reaction is carried out in a solvent such as lower aliphatic alcohol, carbon tetrachloride or chloroform. The starting material is then dissolved in the solvent and the mercuric acetate in solution is also added thereto, together with the addition of ice acetic acid. The mixture is heated to reflux to complete the reaction and the product formed is isolated.
It is preferred to carry out the reaction at a temperature of between about 20 and 125 ° C, where the reaction is ordinarily complete after a period of 10 minutes to about 20 hours.
After completion of the reaction, the product formed can be isolated as follows: The mercury acetate is filtered off while the mixture is still hot, followed by filtration. Concentrate the filtrate, add water and extract the product. with an organic solvent immiscible with water, such as carbon tetrachloride. The extract is washed in the organic solvent with. dilute acetic acid and. with medium alkali, it is dried and filtered. Evaporation of the solvent. under reduced pressure gives the crude product which is recrystallized from a lower aliphatic alcohol; a solid crystalline product having a point is thus obtained. fusion defined.
Example: 1.35 g of 43,5,7,22-ergostatetraen-3-ol acetate is dissolved in 75 em3 of ethanol. To this solution is added a hot solution of 4.05 g of mercuric acetate in 75 kt of ethanol containing 1.6 cm-3 of glacial acetic acid and. the mixture is refluxed for 2 hours in a nitrogen atmosphere. The mercury acetate is filtered off while the mixture is still hot, and the filtrate is concentrated to low volume under reduced pressure.
Water is added and the solution is extracted three times with carbon tetrachloride. The combined extracts are washed with. dilute acetic acid, sodium bicarbonate and water. Dried over magnesium sulfate, filtered with diatomaceous earth and evaporated to dryness under reduced pressure. The residue is taken up in acetone, water is added and. a yellow powder is obtained. Two crystallizations in ethanol and one in a mixture of ethyl acetate and methanol give 0.19 g of @ 3> 5,7,9 (ii), 22-ergost.apentaen-3-ol acetate in the form of yellow crystals; point. melting temperature: 160-7.61 C, maximum = 339, 355 and 375 my (absolute alcohol).
