Verfahren zur Herstellung eines neuen Benzoesäureesters. Die Erfindung bezieht sich auf ein Verfah ren zur Herstellung eines neuen Benzoesäure- esters, nämlich des 1-Cyklohexyla.mino-2-me- t liy 1-propy 1- (2) -benzoats.
Die neue Verbindung sowie deren Salze sind wertvolle technische Produkte und auch wirksame Lokalanästhetika, da. sie eine hohe Wirksamkeit mit verhältnismässig geringer Toxizität und verhältnismässig geringer Reiz wirkung verbinden.
Der genannte Benzoesäureester wird erfin dungsgemäss hergestellt, indem man ein Ben- zoylierungsmittel, das das Radikal

liefert, wie Benzoesäureanhydrid oder ein Benzoylhalogenid, mit 1-Cyklohexylamino-2- inethyl-propa.nol-(2) zum entsprechenden Benz- amid umsetzt, das dann durch Säurebehand lung in das entsprechende Estersalz überge führt. wird, welch letzteres mit Alkali in die freie Base umgewandelt wird.
Die Reaktion zwischen dem Benzoylie- rungsmittel und dem 1-Cyklohexylamino-2- methyl-propanol-(2) kann gewünschtenfalls in einem inerten Lösungsmittel, z. B. einem niedrigen halogenierten Paraffinkohlenwasser stoff, wie Chloroform oder Methylenchlorid, erfolgen.
Eine vorteilhafte Arbeitsweise be steht darin, das Benzoylierungsmittel im iner- ten Lösungsmittel zu lösen und dieser Lösung das 1-Cyklohexylamino-2-methyl-propanol-(2), in einem wässerigen alkalischen Medium ge- löst, zuzusetzen und das Reaktionsgemisch am Rückflusskühler zu erhitzen.
Wenn die Reaktion beendigt ist, kann die Mischung abgekühlt, die Methylenchlorid- schicht abgetrennt und im Vakuum konzen triert werden, wobei das Benzamid als trocke ner Rückstand zurückbleibt. Nach der Um lagerung kann die freie Base erhalten werden, indem man das in Wasser suspendierte Re aktionsprodukt mit einem Alkali, wie Natrium karbonat-Monohydrat, behandelt.
<I>Beispiel:</I> 0,15 Mol Benzoylchlorid in 100 ein:' Me- thylenchlorid wurden rasch zu einer kräftig gerührten Suspension von 1-Cyklohexylamino- 2-methyl-propanol-(2) in 200 em3 5prozentiger Natronlauge gegeben. Die mechanisch kräftig gerührte Mischung wurde am Rückflusskühler auf dem Wasserbad während einer Stunde so hoch erhitzt, dass das Methylenchlorid zum Sieden gelangte. Das Reaktionsgemisch wurde dann abgekühlt, wobei es sieh in zwei Schich ten trennte.
Die wässerige Schicht wurde ein mal mit Methylenchlorid extrahiert und der Extrakt mit der Methylenchloridschicht ver einigt, worauf die vereinigten Methylenchlorid- lösungen zweimal mit Wasser gewaschen und dann im Vakuum zur Trockne verdampft wurden.
Das trockene 1-(Cyklohexyl-benzoylamino)- 2-methyl-propanol (2) wurde einmal aus Äther und Pentan umkristallisiert:, wonach es einen Schmelzpunkt von 77,5 bis 78 C aufwies. Es wurde dann. in das Benzoatchlorhydrat über geführt, indem man eine Lösung von 0,05Mol des Benzamids in 250 ein-' absolutem Alkohol mit, einem 60prozentigen molaren Überschu12) ari konzentrierter Salzsäure während 5 Mi nuten erhitzte. Die Lösung wurde dann ge kühlt. und im Vakuum. zur Trockne einge dampft.
Der Rückstand wurde durch 7u1- satz von Benzol und erneutes Eindampfen im Vakuum weitergetrocknet und hernach aus Aceton umkristallisiert. Das so gereinigte 1- Cy kloliexy lamirio-2-metliyl. - propy l.-\'- benzoat- ehlorhy drat hatte einen Schmelzpunkt. von 162 bis 163 C.
Das Chlorhydrat wurde in einer kleinen Menge Alkohol gelöst, mit Wasser verdünnt und mit einem. über die stöchiometrisehe Menge hinausgehenden Überschuss an Na triumkarbonat behandelt. Die freigesetzte freie Base wurde mit Benzol extrahiert und aus dieser Lösung in bekannter Weise isoliert.
Das erfindungsgemäss erhältliche Amin wird als Anästhetikum zweckmässig in Form eines seiner Salze, z. B. des Chlorhydrats, Sul fats, Sulfamats, Tartrats, Glykolats usw., angewendet, da. die freie Base in Wasser ziem lich unlöslich ist.

An advantageous procedure is to dissolve the benzoylating agent in the inert solvent and to add 1-cyclohexylamino-2-methylpropanol- (2), dissolved in an aqueous alkaline medium, to this solution and to add the reaction mixture to the reflux condenser heat.
When the reaction has ended, the mixture can be cooled, the methylene chloride layer separated and concentrated in vacuo, the benzamide remaining as a dry residue. It was then. into the benzoate chlorohydrate by heating a solution of 0.05 mol of the benzamide in 250 absolute alcohol with a 60 percent molar excess of concentrated hydrochloric acid for 5 minutes. The solution was then cooled. and in a vacuum. evaporated to dryness.
The residue was further dried by adding benzene and again evaporation in vacuo and then recrystallized from acetone. The 1- Cy kloliexy lamirio-2-metliyl purified in this way. - propy l .- \ '- benzoate ehlorhydrate had a melting point. from 162 to 163 C.
The hydrochloride was dissolved in a small amount of alcohol, diluted with water and treated with a. Treated excess sodium carbonate beyond the stoichiometric amount. The released free base was extracted with benzene and isolated from this solution in a known manner.
The amine obtainable according to the invention is conveniently used as an anesthetic in the form of one of its salts, e.g. B. of the chlorohydrate, sulphate, sulfamate, tartrate, glycolate, etc., applied because. the free base is fairly insoluble in water.
