Verfahren zur Herstellung eines Laktons der Cyclopentanopolyhydrophenanthren-Reihe. Es wurde gefunden, dass man zu einem Lakton der Cyclopentanopolyhydrophenan- thren-Reihe gelangen kann, wenn man auf ein Derivat der A20,22_g_Oxy-nor-cholensäure von der Formel

worin X einen durch laktonisierende Mittel Sich abspaltenden Rest und R einen Rest, welcher durch hydrolysierende Mittel' durch die IIydroxylgTuppe ersetzt wird, bedeuten, mit Oxydationsmitteln behandelt,
welche an einem in a-Stellung zu einer Kohlenstoff Kohlenstoff-Doppelbindung stehendem Koh= lenstoffatom eine sauerstoffhaltige Gruppe einzuführen vermögen, welche mit einer Carboxylgruppe zu Iaktonisieren vermag und auf das so erhaltene Reaktionsprodukt ein laktonisierendes und schliesslich ein hydro- 1'isierendes Mittel einwirken lässt. Die Ausgangsstoffe können z.
B. durch Kondensation geeigneter Ketone der Pregnan- reihe mit halogenierten Essigsäurederivaten, wie Chlor-, Brom- oder Jod-essigsäureester und Wasserabspaltung leicht hergestellt wer den. Sie sind ferner z. B. auch durch Bromie- rung von entsprechenden gesättigten Säuren und Bromwasserstoffabspaltung zugänglich. Geeignete Ausgangsstoffe sind z.
B. Ao,22-d- Acyloxy-nor-cholensäureester. Es lassen sich aber auch die entsprechenden freien Säuren oder Amide verwenden. Oxydationsmittel, die geeignet sind, an einem in a-Stellung zu einer Kohlenstoff Kohlenstoff-Doppelbindung stehendem Koh- lenstoffatom eine sauerstoffhaltige Gruppe, welche mit einer Carboxylgruppe zu laktoni- sieren vermag, z. B. eine freie oder veresterte Oxy-Gruppe, einzuführen, sind z. B.
Selen dioxyd oder Bleitetraacylate. Die Behand lung mit diesen Mitteln wird zweckmässig unter Benützung geeigneter Lösungs- bzw. Verdünnungsmittel ausgeführt, gegebenen falls bei erhöhter Temperatur.
Zur Laktonisierung wird das Reaktions produkt z. B. mit Säuren behandelt, oder einer Vakuum-Destillation unterworfen. Die Laktonisierung kann unter Umständen bereits vorher in einem gewissen Umfang eintreten. Sind die Hydroxylgruppen und Carboxyl- gruppen nicht frei, so erfolgt die La.ktonisie- rung z. B. durch eine intermolekulare Ester abspaltung.
Das nach dem neuen Verfahren erhält liche Produkt, das Ao,22-3,21-Dioxy-nor- cholensäure-lakton vom F. 225-227 (14- Desoxy-digitoxigenin), ist identisch mit der gemäss Patent Nr. 240100 herstellbaren Ver bindung. Es soll therapeutische Verwendung finden oder als Zwischenprodukt zur Her- Stellung therapeutisch verwendbarer Ver bindungen dienen.
Beispiel: 4 g A0,22_3-Acetc>xy-nor-chol'ensäure-me- thylester (der z. B. aus 3-Oxy-pregnan-20-on durch Umsetzung mitBromessigsäuremethyl- ester nach Reformatzky und anschliessende Wasserabspaltung leicht zugänglich ist) werden in 300 cm' Essigsäureanhydrid ge löst und bei Siedehitze tropfenweise mit einer Lösung von 4 g .Selendioxyd in 20 em Wasser versetzt. Die Ausscheidung von Selen beginnt augenblicklich.
Nach mehrstündigem Kochen am Rückfluss wird die Lösung im Vakuum auf 50 cm' eingedampft. Man fil triert vom ausgeschiedenen Selen ab, giesst das Filtrat in Wasser und kocht kurz auf. Nach dem Erkalten werden die ausgeschie denen Reaktionsprodukte in Äther auf genommen. Der Äther wird verdampft und der Rückstand einer Destillation im Hoch vakuum unterworfen.
Das Destillat wird in Benzol' gelöst, mit Tierkohle behandelt und zur Entfernung der letzten Spuren von Selen respektive selenhaltigen Anteilen an Alumi niumoxyd ehromatographiert. Die Haupt fraktion des Chromatogrammes kristallisiert nach Zusatz von Methanol. Nach Um kristallisieren aus Alkohol . schmilzt dieses Produkt bei 1ö6-167 . Es liegt das d20,22_3- Acetoxy-21-oxy-nor-cholensäure-lakton vor.
Durch saure Verseifuug erhält man daraus in praktisch quantitativer Ausbeute das 420,22-3,21-Dioxy-nor-cholensäure-lakton <B>vom</B> F. 225-227 (14-Desoxy-digitoxigenin) und der Formel
