Die Ausbeute an 1,3-Diacetyl-4-methyl-imidazolon-(2)-5-a- ketocäpronsäure-inethylester vom Schmelz punkt 70 beträgt 44,2 g.
Process for the preparation of diacetyl-4-bromomethyl-imidazolon- (2) -5-e-ketocaproic acid = methyl ester. It has been found that when N-brominated amides and brominated imides act on compounds of the all- (r formula

in which X is a lower alkyl group, Y is hydrogen, a lower alkyl group,
a Carbalkoxya.lkylgruppe or a Carbalkoxyacylgruppe mean, starting materials for the synthesis of biotin or bio-unlike compounds are obtained. - The acyl groups linked to the nitrogen atoms can mean lower aliphatic acid residues, such as acetyl or propionyl. For the lower alkyl group X in position 4, methyl, ethyl or propyl can be used -erden.
Suitable brominating agents are N-bronisuccinimide and N-bromoacetamide.
The reaction with the brominating agent is conveniently carried out in an inert solvent such as carbon tetrachloride.
In the course of the reaction, a hydrogen atom of the substituents X and / or Y which is in a-sialuig to the ring-shaped core is replaced by a bromine atom; for example, the methyl is converted into a bromomethyl radical.
If there are two substituents X and Y in the 4- and 5-position, which carry hydrogen atoms which can be replaced in the a-position to the nucleus, the reaction can also be carried out in such a way that hydrogen atoms are in one or the other or both in a- Position to the ring-shaped core standing methylene groups are replaced by bromine.
The following general formula applies to the end products:
EMI0001.0051
in which A is an α-bromoalkyl group and B is hydrogen, an alkyl, an α-bromoallyl, a carbalkoxyalloyl, a carbalkoxy-a-bromoalkyl and a carbalkoxyacyl group.
The designation "a" used here is used to identify those carbon atoms which are in the alpha position relative to the ring carbon atoms 4 and 5 in formulas (A) and (B).
Depending on their nature, these compounds can either be isolated in a relatively pure form, or they can be used in the reaction mixture for the production of sub. Institutional products can still be used.
In this context it should be emphasized that the replacement of the bromine in these compounds with other substituents such as acetoxy, ethoxy, mercapto, dicarbethoxymethylene radicals and the like leads to intermediates which are valuable for syntheses.
The subject of the present patent is a process for the preparation of 1,3-di-acetyl-4-bromomethyl-imidazolone- (2) -5-s-keto-caproic acid methyl ester, which is characterized in that 1,3-diacetyl -4-methyl- imidazolon- (2) .- 5.- s-ketocaproic acid methyl ester is reacted with an N-brominated imide.
The previously unknown diacetyl-4-bromomethyl - imidazolone - (2) - 5 - a - ketocaproic acid methyl ester forms colorless needles with a melting point of 58 to 61. It is soluble in the usual organic solvents and is an intermediate product for the manufacture of vitamins or compounds with a vitamin-like effect.
Example: 19.6 g of 4-methyl-imidazolone- (2) are listed under the , for the ethyl ester in the paper by Duschinsky and Dolan (Journal of Ainer. Chem .Soaiety, Vol. 6.7 [1945] -S-. 2082) specified conditions with 35,
7 g of adipic acid monomethyl ester chloride and 80 g of aluminum chloride are reacted in 110 emg of nitrobenzene. The methyl 4-methyl-imidazolon- (2) -5-α-ketocaproate obtained in this way in an amount of 28 g melts at 176 after recrystallization from water with 5% methyl alcohol.
37.9 g of this keto ester are refluxed under reflux with 165 cm acetic anhydride for 20 minutes and then evaporated to a syrup. The boiling and evaporation process is repeated, the residue was dried for 3/4 hours at 100 in a vacuum and then recrystallized from 40 cm of methyl alcohol, cooling in a dry ice-acetone bath.
The yield of 1,3-diacetyl-4-methyl-imidazolon- (2) -5-a-ketocäpronsäure-inethylester with a melting point of 70 is 44.2 g.
16.2 g of this ester are now heated for 1 hour in 95 cm carbon tetrachloride with 8.55 g of N-bromosuccinimide for you. After separating the succinimide (4.44 g) and evaporating the filtrate to give an oil, 50 cm3 of ether are added, whereupon the diacetyl-4-bromomethyl-imidazolone - (2) - 5-8 -ketocaproic acid- methyl ester crystallized in the form of colorless needles with a melting point of 58 to 61.
The yield is 18.5 g.
