Verfahren zur Darstellung eines am indblstickstoff halogenierten Derivates der Lysergsäure-Reihe. Die Herstellung von Halogenverbindim- gen in der Lysergsäure-Reihe begegnet in folge der grossen Empfindlichkeit dieser Stoffe, vor allem gegen Säuren und Oxyda tionsmittel, grossen Schwierigkeiten. Reagen zien, die z.
B. freies Halogen oder Halogen wasserstoff entwickeln, reagieren unter weit gehender Zersetzung und Verharzung des Ausgangsmaterials; wo trotzdem aber in sehr geringer Menge halogenierte Verbindungen gefasst werden können, handelt es sich<B>Um</B> wenig definierte Produkte.
Es wurde nun gefunden, dass man wohl definierte, am Indolstickstoff monohalo- genierte Verbindungen der Lysergsäure-Reihe dadurch herstellen kann, dass man die zu halogenierende Verbindung in einem inerten Lösungsmittel mit einem am Stickstoff halo- genierten Carbon- oder Sulfonsäureamid oder -imid umsetzt.
Als Halogenierungsmittel eignen sich u. a. N-Brom-acetamid, N-Brom-phthalimid, N- Brom-p-toluolsulfamid, N-Brom-di-p-toliiol- sulfimid, N-Brom-succinimid, N-Jod-succin- imid, 2,6-Dichlor-4-nitro-N-chlor-acetanilid und N-Chlor-benzoesäuresulfimid.
Die Halogenierung von Derivaten der Lysergsäure-Reihe wird vorzugsweise in einem inerten Lösungsmittel, wie Dioxan, Äther, Tetraehlorkohlenstoff usw., bei Zimmer- oder etwas erhöhter Temperatur vorgenommen und die neuen Halogenverbindungen am vorteil haftesten durch Chromatographieren ihrer Lösungen von den Begleitstoffen abgetrennt.
Die neuen Verbindungen werden dabei als ölige Substanzen gewonnen, können aber durch Auflösen in einem geeigneten Lösungs mittel, wie Aceton, Benzol, Chloroform, Äther, Methanol usw., zur Kristallisation ge bracht werden, wobei man im allgemeinen schön ausgebildete, gegebenenfalls Kristall lösungsmittel enthaltende Kristalle erhält.
Das Halogenatom tritt bei Verwendung der genannten Halogenierungsmittel an den Indolstiekstoff. Die neuen Verbindungen ent sprechen z. B. den Formeln C15H1Í\T@@ . COR und C"H"N@X. COR (X= Halogen). Einige Vertreter sind in den Tabellen I und II auf geführt.

Das vorliegende Patent betrifft ein Ver fahren zur Herstellung von Brom-ergotamin, welches dadurch gekennzeichnet ist, dass man auf Ergotamin ein am Stickstoff halogenier- tes Carbonsäure- oder Sulfonsäure-amid bzw. -imid einwirken lässt.
<I>Beispiel:</I> 500 mg hochvakuumtrockenes Ergotamin werden in 45 eins Dioxan von 650 C gelöst und eine Lösung von 185 mg Brom-succin- imid in 10 eins Dioxan von 650 C zugesetzt. Das Gemisch, das noch 5 Minuten bei 700 C gehalten wird, färbt sich sofort gelbbraun und innerhalb einer Minute schwarzrot, unter leichter Trübung.
Zur Abscheidung der rohen Reaktionsprodukte wird die Dioxanlösung mit 200 cms Chloroform verdünnt und im Scheidetrichter mit natriumbicarbonathalti- gem Wasser gewaschen. Die Chloroform-Di- oxanschicht wird abgetrennt, mit Natrium sulfat getrocknet und zür Trockne verdampft.
Der Trockenrückstand, das rohe Reaktions produkt enthaltend, wird der chromatogra- phischen Trennung unterworfen. Zu diesem Zweck wird er in 5 cm3 absolutem Chloro form gelöst, die Lösung auf eine Säule von 40g Aluminiumoxyd gegossen und das Chro- matogramm mit abs. Chloroform entwickelt. Aus einer einheitlichen, im ultravioletten Licht schwach leuchtenden Zone gehen 331 mg farbloses Öl ins Filtrat.
Dieses wird zur Trockne verdampft und der Trockenrück stand aus 90%igem Aceton hinkristallisiert. Es werden 260 mg rechteckige -Platten vom Schmelzpunkt 197 bis 1980 C erhalten. Aus Methanol kristallisieren fünf- bis sechseckige Blättchen, die erst bei 207 bis 2080 C schmel zen.
