Verfahren zur Darstellung von Pregnen.ol-3-on-20. Die vorliegende Erfindung betrifft die Darstellung von Pregnenol-3-on-20.
Das .erfindungsgemässe Verfahren ist da durch gekennzeichnet, da.ss man einen 3-0xv- bisnorcholensäureester als Ausgangsstoff ver wendet, die ON-Gruppe in dessen 3-.Stellung intermediär in eine Gruppe verwandelt, die durch Hydrolyse in die 011-Gruppe zurück verwandelt wird.
auf die Estergruppe der Seitenkette in 17-Stellung eine Organo-Metall- verbindung, vorzugsweise Organo-1Vlagnesium- verbindungen. wie Gr-na.rdverbindungen. einwirken lässt, das so erhaltene Reaktions produkt mit Wasser zersetzt und aus dem dabei gebildeten Ca.rbinol die tertiär gebun dene ITvdroxylzruppe und das benachbarte tertiär gebundene Wasserstoffatom in Form von Wasser abspaltet,
die so erhaltene un gesättigte Verbindung, unter intermediärem Sehutz der R.ingdoppelbindung, einer oxyda- tiven Spaltung, zweckmässig mit Ozon, unter wirft, das so erhaltene Keton, unter Regene rierung der Ringdoppelbi.ndung, mit einem verseifenden Mittel behandelt, um die Oxy- gruppe in- 3-Stellung wieder herzustellen,
und aus dem Reaktionsgemisch das P'regnenol- 3-on-20 isoliert. Der Schutz ,der Ringdoppel bindung kann durch Anlagerung von Ha logen oder Halogenwasserstoffen bewirkt werden.
Die Hydroxylgruppe am Kohlenstoff atom 3 wird während der oxydativen Spal tung zum Beispiel durch Veresterung, Ver- ätherung oder dergleichen geschützt er halten und die betreffende Gruppe erst nach her durch Hydrolyse in die Hydroxylgruppe zuriiekverwandelt.
Die hauptsächlichsten Reaktionsvorgänge seien durch die folgenden Formelbilder ver anschaulicht, in denen R- eine 0l1-, 0-Acyl-, 0-Alkyl-, 0-Arylgruppe oder dergl. und R' eine Alkyl- oder Arylgruppe oder de-rgl. be deuten, während mit X, X angelagertes Ha logen oder angelagerter Halogenwasserstoff bezeichnet ist:

Als Anleitung für die praktische Durch- führung .des erfindungsgemässen Verfahrens mögen die nachstehenden Beispiele dienen <I>Beispiel 1:</I> 2,4 g Oxybisnorcholensäuremethylester in 150 cm' abs. Äther werden zu einer Auf- lösung von 3,9 g Magnesium und 25,2 g Brombenzol in 150 cm' abs. Äther unter Rüh ren hinzugegeben.
Das Reaktionsgemisch wird drei ' Stunden am Kühler erhitzt, der Äther abdestilliert und der Rückstand acht Stunden auf .dem Wasserbad erhitzt, dann wird das Reaktionsprodukt mit Eis zersetzt, mit ver dünnter ,Schwefelsäure angesäuert und er schöpfend mit Äther extrahiert. Nach dem Abdestillieren des Äthers wird das Reak- tionsprodukt zur Entfernung von Nebenpro dukten mit Wasserdampf destilliert,
und an schliessend. wird der im Rückstand enthaltene Anteil an nicht umgesetztem Ester durch Kochen mit methylalkoholischem. Kali ver seift. Die Verseifungslösung wird angesäuert. nochmals mit Wasserdampf destilliert und. der Rückstand in Äther aufgenommen und durch Ausschütteln mit verdünnter Natron lauge von den sauren Bestandteilen befreit.
Die ätherische Lösung wird dann ein gedampft, mit dem doppelten Volumen Eis essig versetzt und mehrere Stunden zum Sie den am Kühler gekocht; dann wird der Eis essig abdeztilliert und der Rückstand im Hochvakuum destilliert. Der grösste Teil geht bei 270 bis 280' unter 0,001 mm Druck als gelbliches, bald erstarrendes Öl über. Aus beute 1,2 g.
Durch Erhitzen mit 5 g E'ssig- säureanhy'dri'd wird :daraus ein Acetat vom Smp. 216 bis 217 erhalten.
0,4 g des A.cetates werden in 20 cm' Chloroform gelöst und tropfenweise unter Eiskühlung mit<B>0,1215</B> g Brom (= 2 Atome pro Mol) in 5 cm' Chloroform versetzt. In diese Lösung, die dann :das Dibromid des Acetates enthält, wird für eine iStund'e unter Eiskühlung ein Strom ozonierten Sauer stoffes, :der etwa 6 mg Ozon pro Minute lie fert, eingeleitet.
Nach Entfernung des Chloroforms wird :der Rückstand zur Ent- bromung mit 1 g Zinkstaub und 20 ein' Eis essig versetzt und eine .Stunde auf dem Wasserbad erhitzt. Die Eisessiglösung wird darauf filtriert, mit Wasser verdünnt und mit Äther ausgeschüttelt. Der Ätherauszug wird mit Natronlauge gewaschen, mit Tier kohle entfärbt und 'eingedampft.
Man erhält so :das Acetat des Oxyketons C21gax02 als ölige Kristallmasse, :die nach :dem Umkrietal- lisieren aus verdünntem Alkohol den Smp. 146 bis 147' zeigt.
Durch Umsetzung mit Semi'carbazidacetat erhält man das Semicarbazon vom-S'mp. <B>263'</B> und durch Spaltung desselben das freie Oxy- keton C21113202 vom Smp. <B>190'.</B> Ausbeute zirka 0,15 g. Dasselbe Oxyketon wird auch durch Verseifen des Aceta.tes direkt erhalten.
<I>Beispiel 2:</I> Aus 35 g troekenen Magnesiumspänen. 220 g Brombenzol, 440 cm' über Natrium getrocknetem Äther und einer Spur Jod wird eine ätherische Lösung von Phenyl- ma.gnesiumbromid hergestellt, wobei die Reaktionsflüssigkeit nach .dem Zusammen reben der Reagentien noch 11/2 .Stunden am Sieden erhalten wird.
Zu :dieser Lösung lässt man 20 g 3-Acetoxybisnorcholensäuremethyl- ester. den man -durch Acetylierung von 3-Oxybis:norcholensäuremethylester herstellt, gelöst in 450 cm' trockenem Äther, zu tropfen. Darauf gibt man unter gleichzei tigem Abdestillieren des Äthers 600 cm; über Natrium getrocknetes Toluol zu der Reaktionsflüssigkeit und erheitzt dieselbe zehn :Stunden im siedenden Wasserbade.
Bei den einzelnen Umsetzungen wird :das Reak tionsgut :gerührt.
Zur Aufarbeitung des Reaktionsproduk tes wird die Toluollösung in Eiswasser 'ge gossen und mit verdünnter Schwefelsäure versetzt. Nach Zusatz von Äther wird :die sich abtrennende wässrige Schicht entfernt. und die verbleibende Äther-Toluollö'sung wird im Vakuum zum Trocknen eingedampft.
Nachdem der Rückstand 'durch Kochen mit 5 % iger methylalkoholi'scher Kalilauge ver seift ist, wird die Verseifungslösung durch Zugabe von verdünnter ,S'chwefel'säure kongo sauer gemacht und längere Zeit mit Wasser dampf destilliert. Das dabei erhaltene Car- binol wird darauf fünf Stunden in 150 cm' Eisessig unter Rückfluss .gekocht.
Nach dem Abdestillieren des Eisessigs im Vakuum wird der Rückstand durch kurzes Erhitzen (etwa 10 bis 15 Minuten) mit Essig@s.äureanhydrid acetyliert; das gebildete Acetat wird :durch Zusatz von etwa 100 cm? Alkohol aus der Reaktionslösung abgeschieden, und, wenn er forderlich, aus Chloroform-Äthanol umkri stallisiert.
Dabei werden 16 bis 1,8 .g eines bei 213 bis<B>217'</B> schmelzenden, zweifach un- g o esättigten Acetates erhalten.
Dieses zweifach ungesättigte Acetat wird in 300 em3 Tetrachlorkohlenstoff gelöst und zum Schutze der Ringdoppelhindung -bei :der Oxydation mit der 2 Atomen pro Hol ent sprechenden Menge Brom in 120 cm' Tetra chlorkohlenstoff unter Eiskühlung versetzt. Darnach dampft man den T.etrachlorkohlen- stoff im Vakuum bei etwa 40 .ab und nimmt den Rückstand in 330 cm' Chloroform auf.
Man leitet :dann unter Aussenkühlung mit einer Eiskochsalzmischung etwa sieben Stun- den lang Ozon in .diese Chloroformlösung ein, verdampft darnach das Chloroform im Va kuum bei 40', nimmt :den RüolKsstantl in Eis essig auf und entbromt :
das Reaktionsprodukt durch portionenweise Eintragung von Zink- staub. Darnach wird die Reaktionsflüssig- keit mit Wasser verdünnt, vom Zinkstaub unter wiederholtem Nachwaschen mit Wasser abgegossen und mit Äther extrahiert;
die ätherischen Auszüge werden mit Natronlauge gewaschen, und der nach dem Verdampfen des Äthers verbleibende Rückstand wird mit 5 % iger methylalkoholischer Kalilauge ver seift.
Nach dem Ansäuern. und Ausäthern der Verseifungslösung wird beim Einengen des gewaschenen und .getrockneten Atherextrak- tes ein Teil des Pregnenolons direkt in kri stalliner Form erhalten.
Der in der Mutterlauge verbliebene An teil wird durch Umsetzung mit Semicarbazid- acetat als Semicarbazon abgeschieden.
Die Spaltung des Pregnenolonsemicarba- zons, F. zirka '238 bis 240', erfolgt durch zweistündiges Erhitzen mit einer Spaltungs- lösung, die auf 7'5 cm' Athanol 15 em3 H20 und 10 cm' konzentrierte Schwefelsäure ent= hält. Zweeks Isolierung des Pregnenolons wird das Reaktionsgemisch in der -üblichen Weise mit Wasser versetzt und mit Äther extrahiert:
Es werden so etwa 4 bis 5 g Pregnenolon vom ;Schmelzpunkt <B>190'</B> er halten.
Das verfahrensgemäss erhältliche Pregnen- ol-3-o#n-20 ist ein wertvoller neuer Stoff, der farblose Kristalle vom Schmelzpunkt <B>190'</B> bildet und dessen optische Aktivität raID -i-- <B>30'</B> beträgt; es lässt sich zum Bei spiel auf Pregnendion-3. 2,0, das die Eigen schaften .des Corpus luteum-Hormons besitzt, weiterverarbeiten.
