Solche Verbindungen können beispiels- weise in Anlehnung an das in der britischen Patentschrift Nr. 551216 beschriebene Ver fahren durch Umsetzung eines Esters einer a-Aminosäure mit einem Oxymethylen-bern- steinsäureester hergestellt werden.
Die erste Stufe des erfindringsgeniässeii Verfahrens besteht in der Cyclisierung der Ausgangsverbindung I zu dem Pyridinab- kömmling der Formel 1I bez-#v. IIca durch Einwirkung eines alkalischen Kondensations mittels und nachfolgende Behandlung des Alkalisalzes der entstandenen Verbindung mit Säure Als alkalische Kondensationsmittel kommen vor allem Alkalialkoholate, Alkalimetalle oder Alkaliamide,
insbesondere Natrium- amid, in Frage; es empfiehlt sich, in einem inerten Lösungsmittel zu arbeiten.

Das 2-Methyl-3-oxy-4,5-dicarbomethoxy- pyridin ,schmilzt bei 138-140 C, bildet ein salzsaures Salz vom Schmelzpunkt 165 C und lässt sich zu 2-Methyl-3-oxy-4,5-dicarb- oxy-pyridin vom Schmelzpunkt 265 C ver seifen, z.
B. auf folgende Weise: Die Substanz wird mit der dreifachen molaren Menge 6n Natriumhydroxyd auf dem Wasserbad erwärmt. Die Lösung wird mit 5-6n Salzsäure schwach lackmussauer gestellt, worauf das 2-2Tethyl-3-oxy-4,5-di- carboxy - pyridin auskristallisiert, welches unter Zersetzung bei 259 C schmilzt.
In seinen Eigenschaften stimmt dieses mit einem auf anderem Wege hergestellten, gleichen Pro dukt überein (vergleiche Ichiba & Einote, Sei. Papers, Inst. Phys. Chem. Res., Tokio, Bd. 38 [1941], S. 347). Durch Umkristalli- sieren kann der Schmelzpunkt auf 265 C er höht werden.
Das 2-Methyl-3-oxy-4,5-dicarbomethoxy- pyridin soll als Arzneimittel verwendet werden.
Beispiel .T 3,2 Gewichtsteile pulverisiertes Natrium- amid werden zu einer Lösung von 28,5 Ge wichtsteilen des Kondensationsproduktes von a-Benzylamino-propionsäure-methylester und Oxymethylen - bernsteinsäure - dimethylester von der Formel

und 120 Gewichtsteilen trockenem Benzol hinzugefügt,
worauf die Mischung unter Erhitzen im Wasserbad in einer trockenen Stickstoffatmosphäre gerührt wird. Eine heftige Reaktion setzt ein, wobei das Na- triumamid in Lösung geht und Ammoniak sich entwickelt. Die Lösung wird 1 Stunde unter Rückfluss gekocht und dann gekühlt.
Sie wird mit Eis und Essigsäure, welche etwas Schwefelsäure enthält, gerührt, wobei die Menge der verwendeten Säure derjenigen des Natriumamides entsprechen soll. Der Benzolauszug des so erhaltenen Produktes <B>wird</B> mit Wasser, Natriumbicarbonatlösung und nochmals mit Wasser gewaschen, ge trocknet, worauf das Lösungsmittel abge dampft wird.
Das zurückbleibende 01, wel ches vor allem aus 1-Benzyl-2-methyl-3-oxy- 4,5-dicarbomethoxy-dihydro@pyridin besteht, wird in 170 Gewichtsteilen trockenem Benzol aufgenommen.
Zu dieser Lösung werden unter Rühren 30 Gewichtsteile Sulfuryl- chlorid zugetropft, wobei die Temperatur der Reaktionsmischung auf 35-h0 C gehalten wird. Unter heftiger Reaktion entwickelt sich Chlorwasserstoff, und eine ölige Schicht scheidet sich aus der Benzollösung ab.
Wenn diese Abscheidung vollendet ist, wird das Benzol erbdekantiert und das -Öl während einigen Stunden in einem Teilvakuum auf zirka 40 C erwärmt. Der Rückstand wird mit 30 Gewichtsteilen Aceton digeriert, worauf das 1-Benzyl-2-methyl-3-oxy-4.5-di- carbomethoxy-pyridiniumchlorid als kristal line Verbindung, welche unter Zersetzung bei 148 C schmilzt, erhalten wird.
Dieses Produkt wird durch Filtrieren vom Aceton getrennt und mit dem gleichen Lösungsmittel gewaschen. Eine weitere Menge dieser Verbindung wird durch Zu gabe von trockenem Äther zum Acetonfiltrat erhalten. Die Gesamtausbeute beträgt 60 bis 70 70 der Theorie.
Eine Lösung von 6,5 Gewichtsteilen 1-Benzyl - 2 -methyll-3-oxy-4,5-dicarbometh- oxy-pyridiniumchlorid in 20 Raumteilen Methylalkohol wird mit Wasserstoff bei An wesenheit von 0,5 Gewichtsteilen Palladium- Tierkohle geschüttelt (Verhältnis: 1 Ge wichtsteil Palladium auf 4 Gewichtsteile Tierkohle).
Wenn die berechnete Menge Wasserstoff aufgenommen ist, wird der Ka talysator erbfiltriert und der Methylalkohol unter vermindertem Druck verdampft. Das 2-Methyl - 3- oxy-4, 5-dicarbomethoxy-pyridin- hydrochlorid wird durch Kristallisation aus einer Mischung von Alkohol und Äther in Form farbloser Nadeln, welche unter Zer setzung bei 165 C schmelzen, erhalten.
Eine konzentrierte wässerige Lösung von 2-Methyl-3-oxy-4,5-dicarbomethoxy-pyridin- h.ydrochlorid wird in der gälte mit einer ge sättigten wässerigen Natriumbicarbonat- oder Natriumacetatlösung behandelt. Das gefällte 2 -Methyl <I>- 3 -</I> oxy-4, 5-dicarbomethoxy-pyridin wird erbfiltriert, mit Eiswasser gewaschen.
getrocknet -und aus Methylalkohol oder Li- groin umkristallisiert. worauf es bei<B>138</B> bis 140 C schmilzt.
Beispiel <I>2:</I> Das Verfahren nach Beispiel 1 wird mit der doppelten Menge Natriumamid durch geführt. Die übrigen Aufarbeitungsmethoden sind dieselben, wobei jedoch die vor der Extraktion der cyclisierten Verbindun- zu verwendende Säuremenge verdoppelt wird.
<I>Beispiel 3:</I> Man arbeitet gemäss Beispiel 1, nur ver wendet man an Stelle des dort angegebenen Sirlfurylchlo,rids eine Lösung von 5,9 Ge- wichtsteilen Chlor in 85 Gewichtsteilen Te- trachlorkohlenstoff, wobei man dasselbe Zwi schenprodukt, nämlich 1-Benzyl-2-methyl-3- oxy- 4, 5 - dicarbomethoxy - pyridiniumchlorid, erhält.
<I>Beispiel</I> Zu einer trockenen Benzollösung, welche 30 Gewichtsteile des gemäss Beispiel 1 her gestellten 1-Benzyl-2-methyl-3-oxy-4,5-di- carbomethoxy-dihydropyridins enthält, wird unter Eiskühlung und Rühren die Haupt menge einer Lösung von 16 Gewichtsteilen Brom in 100 Gewichtsteilen Tetrachlor- kohlenstoff zugegeben. Ein dickflüssiges 01 scheidet sich aus der Lösung ab. Sobald keine Abscheidung mehr beobachtet wird, wird mit der Zugabe von Brom aufgehört.
Die an der Oberfläche sich befindende Flüs sigkeit wird abdekantiert und der Rückstand, welcher Bromwasserstoff entwickelt, wäh rend einigen Stunden in einem Teilvakuum stehen gelassen, worauf eine poröse halbfeste Masse zurückbleibt. Diese wird mit 25 Ge- wichtsteilen Aceton digeriert und die erhal tene Lösung mit 40 Gewichtsteilen trockenem Äther behandelt.
Nachdem einige Zeit abge kühlt wurde, wird die abgeschiedene kristal line Verbindung abfiltriert und mit einer Mischung von Aceton und Äther gewaschen, wobei ungefähr 18 Gewichtsteile 1-Benzyl-2- methyl - 3 - oxy - 4,5 - dicarbomethoxy - pyr - idiniumbromid erhalten werden,
welches nach Umkristallisieren aus Methanol und Äther einen Schmelzpunkt von 134 C, unter Zer setzung, aufweist. In dieser Verbindung kann das Brom durch Chlor ersetzt werden, wenn man Silberchlorid in Methanol einv#,ir- ken lässt.
Diese Verbindung kann, nach den für das entsprechende Chloridsalz in Beispiel 1 gemachten Angaben, in 2 - Methyl - 3 - oxy-4,5-dicarbomethoxy-pyridin übergeführt werden.
<I>Beispiel 5:</I> Eine Lösung von 36 Gewichtsteilen des genannten Ausgangsstoffes in 120 Raum teilen trockenem Benzol, in welchem 2,4 Ge wichtsteile pulverisiertes Natrium suspen- diert wurden, wird unter Rüchflusskühlung in einer Stickstoffatmosphäre gekocht. Die ; Auflösung d;#s Natriums wird dadurch er leichtert, dass man eine kleine Menge Na triummethylat oder Natriumäthylat bei gibt.
Es wird während einer Stunde weiter erwärmt, worauf' die Lösung gekühlt und ; mit einer Mischung von Eis und Essigsäure, welche eine kleine Menge Schwefelsäure ent hält, geschüttelt, wobei die Säuremenge der Menge des verwendeten Natriums entspricht. Der Benzolextrakt aus dem Gemische wird , mit Wasser, hernach mit Natriumbicarbonat- lösung und dann erneut mit Wasser ge waschen, getrocknet und vom Lösungsmittel befreit, worauf er mit wasserfreier alkoho lischer Salzsäure behandelt wird. Dabei , wird das Hydrochlorid der cyclisierten Ver bindung erhalten. Dieses scheidet sich nach Zugabe eines Überschusses von wasser freiem Ather als langsam festwerdendes. öliges Produkt ab.
Wird jedoch eine für die, Abscheidung ungenügende Menge Äther zu gegeben und wird die alkoholische Äther lösung wenigstens. während eines Tages der Einwirkung von Luft oder Sauerstoff a.us- gesetzt, dann scheidet sich das 1-Benzyl-2-liie- thyl - 3 - oxv-4,5-dica.rbomethoxy - pyridiniuln- chiorid nach und nach in Form. feiner, farb loser Nadeln ab, welche einen Schmelzpunkt von 146-l48 C aufweisen.
Aus diesem quaternären Salz kann das entsprechende ; phenolische Betgin wie folgt erhalten wer den: Eine kalte, konzentrierte wässerige Lösung von 7 Gewichtsteilen 1-Benzyl-2- methyl - 3 - oxy= 4,5 - dicarbomethoxy-pyridi- niumchlorid wird mit der berechneten Menge , lzalter 2n Natronlauge behandelt. Bald kri- stallisiert die Verbindung aus, wird abfil- triert, mit wenig Wasser gewaschen und im Vakuum getrocknet.
Die Ausbeute beträgt etwas mehr als 6 Gewichtsteile. Aus Wasser, k ristallisiert die Verbindung in flachen Na deln, welche Kristallwasser enthalten und teilweise bei zirka 100 C schmelzen. Beim Erhitzen wird das wasserfreie Produkt gelb und schmilzt bei 138-140 C zu einer röt lichen Flüssigkeit.
Es ist das phenolische Beta.in des 1-Benzyl-2-methyl-3-oxy-4,5-di- c.arbomethoxy-pyridiniumhydroxyds.
Eine Lösung von 6,5 Gewichtsteilen 1-.Benzyl - 2 - methyl - 3 - oxy-4, 5-dicarbometh- oxy-pyridiniumchlorid in 20 Raumteilen Methylalkohol wird mit Wasserstoff in Ge genwart von 0,5 Gewichtsteilen Palladium- Tierkohle (1 : 4) geschüttelt. In wenigen Mi nuten wird die berechnete Menge Wasser stoff aufgenommen.
Nach dem Abfiltrieren des Katalysators und Abdampfen des Methyl alkohols unter vermindertem Druck kristalli siert die Verbindung aus einem Alkohol- _Ither-Gemisch in farblosen Nadeln vom Schmelzpunkt 165 C unter Ziersetzung;
Aus- _beute 4,2 Gewichtsteile. Die gleiche Verbin dung wird erhalten, wenn die Hydrierung des oben genannten phenolischen Betains in -iner methylalkoho-lischen Lösung bei An- Wesenheit einer äquimolaren Menge Chlor- wasserstoff durchgeführt wird.
Das so erhal tene 2 - Methyl - 3 - ox3r,- 4, 5 - dica,rbomethoxy- pyridinhydrochlorid zeigt bei der Phenol Reaktion nach Gibbs Blaufärbung.
<I>Beispiel 6:</I> Bei der Aufarbeitung nach Beispiel 5 werden an Stelle von 2,4 Gewichtsteilen pulverisiertem Natrium 5,6 Gewichtsteile Natriummethylat verwendet. Im übrigen ,wird nach den Angaben in Beispiel 5 gear beitet, wobei dasselbe Endprodukt erhalten wird.
