Verfahren zur Herstellung von Alkalimetallenolaten von 21-Alkoxyoxalyl-ll-keto-progesteron Vorliegende Erfindung bezieht sich -auf die Herstellung von 2'1 -4lkoxyoxalyl-11-keto- progesteron-alkalimetallenolates. Diese neuen Verbindungen entsprechen der Formel

Das neue Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass man 11-Keto- Progesteron mit einem Alkyldiester der Oxal- säure in Gegenwart eines ein Alkalimetall ent haltenden Kondensationsmittels kondensiert, wobei ein 21-Alkoxyoxalyl-11-keto-progest.e- ron-alkalimetallenolat entsteht, ohne dass sich gleichzeitig Allioxyoxalylenolate an andern Stellungen des Moleküls bilden.
Die neuen Verbindungen haben Bedeu tung als stabile Formen der entsprechenden Ketosäuren, die sieh in fester Form befinden und lange Zeit aufbewahren lassen, Ihre Was- serlöslichkeit macht sie geeignet. für weitere Synthesen mittels Reaktionen in wässerigen Medien.
Die entsprechenden Ester, die durch Hydrolyse leicht erhältlichen Säuren und die Enolate selbst, sind wertvolle Zwischen produkte für die Herstellung biologisch ak tiver Verbindungen, wie Cortison und Ken- dalls Verbindung A (2 j1-Acetoxy-11-keto-pro- gesteron), wie im Beispiel 1.4 gezeigt wird.
Die neuen Verbindungen, dne besonderes Interesse besitzen, sind solche der vorstehen den allgemeinen Formel, in der M Kalium oder Natrium und R ein niedriges Alkyl, zum Beispiel Methyl, Äthyl, Propyl, Isopropyl, Butyl, Amyl, Hexyl, Heptyl, Octyl und der gleichen, ist,
wobei #LIethyl imd Äthyl bevor zugt werden.
Das Ausgangsmaterial des vorliegenden Verfahrens ist das 11 Ketc-progesteron, das nach bekannten Methoden (Reichstein und Fuchs., Helv. Chim. Acta, 23, 684 [1943]) her gestellt werden kann.
Zur Durchführung des neuen Verfahrens löst man in der Regel 11-Keto-progesteron im Alkanol, das zur Bildung des Oxalsäureesters verwendet wird oder in einem Lösungsmittel, das unter den Reaktionsbedingungen nicht reagiert, wie zum Beispiel Benzol oder Äther,
und vermischt mit dem gewählten Alkyldiester der Oxalsäure in Gegenwart eines ein Alkali metall enthaltenden Kondensationsmittels. Die bevorzugten Dhester der Oxalsäure sind nied rige Alkylester mit einem bis einschliesslich acht Kohlenstoffatomen, wobei die lulethyl- und Äthylester bevorzugt werden.
Als Alkali metall enthaltende Kondensationsmittel kann man Alkalimetallalkoholate, die Alkalimetalle, Alkalimetallhydride, Natriumamid, 'Triphenyl- methyl-Natrium und andere verwenden.
Von diesen werden die Natrium- und Kaliumalko- holate bevorzugt. Die Alkalialkoholate können ahne Lösungsmittel, in einem inerten Lösungs mittel gelöst oder suspendiert, oder in situ im Alkanol in dem das Alkalimetallalkoholat gebildet wurde, verwendet werden. Verwen- det man Kalium,
so verwendet man vorzugs- weise das nach bekannter Methode hergestellte Reaktionsprodukt mit tert. Butylalkohol.
Die Reaktionsmischiuig hält man dann 30 ylinuten bis 96 Minuten, vorzugsweise etwa eine bis 4 Stunden bei Temperaturen zwischen etwa 0 G und dem ,Siedepunkt der Mischung.
Die so gebildeten Alkalimetallenolate kän- nen dann durch Zugabe eines grossen Volu mens organischer Lösiw.gsmittel; in denen sie nicht löslich sind, wie Äther, abgeschieden werden. Das Enolat fällt so in der Regel als amorphe feste Substanz an und stellt die sta bile Form des entsprechenden Esters dar.
Man kann es auch ohne es zu isolieren als Zwischenprodukt für die Synthese der ent sprechenden 21-Glyoxalsäure oder als Zwi schenprodukt für die Einführung einer 21- Oxy- oder Acyloxygmuppe in das 11-Keto- progesteron verwenden, wie unter A hm Bei spiel 1 beschrieben.
Die folgenden Beispiele sollen das erfin dungsgemässe Verfahren erläutern: Beispieb <I>1</I> Zu einer Miscliimg von 3,4 eins 3y4 molar e_r Nätriummethylatlösung in Methanol, 0,45 cm3 absolutem Äthanol und 20 cm3 trockenem Ben zol,
von der zuerst .8 ems Destillat abdestilliert wurde, worauf man abkühlte, gibt man 2;3 ems Diäthyloxalat und eine Lösung von 3,28 g 11-Keto-progesteron in 38 ems trockenem Benzol. Die Lösung wird trüb, und es scheidet sich .ein gelber Niederschlag ab.
Das Reaktions- gemisch wird' <B>90</B> Minuten gerührt, 55 cm , Äther zugegeben und weitere 60 Minuten ge rührt, worauf man 130 ems Äther zusetzt. Der so entstandene gelbe Niederschlag des Natriiunenolats des 2:1-Ätho.Yyoxalyl-11=keto- progesterons wird filtriert, mehrmals mit je 50 cm3 Äther gewaschen und wiegt nach den i Trocknen 3,;65 g.
Die Waschflüssigkeit enthält 0;54 g nicht umgesetztes 11-Keto-progesteron. Die Ausbeute ist 81% der theoretischen oder, bezogen auf das umgesetzte 11-Keto-progeste- ron, praktisch quantitativ.
Das Vorhanden sein eines Natriimienolats wird durch die ausserordentliche Löslichkeit des Prodaldes in _'G'asser und durch den positiven Ferrichlorid- test für Enole (beim Lösen des Produktes in wässerigen und Ferriehlorid- lösiungen bildet. sich eine hellrote Färbung) bestätigt.
Die Struktur wird ferner bestätigt durch Überführung des Prodidites in das Ace tat von Kendalls Verbindung .1 (21-Acetoxy- 11-Keto-progesteron), wie nachstehend gezeigt. wird.
A. Eine Lösung von 2;65g Natriumenolat des 21-Äthoxyoxalyl-11-keto - progesterons in 25 em3 Methanol wird im Eis-,Salzbad auf etwa -20 'C gekühlt und unter Rühren eine Lösung 1,5 g Jod in 36 cm3 Methanol zugegeben, wozu man ungefähr eine Stunde benötigt. Nachdem man eine weitere Stunde bei 0 C gerührt hat, setzt man 1,7 cm3 einer 3,4 molaren, methanolischen Lösung von Na triummethylat zu.
Man rührt eine weitere Stunde bei '0 C und gibt 200 ems Wasser und 40- g festes. Natriumchlorid zur Mischung. Das ausgefällte rötlichbraune 21-Jod-11-keto- progesteron wird von der Reaktionsmischung abfiltriert, mit Wasser gewaschen, und ohne weitere Reinigung wie folgt in das 21-Acet- oxy-11-keto-progesteron umgewandelt.
Zu einer Mischung von 3 g Kaliumbiear- bonat und 18 g Essigsäure gibt man das feuchte 21- Jod -11- keto - progesteron und 200 ems Aceton. Die Mischung wird eine Stunde am Rückfluss gekocht und dann drei Tage bei Zimmertemperatur stehengelassen.
Das anorganische Material wird durch Fil tration entfernt und mit Aceton gewaschen. Die vereinigten Filtrate und Waschlaugen werden unter vermindertem Druck verdampft und der Rückstand in Benzol gelöst. Diese Lösung wird über 80 g Florisil (syntheti- sehes Magnesiumsilikat) chromatographiert. Die Säule wird mit 100, cm3 Portionen folgen der Lösungsmittel in der angeführten Reihen folge entwickelt:
11 Portionen Benzol, 7 Por tionen Äther, 4 Portionen aus 9 :Teilen Äther und 1 Teil Aceton, 7 Portionen aus 8 Teilen Äther und 2 Teilen Aceton und 1 Portion Aceton. Die Rückstände der ersten drei Por tionen aus 8 Teilen Äther und 2 Teilen Aceton werden vereinigt und aus Äthylacetat bis zum konstanten. Schmelzpunkt 181 bis 182,5 C iun- kristallisiert. Das so erhaltene '2,1-Acetoxy-11- keto-progesteron erweist sich bei der Infrarot analyse als mit einem authentischen Produkt identisch.
<I>Beispiel 2</I> In der gleichen Weise, wie im Beispiel 1 beschrieben, wird 11-Keto-progesteron mit Di- äthyloxalat und Kalium in tert. Butylalkohol in das Kaliumenolat d'es 21-Äthoxyoxalyl-11- keto--progesterons übergeführt.
Das erhaltene Kaliumenolat kann; wie das entsprechende Natriumenolat in das 2@1-Acetoxy-11-keto-pro- gesteron nach der Methode des Beispiels 1t1 übergeführt werden.
<I>Beispiel 3</I> In der gleichen Weise, wie im Beispiel 1 beschrieben, wird 11-Keto-progesteron mit Di- methyloxalat und Natrium in absolutem Me thanol in das 21-Methoxyoxalyl-11-keto-pro- gesteron übergeführt. Das erhaltene Natrium- enol'at kann wie das 21-Äthoxyoxalylderivat nach der Methode des Beispiels 1A in das 21-Acetoxy - 11- keto-progesteron übergeführt werden.
<I>Beispiel 4</I> In der gleichen Weise, wie im Beispiel 1 beschrieben, wird 11-Keto-progesteron durch Reaktion mit Dimethyloxalat -und Kalium in tert. Butylalkohol in das 21-Methoxyoxalyl-11- keto-progesteron übergeführt.
Das so erhaltene Kaliumenolat kann wie das entsprechende Na- triumenolat nach der im Beispiel 1A beschrie- benen Arbeitsweise in das 21-Acetoxy-11-keto- progesteron übergeführt werden.
<I>Beispiel 5</I> In der gleichen Weise, wie im Beispiel 1 beschrieben, wird 11-Keto-progesteron durch Umsetzung mit Dipropyl'oxalat und Natrium in Propylalkohol .in das 21-Propoxyoxalyl-11- Keto-progesteron übergeführt. Das erhaltene Natriumenolat kann wie das 21-Äthoxyoxalyl- .derivat, in das 2'1-Acetoxy-11'-keto@progesteron übergeführt werden.
<I>Beispiel 6</I> Nach der gleichen Arbeitsweise wie im Beispiel 1 wird 11-Keto-progesteron durch Umsetzung mit Diäthyloxalat und Natrium- amid in Benzol in das 21-Äthoxyoxalyl-11- keto-progesteron übergeführt.
In analoger Weise wie in den Beispielen 1 bis 6 werden durch Umsetzung von 11-Keto- progesteron mit den entsprechenden Dialkyl- oxalaten und einem Natrium- oder Kalium- alkoholat in einem Alkanol oder nicht realc- tionsfähigen Lösungsmittel. folgende Verbin dungen hergestellt Na-enolat des 21-Butoxyoxalyl 11-keto-pro-

The new compounds are important as stable forms of the corresponding keto acids, which are in solid form and can be stored for a long time. Their solubility in water makes them suitable. for further syntheses using reactions in aqueous media.
The corresponding esters, the acids easily obtainable by hydrolysis and the enolates themselves, are valuable intermediates for the production of biologically active compounds such as cortisone and Kendall's compound A (2j1-acetoxy-11-keto-progesterone), as shown in example 1.4.

The starting material for the present process is 11 ketc-progesterone, which can be produced by known methods (Reichstein and Fuchs., Helv. Chim. Acta, 23, 684 [1943]).
To carry out the new process, 11-keto-progesterone is usually dissolved in the alkanol that is used to form the oxalic acid ester or in a solvent that does not react under the reaction conditions, such as benzene or ether,
and mixed with the selected alkyl diester of oxalic acid in the presence of an alkali metal-containing condensing agent. The preferred dhesters of oxalic acid are lower alkyl esters with one to eight carbon atoms, inclusive, the lulethyl and ethyl esters being preferred.
As alkali metal-containing condensing agents, alkali metal alcoholates, the alkali metals, alkali metal hydrides, sodium amide, triphenylmethyl sodium and others can be used.
Of these, the sodium and potassium alcoholates are preferred. The alkali metal alcoholates can be used without a solvent, dissolved or suspended in an inert solvent, or in situ in the alkanol in which the alkali metal alcoholate was formed. If you use potassium,

The following examples are intended to explain the method according to the invention: Example <I> 1 </I> To a mixture of 3.4 one 3y4 molar sodium methylate solution in methanol, 0.45 cm3 absolute ethanol and 20 cm3 dry benzene,
8 ems of distillate was first distilled off, after which it was cooled, 2; 3 ems of diethyloxalate and a solution of 3.28 g of 11-keto-progesterone in 38 ems of dry benzene are added. The solution becomes cloudy and a yellow precipitate separates out.
The reaction mixture is stirred for 90 minutes, 55 cm of ether are added and the mixture is stirred for a further 60 minutes, after which 130 ems of ether are added. The yellow precipitate of the sodium enolate of 2: 1-Etho.Yyoxalyl-11 = keto-progesterone formed in this way is filtered, washed several times with 50 cm3 of ether each time and, after drying, weighs 3.65 g.
The washing liquid contains 0.54 g of unreacted 11-keto-progesterone. The yield is 81% of the theoretical or, based on the converted 11-keto-progesterone, practically quantitative.
The presence of a sodium enolate is confirmed by the extraordinary solubility of the prodaldes in water and the positive ferric chloride test for enols (when the product is dissolved in aqueous and ferric chloride solutions, a light red color is formed).
The structure is further confirmed by converting the prodidite to the acetate of Kendall's compound .1 (21-acetoxy-11-keto-progesterone) as shown below. becomes.
A. This solution is chromatographed over 80 g of Florisil (synthetic magnesium silicate). The column is developed with 100 cm3 portions of the solvents in the order listed:
11 portions of benzene, 7 portions of ether, 4 portions of 9 parts of ether and 1 part of acetone, 7 portions of 8 parts of ether and 2 parts of acetone and 1 portion of acetone. The residues of the first three Portions from 8 parts of ether and 2 parts of acetone are combined and removed from ethyl acetate until constant. Melting point 181 to 182.5 C iuncrystallized. The '2,1-acetoxy-11-keto-progesterone obtained in this way proves to be identical to an authentic product in the infrared analysis.

The potassium enolate obtained can; how the corresponding sodium enolate can be converted into 2 @ 1-acetoxy-11-keto-progesterone by the method of example 1t1.
<I> Example 3 </I> In the same way as described in Example 1, 11-keto-progesterone is converted into 21-methoxyoxalyl-11-keto-progesterone with dimethyl oxalate and sodium in absolute methanol . The sodium enolate obtained, like the 21-ethoxyoxalyl derivative, can be converted into 21-acetoxy-11-keto-progesterone by the method of Example 1A.
<I> Example 4 </I> In the same way as described in example 1, 11-keto-progesterone is obtained by reaction with dimethyl oxalate and potassium in tert. Butyl alcohol converted into 21-methoxyoxalyl-11-keto-progesterone.
The potassium enolate obtained in this way, like the corresponding sodium enolate, can be converted into 21-acetoxy-11-keto-progesterone by the procedure described in Example 1A.
<I> Example 5 </I> In the same way as described in Example 1, 11-keto-progesterone is converted into 21-propoxyoxalyl-11-keto-progesterone by reaction with dipropyl oxalate and sodium in propyl alcohol. The sodium enolate obtained can, like the 21-ethoxyoxalyl derivative, be converted into 2'1-acetoxy-11'-keto @ progesterone.
<I> Example 6 </I> Using the same procedure as in Example 1, 11-keto-progesterone is converted into 21-ethoxyoxalyl-11-keto-progesterone by reaction with dietary oxalate and sodium amide in benzene.
