Description
Verfahren zur Darstellung von Salleylfor mylglyeolester. Es wurde gefunden, dass man zu einem therapeutisch wertvollen Ester der Salicy l säure dadurch gelangt, dass man Alkalisalze der Salicylsäure mit Ameisensäure -ss - chlor- äthylester erhitzt. Besonders gut verläuft die Umsetzung, wenn ein Überschuss an Ameisen säure-p-chloräthylester verwendet wird. Der nicht in Reaktion tretende chlorierte Fett säureester kann leicht und vollständig wieder gewonnen werden.
Der neue Salicylsäui,eester ist eine hoch siedende Verbindung von niedrigem Schmelz punkt (26 ) und sehr schwachem Geruch. Er eignet sich deshalb vorzüglich als perku- tan anwendbares Antirheumatika, das sich wegen seiner guten Öllöslichkeit leicht fetten Salben einverleiben lässt. Gleichzeitig in ge eigneter Salbengrundlage mit Histamin zu sammen in die Haut eingerieben, besitzt der Salicylformylglycolester die Eigenschaft, die ser eine erhöhte Durchlässigkeit für Histamin zu verleihen.
Infolge der durch gleichzeitige Verwendung von Salitylformylglycolester her beigeführten erhöhten Durchlässigkeit der Haut, kann die Menge des teuren Histamins herabgesetzt werden, ohne dass der tberapeu- tische Erfolg geringer wird.
Der Salicylformylglycolester soll als Arz neimittel Verwendung finden.
Beispiel: 50 Gewichtsteile trockenes salicylsaures Natrium werden mit 75 Gewichtsteilen Amei- sensäure-p-chloräthylester (dargestellt durch Erwärmen von hochprozentiger Ameisensäure mit Äthylenchlorhydrin bei Gegenwart von wasserfreiem Chlorealcium) 24 Stunden unter Rühren im Ölbad von I55-165 am Rück flusskühler erhitzt. Darin lässt man erkalten und giesst die Reaktionsmasse unter Rühren in 75 Gewichtsteile kaltes Wasser.
In die wässerige Lösung lässt man unter Rühren etwa 6 Gewichtsteile 10 /oige Sodalösung einlaufen, bis die wässerige Phase deutlich erkennbar lackmus-alkalisch reagiert. Dann trennt man das Öl ab und wäscht nochmals mit Wasser. Das verbleibende Öl wird im Vakuum destilliert. Dabei geht bis 100 Wasser und unveränderterAmeisensäurechlor- ätbylester über. Bei höherem Erhitzen de stilliert der Salicylformylglycolester unter etwa 11 mm Druck bei 163-165 . Es wer den 55-56 Gewichtsteile erhalten. Durch nochmalige Destillation unter vermindertem Druck erhält man den Ester völlig rein.
Bei längerem Stehen in einem kühlen Raum er starrt er zu farblosen Kristallen vom Schmelz punkt 26 .
Process for the preparation of Salleylform mylglyeolester. It has been found that a therapeutically valuable ester of salicylic acid can be obtained by heating alkali metal salts of salicylic acid with formic acid -ss-chloroethyl ester. The reaction proceeds particularly well if an excess of formic acid p-chloroethyl ester is used. The non-reacting chlorinated fatty acid ester can be easily and completely recovered.
The new salicylic acid ester is a high-boiling compound with a low melting point (26) and a very faint odor. It is therefore eminently suitable as an anti-inflammatory drug that can be used percutaneously, and because of its good oil solubility, it can be easily incorporated into ointments. At the same time rubbed into the skin in a suitable ointment base with histamine, the salicylformylglycol ester has the property of making it more permeable to histamine.
As a result of the increased permeability of the skin brought about by the simultaneous use of salityl formyl glycol ester, the amount of expensive histamine can be reduced without reducing the therapeutic success.
The salicylformyl glycol ester is said to be used as a medicinal product.
Example: 50 parts by weight of dry sodium salicylic acid are mixed with 75 parts by weight of p-chloroethyl formate (prepared by heating high-percentage formic acid with ethylene chlorohydrin in the presence of anhydrous chlorealcium) for 24 hours on a reflux condenser in an oil bath from 155-165. It is left to cool and the reaction mass is poured into 75 parts by weight of cold water with stirring.
About 6 parts by weight of 10% soda solution are allowed to run into the aqueous solution with stirring until the aqueous phase clearly reacts in a litmus-alkaline manner. Then the oil is separated off and washed again with water. The remaining oil is distilled in vacuo. Up to 100% of water and unchanged formic acid chloroethyl ester are transferred. When heated to a higher temperature, the salicylformyl glycol ester distills under about 11 mm pressure at 163-165. It who received the 55-56 parts by weight. Another distillation under reduced pressure gives the ester completely pure.
