Verfahren zur Darstellung einer Verbindung des 1-Phenyl-2,3-dimethyl-4- isopropyl-5-pyrazolons rnit Isopropylallylbarbitursäui-e. Es ist bekannt, dass sowohl 1-Phenyl-2,3- diinethy 1-5-p@-razolon, als auch 1-Phenyl-2,3- (limethyl-4-dimethylamino-5-pyra.zolon sich mit Dialkyl- und Arylalky lbarbitursäuren verbinden (Patentschriften Nr.
120098, 1?(r444/46, 121981/85 und 9775l, 113103!05, l13()56, 113295e97). Entsprechende Verbin dungen mit 4-Alkylpyrazolonen sind bis ,jetzt noch nicht hergestellt worden.
Wie nun gefunden wurde, verhält sich (las 1-Phenyl-2,3-dimethyl-4-isopropyl-5-pyr- azolon nicht gleich wie das 1-Phenyl-2,3-di- inethyl-5-pyrazolon und das 1-Phenyl-2,3-di- methyl-4-dimethylamino-5-pyrazolon. Es geht nämlich nur mit solchen C,C-disubstituier- ten Barbitursäuren Verbindungen ein, die nur gesättigte oder höchstens nur einen un gesättigten Substituenten besitzen. Sind leide Substituenten ungesättigt, so unter bleibt die Bildung von Verbindungen.
Die Darstellung der neuen Verbindungen gestaltet sich bei der grossen Neigung der Ausgangsstoffe zum Zusammentritt sehr ein fach. Man schmilzt das 1-Phenyl-2,3-dime- thyl-4-isopropyl-5-pyrazolon mit der äqui- molekularen Menge einer Dialkyl- oder Aryl- alkylbarbitursäure zusammen oder lässt die Ausgangsstoffe in Gegenwart eines Lösungs- bezw. Verdünnungsmittels aufeinander ein wirken.
Die Vereinigung des 1-Phenyl-2,3- climeth-#Tl-4-isopropyl-5-pyrazolons mit den Dialkyl- bezw. Arylalkylbarbitursäuren er folgt stets im molekularen Verhältnis auch dann, wenn von dem einen oder andern Be standteil wesentlich grössere Mengen, z. B. 2 Moleküle, vorhanden sind.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung einer Verbin dung des 1-Phenyl-2,3-dimethyl-4-isopropyl- 5-py-razolons mitIsopropylallylbarbitursäure, welches dadurch gekennzeichnet ist, dass man 1. - Phenyl-2,3-dimethyl-4-isopropyl-5-pyrazo- lon und Isopropylallylbarbitursäure im Mo- lekalarverhältnis 1 :1 aufeinander einwir- ken lässt. Die Ausgangsstoffe können zum Beispiel in geschmolzenem Zustand oder in einem Lösungsmittel aufeinander einwirken gelassen werden.
Die Verbindung des 1-Phenyl-2,3-dime- l.liyl-4-isopropyl-5-pyrazolons mit Isopropyl- allylbarbitursäure kristallisiert in feinen farblosen Nadeln, die bei<B>128'</B> schmelzen. Sie lässt sich durch Behandeln mit Lösungs mitteln nicht in die Bestandteile zerlegen. In organischen Lösungsmitteln ist sie leicht. in Wasser schwer löslich. Sie soll als Arz neimittel Verwendung finden, da sie sowohl analgetische bezw. antipyretische, als auch schlafmachende Wirkungen besitzt.
<I>Beispiel 1:</I> Man löst in der Wärme 105 Teile Iso- propylallylbarbitursäure in 600 Teilen Ben zol und 115 Teile 1-Phenyl-2,3-dimethyl-4- isopropyl-5-pyrazolon in 200 Teilen Benzol, giesst die Lösungen warm zusammen und versetzt mit wenig Petroläther bis zur begin nenden Trübung. Beim Abkühlen scheiden sich 200 Teile der neuen Verbindung in fei nen, farblosen Nadeln aus, welche bei 128 schmelzen. Schmelzpunkt und Aussehen ver ändern sich beim Umkristallisieren aus ver dünntem Alkohol oder verdünntem Aceton nicht. Der Rest der Verbindung kann durch Eindampfen der Mutterlauge gewonnen wer den.
<I>Beispiel 2:</I> 105 Teile Isopropyl-allyl-barbitursäure werden zusammen mit 115 Teilen 1-Phenyl- 2,3-dimethyl-4-isopropyl-5-pyrazolon gut verrieben. Der Schmelzpunkt des Gemenges ist unscharf bei 92 bis 100 . Das Gemisch wird nun mit 4000 Teilen Wasser aufge- schlemmt und bei 50 bis 55 verrührt. Nach 2 Stunden ist der Schmelzpunkt scharf bei 1.27 .
Nach einmaligem Umkristallisieren aus verdünntem Alkohol steigt der Schmelz punkt auf<B>128'.</B> <I>Beispiel 3:</I> 210 Teile Isopropyl-allyl-barbitursäure werden mit 230 Teilen 1-Phenyl-2,3-dime- thyl-4-isopropyl-5-pyrazolon gut vermiseht. Das Gemenge wird in einem offenen Gefäss durch ein Ölbad unter Rühren angewärmt. Wenn die Temperatur im 01 auf 130 ge stiegen ist, beginnt das Gemisch langsam zu schmelzen.
Bei einer Ölbadtemperatur von 150 bildet das Ganze einen klaren, fast farblosen Schmelzfluss. Dieser wird unter Rühren abgekühlt und erstarrt bald. Die entstandene, fast farblose Verbindung ist leicht pulverisierbar. Sie schmilzt bei 126 bis 128 . Der Schmelzpunkt steigt durch einmaliges Umkristallisieren aus verdünn tem Alkohol auf 128 . Die in Nadeln kri stallisierende Verbindung ist farblos.
