Verfahren zur Darstellung einer Verbindung des Sparteins mit Isopropylallylbarbitursäure. Verbindungen des Sparteins mit C,C-di- substituierten Barbitursäuren sind noch nicht bekannt. Es wurde nun gefunden, dass Spar tein mit C,C-disubstituierten Barbitursäuren gut kristallisierende, salzartige Verbindungen eingeht. Sie entstehen, wenn man die Aus gangsstoffe in geschmolzenem Zustande oder in einem Lösungsmittel aufeinander einwirken lässt.
Man kann die Verbindungen auch durch doppelte Umsetzung gewinnen, indem man Salze des Sparteins mit Salzen von C,C-di- substituierten Barbitursäuren in einem ge eigneten Lösungsmittel zu9# Reaktion bringt. Je nach der Arbeitsweise erhält man Verbin dungen, die Spartein und Barbitursäuren im Molekularverhältnis 1 : 1 oder 1 : 2 enthalten.
Zur Herstellung von Verbindungen, bestehend aus einem Molekül Spartein und 2 Molekülen einer C,C-disubstituierten Barbitursäure ver fährt man derart, dass man die Ausgangsstoffe im berechneten Verhältnis unmittelbar oder in einem geeigneten Lösungsmittel gegebenen- falls unter Verwendung eines Überschusses des einen Ausgangsstoffes aufeinander ein wirken lässt.
Dass Verbindungen von 1 Molekül Spar tein mit 1 oder 2 Molekülen C,C - disubsti- tuierter Barbitursäuren erhalten werden, geht deutlich aus den Auftau-Schmelzdiagrammen nach Rheinboldt (Journal für praktische Che mie, 2. Folge, Band 111, 1925, Seite 242) hervor. Die Verbindungen kommen aueh in wässeriger Lösung vor, was sich aus der Tatsache ergibt, dass man sie durch Auskri stallisieren aus wässeriger Lösung gewinnen kann.
Spartein wird in der Heilkunde zur Ver besserung der Herz- und Kreislauftätigkeit angewandt. Es hat sich gezeigt, dass die Herz- und Kreislaufwirkung des Sparteins durch gleichzeitige Darreichung von C,0 - disubsti- tuierten Barbitursäuren vorteilhaft unterstützt wird. Durch die Darstellung der neuen Ver bindungen ist die Möglichkeit geboten, die beiden Komponenten in Form von leichtlös lichen, haltbaren Salzen von bestimmter Zu sammensetzung und besonderer Wirksamkeit zu verabfolgen.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Darstellung einer Ver bindung des Sparteins mit Isopropylallyl- barbitursäure, welches dadurch gekennzeich net ist, dass man Spartein und Isopropylallyl- barbitursäure im Molekularverbältnis 1:2 aufeinander einwirken lässt. Die Einwirkung erfolgt in geschmolzenem Zustande oder in einem Lösungsmittel.
Die Verbindung des Sparteins mit Iso- propylallylbarbitursäure schmilzt bei 122 bis 123 . Sie ist in Wasser ziemlich, in Alkohol und Äther leicht und in Petrölgther schwer löslich.
Die neue Verbindung soll als Arzneimittel Verwendung finden.
<I>Beispiel 1:</I> 420 Teile Isopi-opylallylbarbitursä ure und 234 Teile Spartein werden in der Atmosphäre eines indifferenten Gases erwärmt, bis eine klare Lösung entstanden ist. Das Reaktions produkt erstarrt beim Erkalten kristallinisch. Die völlig erstarrte Masse wird zerschlagen und gepulvert. Man erhält ein Pulver vom Schmelzpunkt 122-123 . <I>Beispiel 2:</I> Man erwärmt 420 Teile Isopropylallyl- barbitursäure und 234 Teile Spartein in 200 Teilen absolutem Alkohol, bis alles klar ge löst ist.
Hierauf versetzt man mit 8000 Teilen Benzin und impft. Das Reaktionsprodukt fällt als feines, weisses Kristallmehl aus. Es zeigt die Eigenschaften der unter Beispiel 1 be schriebenen Verbindung.
Process for the preparation of a compound of sparteine with isopropylallylbarbituric acid. Compounds of sparteine with C, C-disubstituted barbituric acids are not yet known. It has now been found that spar teine with C, C-disubstituted barbituric acids forms salt-like compounds which crystallize well. They arise when the starting materials are allowed to act on one another in a molten state or in a solvent.
The compounds can also be obtained by double conversion by bringing salts of sparteine to reaction with salts of C, C-di-substituted barbituric acids in a suitable solvent. Depending on the mode of operation, compounds are obtained which contain sparteine and barbituric acids in a molecular ratio of 1: 1 or 1: 2.
To prepare compounds consisting of one molecule of sparteine and 2 molecules of a C, C-disubstituted barbituric acid, the procedure is such that the starting materials are added to each other in the calculated ratio directly or in a suitable solvent, if necessary using an excess of one starting material works.
The fact that compounds of 1 molecule of spar teine with 1 or 2 molecules of C, C - disubstituted barbituric acids are obtained is clear from the thawing melt diagrams according to Rheinboldt (Journal for practical chemistry, 2nd part, volume 111, 1925, page 242 ). The present invention relates to a process for the preparation of a compound of sparteine with isopropylallyl barbituric acid, which is characterized in that sparteine and isopropylallyl barbituric acid are allowed to act on one another in a molecular ratio of 1: 2. The action takes place in the molten state or in a solvent.
The compound of sparteine with isopropylallylbarbituric acid melts at 122 to 123. It is fairly soluble in water, easily soluble in alcohol and ether, and sparingly soluble in petroleum ether.
The new compound is said to be used as a drug.
<I> Example 1: </I> 420 parts of isopi-opylallylbarbituric acid and 234 parts of sparteine are heated in the atmosphere of an inert gas until a clear solution has formed. The reaction product solidifies in crystalline form on cooling. The completely solidified mass is crushed and powdered. A powder with a melting point of 122-123 is obtained. <I> Example 2 </I> 420 parts of isopropylallylbarbituric acid and 234 parts of sparteine in 200 parts of absolute alcohol are heated until everything has dissolved clearly.
Then 8000 parts of gasoline are added and vaccinated. The reaction product precipitates as fine, white crystal flour. It shows the properties of the compound described in Example 1 be.
