Verfahren zur Herstellung von racemiseliem 1-(in-Oxyplienyl)-2-aminopi#opaiiol(1). Es wurde gefunden, dass man 1-Oxyphenyl- 2-amitioalkohole(1) in Racemforin erhält, wenn man 1-drehende 1- Oxyphenyl - 2 - ketoalko- liole(1) in Gegenwart von Ammoniak oder primären oder sekundären Aminen der kataly tischen Reduktion bei erhöhter Temperatur unterwirft.
Als Katalysatoren können Metalle der Nickelgruppe oder Edelmetalle, wie Platin odet Palladium, oder endlich Kupfer verwandt werden. Bei Verwendung von Edelmetall- katalysatoren wird die Reaktion vorzugsweise bei mässig erhöhter Temperatur, etwa zwischen 40 und 80 , beispielsweise bei etwa l#)0 , ausgeführt. Bei Verwendung eines Kataly- sators der Nickelgruppe wird die Reaktion vorzugsweise bei einer Temperatur zwischen etwa 60 und<B>180'</B> ausgeführt, beispielsweise bei etwa 80 .
In jedem Falle ntuss die Tem peratur der Beschaffenheit des Katalysators angepasst sein, das heisst bei der Anwendung eines Katalysators geringerer Wirksamkeit wird die Reaktion innerhalb des angegebenen Bereiches zweck;nässig bei höherer Temperatur ausgeführt, während sie bei Anwendung eines Katalysators höherer Wirksamkeit bei niederer Temperatur ausgeführt werden kann.
Bei Anwendung von Ammoniak oder Aral- kylaminen erhält man Verbindungen mit pri mären Aminogruppen, während man bei Aü- wendung von Alkylaminen die entsprechenden Alkylaminoverbindungen erhält.
Wenn man jedoch Aralkylamine in Gegenwart eines Katalysators der Nickelgruppe verwendet., erhält man die entsprechende Aralkylamino- verbindung, so dass zur Herstellung der pri mären Aminoverbindung eine weitere Hydrie rung in Gegenwart eines Edelmetallkatalysa- tors notwendig ist; die Aralkylgruppe wird dabei abgespalten.
Bei Anwendung von Ara1- kylaminen in Gegenwart eines Edelmetall- katalysators erhält man die eine pimäre Amitiogruppe enthaltene Verbindung direkt.
Die Reaktion wird zweckmässig in alko holischer Lösung ausgeführt. Da die verwendeten optisch aktiven Oxy- phenylacetylcarbinole sehr leicht zugängliche Ausgangsmaterialien darstellen, so bedeudet das Verfahren einen erheblichen Fortschritt gegenüber den bisher bekannten Gewinnungs weisen der als Heilmittel wichtigen Amino- alkohole.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von racemi- schem 1-(m-Oxyphenyl)-2-ainiiio-propanol(1), das dadurch gekennzeichnet ist, das man 1- m-Oxyphenylacetylcarbinol bei erhöhter Tem peratur in Gegenwart einer Verbindung, welche eine Aminogruppe enthält; mit ;Wasserstoff in Gegenwart eines Edelnietallkatalysators behandelt.
Das neue Verfahrensprodukt bildet in Form seines Chlorhydrats weisse leicht in Wasser lösliche Kristalle vom Schmelzpunkt <B>178</B> o C. Es soll als Arzneimittel Verwendung finden.
Beispiel <I>1:</I> 33 gr 1-m-Oxyphenylacetylcarbinol, 23 gr Benzylamin und 250 cm' Alkohol werden bei 40-50 mit Palladium hydriert. Sobald die Wasserstoffaufnahme zum Stillstand ge kommen ist, wird vom Katalysator abgesaugt, der Alkohol im Vakuum abdestilliert, der Rückstand mit Wasser aufgenommen und Wasserunlösliches mit Äther ausgezogen. Der wässerige Auszug wird mit Salzsäure sauergestellt und im Vakuum bis zur Trockne eingedampft. Die erhaltene kristalline islasse wird aus Propylalkohol und Äther umgelöst.
Erhalten: 30 gr racemisches m-Oxyphenyl- propanolaminchlorhydrat vom Schmelzpunkt 178o C.
<I>Beispiel 2:</I> 8,3 gr 1-ni-Oxyphenylacetylcarbinol werden in 200 cm' einer mindensten 5 o/oigen alko holischen Ammoniaklösung aufgelöst und nach Zusatz von aktiviertem Palladium mit Wasser stoff unter schwachem Erwärmen hydriert. Nach Aufnahme der berechneten Menge Was- serstoff saugt man vom Katalysator ab, de stilliert den Überschuss an Alkohol im Vakuum ab und verrührt den vollständig trocknen Rückstand mit alkoholischer Salzsäure bis zur kongosauren Reaktion.
Auf Zusatz von Aceton kristalliert das racemische m-Oxy- phenylaminopropanolchlorhydrat aus, das nach dem Umkristallieren aus Propylalkohol unter Zusatz von Äther den Schmelzpunkt 178 C zeigt.
