Description
Verfahren zur Gewinnung einer hochkonzentrierten wässerigen Lösung eines Salzes der 3.5-Dijod-4-pyridon-N-essigsäure. Die Verwendung der für pharmazeutische Zwecke, insbesondere für die röntgenbgra- phische Darstellung von Organen im tierischen Körper, wertvollen Halogenpyridoncarbon oder -sulfonsäuren begegnet in der Praxis vielfach infolge der nicht ausreichenden Lös lichkeit ihrer Metallsalze Schwierigkeiten. Im besonderen zeigen die dihalogenierten Pyridonderivate im allgemeinen eine derart geringe Löslichkeit in Wasser, dass ihre prak tische Verwendung dadurch häufig unmöglich wird.
Es wurde nun gefunden, dass man hoch konzentrierte Lösungen von Pyridinderivaten gewinnen kann, wenn man durch zur Salz bildung befähigte saure Gruppen substituierte Oxy- bezw. Oxohalogenpyridone, wie die 3. 5-Dijod-4-pyridon-N-essigsäure, in ihre Salze mit einem aliphatischen Amin umwandelt, wobei als Lösungsmittel Wasser dient.
Gegenstand vorliegender Erfindung ist ein Verfahren zur Herstellung der hochkonzen trierten, als Röntgenkontrastmittel verwend baren Lösung eines Salzes der 3.5-Dijod-4- pyridon-N-essigsäure, das dadurch gekenn- zeichnet ist, dass man die 3. 5-Dijod-4-py ri- don-N-essigsäure in wässerigem Medium mit Diäthylamin behandelt.
<I>Beispiel:</I> 50,6 gr 3,5-Dijod-4-pyridon-N-essigsäure werden mit einer Lösung von 10 gr Diäthyl- amin in 60 cm3 Wasser unter Erwärmen in Lösung gebracht, worauf man die Lösung mit Wasser auf 100 ein' auffüllt. 100 eins dieser Lösung enthalten 60 gr Diäthylaminsalz --, 50,6 gr 3.5-Dijod-4-pyridon-N-essigsäure.
Dagegen enthalten 100 cms einer gesät tigten Lösung des Natriumsalzes nur 33 gr Natriumsalz = 31,3 gr 3,5-Dijod-4-pyridon- N-essigsäure.
Process for obtaining a highly concentrated aqueous solution of a salt of 3,5-diiodo-4-pyridone-N-acetic acid. The use of halopyridone carbon or sulfonic acids, which are valuable for pharmaceutical purposes, in particular for the X-ray graphical representation of organs in the animal body, often encounters difficulties in practice due to the insufficient solubility of their metal salts. In particular, the dihalogenated pyridone derivatives generally have such a low solubility in water that their practical use is often impossible.
It has now been found that highly concentrated solutions of pyridine derivatives can be obtained if oxy- or substituted oxy groups capable of salt formation are used. Oxohalopyridones, such as 3. 5-diiodo-4-pyridone-N-acetic acid, are converted into their salts with an aliphatic amine, water being used as the solvent.
The present invention relates to a process for the preparation of the highly concentrated solution, which can be used as an X-ray contrast medium, of a salt of 3,5-diiodo-4-pyridone-N-acetic acid, which is characterized in that the 3. 5-diiodo-4- treated pyridon-N-acetic acid in an aqueous medium with diethylamine.
<I> Example: </I> 50.6 g of 3,5-diiodo-4-pyridone-N-acetic acid are brought into solution with a solution of 10 g of diethylamine in 60 cm3 of water while warming, whereupon the solution fill up with water to 100 a '. 100 one of these solutions contain 60 grams of diethylamine salt, 50.6 grams of 3.5-diiodo-4-pyridone-N-acetic acid.
In contrast, 100 cms of a saturated solution of the sodium salt contains only 33 grams of sodium salt = 31.3 grams of 3,5-diiodo-4-pyridone-N-acetic acid.
