Verfahren zur Herstellung von Acetyl-I:-stroplianthidin. Das aus den Samen von Strophanthus Kombe oder aus andern Drogen gewonnene und als Herzmittel verwendete k-Strophan- thin, das sowohl in einer amorphen, als auch in seiner luistallisierten Modifikation ein Gemisch von Glykosiden darstellt und des halb in seinen Wirkungen nicht einheitlich ist, liefert bei der Spaltung mit Säuren das k-Strophantidin. Dieses ist in zwei Modifika tionen bekannt, deren Schmelzpunkte etwa, bei<B>173</B> und 232 liegen.
Die pharmakologi schen Wirkungen des k-Strophanthidins glei chen qualitativ denen des k-Strophanthins. quantitativ sind sie geringer.
Feist (Berichte der deutschen chemischen Gesellschaft, Band 33, Seite 2080, 1900) konnte keine charakteristischen Acylierungs- produkte des k-Strophanthidins erhalten. Windaus und Hermanns (Berichte der deut schen chemischen Gesellschaft, Band 48., Seite 986, 1915) gelang es, eine Benzoylver- bindung herzustellen. Deren pharmakologi sche Prüfung durch Straub (Biochemische Zeitschrift, Band 75, Seite 143, 1916) ergab aber, dass sie am Frosch nur noch ein Zehn tel der Wirksamkeit des k-Strophant,hidins besass.
Es wurde gefunden, dass man zu sehr wirksamen Präparaten des k-Strophantliidins gelangen kann, wenn man acylierende Mit tel der aliphatischen Reihe einwirken lässt. -wie Halogenide oder Anhydride aliphatischer Säuren. Vorteilhaft ist dabei die Gegenwart tertiärer Amine, zum Beispiel von Pyridinen oder alkylierten Anilinen. Gegebenenfalls können bei dem Verfahren auch indifferente Lösungsmittel, zum Beispiel Chloroform, be nutzt werden. Besonders reine Produkte er hält man, wenn man bei Ausführung der Reaktion höhere Temperaturen vermeidet.
Bei Anwendung von Acetylchlorid oder Essigsäureanhydrid erhält man Acetyl-k- strophanthidin, das eine stärkere pharmako logische Wirksamkeit besitzt als das k-Stro- phanthidin. Gegenüber dem k-Strophanthi- din besitzt es den Vorzug, dass es chemisch einheitlich und daher in seinen Eigenschaf ten und Wirkungen konstant ist. Beide Mo- difikationen .des li:-Stropha.nthidins liefern dasselbe Acetylprodukt.
Beispiel <I>1:</I> 10 Teile k-Strophanthidin werden in<B>100</B> Teilen Pyridin gelöst und die Lösung bei Zimmertemperatur mit 100 Teilen Essig säureanhydrid versetzt. Nach 24 Stunden wird das Acetylierungsprodukt durch Ein giessen in Eiswasser ausgefällt, abfiltriert und aus 96%igem Alkohol umkristallisiert. Das Acetyl-k-strophanthidin kristallisiert aus Alkohol in farblosen glänzenden Prismen, die bei etwa 248-249' C schmelzen. Der Schmelzpunkt ist bei allen Stoffen dieser Gruppe abhängig von der Dauer und Schnel ligkeit des Erhitzens.
Beispiel <I>2:</I> 3 gr k-Strophanthidin werden mit 30 cm" Essigsäureanhydrid eine Stunde lang am Rückflusskühler gekocht. Es werden dann vorsichtig 30 cm" absoluter Alkohol zugege ben und die Lösung noch einige Zeit im Sieden erhalten. Sodann werden Essigester und Alkohol auf dem Wasserbade abdestil- liert und der Rückstand aus Alkohol um.. kristallisiert. Die Ausbeute an Acetyl-k-. strophanthidin beträgt etwa 85% der Theorie.
<I>Beispiel 3:</I> 3 gr k-St@rophanthidin werden mit 12 ein' Pyridin übergossen und nach Zusatz von 12 cm" Essigsäurea.nhydrid 3 Minuten lang am Rückflusskühler zu gelindem .Sieden er- hitz. Nachdem vorsichtig 20 cm' absoluter Alkohol zugegeben worden sind, wird noch einige Zeit im Sieden erhalten und dann durch die Lösung Wasserdampf geleitet. Der grösste Teil des entstandenen Acetyl-k- strophanthidins scheidet sich dabei kristalli nisch ab.
Der Rest kann durch Extraktion mit Chloroform aus der Mutterlauge abge trennt werden. Die Gesamtausbeute beträgt 85 bis 90 %.
Process for the preparation of acetyl-I: -stroplianthidin. The k-strophanthin obtained from the seeds of Strophanthus Kombe or from other drugs and used as a heart remedy, which is a mixture of glycosides in both an amorphous and its luistallized modification and is therefore not uniform in its effects, supplies k-strophantidine when cleaved with acids. The pharmacological effects of k-strophanthidine are qualitatively similar to those of k-strophanthidine. quantitatively they are lower.
Feist (Reports of the German Chemical Society, Volume 33, Page 2080, 1900) could not obtain any characteristic acylation products of k-strophanthidine. Windaus and Hermanns (Reports of the German Chemical Society, Volume 48., Page 986, 1915) succeeded in producing a benzoyl compound. However, their pharmacological test by Straub (Biochemische Zeitschrift, Volume 75, page 143, 1916) showed that they only had a tenth of the effectiveness of the k-strophant, hidin, on the frog.
It has been found that very effective preparations of k-strophantliidine can be obtained if acylating agents of the aliphatic series are allowed to act. - such as halides or anhydrides of aliphatic acids. The presence of tertiary amines, for example pyridines or alkylated anilines, is advantageous. If necessary, inert solvents, for example chloroform, can also be used in the process. Particularly pure products are obtained if higher temperatures are avoided when carrying out the reaction.
When using acetyl chloride or acetic anhydride, acetyl-k-strophanthidine is obtained, which has a stronger pharmacological activity than k-strophanthidine. It has the advantage over k-strophantidine that it is chemically uniform and therefore constant in its properties and effects. Both modifications of the li: -stropha.nthidin produce the same acetyl product.
Example <I> 1: </I> 10 parts of k-strophanthidine are dissolved in <B> 100 </B> parts of pyridine and 100 parts of acetic anhydride are added to the solution at room temperature. After 24 hours, the acetylation product is precipitated by pouring it into ice water, filtered off and recrystallized from 96% alcohol. The acetyl-k-strophanthidin crystallizes from alcohol in colorless, shiny prisms that melt at around 248-249 ° C. The melting point of all substances in this group depends on the duration and speed of heating.
Example <I> 2: </I> 3 g of k-strophanthidine are boiled with 30 cm "acetic anhydride on the reflux condenser for one hour. 30 cm" of absolute alcohol are then carefully added and the solution is kept boiling for some time. Then ethyl acetate and alcohol are distilled off on the water bath and the residue is recrystallized from alcohol. The yield of acetyl-k-. strophanthidin is about 85% of theory.
<I> Example 3: </I> 3 g of k-st @ rophanthidin are poured over with 12 a pyridine and after addition of 12 cm "acetic anhydride heated to gentle boiling for 3 minutes on the reflux condenser cm 'of absolute alcohol has been added, is kept boiling for some time and then steam is passed through the solution, with the greater part of the acetyl-strophanthidine precipitating out in crystalline form.
The remainder can be separated from the mother liquor by extraction with chloroform. The overall yield is 85 to 90%.
