Abstract

L'invention porte sur la découverte de séquences de rétrovirus caractérisées par RT-PCR sur des amorces <i>gag</i> et <i>env</i>; sur l'ARN de particules rétrovirales RT-positives produites par des lignées de cellules de lymphoblastoïdes B de patients atteints de sclérose en plaque (SEP). On a également découvert des variantes desdites séquences présentant une forte homologie avec le sous-groupe potentiellement fonctionnel RGH de la famille du rétrovirus endogène humain RTVL-H/HERV-H. Ces mêmes séquences ont également été spécifiquement découvertes dans la fraction particulaire d'une série d'échantillons du plasma de patients souffrant de SEP, mais absentes lors des contrôles. Des transferts de South-Western ont montré la présence d'une protéine de fixation d'un acide nucléique dont la taille et la fonction correspondent à celles de la protéine nucléocapsidique Gag NC d'autres rétrovirus. On a également trouvé des indications sur la transmission de rétrovirus à des leucocytes mononucléaires d'humains et d'animaux sains. On a en outre identifié dans des cultures de cellules dérivant du sang périphérique de patients souffrant de SEP de 5 à 15 % de CD3 + cellules T, et une variante épicée spécifique de la séquence <i>env</i> comportant une partie de la région <i>pol</i>.
