Abstract
TRÊS REATORES DE RESINA EM SÉRIE PARA SINTETIZADOR DE PEPTÍDEOA presente invenção refere-se a um dispositivo de Síntese de Peptídeos em Fase Sólida (SPPS) e a um método de uso do mesmo para a fabricação de peptídeos que são ensinados nesta invenção. O sistema compreende pelo menos dois reatores, cada reator incluindo uma quantidade de resina SPPS. Os reatores são posicionados em série. Um agente de desproteção é adicionado ao primeiro reator e depois transferido para os segundo e terceiro reatores, em série, operando assim para desproteger o grupo N protegido. O solvente de lavagem é adicionado ao primeiro reator e depois transferido para o segundo e esta operação é repetida várias vezes. Da mesma forma, uma solução de éster ativado por aminoácido é adicionada, em série, ao primeiro, segundo e terceiro reatores, operando assim para acoplar o aminoácido ao grupo N desprotegido. O solvente de lavagem é adicionado ao primeiro reator e então transferido para o segundo e esta operação é repetida várias vezes antes do próximo ciclo. O uso dos reatores em série reduz o solvente total necessário. A LCMS online também é utilizada para monitorar o progresso e a identidade das reações que acontecem dentro das partículas de resina em fase sólida.
