Abstract

VACINAS UNIVERSAIS CONTRA IMUNÓGENOS DE ORGANISMOS PATOGÊNICOS QUE CONFEREM PROTEÇÃO ESPECÍFICA PARA ORGANISMO E ENTRE GRUPOS. A presente invenção refere-se, em parte, a uma plataforma com base em vetor de estimulação e reforço para desenvolver vacinas contra patógenos que são adaptadas para provocar uma ampla resposta de células T que têm como alvo epítopos virais conservados. As vacinas universais são preparadas contra um imunógeno de um organismo patogênico infeccioso selecionado a partir de um vírus, uma bactéria, um fungo ou um protozoário que compreende pelo menos um polinucleotídeo de ácido ribonucleico (RNA) que compreende um quadro de leitura aberta que codifica pelo menos um antígeno polipeptídico ou um fragmento imunogênico do mesmo, em que o antígeno polipeptídico, ou fragmento imunogênico do mesmo, compreende uma proteína interna conservada que é enriquecida em antígenos de reconhecimento de células T CD8(mais elevado). A eficácia da plataforma de estimulação e reforço é testada em um modelo de camundongo humanizado que compreende um sistema imune humano totalmente funcional.
