Abstract
a presente invenção refere-se a um método para produzir uma proteína de fusão, em que o dito método compreende: a) colocar uma primeira proteína em contato com uma segunda proteína em condições que permitem a formação de uma ligação de isopeptídeo entre as ditas proteínas, em que a dita primeira proteína e a dita segunda proteína compreendem, cada uma, um ligante de peptídeo, em que os ditos ligantes de peptídeo são um par de ligantes de peptídeo que reage para formar uma ligação de isopeptídeo que liga a dita primeira proteína à dita segunda proteína para formar uma proteína ligada; e b) colocar a proteína ligada de (a) em contato com uma terceira proteína em condições que permitem a formação de uma ligação de isopeptídeo entre a dita terceira proteína e a dita proteína ligada, em que a dita terceira proteína compreende um ligante de peptídeo que reage com um ligante de peptídeo adicional na proteína ligada de (a) e em que os ditos ligantes de peptídeo são um par de ligantes de peptídeo que reage para formar uma ligação de isopeptídeo que liga a dita terceira pro-teína à dita proteína ligada para formar uma proteína de fusão, em que o dito par de ligantes de peptídeo usado em (a) é ortogonal ao par de ligantes de peptídeo usado em (b). os ligantes de peptídeo e o uso de pares ortogonais dos ditos ligantes na síntese de proteínas de fusão também são fornecidos. as proteínas recombinantes que compreendem os ditos ligantes, as moléculas de ácido nucleico que codificam as ditas proteínas e os ligantes, os vetores que compreendem as ditas moléculas de ácido nucleico e as células hospedeiras que compreendem os ditos vetores e as ditas moléculas de ácido nucleico também são contemplados.
