invention, peuvent être préparés en faisant réagir un acide 2-amino-nicoti- nique de la formule (II) avec un excès stoéchiométrique de formiamide. La 4-hydroxy-pyrido-(2,3-d)-pyrimidine obtenue peut être ensuite soumise à une nouvelle réaction pour obtenir des dérivés dans lesquels X représente d'au- tres groupes que le groupe hydroxy-. De préférence, cette réaction s'effec- tue à une température dépassant 120 en chauffant les ingrédients pendant 2 à 5 heures.

En variante, les composés peuvent être préparés en traitant une pyrido-(2,3-d)-pyrimidine 2-mercapto-4-substituée de la formule III par du nickel de Raney, afin de remplacer le groupe mercapto- par un atome d'hy- drogène.

Les composés de cette formule (III) font l'objet d'un brevet de même date de la Demanderesse intitulé "Perfectionnements aux composés de py- rimidine".
Le dérivé 4-hydroxy est transformé en dérivé mercapto par traitement par le pentasulfure de phosphore et en dérivé 4-chloro par traitement par le chlorure de phosphoryle. Des réactions avec l'ammoniaque, l'hydrazine et les amines primaires et secondaires fournissent des composés aminoet amino-substitués. Différentes réactions de ces substances et diverses manières d'arriver aux dérivés mentionnés sont décrites dans les exemples qui suivent, à titre d'illustration de l'invention.

On chauffe 50 g d'acide alpha-aminonicotinique et 100 g de for- miamide à 165 -170 (température intérieure) dans un bain d'huile pendant 2 1/2 heures. Le solide refroidi qui est obtenu est recristallisé de 700 cm3 d'eau afin d'obtenir 37,5 g de plaques incolores ayant un point de fusion de 255 -257 . Une seconde recristallisation de l'eau porte le point de fu-

sion à 258 .
Le spectre d'absorption des rayons ultra-violets du produit brut indique l'absence d'acide alpha-aminonicotinique.
EXEMPLE 2.-
4-chloropyido-(2,3-d)-pyrimidine (II) .
.On ajoute à 300 cm3 d'oxychlorure de phosphore 20,0 g de 4-hy- droxypyrido-(2,3-d)-pyrimidine et on chauffe la solution à reflux pendant 1 heure (Il reste une petite quantité de matière goudronneuse insoluble qui ne se dissout plus si l'on poursuit le chauffage à reflux). L'excès d'oxy- chlorure de phosphore est chassé par distillation et le résidu sirupeux ver- sé sur la glace. On isole la 4-chloropyrido-(2,3-d)-pyrimidine par extrac- tion de la solution aqueuse par le chloroforme. Le rendement en produit brut est 9,8 g et celui-ci fond à 135 avec décomposition.Une petite quan- tité du produit est recristallisée de l'heptane (Skellysolve C) ce qui por- te le point de fusion à 137 -138 avec décomposition.

On ajoute à 50 cm3 d'hydroxyde d'ammonium concentré 5,0 g de 4- chloropyrido-(2,3-d)-pyrimidine brute et on chauffe la solution pendant 45 minutes au bain de vapeur.La solution est décolorée à la Norite et le fil- trat est refroidi dans un bain de glace et de sel et saturé d'ammoniaque.
La solution refroidie est ensuite filtrée pour obtenir 3,0 g de 4-aminopy- rido- (2,3-d)-pyrimidine sous forme d'aiguilles incolores.
La recristallisation s'effectue d'une solution isopropanol/eau à 95% et fournit des aiguilles incolores fondant à 301 -302 .
EXEMPLE 4. -

On dissout 300 mg de 2-mercapto-4-aminopyrido-(2,3-d)-pyrimidi- ne (voir le Brevet précité) dans 800 cm3 d'éthanol et on ajoute 50 cm3 d'hy- droxyde d'ammonium concentré, puis 1 g de nickel de Raney et on chauffe la solution pendant 3 heures, on filtre et on évapore jusqu'à siccité au bain de vapeur. Le résidu est extrait par 50 cm3 d'eau et l'évaporation de la so- lution aqueuse fournit 60 mg de produit. Le spectre d'absorption de rayons ultra-violets est identique à celui de la 4-aminopyrido-(2,3-d)-pyrimidine préparée comme dans l'exemple 3.

On ajoute avec précaution 1,0 g de 4-chloropyrido-(2,3-d)-pyri- midine à 1 g d'aniline dans 10 cm3 d'eau. On chauffe la solution pendant 0,5 heure au bain de vapeur puis on la rend basique par de l'hydroxyde d'ammo- nium concentré, on refroidit la solution et on filtre. Le précipité brut est recristallisé de l'éthanol à 95%. Une seconde recristallisation du même sol- vant donne 0,51 g d'aiguilles vert clair fondant à 256 -257 .

On chauffe à 175 -180 pendant 1 1/2 heure 23 g de 4-hydroxy- pyrido-(2,3-d)-pyrimidine, 100 g de pentasulfure de phosphore et 500 oxy de tétraline. On élimine le solvant par filtration; le résidu est traité par l'eau chaude,extrait par un excès d'hydroxyde de sodium dilué (0,1 N) et filtré. Le composé mercapto- est précipité par acidification du filtrat par un léger excès d'acide acétique glacial. Le produit jaune brut est lavé par suspension dans 1 1. d'eau bouillante, puis purifié par extraction au Soxhlet en utilisant de l'éthanol absolu.

On ajoute avec précaution 2 g de 4-chloropyrido-(2,3-d)-pyrimi- dine (II), en refroidissant, à une solution de 15 cm3 d'hydrate d'hydrazine à 95% et 15 cm3 d'eau. La solution est chauffée pendant 30 minutes au bain de vapeur et laissée au repos jusqu'au lendemain pour cristalliser. Le pré- cipité brut est filtré, lavé par un peu d'eau glacée et recristallisé de l'éthanol absolu, ce qui donne des aiguilles rouge-orange ayant un point de fusion de 164-166 (non pesé).

On chauffe le produit brut de l'expérience décrite dans l'exemple 9 avec 50 cm3 d'hydroxyde d'ammonium concentré. Le dérivé amino- est récupéré par évaporation et recristallisé de l'éthanol sous la forme d'aiguilles incolores.

On dissout 3,0 g de 4-chloropyrido-(2,3-d)-pyrimidine dans 150 cm3 de dioxane anhydre. On ajoute 30 cm3 de diéthylamine et on laisse repo- ser la solution jusqu'au lendemain en glacière. Le chlorhydrate de diéthyla- mine est séparé par filtration et l'excès de dioxane et la diéthylamine sont évaporés au bain de vapeur. Le résidu gommeux est extrait plusieurs fois par du Skellysolve "B". La concentration des solutions combinées fournit par refroidissement 0,7 g de cristaux presque incolores ayant un point de fusion de 70 -72 . Une seconde recristallisation du même solvant porte le point de fusion à 72 -73 .

On met en suspension 6 g de 2-mercapto-4-hydroxy-6,7-diméthyl- pyrido-(2,3-d)-pyrimidine dans 1500 cm3 d'éthanol et on ajoute 150 cm3 d'une solution concentrée d'hydroxyde d'ammonium. Après chauffage pour obtenir une solution, on ajoute 20 g de nickel de'Raney et on concentre la solution dans le vide, après filtration, et par refroidissement on obtient 4,1 g de 4-hydroxy-6,7-diméthyl-pyrido-(2,3-d)-pyrimidine incolore. Après recristal- lisation d'un mélange éthanol/eau le composé fond à 360 avec décomposition.

The invention relates to a novel group of pyrido- (23-d) -pyrimidines and a process for preparing these compounds. These substances are useful as intermediates in the preparation of medicaments and as agents for inhibiting microorganisms such as lactic acid bacteria.

In this formula R1, R2 and R3 can be hydrogen, lower alkyl, aralkyl and monocyclic aryl groups, and, 1 is a chloro-, mercapto-, amino-, amino-substituted or arylamino- radical, and if at least one of the radicals R 1, R 2 and R 3 is an alkyl, aralkyl or aryl radical, X can also be a hydroxy group.
According to one of its characteristics, the present invention therefore comprises compounds of general formula (I).
These compounds, according to another characteristic of the present

invention, can be prepared by reacting a 2-amino-nicotinic acid of formula (II) with a stoichiometric excess of formiamide. The obtained 4-hydroxy-pyrido- (2,3-d) -pyrimidine can then be subjected to a further reaction to obtain derivatives in which X represents groups other than the hydroxy group. Preferably, this reaction is carried out at a temperature exceeding 120 by heating the ingredients for 2 to 5 hours.

The compounds of this formula (III) are the subject of a patent of the same date by the Applicant entitled "Improvements to pyrimidine compounds".
The 4-hydroxy derivative is converted into a mercapto derivative by treatment with phosphorus pentasulfide and into a 4-chloro derivative by treatment with phosphoryl chloride. Reactions with ammonia, hydrazine, and primary and secondary amines provide amino and amino-substituted compounds. Various reactions of these substances and various ways of arriving at the derivatives mentioned are described in the examples which follow, by way of illustration of the invention.

The cooled solid which is obtained is recrystallized from 700 cm3 of water in order to obtain 37.5 g of colorless plates having a melting point of 255 -257. The ultra-violet ray absorption spectrum of the crude product indicates the absence of alpha-aminonicotinic acid.
EXAMPLE 2.-
4-Chloropyido- (2,3-d) -pyrimidine (II).
20.0 g of 4-hydroxypyrido- (2,3-d) -pyrimidine are added to 300 cm3 of phosphorus oxychloride and the solution is heated at reflux for 1 hour (a small amount of tarry material remains. insoluble which no longer dissolves if heating is continued under reflux). The excess phosphorus oxychloride is removed by distillation and the syrupy residue is poured onto ice. 4-Chloropyrido- (2,3-d) -pyrimidine is isolated by extraction of the aqueous solution with chloroform. The yield of crude product is 9.8 g and it melts at 135 with decomposition. A small quantity of the product is recrystallized from heptane (Skellysolve C) which brings the melting point to 137 -138. with decomposition.

5.0 g of crude 4-chloropyrido- (2,3-d) -pyrimidine is added to 50 cm3 of concentrated ammonium hydroxide, and the solution is heated for 45 minutes on a steam bath. The solution is decolorized with water. Norite and the filtrate is cooled in an ice-salt bath and saturated with ammonia.
The cooled solution is then filtered to obtain 3.0 g of 4-aminopy-rido- (2,3-d) -pyrimidine in the form of colorless needles.
Recrystallization is carried out from a 95% isopropanol / water solution and gives colorless needles, melting at 301 -302.

The crude precipitate is recrystallized from 95% ethanol. A second recrystallization of the same solvent gives 0.51 g of light green needles melting at 256 -257.

The solvent is removed by filtration; the residue is treated with hot water, extracted with an excess of dilute sodium hydroxide (0.1 N) and filtered. The mercapto compound is precipitated by acidification of the filtrate with a slight excess of glacial acetic acid. The crude yellow product is washed by suspension in 1 l of boiling water, then purified by extraction with a Soxhlet using absolute ethanol.

The crude precipitate is filtered, washed with a little ice-water and recrystallized from absolute ethanol, giving red-orange needles with a melting point of 164-166 (not weighed).

3.0 g of 4-chloropyrido- (2,3-d) -pyrimidine are dissolved in 150 cm3 of anhydrous dioxane. 30 cm3 of diethylamine are added and the solution is left to stand overnight in a cooler. The diethylamine hydrochloride is separated by filtration and the excess dioxane and diethylamine are evaporated on a steam bath. The gummy residue is extracted several times with Skellysolve "B". Concentration of the combined solutions provides on cooling 0.7 g of nearly colorless crystals having a melting point of 70 -72. A second recrystallization of the same solvent brings the melting point to 72 -73.
