Procédé de préparation de solutions aqueuses concentrées de théophylline.
---------------

Process for the preparation of concentrated aqueous solutions of theophylline.
---------------
It is known to increase the solubility of theophylline by means of primary or secondary aliphatic diamines, diethanolamine, salts of organic amines or triethanolamine. Some of these substances are of no therapeutic value. , the others have considerable drawbacks. The theophylline content of solutions produced therapeutically by the method mentioned is 18.7 and 20%.
It has now been discovered that highly concentrated aqueous solutions of theophylline, with a theophylline content of up to 40%, can be obtained by causing theophylline to act on amines of the R1 # R2 # N-alcohol type ( R1 and R2 = H or an alkyl) and dissolving the product obtained in the desired quantity of water. Molecular amounts are advantageously chosen for this purpose, but good solubility is also obtained by the use of other proportions. As particularly favorably active representatives of this group of amines, we can mention) -diethylamino-

EMI2.1
$ thanoiadiethyl- -oy-ethyl3-amine (melting point 35; 75), additionally isopropylaminoethanol or monoethylaminoethanol.
By this process, it is possible to dissolve very large quantities of theophylline in the smallest quantities of water, that is to say to prepare them in the smallest volume for infection applications.
In combination with the good therapeutic action inherent in the mentioned amines, this results in a very effective remedy with a new action.
Examples:
EMI2.2
1.) We introduce in 117 gr. of Ji -diethylam1noethanol 180 gr. powdered theophylline and mixed thoroughly.
The product obtained is brought to a final volume of 450 cc by addition of water and possible slight heating. In this way a concentrated clear solution is obtained having a theophylline content of 40%.
2.) 36 gr. of powdered theophylline are introduced into
EMI2.3
21 gr. of isopropylaminoethanol and mixed thoroughly. The mixture is dissolved in water by heating and made up to 100 oc. The theophylline content of this solution is 36%.
3.) 90 gr. of powdered theophylline and 89 gr. of ethylaminoethanol are dissolved after thorough mixing, with slight heating, in approximately 150 cc of water and then supplemented with water up to 225 cc. This solution, also clear after cooling, has a theophylline content of 40%.
ABSTRACT -----------
Process for the preparation of concentrated aqueous solutions of theophylline, characterized in that theophylline is made to act on amines of the R1 # N-alcohol type and in that the product obtained is dissolved in R2 of water, R1 and R2 represent hydrogen or alkyl.
