<Desc/Clms Page number 2>
<Desc/Clms Page number 3>
<Desc/Clms Page number 4>
<Desc/Clms Page number 5>

<Desc/Clms Page number 7>

<Desc/Clms Page number 9>
<Desc/Clms Page number 10>

EMI1.2
2, 3-, 2, 4-, 2, 5-, 2, 6-, 3, 4- or 3, 5-dihydroxyphenyl: or 2, 3, 4-, 2, 3, 5-, 2, 4, 5-, 2, 4, 6-, 3, 4, 5- or 2, 3, 6 trihydroxyphenyl.
The compounds of formula I can be in the form of trans or cis isomers or mixtures of cis / trans isomers: In general, the trans compounds of formula I are preferred, especially those in which R is the o- or m-hydroxyphenyl
<Desc / Clms Page number 2>
The invention further relates to a process for the preparation of compounds of formula I, pharmaceutical preparations based on compounds of formula I, the compounds of formula I for the treatment and prophylaxis of neoplasias and dermatoses, as well as the use of Use of compounds of formula I for the manufacture of pharmaceutical preparations for the treatment and prophylaxis of such diseases.
The compounds of formula I can be prepared according to the invention by eliminating the protective group from a compound of general formula:
EMI2.1
EMI2.2
11 1 in which R is a residue R, in which the hydroxy group (s) is or are protected.
As protecting groups all conventional protecting groups are taken into account.
Examples of such protecting groups are ethers, in particular 2-tetrahydropyranyl ether and silyl ether, such as trimethylsilyl ether; furthermore, alkyl ethers such as methyl ether and esters, for example the lower saturated carboxylic acid ester such as acetates and carbonates. The removal of the protective groups can be carried out in a manner already known by treatment with acids, bases or reducing agents.
<Desc / Clms Page number 3>
Ether protecting groups such as tetrahydropyranyl or trimethylsilyl are removed by treatment with acids, such as p-toluenesulfonic acid or Lewis acids such as BF- or BBr. Ester protecting groups such as acetate or carbonate are removed by treatment with bases, for example from alcoholic or hydroalcoholic alkali hydroxide solution.
The compounds of general formula II can be obtained by condensing a compound of general formula:
EMI3.1
with a compound of general formula:
EMI3.2
B - R in which either A is one of the residues -CH (CH3) P + (Q) 3Y- or -CH (CH3) P (O) (QAlk) 2 and B is formyl, or A is acetyl and B one of the remains
EMI3.3
-CHP (Q) Y'ou-CHP (OAlk): and Q is an aryl, in particular phenyl
<Desc / Clms Page number 4>
Y- is the anion of an organic or mineral acid, for example Br and Alk is a lower alkyl, for example methyl and R11 has the same meaning as above.
The condensation of the compounds of formulas III and IV can be carried out according to the methods known from Wittig or from Horner.
In the Wittig reaction, that is to say, using a compound of formula III with A = -CH (CH3) P + (Q) 3Y- or of formula IV with B = -CH2P (Q) 3Y, the components are condensed in the presence of an acidity-fixing medium, for example in the presence of a strong base, such as for example butyl lithium, sodium hydride or the sodium salt of dimethyl sulfoxide, but especially in the presence of an ethylene oxide substituted by a lower alkyl such as butylene oxide-1, 2 where appropriate in a solvent, for example in an ether such as diethyl ether or tetrahydrofuran or in an aromatic hydrocarbon such as benzene, in a temperature range between room temperature and the boiling point of the reaction mixture.
Among the acid anions Y - the chloride ion or the bromide ion or the hydrosulfate ion is preferred, among the acid anions the tosyloxy ion is preferred. The aryl radical Q is preferably a phenyl radical or a substituted phenyl radical such as p-tolyl.
In Horner's reaction, that is, using a compound of formula III with
<Desc / Clms Page number 5>
A = -CH (CH3) -P (O) (OAlk) 2 or of formula IV with B = -CH2-P (O) (oAlk) 2 the components are condensed using a base and preferably in presence of an inert organic solvent, for example using sodium hydride in benzene, toluene, dimethylformamide, tetrahydrofuran, dioxane or 1,2-dimethoxyethane or also using a sodium alcoholate in a alkanol, for example sodium methylate in methanol, in a temperature range between 0 and the boiling point of the reaction mixture.
The alkoxy radicals are above all lower alkoxy radicals with 1-6 carbon atoms, like the
EMI5.1
methoxy or ethoxy.
The compounds of formula I can be in trans or cis form.
During preparation, they are mostly in the trans form. The cis contents, if any, can be separated in the manner already known, if desired.
The starting compounds of formulas III and IV, insofar as their preparation is not known or described below, can be prepared by analogy to known methods or described below.
The compounds of formula I are therapeutically effective. They have in particular an anti-seborrheic, anti-keratinizing, anti-neoplastic and anti-allergic anti-inflammatory efficacy, which can be demonstrated using the test procedures described below.
<Desc / Clms Page number 6>
A. The chemically induced prophylaxis action of breast tumors can be determined by the following procedure. Female Sprague-Dawley rats are maintained under controlled conditions of light and temperature, with free access to drinking water and

to food. At the age of 50 days, each spleen is administered, using a stomach tube, 15 mg of dissolved dimethylbenz (a) anthracene. in peanut oil.
Treatment with the test compounds begins 1 day after administration of the carcinogen. The weights of the test animals are recorded and the tumors are palpated weekly and measured with calipers. The volumes are calculated according to the formula
EMI6.2
2 d in which D is the largest diameter and 2 d the smallest diameter, of the ellipsoid of the tumor. The trial is stopped after 11 weeks and evaluates. In this test, in addition to the 30 control animals, which receive exclusively normal food, the following two groups of test animals are used: 1 - 33 rats, which are administered daily, mixed with food, 30 mg / kg of the test compound.
2 - 36 rats, administered daily with food mixtures, 90 mg / kg of the test compound.
B. The action on the tumors can also be determined on transplantable chondrosarcomas of the spleen, according to the following method. The solid tumor of a sacrificed animal is finely ground and suspended in a phosphate / sodium chloride buffer solution.
0.5 ml of the 30% tumor paste is implanted subcutaneously in albino rats.
<Desc / Clms Page number 7>
The transplanted rats are divided into test groups of each 8 animals. The test compounds are suspended in peanut oil and administered for 24 days five times a week using an oral pharynx probe. The tumors are excised on the 24th day and weighed. The results are expressed by the C / T quotient, which is calculated as follows:
EMI7.1
Average weight of the CST - + controls Average weight of the treated C. The anti-metaplastic action can also be determined according to the following procedure.
Female Holtzmann rats weighing close to 100 g, after an acclimatization period of 8 days, are ovariectomized under narcosis by thiogenal and after 14 days are introduced into the test. Two animals are placed in a cage with free access to food, which contains approximately 2,000 IE of vitamin A measured by analysis.
Before the oral administration of the test compound, the animals are treated subcutaneously daily for 6 consecutive days with 1 μg of estradiol benzoate and 250 μl of testosterone propionate dissolved in 0.1 ml of sesame. Parenteral administration of the hormone leads to the formation of a pure scaling stage in the area of the vagina, that is to say a scaly metaplasia. Two days after the oral administration of the substance testaeon again examines the result of the reaction on the vaginal epithelium. To calculate the effective average dose, the surface method according to Behrens and Karber is used.
<Desc / Clms Page number 8>
The compounds of formula I can be used in topical and systemic therapy of benign and malignant neoplasias, premalignant lesions, as well as in systemic and topical prophylaxis of the above-mentioned condition.
They are also suitable for topical and systemic therapy of acne, psoriasis and other dermatoses
EMI8.1
deep with increased keratinization in modified pathology, as well as inflammatory and allergic dermatological conditions. The formula products can also be used to fight mucosal diseases with inflamed or degenerative or metaplastic alterations. The compounds of formula I are characterized in particular by reduced toxicity or better compatibility in comparison with known retinoids.
The products can be administered in the form of galenical enteral, parenteral or topical applications. For enteral application, for example, products in the form of pills, capsules, dragees, syrups, suspensions, solutions and suppositories.
<Desc / Clms Page number 9>
For parenteral application, products in the form of a solution for infusion or injection are suitable.
The dosages in which the preparations are administered may vary according to the mode of use and the route of administration as well as according to the needs of the patients.
In general, adult doses of about 0, 1-50 mg / kg, preferably 1-15 mg / kg, are taken into account.
The preparations can be administered in one or more dosages. A preferred dosage form is the capsule with a content of about 5-200 mg of active ingredient.
The preparations may contain inert or pharmacodynamically active additives. Tablets or granules can contain a series of binders, fillers, vehicles or diluents. Liquid preparations may for example be in the form of a sterile water-miscible solution. The capsules may contain, in addition to the active principle, a filler or a thickener. In addition there may be aromatic additives, as well as the products usually used as preservatives, stabilization, humidification and emulsion, as well as salts for modifying the osmotic pressure, the buffering capacity, and other additives.
The vehicles and diluents mentioned above can be organic or mineral products, for example water, gelatin, lactose,

starch, magnesium stearate, talc, gum arabic, polyalkylene glycols or the like.
The condition is that all the additives used in the manufacture of the preparations are not toxic.
For topical application, the active ingredients are appropriately used in the form of balms, tinctures, creams, solutions, lotions, sprays, suspensions and the like. We prefer balms and creams as well as solutions. These preparations intended for topical applications can be produced by adding to the products of the process as active component of the solid or liquid vehicles customary in such non-toxic, inert preparations suitable for topical treatment.
For topical use, solutions of about 0.1-5%, preferably about 0.3, are suitable.

2%, as well as balms and creams at about 0.1-5%, preferably about 0.3-2%.
To the preparations, an antioxidant can also be added, for example tocopherol, N-methyl-ytocopheramine, as well as butylated hydroxyanisol or butylated hydroxytoluene.
The following examples more fully describe the invention. The temperatures are indicated in degrees Celsius.
