Abstract
The present invention provides A Chinese Herbal Medicine Health-care
Oral Liquid Specially Fermented by Compound Probiotics for Mink and Its
Preparation Method, which is prepared from Chinese herbal medicine extract
and compound probiotics fermentation liquor with a volume ratio of 1:1'-2;
The compound formulation of Chinese herbal medicine and compound
probiotics can achieve the treatment and health care concurrently, i.e. improving
the immunity of animal (mink) while treating the viral diseases; it can not only
treat and prevent diseases, but also promote the symbiotic fermentation of
Lactobacillus and Bacillus in the health liquid, and produce a large number of
intermediate products,such as glucuronic acid, gluconic acid, acetic acid, lactic
acid, amino acid etc.; it can replace antibiotics and chemical antiviral drugs in a
certain range to achieve real ecological breeding.

Method
The present invention relates to A Chinese Herbal Medicine Health
care Oral Liquid Specially Fermented by Compound Probiotics for Mink
and Its Preparation Method.
During the breeding process, farms (farmers) use antibiotics in the
feed for a long time, and ignore the control of the breeding environment,
which result in serious pollution; however probiotics alone can not make
the breeding animals completely resist the infection of bacteria and
viruses. Huge experimental studies have shown that probiotics can
dynamically regulate the gastrointestinal flora ecosystem of animals;
which is with the ability to promote the growth and development of
animals, improve the feed conversion rate, improve the quality of
livestock products, enhance the immunity of animals, improve the
resistance to bacterias and viruses. Meanwhile, huge studies on Chinese

ferment Chinese herbal medicine, to destroy its cell wall and greatly
improve the utilization rate of the active ingredients in Chinese herbal
medicine. During the fermentation process, a variety of growth
promoting factors and new materials also are produced, which can
improve the efficacy of Chinese herbal medicines, thereby further
enhance the immunity of animals, improve product quality, so that
replace antibiotics for disease prevention and treatment, reduce veterinary
drug residues, and improve the environment and ecological benefits, to
truly realize ecological breading, which is a common development trends
all over the world.
Given the deficiencies of the above prior art, the present invention
provides A Chinese Herbal Medicine Health-care Oral Liquid Specially
Fermented by Compound Probiotics for Mink and Its Preparation
Method.
In order to solve the above technical problem, the solution of the
present invention includes:
A Chinese Herbal Medicine Health-care Oral Liquid Specially
Fermented by Compound Probiotics for Mink and Its Preparation
Method, which is prepared from Chinese herbal medicine extract and
compound probiotics fermentation liquor with a volume ratio of 1:1~-2.
The compound probiotic fermented Chinese herbal medicine health
care oral liquid special for mink, wherein the weight ratio of
Lactobacillus and Bacillus of which is 1:1.
The compound probiotic fermented Chinese herbal medicine health
care oral liquid special for mink, wherein the compound probiotics
fermentation liquor includes 10-20 parts of Lactobacillus and 10-20
parts of Bacillus.
The compound probiotic fermented Chinese herbal medicine health
care oral liquid special for mink, wherein the Chinese herbal medicine
extract is prepared from Astragalus membranaceus, Forsythia suspensa,
Radix Isatidis, Flos Lonicerae and Chrysanthemum;
and the parts by mass are as follows: 20-30 parts of Astragalus,
-30 parts of Forsythia, 20-30 parts of Radix isatidis, 20-30 parts of
Flos Lonicerae and 20-30 parts of Chrysanthemum.
A preparation method of the compound probiotic fermented Chinese
herbal medicine health care oral liquid special for mink, wherein the following preparation steps are included: the preparation of compound probiotics fermentation liquor and the Chinese herbal medicine extract respective, and then mixing the prepared compound probiotics fermentation liquor with the Chinese herbal medicine extract in proportion.
The preparation method, wherein the above steps for the preparation
of the compound probiotics fermentation liquor are included:
A. The slant culture preservation is activated, Lactobacillus and
Bacillus are incubated in LB medium respectively, then autoclaved for
-40 min at pH 7-7.5 and temperature of 100-130°C, and cultured
at 30-38C for 12-20 h;
B. Perform the primary liquid expansion culture for Lactobacillus
and Bacillus respectively, with an inoculum amount of 10-20%;
C. Perform the secondary liquid expansion culture for the
Lactobacillus and Bacillus after the primary liquid expansion culture
respectively, with aninoculum amount of 10~20%;
D. Mix the fermentation liquor of Lactobacillus and Bacillus with a
weight ratio of 1:1 to obtain a compound probiotics fermentation liquor.
The preparation method, wherein the above steps for the preparation
of the Chinese herbal medicine extract are included:
E. Crush the corresponding Chinese herbal medicine to 6080
mesh, then mix well to obtain the described Chinese herbal medicine
powder after weighing the corresponding one respectively according to
the mass fraction;
F. Place the above Chinese herbal medicine powder and 300-600
parts of water into the extraction tank, soak for 5-15 h; then heat the
mixture to 40-100°C to extract for 10-12 h, filter and collect the
extract; then add 300-600 parts of water to the extraction tank again for
the extration of the medicine residue at 40-100°C for 10-15 h;
J. The described Chinese herbal medicine extract is obtained by
combining the two extracts and concentrating in vacuum to 10-20% of
the original volume at 3070°C.
The preparation method, wherein the method specifically includes:
add the compound probiotics fermentation liquor to the obtained Chinese
herbal medicine extract to obtain the mixing solution; incubate at
-35°C for 12-36 h; start the stir for 5 min per 12 h at a rotation
speed of 120 r/min; the fermentation process will continuous till the pH
of the mixture drops to 3.0-3.2; the compound probiotic fermented
Chinese herbal medicine health care oral liquid special for mink is
obtained by filtration with 100 mesh filter cloth (4 layers) after stirring
for 30 min.
The present invention provides A Chinese Herbal Medicine Health
care Oral Liquid Specially Fermented by Compound Probiotics for Mink
and Its Preparation Method. The compound formulation of Chinese
herbal medicine and compound probiotics can achieve the treatment and
health care concurrently, i.e. improving the immunity of animal (mink)
while treating the viral diseases; it can replace antibiotics and chemical
antiviral drugs in a certain range to achieve real ecological breeding.
Combining with experiments, the safety and disease prevention and
treatment effects of the compound probiotic fermented Chinese herbal
medicine health liquid of the present invention are illustrated as below.
The acute toxicity tests (rat) of the compound probiotic fermented
Chinese herbal medicine health liquid of the present invention were
conducted in a university in Qingdao, wherein the test group was
assigned 50 rats, which were fed with a dose of 0.6 mL/10 g body weight
for 21 days; and the blank control group was assigned 50 rats, which were
fed as usual.
The results showed that all rats in the test group were alive, and can
move freely, with smooth hair, normal diet, no secretions in the
respiratory tract, eyes and mouth, no symptoms of poisoning, and with
weight gain. Comparing with the blank control group, the weight of the
rats in the test group increased significantly (P < 0.001). At the end of the
21 days observation period, the rats were all put to death by dislocation
for autopsy, and the macroscopic observation result of heart, liver, spleen,
lung, kidney, brain, intestine, small intestine and other organs showed no
obvious abnormalities. In addition, the results of thymus coefficient and
spleen coefficient showed that comparing with the blank control group,
the spleen coefficient of the test group was significantly increased (P <
0.01), suggesting that the compound probiotic fermented Chinese herbal
medicine health liquid of the present invention has the immune
enhancement effect, belongs to the actual non-toxic material with high
safety.
After fermentation of various probiotics, the color, taste and odor of
the Chinese herbal medicine extract of the present invention is improved
obviously. The original bitter components have decreased obviously, but
the sweet taste is obvious, and the taste is mellow and refreshing. The
antioxidant effect of health liquid is significantly enhanced measured by
chemiluminescence method.
The compound probiotic fermented Chinese herbal medicine health
liquid of the present invention contains a large number of Chinese herbal
medicine substances, such as free amino acids, chlorophyll, carotene,
enzymes, betaine, vitamin A, vitamin B, vitamin C, vitamin E, vitamin P,
inorganic salts, and trace elements, etc. (hundreds of extracts in total); it can not only treat and prevent diseases, but also promote the symbiotic fermentation of Lactobacillus and Bacillus in the health liquid, and produce a large number of intermediate products,such as glucuronic acid, gluconic acid, acetic acid, lactic acid, amino acid, etc.
Figure 1 shows the influence of different concentrations of the
compound probiotic fermented Chinese herbal medicine health liquid on
mink body weight;

groups;
The present invention provides A Chinese Herbal Medicine Health
care Oral Liquid Specially Fermented by Compound Probiotics for Mink
and Its Preparation Method. In order to clarify the purposes, technical
proposals and effects of The present invention more clearly and
definitely, the detailed description is as below. It should be note that the
specific embodiments described here are only for interpretation, but not to
limit the present invention.
Embodiment 1
The raw materials for the preparation of the compound probiotic
fermented Chinese herbal medicine health liquid are (parts by mass): 20
parts of Astragalus, 20 parts of Forsythia, 20 parts of Radix isatidis, 20
parts of Flos Lonicerae, 20 parts of Chrysanthemum, 10 parts of
Lactobacillus and 10 parts of Bacillus.
The preparation process of the compound probiotics fermentation
liquor is as follows:
(1) The slant culture preservation is activated,and the Lactobacillus
and Bacillus are incubated (shake flask) in LB medium respectively, then
autoclaved for 20-40 min at pH 7-7.5 and temperature of 100-130°C,
and cultured at 30-38C for 12-15 h;
(2) Perform the primary liquid expansion culture (50 L tank) for
Lactobacillus and Bacillus respectively, with an inoculum amount of
~20%;
(3) Perform the the secondary liquid expansion culture (500 L tank)
for the Lactobacillus and Bacillus after the primary liquid expansion
culture, with aninoculum amount of 10~20%;
(4) Mix the fermentation liquor of Lactobacillus and Bacillus
according to the following ratio: 10 parts of Lactobacillus and 10 parts of
Bacillus.
The preparation process of the Chinese herbal medicine extract is as
follows:
(1) Crush the Chinese herbal medicine each to 60 mesh, weigh
according to the formula, and mix (3D) to obtain the described Chinese
herbal medicine powder;
(2) Place the Chinese herbal medicine powder and 300 parts of water
into the extraction tank, and soak for 5 h;
(3) Heat the mixture to 40-100°C, and extract for 10-12 h;
(4) Filter and collect the extract;
(5) Add 300 parts of water to the medicine residue again, and heat
the mixture to 40-100C to extract for 10 h;
(6) Combine the two extracts and concentrate in vacuum to a 10~
% of the original volume at a temperature of 30-70°C to obtain a
liquid coolant.
(7) Add 10-20 parts of Lactobacillus and 10-20 parts of Bacillus
into the liquid coolant obtained in step (6), culture for 12-36 h at a
temperature of 25-35°C, start the stir for 5 min per 12 h at a rotation speed of 120 r/min; the fermentation process will continuous till the pH of the mixture drops to 3.0-3.2; discharge after a further stirring for 30 min, and the liquid discharged is filtered by 100 mesh filter cloth (4 layers) to obtain the fermentation filtrate, then to obtain the compound probiotic fermented Chinese herbal medicine health liquid after sterile filling and package.
Embodiment 2
The raw materials for the preparation of the compound probiotic
fermented Chinese herbal medicine health liquid are (parts by mass): 25
parts of Astragalus, 25 parts of Forsythia, 25 parts of Radix isatidis, 25
parts of Flos Lonicerae, 25 parts of Chrysanthemum, 15 parts of
Lactobacillus and 15 parts of Bacillus.
The preparation process of the compound probiotics fermentation
liquor is as follows:
(1) The slant culture preservation is activated,and the Lactobacillus
and Bacillus are incubated (shake flask) in LB medium respectively, then
autoclaved for 20-40 min at pH 7-7.5 and temperature of 100-130°C,
and cultured at 30~-38C for 15 h;
(2) Perform the primary liquid expansion culture (50 L tank) for
Lactobacillus and Bacillus respectively, with an inoculum amount of
~20%;
(3) Perform the the secondary liquid expansion culture (500 L tank)
for the Lactobacillus and Bacillus after the primary liquid expansion
culture, with aninoculum amount of 10~20%;
(4) Mix the fermentation liquor of Lactobacillus and Bacillus
according to the following ratio: 15 parts of Lactobacillus and 15 parts of
Bacillus.
The preparation process of the Chinese herbal medicine extract is as
follows:
(1) Crush the Chinese herbal medicine each to 60 mesh, weigh
according to the formula, and mix (3D) to obtain the described Chinese
herbal medicine powder;
(2) Place the Chinese herbal medicine powder and 300 parts of water
into the extraction tank, and soak for 10 h;
(3) Heat the mixture to 40~-100°C, and extract for 10~- 12 h;
(4) Filter and collect the extract;
(5) Add 300 parts of water to the medicine residue again, heat the
mixture to 40-100°C to extract for 12 h;
(6) Combine the two extracts and concentrate in vacuum to a 10~
% of the original volume at a temperature of 30-70°C to obtain a
liquid coolant.
(7) Add 15 parts of Lactobacillus and 15 parts of Bacillus into the
liquid coolant obtained in step (6), culture for 12-36 h at a temperature
of 25-35 0 C, start the stir for 5 min per 12 h at a rotation speed of 120
r/min; the fermentation process will continuous till the pH of the mixture
drops to 3.0-3.2; discharge after a further stirring for 30 min, and the
liquid discharged is filtered by 100 mesh filter cloth (4 layers) to obtain
the fermentation filtrate, then to obtain the compound probiotic fermented
Chinese herbal medicine health liquid after sterile filling and package.
Embodiment 3
The raw materials for the preparation of the compound probiotic
fermented Chinese herbal medicine health liquid are (parts by mass): 30
parts of Astragalus, 30 parts of Forsythia, 30 parts of Radix isatidis, 30
parts of Flos Lonicerae, 30 parts of Chrysanthemum, 20 parts of
Lactobacillus and 20 parts of Bacillus.
The preparation process of the compound probiotics fermentation
liquor is as follows:

into the liquid coolant obtained in step (6), culture for 12-36 h at a
temperature of 25-35C, start the stir for 5 min per 12 h at a rotation
speed of 120 r/min; the fermentation process will continuous till the pH
of the mixture drops to 3.0-3.2; discharge after a further stirring for 30
min, and the liquid discharged is filtered by 100 mesh filter cloth (4
layers) to obtain the fermentation filtrate, then to obtain the compound
probiotic fermented Chinese herbal medicine health liquid after sterile
filling and package.
In order to further describe the influence of compound probiotic
fermented Chinese herbal medicine health liquid for mink on the body
weight, immunity and intestinal health of the mink, the following
grouping controlled studies were performed on minks.
The test minks (Dalian black) were provided by a fur animal
breeding base of an agricultural technology Co., Ltd. in Qingdao. Sixty
cub minks of 150 days old are selected and randomized into 6 groups
according to gender and weight. There are quintuplication (n = 5) in each
group and each duplication sample contained two minks (male and
female are in half). The test has lasted for 35 days from April 20, 2016 to
May 24, 2016. The cub minks in each group are fed in separate cages, and
they could eat and drink freely.
Activity strains of Lactobacillus and Bacillus are isolated and
preserved by the Animal Microbiology and Immunology Laboratory of a
certain university in Qingdao.
Colony count (plate count method)
In the super clean bench, transfer 25 mL of bacterial liquid via a
autoclaved graduated cylinder, into ajar containing 250 mL of normal
saline, then shake well to obtain a 1:10 diluent. Transfer 1.0 mL of the
above diluent via a autoclaved 1.0 mL pipette, into ajar containing 9 mL
of normal saline, it is a 10-fold dilution, and repeat the above operation
until the dilution ratio of bacterial liquid is 109. Transfer 1.0 mL of
bacterial liquid via pipette, into the autoclaved and dried plate,
meanwhile, pour into the nutrient agar cooled to about 46°C into the
plate, and move the plate to make the solution mix well. Place the plate in an incubator for 24 h and count in triplication. The counting formula is: colony forming unit (cfu) per milliliter = Mean colony number of duplicating samples (n = 3) at the same dilution * dilution.
Inoculation
The test animals were randomized into six groups ( group A, B, C,
D, E and F), with quintuplication (n = 5) in each group. The minks were
Inoculated. For the first time, each mink was Inoculated with an equal
volume of mixed antigen and 0.1 mL of Freund's complete adjuvant. One
week later, each mink was Inoculated with an equal volume of mixed
antigen and 0.1 mL of Freund's incomplete adjuvant. After one week
interval, each mink was repeatedly injected with an equal volume of
mixed antigen and 0.1 mL of Freund's incomplete adjuvant.

The influence of different test groups on mink body weight
Results analysis: as shown in Figure 1, the line chart shows that the
initial body weight of minks is close. After one week of gavage treatment,
the weight difference of minks in different test groups is still small, but
the weight difference has been obvious since two weeks later. The body
weight of minks in Lactobacillus group, bacillus group, Chinese herbal
medicine group and compound probiotic fermented Chinese herbal
medicine health liquid group are higher than that of normal saline group,
and the body weight of minks in the compound probiotic fermented
Chinese herbal medicine health liquid group is higher.

Data analysis: Use spss software to conduct statistic analysis on the
internal organs of mink by multiple comparison method. According to
Table 2, the visceral index of the mink in the compound probiotic
fermented Chinese herbal medicine health liquid group is significantly
higher than that of the control group (P < 0.01), and also significantly
higher than other test groups (P < 0.05); the weight of visceral organs in
Bacillus group, Lactobacillus group and Chinese herbal medicine group
are also significantly higher than that of the control group (P < 0.05); and
the compound probiotic fermented Chinese herbal medicine health liquid
group is significantly higher than the control group, probiotics group and
Chinese herbal medicine group (P < 0.05).

Calculation from Table 3 shown that the serum titer of each test
group is increased by 29%, 24%,11%,17%,14%; 30%, 26%,15%, 8%,
12% respectively compared with the control group. It can be concluded that the mink serum bursa antibody content of the compound probiotic fermented Chinese herbal medicine health liquid group is significantly higher than that of other test groups.

According to Table 4 and Figure 2, it can be concluded that the OD
values measured for the first time are close in each group, with an
exception of group E, that is, the antibody contents in the minks body are
not significantly different after the first inoculation; the OD values
measured after the second inoculation are still close, that is, the antibody
contents in minks are not significantly different; After the third
inoculation, the OD values of the compound probiotic fermented Chinese
herbal medicine health liquid group are significantly higher than that of
other test groups, indicating that the antibody content in the mink body of
the compound probiotic fermented Chinese herbal medicine health liquid
group increased more than other test groups.
The influence of different test groups on the mucosal thickness and
villus length of minkjejunum
According to the light microscope observation, the structure of the
mink jejunum mucosa in Bacillus group, Chinese herbal medicine group,
compound probiotic fermented Chinese herbal medicine health liquid
group and control group are relatively complete, with distinct layers, and
intact villi. There are obvious branches and the striated border structure
on the surface of intestinal mucous epithelial cells is uniform. Compared
with the control group, the jejunum mucosal villus of minks in the
compound probiotic fermented Chinese herbal medicine health liquid
group are more closely arranged and the villus are longer.
The results showed that the growth of minks could be promoted by
gavage of Bacillus, Lactobacillus, Chinese herbal medicine, or compound
probiotic fermented Chinese herbal medicine health liquid, but after two
weeks, the weight of mink in the compound probiotic fermented
Chinese herbal medicine health liquid group is much larger than those in
other test groups.
The influence of different test groups on the visceral index of mink.
According to the test result, it can be concluded that, in terms of
promoting the growth of mink internal organs, the compound probiotic
fermented Chinese herbal medicine health liquid group is with
significantly different from the control group (P < 0.01). Comparing with
the control group, the influence of Bacillus group, Lactobacillus group and Chinese herbal medicine group on organ index are with significantly different (P < 0.05). It shows that the compound probiotic fermented
Chinese herbal medicine health liquid group has a more significant
influence on the visceral index of the mink.
The influence of different test groups on the serum titer of bursa
antibody.
The results showed that the antibody titers of minks in different test
groups are significantly different from those in the control group.
However, in the first and second tests, the serum titers of minks in the 11
group increased by 29% and 30% respectively compared with the control
group, and the value changed greatly compared with other test groups.
Therefore, the present invention can obviously promote the growth
and weight gain of mink, the growth of internal organs, the increase of
antibody titer of chicken bursa, the length of intestinal microvilli and the
thickness of intestinal wall, which greatly improves the performance of
mink.
Of course, the above description is only a better embodiment of the
present invention, and it is not limited to listing the above-mentioned
embodiments. It should be noted that all equivalent substitutions and
obvious deformation forms made by any person skilled in the art under the instruction of this specification fall within the substantive scope of the specification and should be protected by the invention.
Claims (12)
1. A compound probiotic fermented Chinese herbal medicine health
care oral liquid special for mink, which is prepared from a Chinese herbal
medicine extract and a compound probiotic fermentation liquor in a
volume ratio of 1:1~-2.
2. The compound probiotic fermented Chinese herbal medicine
health care oral liquid special for mink according to claim 1, wherein the
compound probiotic fermentation liquor comprises Lactobacillus and
Bacillus, and the weight ratio of both is 1:1.
3. The compound probiotic fermented Chinese herbal medicine
health care oral liquid special for mink according to claim 2, wherein the
compound probiotic fermentation liquor comprises Lctobacillus (10-20
parts) and Bacillus (10-20 parts).
4. The compound probiotic fermented Chinese herbal medicine
health care oral liquid special for mink according to claim 1, wherein the
Chinese herbal medicine extract is prepared from Astragalus
membranaceus, Forsythia suspensa, Radix Isatidis, Flos Lonicerae and
Chrysanthemum;
and the parts by mass are as below: 20-30 parts of Astragalus
membranaceus, 20-30 parts of Forsythia suspensa, 20-30 parts of
Radix Isatidis, 20-30 parts of Flos Lonicerae and 20-30 parts of
Chrysanthemum.
5. A preparation method of the compound probiotic fermented
Chinese herbal medicine health care oral liquid special for mink
according to claim 1, wherein the following preparation steps are
included: the preparation of compound probiotics fermentation liquor and
the Chinese herbal medicine extract respective, and then mixing the
prepared compound probiotics fermentation liquor with the Chinese
herbal medicine extract in proportion.
6. The preparation method according to claim 5, wherein the
preparation steps of compound probiotics fermentation liquor are
included:
A. The slant culture preservation is activated, Lactobacillus and
Bacillus are incubated in LB medium respectively, then autoclaved for
-40 min at pH 7-7.5 and temperature of 100-130°C, and cultured
at 30-38C for 12-20 h;
B. Perform the primary liquid expansion culture for Lactobacillus
and Bacillus respectively, with an inoculum amount of 10~20%;
C. Perform the secondary liquid expansion culture for the
Lactobacillus and Bacillus after the primary liquid expansion culture
respectively, with aninoculum amount of 10~20%;
D. Mix the fermentation liquor of Lactobacillus and Bacillus with a
weight ratio of 1:1 to obtain a compound probiotics fermentation liquor.
7. The preparation method according to claim 5, wherein the
preparation steps of the Chinese herbal medicine extract are included:
E. Crush the corresponding Chinese herbal medicine to 60-80
mesh, then mix well to obtain the described Chinese herbal medicine
powder after weighing the corresponding one respectively according to
the mass fraction;
F. Place the above Chinese herbal medicine powder and 300-600
parts of water into the extraction tank, soak for 5-15 h; then heat the
mixture to 40-100°C to extract for 10- 12 h, filter and collect the
extract; then add 300-600 parts of water to the extraction tank again for
the extration of the medicine residue at 40-100°C for 10-15 h;
J. The described Chinese herbal medicine extract is obtained by
combining the two extracts and concentrating in vacuum to 10-20% of
the original volume at 30-70°C.
8. The preparation method according to claim 7, wherein the method
specifically includes: add the compound probiotics fermentation liquor to
the obtained Chinese herbal medicine extract to obtain the mixing
solution; incubate at 25-35°C for 12-36 h; start the stir for 5 min per
12 h at a rotation speed of 120 r/min; the fermentation process will
continuous till the pH of the mixture drops to 3.0-3.2; the compound
probiotic fermented Chinese herbal medicine health care oral liquid
special for mink is obtained by filtration with 100 mesh filter cloth (4
layers) after stirring for 30 min.
