Verfahren zur Herstellung von neuen, in 3-Stellung mit einer einwertigen Schwefelfunktion substituierten Phenothiazin-Derivaten
Es wurde gefunden, dass man zu neuen, in 3-Stellung mit einer einwertigen Schwefelfunktion substituierten Phenothiazin-Derivaten der allgemeinen Formel I :

Gemäss der nicht zum Stande der Technik gehörenden österr. Patentschrift Nr. 198273 wur- den Mercapto-phenothiazin-Derivate der Formel I durch Kondensation von Mercapto-phenothiazinen mit I-Halogen-2- (N-alkyl-piperidyl : -2') - äthan hergestellt.
Gegenstand eines weiteren nicht zum Stande der Technick gehörenden Vorschlages (österr. Patentschrift Nr. 202563) ist ein Verfahren zur Herstellung von Mercapto-phenothiazin-Derivaten der Formel I durch Decarboxylierung der entsprechenden 10 - Phenothiazin - carbonsäure- ester-Derivate.
Das vorliegende Verfahren kann z. B. so durchgeführt werden, dass ein N- [2- (N'-Alkyl-piperi- dyl-2')-äthyl-l]-N- [mercapto-phenyl]-ani- lin-Derivat in einem geeigneten organischen Lösungsmittel wie Benzol, Toluol od. dgl. gelöst und unter Rühren bei Zimmertemperatur oder bei erhöhter Temperatur mit einem Schwefeldihalogenid versetzt wird. Nach beendeter Umsetzung wird das Lösungsmittel unter vermindertem Druck verdampft, und der Eindampfrückstand nach Verrühren mit Wasser und mit Alkalilauge in einem geeigneten mit Wasser nicht mischbaren Lösungsmittel aufgenommen. Der Eindampfrückstand der organischen Schicht wird zunächst durch Fraktionierung im Hochvakuum gereinigt und kann für die weitere Reinigung in das Salz einer geeigneten Säure übergeführt werden.
Dies ist insbesondere dann notwendig, wenn die Phenylkerne des Ausgangsmaterials substituiert sind, so dass bei der Reaktion ein Gemisch verschiedener Isomeren entstehen kann, die sich voneinander durch die Stellung der Substituenten unterscheiden. Die Trennung und Reindarstellung der Komponenten gelingt dann durch fraktionierte Kristallisation ihrer Salze.
Die nach dem vorliegenden Verfahren hergestellten, basischen Verbindungen sind bei Zimmertemperatur ölig oder kristallin und bilden mit Säuren feste, beständige Salze.
Die verfahrensgemäss hergestellten Verbindungen besitzen therapeutisch verwertbare pharmakodynamische Eigenschaften, wie z. B. Potenzierung des Effektes von narkotisch, hypnotisch oder analgetisch wirkenden Pharmaka und eignen sich deshalb zur Narkosevorbereitung. Sie können ausserdem bei allergischen Erkrankungen, sowie als Sedativa, Spasmolytika, Antiemetika oder Neuroplegika verwendet werden, und dienen ausserdem zur Behandlung von verschiedenartigen Erregungszuständen.

In den nachfolgenden Beispielen, die die Ausführung des Verfahrens erläutern, die Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.

According to Austrian Patent No. 198273, which does not belong to the state of the art, mercapto-phenothiazine derivatives of the formula I were obtained by condensation of mercapto-phenothiazines with I-halo-2- (N-alkyl-piperidyl: -2 ') - Ethane made.
The subject of a further proposal not belonging to the state of Technick (Austrian patent specification No. 202563) is a process for the preparation of mercapto-phenothiazine derivatives of the formula I by decarboxylation of the corresponding 10-phenothiazine-carboxylic acid ester derivatives.
The present method can e.g. B. be carried out so that an N- [2- (N'-alkyl-piperidyl-2 ') - ethyl-1] -N- [mercapto-phenyl] -aniline derivative in a suitable organic solvent such as benzene, toluene or the like. This is particularly necessary when the phenyl nuclei of the starting material are substituted so that the reaction can result in a mixture of different isomers which differ from one another in the position of the substituents. The separation and purification of the components then succeed through fractional crystallization of their salts.
The basic compounds produced by the present process are oily or crystalline at room temperature and form solid, stable salts with acids.
The compounds prepared according to the method have therapeutically utilizable pharmacodynamic properties, such as. B. potentiation of the effect of narcotic, hypnotic or analgesic drugs and are therefore suitable for anesthesia preparation. They can also be used for allergic diseases, as well as sedatives, spasmolytics, antiemetics or neuroplegics, and are also used to treat various types of excitement.
