Verfahren zur Herstellung von neuen 9,10-Dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen-Derivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen 9,10-Dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen-Derivaten der allgemeinen Formel I (siehe Formelblatt) und ihren Säureadditions- salzen, worin Ri ein Wasserstoffatom oder die Methylgruppe, Ra ein Wasserstoffatom, R3 eine niedere Alkylgruppe bedeuten, oder R3 mit Rl zusammen die Di-oder Trimethylengruppe, bzw. mit R2 zusammen die Trioder Tetramethylengruppe bedeuten, wobei das andere der Symbole Ri und R2 für ein Wasserstoffatom steht.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der allgemeinen Formel I und ihren Säureadditionssalzen, indem man eine Verbindung der allgemeinen Formel II, worin Ri, R2 und R3 obige Bedeutung besitzen, mit einem Chlorameisensäureester der allgemeinen Formel III, worin R4 eine niedere Alkyl-oder Aralkylgruppe bedeutet, umsetzt, die entstandene Verbindung der allgemeinen Formel IV einer Hydrolyse unterwirft und die erhaltene Verbindung der allgemeinen Formel I gegebenenfalls in ihre Säureadditionssalze überführt.
Die Ausführung des Verfahrens gestaltet sich beispielsweise wie folgt : Die Lösung eines Chlorameisen- säureesters, wie z. B. Chlorameisensäureäthylester oder Chlorameisensäurebenzylester, in einem indifferenten organischen wasserfreien Lösungsmittel, vorzugsweise Benzol, Toluol, Tetrachlorkohlenstoff oder Tetrahydrofuran, wird mit der im gleichen Lösungsmittel gelösten Verbindung der Formel II bei Raumtemperatur versetzt. Zur Vervollständigung der Reaktion erhitzt man das Gemisch noch 1-3 Stunden zum Sieden am Rückfluss oder lässt das Reaktionsgemisch mehrere Stunden bei Raumtemperatur stehen. Die als Zwischenprodukt erhaltene Verbindung der Formel IV wird nach bekannten Methoden isoliert und gereinigt.
In der nächsten Verfahrensstufe wird die Alkoxy carbonyl-oder Aralkoxycarbonylgruppe hydrolytisch durch ein Wasserstoffatom ersetzt. Man verfährt dabei z. z. m der Weise, da).) man die veroindung in emem niederen Alkanol, vorzugsweise n-Butanol, mit einem Alkalihydroxyd, z. B. Kaliumhydroxyd, mehrere Stunden erhitzt.
Die Abspaltung des Alkoxycarbonyl-oder Aralk oxycarbonylrestes gelingt indessen auch in saurem Medium, z. B. durch Erhitzen mit starken anorganischen Säuren, beispielsweise mit 48 % igem Bromwasserstoff.
Die erhaltene Verbindung wird in bekannter Weise aus dem Reaktionsgemisch isoliert und durch Kristallisation oder durch Überführung in ein geeignetes Salz gereinigt. Solche Salze sind beispielsweise die Hydrochloride, Hydrobromide, Phosphate, Sulfate, Acetate, Malonate, Fumarate, Maleinate, Tartrate, Malate, Hexahydrobenzoate, Benzolsulfonate oder p-Toluolsulfonate.
Die neuen Verbindungen der Formel I besitzen wertvolle pharmakodynamische Eigenschaften. So zeichnen sie sich durch starke, für Antidepressiva typische Wirkungen aus, die sich am Tier u. a. in einer Hemmung der durch Reserpin oder Tetrabenazin hervorgerufenen vegetativen und motorischen Symptome äussern. Die Verbindungen potenzieren die Noradrenalinwirkung und besitzen vorwiegend zentral gerichtete anticholinergische Effekte. Die sedativ/neuroleptischen Eigenschaften sind bei den Substanzen nur mässig entwickelt. Die antidepressive Wirkung ist also insofern spezifisch, als die dämpfenden Eigenschaften bei den Verbindungen eher zurücktreten. Die Toxizität der Verfahrensprodukte ist verhältnismässig gering.
Die neuen Verbindungen sollen zur Behandlung neurotischer und psychotischer Störungen, vor allem des depressiven, Formenkreises, Verwendung finden, ebenso können sie zur Therapie psychosomatischer Störungen verwendet werden. Sie werden vorzugsweise in Form ihrer physiologisch verträglichen, wasserlös- lichen Salze verabreicht.
Die als Ausgangssubstanzen verwendeten Verbindungen der allgemeinen Formel II sind neu und können wie folgt hergestellt werden : Die Lösung einer Verbindung der Formel V, wie beispielsweise 4- (3-Di- methylamino-propyliden)-9, 1 0-dihydro-4H-benzo [4,5]cyclohepta [1, 2-b] thiophen in Eisessig wird mit rotem Phosphor und Jodwasserstoffsäure 10 Minuten bis 2 Stunden auf 120 C erhitzt. Nach Filtration des Reaktionsgemisches und Eindampfen des Filtrates wird der Rückstand in Gegenwart von Alkalien, z. B. 20 %- iger Natronlauge, mit einem organischen Lösungsmittel, vorzugsweise Methylenchlorid, ausgeschüttelt.
Aus der organischen Phase wird das Jod mittels Natriumthiosulfat entfernt und das gewünschte Endprodukt in bekannter Weise isoliert und vorzugsweise durch tuber- führung in ein geeignetes Salz gereinigt.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind nicht korrigiert.

Beispiel 1 4- (3-Methylamino-propyl)-9, 10-dihydro-4H- benzo [4,5] cyclohepta [1, 2-b] thiophen a) 4- (3-Methyl-3-äthoxycarbonylamino-propyl)-9, 10 dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen
Zu einer Lösung von 21,0 g Chlorameisensäure- äthylester in 70 cm3 abs. Benzol lässt man während 30 Minuten bei Raumtemperatur eine Lösung von 18,0 g
4- (3-Dimethylamino-propyl)-9, 10-dihydro-4H benzo [4,5] cyclohepta [1, 2-b] thiophen in 70 cm3 abs. Benzol zutropfen.
Anschliessend erhitzt man das Reaktionsgemisch unter Rühren noch 5 Stunden zum Sieden, wäscht es nach dem Abkühlen dreimal mit 2n Salzsäure, dann noch zweimal mit Wasser und trocknet die Lösung über Magnesiumsulfat. Nach dem Eindampfen des Lösungsmittels erhält man das reine 4- (3-Methyl-3-äthoxyvarbonylamino-propyl)-9, 10i dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen. n 2E) = 1,5745.
b) 4- (3-Methylaminopropyl)-9, 10-dihydro-4H benzo [4,5] cyclohepta [1, 2-b] thiophen
Man erhitzt die Lösung von 16,5 g 4- (3-Methyl-3- äthoxycarbonylamino-propyl)-9, 10-dihydro-4H-benzo- [4,5] cyclohepta [1, 2-b] thiophen, 13,5 g Kaliumhydroxyd und 140 cm3 n-Butanol unter Rühren während 4 Stun den in einer Stickstoffatmosphäre zum Sieden. Anschlie ssend dampft man das Lösungsmittel ab, löst den Rück- stand bei 80'ion einem Gemisch von 140 cm3 Wasser und 180 cm3 2n Schwefelsäure und kühlt die Lösung ab.
Diese Lösung wird nun zweimal mit Hexan aus geschüttelt, der organische Extrakt noch zweimal mit
2n Schwefelsäure extrahiert, die wässrigen Extrakte unter
Kühlen mit Natronlauge alkalisch gestellt und dreimal mit Methylenchlorid extrahiert. Nach Trocknen der ver einigten Methylenchloridextrakte über Magnesium sulfat wird das Lösungsmittel unter vermindertem
Druck abgedampft. Die Lösung des Rückstandes in
20 cm3 Athanol versetzt man darauf mit der berechneten Menge äthanolischer Salzsäure. Nach einiger Zeit kristallisiert das Hydrochlorid aus. Es wird aus Äthanol umkristallisiert. Smp. 210,5-212 .
Das als Ausgangsmaterial verwendete 4- (3-Dime- thylamino-propyl)-9, 10-dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen wird wie folgt hergestellt :
Die Lösung von 12,4 g 4- (3-Dimethylamino-propyl- iden)-9,10-dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen-hydrochlorid in 240 cm3 Eisessig wird mit 12,0 g rotem Phosphor und 64 cm3 56% iger Jodwasserstoff- säure versetzt und das Gemisch unter Rühren wäh- rend 15 Minuten auf 120 erhitzt. Anschliessend wird das Reaktionsgemisch filtriert und das Filtrat unter vermindertem Druck eingedampft.
Der ölige Rückstand wird in 20% iger Natronlauge und in Methylenchlorid gelöst, die organische Phase abgetrennt und der wässrige Teil noch zweimal mit Methylenchlorid extrahiert. Die vereinigten Methylenchloridextrakte werden zweimal mit
5 % iger Natriumthiosulfatlösung, dann noch zweimal mit Wasser gewaschen, über Kaliumcarbonat getrocknet und unter vermindertem Druck eingedampft. Die Lösung des Rückstandes-in 35 cm3 Aceton versetzt man darauf mit der berechneten Menge Chlorwasserstoff in Aceton und lässt die Lösung langsam auf Raumtemperatur abkühlen. Das ausgefallene Hydrochlorid wird abfiltriert und aus Aceton kristallisiert. Smp. 158,5 bis

Beispiel 2 4 (3-Methylamino-2-methyl-propyl)-9, 10-dihydro
4H-benzo [4,5] cyclohepta [l, 2-b] thiophen
Analog Beispiel 1 erhält man durch Umsetzen von 4- (3-Dimethylamino-2-methyl-propyl)-9, 10-dihydro-4Hbenzo [4,5] cyclohepta [1, 2-b] thiophen mit Chlorameisen- säureäthylester und anschliessender Hydrolyse die ge wünschte Verbindung. Das Hydrochlorid schmilzt nach Umkristallisieren aus Äthanol bei 210,5-212 (Zers.).
Das als Ausgangsmaterial verwendete 4- (3-Dimethyl- amino-2-methylpropyl)-9, 10-dihydro-4H-b, enzo [4,5]cyclohepta [l, 2-b] thiophen wird wie folgt hergestellt :
Die Lösung von 1,0 g 4- (3-Dimethylamino-2-methyl- propyliden)-9,10-dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiopen in 18 cm3 Eisessig wird mit 0,9 g rotem Phosphor und 5,2 cm3 56 % iger Jodwasserstoffsäure versetzt und das Gemisch unter Rühren während 10 Minuten auf 120 erhitzt. Anschliessend wird das Reaktionsgemisch filtriert und das Filtrat unter vermindertem Druck eingedampft.
Der ölige Rückstand wird zwischen 20 % iger Natronlauge und Methylenchlorid verteilt, die organische Phase abgetrennt und der wäss- rige Teil noch zweimal mit Methylenchlorid extrahiert.
Die vereinigten Methylenchloridextrakte werden zweimal mit 5 % iger Natriumthiosulfatlösung, dann noch zweimal mit Wasser gewaschen, über Kaliumcarbonat getrocknet und unter vermindertem Druck eingedampft.
Die Lösung des Rückstandes in Aceton versetzt man darauf mit der berechneten Menge Chlorwasserstoff in Aceton und lässt die Lösung langsam auf Raumtem- peratur abkühlen. Das ausgefallene Hydrochlorid (Diastereoisomerengemisch) wird abfiltriert und aus Isopropanol umkristallisiert. Smp. 186,5-188,5 (Zersetzung).
Beispiel 3 4- [2- (2-Piperidyl)-äthyl]-9, 10-dihydro-4H- benzo [4,5] cyclohepta [1, 2-b] thiophen
Analog Beispiel 1 erhält man durch Umsetzen von 4- [2-(1-Methyl-2-piperidyl)-äthyl]-9,10-dihydro-
4H-benzo [4,5] cyclohepta [1, 2-b] thiophen mit Chlorameisensäureäthylester und anschliessender Hydrolyse die im Titel genannte Verbindung. Das Iso merengemisch destilliert bei einem Druck von 0,08 mm Hg bei 165-170 als gelbliches Öl.
Hydrochlorid : Eine acetonische Lösung der destillierten Base versetzt man mit der berechneten Menge acetonischer Salzsäure. Nach mehreren Stunden filtriert man das ausgefallene Hydrochlorid ab und kristallisiert es aus Aceton um. Smp. 164-169 (Zers.) (Isomerengemisch).
Das als Ausgangssubstanz verwendete 4- [2- (l-Me- thyl-2-piperidyl)-äthyl]-9, 10-dihydro-4H-benzo [4,5]cyclohepta [1, 2-b] thiophen wird analog der in den Beispielen 1 und 2 verwendeten Ausgangsmaterialien erhalten, indem man 1,0 g 4- [2- (l-Methyl-2-piperidyl)- äthyliden]-9, 10-dihydro-4H-benzo [4,5] cyclohepta [1, 2-b'thiophen mit 1,0 g rotem Phosphor, 5,6 cm3 56% iger Jodwasserstoffsäure und 60 ce Eisessig während 10 Minuten zum Sieden erhitzt. Das Hydrochlorid (Diastereoisomerengemisch) schmilzt nach Umkristallisieren aus Aceton bei 208,5-210,5 (Zers.).
Beispiel 4 4- (3-Methylamino-propyl)-9, 10-dihydro-4H- benzo [4,5] cyclohepta [1, 2-b] thiophen
Die Abspaltung der Athoxycarbonylgruppe aus der gemäss Beispiel 1 a hergestellten Verbindung kann auch mit Bromwasserstoff ausgeführt werden.
Die Lösung von 2 g 4- (3-Methyl-3-äthoxycarbonyl- aminopropyl)-9,10-dihydro-4H-benzo [4,5] cyclohepta [1, 2-b] thiophen in 25 cm3 48% iger Bromwasserstoffsäure wird in einer Stickstoffatmosphäre während 30 Minuten zum Sieden erhitzt. Dann giesst man in 250 cin Eiswasser, stellt die erhaltene Lösung mit Natronlauge alkalisch und schüttelt die Lösung dreimal mit Methylenchlorid aus. Die organischen Extrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel eingedampft. Die Lösung des Rück- standes in Äthanol versetzt man darauf mit der berech- neten Menge äthanolischer Salzsäure. Nach einiger Zeit kristallisiert das Hydrochlorid aus.
Smp. 210,5-212 aus Äthanol.
Process for the preparation of new 9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene derivatives
The present invention relates to a process for the preparation of new 9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene derivatives of the general formula I (see formula sheet) and their acid addition salts, in which Ri is a hydrogen atom or the methyl group, Ra is a hydrogen atom, R3 is a lower alkyl group, or R3 and Rl together are the di- or trimethylene group, or with R2 together are the tri or tetramethylene group, the other of the symbols Ri and R2 being a hydrogen atom .

The compounds potentiate the norepinephrine effect and have mainly centrally directed anticholinergic effects. The sedative / neuroleptic properties of the substances are only moderately developed. The antidepressant effect is specific in that the depressant properties of the compounds tend to recede. The toxicity of the process products is relatively low.
The new compounds should be used for the treatment of neurotic and psychotic disorders, especially the depressive ones, and they can also be used for the therapy of psychosomatic disorders. They are preferably administered in the form of their physiologically compatible, water-soluble salts.
The compounds of general formula II used as starting substances are new and can be prepared as follows: The solution of a compound of formula V, such as 4- (3-dimethylamino-propylidene) -9, 10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene in glacial acetic acid is heated to 120 ° C. for 10 minutes to 2 hours with red phosphorus and hydriodic acid. After filtration of the reaction mixture and evaporation of the filtrate, the residue in the presence of alkalis, eg. B. 20% sodium hydroxide solution, extracted with an organic solvent, preferably methylene chloride.
The iodine is removed from the organic phase by means of sodium thiosulfate and the desired end product is isolated in a known manner and preferably purified by transferring it to a suitable salt.

To a solution of 21.0 g of ethyl chloroformate in 70 cm3 of abs. Benzene is left in a solution of 18.0 g for 30 minutes at room temperature

The solution of 16.5 g of 4- (3-methyl-3-ethoxycarbonylamino-propyl) -9, 10-dihydro-4H-benzo- [4,5] cyclohepta [1,2-b] thiophene, 13, 5 g of potassium hydroxide and 140 cm3 of n-butanol with stirring for 4 hours in a nitrogen atmosphere to boiling. The solvent is then evaporated off, the residue is dissolved at 80% in a mixture of 140 cm3 of water and 180 cm3 of 2N sulfuric acid and the solution is cooled.
This solution is then shaken out twice with hexane, the organic extract with twice more
2n sulfuric acid extracted, the aqueous extracts under

5% sodium thiosulphate solution, then washed twice with water, dried over potassium carbonate and evaporated under reduced pressure. The solution of the residue in 35 cm 3 of acetone is then mixed with the calculated amount of hydrogen chloride in acetone and the solution is allowed to cool slowly to room temperature. The precipitated hydrochloride is filtered off and crystallized from acetone. M.p. 158.5 to

Example 2 4 (3-methylamino-2-methyl-propyl) -9,10-dihydro
4H-benzo [4,5] cyclohepta [1,2-b] thiophene
Analogously to Example 1, reaction of 4- (3-dimethylamino-2-methyl-propyl) -9, 10-dihydro-4Hbenzo [4,5] cyclohepta [1,2-b] thiophene with ethyl chloroformate and subsequent hydrolysis is obtained the desired connection. After recrystallization from ethanol, the hydrochloride melts at 210.5-212 (decomp.).
The 4- (3-dimethylamino-2-methylpropyl) -9, 10-dihydro-4H-b, enzo [4,5] cyclohepta [1,2-b] thiophene used as starting material is prepared as follows:
The solution of 1.0 g of 4- (3-dimethylamino-2-methylpropylidene) -9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene in 18 cm3 of glacial acetic acid is added 0.9 g of red phosphorus and 5.2 cm3 of 56% strength hydriodic acid were added and the mixture was heated to 120 for 10 minutes while stirring. The reaction mixture is then filtered and the filtrate is evaporated under reduced pressure.
The oily residue is distributed between 20% strength sodium hydroxide solution and methylene chloride, the organic phase is separated off and the aqueous part is extracted twice more with methylene chloride.
The combined methylene chloride extracts are washed twice with 5% sodium thiosulphate solution, then twice more with water, dried over potassium carbonate and evaporated under reduced pressure.
The solution of the residue in acetone is then mixed with the calculated amount of hydrogen chloride in acetone and the solution is slowly allowed to cool to room temperature. The precipitated hydrochloride (mixture of diastereoisomers) is filtered off and recrystallized from isopropanol. M.p. 186.5-188.5 (decomposition).
Example 3 4- [2- (2-piperidyl) ethyl] -9, 10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene
Analogously to Example 1, by reacting 4- [2- (1-methyl-2-piperidyl) ethyl] -9,10-dihydro-
4H-benzo [4,5] cyclohepta [1, 2-b] thiophene with ethyl chloroformate and subsequent hydrolysis of the compound named in the title. The mixture of isomers distilled at a pressure of 0.08 mm Hg at 165-170 as a yellowish oil.
Hydrochloride: An acetone solution of the distilled base is mixed with the calculated amount of acetone hydrochloric acid. After several hours, the precipitated hydrochloride is filtered off and recrystallized from acetone. M.p. 164-169 (decomp.) (Mixture of isomers).
The 4- [2- (l-methyl-2-piperidyl) -ethyl] -9, 10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene is used as the starting substance analogously to that Starting materials used in Examples 1 and 2 obtained by adding 1.0 g of 4- [2- (1-methyl-2-piperidyl) ethylidene] -9, 10-dihydro-4H-benzo [4.5] cyclohepta [ 1,2-b'thiophene with 1.0 g of red phosphorus, 5.6 cm3 of 56% strength hydriodic acid and 60 ce of glacial acetic acid was heated to the boil for 10 minutes. The hydrochloride (mixture of diastereoisomers) melts after recrystallization from acetone at 208.5-210.5 (decomp.).
Example 4 4- (3-methylamino-propyl) -9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene
The cleavage of the ethoxycarbonyl group from the compound prepared according to Example 1a can also be carried out with hydrogen bromide.
The solution of 2 g of 4- (3-methyl-3-ethoxycarbonylaminopropyl) -9,10-dihydro-4H-benzo [4,5] cyclohepta [1,2-b] thiophene in 25 cm3 of 48% hydrobromic acid is heated to boiling in a nitrogen atmosphere for 30 minutes. It is then poured into 250 cc ice water, the solution obtained is made alkaline with sodium hydroxide solution and the solution is extracted three times with methylene chloride. The organic extracts are washed with water, dried over sodium sulfate and the solvent is evaporated. The solution of the residue in ethanol is then mixed with the calculated amount of ethanolic hydrochloric acid. After a while, the hydrochloride crystallizes out.
