Verfahren zur Darstellung basisch substituierter Äther bzw. Thioäther.
In den deutschen Patentschriften Erz 499826, 06 cl4747 und 541474 sind Verfahren zur Darstellung von an ihrer Aminogruppe durch basisehe Reste substituierten Aminooxy-und Polyamino- verbindungen der aromatischen Reihe bzw. deren Abkömmlingen beschrieben, die zu Verbindungen führen. welche teils die Atmung und den Blutdruck verbessern, teils in ausgesprochener Weise gegen Blutparasiten wirksam sind.
Es wurde gefunden, dass man zu Verbindungen mit ausgesprochen anästhesierender und AmoebenWirkung gelangt wenn man nach den in den genannten Patentschriften angegebenen Methoden in die ringständige Aminogruppe von aromatischen. alicyclischen oder heterocyclischen Aminoverbindungen. deren Kern mit einem aromatischen, alicyclischen oder heterocyclischen Rest äther-oder thioätherartiz ein-oder mehrfach verbunden ist. nach an sich üblichen Methoden aliphatische Reste einführt, die, gegebenenfalls heterocyclisch gebundene. Aminogruppen enthalten.
Der die ringständige Aminogmppe substituierende basische Rest kann ein oder mehrere Stickstoffatome enthalten und durch weitere Substituenten, z. B. Hydroxylgruppen, substituiert sein : er kann ferner äther- oder thioätherartige Bindungen enthalten und sich aus aliphatischen. isocyclischen und heteroeyclischen Bestandteilen zusammensetzen.
Die neuen Verbindungen können die die eyclischen Reste ätherartig verbindenden Sauerstoffbzw. Sehwefelatome ein oder mehrfach enthalten : die cyelischen Reste können unter sich gleich oder verschieden und auch mehrfach ätherartig miteinander verbunden sein, wie z. B. im Diphenylendioxyd.

Nacli dem Abkühlen trennt man die Benzolschirht von dem zähflüssigen Reaktionsprodukt, löst das letztere in Wasser macht alkalisch und nimmt den entstandenen 4-[ss-Diäthylaminoäthylamino]-phenolphenyläther in Äther auf.
Nach dem Abdampfen des Äthers wird das Reaktionsprodukt als hellgelbes Öl vom Kp ! = 1750 erhalten, dessen salzsaures Salz farblos und äusserst hygroskopisch ist.
Zum 4-[Bis-(ss-diäthylaminoäthylamino)]-phenolphenyläther gelangt man, wenn man 185 g 4-Amino- diphenyläther in 400 cm3 Nitrobenzol mit etwa 300 g 2-Chlor-1-diäthylaminoäthan während 10 Stunden auf 120-130"erhitzt. Das Nitrobenzol wird bei kongosaurer Reaktion mit Wasserdampf abgeblasen. gegebenenfalls nicht umgesetzter Ausgangsstoff aus der essigsauer gemachten Lösung durch Ausschütteln mit Äther entfernt und das Reaktionsprodukt nach Alkalisehmachen in Äther aufgenommen. Durch fraktionierte Destillation allein lässt sich nicht immer eine völlige Trennung von der stets als Nebenprodukt entstehenden sekundären Base erzielen.
Das noch sekundäre Base enthaltende Rohprodukt wird dann entsprechend dem analytisch festgestellten Gehalt an dieser mit wenig Benzol und etwa 20% mehr als der berechneten Menge Phthalsäureanhydrid 2 Stunden unter Rühren im siedenden Wasserbad erhitzt. Nach Zugabe von wenig mehr als der für die entstandene Phthalamidsäure berechneten Menge verdünnter Natronlauge wird mit Äther ausgeschüttelt. Aus der alkalischen Lösung kann die Phthalamidsäure mit verdünnter Essigsäure gefällt und aus ihr durch Verseifen nach üblichen Methoden die reine sekundäre Base gewonnen werden.
In der ätherischen Lösung befindet sich die tertiäre Base neben wenig Phthalsäureanhydrid. Man reinigt sie weiter durch Ausschütteln mit verdünnter Salzsäure, aus der sie nach Alkalischmachen wieder in Äther gebracht wird. Durch Destillation des Ätherrüekstandes erhält man die analytisch reine disubstituierte Base in Form eines fast farblosen Öles vom Kpt == 195. Für diese Trennung können sinngemäss auch alle sonstigen für die Trennung sekundärer und tertiärer Amine üblichen Methoden. z. B. die mit Chlorsulfonsäure, zur Anwendung gelangen.
An Stelle des 4-Aminodiphenyläthers können auch dessen Homologe oder Substitutionsprodukte zur Anwendung gelangen.
Beispiel 2: 37 g 4-Aminodiphenyläther, 32 g 4-Brom-1-diäthylaminopentan-hydrobromid und 25 m Wasser werden 8 Stunden bei 1 120-1300 verschmolzen. Das Reaktionsprodukt wird in verdünnter Salzsäure aufgenommen, die Base durch Alkalizusatz in Freiheit gesetzt und in Äther aufgenommen. Nach dem Trocknen und Verdampfen des Äthers wird durch Fraktionieren von überschüssigem 4-Amino- diphenyläther getrennt. Der 4-[(α-Methyl-#-diäthylaminobutyl)-amino]-diphenyläther destilliert unter 1 mm Druck bei 196 als hellgelbes Öl ; er liefert ein farbloses, stark hygroskopisches Hydrochlorid.
Beispiel 3 : 46 g 2'-Methoxy-2.4-diaminodiphenyläther (F = 106 ; gewonnen durch Umsetzung von Guajacolnatrium mit 4-Chlor-1. 3-dinitrobenzol und Reduktion der entstandenen Dinitroverbindung).
