Verfahren zur Herstellung von Alkaminestern Es wurde gefunden, daß
die aromatischen Ester der Aminoalkohole von der Formel
(wobei R und R' einen Alkylrest bedeuten, R" einen Alkylrest oder Wasserstoff bedeutet)
als Mittel zur örtlichen Schmerzbetäubung besonders günstige Eigenschaften zeigen.
Die als Ausgangsstoffe dienenden Alkoholbasen entstehen aus den nach dem Verfahren
des Patents 542 253, K1. i2o, erhaltenen Aldehydbasen durch Reduktion mit Natriumamalgam
in schwach essigsaurer Lösung. Bei der Einwirkung von aromatischen Säuren oder Derivaten,
vorzugsweise von Säurechloriden, auf die Alkoholbasen entstehen glatt und mit guter
Ausbeute die Ester, welche gut kristallisierte, auch in heißem Wasser beständige
Hydrochloride bilden. Beispiel i 5,5 Teile 3=Dimethylamino-2-dimethylpropan-i-ol
(Siedepunkt 166 bis 168° bei 76o mm) werden in 6 Teilen Chloroform gelöst und unter
Kühlung mit einer Lösung von 6 Teilen Benzoylchlorid in wenig Chloroform versetzt.
Man erwärmt sodann i Stunde auf ¢o° und fällt mit Äther. Das ausfallende Hydrochlorid
wird aus Isopropylalkohol umkristallisiert und schmilzt bei 153°. Beispiel e . 5,5
Teile 3-Dimethylamino-2-dimethylpropan-i-ol werden in 6 Teilen Chloroform gelöst
und mit einer Lösung von 12 Teilen p-Nitrobenzoylchlorid versetzt. Nach istündigem
Erwärmen auf 4.o° wird das Chloroform im Vakuum entfernt und der Rückstand mit Äther
verrührt, bis das Salz kristallinisch wird. Aus Alkohol umkristallisiert, schmilzt
es gegen. i8o°. i Teil des so gereinigten Produktes wird in 2o Teilen Wasser gelöst,
mit 0,3 Teilen i°joiger Palladiumchlorürlösung versetzt und mit Wasserstoff
bei 2 Atmosphären Druck geschüttelt. Wenn etwa 22o ccm Wasserstoff aufgenommen sind,
filtriert man und fällt aus der sauren Lösung die Base mit Natronlauge -aus. Sie
kristallisiert aus Petroläther mit dem Schmelzpunkt 79 bis 8o°. Auf die übliche
Weise wird daraus mit alkoholischer Salzsäure und Äther das kristallinische Hydrochlorid
vom Schmelzpunkt 930 gewonnen. Beispiel 3 ¢ Teile 3-Diäthylamino-2-dimethylpropani-ol
(Siedepunkt go bis g1° bei 12 min) werden mit 5 Teilen Chloroform verdünnt und unter
Kühlung mit 6 Teilen Benzoylchlorid versetzt. Man läßt einige Stunden stehen und
fällt mit Äther. Das ausfallende Chlorhydrat des 3-Diäthylamino-2-dimethyl-i-benzoyloxypropans
wird aus absolutemAlkohol unterZusatz von Äther umkristallisiert und schmilzt bei
131 bis 132°.
Beispiel a Fügt man . eine Lösung von 5,5 Teilen p-Nitrobenzoylchlorid
in Chloroform zu Teilen ` 3-Diäthylamino-2-dimethylpropani-ol, so bildet sich unter
Erwärmung das Hydrochlorid des 3-Diäthylamino-2-dimethyli-p-nitrobenzoyloxypropans.
Man läßt einige Stunden stehen, fällt die gebildete Verbindung mit Äther aus und
kristallisiert sie aus Isopropylalkohol um. Schmelzpunkt i6o°. i Teil dieses Zwischenproduktes
wird in 15 Teilen Wasser gelöst und nach Zusatz von 0,3 Teilen einer i°1oigen Palladiumchlorürlösung
und o, i Teil Tierkohle bei 2 Atmosphären Druck hydriert. Nach beendigter Wasserstoffaufnahme
wird filtriert und die Lösung im Vakuum eingeengt. Der Rückstand wird aus Isopropylalkohol
umkristallisiert. Das 2-Dimethyl-3-diäthvlamino-i-paminobenzoyloxypropanhydrochlorid
schmilzt bei 195 bis 196°. Beispiels Zu einer Lösung von q. Teilen 3=DimethylaMino-2-isopropylpropan-i-ol
in 5 Teilen Chloroform gibt man eine Lösung von 5 Teilen p-Nitrobenzoylchlorid in
möglichst wenig Chloroform langsam unter Kühlung. Im Laufe von einigen Stunden .kristallisiert
das Hydrochlorid des 3-Dimethylamino-2-isopropyl-i-p-nitrobenzoyloxypropans als
schwach gelblich gefärbte Nädelchen vom Schmelzpunkt 17q.° aus. Es kann in der üblichen
Weise, am besten durch katalytische Hydrierung, in die p-Aminoverbindung vom Schmelzpunkt
95° umgewandelt werden. Das Hydrochlorid dieser Base schmilzt bei 17o bis 171°.
Beispiel 6 5 Teile Benzoesäure und 5 Teile 3-Dimethylamino-2-dimethylpropan-i-ol
werden 3 Stunden unter ständigem Durchleiten von trockenem Chlorwasserstoffgas bei
ioo° gehalten. Sodann wird die Schmelze in Wasser aufgenommen, die überschüssige
Benzoesäure mit Äther entfernt, der wässerige Teil mit Natronlauge alkalisch gemacht
und ausgeäthert. Der Äther wird nach dem Trocknen abdestilliert, die unveränderte
Ausgangsbase bei 6q.° und 12 mm Druck abdestilliert und der Rückstand nach Zusatz
von alkoholischer Salzsäure und Äther als Hydrochlorid gewonnen. Man erhält die
Verbindung als farblose, silberglänzende Blättchen vom Schmelzpunkt i53°. Beispiel
? Man löst 2i2 Teile 3-Diäthylamino-2-dimethylpropan-i-ol in 318 Teilen Chloroform
und läßt eine Lösung von 246 Teilen o-Nitrobenzoylchlorid in 5oo Teilen Chloroform
unter Kühlung zutropfen. Nach mehrstündigem Stehen wird das Chlordform abdestilliert.
Das Hydrochlorid des o-Nitrobenzoesäureesters des Diäthylaminodimethylpropanols
hinterbleibt als ein in Wasser leicht lösliches Öl. Durch Reduktion mit Zinnchlorür
und Salzsäure gewinnt man daraus in üblicher Weise die o-Aminoverbindung, deren
Hydrochlorid aus Aceton mit dem Schmelzpunkt 13o° kristallisiert. ' PATENTANSPRUCH:
