Abstract
Un kit de diagnóstico de fundamento molecular para la detección de secuencias nucleotídicas de ADN, ADN copia y ARN, provenientes de agentes infecciosos, transgenes, alelos o secuencias no codificantes, sin necesidad de realizar pasos de aislamiento inicial, que comprende: al menos un primer frasco gotero calibrado con sistema anticontaminación, conteniendo una solución de la enzima polimerasa, agentes caotrópicos, sales y desoxitibonucleótidos; un segundo frasco gotero calibrado con sistema anticontaminación, conteniendo un set de al menos 4 primers, con al menos parte de sus secuencias conservadas en el fragmento de ácido nucleico que desea detectarse; un gotero con la mezcla de reactivos de revelado; un soporte para colocar la muestra a evaluar; al menos un sistema de revelado (por LFD o por reacción colorimétrica o fluorogénica); controles positivos y negativos; donde la enzima polimerasa se selecciona de la familia de enzimas BST, que está formada por DNA polimerasas / helicasas, sin actividad exonucleasa; y el set de primers se diseña a partir de la secuencias génica que desea detectarse, ya sea de naturaleza desoxirribonucleica o ribonucleica. El soporte puede ser un tubo o una bandeja de reacción o bien un papel de filtro o similar impregnado con la muestra, colocado en uno de dichos continentes. El método para detectar agentes infecciosos comprende los pasos de: colocar la muestra dentro del soporte, colocar al menos una gota del primer gotero dentro del continente de reacción, colocar al menos una gota del segundo gotero dentro del continente de reacción, tapar el continente, mezclar hasta homogeneizar los componentes de la solución, dar lugar a la reacción isotérmica de amplificación, mezclar y leer el resultado por cambio de color o fluorescencia. Si el método de lectura es por cromatografía en tiras reactivas, adicionar a la reacción una gota del gotero del revelado, introducir una tira reactiva LFD dentro del tubo, dejar que el contenido de la reacción de amplificación avance por capilaridad a lo largo de la LFD por unos 2 - 5 minutos, y leer el resultado. Si el soporte es un papel de filtro la parte con la muestra se coloca dentro del tubo de reacción o la bandeja de reacción.
