Abstract

La presente descripción proporciona métodos para detectar e identificar eventos de plantas que contienen loci (lugares o sitios) genómicos elegidos con precisión, y plantas y células de plantas que comprenden tales loci genómicos elegidos. El método puede llevarse a cabo como un proceso de alto rendimiento utilizado para seleccionar la condición intacta o la disrupción de un loci genómico elegidos y opcionalmente para detectar una inserción de polinucleótidos de ADN donante en los loci genómicos elegidos. Los métodos son fácilmente aplicables para la identificación de eventos de plantas producidos vía un método de determinación que resulta del uso de una nucleasa específica de sitio. Reivindicación 1: Un método para identificar la presencia de un polinucleótido de ADN donante exógeno insertado dentro de un locus o sitio genómico objetivado, que comprende: a) amplificar en una primera reacción de amplificación una muestra de ADN genómico que comprende el locus genómico objetivado utilizando una primera pluralidad de oligonucleótidos que se unen en condiciones de hibridación cerca del locus genómico objetivado, para así generar un primer amplicón que comprende el locus genómico objetivados; y b) detectar la presencia o ausencia del primer amplicón, en donde la ausencia del primer amplicón indica la presencia del polinucleótido de ADN donante dentro del locus genómico objetivado.
