Abstract
Se proporcionan composiciones y métodos para proteger una planta de un insecto plaga. Se proporcionan moléculas de ácidos nucleicos que codifican protoxinas de insectos o toxinas de insectos modificadas para que comprendan al menos un sitio de activacion proteolítica que es sensible a una proteasa intestinal de los insectos. El corte de una protoxina de insecto modificada en el sitio de activacion proteolítica por una proteasa intestinal de los insectos produce una toxina de insecto activa en el intestino del insecto plaga. El corte de una toxina de insecto modificada de la presente en un sitio de activacion proteolítica resulta en la produccion de una toxina de insecto activa en el intestino del insecto, que presenta una actividad plaguicida mejorada respecto de la toxina del insecto que carece del sitio de activacion proteolítica. Se proporcionan métodos para usar las secuencias de la protoxina de insecto modificada y la toxina de insecto modificada, y los polipéptidos que codifican, para proteger la planta el dano provocado por el insecto plaga. En realizaciones particulares de la presente se proporcionan otras composiciones y formulaciones de protoxinas de insectos modificadas y toxinas de insectos modificadas, cassettes de expresion y plantas, células vegetales y semillas transformadas. En la presente documentacion también se describen proteasa intestinales de insectos y moléculas de ácidos nucleicos que las codifican.
