  • Skin is the body's first line of defense against infections and environmental stressors. It acts as a major physical and immunological protective barrier, but also plays a critical role in temperature regulation, water holding, vitamin D production, and sensing. Skin care compositions are disclosed herein. In accordance with an aspect, provided is a skin care composition including a skin care composition comprising: palmitoyl sh-tripeptide-3 amide; hyaluronic acid and/or a salt thereof; a lipid mimetic; and an amino acid system.

BACKGROUND
[0002] Skin is the body's first line of defense against infections and environmental stressors. It acts as a major physical and immunological protective barrier, but also plays a critical role in temperature regulation, water holding, vitamin D production, and sensing. Its outermost surface consists of a lipid- and protein-laden cornified layer dotted with hair follicles and eccrine glands that secrete lipids, antimicrobial peptides (AMPs), enzymes, salts, etc. It harbors microbial communities living in a range of physiologically and anatomically distinct niches. Overall, this constitutes a highly heterogeneous and complex system.
[0003] Common skin diseases or disorders include pressure ulcers caused by friction and shearing, diaper or infant rashes, incontinence dermatitis and abraded, excoriated skin, eczema, psoriasis and general dermatitis, including contact dermatitis, atopic dermatitis and seborrheic dermatitis. Other common skin injuries include flame, sun, and radiation induced burns, bites from insects and animals, sports chafing and heat rash injuries, and infections from ingrown hairs or nails in the nailbed, along with dry, cracked feet, hands and extremities due to both disease and environmental factors.
[0004] The wound healing process, depending on the type of injury to the skin, is an intricate process that involves the steps of inflammation, proliferation, and remodeling. During inflammation, bacteria and debris are phagocytized and removed, and factors are released that cause the migration and division of cells involved in the proliferative phase. The proliferative phase is characterized by angiogenesis, deposition of collagen, formation of granular tissue, re- epithelialization, and wound contraction. In angiogenesis, new blood vessels are formed by vascular endothelial cells. In fibroplasia and formation of granular tissue, fibroblasts grow and form a new, provisional extracellular matrix by excreting collagen and fibronectin. Simultaneously, re-epithelialization occurs, providing a new epithelial layer. It is at this stage that oxygenation of the skin is critical to wound healing. In the remodeling phase, collagen is remodeled and realigned along tension lines and cells that are no longer needed are removed by apoptosis.
[0005] The health of wounded skin can be improved by promoting the healing process of such wound skin. In accordance with at least one aspect of the instant invention, provided is skin care compositions that promote and/or increase the healing of skin wounds and/or improve the appearance of skin.
BRIEF SUMMARY

[0007] The present disclosure relates skin care compositions adapted to improve the appearance and/or health of skin. For example, the inventors discovered that skin care compositions containing certain combinations of palmitoyl sh-tripeptide-3 amide, a hyaluronic acid and/or a salt thereof, a lipid mimetic, and an amino acid system can achieve enhanced wound healing. Additionally, it was unexpectedly discovered that skin care compositions comprising specific combinations of palmitoyl sh-tripeptide-3 amide; andiroba oil; and a lipid mimetic in certain amounts increase proteins associated with cellular proliferation in wound healing.

EXAMPLES
Example 1
[0118] A non-limiting example composition (Example Composition A) was prepared in the form of a serum in accordance with aspects of the invention. The formulation for Example Composition A is shown in Table 1.

[0119] In vivo evaluations were performed to assess Example Composition A on wounded skin. Specifically, human skin explants were wounded using an ablative laser. The ablative laser treatments produced 200 pm wounds covering a 14x14 mm2 area with 10% coverage. After laser wounding, 20pL samples of either water, Example Composition A, or a Placebo Composition were applied and spread with an inoculation loop. The Placebo Composition was similar to Example Composition A, but did not include the inventive combination of ingredients included in Example Composition A. Tissues were harvested at different time-points over 48 hours and processed for the hematoxylin and eosin staining to visualize the wound healing. FIG. 1 provides images of the skin explants after receiving the application of water (a control), the Placebo Composition, or Example Composition A.
Example 3 [0120] Example Composition A was further evaluated to assess its effect on wounded skin. Specifically, human skin explants were washed, dried, and the epidermis was carefully and partially removed using a diamond burr tip to wound the skin samples. From the de-epidermized skin explant, samples having a size of 8x3 mm (0 x thickness) were collected, in order to maintain the cut in the middle, with an electronic puncher and cultured up to the planned endpoints. The skin samples were cultured in an air-liquid interface in a perforated ring of stainless steel in contact with culture medium (modified Williams’ E medium) up to planned time-points.
[0121] A 6 pl sample of water, Example Composition A, or Placebo Composition was applied to the skin samples. The skin samples were then stained using hematoxylin eosin staining and then statistically analyzed. The measures of variation as standard deviation and standard error of mean (sem) were applied to the original scores. The reduction of the wound width for the skin samples after receiving Example Composition A, Placebo Composition, or water is summarized and shown in FIG. 2.

Claims

CLAIMS What is Claimed is:
1. A skin care composition comprising: palmitoyl sh-tripeptide-3 amide; a hyaluronic acid and/or a salt thereof; a lipid mimetic; and an amino acid system.
2. The skin care composition according to claim 1, wherein the palmitoyl sh-tripeptide-3 amide is present in an amount from about 0.005 to about 1 wt.%, the hyaluronic acid and/or the salt thereof is present in an amount from about 0.1 to about 1.5 wt.%, the lipid mimetic is present in an amount from about 1 to about 9 wt.%, and the amino acid system is present in an amount from about 1 to about 5 wt.%, wherein all weight percentages are based on the total weight of the skin care composition.
3. The skin care composition according to claim 1 or claim 2, wherein the hyaluronic acid and/or the salt thereof is selected from an acetylated hyaluronic acid and/or a salt thereof.
4. The skin care composition according to any foregoing claim, wherein the lipid mimetic comprises one or more of jojoba oil/macadamia seed oil ester, squalene, phytosterylmacadamiate, or phytosterol.
5. The skin care composition according to any foregoing claim, wherein the lipid mimetic comprises jojoba oil/macadamia seed oil ester, squalene, and phytosterylmacadamiate.
6. The skin care composition according to claim 4 or claim 5, wherein the jojoba oil/macadamia seed oil esters and squalene are present in the skin care composition in a weight ratio of the jojoba oil/macadamia seed oil esters to the squalene of about 1 :2 to about 1 : 15.

The skin care composition according to any foregoing claim, wherein the amino acid system comprises two or more amino acids selected from the group consisting of arginine, lysine, serine, citrulline, ornithine, creatine, histidine, diaminobutanoic acid, diaminoproprionic acid, alanine, aminobutyrate, asparagine, cysteine, cystine, glutamine, glycine, hydroxyproline, isoleucine, leucine, methionine, phenylalanine, proline, serine, taurine, threonine, tryptophan, tyrosine, valine, and a combination of two or more thereof.
8. The skin care composition according to any foregoing claim, wherein the amino acid system comprises two or more of arginine, lysine, glycine, taurine, or tyrosine.
9. The skin care composition according to any foregoing claim, wherein the amino acid system comprises arginine, glycine, and taurine.
10. The skin care composition according to any foregoing claim further comprising: a fatty alcohol, optionally, present in an amount from about 1 to about 9 wt.%, based on the total weight of the skin care composition.
11. The skin care composition according to claim 10, wherein the fatty alcohol comprises octyldodecanol.
12. The skin care composition according to any foregoing claim further comprising: a polyol, optionally, present in an amount from about 2 to about 15 wt.%, based on the total weight of the skin care composition.
13. The skin care composition according to claim 12, wherein the polyol is selected from ethylene glycol, propylene glycol, butylene glycol, hexylene glycol, glycerin, diglycerin, diethylene glycol, dipropylene glycol, sorbitol, pentylene glycol, caprylyl glycol, 1,2-hexanediol, 1,2-pentanediol, and a combination of two or more thereof.
14. The skin care composition according to any foregoing claim further comprising: from about 60 to about 90 wt.% of water, based on the total weight of the skin care composition.
15. A skin care composition comprising: palmitoyl sh-tripeptide-3 amide; an acetylated hyaluronic acid and/or a salt thereof; a lipid mimetic; an amino acid system; and from about 60 to about 90 wt.% of water, wherein all weight percentages are based on the total weight of the skin care composition.
16. The skin care composition according to any foregoing claim further comprising: an andiroba oil, optionally, present in an amount from about 0.1 to about 5 wt.%.
17. A skin care composition comprising: palmitoyl sh-tripeptide-3 amide; a lipid mimetic; from about 0.1 to about 10 wt.% of a hydrophobic component, the hydrophobic component comprising andiroba oil; and from about 60 to about 90 wt.% of water, wherein all weight percentages are based on the total weight of the skin care composition.
18. The skin care composition according to claim 17 further comprising: an amino acid system, optionally, present in an amount from about 1 to about 5 wt.%.
19. The skin care composition according to claim 17 or claim 18, wherein the amino acid system comprises arginine, glycine, and taurine.
20. The skin care composition according to any one of claims 17 to 19, wherein the hydrophobic component further comprises octyldodecanol.
