Abstract
본 발명은 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질; 및 분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하는 나노구조체 및 이의 용도에 관한 것이다.
Description
본 발명은 사이클로덱스트린 유래 고분자 물질 및 게스트 물질을 포함하는 나노구조체 및 이의 용도에 관한 것이다.
고분자 물질에 안정적으로 기능성을 도입하기 위해서는 화학적 결합을 통한 직접 결합 방법과 물리적 결합을 통한 방법이 존재한다.
화학적 결합의 경우 안정적이고 강력하게 결합되지만 반응이 번거롭고 반응마다 재현성이 높지 못한다는 한계가 있다. 한편, 물리적 결합의 경우 간단하지만 화학적 결합에 비해 정확도가 떨어지며 대부분 다중 상호작용으로 인해 구조를 예측하는 것이 쉽지 않다는 한계가 있다.

그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
본 발명은 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질; 및 분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하는 나노구조체를 제공한다.
상기 사이클로덱스트린 유래 작용기는 알파-사이클로덱스트린 유래 작용기일 수 있다.
상기 고분자 물질은 카르복실기, 하이드록시기, 아민기, 아크릴기, 티올기 및 비닐기로 이루어진 군으로부터 선택된 하나 이상의 작용기를 가지는 천연 또는 합성 고분자 물질일 수 있다.
상기 고분자 물질은 알지네이트, 셀룰로오스, 키토산, 히알루론산, 덱스트란, 폴리아크릴산, 카르복시메틸셀룰로오스 및 콘드로이틴 설페이트로 이루어진 군으로부터 선택된 하나 이상을 포함할 수 있다.

상기 제2 모이어티는 폴리에틸렌글리콜계 친수성 고분자 유래일 수 있다.
상기 사이클로덱스트린 유래 작용기 및 상기 게스트 물질 간에 호스트-게스트 상호작용을 이룰 수 있다.
상기 나노구조체의 평균 입자크기는 10 nm ~ 200 nm일 수 있다.
본 발명의 다른 구현예로, 상기 나노구조체; 및 상기 나노구조체 내부에 담지되거나, 상기 나노구조체와 화학적으로 결합된 생리활성물질을 포함하는 생리활성물질 전달체를 제공한다.
상기 나노구조체 내부에 담지된 생리활성물질은 소수성 약물일 수 있다.
상기 나노구조체와 화학적으로 결합된 생리활성물질은 양이온성 생리활성물질일 수 있다.
본 발명에 따른 나노구조체는 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질; 및 분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하는 것을 특징으로 한다. 상기 게스트 물질은 양친성 물질이면서, 상기 사이클로덱스트린 유래 작용기 및 상기 게스트 물질 간에 호스트-게스트 상호작용을 이룸에 따라, 균일한 입자크기 및 입자분포를 가질 수 있다.
이로써, 본 발명에 따른 나노구조체는 생리활성물질 전달체, 부형제, 생체 재료 등으로 다양하게 활용가능하고, 특히, 상기 나노구조체 내 담지되거나 화학적으로 결합된 약물 등 다양한 생리활성물질의 체내 방출 속도를 효과적으로 조절할 수 있는 이점이 있다.

본 발명자들은 다양한 고분자 물질에 사이클로덱스트린을 도입한 다음, 이와 호스트-게스트 상호작용을 이루는 게스트 물질과 혼합하여 균일한 입자크기 및 입자분포를 가지는 나노구조체를 제조하였다.
이하, 본 발명을 상세히 설명한다.
사이클로덱스트린 유래 고분자 물질 및 게스트 물질을 포함하는 나노구조체
본 발명은 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질; 및 분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하는 나노구조체를 제공한다.
먼저, 본 발명에 따른 나노구조체는 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질을 포함하고, 상기 고분자 물질은 하기 화학식 1로 표시될 수 있다:

상기 화학식 1에서, 상기 R1은 고분자 물질의 단위 단량체이고, R2는 고분자 물질에 있어서, 사이클로덱스트린 유래 작용기(특히, 하이드록시기)와 화학적으로 결합된 작용기이다.
상기 사이클로덱스트린 유래 작용기는 글루코피라노오스 단위들이 α-(1,4) 결합을 통해 고리 구조를 하고 있으며, 6개 단위의 알파-사이클로덱스트린 유래 작용기, 7개 단위의 베타-사이클로덱스트린 유래 작용기 및 8개 단위의 감마-사이클로덱스트린 유래 작용기를 포함한다. 상기 사이클로덱스트린 유래 작용기의 고리 구조에 있어서, 내부는 소수성 특성을 가지고, 외부는 하이드록시기의 존재로 인해 친수성 특성을 가지는 것을 특징으로 한다.
상기 사이클로덱스트린 유래 작용기는 고리의 지름을 고려할 때, 알파-사이클로덱스트린 유래 작용기인 것이 바람직하나, 이에 한정되지 않는다.
상기 고분자 물질은 천연 또는 합성 고분자 물질로서, 상기 사이클로덱스트린 유래 작용기(특히, 하이드록시기)와 화학적으로 결합가능한 작용기를 가진다.
구체적으로, 상기 고분자 물질은 카르복실기, 하이드록시기, 아민기, 아크릴기, 티올기 및 비닐기로 이루어진 군으로부터 선택된 하나 이상의 작용기를 가지는 천연 또는 합성 고분자 물질일 수 있고, 카르복실기를 가지는 천연 또는 합성 고분자 물질인 것이 바람직하나, 이에 한정되지 않는다. 보다 구체적으로, 상기 고분자 물질은 알지네이트, 셀룰로오스, 키토산, 히알루론산, 덱스트란, 폴리아크릴산, 카르복시메틸셀룰로오스 및 콘드로이틴 설페이트로 이루어진 군으로부터 선택된 하나 이상을 포함할 수 있고, 카르복실기를 가지는 알지네이트, 폴리아크릴산 또는 카르복시메틸셀룰로오스인 것이 바람직하나, 이에 한정되지 않는다. 이때, 상기 고분자 물질은 중량평균분자량이 0.5 kDa 내지 1,000 kDa일 수 있고, 30 kDa 내지 250 kDa인 것이 바람직하나, 이에 한정되지 않는다.
다음으로, 본 발명에 따른 나노구조체는 분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하고, 상기 게스트 물질은 하기 화학식 2로 표시될 수 있다:

상기 화학식 2에서, 상기 R3은 게스트 물질의 제1 모이어티이고, R4는 게스트 물질의 제2 모이어티이다.
상기 게스트 물질은 양친성 물질일 수 있고, 구체적으로, 상기 제1 모이어티는 분자의 지름이 4.7 Å 보다 큰 소수성 모이어티일 수 있고, 상기 제2 모이어티는 분자의 지름이 4.7 Å 보다 작은 친수성 모이어티일 수 있으며, 상기 제1 모이어티 및 상기 제2 모이어티는 서로 화학적인 결합을 통해 연결될 수 있다.
상기 제1 모이어티는 레티놀, 베타-카로틴, 알파-카로틴, 트레티노인, 7-디히드로콜레스테롤, 프리비타민 D3, 콜레칼시페롤, 칼시디올, 칼시트리올, 칼시트로익산, 에고스테롤, 에고칼시페롤, 디히드로타취스테롤, 칼시포트리올, 타칼시톨, 디히드로에고칼시페롤, 토코페롤, 토코트리에놀, 나프토퀴논, 필로퀴논, 메나테트레논, 피토나디온, 콜레스테롤, 캄페스테롤, 시토스테롤, 스티그마스테롤 및 에르고스테롤로 이루어진 군으로부터 선택된 하나 이상의 소수성 물질 유래일 수 있다. 즉, 상기 제1 모이어티는 소수성을 가지면서, 상기 사이클로덱스트린 유래 작용기의 고리 구조에 비해 분자의 지름이 큰 것을 특징으로 하는바, 사이클로덱스트린 유래 작용기의 고리 구조를 통과하지 못하고 말단에 걸리게 된다. 또한, 상기 제1 모이어티가 소수성을 가짐에 따라, 소수성 생리활성물질을 용이하게 물리적으로 담지할 수 있다.
상기 게스트 물질 중에서 상기 제2 모이어티는 상기 고분자 물질 간에 호스트-게스트 상호작용을 이룰 수 있다. 구체적으로, 상기 제2 모이어티는 폴리에틸렌글리콜계 친수성 고분자 유래일 수 있다. 이때, 상기 제2 모이어티는 중량평균분자량이 50 Da 내지 100,000 Da일 수 있고, 500 Da 내지 1,000 Da인 것이 바람직하나, 이에 한정되지 않는다. 즉, 상기 제2 모이어티는 상기 사이클로덱스트린 유래 작용기의 고리 구조에 비해 분자의 지름이 작은 것을 특징으로 하는바, 사이클로덱스트린 유래 작용기의 고리 구조를 통과할 수 있다. 또한, 상기 제2 모이어티는 친수성을 가짐에 따라, 상기 사이클로덱스트린 유래 작용기의 고리 구조 외부와 호스트-게스트 상호작용을 이룰 수 있다. 또한, 상기 제2 모이어티가 친수성을 가짐에 따라, 친수성 생리활성물질을 용이하게 물리적으로 담지할 수 있다.
다시 말해, 상기 사이클로덱스트린 유래 작용기 및 상기 게스트 물질 간에 호스트-게스트 상호작용을 이룰 수 있다.
상기 호스트-게스트 상호작용은 화학적 결합(예컨대, 공유 결합)에 비해 약하나, 단순 물리적 결합에 비해 강력한 것을 특징으로 하며, 제어가능한 것을 특징으로 한다. 상기 호스트-게스트 상호작용은 상기 사이클로덱스트린 유래 작용기의 고리 구조 외부와 상기 제2 모이어티 간에 이루어질 수 있다. 또한, 호스트-게스트 상호작용으로 인하여, 균일한 입자크기 및 입자분포를 가질 수 있다. 특히, 상기 나노구조체의 평균입자크기는 상기 고분자 물질의 중량평균분자량, 소니케이션 조건 등에 따라 다양하게 제어가능하고, 일 예로, 상기 나노구조체의 평균입자크기는 10 nm ~ 200 nm인 것이 바람직하나, 이에 한정되지 않는다. 뿐만 아니라, 이러한 호스트-게스트 상호작용으로 인하여 나노구조체 내 담지되거나 화학적으로 결합된 약물 등 다양한 생리활성물질의 체내 방출 속도를 효과적으로 조절할 수 있는 이점이 있다.
생리활성물질 전달체 등
본 발명은 상기 나노구조체; 및 상기 나노구조체 내부에 담지되거나, 상기 나노구조체와 화학적으로 결합된 생리활성물질을 포함하는 생리활성물질 전달체를 제공한다.
먼저, 본 발명에 따른 생리활성물질 전달체는 상기 나노구조체 기반인 것으로, 상기 나노구조체에 대해서는 전술한바 있으므로, 중복 설명을 생략하기로 한다.
또한, 본 발명에 따른 생리활성물질 전달체는 상기 나노구조체 내부에 담지되거나, 상기 나노구조체와 화학적으로 결합된 생리활성물질을 포함한다.
먼저, 상기 생리활성물질은 상기 나노구조체 내부에 물리적으로 담지될수도 있고, 이때, 상기 생리활성물질은 소수성 약물일 수 있고, 낮은 용해도 및 낮은 투과성을 동시에 가진 BCS 클래스 Ⅳ 약물인 것이 바람직하나, 이에 한정되지 않는다. 이러한 소수성 약물은 상기 나노구조체의 소수성 부분과 소수성 상호작용을 할 수 있다. 구체적으로, 상기 소수성 약물은 독소루비신(Doxorubicin), 클로르타리돈(Chlorthalidone), 클로파미드(Clopamide), 푸로세미드(Furosemide), 니트로푸란토인(Nitrofurantoin), 파크리탁셀(Paclitaxel), 도세탁셀(Docetaxel), 이리노테칸(Irinotecan), 에토포사이드(Etoposide) 다우노루비신(Daunorubicin), 캠토테신(Camptothecin), 레티노이드(Retinoids), 수니티닙(Sunitinib) 및 레날리도마이드(Lenalidomide)로 이루어진 군으로부터 선택된 하나 이상일 수 있고, 독소루비신(Doxorubicin)인 것이 바람직하나, 이에 한정되지 않는다.
한편, 상기 생리활성물질은 상기 나노구조체의 고분자 물질 또는 게스트 물질과 화학적으로 결합(예컨대, 이온성 결합)될수도 있고, 이때, 상기 생리활성물질은 이온성 생리활성물질일 수 있고, 양이온성 생리활성물질인 것이 바람직하나, 이에 한정되지 않는다. 이러한 양이온성 생리활성물질은 예컨대, 단백질, 폴리펩타이드, 펩타이드, 백신, 유전자, 호르몬 등의 약물이거나, 치료용 세포일 수 있다. 구체적으로, 상기 나노구조체와 화학적으로 결합(예컨대, 이온성 결합)된 생리활성물질은 단백질 약물 중에서도 성장인자일 수 있고, 신경 성장인자(NGF), 전환 성장인자(TGF-β), 섬유아세포 성장인자(FGF), 골 형성 단백질(BMP), 혈관내피 성장인자(VEGF), 표피 성장인자(EGF), 인슐린-유사 성장인자(IGF), 혈소판-유래 성장인자(PDGF), 간세포 성장인자(HGF), 상피 성장인자, 안지오포이에틴-1, 안지오포이에틴-2, 뉴로트로핀, 태반 성장인자(PIGF), 과립구 콜로니 자극인자(G-CSF) 및 과립구 대식세포 콜로니 자극인자(GM-CSF)로 이루어진 군으로부터 선택된 하나 이상일 수 있고, 신경 성장인자(NGF)인 것이 바람직하나, 이에 한정되지 않는다.
상기 생리활성물질 전달체가 상기 생리활성물질로서 약물을 포함하는 경우, 상기 생리활성물질 전달체 전체 부피를 기준으로, 상기 약물의 부피는 1 × 10-8 내지 50 부피%, 바람직하게는 1 × 10-4 내지 20 부피%인 것이 바람직하나, 이에 한정되지 않는다. 한편, 상기 생리활성물질 전달체가 상기 생리활성물질로서 치료용 세포를 포함하는 경우, 상기 생리활성물질 전달체 전체 부피를 기준으로, 상기 치료용 세포의 부피는 1 × 10-8 내지 50 부피%인 것이 바람직하나, 이에 한정되지 않는다. 이와 같은 약물 또는 치료용 세포의 함량이 상기 범위보다 작은 경우에는 원하는 약물 또는 치료용 세포의 효과를 얻을 수 없고, 상기 범위보다 큰 경우에는 나노구조체 물성에 영향을 미칠 수 있어 바람직하지 않다.
상기 생리활성물질 전달체는 상기 생리활성물질의 효용성을 증가시키기 위한 첨가제를 추가로 포함할 수 있다. 이를 위해, 상기 첨가제는 고분자 물질 및 게스트 물질의 비율을 조절하여 원하는 물성을 얻거나, 나노구조체 내에서 생리활성물질의 안정성을 유지시키거나, 나노구조체 및 생리활성물질 사이에서 화학적 결합에 따른 상호 작용을 유도하여 생리활성물질의 체내방출 속도를 제어하거나, 치료용 세포 등 생리활성물질의 생체내 활성을 증가시킬 수 있다.
상기 생리활성물질 전달체가 상기 첨가제를 추가로 포함하는 경우, 상기 생리활성물질 전달체 전체 중량을 기준으로, 상기 첨가제의 중량은 1 × 10-6 내지 30 중량%, 바람직하게는 1 × 10-3 내지 10 중량%인 것이 바람직하나, 이에 한정되지 않는다. 구체적으로, 상기 첨가제는 양이온성 고분자(예컨대, 중량평균분자량 200 내지 750,000), 음이온성 고분자(예컨대, 중량평균분자량 200 내지 750,000), 아미노산, 펩타이드, 단백질, 지방산, 인지질, 비타민류, 약물, 폴리에틸렌글리콜 에스테르(예컨대, 중량평균분자량 300 내지 50,000), 스테로이드, 아민 화합물, 아크릴계 공중합체(예컨대, 중량평균분자량 300 내지 500,000), 유기용매, 보존제, 당류, 폴리올, 당함유 폴리올, 당함유 아미노산, 계면활성제, 당함유 이온, 규산염, 금속염 및 암모늄염으로 이루어진 군으로부터 선택된 하나 이상일 수 있다.
상기 검토한 바와 같이, 본 발명에 따른 나노구조체는 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질; 및 분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하는 것을 특징으로 한다. 상기 게스트 물질은 양친성 물질이면서, 상기 사이클로덱스트린 유래 작용기 및 상기 게스트 물질 간에 호스트-게스트 상호작용을 이룸에 따라, 균일한 입자크기 및 입자분포를 가질 수 있다.
이로써, 본 발명에 따른 나노구조체는 생리활성물질 전달체, 부형제, 생체 재료 등으로 다양하게 활용가능하고, 특히, 상기 나노구조체 내 담지되거나 화학적으로 결합된 약물 등 다양한 생리활성물질의 체내 방출 속도를 효과적으로 조절할 수 있는 이점이 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1: 알파-사이클로덱스트린 및 알지네이트 결합 고분자 물질(Mw = 50 kDa)의 제조(도 1)
3L 플라스크에 알파-사이클로덱스트린(alpha-cyclodextrin)(0.115 mol)을 넣은 후 MES 버퍼(pH 5.5) 1.5 L를 넣어 투명해질 때까지 용해시켰다.
한편, 알긴산 소듐(alginic acid sodium)(출처:Sigma-aldrich)(0.046 mol)을 MES 버퍼(pH 5.5) 1 L에 용해시켰다. 여기에 소량의 4-디메틸아미노피리딘(4-dimethylaminopyridine; DMAP)(0.00828 mol) 및 1-에틸-3-(3-디메틸아미노프로필)카보디아미드 하이드로클로라이드(1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride; EDC)(0.00828 mol)를 넣고 10분 동안 활성화시켜준 다음, 이를 알파-사이클로덱스트린이 용해된 3L 플라스크에 천천히 적하시켜(dropwise) 5일 동안 상온에서 반응을 진행하였다. 반응이 끝난 후 아래 방법을 통해 정제하였다: 1) 생성물을 에탄올에 침전시킨 후, 글래스 필터로 필터링시켰다. 2) 디메틸포름아미드(Dimethylformamide)로 3번 세척하였다. 3) 아세톤으로 2번 세척한 후, 증류수(D.W)로 용해시켰다. 4) D.W에 용해된 생성물을 원심분리기(9000 rpm, 20 분, 4℃)를 통해 불순물을 제거하였다. 5) 12~14 KDa MWCO 투석 멤브레인을 이용하여 3일 동안 D.W 투석을 진행한 후, 동결건조하였다. 동결건조된 생성물 구조를 1H-NMR을 통해 확인하였다:

실시예 2: 알파-사이클로덱스트린 및 폴리아크릴산 결합 고분자 물질(Mw = 30 kDa, 100 kDa)의 제조(도 2)
2L 플라스크에 알파-사이클로덱스트린(0.021 mol)을 넣은 후 MES 버퍼(pH 5.5) 600 mL를 넣어 투명해질 때까지 용해시켰다.
한편, 폴리아크릴산(polyacrylic acid)(출처:Polysciences)(0.0087 mol)을 MES 버퍼(pH 5.5) 200 mL에 용해시켰다. 여기에 소량의 DMAP(0.0015 mol) 및 EDC(0.0015 mol)를 넣고 10분 동안 활성화시켜준 다음, 이를 알파-사이클로덱스트린이 용해된 2L 플라스크에 천천히 적하시켜 5일 동안 상온에서 반응을 진행하였다. 반응이 끝난 후 아래 방법을 통해 정제하였다: 1) 생성물을 에탄올에 침전시킨 후, 글래스 필터로 필터링시켰다. 2) 아세톤으로 2번 세척한 후, 에틸아세테이트로 1번 세척한 다음, 증류수(D.W)로 용해시켰다. 3) D.W에 용해된 생성물을 원심분리기(9000 rpm, 20 분, 4℃)를 통해 불순물을 제거하였다. 4) 12~14 KDa MWCO 투석 멤브레인을 이용하여 3일 동안 D.W 투석을 진행한 후, 동결건조하였다. 동결건조된 생성물 구조를 1H-NMR을 통해 확인하였다:

실시예 3: 알파-사이클로덱스트린 및 카르복실메틸셀룰로오스 결합 고분자 물질(Mw = 250 kDa)의 제조(도 3)
3L 플라스크에 알파-사이클로덱스트린(0.0067mol)을 넣은 후 MES 버퍼(pH 5.5) 1.5 L를 넣어 투명해질 때까지 용해시켰다.
한편, 카르복실메틸셀룰로오스(carboxymethyl cellulose)(출처:Sigma -aldrich)(0.0268 mol)을 MES 버퍼(pH 5.5) 1.5 L에 용해시켰다. 여기에 소량의 DMAP(0.0268 mol) 및 EDC(0.0268 mol)를 넣고 10분 동안 활성화시켜준 다음, 이를 알파-사이클로덱스트린이 용해된 3L 플라스크에 천천히 적하시켜 5일 동안 상온에서 반응을 진행하였다. 반응이 끝난 후 아래 방법을 통해 정제하였다: 1) 생성물을 에탄올에 침전시킨 후, 글래스 필터로 필터링시켰다. 2) 디메틸포름아미드(Dimethylformamide)로 3번 세척하였다. 3) 아세톤으로 2번 세척한 후, 증류수(D.W)로 용해시켰다. 4) D.W에 용해된 생성물을 원심분리기(9000 rpm, 20 분, 4℃)를 통해 불순물을 제거하였다. 5) 12~14 KDa MWCO 투석 멤브레인을 이용하여 3일 동안 D.W 투석을 진행한 후, 동결건조하였다. 동결건조된 생성물 구조를 1H-NMR을 통해 확인하였다:

실시예 4: 토코페롤 및 폴리에틸렌글리콜 결합 물질(Mw = 500, 750, 1,000 Da)의 제조
1L 갈색 플라스크에 토코페롤 석시네이트(tocopherol succinate)(출처:TCI) (0.018839mol)을 넣은 후 DCM(Dichloromethane) 용매 200 mL에 용해시켰다.
한편, 500mL 플라스크에 폴리에틸렌글리콜(0.018839mol)(출처: Sigma-aldrich)을 넣은 후 DCM 300mL에 용해시켰다. 여기에 소량의 DMAP(0.00942mol) 및 N,N'-디이소프로필카보디이미드(N,N'-Diisopropyl Carbodiimide; DIC)(0.00942mol)를 넣고 10분 동안 활성화시켜준 다음, 이를 토코페롤 석시네이트 용액에 천천히 적하시켜 2일 동안 45℃에서 반응을 진행하였다. 반응이 끝난 후 아래 방법을 통해 정제하였다: 1) 생성물을 글래스 필터로 필터링시킨 후, 증발시켰다. 2) 클로로포름에 용해시킨 후, 1M HCl, NaHCO3, 브린(brine) 순으로 추출한 후, MgSO4로 처리하였다. 3) 글래스 필터로 필터링시킨 후, 증발시켰다. 4) 헥산에 용해시킨 후 원심분리기(9000 rpm, 20 분, 4℃)를 통해 불순물을 제거하였다. 5) 2K 투석 멤브레인을 D.W 투석을 3일 동안 진행한 후, 동결건조하였다. 동결건조된 생성물 구조를 1H-NMR을 통해 확인하였다:

실시예 5: 콜레스테롤 및 폴리에틸렌글리콜 결합 물질(Mw = 500, 750, 1,000 Da)의 제조
1L 갈색 플라스크에 콜레스테릴 클로로포메이트(cholesteryl chloroformate) (출처: TCI)(0.0222mol)을 넣은 후 DCM(Dichloromethane) 용매 200 mL에 용해시켰다. 여기에 메톡시폴리에틸렌글리콜로부터 합성된 아미노폴리에틸렌글리콜(0.0222mol)을 넣은 후, 트리에틸아민(triethylamine)(TEA)(0.0289 mol)을 넣어 2일 동안 45℃에서 반응을 진행하였다. 반응이 끝난 후 아래 방법을 통해 정제하였다: 1) 생성물을 글래스 필터로 필터링시킨 후, 증발시켰다. 2) 헥산에 원심분리기(9000 rpm, 20 분, 4℃)를 통해 불순물을 제거하였다. 3) 2K 투석 멤브레인을 D.W 투석을 3일 동안 진행한 후, 동결건조하였다. 동결건조된 생성물 구조를 1H-NMR을 통해 확인하였다:

실시예 6: 콜레칼시페롤 및 폴리에틸렌글리콜 결합 물질(Mw = 500, 750, 1000)의 제조
1L 갈색 플라스크에 콜레칼시페롤(cholecalciferol - SA)(출처:ThermoFisher SCIENTIFIC)(0.0561mol)을 넣은 후 DCM(Dichloromethane) 용매 300 mL에 용해시켰다.
한편, 500mL 플라스크에 폴리에틸렌글리콜(출처: Sigma-aldrich)(0.0561 mol) 4.2 g을 넣은 후 DCM 300mL에 용해시켰다. 여기에 소량의 DMAP(0.0561mol) 및 N,N'-디이소프로필카보디이미드(N,N'-Diisopropyl Carbodiimide(0.112mol); DIC)를 넣고 10분 동안 활성화시켜준 다음, 이를 콜레칼시페롤 용액에 천천히 적하시켜 2일 동안 45℃에서 반응을 진행하였다. 반응이 끝난 후 아래 방법을 통해 정제하였다: 1) 생성물을 글래스 필터로 필터링시킨 후, 증발시켰다. 2) 클로로포름에 용해시킨 후, 1M HCl, NaHCO3, 브린(brine) 순으로 추출한 후, MgSO4로 처리하였다. 3) 글래스 필터로 필터링시킨 후, 증발시켰다. 4) 헥산에 용해시킨 후 원심분리기(9000 rpm, 20 분, 4℃)를 통해 불순물을 제거하였다. 5) 2K 투석 멤브레인을 D.W 투석을 3일 동안 진행한 후, 동결건조하였다. 동결건조된 생성물 구조를 1H-NMR을 통해 확인하였다:

실시예 7: 고분자 물질 및 게스트 물질의 혼합에 따른 나노구조체의 형성
실시예 2에서 최종 제조된 고분자 물질(PAA-α-CD) 0.1g을 1 (w/w)%의 농도로 4℃에서 인산완충식염수(pH 7.4)에 용해시킨 다음, 여기에 실시예 5에서 제조된 콜레스테롤 및 폴리에틸렌글리콜 결합 물질(Mw = 500, 750)(CPEG500, CPEG750) 0.0093g을 혼합한 후 소니케이션(40 KHZ, 30 분, 4℃)을 진행하여 나노구조체(PAA-α-CD-CPEG500, PAA-α-CD-CPEG750)를 형성하였다. 본 발명에 따라 형성된 나노구조체(PAA-α-CD-CPEG500, PAA-α-CD-CPEG750)를 나노입도분석기(제타사이저)로 관찰하였다(도 4).
도 4에 나타난 바와 같이, 실시예 5에서 제조된 콜레스테롤 및 폴리에틸렌글리콜 결합 물질(Mw = 500, 750)(CPEG500, CPEG750)은 양친성 물질로서, 벌키한 구조의 실시예 2에서 최종 제조된 고분자 물질과 혼합하더라도, 호스트-게스트 상호작용으로 인하여 입자크기 및 입자분포가 크게 변화하지 않는 것으로 확인된다. 따라서, 사이클로덱스트린 유래 폴리아크릴산 및 게스트 물질을 포함하는 나노구조체(PAA-α-CD-CPEG500, PAA-α-CD-CPEG750)의 평균입자크기는 약 10 nm 내지 60 nm, 특히, 약 10 nm 내지 40 nm로 균일한 입자크기 및 입자분포를 가질 수 있다.
그밖에, 사이클로덱스트린 유래 알지네이트 및 게스트 물질을 포함하는 나노구조체의 평균입자크기는 약 20 nm 내지 110 nm이고, 사이클로덱스트린 유래 카르복실메틸셀룰로오스 및 게스트 물질을 포함하는 나노구조체의 평균입자크기는 약 10 nm 내지 40 nm로 마찬가지로 균일한 입자크기 및 입자분포를 가질 수 있다.
실시예 8: 나노구조체를 이용한 독소루비신 전달체 제조
실시예 1에서 최종 제조된 고분자 물질(PAA-α-CD) 0.1g을 1 (w/w)%의 농도로 4℃에서 인산완충식염수(pH 7.4)에 용해시킨 다음, 여기에 실시예 4에서 제조된 토코페롤 및 폴리에틸렌글리콜 결합 물질(Mw = 750)(TPGS750) 0.0093g을 혼합한 후 소니케이션(40 KHZ, 1 시간, 4℃)을 진행하여 나노구조체(PAA-α-CD-TPGS750)를 형성하였다. 이후, 독소루비신(Doxorubicin; DOX) 5㎍을 추가 혼합한 후 소니케이션(40 KHZ, 1 시간, 4℃)을 추가 진행하여 독소루비신 전달체(PAA-α-CD-TPGS750 + DOX 5㎍)를 제조하였다. 본 발명에 따라 형성된 나노구조체(PAA-α-CD-TPGS750)와 독소루비신 전달체(PAA-α-CD-TPGS750 + DOX 5㎍)를 나노입도분석기(제타사이저)로 관찰하였다(도 5).
도 5에 나타난 바와 같이, 실시예 4에서 제조된 토코페롤 및 폴리에틸렌글리콜 결합 물질(Mw = 750)(TPGS750)은 양친성 물질로서, 벌키한 구조의 실시예 1에서 최종 제조된 고분자 물질과 혼합하더라도, 호스트-게스트 상호작용으로 인하여 입자크기 및 입자분포가 크게 변화하지 않는 것으로 확인된다. 따라서, 사이클로덱스트린 유래 폴리아크릴산 및 게스트 물질을 포함하는 나노구조체(PAA-α-CD-TPGS750)의 평균입자크기는 약 60 nm 내지 120 nm로 균일한 입자크기 및 입자분포를 가질 수 있다.
한편, 독소루비신 그자체는 물에 녹지 않아 침전이 생기는 문제점이 있다. 이러한 독소루비신을 나노구조체에 물리적으로 담지한 경우, 그 독소루비신 전달체(PAA-α-CD-TPGS750 + DOX 5㎍)의 평균입자크기 역시 약 50 nm 내지 250 nm로 나노구조체 단독과 유사한 것으로 확인된다. 따라서, 독소루비신이 나노구조체의 소수성 부분과 소수성 상호작용을 통해 잘 담지된 것으로 볼 수 있다.
따라서, 나노구조체 내 담지된 BCS 클래스 Ⅳ 약물의 체내 방출 속도를 효과적으로 조절할 수 있는 이점이 있다.
실시예 9: 나노구조체를 이용한 신경 성장인자 전달체 제조
실시예 1에서 최종 제조된 고분자 물질(PAA-α-CD) 0.1g을 1 (w/w)%의 농도로 4℃에서 인산완충식염수(pH 7.4)에 용해시킨 다음, 여기에 실시예 4에서 제조된 토코페롤 및 폴리에틸렌글리콜 결합 물질(Mw = 750)(TPGS750) 0.0093g을 혼합한 후 소니케이션(40 KHZ, 1 시간, 4℃)을 진행하여 나노구조체(PAA-α-CD-TPGS750)를 형성하였다. 이후, 신경 성장인자(NGF) 5ng을 추가 혼합한 후 소니케이션(40 KHZ, 1 시간, 4℃을 추가 진행하여 신경 성장인자 전달체를 제조하였다.
본 발명에 따라 형성된 나노구조체(PAA-α-CD-TPGS750)와 신경 성장인자 전달체(PAA-α-CD-TPGS750 + NGF 5ng)를 나노입도분석기(제타사이저)로 관찰하였다(도 6).
도 6에 나타난 바와 같이, 실시예 4에서 제조된 토코페롤 및 폴리에틸렌글리콜 결합 물질(Mw = 750)은 양친성 물질로서, 벌키한 구조의 실시예 1에서 최종 제조된 고분자 물질과 혼합하더라도, 호스트-게스트 상호작용으로 인하여 입자크기 및 입자분포가 크게 변화하지 않는 것으로 확인된다. 따라서, 사이클로덱스트린 유래 폴리아크릴산 및 게스트 물질을 포함하는 나노구조체(PAA-α-CD-TPGS750)의 평균입자크기는 약 108.9 nm로 균일한 입자크기 및 입자분포를 가질 수 있다.
한편, 신경 성장인자 그자체(NGF 단독)는 균일한 크기를 보이지 않는 것으로 확인된다. 이러한 신경 성장인자를 나노구조체와 화학적으로 결합한 경우, 그 신경 성장인자 전달체(PAA-α-CD-TPGS750 + NGF 5ng)의 평균입자크기는 약 97.58 nm로 나노구조체 단독(PAA-α-CD-TPGS750) 보다 오히려 크기가 감소한 것으로 확인된다. 이는 이온성 결합에 따른 결과로 볼 수 있다.
따라서, 나노구조체 내 담지된 성장인자의 체내 방출 속도를 효과적으로 조절할 수 있는 이점이 있다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예는 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
Claims (11)
- 사이클로덱스트린 유래 작용기가 화학적으로 결합된 고분자 물질; 및분자의 지름이 4.7 Å 보다 큰 제1 모이어티 및 분자의 지름이 4.7 Å 보다 작은 제2 모이어티가 서로 결합된 게스트 물질을 포함하는 나노구조체.
- 제1항에 있어서,상기 사이클로덱스트린 유래 작용기는 알파-사이클로덱스트린 유래 작용기인 것을 특징으로 하는, 나노구조체.
- 제1항에 있어서,상기 고분자 물질은 카르복실기, 하이드록시기, 아민기, 아크릴기, 티올기 및 비닐기로 이루어진 군으로부터 선택된 하나 이상의 작용기를 가지는 천연 또는 합성 고분자 물질인 것을 특징으로 하는, 나노구조체.
- 제1항에 있어서,상기 고분자 물질은 알지네이트, 셀룰로오스, 키토산, 히알루론산, 덱스트란, 폴리아크릴산, 카르복시메틸셀룰로오스 및 콘드로이틴 설페이트로 이루어진 군으로부터 선택된 하나 이상을 포함하는 것을 특징으로 하는, 나노구조체.
- 제1항에 있어서,상기 제1 모이어티는 레티놀, 베타-카로틴, 알파-카로틴, 트레티노인, 7-디히드로콜레스테롤, 프리비타민 D3, 콜레칼시페롤, 칼시디올, 칼시트리올, 칼시트로익산, 에고스테롤, 에고칼시페롤, 디히드로타취스테롤, 칼시포트리올, 타칼시톨, 디히드로에고칼시페롤, 토코페롤, 토코트리에놀, 나프토퀴논, 필로퀴논, 메나테트레논, 피토나디온, 콜레스테롤, 캄페스테롤, 시토스테롤, 스티그마스테롤 및 에르고스테롤로 이루어진 군으로부터 선택된 하나 이상의 소수성 물질 유래인 것을 특징으로 하는, 나노구조체.
- 제1항에 있어서,상기 제2 모이어티는 폴리에틸렌글리콜계 친수성 고분자 유래인 것을 특징으로 하는, 나노구조체.
- 제1항에 있어서,상기 사이클로덱스트린 유래 작용기 및 상기 게스트 물질 간에 호스트-게스트 상호작용을 이루는 것을 특징으로 하는, 나노구조체.
- 제1항에 있어서,상기 나노구조체의 평균 입자크기는 10 nm ~ 200 nm인 것을 특징으로 하는, 나노구조체.
- 제1항에 따른 나노구조체; 및상기 나노구조체 내부에 담지되거나, 상기 나노구조체와 화학적으로 결합된 생리활성물질을 포함하는 생리활성물질 전달체.
- 제9항에 있어서,상기 나노구조체 내부에 담지된 생리활성물질은 소수성 약물인 것을 특징으로 하는, 생리활성물질 전달체.
- 제9항에 있어서,상기 나노구조체와 화학적으로 결합된 생리활성물질은 양이온성 생리활성물질인 것을 특징으로 하는, 생리활성물질 전달체.
