Description

細胞剥離方法及び細胞剥離装置
 本開示は、細胞剥離方法及び細胞剥離装置に関する。
 近年、再生医療で用いられる細胞や、バイオ医薬品などの製造に用いられる細胞を培養する細胞培養が盛んに行われている。細胞培養は、接着培養と浮遊培養に大別され、そのうち接着培養は様々な細胞種に適した培養方法であり、広く用いられている。しかしながら、細胞を回収する際は、細胞を剥離するために、酵素処理を行う必要があり、処理方法によっては細胞にダメージを与える場合がある。そのため、酵素処理による細胞の剥離は細胞品質のバラツキを生じる原因ともなっている。それ故に、細胞にダメージを与えずに、高い剥離率で回収する剥離条件の検討がなされている。
 超音波や音響放射圧を利用した細胞剥離技術の場合、音響伝達媒体として一般的に水を用いられる場合が多く、この場合、水が雑菌繁殖の懸念が強く、医療現場においては特に忌避されている傾向が高い。
 特許文献1に、培養基材に音響放射圧を伝達する媒体を介して細胞を特定の位置に配列するパターニング装置が提案されている。音響伝達媒体の例示として、シリコーンゴムが記載されているが、この材料で培養基材に音響放射圧を伝達させたところ、水に比べて細胞剥離性能が十分ではなく、さらなる細胞剥離性能の向上、及びバラツキ低減が求められている。また、特許文献2に密閉体で構成される医療用超音波伝達用具が提案されている。超音波探触子と対象部位との間の音響結合に用いられているものの、この超音波伝達手段を用いて超音波振動を伝達させ、細胞を剥離する記載がない。

 本開示は上述の課題に対処するためになされたもので、対象細胞に合わせた振動設計を可能とし、高い剥離率で接着細胞を剥離可能な細胞剥離方法を提供するものである。
 本開示は、培養基材の培養面に付着した細胞を前記培養面から剥離する細胞剥離方法であって、
 前記培養基材と振動体との間に音響伝達媒体を配置した状態で、前記振動体を振動させて、前記培養面から前記細胞を剥離する細胞剥離方法、
 ただし、前記音響伝達媒体は、第一の音響伝達媒体及び第二の音響伝達媒体を含み、
 前記第一の音響伝達媒体は室温において弾性を有する固体であり、

 培養基材設置部を、この順に有し、
 前記振動体の振動が前記音響伝達媒体を経由して前記培養基材に伝達されるように構成され、

 前記第一の音響伝達媒体は室温において弾性を有する固体であり、

 本開示によれば、対象細胞に合わせた振動設計を可能とし、高い剥離率で接着細胞を剥離可能な細胞剥離方法を提供することができる。

 以下に実施例を挙げて、本開示の細胞剥離方法を説明するが、本開示は、以下の実施例により限定されるものではない。
(基本構成)
 本開示の細胞の剥離方法は、
 培養基材の培養面に付着した細胞を前記培養面から剥離する細胞剥離方法であって、
 前記培養基材と振動体との間に音響伝達媒体を配置した状態で、前記振動体を振動させて、前記培養面から前記細胞を剥離する細胞剥離方法、
 ただし、前記音響伝達媒体は、第一の音響伝達媒体及び第二の音響伝達媒体を含み、
 前記第一の音響伝達媒体は室温において弾性を有する固体であり、
 前記第二の音響伝達媒体は前記第一の音響伝達媒体に内包され、室温において液体であるか、または室温で前記第二の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体のtanδよりも大きいことを特徴とする、細胞剥離方法である。
 本開示の細胞剥離方法の概念図を図7Aから図7Dに示す。
 図7Aは音響伝達媒体13を配置する工程を示す。なお、音響伝達媒体13は、あらかじめ配置されていてもよく、本工程は本開示の細胞剥離方法に必須の工程ではない。
 図7Aでは、音響伝達媒体13を振動体101の上に配置している。図7Bでは、配置された音響伝達媒体13上に、培養基材21を配置している。図7Cは加圧工程を示し、培養基材21上から加圧している。なお、加圧工程は本開示の剥離方法に必須ではない。図7Dは、剥離工程を示す。図7Dでは、培養基材21上から加圧しながら振動体101を駆動させて超音波振動を付与して細胞を剥離している。加圧工程によって、第一の音響伝達媒体が培養基材や振動体に整合させやすくなるとともに、剥離行程においても、第一の音響伝達媒体が培養基材や振動体に整合した状態で振動を伝達することができる。ここで、第一の音響伝達媒体と、培養基材や振動体とが整合しているとは、第一の音響伝達媒体や第二の音響伝達媒体を介して、振動体の振動を培養基材に十分に伝達するように十分に接触しているが、第一の音響伝達媒体と、培養基材や振動体との間に、本発明の課題を解決できる程度のきわめて微小な隙間や、きわめて微量な物質が介在していてもよいことを意味する。例えば、第一の音響伝達媒体と培養基材との間に数分子程度の水が介在していても、第一の音響伝達媒体と培養基材とが接触しているとみなすことができる。
 本開示の細胞剥離方法において、振動体の振動を培養基材に伝達する際に、これらの間に音響伝達媒体を設ける。音響伝達媒体は、室温で、弾性を有し、かつ固体である第一の音響伝達媒体が、室温において液体であるか、または室温で第二の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体のtanδ(室温で第一の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδ)よりも大きい第二の音響伝達媒体を内包している。振動発生部である振動体は、圧電体、振動板を備え、その振動機構は、自身が伸縮する圧電体と接着された振動板との硬さの組み合わせによって振動を増幅させた屈曲振動を発生させるものであり、本開示では振動体の構造要因による共振を利用するもので、共振周波数で大きな振動を得ることができる。
(音響伝達媒体)
 本開示の細胞剥離方法において、振動を発生する振動体と振動を伝える対象の培養基材との整合に、音響伝達媒体が用いられる。整合には音響インピーダンスなどの物性的な整合の他、密着性などの機械的な整合も重要である。音響伝達媒体と、培養基材や振動体との間で、機械的な整合や物性的な整合がとれて、本発明の課題を解決できている構成となっているとよい。なお、本実施形態において音響伝達媒体は音響伝達材料と言い換えることもできる。
 振動体と培養基材との物性的な整合に関しては、「音響インピーダンスZ」と呼ばれる物理量によって規定することができる。
 具体的には、音響伝達媒体の音響インピーダンスZは、
  Z=(音響伝達媒体の密度)×(音響伝達媒体内での音速)
で定義される。
 また、2つの物質の境界に音波が到達した時、音波の透過率は、
  透過率=(4×Z1×Z2)/(Z1+Z2)^2
で定まる。なお、Z1,Z2は、音波を伝達する1つ目及び2つ目の物質の音響インピーダンスである。
 例えば、水と空気とにおける透過率、及びシリコーンゴムと空気とにおける透過率は、それぞれ、
 水の音響インピーダンスZ1=1.5×10^6[kg/m^2/s]
 空気の音響インピーダンスZ2=4.08×10^2[kg/m^2/s]
 シリコーンゴムの音響インピーダンスZ3=1.0×10^6[kg/m^2/s]
である。上述の透過率の式に水と空気のインピーダンスを代入すると、透過率はほぼゼロとなる。
 即ち、水と空気、シリコーンゴムと空気との間では、音波の透過率はほぼゼロであり、全反射することとなる。
 培養基材の材料によく用いられるポリスチレンの音響インピーダンスは、Z4=2.4×10^6[kg/m^2/s]である。したがって、本開示の音響伝達媒体における、振動体と培養基材との物性的な整合に関しては、上記の値に近い音響インピーダンスをもつ材料を第一、第二の音響伝達媒体とすることが好ましい。
 振動体と培養基材との機械的な整合に関しては、固体を介して密着している状態が好ましい。特に本開示では、振動体の振動波形を培養基材の細胞培養面に形成するため、広い面積で密着できるような機械的な整合が重要である。
 培養基材として、市販のディッシュ型の容器を用いた場合、音響伝達媒体として水や低荷重で容易に変形するグリセリンなどの低分子材料では、培養基材を振動体上に載置したとき、培養基材の突出部が直接振動体に接触し、振動伝達経路が複数存在することになり、発生した振動が複雑化し、制御が困難となる場合がある。本開示の細胞剥離方法は、主体的な振動の制御を行う。すなわち、振動体と培養基材とを固体で密着させることで振動伝達経路を一定に保ち、振動制御が容易にする。
 本開示の音響伝達媒体で振動体と培養基材とを機械的に整合する場合、固体材料としては高分子材料であるゴムやゲルのように弾性を有する材料であることが好ましく、具体的には、シリコーン樹脂、ウレタン樹脂、ジアリル二レート樹脂、不飽和ポリエステル、エポキシ樹脂、フェノール樹脂、ユリア樹脂、メラニン樹脂、天然ゴムなどが挙げられる。これらの中でも、駆動周波数(振動周波数)の周波数帯域での減衰の少ない物性であることで伝達効率が高く、さらに音響伝達媒体自身の発熱が抑制できることから、シリコーンゴムが好ましい。
(第一の音響伝達媒体)
 第一の音響伝達媒体は室温において弾性を有する固体材料からなる。なお、本明細書中で、室温とは、20℃以上30℃以下であり、好ましくは25℃である。
 弾性を有する固体材料の例としては高分子材料であるゴムやゲルを挙げることができる。
 高分子のゴム材料としては、例えばシリコーン樹脂、ウレタン樹脂、ジアリル二レート樹脂、不飽和ポリエステル、エポキシ樹脂、フェノール樹脂、ユリア樹脂、及びメラニン樹脂で構成される群から選択される少なくとも一種の樹脂を含むゴム、天然ゴム、などが挙げられる。より具体的には、シリコーンゴム、ウレタンゴム、ブチルゴム、ニトリルゴム、ブタジエンゴム、ラテックスゴムが挙げられる。
 高分子のゲル材料としてはハイドロゲルや高分子ゲルが挙げられる。ハイドロゲルとして、例えばポリビニルアルコールやポリアクリル酸などの親水性高分子材料とこれらに対応する架橋剤とを組み合わせて得られるハイドロゲルが挙げられる。架橋剤として例えば、無機系の架橋剤であるホウ砂や層状粘土鉱物(クレイ)などや、有機系の架橋剤である樹状デンドリティック高分子などの多官能の高分子材料などが挙げられる。高分子ゲルとして例えば、アクリル樹脂やウレタン樹脂などの高分子材料と、これらに対応する架橋剤である多官能化合物とを組み合わせて得られる高分子ゲルなどが挙げられる。
 第一の音響伝達媒体は、周波数1Hzの振動を付与した場合であって、室温における動的粘弾性測定で得られるtanδが1.0未満であるものを用いることができる。また、第二の音響伝達媒体が、室温で固体である場合、周波数0.1Hzの振動を付与した場合であって、室温における動的粘弾性測定で得られる第一音響伝達媒体及び第二音響伝達媒体のtanδをそれぞれtanδi、tanδiiとしたとき、tanδi<tanδiiを満たす、及び/またはtanδi≧1であることが好ましい。
 第一の音響伝達媒体は、シリコーンゴム、天然ゴム、ウレタンゲル、及びゲル材料からなる群から選択される少なくとも一種を含むことが好ましく、中でも、シリコーンゴムを含むことは好ましい。シリコーンゴムは、駆動周波数の周波数帯域での減衰の少ない物性であることで伝達効率が高く、さらに音響伝達媒体自身の発熱が抑制できる。
 また、取り外しを容易にする目的で、第一の音響伝達媒体の培養基材あるいは振動体に対向する面に例えばフッ素コートのような表面処理を施しても構わない。
 また、音響伝達媒体は、細胞剥離状況を直接観察するためには、可視光の波長領域の光に対して透明であることが好ましい。
 第一の音響伝達媒体には、粘度、第二の音響伝達媒体との親和性、第二の音響伝達媒体との音響インピーダンス、保存安定性、抗菌作用などの物性値を制御する目的で各種添加剤を加えてもよい。なお、第二の音響伝達媒体との親和性とは、例えば、第一の音響伝達媒体と第二の音響伝達媒体に関する接触角や濡れ性のような特性をいう。
 第一の音響伝達媒体の厚みについては、厚すぎると超音波振動伝達において界面で超音波を反射しやすくなり、また、振動減衰が無視できなくなる。薄すぎると第二の音響伝達媒体を内包する膜としての耐久性が悪くなり、また、破れたり、穴が開いて内包物が外に出てしまう。従って第一の音響伝達媒体の厚みの合計としては振動体を振動させる際に5mm以下であることが好ましい。
(第二の音響伝達媒体)
 第二の音響伝達媒体は室温において液体であるか、または室温で前記第二の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体よりも大きい。
 室温で液体であるとは、20℃以上30℃以下で液体であることを指し、特に、室温25℃における固有粘度[η]または周波数1Hzの振動が付与された場合であって、室温25℃における動的粘弾性測定で得られる複素粘度η*、またはB型粘度計やレオメーターによってせん断速度を変化させることで測定される粘度ηが小さいものとすることができる。
 また、室温で固体である場合、室温で周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体よりも大きい。好ましくは、周波数0.1Hz振動が付与された場合であって、室温における動的粘弾性測定で得られるtanδが1以上となる固体である。すなわち、第二の音響伝達媒体が固体である場合、周波数0.1Hzの振動が付与された場合であって、室温における動的粘弾性測定で得られる第一音響伝達媒体及び第二音響伝達媒体のtanδをそれぞれtanδi、tanδiiとしたとき、tanδi<tanδiiを満たす、及び/またはtanδi≧1であることが好ましい。
 第二の音響伝達媒体となり得る固体として、可撓性のあるゲルを挙げられる。可撓性のあるゲルは、第一の音響伝達媒体として例示したゲルと同じく、例えばポリビニルアルコールやポリアクリル酸などの親水性高分子材料と、架橋成分であるホウ砂や層状粘土鉱物(クレイ)、樹状デンドリティック高分子などの多分岐高分子材料などを組み合わせたハイドロゲルや、アクリル樹脂やウレタン樹脂などの高分子材料と対応する架橋剤である多官能化合物とを組み合わせた高分子ゲル、などが挙げられる。
 tanδは、音響伝達媒体で振動体と培養基材とを機械的に整合する際の温度、時間スケールにおける値を使用することが好ましい。すなわち、tanδの測定は、室温、より好ましくは25℃で、周波数1(Hz)あるいは0.1(Hz)の振動を付与したときの動的粘弾性G’及びG”をレオメーターなどで測定し、tanδ=G”/G’の関係式を用いて計算することでtanδを算出することができる。ここでGはせん断弾性率を表し、G’は貯蔵剪断弾性率、G”は損失剪断弾性率を表す。ただし、第二の音響伝達媒体について、tanδが測定される場合に以上を満たせばよく、第二の音響伝達媒体については、tanδが測定できない場合もある。第二の音響伝達媒体は室温で液体であってもよい。この場合tanδは測定できない場合もある。
 第二の音響伝達媒体が液体である場合、音響伝達を損なわない程度に粘度が高くても構わない。
 粘度は、室温、より好ましくは25℃でB型粘度計やレオメーターで粘度測定することができる。さらには、25mmのパラレルプレートを使用し、ギャップ1.0mmで測定する場合のせん断速度が1.0(1/s)における粘度の値を採用することが好ましい。液体の粘度によっては前記条件では適切なトルク値が得られない場合があるが、その際は同程度のせん断速度となるよう、プレートの径やギャップなどの値を変更することで測定することが可能である。
 液体としては水、生理食塩水、エタノール、希釈エタノールなどの低粘度液体や、これらの混合物、さらにグリセリンやシリコーンオイル、高分子溶液などの高粘性液体やゲル材料、さらにはこれらの混合物などが挙げられる。ゲルを水などの液体に分散、膨潤させたポリマー溶液などでもよく、より具体的には、グリセリンなどの液体にPVAゲルなどの増粘剤などの添加剤を添加して粘弾性を調整した超音波ジェルも好ましい。なお、ゲル材料は固体と分類される場合と液体と分類される場合がある。固体としてのゲルは架橋成分の含有比率が高く、液体としてのゲルは架橋成分の含有比率が低い。本明細書においては、発明者らが行った測定方法においてtanδが測定可能であったものを固体、そうでないものを液体としている。これは、弾性体としての反発と関係していると考えられる。以下の実施例においては、PVAは固体としての可撓性のあるゲルであり、超音波ジェルは液体としてのゲルである。
 第二の音響伝達媒体を内包している第一の音響伝達媒体に傷がついた際に中身が漏れ出ることを防ぐために、第二の音響伝達媒体は、室温で周波数0.1Hzの振動が付与された場合におけるtanδが1.0以上の固体であることが好ましい。
 第二の音響伝達媒体は、粘度や第一の音響伝達媒体との親和性や音響インピーダンス、表面張力や保存安定性、抗菌作用などの物性値を制御する目的で各種添加剤を含んでもよい。
 添加剤の例として、以下を挙げることができる。
 表面張力の調整のための添加剤として、アニオン性界面活性剤として、ドデシル硫酸ナトリウム、ラウリル硫酸ナトリウムなどを挙げられる。
 カチオン性界面活性剤として、エチルトリメチルアンモニウムブロマイド、ベンザルコニウム塩化物、ステアリン酸ジメチルアミノプロピルアミドなどを挙げられる。
 非イオン性界面活性剤として、コンタミノン(登録商標)、コンタミノン(登録商標)US、プルロニック(登録商標)F-68、Tween(登録商標)20、Tween(登録商標)80などを挙げられる。
 両性界面活性剤として、CHAPS、CHAPSOなどを挙げられる。
 保存安定剤として、ヒンダードアミン、ベンゾフェノン、ベンゾトリアゾールなどの光安定剤、アミン系、フェノール系、硫黄系、リン系などの酸化防止剤を挙げられる。
 抗菌作用を示すものとして、ベンザルコニウム塩化物、アジ化ナトリウム、及びアンピシリン、ペニシリン、ストレプトマイシンなどの抗生物質、などが挙げられる。
(音響伝達媒体の内包)
 本開示の細胞剥離方法に用いられる音響伝達媒体は、第一の音響伝達媒体によって第二の音響伝達媒体が内包された構成となる。
 内包とは振動体と培養基材との間で第一の音響伝達媒体の内側に第二の音響伝達媒体が存在していればよい。例えば袋状の第一の音響伝達媒体の中に第二の音響伝達媒体が密閉された形態や、振動伝達を損なわないように、袋状の第一の音響伝達媒体に循環用の管がついていて、その中を通して第二の音響伝達媒体を供給し、循環できるような形態であってもよい。第一の音響伝達媒体の中で第二の音響伝達媒体が循環できるような形態である場合、後述の配置工程で第二の音響伝達媒体を第一の音響伝達媒体に内包させたのち、加圧、剥離工程においては弁などで循環しないような状態にすることが好ましい。
 内包させる方法については、例えば、袋状の第一の音響伝達媒体を作製しておき、中に第二の音響伝達媒体を注入後、注入口を封じる方法が挙げられる。封じる方法としては、熱で溶融させて封じる方法、圧力で圧着する方法、接着剤や第一の音響伝達媒体の原材料化合物などで反応させて封じる方法、物理的にクランプして封じる方法などが挙げられる。
 その他の内包する方法として、上面、下面の第一の音響伝達媒体のシートを作製しておき、第二の音響伝達媒体を内包させながら上下面を密着させて封じる方法が挙げられる。本開示の細胞剥離方法は、内包工程として、第二の音響伝達媒体を第一の音響伝達媒体に内包させる内包工程を含むことができる。
 第一の音響伝達媒体は第二の音響伝達媒体と対向する方向に凸となる突起部を少なくとも1つ有することができる。突起部は加圧工程で加圧した際に、向かい合う反対側の面にある第一の音響伝達媒体と物理的に密着する形状であることが好ましく、例えば円柱状、半球状、円錐状、円柱または円錐と半球を組み合わせた構造、などとすることができる。突起部は第一の音響伝達媒体と同じ材料であることが好ましい。
 突起部の位置については付与する超音波振動パターンの振動の強い腹部(振幅値の大きい領域)に相当する位置が好ましいが、振動の強くない部分や、振動しない部分を含んでいてもよい。突起部の数についても、少なくとも一つであれば良く、例えば、複数の振動パターンに対応するため、各振動パターンの腹部に相当する位置に複数個の突起部が配置されていてもよい。
 第一の音響伝達媒体は前記第二の音響伝達媒体を横切ってつながっている柱部を少なくとも1つ有することができる。
 本開示の音響伝達媒体のうち、振動体と対向する面は培養基材と対向する面よりもヤング率が高いことができる。ヤング率が高いということは、弾性率が高いことと同義であり、例えば、第一の音響伝達媒体の厚みが大きい、または第一の音響伝達媒体の製造時の分岐成分量を多くすることなどで、ヤング率を高くすることができる。
 第一の音響伝達媒体と第二の音響伝達媒体の音響インピーダンス差が少ない方が好ましい。また、音響伝達媒体は空気を内包しないことが好ましく、したがって、第二の音響伝達媒体を第一の音響伝達媒体に内包させる時は空気が含まれないように内包することが好ましい。
 本開示の音響伝達媒体の厚みは振動減衰を低減し、また、界面で超音波を反射しないようにするという観点から、10mm以下であることが好ましく、振動体に振動を発生させる際には5mm以下であることが好ましい。
 また、音響伝達媒体のアスペクト比(厚み/対角線の大きさ)は0.3以下であることが好ましい。0.3以下となることで設置時の安定性が高まる。
 本開示の音響伝達媒体の振動体と培養基材と対向する面に、整合性を高めるために、水などの液体を付与してから使用してもよい。液体としては好ましくは表面張力の低い液体である。
 本開示の音響伝達媒体のうち、振動体と対向する面には整合性を高めるために、接着剤や粘着剤などを用いて振動体と全面、または部分的に接着させてもよい。接着剤や粘着剤については公知の接着剤や粘着剤、両面テープなどが用いられる。
(培養基材)
 本開示の細胞剥離方法は培養基材の培養面に付着した細胞を培養基材から剥離するものである。細胞が付着するとは、接着性の細胞が接着することのみでなく、細胞が接触して存在することを含む。すなわち、細胞が培養基材に付着するとは、接着性の細胞が接着することのみでなく、細胞が接触して存在することを含む。すなわち、細胞が培養基材に細胞が接着因子を介して接着することを含むが、接着に限定されず、細胞が培養基材との弱い相互作用によって留まっていること、あるいは、細胞が重力などの外力によって単に留まっていることを含む。
 本開示の培養基材とは、細胞培養に用いる基材であり、細胞が付着する面を含むあらゆる基材を指す。培養基材は例えば、細胞培養用途の容器あるいはその一部を含む。細胞容器の例として、ディッシュ(シャーレともいう)、フラスコ、プレート、チューブなどを挙げられる。これらはいずれも、例えばコーニング社、サーモフィッシャーサイエンティフィック社、AGCテクノガラス社などの製品が汎用のディスポーザブル容器として用いられる。中でも本開示の培養基材の例として、ディッシュは典型的である。培養基材は取り外し可能な蓋がついているものが汎用される。本明細書中で培養基材は、蓋のような部品を含んでもよいし、含まなくてもよい。ディッシュの形状の例は図8の概略図の通りである。この中で特に底面反り高さ300はメーカーごとの型番違いや、ロット違い、さらにはロット内でもバラツキが大きい。培養基材を音響伝達媒体と機械的に整合するためには底面反り高さのバラツキを考慮する必要があるが、本開示の第一の音響伝達媒体と第二の音響伝達媒体とを併用することで機械的な整合が容易となる。
 図8の概略図のように市販の培養基材底面には、外周部に突出部がある。突出部の主な役割は、基材底面を傷つけないこと、容器同士を重ねることを可能とすること、作業台上に置いた際、直接基材底面に作業台の温度が伝わらないことなどが考えられる。培養基材と音響伝達媒体とを機械的に整合する際には、この突出部を避けて整合させてもよいし、突出部を覆う形で整合させてもよい。
 基材の材質は、化学的に安定であり、所望の細胞を培養可能な材質であればよく、例えば、ポリエチレン、ポリプロピレン、ポリカーボネート、ポリスチレン、ポリ塩化ビニル、ナイロン、ポリウレタン、ポリウレア、ポリ乳酸、ポリグリコール酸、ポリビニルアルコール、ポリ酢酸ビニル、ポリ(メタ)アクリル酸、ポリ(メタ)アクリル酸誘導体、ポリアクリロニトリル、ポリ(メタ)アクリルアミド、ポリ(メタ)アクリルアミド誘導体、ポリスルホン、セルロース、セルロース誘導体、ポリシリコーン、ポリメチルペンテン、ガラス、金属などが挙げられる。この中でもポリスチレンが好ましい。
(振動体)
 本開示の振動体は振動を発生するものであれば、限定されることなく用いられるが、例えば圧電体と振動板とを接着したものが挙げられる。圧電体が円形型の場合は振動板はガラス、SUS、石英であることが好ましい。振動板をガラス、SUS、石英にすることで、振動体として破損することなく超音波領域の比較的高い駆動周波数(振動周波数)で大きい振幅を出力することができる。振動体は超音波振動子を含むことができる。
 リング形状の圧電体の場合、振動板の外径寸法は圧電体の外径寸法と等しいことが好ましい。
 振動板の厚みは、圧電体と振動板とが接着され、撓み振動した際に撓みの厚さ方向の中点、つまり撓みの際、引っ張り、圧縮のどちらもならない中立面が振動板側にあることが、圧電体の歪を効率良く撓みに利用するために好ましい。
(圧電体)
 本開示の振動体に用いられる圧電体はリング形状の圧電体が好ましいが、振動体を撓ませる圧電体の形状は、円環形状、円板形状であっても構わない。細胞剥離状況を直接観察するためには、培養基材下部の視野を確保できる円環形状が好ましい。
 圧電体は、分極処理を施すことで圧電特性を発揮するが、図10の通り、電極パターンに応じて分極の極性を変えて構わない。分極処理した圧電体は、入力した交流電圧の位相を電極パターン毎に制御することで、定在波モード、進行波モードの振動モードを励振することができる。プラスに分極した電極に対して、マイナスに分極した電極に180度の位相差を付けた交流電圧を印加することで定在波が発生する。これに対し、図10の様に、左右にA相、B相と電極を分けたとき、A相とB相の位相差を90度にした交流電圧を印加すると、2相駆動の進行波が発生する。図10でGNDはグランドを指し、裏面に配置された電極の接地に用いる。
 電極パターンは、Agを印刷法で形成する例を挙げられるが、電極材料にはAgの他、Au、Pt、Pdなどの貴金属やCuなどの卑金属でも構わない。形成方法は印刷法、メッキ法、スパッタ法であっても構わない。
 振動体に用いた圧電体の材料組成はPbZrTiO(PZT)を用いたが、環境規制を考慮して実質的にPbを用いない非鉛圧電材料であることが好ましい。非鉛圧電材料には、主成分がBaTiO(BT)、NaNbO、BiNaTiO、BiFeOなどが挙げられ、それぞれの組み合わせや、主成分に金属元素が添加されていても構わない。特に、BaTiO(BT)系材料で、PbZrTiO(PZT)と同等の振動性能を確認した。また、セラミックス以外の材料として、単結晶材料、高分子系圧電材料でも構わない。
 振動体の圧電体の厚みは、接着し、積層された圧電体と振動板が、撓み振動した際に、撓みの厚さ方向の中点、つまり、撓みの際、引っ張り、圧縮のどちらともならない中立面が振動板側にあることが、圧電体素子の歪を効率良く撓みに利用するために好ましいため、圧電体と振動板の厚さの組み合わせは、圧電体の硬さと振動板の硬さの関係で算出した。
(工程)
 本開示の細胞剥離方法は以下の工程を有することができる。
配置工程:
 振動体上に音響伝達媒体を設置し、必要に応じて、第二の音響伝達媒体が第一の音響伝達媒体に内包されるよう供給し、さらに培養基材を設置する工程
加圧工程:
 培養基材及び振動体が音響伝達媒体に接するように、培養基材または振動体側から加圧する工程
剥離工程:
 培養基材と振動体が音響伝達媒体に接している状態で振動体を振動させる工程。
 本開示の細胞剥離方法の具体例の一形態を図5に示す。
 まず、配置工程として第一の音響伝達媒体を振動体上に設置する(ステップS210)。
 その後、第二の音響伝達媒体が第一の音響伝達媒体に内包されるよう供給する(ステップS220)。
 さらにその後、音響伝達媒体上に培養基材を設置する(ステップS230)。
 次いで、加圧工程として、培養基材上面より重りなどで加圧する(ステップS240)。
 剥離工程として、超音波付与条件を設定する(ステップS250)。
 次いで、超音波付与を開始する(ステップS260)。
 最後に、剥離された細胞を回収する(ステップS270)。
 なお、第二の音響伝達媒体が第一の音響伝達媒体に内包されるよう供給すること、すなわち、内包工程は本開示の細胞剥離方法に必須の工程ではなく、供給は別途行われていてもよく、あるいは、あらかじめ、第二の音響伝達媒体が第一の音響伝達媒体に内包された音響伝達媒体を用いてもよい。また、加圧すること、すなわち加圧工程も本開示の細胞剥離方法に必須の工程ではない。
(配置工程)
 配置工程では、振動体の上に音響伝達媒体を設置し、必要に応じて、第二の音響伝達媒体が第一の音響伝達媒体に内包されるよう供給し、培養基材を設置する工程となる。
 音響伝達媒体が第二の音響伝達媒体を供給して循環できる構造の場合、本工程において供給、循環させておき、この後の加圧工程、剥離工程の際に弁を閉じて供給、循環をストップさせる。供給、循環させることで音響伝達媒体中の第二の音響伝達媒体の量を制御でき、培養基材の種類や形状に応じて適切に接触させることができ、さらに循環させることで音響伝達媒体の温度も制御可能となる。
(加圧工程)
 本実施形態における加圧工程は、培養基材と振動体とが近づくように加圧する工程であればよく、培養基材を変位させて振動体に近づけても、振動体を変位させて培養基材に近づけても良く、培養基材と振動体の両方を変位させて両者が近づくようにしても良い。
 加圧工程では、培養基材と振動体が音響伝達媒体に接触した状態で、培養基材と音響伝達媒体とを機械的に整合するように、培養基材側または振動体側から加圧する。
 加圧工程を実行するための加圧手段としては、例えば培養基材の蓋上に重りを設置する、培養基材の容器の蓋上からバネや板バネなどといった荷重方法で均一に上からの荷重をかけて培養基材の蓋上部を上部から動かないよう固定しておき、振動体ごと下から上方向への荷重をかける、などの方法が挙げられる。
 加圧手段については振動の絶縁性を考慮すると重りによる荷重が好ましいが、振動を阻害しない程度に他の荷重方法を選択することができる。加圧手段として例えば定荷重ばねを用いることができる。
 培養基材の蓋上に設置する重りの形状としては、例えば容器内の細胞を観察しやすいように円環形状の重りであることが好ましい。
 加える荷重としては、培養基材の大きさや、付与する振動パターンによって変化するが、例えば35mmディッシュの場合、10gから300gの間であることが好ましく、60mmディッシュの場合、100gから600gであることが好ましい。荷重は重りを使用しても良いし、培養容器自体の重量としても良い。
 重りが重過ぎると振動そのものを押さえつけ、阻害することの他に、培養基材自体が撓んでしまうことで振動伝達状態が不均一になり、剥離率低下の要因となる。
(剥離工程)
 剥離工程においては、培養基材または振動体が音響伝達媒体に接している状態で振動体を振動させて、細胞を剥離する。さらに、剥離工程は振動体に超音波帯域の振動を励起させる工程を含むことができる。
 配置工程においては、振動体の振動は開始しておらず、剥離工程で振動体の振動を開始する。例えば、図6に示す装置の例においては、共振周波数の電圧を供給することで、振動体を駆動させて所望の超音波振動モードを発生させる。この場合、剥離工程において、振動体に共振周波数の電圧を供給することで、振動体を駆動させて所望の超音波振動モードを発生させる。なお、上記電圧を付与する電源は図6において不図示である。
 振動パターンは、細胞種や剥離結果に応じて自由に選択することができる。また、剥離工程においてこれらの振動パターンを単一で用いてもよいし、周期的に変化させて入力させてもよい。
 振動波形は培養基材の細胞培養面を一定の振幅、例えば1μmの振幅で振動することができるものが好ましい。基本的に、低周波から高周波まで、例えば超音波領域では20kHzから150kHzまで周波数掃引した際に、共振する振動が検出できるが、低周波側で検出した振動は振幅が大きいが加速度は小さい。これに対し、高周波側で検出した振動は、振幅が小さいが加速度は大きい。このことから、培養基材に対する培養細胞の接着力を弱める力を付与する条件は、細胞の接着力、細胞のサイズや形状によるところが大きい。さらに、剥離液に生じる流動についても振幅、加速度によって異なるため、様々な組み合わせが存在する。また、単純に大きな振幅、大きな加速度では、細胞の生存率を低下の要因となるキャビテーションの発生を促進することから好ましくないことがある。
 振動波形は、円板状の振動板の中心から同心円状、または円周方向に次数を持つようなパターンとして形成される定在波モードと、圧電体を径方向に位相を変えて振動させることで、振動板上に径方向の振動波形を形成する進行波モードとすることができる。それぞれ振動体に共振周波数で電圧を印加することで振動波形が形成でき、その共振周波数、振動波形形状は振動体の形状と硬さ、密度などの材料物性値を用いた構造計算で算出できる。
 また、振動体を連続駆動すると圧電体自身の発熱から共振周波数が低周波側にシフトするため、周波数を固定して駆動し続けると振幅が小さくなる。このため、一定時間振幅させた後、休止時間を設け、自己発熱による温度上昇を抑制する駆動方法か、あるいは、一定の周波数幅で高周波側から低周波側へ掃引を繰り返し、たとえ発熱しても必ず共振周波数を経る駆動方法を用いて駆動させることで十分な振幅を得ることができる。
(振動評価)
 振動板の振動波形はレーザードップラー振動計(グラフテック社AT7200)で計測することができる。図6に示す細胞剥離装置を例に、計測手法を説明する。所望の共振周波数、電圧で細胞剥離装置を駆動し、振動板11の表面をレーザーでXY方向の2次元的にスキャンすると、各点の振動速度が得られ、計算よりZ方向の振幅を算出することができる。この結果、振動板11の振動波形を3次元的に構成することができ、駆動時の振動の振幅の大きい腹と振動しない節の位置を動的に把握することができる。
 培養基材21の細胞培養面の振動計測も同様の構成で計測することができる。振動板11上に音響伝達媒体13を設置し、培養基材21をレーザーが反射するように金属、例えばAgを蒸着法で培養基材21内面に成膜し、培養基材21と音響伝達媒体13との間に空気層が入らないようにし、設置することが好ましい。なお、図6では、第一の音響伝達媒体および第二の音響伝達媒体をまとめて音響伝達媒体13として示している。
 音響伝達媒体13上に設置した培養基材21を細胞剥離装置で、駆動させる目的の共振が同心円に振幅する定在波モードの場合、培養基材21の中央部の振幅を目的の共振周波数付近で掃引し、その時、最も振幅の大きいときの周波数で培養基材21の細胞培養面全体を2次元的にスキャンすることで、培養基材21の細胞培養面に発生した振動波形を3次元的に把握することができる。
(細胞)
 本開示の細胞剥離方法は、限定なくあらゆる種類の細胞に適用することができる。例として、チャイニーズハムスター卵巣由来CHO細胞やマウス結合組織L929細胞、マウス骨格筋筋芽細胞(C2C12細胞)、ヒト胎児肺由来正常二倍体線維芽細胞(TIG-3細胞)、ヒト胎児腎臓由来細胞(HEK293細胞)、ヒト肺胞基底上皮腺癌由来A549細胞やヒト子宮頸癌由来HeLa細胞、昆虫由来の細胞株、などの種々の培養細胞株、生体内の各組織、臓器を構成する上皮細胞や内皮細胞、収縮性を示す骨格筋細胞、平滑筋細胞、心筋細胞、神経系を構成するニューロン細胞、グリア細胞、繊維芽細胞、生体の代謝に関与する肝実質細胞、肝非実質細胞や脂肪細胞などの初代培養細胞、誘導多能性幹(iPS)細胞、胚性幹(ES)細胞、胚性生殖(EG)細胞、胚性癌(EC)細胞、間葉系幹細胞、肝幹細胞、膵幹細胞、皮膚幹細胞、筋幹細胞、生殖幹細胞などの各種幹細胞、または各組織の前駆細胞、さらにはそれらから分化誘導した細胞などが挙げられる。
 細胞の培養条件は、培養する細胞に従って適宜選択し得る。一般的には、培養基材21内に適切な培地を添加し、そこに1.0×10cells/cm~5.0×10cells/cm程度の細胞を播種し、37℃、CO濃度5%の環境下で培養する。この際、培養基材21中の細胞占有面積率が7割~8割程度、いわゆるサブコンフルエントの状態になるまで培養するのが好ましい。
 培養した細胞の培養液(培地)は、細胞剥離工程では細胞剥離液に置換することができる。
 一般的には、剥離液には、タンパク質分解酵素を含有している。タンパク質分解酵素には、例えばトリプシン、アキュターゼ、コラゲナーゼ、天然プロテアーゼ、キモトリプシン、エラスターゼ、パパイン、プロナーゼ、もしくはそれらの組み換え体などが挙げられる。しかし、タンパク質分解酵素は、細胞を溶かしてしまうことから細胞への悪影響が懸念されるものである。これに対し、本開示の細胞剥離手法では、タンパク質分解酵素を用いない、用いても極少量で同様の効果を発揮することから細胞への悪影響を抑制する効果がある。
 細胞剥離液は、細胞の剥離時に用いる液であり、細胞の接着力を低下させる作用があってもよい。
 細胞剥離液のpHは中性領域であることが好ましい。中性領域は細胞培養に適しており、細胞の生存率を安定的に高く保つことができるためである。pHは塩酸や水酸化ナトリウム等で適宜調整可能である。また、pHを安定的に保つために各種緩衝液が好適に用いられる。また、剥離液は培地であっても構わない。
 細胞剥離液の粘度はポリマーや糖類の添加などによって適宜調整可能である。
 細胞剥離液として、金属イオンキレート剤(以後、キレート剤と呼ぶことがある)を含む溶液が、特に好ましい。キレート剤を含む細胞剥離液を用いることによって、細胞の接着力を効果的に低下させることが出来るからである。
 キレート剤としては、特に限定はないが、例えば、エチレンジアミン四酢酸(以後、EDTAと呼ぶことがある)、エチレンジアミン、エチレンジアミンテトラメチレンホスホン酸、グリコールエーテルジアミン四酢酸、ニトリロトリ酢酸、ジエチレントリアミン五酢酸、イミノ二酢酸、ジヒドロキシエチルグリシン、ジカルボキシメチルグルタミン酸、エチレンジアミンジコハク酸、エチドロン酸、クエン酸、グルコン酸、ホスホノブタン三酢酸等を挙げることができる。この中でも二価の陽イオンとキレートを形成するキレート剤が好ましく、Ca2+及びMg2+とキレートを形成するキレート剤が特に好ましく、エチレンジアミン四酢酸が最も好ましい。キレート剤としてエチレンジアミン四酢酸を用いた場合、細胞剥離液のpHは7.0以上8.0以下であることが好ましい。これは細胞生存率を高く保つことができる中性領域の中でも高めのpHであることにより、エチレンジアミン四酢酸のキレート能を高めることができ、細胞の接着力の低下効率をより高くすることが可能であるためである。なおキレート剤は単独で用いても、或いは2種類以上を複合して用いてもよい。
 キレート剤の含有量としては0.01mM以上5.0mM以下であることが好ましい。
 この範囲であることにより、キレート効果を確実に得ることができ、過剰なキレート剤の存在による活性低下も抑えることができる。
 細胞剥離液として、ポリアルキレングリコール構造を含む親水性ポリマーを含んでいてもよい。ポリアルキレングリコール構造を含む親水性ポリマーの例として、ポリエチレングリコールがあげられる。親水性ポリマーは、ゲルパーミエーションクロマトグラフィーにより測定されるピーク分子量Mpが800以上50000以下であることが好ましく、1200以上20000以下であることがさらに好ましい。これは、ポリマーの細胞への影響が少なく、かつ、ポリマーによる増粘効果が抑制できるためである。
(剥離試験)
 本開示の細胞剥離方法を評価するために剥離試験を行うことができる。剥離試験は例えば次のように行うことができる。
 細胞を培養し、必要に応じ、培養液(培地)を剥離液に交換した基材に対し、配置工程を行い、必要に応じ、保温機内で一定温度、例えば25℃で保持した状態で振動体を駆動することもできる。駆動時間は細胞剥離に十分な時間で駆動する。駆動後、スポイトで剥離した細胞を回収し、回収できず培養基材21にまだ貼り付いている細胞をピペッティング法で回収する。
 以下で用いた剥離率は、振動による剥離細胞数とピペッティング法による剥離細胞数との合算数に対する、振動による剥離細胞数の割合を%表記したものである。バラツキはn=10で剥離率を求め、剥離率の標準偏差を5%未満がA、5%以上7.5%未満がB、7.5%以上10%未満がC、10%以上がD評価とした。
 細胞剥離後の基材の底面に残った残細胞をバックライトによる散乱光を用いて観察することができる。
(細胞剥離装置)
 本開示の細胞剥離装置は培養基材の培養面に設けられた細胞に振動体が発生した振動を伝達することで、前記細胞を剥離する細胞剥離装置であって、振動体、音響伝達媒体、及び培養基材設置部を、この順に有し、前記振動体の振動が前記音響伝達媒体を経由して前記培養基材に伝達されるように構成され、前記音響伝達媒体は、第一の音響伝達媒体及び第二の音響伝達媒体を含み、前記第一の音響伝達媒体は室温において弾性を有する固体であり、前記第二の音響伝達媒体は前記第一の音響伝達媒体に内包され、室温において液体であるか、または室温で前記第二の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体よりも大きいことを特徴とする細胞剥離装置である。
 本開示の装置の構成を図6に示す。
 図6に示すように、本開示の細胞剥離装置は、振動体101、音響伝達媒体13を含む。培養基材設置部は、使用者が培養容器を置くことが認識できればよく、図6の例では、振動体101と上カバー15、緩衝材16で形成される開口が、汎用されるディッシュを設置するのに適しているため、この開口が培養基材設置部に相当する。
 図6の細胞剥離装置には、振動発生部である振動体101として、圧電体12、振動板11を備える例を示す。圧電体12、振動板11を備える振動体101は、自身が伸縮する圧電体12と、接着された振動板11との硬さの組み合わせによって振動を増幅させた屈曲振動を発生する。この例では振動体101の構造要因による共振を利用するもので、共振周波数で大きな振動を得ることができる。振動体は超音波振動子などの振動子を含むことができる。なお、圧電体12に不図示の電源を用いて電圧を印加することで、上記伸縮をさせる。
 図6では、代表的な振動体101として、円環型振動子を記しているが、大振幅が得られるランジュバン振動子、矩形型振動子であっても構わない。
 共振によって振動板に形成した振動を、細胞を培養させた培養基材21に効率的に伝達するために、音響伝達媒体13を振動板11と培養基材21の間に介在させる。直接、振動板11に培養基材21を設置すると、接触部が点接触となり、鳴き、発熱の発生など振動以外のエネルギーとして消費されるため効率悪化の要因となる。音響伝達媒体13の設置は、基本的には培養基材21に付着した細胞の、剥離する面積を十分に満たす面積で設置されていることが好ましい。
 振動体101を固定するために、図6の上カバー15、下カバー17、緩衝材16、ボルト18によって、振動体101の端部を挟み込む構造をとるが、端部の挟み込む圧力は振動板に発生する振動を阻害しない程度に締め付ける。
 培養基材の上部には振動体の下部には加圧工程用の加圧手段である重り14を備えた例を示す。重り14が上下することで加圧、除圧が可能となる。
 共振周波数の電圧を供給することで、振動体を駆動させて所望の超音波振動モードを発生させる。
 振動体の駆動には、共振周波数の電圧を供給する交流電源を用いて、培養基材の細胞培養面に所望の振動波形が形成できる振動モードの周波数で、かつ、所望の振幅が得られる電圧を印可することで細胞剥離装置を駆動させることができる。この時、振動体に備えるセンサ相であるS相の電圧をオシロスコープで計測すると、振動体自身が歪むことで発生させた電圧を直接モニタすることができ、振動体の振動状態を確認しながら駆動することができる。また、細胞剥離工程中の温度を計測できる熱電対を振動に影響ない箇所に設置することで、モニタしながら駆動させることができる。温度測定には赤外線温度計測器であっても構わない。
 本開示の細胞剥離方法に用いられる音響伝達媒体の例を図1Aから図1C、図2Aから図2C、図3Aから図3C、図4Aから図4C、及び図9Aから図9Dに示す。
(第一例)
 音響伝達媒体の第一例の構成図を図1Aから図1C、及び図9Aに示す。
 音響伝達媒体の第一例は、第二の音響伝達媒体が、第一の音響伝達媒体に内包されている。
 図1Aは、底面が平面の培養基材及び振動体と接している構成を示す。
 図1Bは、底面が反った形状の培養基材及び振動体と接している構成を示す。
 図1Cは、さらに、培養基材または振動板から加圧することで培養基材の底面の形状などに音響伝達媒体が追従しているところを示す。
 図9Aは第一例の音響伝達媒体の上面図と断面図を例示する。
(第二例)
 音響伝達媒体の第二例の構成図を図2Aから図2C、及び図9Bに示す。
 音響伝達媒体の第二例は、第二の音響伝達媒体が、第一の音響伝達媒体に内包され、かつ、第一の音響伝達媒体が第二の音響伝達媒体と対向する方向に凸となる突起部を有する。
 図2Aにおいては、第一の音響伝達媒体側の上面側に突起部を有する音響伝達媒体が、底面が平面の培養基材及び振動体と接している構成を示す。
 図2Bにおいては、第一の音響伝達媒体側の上面側に突起部を有する音響伝達媒体が、底面が反った形状の培養基材及び振動体と接している構成を示す。
 図2Cは、さらに、培養基材または振動板から加圧することで培養基材の底面の形状などに本開示の音響伝達媒体が追従しているところを示す。図示するように、突起部は加圧された状態で対面の第一の音響伝達媒体に接する構造となることが好ましい。
 図9Bは第二例の音響伝達媒体の上面図と断面図を例示する。図中の207は突起径を、208は突起高さを示す。
(第三例)
 音響伝達媒体の第三例の構成図を図3Aから図3C、及び図9Cに示す。
 音響伝達媒体の第三例は、第二の音響伝達媒体が、第一の音響伝達媒体に内包され、かつ、第一の音響伝達媒体は前記第二の音響伝達媒体を横切ってつながっている柱部を有する。
 図3Aにおいては、第一の音響伝達媒体が第二の音響伝達媒体を横切ってつながっている柱部を有する音響伝達媒体が、底面が平面の培養基材及び振動体と接している構成を示す。
 図3Bにおいては、第一の音響伝達媒体が第二の音響伝達媒体を横切ってつながっている柱部を有する音響伝達媒体が、底面が反った形状の培養基材及び振動体と接している構成を示す。

 音響伝達媒体の第四例は、第二の音響伝達媒体を第一の音響伝達媒体に内包するための第二の音響伝達媒体供給管133を有する。
 図4Aにおいては、第二の音響伝達媒体供給管133を有する音響伝達媒体が、底面が平面の培養基材及び振動体と接している構成を示す。
 図4Bにおいては、第二の音響伝達媒体供給管133を有する音響伝達媒体が、底面が反った形状の培養基材及び振動体と接している構成を示す。
 図4Cは、さらに、培養基材または振動板から加圧することで培養基材の底面の形状などに本開示の音響伝達媒体が追従しているところを示す。この状態においては、第二の音響伝達媒体供給管133の弁が閉じている状態が好ましい。
 図9Dは第四例の音響伝達媒体の上面図と断面図を例示する。
(実施例1)
 以下の実施例では、図1Aの概略図で示す音響伝達媒体の第一の例の構成を用いた例を示す。培養基材には、サーモフィッシャーサイエンティフィック社製Nunc(登録商標)Φ35mmディッシュを用いた。細胞剥離装置の振動板は、外径70mm、厚さ4mmのガラスを用いた。圧電体の材料組成はPZT、外径は振動板と同じ70mm、内径は57mm、厚さは中立面が振動板側にあることを考慮し、2mmとした。図6のように細胞剥離装置を組立て、振動板上には、音響伝達媒体は、図9Aの概略図で示す、高さ(205)=2.0mm、上面高さ(204)=1.0mm、下面高さ(206)=1.0mm、上面厚み(201)=0.2mm、下面厚み(202)=0.2直径(203)=30mmの形状で中に水を封止したものを使用した。
 音響伝達媒体の作製方法は音響伝達媒体の上面形状、下面形状に相当する金型を用意し、そこにシリコーンゴム原料(製品名:KE-1950-10;信越化学製)を調製して流し込み、加熱硬化させて上下面のシリコーンゴムを得、これを第一の音響伝達媒体とした。次いで、両面のゴムを接着剤で貼り合わせ、中に第二の音響伝達媒体として水を注入口より注入し、注入口を接着剤で封止した。余分な注入口を切除して音響伝達媒体を得た。
 得られた音響伝達媒体を振動板と中心が合うように設置し、次いで、細胞を培養した培養基材のサーモフィッシャーサイエンティフィック社製Nunc(登録商標)Φ35mmディッシュの中心を振動板の中心に合わせてゆっくりと音響伝達媒体上に設置した。このときの構成を表す概略図は図1Bであった。その後、培養基材の蓋上に100gの重りを設置して加圧した。加圧後の音響伝達媒体の構成を表す概略図は図1Cであった。
 細胞の培養は以下のように行った。CHO(Chinese Hamster Ovary)細胞をサーモフィッシャーサイエンティフィック社製Nunc(登録商標)Φ35mmディッシュに10000cells/cmの密度で播種し、37℃、CO濃度5%の環境下で培養した。培地はHam’s F12(サーモフィッシャーサイエンティフィック社製)にFetal Bovine Serum(シグマアルドリッチ社製)を10%、ペニシリン-ストレプトマイシン(10000U/ml、サーモフィッシャーサイエンティフィック社製)を1%添加したものを用いた。培養は48時間行ない、位相差顕微鏡で細胞の様子を観察し、細胞接着ならびに増殖を確認した。ディッシュの細胞占有面積率は8割程度であった。
(細胞の剥離)
 ディッシュ内の培地を除去し、PBS(-)で細胞を洗浄後、PBS(-)剥離液に3分間浸漬させた。
 その後、培養基材を環境温度25℃下で細胞剥離装置に設置し、環境温度25℃において定在波モードで2分間周波数掃引振動(周波数22-27kHz、周波数掃引周期1s、電圧90V)で細胞の剥離を行った。
 剥離した細胞を回収した後、超音波剥離後のディッシュに対して、セルスクレーパーを用いて超音波で剥離できなかった細胞を剥離した。剥離した細胞の数を血球計算盤測定した。超音波で剥離した細胞数とその後セルスクレーパーで剥離した細胞数を合わせて総剥離細胞数とし、総剥離細胞数に対する超音波剥離細胞数の割合を剥離率と定義し、算出した。バラツキについてはn=10で剥離率を求め、剥離率の標準偏差を5%未満がA、5%以上7.5%未満がB、7.5%以上10%未満がC、10%以上がDとして評価した。
 評価結果は、剥離率は95.4%、バラツキはA評価であった。
(実施例2)
 実施例2では第二の音響伝達媒体の種類を以下のように変更した以外は、実施例1と同様の条件で行った。すなわち、本実施例では、第二の音響伝達媒体として超音波ジェル(超音波検査用プロゼリー ノーマルタイプ;ジェクス社製)を使用した。
 評価結果は、剥離率は96.1%、バラツキはA評価であった。
(実施例3)
 実施例3では第二の音響伝達媒体の種類を以下のように変更した以外は、実施例1と同様の条件で行った。すなわち、第二の音響伝達媒体としてPVAゲルを使用した。
 PVAゲルは以下の方法で作製した。デンカポバール(登録商標)K-24E(デンカ株式会社製)を3.7質量%となるよう秤量し、純水を加えて調製し、合計80mLとした。恒温槽で熱を加えながら攪拌し、PVAを完全に溶解させ、PVA水溶液を得た。次いで、ホウ砂(特級四ほう酸ナトリウム(10水和物);キシダ化学製)を秤量し、純水を加えて1.0質量%となるように調製し、合計16mLとした。PVA水溶液を攪拌しながら、徐々に前記ホウ砂水溶液を滴下し、均一になるまで攪拌することでPVAゲルを得た。
 評価結果は、剥離率は93.0%、バラツキはA評価であった。
(実施例4)
 実施例4では第一の音響伝達媒体の形状を以下のように変更した以外は、実施例1と同様の条件で行った。すなわち、音響伝達媒体として、図9Aの概略図で示す、高さ(205)=2.0mm、上面高さ(204)=1.0mm、下面高さ(206)=1.0mm、上面厚み(201)=0.2mm、下面厚み(202)=0.4mm、直径(203)=30mmの形状のものを使用した。
 評価結果は、剥離率は94.6%、バラツキはA評価であった。
(比較例1)
 比較例1では音響伝達媒体として、シリコーンゴムを使用した以外は実施例1と同様の条件で検討を行った。
 音響伝達媒体として、厚さ2.0mm、硬度30°のシリコーンゴムを使用した。
 評価結果は、剥離率は80.1%、バラツキはD評価であった。
(まとめ)
 上記実施例、及び比較例の各構成をまとめた結果を表1に示す。
 本発明は上記実施の形態に制限されるものではなく、本発明の精神及び範囲から離脱することなく、様々な変更及び変形が可能である。従って、本発明の範囲を公にするために以下の請求項を添付する。
 本願は、2023年7月20日提出の日本国特許出願特願2023-118584を基礎として優先権を主張するものであり、その記載内容の全てをここに援用する。

Claims (15)

  1.  培養基材の培養面に付着した細胞を前記培養面から剥離する細胞剥離方法であって、
     前記培養基材と振動体との間に音響伝達媒体を配置した状態で、前記振動体を振動させて、前記培養面から前記細胞を剥離する細胞剥離方法、
     ただし、前記音響伝達媒体は、第一の音響伝達媒体及び第二の音響伝達媒体を含み、
     前記第一の音響伝達媒体は室温において弾性を有する固体であり、
     前記第二の音響伝達媒体は前記第一の音響伝達媒体に内包され、室温において液体であるか、または室温で前記第二の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体のtanδよりも大きいことを特徴とする、細胞剥離方法。
  2.  前記第一の音響伝達媒体は前記第二の音響伝達媒体と対向する方向に凸となる突起部を少なくとも1つ有することを特徴とする請求項1に記載の細胞剥離方法。
  3.  前記第一の音響伝達媒体は前記第二の音響伝達媒体を横切ってつながっている柱部を少なくとも1つ有することを特徴とする請求項1に記載の細胞剥離方法。
  4.  前記第一の音響伝達媒体は、シリコーンゴム、天然ゴム、ウレタンゲル、及びゲル材料からなる群から選択される少なくとも一種を含むことを特徴とする請求項1から3のいずれか1項に記載の細胞剥離方法。
  5.  前記第二の音響伝達媒体は、室温で、周波数0.1Hzの振動が付与された場合におけるtanδが1.0以上の固体であることを特徴とする請求項1から4のいずれか1項に記載の細胞剥離方法。
  6.  前記第二の音響伝達媒体は、室温で、液体であることを特徴とする請求項1から5のいずれか1項に記載の細胞剥離方法。
  7.  前記第二の音響伝達媒体は、液体、ポリマー溶液、ゲル材料からなる群から選択される少なくとも一種を含むことを特徴とする請求項1から6のいずれか1項に記載の細胞剥離方法。
  8.  前記第一の音響伝達媒体がシリコーンゴムを含み、前記第二の音響伝達媒体が水を含むことを特徴とする請求項1から7のいずれか1項に記載の細胞剥離方法。
  9.  前記音響伝達媒体の、前記振動体と対向する面は、前記音響伝達媒体の、前記培養基材と対向する面よりもヤング率が高いことを特徴とする請求項1から8のいずれか1項に記載の細胞剥離方法。
  10.  前記第二の音響伝達媒体を前記第一の音響伝達媒体に内包させる内包工程を含むことを特徴とする請求項1から9のいずれか1項に記載の細胞剥離方法。
  11.  前記振動体に超音波帯域の振動を発生させることを特徴とする請求項1から10のいずれか1項に記載の細胞剥離方法。
  12.  前記培養基材と前記振動体を近づけるように加圧する加圧工程を有することを特徴とする請求項1から11のいずれか1項に記載の細胞剥離方法。
  13.  培養基材の培養面に設けられた細胞に振動体が発生した振動を伝達することで、前記細胞を剥離する細胞剥離装置であって、
     振動体、
     音響伝達媒体、及び
     培養基材設置部を、この順に有し、
     前記振動体の振動が前記音響伝達媒体を経由して前記培養基材に伝達されるように構成され、
     前記音響伝達媒体は、第一の音響伝達媒体及び第二の音響伝達媒体を含み、
     前記第一の音響伝達媒体は室温において弾性を有する固体であり、
     前記第二の音響伝達媒体は前記第一の音響伝達媒体に内包され、室温において液体であるか、または室温で前記第二の音響伝達媒体に周波数0.1Hzの振動が付与された場合におけるtanδが、前記第一の音響伝達媒体のtanδよりも大きいことを特徴とする細胞剥離装置。
  14.  前記振動体は超音波振動子を含むことを特徴とする請求項13に記載の細胞剥離装置。
  15.  前記培養基材と前記振動体を近づけるように加圧する加圧手段を有することを特徴とする請求項13または14に記載の細胞剥離装置。
