본 발명은 신규한 고리 구조의 트리아졸 화합물, 이의 제조방법 및 이의 용도에 관한 것으로, 본 발명의 신규한 고리 구조의 트리아졸 화합물은 ASK1에 대하여 높은 억제능을 나타내고, 신경세포의 사멸을 억제하고 있으므로, 신경퇴행성 질환, 심혈관 질환, 자가면역질환 및 간 질환을 포함하는 ASK1 관련 질환의 예방 또는 치료에 사용될 수 있다.

Description

신규한 고리 구조의 트리아졸 화합물, 이의 제조방법 및 이의 용도
본 발명은 신규한 고리 구조의 트리아졸 화합물, 이의 제조방법, 및 이의 용도에 관한 것이다.
ASK1(Apoptosis signal-regulating kinase 1; ASK1)은 c-Jun N-말단 단백질 키나제(JNK) 및 스트레스에 반응하는 미토겐-활성화 단백질 키나제(mitogen-activated protein kinase; MAP3K) 계열의 구성원으로, MAP3K5라고도 알려져 있다. ASK1은 세포 사멸의 중심 조절인자이고, 산화 스트레스, 활성 산소 종(ROS), 소포체(ER) 스트레스 및 미접힘 단백질 반응(UPR), 미토콘드리아 스트레스 등 다양한 형태의 스트레스에 의해 유발되는 여러 스트레스-유발 및 수용체-매개 세포 사멸 경로에 참여하는 것으로 알려져 있다.
또한, ASK1은 세포 사멸 경로 뿐만 아니라 사이토카인 반응 및 세포 분화를 포함한 염증 및 선천 면역 반응에서도 필수적인 역할을 한다. ASK1 단백질의 인산화는 세포 유형에 따라 세포자연사(apoptosis) 또는 기타 세포 반응을 유발할 수 있다. 특히, ASK1 활성화 및 시그널링은 신경퇴행성, 심혈관, 염증성, 자가면역 및 대사 질환을 포함한 광범위한 질환에서 중요한 역할을 하는 것으로 보고되었다. 또한, ASK1은 심장, 뇌 및 신장의 허혈 및 재관류에 따른 장기 손상을 중재하는데 관련이 있다고 알려져 있다(Zhang et al. J Clin Invest.2003; 111(12): 1933-1943.).
한편, 파킨슨병은 주로 노년층에서 발생하는 신경퇴행성 질환이다. 파킨슨병은 뇌의 중간에 존재하는 뇌흑질의 도파민성 신경세포가 사멸하여 행동에 장애를 일으킨다. 또한, 뇌와 말초신경의 여러 부위에 알파 시누클레인 단백질이 침착되어 생긴 루이소체가 파킨슨병 환자의 신경병리학적 주된 특징이다. 파킨슨병의 대표적인 증상으로 몸의 움직임이 비정상적으로 느려지는 서동, 손이나 발이 떨리는 진전 및 근육과 관절이 뻣뻣해지는 경직이 있으며, 비운동성 증상으로 정신장애, 자율신경 기능 장애 등이 나타난다.
이때, 과도한 양의 ASK1은 심장질환, 염증질환, 간질환, 감염병 및 퇴행성 뇌질환과 밀접하게 연관되어 있음이 보고되고 있으며, ASK1 저해 기반 파킨슨병 치료제로서의 가능성이 다각도로 제기되고 있다. 알파 시누클레인 PFF(alpha-synuclein pre-formed fibrils)에 의한 파킨슨병 유발 시, ASK1 결손 마우스에서는 선조체와 대뇌피질에서 인산화된 알파 시누클레인(p-alpha-synuclein)의 축적이 감소하였다. 나아가 신경염증도 완화되었으며, 이는 행동학적 개선으로도 이어졌다. 또한, 파킨슨병의 환자의 대뇌 흑질 부분의 신경세포에서 정상인 대비 4배 이상의 p-ASK1의 활성화가 관찰되었다.

나아가, 본 발명은 상기 화학식 1로 표시되는 화합물 및 이의 약학적으로 허용가능한 염 뿐만 아니라, 이로부터 제조될 수 있는 용매화물, 입체이성질체, 수화물 등을 모두 포함한다.
본 명세서에서 사용된 용어, "수화물(hydrate)"은 비공유적 분자간력(non-covalent intermolecular force)에 의해 결합된 화학양론적(stoichiometric) 또는 비화학양론적(non-stoichiometric) 량의 물을 포함하고 있는 본 발명의 화합물 또는 그것의 염을 의미한다. 본 발명의 상기 화학식 1로 표시되는 화합물의 수화물은 비공유적 분자간 힘으로 결합되는 화학양론적 또는 비화학양론적 양의 물을 포함할 수 있다. 상기 수화물은 1당량 이상, 바람직하게는, 1 당량 내지 5당량의 물을 함유할 수 있다. 이러한 수화물은 물 또는 물을 함유하는 용매로부터 본 발명의 상기 화학식 1로 표시되는 화합물, 이의 입체이성질체, 이의 용매화물, 이의 수화물 또는 이들의 약제학적으로 허용 가능한 염을 결정화시켜 제조될 수 있다.

본 명세서에서 사용된 용어, "이성질체(isomer)"는 동일한 화학식 또는 분자식을 가지지만 구조적 또는 입체적으로 다른 본 발명의 화합물 또는 그것의 염을 의미한다. 이러한 이성질체에는 호변이성질체(tautomer)까지 포함하는 구조이성질체와, 입체이성질체(stereoisomer)가 포함되며, 입체이성질체에는 비대칭 탄소 중심을 1개 이상 가짐으로써 나타나는 광학이성질체(enatiomer) 및 부분입체이성질체, 그리고 기하이성질체(트랜스, 시스)가 모두 포함된다. 이들 모든 이성체 및 그것의 혼합물들 역시 본 발명의 범위에 포함된다.

또한, 상기 화합물, 이의 입체이성질체, 이의 용매화물, 이의 수화물 또는 이의 약학적으로 허용가능한 염은 신경세포의 사멸을 억제할 수 있다.
본 발명의 일 측면은, 상기 화학식 1로 표시되는 화합물, 이의 입체이성질체, 이의 용매화물, 이의 수화물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 ASK1 관련 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
약학적 조성물은, 상기 유효성분 외에 약학적으로 허용 가능한 담체를 더 포함할 수 있다.
상기 ASK1 관련 질환은 신경퇴행성 질환, 심혈관 질환, 자가면역 질환, 및 간 질환을 포함한다.
상기 신경퇴행성 질환은 알츠하이머병, 해마 경화증, 전두측두엽 치매 (FTD), 전두측두엽 변성 (FTLD), 헌팅턴병, 피질기저핵 변성, 근위축성 측삭경화증, 척수성 근위축, 운동 뉴런 질환, 봉입체 근염, 파킨슨병, 루이소체 치매, 루이소체 질환, 다계통 위축, 진행성 핵상 마비, 피크병, 프라이온병, 외상성 뇌 손상, 허혈성 및 출혈성 뇌졸중, 대뇌 허혈, 저산소증, 및 글루타메이트 신경독성을 포함한다.
상기 심혈관 질환은 심부전, 허혈, 재발성 허혈, 심근경색, 부정맥, 급성 관상동맥 증후군, 당뇨병, 죽상경화증 및 간헐적 파행을 포함한다.
상기 자가면역 질환은 류마티스 관절염, 섬유근육통, 전신홍반루푸스, 다발성 경화증, 당뇨병, 전신경화증, 그레이브스병, 길랑-바레 증후군, 중증근무력증, 건선, 크론병, 궤양성 대장염, 시신경염, 및 쇼그렌 증후군을 포함한다.
상기 간 질환은 비알코올성 지방간(NASH), 알코올성 지방간, 간 섬유화, 간암, 간독성, 담즙울체, 간경변, 간허혈, 간 농양, 간성 혼수, 및 간 위축증을 포함한다.
상기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용가능한 염은 임상 투여시에 경구 및 비경구의 여러 가지 제형으로 투여될 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등 이 포함되며, 이러한 고형제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용된다. 경구투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제가 포함된다. 비수성용제, 현탁용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테로 등이 사용될 수 있다.
상기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용가능한 염을 유효 성분으로 하는 약학적 조성물은 비경구 투여할 수 있으며, 비경구 투여는 피하주사, 정맥주사, 근육 내 주사 또는 흉부 내 주사를 주입하는 방법에 의한다.
이때, 비경구 투여용 제형으로 제제화하기 위하여 상기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용가능한 염을 안정제 또는 완충제와 함께 물에 혼합하여 용액 또는 현탁액으로 제조하고, 이를 앰플 또는 바이알 단위 투여형으로 제조할 수 있다. 상기 조성물은 멸균되고/되거나 방부제, 안정화제, 수화제 또는 유화 촉진제, 삼투압 조절을 위한 염 및/또는 완충제 등의 보조제, 및 기타 치료적으로 유용한 물질을 함유할 수 있으며, 통상적인 방법인 혼합, 과립화 또는 코팅 방법에 따라 제제화할 수 있다.
경구 투여용 제형으로는 예를 들면 정제, 환제, 경/연질 캅셀제, 액제, 현탁제, 유화제, 시럽제, 과립제, 엘릭시르제, 트로키제 등이 있는데, 이들 제형은 유효성분 이외에 희석제(예: 락토즈, 덱스트로즈, 수크로즈, 만니톨, 솔비톨, 셀룰로즈 및/또는 글리신), 활택제(예: 실리카, 탈크, 스테아르산 및 그의 마그네슘 또는 칼슘염 및/또는 폴리에틸렌 글리콜)를 함유하고 있다. 정제는 마그네슘 알루미늄 실리케이트, 전분 페이스트, 젤라틴, 메틸셀룰로즈, 나트륨 카복시메틸셀룰로즈 및/또는 폴리비닐피롤리딘 등과 같은 결합제를 함유할 수 있으며, 경우에 따라 전분, 한천, 알긴산 또는 그의 나트륨 염 등과 같은 붕해제 또는 비등 혼합물 및/또는 흡수제, 착색제, 향미제, 및 감미제를 함유할 수 있다.
본 발명의 상기 화학식 1로 표시되는 화합물, 이의 입체이성질체, 이의 용매화물, 이의 수화물 또는 이의 약학적으로 허용가능한 염 또는 이를 포함하는 약학적 조성물은 “약학적으로 유효한 양”으로 투여된다. 본 명세서에 있어서, 용어 “약학적으로 유효한 양”이란, 의학적 치료 또는 개선에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 예를 들어, 0.001 mg/kg 내지 1000 mg/kg, 0.01 mg/kg 내지 100 mg/kg 또는 0.1 내지 20 mg/kg 또는 0.1 내지 500 mg/kg의 유효한 양이 포함된다. 본 발명의 화합물 또는 약학적 조성물의 양은 당업자가 적절한 범위 내에서 선택하여 실시할 수 있다.
본 발명의 일 측면은, 상기 화학식 1로 표시되는 화합물, 이의 입체이성질체, 이의 용매화물, 이의 수화물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 ASK1 관련 질환의 예방 또는 개선용 건강기능식품을 제공한다.
본 발명에 따른 화학식 1로 표시되는 화합물은, 높은 ASK1 억제능을 나타냄으로써, ASK1 관련 질환의 예방 또는 개선용 건강기능식품 조성물로 약학적 조성물에 준하여 제조되거나, 식품, 음료 등의 건강기능보조 식품에 첨가할 수 있다.
본 발명에 따른 상기 화학식 1로 표시되는 화합물은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분의 혼합량은 그의 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 일반적으로, 건강식품 중의 상기 화합물의 양은 전체 식품 중량의 0.1 내지 90 중량부로 가할 수 있다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.

상기 투여는 경구 또는 비경구 투여일 수 있다. 경구 투여시는 활성성분을 기준으로 하루에 체중 1 kg당 0.01 내지 1000 mg, 보다 구체적으로는 0.1 내지 300 mg의 양으로 투여되도록, 비경구 투여시는 활성성분을 기준으로 하루에 체중 1 kg당 0.01 내지 100 mg, 보다 구체적으로는 0.1 내지 50 mg의 양으로 투여되도록 1회 내지 수회에 나누어 투여할 수 있다. 특정 개체 또는 환자에 대한 투여 용량은 환자의 체중, 연령, 성별, 건강 상태, 식이, 투여 시간, 투여 방법, 질환의 중증도 등의 여러 관련 인자에 비추어 결정되어야 하는 것이고 전문가에 의해 적절하 가감될 수 있다.
본 명세서에서 용어, "개체"는 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는, 인간 또는 비-인간인 영장류, 생쥐(mouse), 개, 고양이, 말, 및 소 등의 포유류를 의미한다.
본 발명의 일 측면은, ASK1 관련 질환의 예방 또는 치료에 사용하기 위한, 상기 화학식 1로 표시되는 화합물, 이의 입체이성질체, 이의 용매화물, 이의 수화물 또는 이의 약학적으로 허용가능한 염의 용도를 제공한다.

제조예 1. 6-(1H-벤조[d][1,2,3]트리아졸-1-일)피리딘-2-아민(중간체 1)

상기 반응식에 따라, 중간체 1을 얻었다.
DMSO (6 mL) 내에서 6-플루오로피리딘-2-아민 (200 mg, 1.78 mmol), 벤조트리아졸 (255 mg, 2.14 mmol) 및 K2CO3 (740 mg, 5.34 mmol)의 혼합물을 12시간 동안 180℃에서 가열하였다. 상온에서 냉각 후, 1 M Na2CO3 용액을 추가하였다. 생성된 고체는 여과하고 감압하에 건조하여, 6-(1H-벤조[d][1,2,3]트리아졸-1-일)피리딘-2-아민을 수득하였다(중간체 1, 230 mg, 61%). 1H NMR (500 MHz, Chloroform-d) δ 8.45 (d, J = 8.4 Hz, 1H), 8.11 (d, J = 8.3 Hz, 1H), 7.72 (t, J = 8.0 Hz, 1H), 7.59 (t, J = 7.7 Hz, 1H), 7.48 (d, J = 7.7 Hz, 1H), 7.44 (t, J = 7.7 Hz, 1H), 6.68 (d, J = 8.2 Hz, 1H); LCMS: 212.4 [M+H+].
제조예 2. 6-(5,6-디하이드로사이클로펜타[ d ][1,2,3]트리아졸-1(4 H )-일)피리딘-2-아민(중간체 2)
하기 반응식에 따라 중간체 2를 얻었다.

단계 1: 1-아지도-4-니트로벤젠
4-니트로아닐린 (1.00 g, 7.24 mmol)은 무수 아세토니트릴 (20 mL)에 용해된 후, 아이스 배스에서 0℃로 냉각시켰다. 교반 용액에, 터트-부틸 니트라이트(1.03 mL, 8.69 mmol)를 첨가하였고, 생성된 혼합물은 20분 동안 교반시켰다. 그 다음, 아지도트리메틸시레인 (1.44 mL, 10.86 mmol)을 10분동안, 한 방울씩 첨가하였고, 갈색의 최종 용액을 12시간 동안 상온에서 교반시켰다. 용매는 감압 하에서 제거하였다. 잔여물은 실리카겔을 이용한 플래시 컬럼 크로마토그래피(flash column chromatography) (헥산/에틸 아세테이트 100/0->80/20)을 이용하여 정제하여, 1-아지도-4-니트로벤젠을 수득하였다(1.16 g, 97%).

단계 2: 1,4,5,6-테트라하이드로사이클로펜타[ d ][1,2,3]트리아졸
사이클로펜탄온 (1.08 mL, 12.2 mmol), NH4OAc (4.69 g, 60.9 mmol), 및 1-아지도-4-니트로벤젠을 마그네틱 교반바를 장착한 스크류-캡이 있는 반응 튜브에 첨가하였다. 생성된 혼합물은 DMF (20 mL)에서 용해시켰고 12시간 동안 80℃에서 교반하였다. 완료시 감압 하에서 DMF를 제거하였고, 실리카겔을 이용한 플래시 컬럼 크로마토그래피 (DCM 100% 후, 헥산/에틸 아세테이트 100/0->50/50)을 이용하여 정제하여, 1,4,5,6-테트라하이드로사이클로펜타[d][1,2,3]트리아졸을 수득하였다(164 mg, 61%).

단계 3: 6-(5,6-디하이드로사이클로펜타[ d ][1,2,3]트리아졸-1(4 H )-일)피리딘-2-아민
DMSO (20 mL) 내에서 6-플루오로피리딘-2-아민 (940 mg, 8.32 mmol), 1,4,5,6-테트라하이드로사이클로펜타[d][1,2,3]트리아졸 (1.00 g, 9.16 mmol) 및 K2CO3 (3.44 g, 25.0 mmol)의 혼합물을 12시간 동안 160℃에서 가열하였다. 상온에서 냉각 후, 혼합물은 물로 희석되었고 아세틸 아세테이트로 두 번 추출되었다. 혼합된 유기층은 염수로 세척되었고, MgSO4로 건조시킨 후, 여과하고 농축하였다. 잔여물은 실리카겔을 이용한 플래시 컬럼 크로마토그래피 (헥산/에틸 아세테이트 100/0->60/40)을 이용하여 정제하여, 6-(5,6-디하이드로사이클로펜타[d][1,2,3]트리아졸-1(4H)-일)피리딘-2-아민(중간체 2, 700 mg, 42%)을 수득하였다.

단계 1. 2,2-디메톡시사이클로헥산-1-온
메탄올 (500 ml) 내에 트리메틸실일 클로라이드 (7.0 ml, 58.9 mmol) 용액에 메탄올 (50 ml) 내에 사이클로엑산-1,2-디온 (6.0 g, 53.5 mmol)의 용액을 천천히 추가하였고, 생성된 혼합물은 16시간 동안 상온에서 교반하였다. 반응 혼합물은 물에 붓고, 디에틸 에테르로 추출하였다. 유기층은 포화된 암모늄 카보네이트용액 및 물, 염수로 세척하였다. MgSO4로 건조 후, 용매는 감압하에 제거하여, 2,2-디메톡시사이클로헥산-1-온을 수득하였다(quantitative yield).

단계 2. 6-(4,5,6,7-테트라하이드로-1 H -벤조[ d ][1,2,3]트리아졸-1-일)피리딘-2-아민

상기 반응식에 따라서 실시예 1 화합물을 제조하였다.
DMF (3 mL) 중 6-(1H-벤조[d][1,2,3]트리아졸-1-일)피리딘-2-아민 (중간체 1, 70 mg, 0.33 mmol), DMAP (400 mg, 3.30 mmol) 및 5-(4-사이클로프로필-1H-이미다졸-1-일)-2-플루오로-4-메틸벤조익 산 (130 mg, 0.50 mmol) [purchased from AA blocks (5 g), Labnetwork (10 g), Chemscene; reference: WO 2013/112741]의 교반된 용액에, HATU (1.25 g, 3.30 mmol)을 실온에서 첨가하였다. 생성된 혼합물을 12시간 동안 상온에서 교반한 후, 에틸 아세테이트 및 포화 NaHCO3 수용액으로 희석하였다. 분리된 유기층은 염수로 세척하고, 무수 MgSO4로 건조시킨 후, 여과하였다. 유기 여과액은 감압 하에서 증발시켰다. 수득한 물질은 실리카겔을 이용한 플래시 컬럼 크로마토그래피(flash column chromatography) (에틸 아세테이트/메탄올 100/0->90/10) 및 C18-역상 HPLC (완충 용매로 H2O 중 % TFA 및 ACN 중 0.1% TFA 이용)을 이용하여 정제하여, N-(6-(1H-벤조[d][1,2,3]트리아졸-1-일)피리딘-2-일)-5-(4-사이클로프로필-1H-이미다졸-1-일)-2-플루오로-4-메틸벤즈아미드를 수득하였다(1, 35 mg, 23%).

시판되는 5-(4-사이클로프로필-1H-이미다졸-1-일)-2-플루오로-4-메틸벤조산(0.38g, 1.3mmol)의 교반 용액을 포함하고 있는 NMP(3.0ml)에 DMAP(0.36g, 3.0mmol)를 가한 후 실온에서 T3P(50% 에틸 아세테이트)(1.27ml, 2.0mmol)를 천천히 가하였다. 결과 혼합물을 10분간 교반하고 6-(5,6-디하이드로사이클로펜타[d][1,2,3]트리아졸-1(4H)-일)피리딘-2-아민(0.2g, 1.0mmol)을 첨가하고 12시간 동안 교반하였다. 반응 혼합물을 얼음 냉수로 퀀칭하고 포화 NaHCO3 수용액으로 염기화(pH 7-8)하였다. 얻어진 고체를 여과하고 물로 세척하고 건조시켰다. 고체를 에틸 아세테이트로 삼투시켜 원하는 5-(4-시클로프로필-1H-이미다졸-1-일)-N-(6-(5,6-디하이드로사이클로펜타[d][1,2,3]트리아졸-1(4H)-일)피리딘-2-일)-2-플루오로-4-메틸벤자마이드 화합물을 순수한 백색 고체(0.3g, 68%)로 수득하였다.

시판되는 5-(4-사이클로프로필-1H-이미다졸-1-일)-2-플루오로-4-메틸벤조산(3.93 g, 15.1 mmol)의 교반 용액을 포함하고 있는 NMP(30 ml)에 DMAP(4.25 g, 34.8 mmol)를 가한 후 실온에서 T3P(50% 에틸 아세테이트)(14.7 g, 23.2 mmol)를 천천히 가하였다. 결과 혼합물을 10분간 교반하고 6-(4,5,6,7-테트라하이드로-1H-벤조[d][1,2,3]트리아졸-1-일)피리딘-2-아민(2.5g, 11.6mmol)을 첨가하고 12시간 동안 교반하였다. 반응 혼합물을 얼음 냉수로 퀀칭하고 포화 NaHCO3 수용액으로 염기화(pH 7-8)하였다. 얻어진 고체를 여과하고 물로 세척하고 건조시켰다. 고체를 메탄올로 삼투시켜 원하는 5-(4-사이클로프로필-1H-이미다졸-1-일)-2-플루오로-4-메틸-N-(6-(4,5,6,7-테트라하이드로-1H-벤조[d][1,2,3]트리아졸-1-일)피리딘-2-일)벤자미드 화합물을 백색 고체(5.0g, 94%)로 수득하였다.

중간체 1 대신 중간체 4를 사용한 것을 제외하고 실시예 1과 동일한 방법을 수행하여 5-(4-사이클로프로필-1H-이미다졸-1-일)-2-플루오로-4-메틸-N-(6-(5,6,7,8-테트라하이드로사이클로헵타[d][1,2,3]트리아졸-1(4H)-일)피리딘-2-일)벤즈아미드를 수득하였다(13%).

실험예 1. ASK1 저해 활성 확인
본 발명에 따른 화학식 1로 표시되는 화합물의 ASK1 활성 억제능을 측정하기 위하여 하기와 같은 실험을 수행하였다.
재조합 인간 ASK1 단백질은 반응액 (8 mM MOPS pH 7.0, 0.2 mM EDTA, 0.33 mg/mL myelin basic protein, 10 mM Mg Acetate and [gamma-33P]-ATP)에서 Mg/ATP 혼합액을 첨가함에 의해 반응을 개시하고, 실온에서 40분간 반응한 후 0.5% phosphoric acid를 첨가하여 반응을 종료시켰다. 이후 P30 점적필터에 10uL의 반응액을 떨어뜨린 후 0.425% phosphoric acid에서 4분간, 총 4회 세척하여 methanol에 넣어 말리고 신틸레이션 계수(scintillation counting)을 통해 ASK1의 활성을 측정하였다. 화합물이 포함되지 않은 군에서 나온 활성 값을 100%로 정하여 화합물에 의한 ASK2 저해 정도를 환산하고 하기 표 2에 실시예 화합물의 ASK1 저해 활성을 정리하였다.

상기 표 2를 살펴보면, 본 발명에 따른 화학식 1로 표시되는 화합물은 ASK1 저해 활성을 가지는 물질임을 확인할 수 있었다. 따라서, 본 발명의 화합물은 ASK1으로부터 유발되는 신경퇴행성 뇌질환의 예방 또는 치료용 조성물로 유용하게 사용될 수 있다.
실험예 2. MPP+ 유도된 인간 신경세포에서의 세포보호효과 평가
본 발명에 따른 화학식 1로 표시되는 화합물에 대해 도파민성 신경세포의 손상을 보호하는 효과를 평가하기 위하여 하기와 같은 실험을 수행하였다.
SH-SY5Y 세포를 ATCC에서 구입하였으며, 10% FBS 및 1% 페니실린(penicillin) 및 스트렙토마이신(streptomycin)이 함유된 RPMI 배지에서 37℃, 5% CO2 조건하에서 배양하였다. MPP+는 sigma aldrich에서 구입하였으며, 5 mM로 처리하여 세포독성을 유발하였다. 세포생존율은 CCK-8 시약을 처리하고 1-3시간 후 540nm에서의 흡광도를 측정하여, 그 결과를 하기의 표 3에 나타내었다.

상기의 표 3 및 도 1에 나타난 바와 같이, SH-SY5Y 세포에서 본 발명의 실시예 화합물은 우수한 세포 보호 효과를 나타내었으며, 특히 실시예 2 및 3은 비교예 1(Selonsertib) 보다 우수한 EC50 값을 보였다.
실험예 3. 세포 생존율 평가
화합물 처리시 세포 생존율을 평가하기 위해 BV2세포를 96-웰 플레이트에 웰 당 2x104개씩 시딩(seeding)하고 하루 동안 배양한 후, 화합물을 각각 0.1 μM, 0.3 μM 및 1 μM로 처리 후 37℃의 5% CO2에서 72시간 동안 배양하였다. 세포 생존율을 배양이 끝난 세포에 Cell Counting Kit-8 (CCK-8; LPS solution, Seoul, Korea)을 첨가한 후 Hidex sense microplate reader (Hidex, Turku, Finland)를 사용해 565 nm 파장에서 측정된 흡광도를 사용해 정량화 하였다.
도 2에 나타난 바와 같이, 본 발명에 따른 실시예 화합물은 비교예 1(selonsertib)과 유사한 수준의 세포 생존율을 보였다.
실험예 4. 항염증 효능 평가
BV2 세포(well 당 2 x 104 세포)를 96-well 배양 플레이트에서 24시간 동안 배양하였다. 그 후, 각각 0.1 μM, 0.3 μM 및 1 μM로 1시간 동안 실시예 화합물을 처리한 후 LPS 100 pg/ml을 첨가하여 24시간 동안 배양하였다. 배양 후 conditioned medium (CM)을 수확하여 2000 rpm에서 원심분리 하였다. 원심분리가 끝난 용액의 상층액을 분리한 후 그 상층액에서 세포 배양 배지에서 분비되는 TNF-α는 해당 ELISA 키트(LABISKOMA, Seoul, Korea)를 사용하여 측정하였다.
도 2에 나타난 바와 같이, 본 발명에 따른 실시예 화합물은 selonsertib(비교예 1)과 유사한 수준으로 LPS에 의해 유도되는 염증성 사이토카인 TNF-α의 분비를 감소시켰다. 특히 실시예 3은 selonsertib과 비교하여 모든 농도에서 TNF-α 분비를 감소시키는 것을 확인하여, 우수한 항염증 효능을 가진다는 것을 확인하였다.
실험예 5. 신경세포 보호 효능 평가
신경세포 독성을 일으키는 물질인 6-OHDA(6-hydroxydopamine hydrobromide)와 MPP+(1-methyl-4-phenylpyridinium)은 Sigma-Aldrich (Saint Louis, MO, USA)에서 구매해 100 mM DMSO stock으로 만들어 사용하였다. PC12 세포를 96-웰 플레이트에 웰당 1x104개씩 시딩하고 하루 간 배양하였다. 세포를 각 농도의 실시예 화합물과 함께 1시간 동안 전처리하였다. 그 후, 각각 40 μM 6-OHDA와 2 mM의 MPP+를 첨가해 24시간 동안 배양하였다. 그 후 MTT (Invitrogen, MA, USA)를 이용해 세포 생존율을 측정했다.
표 4 및 5에 나타난 바와 같이, 본 발명에 따른 실시예 화합물은 비교예 1(selonsertib)보다 뛰어난 세포 생존율을 보였으므로, PC-12 cell line에서 뛰어난 신경세포 보호 효과가 있음을 확인하였다.

실험예 6. TG 합성 억제 효과 평가
Triglyceride(TG) 측정은 인간 간암세포중인 HepG2 세포를 96-웰 플레이트에 각 웰 당 2x104 세포를 분주해 24시간 동안 배양하였다. 배양된 세포는 농도 별로 실시예 화합물을 2시간 전처리 한 후, Palmitic acid (PA, sigma, #P9767)와 Oleic acid (OA, sigma, #O7501)를 1:2 비율로 혼합한 Free fatty acid (FFA)를 1mM 첨가한 후 48시간 동안 배양하였다. 배양 후 상층액을 분리해 Triglyceride Quantification Kit(sigma, #MAK266)을 이용해 570nm의 흡광도에서 측정하였다.
하기 표 6에 나타낸 것과 같이, 항-NASH(비알콜성지방간염) 효능은 TG 합성 억제를 이용해 검증하였다. 실험 결과, 실시예 3 화합물은 처리 농도가 증가할수록 더 우수한 TG 합성 억제 효과를 나타내었다.

실험예 7. MDR-MDCK 세포막 투과도 평가
MDR-MDCK 세포 단일층을 통한 투과도를 측정하기 위해 MDCK-MDR1 세포(5Х105 cells/mL)를 24-transwell plate(Corning, NY, USA)에 4일 동안 배양하였다. 배지는 이틀에 한 번 새 배지로 교체하였으며, 상피조직 간 전기저항(TEER) 값은 Millicell® ERS-2 Volohmmeter(Merck KGaA, Darmstadt, Germany)를 이용하여 측정하여, 이 값이 300 Ohm.cm2 이상임을 확인한 후 세포막 투과도 시험을 수행하였다.
apical 및 basolateral chamber는 DPBS buffer, 10 mM Hydroxyethyl piperazine Ethane Sulfonic acid(HEPES)를 포함한 Hanks' Balanced Salt Solution (HBSS) 순서로 각각 2번씩 세척하였으며, 마지막 세척 단계에서는 HBSS를 채워 37℃에서 30분 동안 안정 상태를 유지하였다. 30분 경과 후 시험물질을 각 chamber에 점적한 후 37℃에서 2시간 동안 방치하며 이동한 층의 배양액을 30분 간격으로 수거하였고, 수거한 양만큼 새 HBSS로 보충해 주었다.

표 7에 나타난 바와 같이, 비교예 1 화합물(Selonsertib)은 흡수 방향과 유출방향의 겉보기 계수의 큰 차이로 인해 27의 유출계수를 나타내 유출 수송체의 기질임이 확인된 반면, 본 발명에 따른 실시예 화합물들은 흡수 및 유출방향 모두 매우 높은 겉보기 배수 값과 1에 가까운 유출 비율을 나타내어 세포막 투과가 용이하며, 유출수송체의 기질이 아님을 확인하였다.
실험예 8. 마우스 뇌 내 분포
ICR 마우스에서의 혈액-뇌 장벽 투과율은 10 mg/kg의 시험물질을 경구 투여하여 확인하였다. 수컷 C57BL/6 마우스는 투여 전 15시간 동안 절식시켰다. 시험물질은 10 mg/kg의 용량과 5 mL/kg의 부피로 투여되었으며, 투여 용매 조성은 N-Methyl-2-pyrrolidone, NMP), Polyethylene glycol 400(PEG 400), 3차 증류수 (Distilled water, D.W)를 각각 10:40:50 (%)의 부피비로 제조하였다. 투여 후에는 1, 4, 8시간에 해당하는 타임 포인트 별로 마우스를 이산화탄소 흡입법으로 희생시켜 혈액 및 뇌조직 샘플을 취하였다. 전혈은 복대정맥에서 채취되었으며, 식염수로 전신 관류를 시켜 혈액을 최대한 제거한 후 뇌 샘플을 수거하였다.
마우스 혈장 및 뇌에서의 분포 비율은 각 조직에서의 농도 정량 후 AUC를 산출하여 계산하였다.

표 8에서 나타난 바와 같이, 본 발명에 따른 실시예 3은 비교예 1 (Selonsertib)에 비해 혈장 및 뇌 중 농도와 Kp 값이 상대적으로 매우 높았으므로, 뇌질환 치료를 위해 우수한 약동학적 특성을 가지고 있음을 확인하였다.
실험예 9. MPTP-유도 파킨슨병 마우스 모델에 대한 효능 평가
실험예 9.1. 실험 준비
본 명세서에 기재된 모든 동물 실험은 윤리위원회(IACUC)의 승인을 받아 식약처의 실험동물 관리 및 사용에 관한 지침에 따라 수행하였다.
코아텍(Koatech, 한국)에서 생후 6주령의 C57BL/6 수컷 마우스를 구입하였다. 마우스는 케이지당 최대 4마리씩 그룹을 지어 온도 (22±1℃)와 습도 (30~50%)가 조절된 환경에서 일반적인 명암 주기(08:00~20:00)로 사육했으며 음식과 물을 무제한(Ad libitum)으로 제공하였다. 실험 시작 전 1주일 동안 마우스가 적응할 수 있도록 하였다.
정상 마우스 및 MPTP(1-메틸-4-페닐-1,2,3,6-테트라하이드로피리딘)에 의해 유도된 파킨슨병(PD) 마우스 모델에 대하여 시험 약물을 투여하고 효능을 평가하였다. 실시예의 화합물에 대하여 10% NMP, 40% PEG 400 및 50% D.W.(증류수)를 전달체(vehicle)로 사용하여 시험 약물을 준비하였다.
평가 결과에서 모든 값은 평균 ± 표준 오차(SEM)로 표시하였으며, LSD(Least Significant Difference: 최소 유의차) 사후 분석(post hoc)을 통한 일원 분산 분석(one-way ANOVA)으로 통계적으로 분석되었다. 평가 결과의 유의 수준(Significance level)은 하기 기준에 따라 책정하였다.

실험예 9.2. 실험 설계
도 3은 MPTP-유도 PD 마우스 모델에 대한 효능 실험의 모식도이다.
생후 7주령의 마우스를 아래 표 9과 같이 7개군으로 나누어 급성 MPTP 모델 생성 및 약물 투여를 진행하였다: 대조군 + vehicle (G1), MPTP + vehicle (G2), MPTP + 닐로티닙 (Nilotinib) 40 mg/kg (G3), MPTP + 실시예3 2 mg/kg (G4), MPTP + 실시예3 10 mg/kg (G5), MPTP + 실시예3 50 mg/kg (G6) 및 MPTP + 실시예2 50 mg/kg (G7). 시험 약물을 1일차부터 7일차까지 1일 1회(QD) 10 ml/kg씩 경구 투여(PO)하였으며. 급성 MPTP 모델은 대조군을 제외한 모든 그룹은 0일차에 2시간 간격으로 MPTP.HCl (유리 염기 20 mg/kg)을 복강 내(IP) 주사로 4회 투여하였다. 대조군에는 동일한 양의 0.9% 식염수를 투여하였다. 폴 테스트(Pole test)는 4일차 (2일차 및 3일차 훈련 후)에 실시하였다. 7일차에 모든 마우스를 안락사시키고 뇌를 채취하였다.
연구 기간 동안 매일 체중을 측정하고 마우스의 상태에 이상이 없는지 검사하였다. 마우스의 전반적인 건강 상태가 현저히 악화되는 경우, 마우스를 안락사 시키고 연구 대상에서 제외하였다.

실험예 9.3. 행동 결함 개선 효능 평가
케이지에 수직 나무 기둥(마우스에 그립감을 제공하는 직경 0.8 cm 내지 1 cm, 길이 80 cm)을 바닥에 놓고 테스트하였다. 마우스는 이틀 연속(2일차, 3일차) 훈련을 받았으며 각 훈련 세션은 세 번의 시험으로 구성하였다. 3일차 (실제 시험일인 4일차)에는 마우스를 기둥 끝부분에 놓고 꼭대기를 돌아 기둥 아래까지 도달하는 총 시간을 측정하되, 꼭대기에서 회전하기(turn down, 마우스가 고개를 아래로 돌리는 데 걸린 시간), 내려오기(climb down, 고개를 숙인 시점부터 바닥에 닿는 데 걸린 시간) 및 바닥에 닿는 데 걸린 총 시간(total time)을 구분하여 기록하고 도 4의 그래프를 얻었다.
도 4는 MPTP-유도 PD 마우스 모델에 대한 행동 결함 평가(폴 테스트) 결과 그래프이다. 폴 테스트에서 MPTP를 처리한 그룹(G2)은 대조군(G1)에 비해 회전(도 4의 a), 하강(도 4의 b) 및 지면에 도달하는 총 시간(도 4의 c)이 훨씬 더 많이 소요된 것이 확인되었다. MPTP와 실시예 화합물을 처리한 그룹(G4 내지 G7)은 그룹 G2에 비해 회전하는 데 필요한 시간이 현저히 감소하였다. 특히, 그룹 G4 및 G5에서 현저한 회전, 하강 및 지면 도달 시간이 감소하였다. 이를 통해, 실시예 화합물이 유의미하게 행동 결함을 개선시키는 것이 확인되었다.
실험예 9.4. TH 염색 정도 평가
조직학적 절편의 염색 및 분석을 위한 조직 샘플 수집을 위해 마우스를 졸레틸 투여로 안락사 시켰다. 마우스의 심장에서 혈액을 채취한 후 PBS(인산염 완충 식염수)로 잔여 혈액을 관류하였다. 절반의 마우스(n=6/그룹)에 대해 뇌를 선조체, 흑질 및 나머지 뇌로 절개하여 급속 냉동한 후 -80℃에서 보관하였다. 나머지 마우스(n=6/그룹)는 뇌를 분리하여 4% 파라포름알데히드에 4℃에서 24시간 동안 고정시킨 다음, 0.02% 소듐 아자이드(sodium azide)가 포함된 PBS(인산염 완충 식염수)에 30% 자당 (sucrose)을 넣고 4℃에서 3일 동안 보관하였다. OCT(Optimal cutting temperature) 화합물이 내장된 뇌 샘플에 대해 면역조직화학적 절편을 수행하였다. 샘플은 Leica CM1950(독일 라이카 마이크로시스템즈)을 사용하여 20μm 두께로 절편화하고, 항체로 염색할 때까지 보존액에 담아 4℃에서 보관하였다.
TH(티로신 하이드록실라제)는 아미노산인 L-티로신을 L-3,4-디하이드록시 페닐알라닌(L-DOPA)으로 전환하는 촉매 효소로서 도파민(DA)을 포함한 카테콜아민 생합성에서 가장 중요한 요소 중 하나이다. TH의 발현이나 활성의 변화는 DA 생성에 큰 영향을 미치므로 파킨슨병의 발병 기전에서 매우 중요하다.
뇌의 선조체와 흑질을 염색하고 현미경 슬라이드에 장착한 후, 슬라이드를 0.3% 트리톤 X-100과 퍼옥시다아제 블록을 함유한 PBS로 실온에서 15분 동안 세척하였다. 슬라이드를 0.3% 트리톤 X-100을 함유한 PBS로 3회 세척한 후 PBS, 5% 정상 염소 혈청 (normal goat serum) 및 0.3% 트리톤 X-100을 함유한 용액에서 1시간 동안 RT(실온)에서 블로킹하였다. 티로신 하이드록실라제 항체를 1:1000으로 희석하고 4℃에서 하룻밤동안 배양(incubation) 하였다. 슬라이드를 PBS 용액으로 세척하고 2차 항체와 RT에서 1시간 동안 배양하였다. 슬라이드를 PBS 용액으로 세척하고 섹션을 용액에 넣어 DAB(디아미노벤지딘) 반응을 진행하였다.
현미경으로 2분 내지 3분 동안 DAB 반응을 모니터링하고 DAB 반응을 멈추기 위해 슬라이드를 PBS로 세척하고 탈수한 후 자일렌을 사용하여 마운팅하였다. 올림푸스 현미경(Olympus, SZ61, BX51)을 사용하여 TH 염색 이미지를 시각화하였다. 양성 TH 염색의 OD(optical density) 값을 선조체와 흑질에서 측정하고 Image J 소프트웨어를 사용하여 분석하였다.
도 5a 및 도 5b는 선조체에 대한 TH 염색 면역조직화학 분석 결과이며, 도 6a 및 도 6b는 흑질에 대한 TH 염색 면역조직화학 분석 결과이다. 선조체 및 흑질에서 MPTP를 처리한 그룹(G2)의 TH 수준은 대조군(G1)에 비해 유의하게 낮게 나타났다. 닐로티닙을 처리한 그룹(G3)의 선조체 TH 수준은 G2에 비해 유의하게 높게 나타났다. 실시예3 또는 실시예2을 처리한 그룹은 G2에 비해 선조체와 흑질 모두에서 TH 수준이 유의하게 증가하였다. 또한, G5 (MPTP + 10 mg/kg 실시예3) 및 G6 (MPTP + 50 mg/kg 실시예3)에서 선조체의 TH 수준과 G5, G6 및 G7 (MPTP + 50 mg/kg 실시예2)에서 흑질의 TH 수치는 대조군(G1)에 비해 유의하게 높게 나타났다.
실험예 9.6. 도파민 증가 효능 평가
적출한 뇌조직의 선조체와 흑질에 대하여 Agilent 1290 (LC)와 SCIEX QTARP 6500 + (QQQ)를 이용하여 LC-MS/MS 분석을 통해 도파민 양을 측정하였다.
도 7은 MPTP-유도 PD 마우스 모델의 선조체와 흑질에서의 그룹별 도파민 양 그래프이다. 도 7을 참고하면, 실시예2 50 mg/kg 투여군에서는 선조체와 흑질 모두에서, 실시예3 2 및 10 mg/kg 투여군에서는 흑질에서 MPTP 단독 투여군 대비 도파민 농도가 유의하게 증가하였다. 이를 통해, 실시예 화합물이 도파민 양을 유의미하게 증가시키는 것이 확인되었다.
실험예 10. α-Syn 형질전환 파킨슨병 마우스 모델에 대한 효능 평가
실험예 10.1. 실험 준비
미국 샌디에이고 UCSD(University of California San Diego)에서 획득한 mThy-1α-syn 형질전환 (α-syn TG; Line 61번) 암컷과 C57BI/6 X DBA/2 F1 WT (야생형)수컷을 교배하여 오송첨단의료산업진흥재단 (KBIOHealth) 동물센터에서 사육하였다.
모든 마우스는 온도(22±1℃)와 습도(30~50%)가 조절된 환경에서 일반적인 명암 주기(08:00~20:00)로 사육했으며 음식과 물을 무제한 (Ad libitum)으로 제공하였다. 생후 15일령 내지 21일령에 귀 표식을 하고 귀 또는 꼬리 샘플을 채취하여 유전자형 분석을 위한 PCR(polymerase chain reaction) 분석을 실시하였다. 또한 필요한 경우 유전자형 확인을 위해 종료 지점에서 꼬리 샘플을 수집하였다.
정상 마우스 및 α-Syn(알파-시누클레인) 형질전환 파킨슨병 마우스 모델에 대하여 시험 약물을 투여하고 효능을 평가하였다. 실시예의 화합물에 대하여 10% NMP, 40% PEG 400 및 50% D.W.를 전달체로 사용하여 시험 약물을 준비하였다.
실험예 10.2. 실험 설계 및 마우스 체중 변화
도 8은 α-Syn 형질전환 파킨슨병 마우스 모델에 대한 효능 실험의 모식도다.
생후 4개월 령의 마우스를 7개 그룹으로 아래 표 10과 같이 나누었다. 생후 4개월부터 생후 9개월까지 5개월 동안 5 mL/kg의 용액으로 경구 투여(PO)를 통해 1일 1회(QD) 시험 약물을 투여하였다. 운동 행동 테스트(빔 테스트) 및 신경학적 점수(modified Irwin test)는 4개월(투여 전), 7개월 및 9개월에 실시하였다. 최종 행동 테스트 후 조직 샘플을 채취하였다. 생후 4개월부터 연구가 끝날 때까지 일주일에 한 번 동일 요일에 체중을 측정하였다. 전반적인 건강 상태가 현저히 악화된 마우스는 연구에서 제외하였다.

도 9는 α-Syn 형질전환 파킨슨병 마우스 모델의 그룹별 체중 변화에 대한 그래프이다. 도 9를 참고하면, WT 마우스(G1)의 체중은 시간이 지남에 따라 증가한 반면, TG 그룹 (G2-7)의 체중 증가는 정체된 것이 확인되었다. WT 마우스와 TG 마우스 간의 체중 차이는 생후 약 21주 내지 22주 이후부터 뚜렷해졌으며, 약물투여는 TG 마우스의 체중에 큰 영향을 미치지 않는 것으로 확인되었다.
실험예 10.3. 신경학적 행동 개선 효과
생후 4개월 (투여 전, 기준선), 7개월 및 9개월에 마우스당 1분 내지 2분 동안 아래 표 11에 열거된 신경학적 장애 행동을 관찰하였다. 깜짝 놀람 반사 (startle reflex), 꼬리 꼬집기 반사 (tail pinch) 및 정위 반사 (righting reflex)를 제외한 신경학적 행동 장애에 대해 케이지에 있는 마우스를 직접 관찰하여 점수를 기록하였다. 깜짝 놀람 반사를 테스트하기 위해 작은 리모콘을 사용하여 큰 소리를 내고 점프, 정지, 빠른 눈 깜빡임 등의 행동을 확인하였다. 정위 반사의 경우, 각 마우스를 케이지에서 꺼내서 등이 바닥을 향하도록 놓고 마우스가 스스로 자세를 바로잡을 수 있도록 하였다. 꼬리 꼬집기 반사는 집게로 꼬리 끝을 부드럽게 쥐는 방법으로 테스트하였다.
운동 활동과 같은 정상적인 특징이 있거나 이상 특징이 없는 경우 0점, 경미한 이상이 관찰되면 1점, 심각한 이상이 관찰되면 2점 내지 3점을 부여하였다. 각 마우스에 대해 관찰된 모든 행동에 대한 점수를 더하고, 각 그룹에 대한 추가된 점수의 평균을 계산하였다.

도 10은 α-Syn 형질전환 파킨슨병 마우스 모델의 그룹별 신경학적 행동 점수에 대한 그래프이다. 생후 4개월과 7개월에 G2의 신경학적 점수 (TG + 전달체)는 G1에 비해 유의하게 낮게 나타났다. 실시예3 또는 실시예2를 투여한 군은 G2에 비해 TG 마우스의 신경학적 점수가 유의하게 감소하였다. 그룹 G4 내지 G7의 신경학적 점수는 연구 기간 내내 전반적으로 매우 낮게 유지되었으며, 약물에 의한 신경학적 독성이 없는 것이 확인되었다.
실험예 10.4. 걸음 오류 개선 효능 평가(빔 테스트)
빔 테스트(Beam test, Challenging Beam Traversal)로 플레밍 등이 설명한 절차(J Neurosci, 24:9434-40, 2004)를 채택하였다. 운동 능력은 기존의 빔 워킹 테스트를 변형한 새로운 빔 테스트로 측정하였다. 빔은 플렉시글라스 (Plexiglas)로 제작되었으며, 각 섹션의 폭이 다른, 4개의 섹션(각 25 cm, 총 길이 1 m)으로 구성하였다. 빔은 3.5cm 너비에서 시작하여 0.5 cm × 1 cm 단위로 점차 좁아지도록 설계하였다. 빔의 상단 표면에서 1.0 cm 아래에 폭 1 cm의 선반을 배치하였다.
마우스를 가장 넓은 구간에서 시작하여 가장 좁고 어려운 구간까지 빔의 길이를 가로지르도록 훈련시켰다. 빔의 좁은 끝은 마우스의 집 케이지로 바로 연결되도록 설계하였다. 마우스를 테스트 전에 2일간 훈련시켰으며, 모든 훈련은 메쉬 그리드 없이 진행하였다. 첫날에는 마우스를 빔에 올려놓고 홈 케이지를 마우스와 가까운 곳에 배치하는 두 번의 보조 훈련을 수행하였다. 이를 통해 빔을 따라 앞으로 움직이도록 유도하였다. 두 번의 보조 훈련 후, 마우스가 빔의 전체 길이를 도움 없이 횡단할 수 있었다. 훈련 1일차에는 모든 마우스가 빔의 전체 길이에 걸쳐 5번의 보조 없이 달리기를 완료한 것을 확인하였다. 훈련 2일차에는 마우스가 5번의 시험을 수행하였다. 시험 당일에는 난이도를 높이기 위해 빔 표면 위에 해당 폭의 메쉬 격자(1 cm 정사각형)를 배치하고 격자와 빔 표면 사이에 약 1cm의 공간을 두었다. 아래 선반은 사지가 그리드에서 미끄러질 때 마우스가 사용할 수 있는 지지대로 제공했으며, 마우스가 테스트를 완료하기 위해 보상 운동 전략을 사용할 필요없이 평가할 수 있도록 하였다. 총 5회에 걸쳐 격자무늬 빔을 가로지르는 마우스의 모습을 비디오로 촬영하였다.
비디오 테이프 분석은 마우스 유전자형/그룹을 모르는 연구자가 오류에 대한 슬로모션을 확인하고, 각 마우스의 걸음 수, 5번의 실험에 걸친 횡단 시간을 평가하였다. 오류는 앞으로 이동하는 동안 사지(앞다리 또는 뒷다리)가 그리드를 통과하여 그리드와 빔 표면 사이에 보이는 경우 계수하였다. 개별 마우스는 한 걸음당 최대 4번까지 미끄러질 수 있었다. 각 사지 미끄러짐을 개별적으로 점수화하여 오류를 측정하였다. 예를 들어, 한 걸음에서 세 개의 팔다리가 그리드에서 미끄러진 마우스는 오류 점수 3점으로 평가하고, 한 걸음에서 한 개의 팔다리만 그리드에서 미끄러진 마우스는 해당 걸음에서 오류 점수 1점으로 평가하였다. 마우스가 앞으로 움직이지 않거나 마우스의 머리가 빔의 왼쪽이나 오른쪽을 향하는 경우 미끄러짐으로 계수하지 않았다. 걸음 오류의 수와 횡단하는 데 걸린 시간은 다섯 번의 실험 모두에서 측정하여 평균을 내었다.
도 11은 α-Syn 형질전환 파킨슨병 마우스 모델에서 그룹별 걸음 오류 평가 결과를 나타내는 그래프이다. 도 11을 참고하면, 그룹 G2는 빔 테스트에서 그룹 G1에 비해 횡단하는 동안 훨씬 더 많은 오류를 범하였다. 메만틴(G3), 실시예3(G4 내지 G6) 또는 실시예2(G7) 투여군은 7개월 및 9개월에 그룹 G2에 비해 걸음 오류 수가 현저히 감소하였다. 이를 통해, 실시예 화합물이 행동 오류를 개선시키는 것이 확인되었다.
실험예 10.5. 도파민성 신경퇴화 개선 효능 평가
생후 9개월에 측정 기간이 끝나고 마우스를 마취한 다음 IACUC 지침에 따라 심장 천자를 통해 실험을 종료하였다. 조직 염색을 위해 마우스에 0.9% 식염수를 경심 관류한 다음 4% 파라포름알데히드로 마우스가 고정될 때까지 관류하였다. 뇌를 분리하여 4% 파라포름알데히드에 4℃에서 24시간 동안 담가 고정시킨 다음, 0.02% 소듐 아자이드를 함유한 PBS에 30% 자당을 넣고 4℃에서 3일 동안 보관하였다. OCT 화합물에 내장된 뇌 샘플을 라이카 CM1950 (독일 라이카 마이크로시스템즈)을 사용하여 20μm 두께로 절편화하였다. 절편은 항체에 의해 염색될 때까지 보존액에 담아 4℃에서 보관하였다.
뇌 조직은 조직을 부드럽게 흔들면서 PBS로 10분씩 3회 세척하였다. 그 후 조직을 PBS, 10% 정상 염소 혈청 및 0.3% 트리톤 X-100을 함유한 차단 완충액에서 실온에서 1시간 동안 반응시켰다. 조직을 아래 표의 항체 및 희석률에 따라 1차 항체 (Iba-1, TH)와 함께 4℃에서 하룻밤 배양하였다. 1차 항체 배양 후, 조직을 PBS 용액으로 세척하고 실온에서 1시간 동안 2차 항체와 반응시켰다. 그 후, 섹션을 PBS 용액으로 10분씩 3회 세척하고 슬라이드와 커버슬립에 DAPI 마운팅 용액 (VECTASHIELD, #H1500)를 사용하여 마운팅하였다.

도 12a 및 도 12b는 α-Syn 형질전환 파킨슨병 마우스 모델의 그룹별 도파민성 신경퇴화 개선 효능 평가 결과를 나타내는 그래프 및 면역형광분석 이미지이다. 도 12a 및 도 12b를 참고하면, 그룹 G2 선조체의 TH 염색 강도는 그룹 G1에 비해 낮은 경향을 보였다. 메만틴이 아닌 실시예3 또는 실시예2 투여군(그룹 G4 내지 G7)의 TH 염색 강도는 그룹 G2에 비해 유의하게 높게 나타났다. 그룹 G7 (TG + 실시예2)의 TH 염색 강도 또한 그룹 G2에 비해 유의하게 높게 나타났다. 이로써, 실시예 화합물이 도파민성 신경퇴화를 유의미하게 개선시킬 수 있는 것이 확인되었다.
실험예 10.6. 미세아교세포 감소 효능 평가
피질의 미세아교세포증을 평가하기 위해 올림푸스의 면역 형광 현미경(Olympus, BZ21)을 사용하여 실험예 10.5.에서 Iba-1 염색된 뇌 조직을 이미지화하였다. Iba1 정량화를 하기 위해 각 마커를 수동으로 카운트하였다.
도 13a 및 도 13b는 α-Syn 형질전환 파킨슨병 마우스 모델의 그룹별 미세아교세포 감소 효능 평가 결과를 나타내는 그래프 및 면역형광분석 이미지이다. 도 13a 및 도 13b를 참고하면 대뇌 피질에서 Iba-1 양성 세포의 수는 메만틴(G3), 실시예3 2 mg/kg 및 10 mg/kg (G4 및 G5) 및 실시예2 2 mg/kg (G7) 투여 그룹에서 유의하게 감소하였다. 이를 통해, 실시예 화합물이 미세아교세포를 유의미하게 감소시키는 것이 확인되었다.
실험예 11. SOD1 G93A 형질전환 근위축성 측삭경화증 마우스 모델에 대한 효능 평가
실험예 11.1. 실험 준비
잭슨 연구소에서의 G93A SOD1 (high-copy SOD1-G93A, 25 TG 카피의 G1H, 재고번호 002726) 수컷과 B6SJL 균주 배경 (non-carrier, WT) 암컷을 교배하여 실험용 F1 마우스를 생성하였다. 모든 마우스는 온도(22±1℃)와 습도(30~50%)가 조절된 환경에서 일반적인 명암 주기(08:00~20:00)로 사육했으며 음식과 물을 무제한(Ad libitum)으로 제공하였다. 생후 15~21일령에 귀 표식을 하고 귀 또는 꼬리 샘플을 채취하여 유전자형 분석을 위한 PCR 분석을 실시하였다. 또한 필요한 경우 유전자형 확인을 위해 종료 지점에서 꼬리 샘플을 수집하였다.
정상 마우스 및 SOD1 G93A 형질전환 근위축성 측삭경화증(ALS: Amyotrophic Lateral Sclerosis) 마우스 모델에 대하여 시험 약물을 투여하고 효능을 평가하였다. 실시예의 화합물에 대하여 10% NMP, 40% PEG 400 및 50% D.W.를 전달체로 사용하여 시험 약물을 준비하였다.
실험예 11.2. 실험 설계
도 14는 SOD1 G93A 형질전환 ALS 마우스 모델에 대한 효능 실험의 모식도다.
생후 8주령의 마우스를 아래 표 13의 3개 그룹으로 나누었다. 생후 8주령에서 사망할 때까지 5 mL/kg의 용액으로 경구 투여(PO)를 통해 1일 1회(QD) 시험 약물을 투여하였다. 체중은 매주 측정하고, 질병 발병(disease score)는 8주부터 동물이 사망할 때까지 매일 측정하였다. 운동 행동 테스트(악력, grip strength) 및 신경학적 점수 테스트(modified Irwin test)는 9, 11, 13, 15, 17, 19 및 21주에 각각 실시하였다. 마우스의 생존 여부는 매일 모니터하였다. 마우스의 전반적인 건강 상태가 현저히 악화된 경우, 연구에서 제외시켰다.

도 15는 SOD1 G93A 형질전환 ALS 마우스 모델의 그룹별 체중 변화에 대한 그래프이다. 도 15를 참고하면, Tg 그룹(G2)의 체중은 WT 그룹(G1)보다 현저히 낮게 나타났다. 반면 TG (G2)와 약물 처리군 (G3)간의 유의미한 차이는 없었으며, 실시예 화합물이 체중을 유의미하게 감소시키지 않는 것이 확인되었다.
실험예 11.3. 신경학적 행동 개선 효능 평가
실험예 10.3.과 동일한 방법으로 생후 9, 11, 13, 15, 17, 19 및 21주령에 마우스 당 1분 내지 2분 동안 신경학적 장애 행동을 관찰하였다.
도 16은 SOD1 G93A 형질전환 ALS 동물 모델의 그룹별 신경학적 점수에 대한 그래프이다. 도 16을 참고하면, WT 그룹 (G1)의 신경학적 점수는 연구 기간 내내 1 미만으로 매우 낮게 나타났다. 13주 이후부터 TG 그룹의 신경학적 점수는 G1 그룹보다 유의하게 높게 나타났다. 생후 19주째 이후부터 실시예3을 처리한 그룹(G3)은 G2 대비 신경학적 점수가 유의하게 감소하였다. 이를 통해, 실시예 화합물이 유의미하게 신경학적 행동을 개선시키는 것이 확인되었다.
실험예 11.4. 질병 증상 개선 효능 평가
생후 8주부터 ALSTDI(ALS Therapy Development Institute) 방법으로 개발된 신경학적 점수 시스템을 사용하여 마우스의 질병 증상을 평가하였다. 양쪽 뒷다리에 대하여 아래 표 14의 기준으로 채점하였다.

도 17은 SOD1 G93A 형질전환 ALS 마우스 모델의 그룹별 질병 점수 변화에 대한 그래프이다. 도 17을 참고하면, TG 그룹(G2 및 G3)의 질병 발병 점수는 WT 그룹(G1)보다 유의하게 증가하였으며 대략 15주령에 차이가 두드러지게 나타났다. 실시예3을 처리한 그룹(G3)은 G2 대비 질병 증상 점수가 관찰 기간동안 낮은 경향을 보였으며, 20주에 유의하게 감소하였다. 이를 통해, 실시예 화합물이 유의미하게 질병 증상을 개선시키는 것을 확인하였다.
실험예 11.5. 악력 개선 효능 평가
생후 9, 11, 13, 15, 17, 19 및 21주령의 마우스를 대상으로 악력 측정을 실시하였다. 마우스가 앞발로 작은 그물망 손잡이를 잡는 방식으로 마우스를 악력 측정 장치 (미국 샌디에이고, 샌디에이고 Instruments)에 올려놓았다. 마우스를 플랫폼으로 내려놓은 다음 마우스가 손잡이를 놓을 때까지 꼬리로 손잡이를 천천히 잡아당겼다. 장비는 자동으로 마우스의 악력 강도를 그램 단위로 측정하였다. 하루 세션에 마우스당 5개의 점수를 연속적으로 기록하였다.
도 18은 SOD1 G93A 형질전환 ALS 마우스 모델의 그룹별 악력 변화에 대한 그래프이다. 도 18을참고하면, TG 그룹의 악력은 일반적으로 시간이 지남에 따라 감소했으며 그룹 G1보다 유의하게 낮게 나타났다. 실시예3을 처리한 그룹(G3)은 그룹 G2 대비 악력이 증가하는 경향을 보이고 19주부터 유의하게 악력이 증가하였다. 이로써, 실시예 화합물이 유의미하게 악력을 개선시키는 것이 확인되었다.
실험예 11.1. 생존율 개선 효능 평가
매일 마우스의 생존 여부를 모니터링하였다. 마우스의 생존 연령을 기록하여 카플란 마이어 생존 플롯으로 표시하였다(Kaplan Meier survival plots).
도 19는 SOD1 G93A 형질전환 ALS 마우스 모델의 그룹별 생존율에 대한 그래프이다. 도 19를 참고하면, TG 그룹의 생존율은 생후 약 110일 내지 120일 사이부터 감소하기 시작하였으며, 실시예3을 처리한 그룹 (G3)은 그룹 G2 대비 유의성은 없으나 생존율이 개선되는 경향을 보였다. 이로써, 실시예 화합물이 생존율을 개선시키는 것이 확인되었다.
실험예 12. Reserpine 유도 섬유근육통 랫트 모델에 대한 효능 평가
실험예 12.1. 실험 준비
6주령 Sprague Dawley(SD) 수컷 랫트(rat)를 사용하였다. 모든 랫트는 온도(22±1℃)와 습도(30~50%)가 조절된 환경에서 일반적인 명암 주기(08:00~20:00)로 사육했으며 음식과 물을 무제한(Ad libitum)으로 제공하였다.
정상 랫트 및 Reserpine 유도 섬유근육통(Fibromyalgia) 랫트 모델에 대하여 시험 약물을 투여하고 효능을 평가하였다. 실시예의 화합물에 대하여 10% NMP, 40% PEG 400 및 50% D.W.를 전달체로 사용하여 시험 약물을 준비하였다.
실험예 12.2. 실험 설계
도 20은 Reserpine 유도 섬유근육통 랫트 모델에 대한 효능 실험의 모식도다.
Reserpine 유도 섬유근육통 모델 생성 및 약물 투여는 생후 6주령 수컷 SD 랫트를 아래 표 15와 같이 6개군으로 나누어 진행하였다: 대조군(G1), reserpine + 전달체(G2), reserpine + 가바펜틴(Gabapentin) 50 mg/kg(G3), reserpine + 실시예2 3 mg/kg(G4), 10 mg/kg(G5), 및 30 mg/kg(G6). 랫트 섬유근육통 모델을 만들기 위해, 0.5% 아세트산에 녹인 reserpine(Sigma-aldrich, 1mg/kg)을 실험 시작 3일 전부터 3일 동안 피하로 1회씩 투여하였다. 1일차부터 1일 1회(QD) 전달체, gabapentin 및 실시예2을 실험수행 2시간 전에 복강(IP) 또는 경구(PO) 투여하였다.

실험예 12.3. 통증 개선 효능 평가
오른쪽 뒷발 촉각 반응 테스트 (von Frey test)는 5회 (섬유근육통 모델 생성 전, 모델 생성 후 1일차, 2일차, 4일차 및 6일차) 실시하였다. 모든 행동 테스트 전에 30분 동안 랫트에게 환경에 익숙해지도록 하였고, 행동 테스트를 수행하는 모든 실험자는 약물 투여 정보를 알지 못하도록 진행하였다. 모델 생성 전에 모든 랫트에 대해 von Frey 테스트를 실시하여 기준선 측정을 수행하였다. 랫트를 유선 매쉬 위에 올려놓고 투명한 플라스틱 상자 (20 x 20 x 14cm)에 넣고, 기계적 이질통을 평가하기 위해 오른쪽 뒷발 중앙에 von Frey 필라멘트 (오름차순 무게: 0.4, 0.6, 1.4, 2, 4, 6, 8 및 15g, North Coast)를 적용하여 발 인출 역치를 측정하였다.
도 21은 Reserpine 유도 섬유근육통 랫트 모델에 대한 통증 개선 효능 평가 결과 그래프이다. 대조군 (G1)에 비해 reserpine 처리 그룹(G2)은 높은 민감도를 보였으며, 모델 생성 후 가바펜틴 처리 그룹(G3)과 실시예2 처리 그룹 (G4 내지 G6)은 시간이 지나면서 민감도가 점진적으로 개선된 것이 확인되었다. 실시예2은 용량 의존적으로 회복되는 경향을 보였으며, 30mg/kg에서 유의성 있는 효능을 나타내었다. 이를 통해, 실시예의 화합물이 유의미하게 통증을 개선시키는 것이 확인되었다.
실험예 13. CDAHFD 유도 비알코올성 지방간염 마우스 모델에 대한 효능 평가
실험예 13.1. 실험 준비
생후 3주령의 C57BL/6 수컷 마우스(Koatech, 한국)를 구입하였다. 마우스는 케이지당 최대 5마리씩 그룹을 지어 온도 (22±1℃)와 습도 (30~50%)가 조절된 환경에서 일반적인 명암 주기(08:00~20:00)로 사육했으며 음식과 물을 무제한 (Ad libitum)으로 제공하였다. 실험 시작 전 1주일 동안 마우스가 적응할 수 있도록 하였다.
정상 마우스 및 CDAHFD(A choline-deficient, L-amino acid-defined, high-fat diet: 콜린 결핍, L-아미노산 정의 고지방 식단)-유도 비알코올성 지방간염(NASH) 마우스 모델에 대하여 시험 약물을 투여하고 효능을 평가하였다. 실시예의 화합물에 대하여 10% NMP, 40% PEG 400 및 50% D.W.를 전달체로 사용하여 시험 약물을 준비하였다.
CDAHFD는 0.1% 메티오닌(methionine)과 60 kcal% 지방을 포함하고, 콜린(choline)이 결핍된 고지방 식단으로 제조하였다. CDAHFD를 먹인 C57BL/6J 마우스 모델은 짧은 기간에 섬유화를 동반한 지방간염이 빠르게 진행된다.
실험예 13.2. 실험 설계
도 22는 CDAHFD 유도 비알코올성 지방간염 마우스 모델에 대한 효능 실험의 모식도다.
CDAHFD 유도 NASH 모델 생성 및 약물 투여는 생후 3주령 C57BL/6 수컷 마우스를 아래 표 16과 같이 7개군으로 나누어 진행하였다. 대조군 (G1)은 표준 사료와 물을 먹였다. 그룹 G2 내지 그룹 G7은 CDAHFD (Research Diet Inc, #A06071302) 식이를 4주와 8주 동안 먹였다. 대조군을 제외한 각 그룹은 5주령부터 1일 1회씩 (QD) 전달체, 셀론설팁 30 mg/kg, 실시예3 10 mg/kg, 실시예3 30 mg/kg, 실시예2 10 mg/kg 및 실시예2 30 mg/kg을 경구 투여하였다.

실험예 13.3. 간 섬유화 개선 효능 평가
콜라겐 섬유의 조직학적 시각화를 위해 피크로시리우스 레드 염색(Picrosirius red staining)을 이용하였다. 조직병리학 분석을 통해 간 섬유화 개선 효능을 평가하였다.
각 군의 마우스에 대하여 4% PFA(파라포름알데히드)로 고정된 간 세 절편을 파라핀 블록으로 제작하였다. 파라핀 블록은 5μm 두께로 절편화하고(Histocore AUTOCUT R, 라이카 마이크로시스템즈, 독일), 섹션을 실란 코팅 슬라이드(MUTO, JPN)에 장착 후 염색할 때까지 보관하였다.
자일렌을 사용하여 슬라이드에서 파라핀을 제거하고 슬라이드에 수분을 공급하고 세척하였다. 슬라이드를 피크로시리우스 레드 용액 (ab150681, Abcam, 영국)으로 염색하고 아세트산 용액으로 씻어내었다. 슬라이드는 알코올과 자일렌을 사용하여 탈수 및 세척 후 장착하였다. 각 섹션에 대해 현미경(Axioscan7, 자이스, 독일)을 사용하여 서로 다른 영역의 이미지를 2장씩 촬영하였다. 섬유화 면적은 image J 소프트웨어를 사용하여 전체 면적 대비 섬유화 면적을 측정하였다.
도 23a 및 도 23b는 CDAHFD 유도 비알코올성 지방간염 마우스 모델의 간에서의 Picrosirius red 염색 면적비에 대한 그래프 및 이미지이다. 도 23a 및 도 23b를 참고하면, 그룹 G1에 비해 CDAHFD 군은 Picrosirius red 로 염색된 상대적 면적이 유의하게 높아 NASH 모델이 적절하게 형성된 것을 나타내었다. 비교약물 셀론설팁(G3)과 실시예3(G4 및 G5), 실시예2(G6 및 G7)에서 Picrosirius red 염색 면적이 유의하게 감소하였으며, CDAHFD 식이 4주 후, 실시예2 30mg/kg 투여군에서 가장 개선된 효능을 확인하였다. 이로써 실시예 화합물이 유의미하게 간 섬유화를 개선시키는 것이 확인되었다.

    상기 신경퇴행성 질환은 알츠하이머병, 해마 경화증, 전두측두엽 치매 (FTD), 전두측두엽 변성 (FTLD), 헌팅턴병, 피질기저핵 변성, 근위축성 측삭경화증, 척수성 근위축, 운동 뉴런 질환, 봉입체 근염, 파킨슨병, 루이소체 치매, 루이소체 질환, 다계통 위축, 진행성 핵상 마비, 피크병, 프라이온병, 외상성 뇌 손상, 허혈성 및 출혈성 뇌졸중, 대뇌 허혈, 저산소증, 및 글루타메이트 신경독성으로 이루어진 군으로부터 선택되는 1종 이상인 것을 특징으로 하는, 약학적 조성물.
  12. 제10항에 있어서,
    상기 심혈관 질환은 심부전, 허혈, 재발성 허혈, 심근경색, 부정맥, 급성 관상동맥 증후군, 당뇨병, 죽상경화증 및 간헐적 파행으로 이루어진 군으로부터 선택되는 1종 이상인 것을 특징으로 하는, 약학적 조성물.
  13. 제10항에 있어서,
    상기 자가면역 질환은 류마티스 관절염, 섬유근육통, 전신홍반루푸스, 다발성 경화증, 당뇨병, 전신경화증, 그레이브스병, 길랑-바레 증후군, 중증근무력증, 건선, 크론병, 궤양성 대장염, 시신경염, 및 쇼그렌 증후군으로 이루어진 군으로부터 선택되는 1종 이상인 것을 특징으로 하는, 약학적 조성물.
  14. 제10항에 있어서,
    상기 간 질환은 비알코올성 지방간(NASH), 알코올성 지방간, 간 섬유화, 간암, 간독성, 담즙울체, 간경변, 간허혈, 간 농양, 간성 혼수, 및 간 위축증으로 이루어진 군으로부터 선택되는 1종 이상인 것을 특징으로 하는, 약학적 조성물.
  15. 제1항 내지 제6항 중 어느 한 항의 화합물, 또는 이의 용매화물, 입체이성질체 또는 약학적으로 허용가능한 염을 개체에게 투여하는 단계를 포함하는 ASK1 관련 질환의 예방 또는 치료 방법.
  16. ASK1 관련 질환의 예방 또는 치료에 사용하기 위한 제1항 내지 제6항 중 어느 한 항에 따른 화합물, 이의 용매화물, 입체이성질체 또는 약학적으로 허용가능한 염의 용도.
  17. ASK1 관련 질환의 예방 또는 치료용 의약을 제조하기 위한 제1항 내지 제6항 중 어느 한 항에 따른 화합물, 이의 용매화물, 입체이성질체 또는 약학적으로 허용가능한 염의 용도.
