Abstract

内部に気泡が残らない自動分析装置などを提供する。本発明に係る自動分析装置(1)は、液体を吸引・吐出するノズル(サンプル分注ノズル(10)、試薬分注ノズル(11)、分注ノズル(102)、分注ノズル(115))と、該ノズルに接続され液体を吸引・吐出させるための圧力を発生させる圧力発生手段(給水ポンプ(111))と、前記ノズルおよび前記圧力発生手段を含めた流路(104)内の圧力を測定する圧力センサユニット(112)と、を備えた分注機構(100)を有し、前記圧力センサユニット(112)の内面(203a)に、シリカ粒子およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含んでなる親水膜(305)が形成されており、前記内面(203a)の基材(301)の算術平均粗さRa0と、前記親水膜(305)形成後の内面(203a)の算術平均粗さRa1との関係が、0.8×Ra0≦Ra1である。

Description

自動分析装置、自動分析方法および圧力センサユニットの製膜方法
 本発明は、自動分析装置、自動分析方法および圧力センサユニットの製膜方法に関する。
 血液をはじめとする体内成分を分析し、生体成分の異常値を見つけたり、病気の有無を判断可能な生体成分の分析を行ったりする自動分析装置は、病院や多くの医療研究機関で使用されている。
 自動分析装置内には、液体の試料が入っているサンプル容器や試薬が入っている試薬容器から、試料や試薬を適切な割合で混合するための反応容器へ分注と称する吸引・吐出操作により投入するための分注機構が備えられている。
 分注機構は極めて高い分注精度が求められるため、分注機構内のシリンジ、ダイアフラムなどの圧力発生手段で発生させた圧力を正確に伝達すべく、通常は分注流路内が精製水などの液体(以下、システム水と記す)で充填されている。また、分注機構は、その高い分注精度での分注を具現するため、圧力センサユニットを備えている。
 圧力センサユニットは、ダイアフラムで圧力を計測するセンサ部分と、システム水をダイアフラムの受圧面から分注ノズル方向およびシリンジ方向に分岐する接続容器とからなる。圧力センサユニットはおおよそ-100~100kPa程度の範囲の圧力を高感度で測定するため、その先端に付いているダイアフラムの直径は大きくする必要がある。ダイアフラムの直径は具体的には概ね10~15mmである。また、圧力センサユニットの接続容器につながっている配管(流路)の直径は数mmであり、ダイアフラムに比べて小さい。
 そのため、圧力センサユニットの接続容器内で流路の直径が数mmから10~15mmに大きく拡大すると、その部分のシステム水の流れが乱れて流速が減少する。これにより、流路内の気泡が接続容器内や圧力センサユニットの内部に付着しやすくなる。
 付着した気泡は分注操作を行う際、圧力センサユニットにかかる圧力が上昇した場合に圧力の一部が気泡を縮ませるため、正確な圧力計測ができなくなる。そのため、正確な量の分注を行うことができなくなる。
 そこで、圧力センサユニットの接続容器に2か所ある液体の流入出口の内の1か所をダイアフラム近傍に設け、システム水が接続容器内に流入する際にダイアフラム表面近傍で渦を巻くように動くようにすることで気泡を除去する構造にする方法が特許文献1に記されている。
 この構造を採ることにより、圧力センサユニット内部の気泡はある程度除去される。

 しかしながら、特許文献1の技術を適用しても圧力センサユニットの構造に由来して内部には気泡が残り、これが圧力の測定精度を下げてしまう場合がある。このことから現状、構造を改良することが望ましいものの、一般品を活用するうえでは構造変更が難しい場合が多い。
 本発明は前記状況に鑑みてなされたものである。本発明の目的は、内部に気泡が残らない自動分析装置、自動分析方法および圧力センサユニットの製膜方法を提供することにある。
 本発明者らは、前記課題を解決するために鋭意研究開発した結果、圧力センサユニットの内面に水との接触角が15°以下の親水膜を形成することにより、前記課題を解決できることを見出し、本発明を完成するに至った。
 前記課題を解決した本発明に係る自動分析装置は、液体を吸引・吐出するノズルと、該ノズルに接続され液体を吸引・吐出させるための圧力を発生させる圧力発生手段と、前記ノズルおよび前記圧力発生手段を含めた流路内の圧力を測定する圧力センサユニットと、を備えた分注機構を有し、前記圧力センサユニットの内面に、シリカ粒子およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含んでなる親水膜が形成されており、前記内面の基材の算術平均粗さRa0と、前記親水膜形成後の内面の算術平均粗さRa1との関係が、0.8×Ra0≦Ra1であること、としている。
 本発明によれば、内部に気泡が残らない自動分析装置、自動分析方法および圧力センサユニットの製膜方法を提供することができる。
 前述した以外の課題、構成および効果は以下の実施形態の説明により明らかにされる。

 以下、適宜図面を参照して、本発明の一実施形態に係る自動分析装置、自動分析方法および圧力センサユニットの製膜方法について詳細に説明する。
 (1)自動分析装置
 図1は、本発明の一実施形態に係る自動分析装置の全体構成の一例を示した説明図である。図2は、本発明の一実施形態に係る自動分析装置の全体構成の他の一例を示した説明図である。
 図1に示すように、自動分析装置1は、検体のサンプルと試薬を反応させて検体中の特定の成分を分析するものである。自動分析装置1の概略は、試薬を充填している試薬容器2が装てんされる試薬容器装てん部3と、試薬容器装てん部3を内包する試薬保冷庫4と、サンプルが保持されているサンプル容器5が架設されたラック6の搬送ラインであるラック搬送ライン7とを有した構成である。
 また、自動分析装置1には、多くの反応容器8を設置可能な円盤状のインキュベーターディスク9と、サンプルを分注するノズルであるサンプル分注ノズル10と、試薬を分注するノズルである試薬分注ノズル11と、反応容器8を搬送する機構である反応容器搬送機構12とが設けられている。
 インキュベーターディスク9には、前述のように複数の反応容器8が設置可能である。インキュベーターディスク9は、円周方向に設置された反応容器8を回転によりそれぞれ所定の位置まで移動が可能なように取り付けられている。
 自動分析装置1では、ラック6には前述のようにサンプルを保持するサンプル容器5が架設されており、ラック搬送ライン7によってサンプル分注ノズル10の近傍のサンプル分注位置13までサンプル容器5を移動させる。
 インキュベーターディスク9の近傍には、サンプル分注チップ/反応容器搬送機構14がX軸、Y軸、Z軸の3方向に移動可能な形で設置されている。サンプル分注チップ/反応容器搬送機構14は、サンプル分注チップ/反応容器保持部材15、および反応容器攪拌機構16、サンプル分注チップ/反応容器廃棄孔17、サンプル分注チップ装着位置18、インキュベーターディスク9の所定箇所、の範囲を移動可能なように取り付けられており、これによりサンプル分注チップおよび反応容器8の搬送を行う。
 サンプル分注チップ/反応容器保持部材15には、未使用の反応容器8とサンプル分注チップが複数設置されている。したがって、サンプル分注チップ/反応容器搬送機構14は、サンプル分注チップ/反応容器保持部材15の上方に移動し、その後、下降して未使用の反応容器8を掴むと上昇する。その後、サンプル分注チップ/反応容器搬送機構14は、インキュベーターディスク9の所定位置の上方に移動後、下降してインキュベーターディスク9に未使用の反応容器8を装着する。
 また、サンプル分注チップ/反応容器搬送機構14は、サンプル分注チップ/反応容器保持部材15の上方に移動し、その後、下降して未使用のサンプル分注チップを掴み上昇する。さらに、サンプル分注チップ/反応容器搬送機構14は、サンプル分注チップ装着位置18の上方に移動し、そこで下降してサンプル分注チップをサンプル分注チップ装着位置18に設置する。
 また、サンプル分注ノズル10は、回転および上下動可能である。サンプル分注ノズル10は、サンプル分注チップ装着位置18の上方に回転移動した後、下降してサンプル分注ノズル10の先端にサンプル分注チップを装着する。その後、サンプル分注チップを装着したサンプル分注ノズル10は、ラック6に保持されたサンプル容器5の上方に移動した後、下降してサンプル容器5に保持されたサンプルを所定量吸引する。
 次に、前記サンプルを吸引したサンプル分注ノズル10は、インキュベーターディスク9の上方に移動した後に下降して、インキュベーターディスク9に保持された未使用の反応容器8にサンプルを吐出する。サンプル分注ノズル10は、前記サンプルの吐出が完了すると、サンプル分注チップ/反応容器廃棄孔17からサンプル分注チップを廃棄する。
 また、試薬保冷庫4の内部に配置された試薬容器装てん部3には複数の試薬容器2が装てんされている。さらに、試薬容器装てん部3の上部には試薬保冷庫4の蓋19が設けられ、試薬保冷庫4によってその内部は所定の温度に保温することができる。
 蓋19は保冷庫の一部を覆っていないが、ここは試薬容器装てん部3を上に持ち上げ出し入れするためのローダー(図1では図示していない)がある。
 また、蓋19の一部には、試薬吸引用の貫通孔である試薬吸引孔20が設けられている。なお、試薬分注ノズル11は回転と上下動が可能であり、試薬保冷庫4の蓋19に設けられた試薬吸引孔20の上方に回転移動した後に下降し、試薬吸引孔20を通過する。その後、試薬分注ノズル11は、試薬吸引孔20を通った試薬分注ノズル11の先端を所定の試薬容器2内の試薬に入れ、所定量の試薬を吸引する。その後、試薬分注ノズル11は上昇および回転してインキュベーターディスク9の所定位置の上方に移動し、インキュベーターディスク9に設置された反応容器8内に試薬を吐出する。
 サンプルおよび試薬が吐出された反応容器8は、インキュベーターディスク9の回転によって所定位置に移動し、さらにサンプル分注チップ/反応容器搬送機構14によって反応容器攪拌機構16に搬送される。反応容器攪拌機構16は、反応容器8に対して回転運動を加えることで反応容器8内のサンプルと試薬を攪拌して混和させる。なお、攪拌の終了した反応容器8はサンプル分注チップ/反応容器搬送機構14によって、インキュベーターディスク9の所定位置に戻される。
 反応容器搬送機構12は、回転と上下動が可能であり、サンプルと試薬の分注、および攪拌が終了し、且つインキュベーターディスク9で所定の反応時間が経過した反応容器8の上方に移動してから下降し、反応容器8を保持する。その後、反応容器8は、反応容器搬送機構12の回転移動によって検出ユニット21に搬送され、検出ユニット21においてサンプル中の特定成分の分析結果が決定される。
 なお、以上の各種構成部材の駆動および駆動タイミングについては、図1には図示されていない自動分析装置1のパソコンなどの制御ユニット103(図3、図4参照)によって制御されている。
 図2に示す自動分析装置22も多くの部分が図1に示す自動分析装置1と共通であるので、異なる部分を記す。
 まず、この自動分析装置22は、サンプル分注ノズル23と試薬分注ノズル24の先にチップが付いていないタイプである。自動分析装置22は、これらのノズルでのサンプルおよび試薬の吸引、排出を終えた後、チップを交換しない代わりに、ノズルの先端を洗浄するためのノズル洗浄機構25に先端を入れて洗浄する。
 また、この自動分析装置22では反応容器8内の試薬・サンプル混合物の発色強度、濁度などを測定ユニット26で定量する。
 (2)分注機構
 図3は、本発明の一実施形態に係る自動分析装置における分注機構の一例を示す概略模式図である。図4は、本発明の一実施形態に係る自動分析装置における分注機構の他の一例を示す概略模式図である。
 図3に示すように、分注機構100は、着脱可能なチップ101(前述したサンプル分注チップやチップに相当)を装着したサンプルおよび試薬を分注する分注ノズル102(前述したサンプル分注ノズル10や試薬分注ノズル11に相当)などは、図1で説明したような動きを行う。これは、制御ユニット103で制御されている。
 分注ノズル102は、流路104を介してシリンジ105に接続され、これらの内部はシステム水106で満たされている。シリンジ105は、シリンダ107およびプランジャ108を有する。プランジャ108は、制御ユニット103により駆動を制御されている。制御ユニット103は、プランジャ108をシリンジ105に対して上下方向に駆動させ、これにより分注ノズル102に装着されたチップ101での液体サンプルや液体試薬の吸引および吐出を行う。シリンジ105は、給水タンク109と通じる流路118を有している。この流路118には、電磁弁110および圧力発生手段である給水ポンプ111が設けられている。給水タンク109にはシステム水106が貯留されており、給水ポンプ111の駆動によりシステム水106を分注ノズル102から吐出することで分注ノズル102の内部を洗浄することができる。この洗浄は、サンプルや試薬を分注する前に行うことにより、分注時の分注ノズル102の汚れがサンプルや試薬に混入することを極力抑制することができる。
 圧力センサユニット112は、分注ノズル102とシリンジ105との間の流路104中にある。圧力センサユニット112は、ノズル(分注ノズル102)および圧力発生手段(給水ポンプ111)を含めた流路104内の圧力を圧力センサ113で測定し、その結果を制御ユニット103に送る。
 図4に示す分注機構114も多くの部分が図3に示す分注機構100と共通であるので、異なる部分を記す。
 まず、この分注機構114は、分注ノズル115の先にチップが付いていないタイプである。分注ノズル115は、サンプルや試薬の吸引および吐出を終えた後、チップを交換しない代わりに、ノズルの先端を洗浄するための洗浄水116の入ったノズル洗浄機構117にノズルの先端を入れて洗浄する。
 (3)圧力センサユニット
 前述した圧力センサユニット112について詳述する。
 図5は、本発明の一実施形態に係る自動分析装置における圧力センサユニットの一例を示す断面模式図である。図6は、本発明の一実施形態に係る自動分析装置における圧力センサユニットの他の一例を示す断面模式図である。図7は、本発明の一実施形態に係る自動分析装置における圧力センサユニットのさらなる他の一例を示す断面模式図である。
 図5に示す圧力センサユニット112は、図3、図4を参照して説明したように、分注ノズル102(分注ノズル115)とシリンジ105の間の流路104中にある。圧力センサユニット112は、圧力を計測するダイアフラム201を有する圧力センサ202と、圧力センサユニット112の筐体である接続容器203とを含んで構成されている。また、圧力センサ202と接続容器203との間には、システム水106が漏れないようにするため、O-リング204が装着されている。O-リング204が接続容器203内部に入り込む構造にすることで、より耐圧性が向上する。
 本実施形態では、圧力センサユニット112の(具体的には、接続容器203の)内面203aに、シリカ粒子またはアルミナ粒子と、ケイ素化合物と、を含んでなる、または、シリカ粒子またはアルミナ粒子と、ケイ素化合物と、からなる親水膜305(図8、図12参照)が形成されている。このように、圧力センサユニット112の内面203aを親水膜305によって親水化することにより、内面203aは水と親和し、空気をはじく撥空気面になる。そのため、気泡が付着しなくなり、気泡レス化が進行する。
 また、本実施形態では、内面203aの基材301(図8や図12A~C参照)の算術平均粗さRa0と、親水膜305形成後の内面203aの算術平均粗さRa1との関係が、0.8×Ra0≦Ra1であることとしている。内面203aがこのような態様で粗化されることにより、親水性はさらに向上し、気泡レス化がさらに達成される。この効果をより得る観点から、本実施形態では、0.85×Ra0≦Ra1が好ましく、0.9×Ra0≦Ra1がより好ましく、0.95×Ra0≦Ra1がさらに好ましく、0.97×Ra0≦Ra1がさらにより好ましい。
 なお、O-リング204が接続容器203内部に入り込む構造にすると、すなわち、O-リング204がシステム水106と接する箇所があると、システム水106や流路104などで発生した気泡がO-リング204にも接触する。そのため、O-リング204表面に気泡が残り、圧力のセンシングに影響を与える可能性がある。したがって、その際はO-リング204にも親水膜305を形成することで、接続容器203内をより気泡レス化することができる。
 分注ノズル102(分注ノズル115)およびシリンジ105からのそれぞれの流路104の圧力センサユニット112との接続部位205、206は、圧力センサユニット112内にシステム水106が流入または流出される流入出口205a、206aを有している。流入出口205a、206aの開口部は、流路104の開口部と連通する。なお、図5に示す接続部位205、206は、流路104との接続をねじ込み式としているが、耐圧性のある接続方法であれば押し込むなどの方法で流路104と接続することができる。
 本実施形態では、圧力センサユニット112の液体の流入出口205a、206aの1つあたりの断面積、すなわち、接続部位205、206における流路開口面積をα、圧力センサユニット112の受圧面(ダイアフラム201)の面積をβとしたとき、α<βとすることが好ましい。このようにすると、圧力センシングの感度が向上する。
 図5では、圧力センサユニット112の内面形状(接続容器203の内面形状)は円錐状になっており、円錐頂点が鉛直上方となるように設置される。円錐の頂点部にノズル(分注ノズル102)と接続される液体の流入出口205aが設けられている(図5の上図)。円錐の底周面の接線方向に圧力発生手段(給水ポンプ111)と接続される液体の流入出口206aが設けられている(図5の下図)。また、円錐の底面に圧力センサユニット112の受圧面(ダイアフラム201)が配置されている。
 底周面の接線方向に設けられている流入出口206aから接続容器203内に流入したシステム水106は、接続容器203内の円周面に沿って旋回流を起こしながら、頂点部の流入出口205aに向かって流れ、頂点部の流入出口205aから流出する。圧力センサユニット112は、この旋回流により接続容器203内の気泡が除去される。
 接続容器203は耐圧を考慮し、アクリル、ポリカーボネートなどの樹脂を用いるとよい。接続容器203の製造の際は、樹脂のブロックを機械加工により成型することができるが、その際、発生する熱による変形を防ぐため、ブロック自体を水に沈めて加工したり、ブロック表面に十分散水して冷却しながら加工したりするとよい。本実施形態で用いる接続容器203の内面の算術平均粗さ(Ra)を測定したところ、ロットにより差はあるが100~300nmであった。
 本実施形態では、接続容器203の円錐状の内面203aに親水膜305を形成し、接続容器203内の気泡を除去することにより、接続容器203内部の気泡の滞留を減らせるので、ダイアフラム201での圧力計測の精度がより向上する。
 なお、圧力センサユニット112の形状(接続容器203の形状)は、図5に示すような円錐状に限定されない。
 圧力センサユニット112は、例えば、図6に示すように、接続容器203の内部を円錐と円筒とを繋げた構造とすることができる。この場合、流入出口206aは円錐の部分ではなく、円筒の部分に形成するとよい。円錐部分より円筒部分の方が、流入出口206a形成の加工がしやすいメリットがある。
 また、圧力センサユニット112は、例えば、図7に示すように、接続容器203の内部を円筒状としつつ、上部の角部を丸めた構造とすることができる。このような丸みを帯びた構造とした場合も、上部の角部における気泡の滞留を抑制することができる。
 図5~図7のうち、圧力センサユニット112の上部の流入出口205aから圧力センサ202の受圧部であるダイアフラム201まで接続容器203内の空隙の断面が徐々に大きくなっていく方が、接続容器203内でシステム水106の乱流を生じにくい。その観点からすると、圧力センサユニット112は、図5に示す円錐状の構造が最も好適であると言える。
 (4)親水膜
 a)親水膜用コート液の組成・形成方法の概略
 本実施形態における親水膜305は、この膜を形成する親水膜用コート液を接続容器203内部に塗布した後、硬化させることにより製する。親水膜用コート液の材料組成は、具体的には親水性の材料として二酸化ケイ素(シリカ)製の粒子(シリカ粒子)および三酸化二アルミニウム(アルミナ)製の粒子(アルミナ粒子)のうちの少なくとも一方と、これらの粒子を保持するバインダと、これらを分散または溶解する有機溶剤とからなる。なお、バインダは、加熱すると主成分がシリカになるアルコキシシラン化合物であることが好ましい。
 本実施形態においては、シリカ粒子やアルミナ粒子のサイズを規定することにより親水膜305の親水性がより向上し、より気泡レス化に寄与する。また、シリカ粒子やアルミナ粒子の添加率を規定することにより親水膜305の親水性がより向上し、より気泡レス化に寄与する。
 図8は、シリカ粒子とシリカバインダとを含んでなる親水膜の断面図(走査電子顕微鏡像(SEM像))である。図8に示す親水膜305は、親水性の材料としてシリカ粒子302を用いている。
 図8に示すように、親水膜305は、基材301の上に形成されている。親水膜305は、シリカ粒子302とシリカバインダ304(図12A~図12C参照)とその間にある空隙303とを含んでなる。シリカバインダ304は、複数のシリカ粒子302の間および基材301に薄く付着して膜としての構造を保持している。シリカ自身の水との接触角は30°程度であるが、親水膜305表面のシリカ粒子302による細かな凹凸、および表面の空隙303により、水との接触角が低減する。さらに、シリカ粒子302間の隙間、および空隙303に対して毛細管現象により、結露水が染み込むことにより、親水膜305表面の水との接触角がさらに低減する。これにより、水との接触角が15°以下の親水膜305が形成可能となる。
 なお、シリカ粒子302に代えてアルミナ粒子を用いた場合も同様の膜構造となり、同様の効果が得られる。
 b)親水膜を形成する親水膜用コート液とその組成
 この親水膜用コート液は、シリカ粒子およびアルミナ粒子のうちの少なくとも一方と、シリカバインダの前駆体とを有機溶剤に分散および溶解することにより製する。有機溶剤は、アルコール系の溶剤である。これら3種類の材料について説明する。
 b-1)親水性の材料
 親水性の材料として、本実施形態ではシリカ粒子およびアルミナ粒子のうちの少なくとも一方を用いる。これらの粒子の平均粒子径は、好ましくは10~50nm、より好ましくは10~20nmである。以下にこの理由を記す。
 なお、これらの粒子の平均粒子径は、堀場製作所製粒度分析装置LA-950V2で測定することができる。
・親水性:
 シリカ粒子およびアルミナ粒子の親水性は、粒子の表面により発揮される。例えば、シリカ粒子の場合は、ケイ素と酸素の結合の一部が切れて水酸基が露出し、これが親水性を発揮すると考えられている。また、アルミナの場合は、アルミニウムと酸素の結合の一部が切れて水酸基が露出し、これが親水性を発揮していると考えられている。そのため、単位質量当たりの粒子の表面積が大きいほど、単位質量当たりの表面に露出している水酸基の数が増えるので親水性が向上する。粒子径が小さい方が単位質量当たりの表面積が大きくなるため、親水性をより強く発揮する。
 一方、粒子径が大きくなると成膜後の粒子間の隙間の間隔が広くなり、粒子間の隙間の数が少なくなるので、粒子径の小さな粒子を用いた場合に比べて毛細管現象が起こりにくくなる。そのため、粒子径が大きくなると、結果的に親水性が向上しにくくなる。
・親水膜用コート液中での分散安定性:
 これらの粒子の密度は、シリカ粒子が2.5~2.6g/cm3、アルミナ粒子が3.9~4.1g/cm3であり、後述する溶剤の0.8~1.0g/cm3に比べてかなり大きい。そのため、これらの粒子の粒子径が大きいと親水膜用コート液中で沈降する。サイズ的にはシリカ粒子の場合は70nm以上、アルミナ粒子の場合は50nm以上で沈降が顕著に見られるようになる。そのため、粒子径が大きい場合は使用前に攪拌し、親水膜用コート液全体に分散させてから使用する必要がある。よって、上記理由から好ましい粒子の平均粒子径は50nm以下となる。
 なお、上記は粒子形状が球状であることを前提で記しているが、粒子によっては球状粒子が数個~数十個つながった形状のものもある。この粒子の場合、1個の球状粒子のサイズが50nm以下であれば、分散性は十分であり、好適である。この観点からも、上記理由により、好ましい平均粒子径は50nm以下となる。
・基材に対するレベリング性:
 表面に凹凸のある基材301にコーティングした場合、粒子が谷の部分をある程度埋めるので表面凹凸が小さくなる。これは算術表面粗さ(Ra)が小さくなることで確認できる。Raが小さくなると、実表面積が小さくなるので、投影面積の単位面積当たりのシリカ粒子やアルミナ粒子由来の水酸基の数が減り、所望の親水性が得られない可能性がある。粒子が大きいほど、具体的には平均粒子径が30nm以上の粒子を用いた場合、谷を埋める傾向が強い。平均粒子径が10~20nmの場合は、谷を埋める効果はわずかであり、凹凸の形状に沿って、谷を埋めずにコーティングされるので、Raはほとんど変化がなく、高い親水性を付与することが可能である。この観点からは、上記理由により、より好ましい平均粒子径は10~20nmとなる。
・ハンドリング:
 粒子が小さすぎると、具体的には平均粒子径が10nm未満の場合、秤量や混合等の際、空気中に粉塵として広がる恐れがある。空気中に漂っているこれら粒子を人間が吸うと呼吸障害などが発生する恐れがある。したがって、用いるシリカ粒子およびアルミナ粒子の平均粒子径は10nm以上が好ましい。この観点からは、上記理由により、好ましい平均粒子径は10nm以上となる。
 b-2)バインダ
 一実施形態に係る親水膜305は、製膜後の主成分がシリカのバインダである。シリカは有機溶剤には不溶であるが、その前駆体は有機溶剤に溶解するものがある。本実施形態では、有機溶剤に溶解するシリカの前駆体を用いる。
 このようなシリカの前駆体としては、具体的には、加水分解によってシリカに変化する加水分解性のケイ素化合物が挙げられる。もっとも一般的な加水分解性のケイ素化合物は通常シリカゾルと呼ばれている。これは複数のテトラアルコキシシランが一部加水分解してアルコキシ基が外れ、複数の分子同士がケイ素-酸素-ケイ素の結合を形成して平均分子量が数千~数万の重合体になったものである。
 シリカゾルは、メタノール、エタノールなどのアルコール系溶剤に可溶である。テトラアルコキシシランとしては、テトラエトキシシランが最も使いやすい。これよりアルキル鎖が短いテトラメトキシシランは水との反応性が高いため、高湿環境で長期間保管すると親水膜305を形成する親水膜用コート液が固まってしまう可能性がある。また、テトラエトキシシランよりアルキル鎖の長いテトラプロポキシシラン、テトラブトキシシランなどは加水分解反応が進行しにくいので、塗布後の加熱硬化時間が長くなったり、加熱温度を高くしないと熱硬化反応の進行が遅くなったりするなどの傾向がある。したがって、シリカゾルを形成するためのテトラアルコキシシランとしては、テトラエトキシシランが好ましい。
 アルコキシシランの重合体であるシリカゾル以外には、分子中の4つの結合基を有するケイ素原子に3つのアルコキシ基と、それ以外にベンゼン環やアルキル鎖などのアルコキシ基以外の結合基を有する化合物も、シリカバインダ304の前駆体として用いることが可能である。ただし、この材料を使った場合、親水膜305の主成分はシリカではあるが、これのみではなく、炭素や窒素、イオウなどの原子も含まれた膜となる。
 そのような化合物として具体的には、メチルトリメトキシシラン、ジメチルジメトキシシラン、フェニルトリメトキシシラン、フェニルトリエトキシシラン、メチルトリエトキシシラン、n-プロピルトリメトキシシラン、n-プロピルトリエトキシシラン、ビニルトリメトキシシラン、ビニルトリエトキシシラン、3-アミノプロピルトリメトキシシラン、3-アミノプロピルトリエトキシシラン、N-2-(アミノエチル)-3-アミノプロピルトリメトキシシラン、N-2-(アミノエチル)-3-アミノプロピルトリエトキシシラン、3-イソシアネートプロピルトリメトキシシラン、3-イソシアネートプロピルトリエトキシシラン、3-メルカプトプロピルトリメトキシシラン、3-メルカプトプロピルトリエトキシシランなどが挙げられる。
 b-3)溶剤
 親水膜用コート液の溶剤は、通常の有機溶剤を用いる。但し、圧力センサユニット112の筐体である接続容器203は機械加工のし易さからアクリルやポリカーボネートといった樹脂が用いられることが多い。しかし、これらの樹脂は溶剤の種類によっては溶解したり、膨潤したりしてしまう恐れがある。そこで、これらの樹脂を溶解、膨潤させない、或いはほとんどさせないという点で、アルコール系の溶剤が好適である。具体的には、炭素数が1~4のメタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、tert-ブタノールなどが好適である。これらの溶剤は沸点が60~120℃程度であるため、コーティング後の揮発が敏速であり、製膜に要する時間を短くできるメリットがある。ブタノールより炭素数が多いと加水分解性のケイ素化合物を溶解しにくくなり、しかも塗布後の乾燥に時間がかかるので、炭素数が5以上のアルコールは好適ではない。
 これらのアルコール以外には、エチレングリコールの水酸基の1個がモノアルコキシ基となったセロソルブ系の溶剤も加水分解性のケイ素化合物を溶解しやすい点で好適である。ただ、これらの多くは沸点が高く揮発しにくいので、コーティング後は何らかの加熱手段、或いは減圧手段により揮発を早めることが望まれる。
 c)膜厚(平均膜厚)
 図9は、親水膜の膜厚(nm)と水との接触角(°)との関係を示したグラフである。
 図9のグラフは、シリカ粒子302とシリカの前駆体との質量比率が1:1の親水膜用コート液で親水膜305を製膜した結果を示している。また、基材301は、算術表面粗さ(Ra)が10nmであり、目視では平坦なアクリル板である。図9に示すように、親水膜305の膜厚が40nm以上になると、水との接触角が15°以下まで低下し、十分な親水性を発揮する。さらに、親水膜305の膜厚が50nm以上になると水との接触角は10°まで低下し、より高い親水性を発揮する。シリカ粒子302に代えてアルミナ粒子を用いた場合もシリカ粒子302を使った場合とほぼ同様の結果であった。したがって、本実施形態における親水膜305の膜厚は50nm以上が好ましい。なお、膜厚は、複数の任意の箇所の膜厚を測定し、その平均値(平均膜厚)で管理することができる。
 d)混合比率
 親水膜305を形成する親水膜用コート液中の固形分は、親水性の材料であるシリカ粒子302およびアルミナ粒子のうちの少なくとも一方と、バインダであるシリカの前駆体とである。この両者の比率が異なると親水膜305の性能、すなわち、水との接触角が変化する。シリカ粒子302とシリカの前駆体との比率を変えて膜厚がほぼ100nmになるよう製膜した親水膜305の水との接触角を調べた。結果を図10に示す。
 図10は、親水膜中のシリカ粒子の割合(質量%)と水との接触角(°)との関係を示したグラフである。
 図10の横軸はシリカ粒子302の割合を示している。シリカバインダ304の前駆体の割合は、100%からシリカ粒子302の割合を引いた値となる。なお、ここで用いたシリカ粒子302の平均粒子径は、20nmであった。シリカ粒子302の平均粒子径は、堀場製作所製粒度分析装置LA-950V2で測定した。接触角は、協和界面科学株式会社製接触角計(型式:CA-D)を用いて測定した。
 親水性のシリカ粒子302の含有率が高くなるにつれ、水との接触角は低下していく。Raが5nmの実質平坦なアクリル板に親水膜305を製膜した(a)の場合は、シリカ粒子302を30質量%以上含有していれば、接触角は10°まで下がった。しかし、シリカ粒子302の含有率が30~90質量%まで接触角は一定であり、これ以上は下がらなかった。
 一方、Raが100nmのアクリル板に親水膜305を製膜した(b)の場合は、Raが5nmの場合よりもさらに接触角は下がった。シリカ粒子302の含有率が30~60質量%では接触角が4~5°まで低下した。すなわち、Raが大きい基板に製膜した方が、親水性が向上した。
 表面の実面積が投影面積より大きくなると、撥水性の表面はより撥水性に、親水性の表面はより親水性になることが知られている。したがって、Raが大きくなること、すなわち、投影面積に比べて実面積が大きくなることにより、親水性が向上し、接触角が低下したものと考えられる。
 なお、(b)の場合において、親水性のシリカ粒子302を70質量%以上にすると接触角が上昇した。すなわち、親水性が低下する傾向が見られた。これは、シリカ粒子302の割合を増やすことでシリカ粒子302が粗化表面の谷の部分に入り込み、Raを下げてしまったことで接触角がやや上昇したものと考えられる。
 親水膜305形成後のRaを測定したところ、平均粒子径20nmのシリカ粒子302を30質量%含有する親水膜305のRaは92nmであった。前記シリカ粒子302を60質量%含有する親水膜305のRaは82nmであった。また、前記シリカ粒子302を65質量%含有する親水膜305のRaは61nmであった。これらのことから、シリカ粒子302の割合が増えてRaが小さくなったことが接触角を高める要因になっていることが判った。十分な親水性を得る観点からは、親水膜305のRaは80nm以上(0.08μm以上)であることが好ましいと言える。
 Raが300nm、500nmのアクリル板を用いた場合も上記と同様、シリカ粒子302の含有率が30~60質量%であると、シリカ粒子302の含有率が20質量%および70質量%のときに比べて接触角が約3°低い4~5°まで低下することが判った。
 以上の結果から、親水膜用コート液中の固形分のうち、親水性の材料であるシリカ粒子302の含有率は30~60質量%が好適であることが判った。
 アルミナ粒子(平均粒子径20nm)でも同様の実験を行ったところ、シリカ粒子302と同様、粒子の含有率は30~60質量%が好適であることが判った。なお、アルミナ粒子の平均粒子径も堀場製作所製粒度分析装置LA-950V2で測定した。
 また、平均粒子径が20nmと50nmの2種類のシリカ粒子302を用いて、Raが100nmのアクリル板に親水膜305を製膜して上記と同様の実験を行った。その結果を図11に示す。
 図11は、親水膜中のSiO2粒子の割合(質量%)と水との接触角(°)との関係を示したグラフである。
 図11に示すように、平均粒子径50nmのシリカ粒子302を用いた(b)の場合、含有率15質量%以上で接触角は12°から下がらなくなった。一方、平均粒子径20nmのシリカ粒子302を用いた(a)の場合、含有率30~60質量%で接触角は4~5°まで低下した。
 親水膜305形成後のRaを測定したところ、平均粒子径20nmのシリカ粒子302を30質量%含有する親水膜305のRaは92nmであった。平均粒子径20nmのシリカ粒子302を60質量%含有する親水膜305のRaは82nmであった。
 一方、平均粒子径50nmのシリカ粒子302を30質量%含有する親水膜305のRaは55nmであった。平均粒子径50nmのシリカ粒子302を60質量%含有する親水膜305のRaは30nmであった。
 図10の結果と同様、Raが小さくなったことが接触角を高める要因になっていることが判った。
 上記結果より、親水膜305成膜後のRaは、基材301のRaの概ね8割程度あれば接触角が4~5°まで低下させることができると考えられる。これ以上Raが小さくなると接触角はこれほどは下がらないことが判った。
 以上の結果と原因を図12A~図12Cに模式的に示す。
 図12A~図12Cは、親水膜を形成した粗化面の断面の模式図である。
 図12Aは、平均粒子径が10~20nm、粒子添加率が30~60質量%の場合であり、シリカ粒子302またはアルミナ粒子とシリカバインダ304とを含んでなる親水膜305が、基材301の凹凸に沿って製膜されている。
 一方、図12Bは、平均粒子径が40nm以上、粒子添加率が30~60質量%の場合であり、シリカ粒子302またはアルミナ粒子とシリカバインダ304とを含んでなる親水膜305が、基材301の凹凸に沿って製膜されている。
 図12Cは、平均粒子径が10~20nm、粒子添加率が60質量%を超える場合であり、シリカ粒子302またはアルミナ粒子とシリカバインダ304とを含んでなる親水膜305が、基材301の凹凸に沿って製膜されている。
 図12Bおよび図12Cに示すように、いずれも基材301の凹凸の谷を埋めるような形で親水膜305が製膜されている。そのため、Raが低下して表面の実面積が小さくなり、親水性が低下すると考えられる。
 シリカ粒子302やアルミナ粒子が大きいと谷が埋まる理由は、以下のように考えられる。
 粒子径が小さい場合、単位質量当たりの粒子の表面積が大きいので粒子の表面と基材301表面の相互作用が大きく、基材301の表面に沿った形で粒子が並んで製膜される。
 これに対し、粒子が大きくなると、単位質量当たりの粒子の表面積が小さくなるので、基材301表面の相互作用が小さくなる。そのため、粒子の重力が支配的になり、基材301の谷の部分に沈降する。その結果、図12Bのような膜構造になると考えられる。
 また、粒子の割合が高くなると谷の部分が埋まる理由は、粒子分が高くなるとコート液のチキソトロピック性が高まり、基材301の谷の部分にコート液が溜まりやすくなったのではないかと推定する。
 e)圧力センサユニットの製膜方法
 圧力センサユニット112の製膜は、親水膜用コート液の塗布および熱硬化により完了する。
 (i)塗布(ねじ込み工程、塗布工程)
 親水膜用コート液をスポイトなどで接続容器203内に塗布する。このとき、接続容器203と配管(流路104)とを接続する接続部位205、206は、図5~7に示すように、ねじ込み式の接続になっている。しかし、このねじ込み部分に親水膜用コート液が付着して親水膜305が形成されると、製膜後、圧力センサユニット112に高圧がかかった場合に、接続部位205、206からシステム水106が漏れてくる可能性がある。これはシステム水106が親水膜305内に形成される空隙303内を毛細管現象によって移動して生じると考えられる。
 この懸念は、親水膜用コート液を塗布する際に接続部位205、206に予め配管または樹脂製のねじ207(図13A参照)などをねじ込んでおき、親水膜用コート液塗布時に親水膜用コート液がねじ込み部分に付着しないようにすることで解消できる。つまり、流入出口205a、206aにおける流路104と接する面(より具体的には、流路104と接する接続部位205、206の表面)には、前記親水膜305が形成されないようにすることで解消できる。なお、親水膜用コート液塗布後、溶剤が揮発すれば、ねじ込み部分への親水膜用コート液の付着の恐れは無くなるので、その時点で配管または樹脂製のねじ207を外し、後述する熱硬化工程に進むことができる。
 (ii)熱硬化(製膜工程)
 親水膜用コート液を塗布後、熱硬化を行い、製膜する。なお、熱硬化前かつ配管または樹脂製のねじ207を外す前に、内面203aに付着した余分な親水膜用コート液を除去するとよい。これは、紙などの液体を吸収する能力のあるものを親水膜用コート液に接触させることで行うことができる。
 熱硬化では、接続容器203の材質が加熱により変形しないよう耐熱温度以下で加熱する必要がある。材質がアクリルの場合は70℃以上で変形の恐れがあるので、マージンをとって60℃で加熱する。材質がポリカーボネートの場合は120℃以上で変形の恐れがあるので、マージンをとって100℃で加熱する。
 熱硬化の進行は加熱温度が高いほど早く進行するので、基材301の耐熱温度が高いほど、熱硬化に要する時間も短時間で済む。材質として比較的耐熱温度の低いアクリルの場合、60℃で1時間加熱すると、鉛筆硬度でB程度の硬度の親水膜305が得られる。この後、相対湿度が70~90%程度の高湿環境に1時間程度放置し、再び60℃で1時間加熱すると、鉛筆硬度でHB程度の硬度の親水膜305が得られる。もう一度、相対湿度が70~90%程度の高湿環境に1時間程度放置し、再び60℃で1時間加熱すると、鉛筆硬度でH程度の硬度の親水膜305が得られる。基材301のアクリルの鉛筆硬度がH程度であるので、この程度まで硬化すれば親水膜305は基材301と同程度の硬度となり、実用上問題ないレベルと判断される。
 高湿環境に放置する工程を入れる理由は以下の通りである。親水膜用コート液の熱硬化反応が加水分解反応なので、反応中に水分を供給することが効果的であるが、60℃の恒温槽中では空気中の水分は極僅かであるため空気中から水分を供給できない。そこで、途中で熱硬化を中断し、高湿環境に放置することで空気中の水分が熱硬化途上の親水膜305に付着・吸収され、再び加熱した際に熱硬化反応を効率良く進行させることができる。
 以上に説明したように、本実施形態に係る圧力センサユニット112の製膜方法は、圧力センサユニット112の内面203aに、シリカ粒子302およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含んでなる、または、からなる親水膜305を形成することができる。そして、このようにして製膜されると、内面203aの基材301の算術平均粗さRa0と、親水膜305形成後の内面203aの算術平均粗さRa1との関係は、0.8×Ra0≦Ra1となる。したがって、この製膜方法によって得られた圧力センサユニット112は、内部に気泡を残さずに、液体を吸引・吐出することができる。
 f)自動分析方法
 本実施形態に係る自動分析方法は、前記圧力センサユニット112を備えた分注機構100、114に接続されたノズル(分注ノズル102、115、サンプル分注ノズル10、試薬分注ノズル11)により液体を吸引・吐出する、というものである。本実施形態に係る自動分析方法で用いる圧力センサユニット112は、前述したように、内面203aにシリカ粒子302およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含んでなる、または、からなる親水膜305が形成されている。そして、前記内面203aの基材301の算術平均粗さRa0と、前記親水膜305形成後の内面203aの算術平均粗さRa1との関係が、0.8×Ra0≦Ra1となっている。このように、自動分析方法は、前記圧力センサユニット112を用いているので、内部に気泡を残さずに、液体を吸引・吐出することができる。
 次に、実施例を示しながら、本発明を説明する。なお、本発明はこれらの実施例に限定されるものではない。
 (1)親水膜を形成するコート液の調製
 平均粒子径が10nmのシリカ粒子(15g)、および分散剤としてジエチレングリコールモノアセテート(0.5g)、溶剤としてエタノール(84.5g)を内容量200mlのポリプロピレン製のビンに入れ、これらをオーバーヘッドスターラ―で攪拌することにより15質量%シリカ粒子の分散液Aを100g調製した。
 加水分解後の固形分濃度が2質量%になるよう調製した溶剤がエタノールのシリカゾル溶液Bを750g調製した。
 内容量1000mlのポリプロピレン製のビンに分散液A(1.0g)、シリカゾル溶液B(7.5g)、エタノール(21.5g)を加え、ビンを密栓後数回攪拌して、固形分が1.0質量%で、且つシリカ粒子が固形分中の50質量%の親水膜305を形成するコート液(親水膜用コート液CS1)(30g)を調製した。
 なお、本実施例で調製した親水膜用コート液CS1を用いてRaが10nmのアクリル板に製膜された親水膜305の水との接触角は10°であった。
 (2)圧力センサユニット112の接続容器203への親水膜305の製膜プロセス
 図13A~図13Cを参照して、このプロセスについて説明する。
 図13A~図13Cはいずれも、接続容器の内面への親水膜成膜プロセスを説明する説明図である。なお、図13Aはコーティング前の状態を示している。図13Bはコーティングおよび熱硬化後、O-リング204を装着した状態を示している。図13Cは圧力センサ202を設けた状態、つまり、圧力センサユニット112の状態を示している。
 図13Aに示すように、本実施例で用いる圧力センサユニット112の接続容器203を円錐状の頂点の部分を下にして平坦な台に置いた。この接続容器203の内面203aのRaは100nmである。接続容器203の接続部位205、206は親水膜用コート液CS1が付着しないようあらかじめ接続部位205、206にフィットする樹脂製のねじ207をねじ込んでおいた。この状態で親水膜用コート液CS1をスポイトなどで接続容器203の内面203aに塗布した。
 その後、接続容器203をひっくり返し、円錐状の頂点の部分を上にして余分な親水膜用コート液CS1を吸収する紙を敷いた上に置いた。約10分後、接続容器203を60℃に調整した恒温槽に入れ、1時間その中に放置した。その後、接続容器203を温度25℃、湿度80%に制御された恒湿恒温槽に入れ、1時間その中に放置した。再び、接続容器203を60℃に調整した恒温槽に入れ、1時間その中に放置した。その後、温度25℃、湿度80%に制御された恒湿恒温槽に接続容器203を入れ、1時間その中に放置し、最後に再び60℃に調整した恒温槽に入れ、1時間その中に放置することで接続容器203の内面203aに所望の親水膜305を形成した。
 次に、図13Bに示すように、接続部位205、206から樹脂製のねじ207を外し、接続容器203の円錐状の底周面の周縁に沿うようにO-リング204を装着した。
 次に、図13Cに示すように、圧力センサ202をダイアフラム201が接続容器203の内面203aに向くようにセットして、圧力センサユニット112を完成させた。
 (3)O-リングへの親水膜の製膜プロセス
 上記で使ったO-リング204は、接続容器203の内面203aと圧力センサ202との隙間を塞ぎ、高圧のシステム水106が漏れないようにする役割がある。そのため、極僅かであるがシステム水106との接触がある。したがって、接続容器203の内面203aに親水膜305を形成しても、O-リング204上に気泡が付着する可能性がある。そのため、O-リング204表面も親水膜305を形成することが望ましい。
 そこで、O-リング204が完全に浸る量の前記親水膜用コート液CS1をシャーレに入れ、その中にO-リング204を10秒間浸した。その後、O-リング204をピンセットで引上げ、接続容器203と同様、60℃で1時間加熱し、その後、温度25℃、湿度80%の環境に1時間放置するプロセスを2回行った後、60℃で1時間加熱することで、表面に親水膜305を形成したO-リング204を製した。
 そして、前述したように、このO-リング204を内面203aに親水膜305を形成した接続容器203に装着し、その後、圧力センサ202を装着して本実施例の圧力センサユニット112を完成させた。
(4)親水膜形成の効果確認
 上記(1)~(3)のプロセスで製した圧力センサユニット112を図3に示す分注機構100に取り付け、これを図1の自動分析装置1にセットし、試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に目視では気泡は確認されず、分注の誤差も基準以下であった。
 これに対し、親水膜305を形成しない圧力センサユニット112を用いて試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に気泡が数個確認され、O-リング204にも気泡の付着が確認された。分注の誤差は基準を超えていた。
 以上より、接続容器203およびO-リング204に親水膜305を形成した圧力センサユニット112を用いることで、精度の高い分注操作が可能な自動分析装置1を具現できることが確認された。
 なお、上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため図14の垂直面切断箇所208と傾斜面切断箇所209で切断し、垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。なお、図14は、接続容器の内面の表面粗さ測定時の切断部分を示す説明図である。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では90~95nm、傾斜面の粗さ測定部位211では86~91nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。
 また、接続容器203に形成された親水膜305の厚さは切断面をSEMで観察・撮影し、得られた画像から測定した。その結果、垂直面の粗さ測定部位210では120~150nm、傾斜面の粗さ測定部位211では170~200nmであった。
 内容量1000mlのポリプロピレン製のビンに実施例1で調製した15質量%シリカ粒子の分散液Aを1.0gから0.6gに変更し、シリカゾル溶液Bを7.5gから10.5gに変更し、エタノールを21.5gから18.9gに変更して加え、ビンを密栓後数回攪拌して、固形分が1.0質量%で、且つシリカ粒子が固形分中の30質量%の親水膜305を形成するコート液(親水膜用コート液CS2)(30g)を調製した。
 親水膜用コート液CS1の代わりに上記の親水膜用コート液CS2を用いる以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に目視では気泡は確認されず、分注の誤差も基準以下であった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では95~97nm、傾斜面の粗さ測定部位211では90~95nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。
 また、接続容器203に形成された親水膜305の厚さを実施例1と同様に測定すると、垂直面の粗さ測定部位210では100~120nm、傾斜面の粗さ測定部位211では130~150nmであった。
 内容量1000mlのポリプロピレン製のビンに実施例1で調製した15質量%シリカ粒子の分散液Aを1.0gから1.2gに変更し、シリカゾル溶液Bを7.5gから6.0gに変更し、エタノールを21.5gから22.8gに変更して加え、ビンを密栓後数回攪拌して、固形分が1.0質量%で、且つシリカ粒子が固形分中の60質量%の親水膜305を形成するコート液(親水膜用コート液CS3)(30g)を調製した。
 親水膜用コート液CS1の代わりに上記の親水膜用コート液CS3を用いる以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に目視では気泡は確認されず、分注の誤差も基準以下であった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では90~92nm、傾斜面の粗さ測定部位211では82~88nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。
 また、接続容器203に形成された親水膜305の厚さを実施例1と同様に測定すると、垂直面の粗さ測定部位210では140~180nm、傾斜面の粗さ測定部位211では200~230nmであった。
 平均粒子径が10nmのシリカ粒子(15g)を平均粒子径が20nmのシリカ粒子(15g)に代える以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に目視では気泡は確認されず、分注の誤差も基準以下であった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では86~90nm、傾斜面の粗さ測定部位211では82~84nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。

 平均粒子径が10nmのシリカ粒子(15g)を平均粒子径が20nmのアルミナ粒子(15g)に代える以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に目視では気泡は確認されず、分注の誤差も基準以下であった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では86~90nm、傾斜面の粗さ測定部位211では82~84nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。

 平均粒子径が10nmのシリカ粒子(15g)を平均粒子径が40nmのシリカ粒子(15g)に代える以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、分注の誤差は試薬分注およびサンプル分注いずれの場合も分注量の質量比で1/100~5/100であった。
 用いる粒子径が大きいため親水膜305の親水性が低くなり分注の誤差が若干大きくなったものの、分注の誤差は基準以下であった。また、圧力センサユニット112の接続容器203内に気泡は目視では確認されなかった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では84~86nm、傾斜面の粗さ測定部位211では80~82nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。
 また、接続容器203に形成された親水膜305の厚さを実施例1と同様に測定すると、垂直面の粗さ測定部位210では180~220nm、傾斜面の粗さ測定部位211では250~300nmであった。
 内容量1000mlのポリプロピレン製のビンに実施例1で調製した15質量%シリカ粒子の分散液Aを1.0gから1.3gに変更し、シリカゾル溶液Bを7.5gから5.25gに変更し、エタノールを21.5gから23.45gに変更して加え、ビンを密栓後数回攪拌して、固形分が1.0質量%で、且つシリカ粒子が固形分中の65質量%の親水膜を形成するコート液(親水膜用コート液CS4)(30g)を調製した。
 親水膜用コート液CS1の代わりに上記の親水膜用コート液CS4を用いる以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、圧力センサユニット112の接続容器203内に目視では気泡は確認されなかった。
 用いる粒子の添加率が高いため親水膜305の親水性が低くなり分注の誤差が若干大きくなったものの、分注の誤差は基準以下であった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では88~90nm、傾斜面の粗さ測定部位211では80~84nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。
 また、接続容器203に形成された親水膜305の厚さを実施例1と同様に測定すると、垂直面の粗さ測定部位210では160~200nm、傾斜面の粗さ測定部位211では220~250nmであった。
 内容量1000mlのポリプロピレン製のビンに実施例1で調製した15質量%シリカ粒子の分散液Aを1.0gから0.5gに変更し、シリカゾル溶液Bを7.5gから11.25gに変更し、エタノールを21.5gから18.25gに変更して加え、ビンを密栓後数回攪拌して、固形分が1.0質量%で、且つシリカ粒子が固形分中の25質量%の親水膜305を形成するコート液(親水膜用コート液CS5)(30g)を調製した。
 親水膜用コート液CS1の代わりに上記の親水膜用コート液CS5を用いる以外は実施例1と同様のプロセスで圧力センサユニット112を試作し、自動分析装置1にセットして試薬分注およびサンプル分注の操作を行った。その結果、分注の誤差は試薬分注およびサンプル分注いずれの場合も分注量の3/100~5/100であった。
 用いる粒子の添加率が低いため親水膜305の親水性が低くなり分注の誤差が若干大きくなったものの、分注の誤差は基準以下であった。また、圧力センサユニット112の接続容器203内に気泡は目視では確認されなかった。
 上記で作製した圧力センサユニット112を分解して接続容器203を回収し、内面203aのRaを求めるため実施例1と同様に垂直面の粗さ測定部位210、傾斜面の粗さ測定部位211を準備した。
 これらの接続容器203の内面203aにあたる面のRaを測定したところ、垂直面の粗さ測定部位210では92~97nm、傾斜面の粗さ測定部位211では88~93nmであった。つまり、親水膜305形成後のRaは親水膜305形成前のRa(100nm)の0.8倍以上であることが確認された。
 また、接続容器203に形成された親水膜305の厚さを実施例1と同様に測定すると、垂直面の粗さ測定部位210では100~120nm、傾斜面の粗さ測定部位211では140~170nmであった。
 従来は内面に気泡が付着することで正確な圧力測定が困難であった圧力センサユニットは、実施例1~8に示すように、接続容器203の内面203aに適切な親水膜305を設けて、内面203aの基材301の算術平均粗さRa0と、親水膜305形成後の内面203aの算術平均粗さRa1との関係が、0.8×Ra0≦Ra1であることとすることにより、気泡レス化が可能となった。そして、それにより、圧力センサユニット112を備えた自動分析装置1は、正確な圧力測定が可能となり、その結果、分注機構100での正確な分注を行えることが確認された。
 以上、本発明に係る自動分析装置1、自動分析方法および圧力センサユニット112の製膜方法について実施形態および実施例により詳細に説明したが、本発明は前記した実施形態および実施例に限定されるものではなく、様々な変形例が含まれる。例えば、前記した実施形態は本発明を分かり易く説明するために詳細に説明したものであり、必ずしも説明した全ての構成を備えるものに限定されるものではない。また、ある実施形態の構成の一部を他の実施形態の構成に置き換えることが可能であり、また、ある実施形態の構成に他の実施形態の構成を加えることも可能である。また、それぞれの実施形態の構成の一部について、他の構成の追加・削除・置換をすることが可能である。

Claims (11)

  1.  液体を吸引・吐出するノズルと、該ノズルに接続され液体を吸引・吐出させるための圧力を発生させる圧力発生手段と、前記ノズルおよび前記圧力発生手段を含めた流路内の圧力を測定する圧力センサユニットと、を備えた分注機構を有し、
     前記圧力センサユニットの内面に、シリカ粒子およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含んでなる親水膜が形成されており、
     前記内面の基材の算術平均粗さRa0と、前記親水膜形成後の内面の算術平均粗さRa1との関係が、0.8×Ra0≦Ra1であること、
     を特徴とする自動分析装置。
  2.  前記圧力センサユニットの液体の流入出口の1つあたりの断面積をα、前記圧力センサユニットの受圧面の面積をβとしたとき、α<βであること、
     を特徴とする請求項1に記載の自動分析装置。
  3.  前記圧力センサユニットの内面形状が円錐であり、
     前記円錐の頂点部が鉛直上方となるよう配置され、
     前記円錐の底周面の接線方向に前記圧力発生手段と接続される液体の流入出口が設けられ、
     前記頂点部に前記ノズルと接続される液体の流入出口が設けられ、
     前記円錐の底面に前記圧力センサユニットの受圧面が配置されていること、
     を特徴とする請求項1に記載の自動分析装置。
  4.  前記シリカ粒子および前記アルミナ粒子の平均粒子径が10~20nmであること、
     を特徴とする請求項1に記載の自動分析装置。
  5.  前記シリカ粒子および前記アルミナ粒子の割合が30~60質量%であること、
     を特徴とする請求項1に記載の自動分析装置。
  6.  前記圧力センサユニットの圧力センサと前記圧力センサユニットの筐体である接続容器との間にO-リングが装着されており、前記O-リングの表面に前記親水膜が形成されていること、
     を特徴とする請求項1に記載の自動分析装置。
  7.  前記流入出口における前記流路と接する面には前記親水膜が形成されていないこと、
     を特徴とする請求項3に記載の自動分析装置。
  8.  前記親水膜の膜厚が50nm以上であること、
     を特徴とする請求項1に記載の自動分析装置。
  9.  前記親水膜の算術平均粗さが0.08μm以上であること、
     を特徴とする請求項1に記載の自動分析装置。
  10.  内面にシリカ粒子およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含んでなる親水膜が形成されており、前記内面の基材の算術平均粗さRa0と、前記親水膜形成後の内面の算術平均粗さRa1との関係が、0.8×Ra0≦Ra1である圧力センサユニットを備えた分注機構に接続されたノズルにより液体を吸引・吐出すること、
     を特徴とする自動分析方法。
  11.  圧力センサユニットのすべての流入出口にねじ部材をねじ込むねじ込み工程と、
     前記流入出口に前記ねじ部材をねじ込んだ圧力センサユニットの内面に、シリカ粒子およびアルミナ粒子のうちの少なくとも一方と、ケイ素化合物と、を含む親水膜用コート液を塗布する塗布工程と、
     加熱して、前記内面に塗布した親水膜用コート液を親水膜にする製膜工程と、
     を有し、
     前記内面の基材の算術平均粗さRa0と、前記親水膜形成後の内面の算術平均粗さRa1との関係が、0.8×Ra0≦Ra1であること、を特徴とする圧力センサユニットの製膜方法。
