Abstract
본 발명은 LMNA 유전자가 변형된 줄기세포주 및 이를 이용하에 제작한 심근병증 심장 오가노이드에 관한 것으로, 본 발명에서는 LMNA 유전자 서열의 특정 염기 서열을 유전자 편집 기술로 변형함으로써 확장성 심근병증을 모델링한 세포주를 제작하였으며, 이를 이용하여 확장성 심근병증의 특성이 잘 반영된 확장성 심근병증 심장 오가노이드를 제작하였으므로, 이를 LMNA 유전자 변형에 기인한 심근병증의 모델링에 이용할 수 있으며, 심근병증 치료제 스크리닝에 유용하게 활용할 수 있다.
Description
본 발명은 LMNA 유전자가 변형된 줄기세포주 및 이를 이용하에 제작한 심근병증 심장 오가노이드에 관한 것이다.
우리 몸에 혈액을 내보내는 역할을 하는 심장은 심근(심장근육)으로 구성되어 있는데, 심근의 기능에 이상이 발생하면 제대로 펌프 역할을 할 수 없게 되고, 이로 인해 혈액 순환이 제대로 이루어지지 않으므로 생명이 위협받게 된다. 이러한 심근 기능의 이상에 의해 발생하는 병을 총칭하여 심근병증(cardiomyopathy)이라 하며, 정상적으로 수축하는 심장 근육의 부분 혹은 전체가 변화하는 질환이다. 심근병증은 원인을 밝힐 수 없는 경우가 많으므로, 대개 그 형태적인 특징에 따라 확장성, 제한성, 비후성 등으로도 구분한다. 확장성 심근병증은 좌심실이 확장되면서 수축 기능이 저하되는 경우이고, 제한성 심근병증은 좌심실벽이 두꺼워지면서 주로 좌심실의 이완기 충만에 문제가 발생하는 경우이며, 비후성 심근병증은 좌심실의 일부 또는 전체가 특별한 이유 없이 두꺼워지는 경우이다. 일부 환자에서는 이 세 가지 형태 중 두 가지 이상이 한꺼번에 나타나거나 또는 순차적으로 나타날 수도 있다. 우리나라에서는 매년 약 10,000여명의 새로운 심근병증 환자가 발생된다고 알려져 있으며, 심근병증에 의한 사망자는 대부분 급사자로 총 사망자의 약 1%를 차지한다고 추정될 뿐, 아직 국내 역학이나 통계가 정확히 발표된 바가 없다. 그러나, 미국 질병관리센터(center for disease control)에서 35세 이상의 사망자를 대상으로 조사한 자료에 따르면 심장 돌연사의 발생률은 연 0.1~0.2%로 1년에 20~40만명이 심장 돌연사로 사망하고 있으며, 이러한 심장 돌연사의 원인은 약 80%가 관동맥질환이다 (Zipes DP et al., 1998, Circulation, 98:2334-2351.). 즉, 심장에 혈액을 공급하는 관상동맥의 심한 협착 또는 폐색 등이 발생하면 심근세포의 생존에 필요한 산소 및 영양소 공급이 차단되고, 그 결과 심근세포가 사멸하거나 심각한 심근 기능장애(심근병증)가 나타나게 되며, 이로 인해 심장 돌연사가 발생하게 됨을 알 수 있다. 특히, 심근세포는 재생, 증식하는 능력이 미약한 세포로서, 산소 및 영양소 공급의 차단으로 인한 심근세포의 사멸은 심장기능의 악화에 큰 영향을 미친다.
이 중, 확장성 심근병증(dilated cardiomyopathy, DCM)은 심장비대증으로도 불리며 좌심실의 벽이 얇아지고 늘어나면서 심실내강이 비정상적으로 확장되는 질환이다. 혈액을 온몸으로 순환시킬 수 있도록 하는 이완과 수축을 제대로 하지 못하는 증상을 가지고 있으며, 국내에서는 10만 명 당 1~2명이 이 질환을 앓는 것으로 알려졌다. 이 중 14.3%가 LMNA 유전자의 변형에 인한 것으로 알려져 있다. LMNA는 Lamin A/C를 코딩하는 유전자로서 핵 내부에 위치하여 층을 이루고 있다. Lamin A/C는 세포핵의 물리적인 안정성을 유지하는 핵막의 기능을 담당한다. 또한, 세포핵 내부의 유전자 전사 조절, 세포분열, 세포분화, 세포내 운반 등 생리학적 기능도 수행한다. Lamin A/C은 Lamin-associated domains(LADs)와 상호작용하여 세포핵의 구조와 기능을 유지하는 데에 중요한 역할을 한다. 이외에도 Lamin A/C는 핵골격과 세포골격 사이의 연결을 담당하는 단백질인 LINC(Linker of Nucleoskeleton and Cytoskeleton)와 상호작용하며 세포내의 이동, 세포 외부와의 상호작용 등에도 관여한다. LMNA에 이상이 있을 경우, 심장근육세포의 구조적 안정성을 손상시키고 심근병증을 유발할 수 있다.
인체의 손상된 조직을 재건하기 위한 이식방법에는 이종이식, 동종이식, 자가이식이 있다. 이종이식은 면역 부적합성 및 레트로바이러스를 포함하는 인수공통 병원체 전달의 문제를 갖는다. 또한, 동종이식은 제공자의 면역거부 및 이용불가능성의 문제를 가지며, 자가이식은 필요한 양만큼의 적절한 조직을 얻기 어려우며 환자에 대한 외상의 증가의 문제를 가진다. 이러한 문제점을 해결하기 위하여, 인공 대체물이나 세포를 배양하여 조직화시킨 것을 그대로 이식하려고 하는 기술이 주목받고 있다. 여러 종류의 세포로 분화할 수 있는 미분화 세포인 인간 다능성 줄기세포는 심근세포로 분화 가능하며, 2차원 배양 시스템에서 생성된 인간 다능성 줄기세포 유래 심근세포에 관한 많은 연구를 진행하고 있다. 이러한 인간 다능성 줄기세포 유래 심근세포는 심장발달, 약물 스크리닝, 질병 모델링 및 심장 회복 연구에 광범위하게 사용될 수 있다. 하지만, 기존 2차원 배양 시스템으로부터 생성된 인간 다능성 줄기세포 유래 심근세포는 배아 또는 태아 단계와 유사한 구조적 및 기능적으로 미성숙한 특성을 나타내기 때문에 성숙한 성인 심근세포와 달리 질병 모델링, 약물 스크리닝 및 심장 독성을 위한 활용에 한계점을 갖고 있다. 2004년 순수한 세포 자체를 이용하여 제작한 셀 시트(cell sheet)를 이용한 망막 재생에 관한 연구가 New England journal of Medicine에 발표된 뒤 조직공학 분야에서 세포만을 이용한 3차원 인공조직에 관한 연구가 활발히 진행되고 있다.
조직공학 분야에서 최근 인공 조직보다 주목받고 있는 것이 인공 장기 또는 미니 장기라고 불리는 오가노이드(organoid)이다. 현재 생체 내 여러 장기의 줄기세포에서 유래하는 다양한 종류의 오가노이드 모델이 구축되어 있으며, 재생의학이나 세포치료제로써 오가노이드에 대한 연구가 활발하게 진행되고 있다. 오가노이드는 줄기세포로부터 발생 및 분화를 통하여 형성된 3차원 세포 구조체로서, 해당 장기의 구조적, 기능적 특성을 재현하는 소형 장기 모사체이다. 오가노이드는 3차원 세포 구조체로서 조직을 구성하고 있는 다양한 세포로 구성되어 있고 세포외 기질(ECM, extracellular matrix), 세포 유형 및 가용성 인자를 포함하는 미세 환경을 제공해 주어 기존의 2차원 배양 시스템보다 우수한 신체와 동일한 구조로 만들어 맞춤형 치료제 개발 및 임상 실험 대체 가능성이 있는 후보 물질로 각광받고 있다. 따라서, 기초 생물학 연구 분야에서부터 신약개발, 질병 모델링, 재생 치료 등 다양한 응용 연구 분야에 이르기까지 폭넓게 활용되고 있다. 현재 다양한 종류의 오가노이드가 생체 내 여러 장기의 줄기세포 및 조직에서 유래하여 모델링 되어 있으며, 재생의학이나 세포치료제로써 오가노이드에 대한 연구가 활발하게 진행되고 있다.

본 발명에서는 LMNA 유전자 서열의 특정 염기 서열을 유전자 편집 기술로 변형함으로써 확장성 심근병증을 모델링한 세포주를 제작하였으며, 이를 이용하여 확장성 심근병증의 특성이 잘 반영된 확장성 심근병증 심장 오가노이드를 제작하였으므로, 이를 LMNA 유전자 변이에 기인한 심근병증의 모델링에 이용할 수 있으며, 심근병증 치료제 스크리닝에 유용하게 활용할 수 있다.
도 1 및 2는 염기 교정기법 및 이를 이용한 인간 유래 배아 줄기세포 세포주 및 인간 유래 섬유아세포주의 LMNA 유전자에 단일염기서열 변이를 유도하고, 표적심층서열분석 방법으로 C에서 T로의 염기 치환 효율을, 생어 시퀀싱(Sanger sequencing)으로 선별한 세포 콜로니의 세포의 유전자형을 확인한 도이다.

본 발명에서 용어, "오가노이드 (organoid)"는 줄기세포를 이용해 최소 기능을 할 수 있도록 만든 '미니 유사 장기’로서, 3차원 구조로 만들어져 실험실에서도 실제 신체 기관과 비슷한 환경을 만들 수 있는 것이 특징이다. 즉, "오가노이드(organoid)"는 3D 입체구조를 가지는 세포를 의미하며, 동물 등에서 수집, 취득하지 않은 인공적인 배양 과정을 통하여 제조한 신경, 장 등의 장기와 유사한 모델을 의미한다. 이를 구성하는 세포의 유래는 제한되지 않는다. 상기 오가노이드 (organoid)는 세포의 성장 과정에서 주변 환경과 상호 작용하도록 허용되는 환경을 가질 수 있다. 2D 배양과는 달리, 3D 세포 배양은 체외에서 세포가 모든 방향으로 성장할 수 있다. 이에 따라 본 발명에서 3D 오가노이드는 실제로 생체 내에서 상호 작용을 하고 있는 장기를 거의 완벽히 모사하여, 질병의 치료제 개발 및 등을 관찰할 수 있는 훌륭한 모델이 될 수 있다.
본 발명에서 용어, "분화"는 세포가 분열하여 증식하며 전체 개체가 성장하는 동안에 세포의 구조나 기능이 특수화되는 현상을 의미한다. 즉, 생물의 세포, 조직 등이 각각에게 주어지는 역할을 수행하기 위해 적합한 형태 및 기능으로 변하는 과정을 말하며, 예를 들어, 만능 줄기세포가 외배엽, 중배엽 및 내배엽 세포로 변하는 과정뿐 아니라 전구세포가 특정 분화형질을 발현하게 되는 것도 모두 분화에 포함될 수 있다.

본 발명에서 용어, "투여"란, 어떠한 적절한 방법으로 대상에게 특정한 조성물을 도입하는 것을 의미하며, 본 발명에서 용어, "도입"은 "처리", "주입", "주사" 또는 "감염"과 혼용되어 사용할 수 있다.
본 발명에서, 본 발명의 동물 모델에 물질을 투여하는 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 예를 들어, 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여될 수 있으나, 이에 제한되지는 않는다.
본 발명에서 "스크리닝"이란, 여러 물질로 이루어진 후보군으로부터 목적으로 하는 어떤 특정한 성질을 갖는 물질을 특정한 조작 또는 평가 방법으로 선별하는 것이다. 본 발명의 목적상, 본 발명의 스크리닝은, 본 발명에 의한 심근병증 모델 세포주, 오가노이드 또는 동물모델에 심근병증 예방용 또는 치료용 후보 물질을 처리하고, 상기 후보 물질에 의하여 심근병증이 예방되거나, 치료되거나, 대조군 물질에 비하여 예후가 증진된 경우에 후보 물질을 심근병증 예방제 또는 치료제로 결정하는 것이다.
본 발명에서 "치료" 또는 "경감"이란 시험 물질 또는 후보 물질의 투여로 심근병증의 증세를 치료, 호전시키거나 이롭게 변경하는 모든 행위를 의미한다. 구체적으로, "치료"는 이롭거나 바람직한 임상적 결과를 수득하기 위한 접근을 의미한다. 본 발명의 목적을 위해서, 이롭거나 바람직한 임상적 결과는 비제한적으로, 증상의 완화, 질병 범위의 감소, 질병 상태의 안정화 (즉, 악화되지 않음), 질병 진행의 지연 또는 속도의 감소, 질병 상태의 개선 또는 일시적 완화 및 경감 (부분적이거나 전체적으로), 검출가능 여부를 포함한다. "치료"는 치료학적 치료 및 예방적 또는 예방조치 방법 모두를 가리킨다. 상기 치료들은 예방되는 장애뿐만 아니라 이미 발생한 장애에 있어서 요구되는 치료를 포함한다. 질병을 "완화 (palliating)"하는 것은 치료를 하지 않은 경우와 비교하여, 질병상태의 범위 및/또는 바람직하지 않은 임상적 징후가 감소되거나 및/또는 진행의 시간적 추이(time course)가 늦춰지거나 길어지는 것을 의미한다.
본 발명의 스크리닝 방법에 의해 분석되는 심근병증 치료제 후보 물질 (또는 시험 물질)은 단일 화합물, 화합물들의 혼합물 (예컨대, 천연 추출물 또는 세포 또는 조직 배양물), 또는 바이오의약품 (예컨대, 단백질, 항체, 펩타이드, DNA, RNA, 안티센스 올리고뉴클레오티드, RNAi, 앱타머, RNAzyme 및 DNAzyme) 일 수 있다. 상기 시험 물질은 합성 또는 천연 화합물의 라이브러리로부터 얻을 수 있으며 이러한 화합물의 라이브러리를 얻는 방법은 당업계에 공지되어 있다.
이러한 측면에서 본 발명의 방법은 대조군 세포주, 오가노이드 또는 동물 모델을 제조하는 단계, 대조군 샘플에 후보 물질을 처리하는 단계 및 후보 물질이 심근병증의 치료 또는 경감의 효과가 있는지를 대조군과 비교 평가하여 선정하는 단계를 추가로 포함할 수 있다.
일 측면에서, 본 발명은 심근병증 모델로 사용하기 위한, 본 발명의 심근병증 심장 오가노이드의 용도에 관한 것이다.
하기의 실시예를 통하여 본 발명을 보다 상세하게 설명한다. 그러나 하기 실시예는 본 발명의 내용을 구체화하기 위한 것일 뿐 이에 의해 본 발명이 한정되는 것은 아니다.
실시예 1. LMNA 유전자 변이 유도 및 확인

유전자 변이 유도
GWAS 연구를 통해 LMNA 유전자에서 종래에 확장성 심근병증(DCM)과의 관련성에 대해 보고되지 않은 SNP(rs28928901)를 심근병증 모델의 세포주 제작을 위한 타겟으로 선정하고, LMNA-rs28928901-sg Gx20 (+): 5'-CCGT" C "ATGAGACCCGACTGG TGG -3' (서열번호 3)에서 5'-CCGT" T "ATGAGACCCGACTGG TGG -3' (서열번호 2)로 단일염기변이 (C>T, c.665C>T) (H222Y)를 유도하기 위한 sgRNA를 설계한 뒤, sgRNA의 적용가능성을 다양한 세포주에서 확인하기 위해, Cas9 단백질, PAM 서열 5'-TGG-3' 및 설계된 sgRNA (5'-CCACCAGUCGGGUCUCAUAACGG-3', 서열번호 1)를 포함하는 시토신 염기교정기(cytocine base editor: CBE)를 이용하여 LMNA 유전자 돌연변이 세포주를 제작하였다. 구체적으로, 인간 유래 섬유아세포, 인간 유래 배아 줄기세포 및 인간 유래 유도만능 줄기세포를 각각 sgRNA 발현 벡터로 형질전환한 후 단일 세포로 분리하여 콜로니(colony)로 배양하고, 각 콜로니에서 세포를 채취하여 표적심층서열분석으로 C에서 T로의 염기 치환을 확인하였다.
LMNA 유전자 돌연변이 유도를 위한 sgRNA 염기서열 및 이를 이용한 형질전환 결과 인간 유래 배아줄기세포에서 유전자 편집이 이루어지지 않은 야생형(wild type), 50% 치환이 발생한 이형(hetero type), 100% 치환이 발생한 동형(homo type)의 콜로니를 확인할 수 있었다 (도 1). 또한, 인간 유래 섬유아세포에서 유전자 편집이 이루어지지 않은 야생형, 100% 치환이 발생한 동형의 세포를 확인할 수 있었으며, 각각의 유전자형을 생어 시퀀싱(Sanger sequencing)으로 다시 확인하였다 (도 2). LMNA 유전자 편집을 위한 sgRNA는 인간 유래 유도만능줄기세포에서는 유전자 편집을 유도하지 않았다 (데이터 미도시).
1-2. 표적 서열 외 변이 유무 확인
상기 실시예 1-1에서 제작한 돌연변이 세포주의 LMNA 표적에 대한 안전성을 검증하기 위해 표적 외 절단 유무를 확인하였다. 구체적으로, 세포 수준에서 LMNA 표적에 대한 표적 외 절단 가능성이 있을 것으로 판단되는 표적들을 RGEN tools 프로그램을 통해 3개의 염기 불일치까지 허용하면서 표적 외 절단 가능성이 있는 서열들을 선별하였다. 또한, LMNA 표적에 대해 디자인된 라이브러리 프라이머(library primer)를 이용하여 NGS 라이브러리를 제작하고, 원하는 유전자 변이가 도입된 세포주를 통해 선별된 표적 외 절단 서열들을 표적심층서열 분석 방법(Targeted deep sequencing)으로 분석하였다.
그 결과, 5개의 편집 가능성이 유망한 표적 서열들을 확인하였으며, 표적 외 절단 표적 서열 5영역 모두에서 염기 치환이 발생하지 않았음을 확인하였다 (도 3).

LMNA
유전자가 변이된 줄기세포주의 형태학적 관찰
LMNA 유전자 교정용 sgRNA 및 BE4max를 이용하여 유전자 편집을 수행한 인간 유래 배아줄기세포의 동형(Homo type) 콜로니의 세포 (H222Y)와 교정하지 않은 정상 배아줄기세포 (Control)의 형태를 비교하기 위해, 10cm 배양 디쉬에 약 50% 밀도로 세포를 배양하고 배아줄기세포의 특성인 콜로니 형성을 현미경으로 관찰하였다.
그 결과, 대조군 줄기세포주와 LMNA 변이 줄기세포주를 형태학적으로 관찰한 결과 두 세포주 모두 콜로니의 형성이 잘 된 것을 확인할 수 있었다 (도 4).
실시예 2. 확장성 심근병증 심장 오가노이드 모델 제작
2-1. 심장 오가노이드 제작
상기 실시예 1에서 구축한 세포주 중에서 LMNA 변이 동형 줄기세포주로 확장성 심근병증(dilated cardiomyopathy, DCM)을 모델링하는 심장 오가노이드 (H222Y)를 제작하고, 정상 WT형 줄기세포주로 대조군 심장 오가노이드 (Control)를 제작하였다. 구체적으로, 100mm 배양 플레이트에 마트리젤 (Matrigel In DMEM F-12)을 코팅한 후, E8 배지 (STEMCELL Technologies) 10ml과 정상 줄기세포주 또는 LMNA 유전자 변이 줄기세포주를 분주하고, 3일 동안 매일 E8 배지를 교체하여 배양하였다. 이 후, 아큐테이즈로 각각 단일세포로 분리한 뒤, 96-웰 U 플레이트에 2μM의 Thiazovivin (Rho kinase 억제제)이 포함된 E8 배지를 이용하여 한 웰당 5000, 7500 또는 10000개의 세포를 분주하고, 4℃에서 3분 동안 1000rpm으로 원심분리한 뒤 인큐베이터에서 24시간 동안 배양하여 세포 응집체를 형성하였다. 24시간 후, 6μM의 농도의 GSK-3 억제제인 CHIR99021 (Sigma Aldrich)을 첨가한 RPMI1640+B27-인슐린 배지 (B-27 Supplement, minus insulin RPMI1640)에서 48시간 동안 배양하여 분화를 유도하고, 분화 2일차에, 2μM IWP2를 배지에 추가하여 48시간 동안 배양하였다. 분화 4일 및 5일차에 RPMI1640+B27-인슐린 배지로 교체하여 배양하고, 분화 7일차에 2일에 한 번씩 RPMI1640+B27-비타민 A 배지 (B-27 Supplement, minus vitamin A RPMI1640)로 교환해주면서 분화 20일차까지 배양하였다 (도 5a).
2-2. 세포수에 따른 분화율 분석
세포 응집체 형성시 분주된 세포수에 따른 분화율을 확인하기 위해, 분화 유도 후 8일, 15일 및 20일에 각 오가노이드의 박동을 백분율로 분석하였다. 또한, cTnT 및 MLC2v의 발현을 중합효소연쇄반응(qRT-PCR)로 분석하여 분화율을 확인하였다.
그 결과, 분주된 세포수가 10000개인 경우에 비해, 5000 및 7500개인 오가노이드에서 분화 8일차에 박동율이 현저히 높게 나타났으며 (도 5b), qRT-PCR 분석 결과, cTnT 및 MLC2v의 발현이 세포수 5000에 비해 7500인 경우의 DCM 심장 오가노이드 (H222Y)에서 특이적으로 증가한 것으로 나타나 (도 5c 및 d), DCM 심장 오가노이드 제작을 위한 최적 세포수를 7500개 (2.4 x 103개/cm2)로 선정하고 이후의 실험에 적용하였다.
실시예 3. DCM 심장 오가노이드 모델의 심장 마커 발현 및 심근세포 타입 확인
정상 WT형 줄기세포주로 제작한 대조군 심장 오가노이드와 DCM 심장 오가노이드의 분화율을 비교하기 위해, 심장 마커인 cTnT,(cardiac troponin T) MLC2a(Myosin light chain 2a) 및 MLC2v(Myosin light chain 2v)의 단백질 발현 분석을 웨스턴 블롯 및 면역형광염색법을 이용하여 분석하였다. 구체적으로, 항-cTnT 항체 (1:1000), 항-MLC2a 항체 (1:2000) 및 항-MLC2v 항체 (1:2000)를 사용하여 웨스턴 블롯 분석을 수행하였다. 또한, 면역형광염색을 위해, 심근세포 오가노이드를 4% PFA를 이용하여 한 시간 동안 고정한 후, PBS로 세척하고, 항-cTnT 항체 (1:500), 항-MLC2a 항체 (1:500) 및 항-MLC2v 항체 (1:400)를 1차 항체로 사용하여 세포를 12시간 동안 4℃에서 각각 반응시켰다. 이 후, 2차 항체로서 goat 항-mouse IgG1 Alexa 488 및 goat 항-rabbit IgG Alexa 594 항체와 상온에서 각각 30분간 반응시키고 추가로 DAPI와 함께 30분 더 반응시키고, 형광현미경으로 관찰하였다.
웨스턴 블롯 분석을 수행한 결과, cTnT, MLC2a 및 MLC2v의 단백질 발현이 두 심장 오가노이드 사이에서 유의적인 차이가 없는 것으로 나타났다 (도 6a). 또한, 면역형광염색법으로 분석한 결과, 웨스턴 블롯 분석과 같이 cTnT, MLC2a 및 MLC2v의 단백질 발현 정도가 두 심장 오가노이드 사이에서 유의적인 차이가 없는 것으로 나타났다 (도 6b 및 c).
실시예 4.
DCM 심장 오가노이드 모델의 DCM 질병 특성 분석
4-1. LADs 마커 발현 분석
대조군 심장 오가노이드와 DCM 심장 오가노이드에서 LADs(Lamin A/C 관련 도메인) 마커들 발현 정도를 비교하기 위해, LMNA(Lamin A/C), LB1(Lamin B1), Emerin, SUN 및 LMNA와 상호작용하는 크로마틴 H3k9me2의 발현을 항-Lamin A/C 항체 (1:2000), 항-LB1 항체 (2:1000), 항-Emerin 항체 (1:1000), 항-SUN 항체 (1:1000) 및 항-H3k9me2 항체 (1:1000)를 사용하여 웨스턴 블롯 분석으로 확인하고, 항-Lamin A/C 항체 (1:2000), 항-LB1 항체 (2:1000), 항-Emerin 항체 (1:1000), 항-SUN 항체 (1:1000) 및 항-H3k9me2 항체 (1:1000)를 사용하여 면역형광염색법으로 분석하였다.
웨스턴 블롯 분석 결과, Emerin을 제외한 Lamin A/C, LB1, SUN 및 H3k9me2가 대조군 심장 오가노이드에 비해 DCM 심장 오가노이드에서 상대적으로 낮은 발현율을 나타냈다 (도 7a). 또한, 면역형광염색법으로 LADs의 발현을 비교한 결과, 웨스턴블롯 결과와 마찬가지로 Emerin을 제외한 Lamin A/C, LB1, SUN 및 H3k9me2가 대조군 심장 오가노이드 (Control)에 비하여 DCM 심장 오가노이드 (H222Y)에서 상대적으로 낮은 발현율을 나타냈다 (도 7b 및 c).
4-2. 핵 모양 관찰
대조군 심장 오가노이드와 DCM 심장 오가노이드를 구성하는 세포의 핵 모양을 비교하기 위해, 오가노이드 분화 20일차에 아큐테이즈를 이용하여 각각 단일 세포로 만든 뒤, 챔버 세포 컬쳐 슬라이드 웰 당 1x105개로 분주하여 최대 3일 동안 안정화시켰다. 그 후, DAPI로 상온에서 30분간 염색하여 콘포칼 현미경으로 이미지를 얻어 Image J로 정량하였다. 그 결과, 대조군 오가노이드에 비해 DCM 오가노이드의 핵 모양이 비정상적으로 변형된 것을 확인할 수 있었다 (도 8).
4-3. 혈관 형성 분석
대조군 심장 오가노이드와 DCM 심장 오가노이드에서의 혈관 생성의 유무를 확인하기 위해, 혈관 관련 유전자인 vWF, CD31, PDGFR-ß및 α-SMA의 발현을 웨스턴 블롯 분석 및 면역형광염색으로 확인하였다.
웨스턴 블롯 분석 결과, vWF, CD31, PDGFR-ß및 α-SMA의 발현 정도가 대조군 심장 오가노이드에 비해 DCM 심장 오가노이드에서 현저히 낮은 것으로 나타났다 (도 9a). 또한, 면역형광염색법으로 CD31의 발현을 확인한 결과, 대조군 심장 오가노이드에 비해 DCM 심장 오가노이드에서 혈관 형성 유도가 되지 않은 것을 확인하였다 (도 9b).
DCM 심장 오가노이드 모델에서 혈광 형성이 억제된 것을 확인함으로써, DCM 질병의 특성이 본 발명의 LMNA 유전자 변형된 세포주 유래 DCM 심장 오가노이드 모델에 잘 반영된 것을 확인하였다.
상기 실시예에서와 같이, 본 발명에서는 인간 줄기세포인 배아 줄기세포를 사용하여 확장성 심근병증 유발 유전자 변이를 선별하였고, 크리스퍼 유전자 편집 방법 중 염기 수준에서 변이 유도가 가능한 염기 교정 방법을 적용하여 인간의 확장성 심근병증 유도 유전자 변이 표적인 LMNA 유전자가 변이된 세포주를 확보하였으며, 확보된 세포주를 이용하여 심장 오가노이드로 분화 유도함으로써 확장성 심근병증 오가노이드 모델을 제작하였다.
Claims (21)
- LMNA(lamin A/C) 유전자에 단일염기서열 변이(single nucleotide variant, SNV)를 포함하는 심근병증(cardiomyopathy) 모델 세포주.
- 제 1항에 있어서, LMNA 유전자의 665번째 cDNA(coding DNA)인 사이토신이 타이민으로 (c.665C>T) 단일 염기 편집된, 심근병증 모델 세포주.
- 제 2항에 있어서, 유전자 편집이 p.H222Y(His222Tyr)를 유도하는, 심근병증 모델 세포주.
- 제 1항에 있어서, SNP rs28928901를 포함하는, 심근병증 모델 세포주.
- 제 1항에 있어서, 세포는 인간 유래 섬유아세포 또는 인간 유래 다능성 줄기세포인, 심근병증 모델 세포주.
- 제 5항에 있어서, 인간 유래 다능성 줄기세포는 배아줄기세포(embryonic stem cells, ESCs) 또는 유도 만능 줄기세포 (induced pluripotent stem cells, iPSCs, 역분화 줄기세포)인, 심근병증 모델 세포주.
- a) 세포주에 시토신 염기 교정기를 이용하여 LMNA 유전자에 C>T의 단일염기변이를 유도하는 단계; 및b) 동형(homo type)의 콜로니를 분리하는 단계를 포함하는 심근병증 모델 세포주의 제조방법.
- 제 7항에 있어서, 시토신 염기 교정기는 가이드 RNA, Cas9 단백질 및 시토신 디아미네이즈(deaminase)를 포함하는, 심근병증 모델 세포주의 제조방법.
- 제 8항에 있어서, 가이드 RNA는 서열번호 1의 염기서열을 포함하는, 심근병증 모델 세포주의 제조방법.
- a) 제 1항의 심근병증 모델 세포주를 분주 및 배양하여 세포 응집체를 형성하는 단계; 및b) 세포 응집체를 심장 오가노이드로 분화시키는 단계를 포함하는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항에 있어서, 심근병증 모델 세포주를 1 x 103개/cm2 내지 1 x 105개/cm2의 밀도로 분주하는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항에 있어서, 심근병증 모델 세포주를 분주하여 원심분리한 뒤 배양하는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항에 있어서, 심근병증 모델 세포주를 Rho kinase 억제제가 포함된 배지에서 배양하여 세포 응집체를 형성하는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항에 있어서, 심근병증 모델 세포주를 0.5 내지 2일 동안 배양하여 세포 응집체를 형성하는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항에 있어서, 세포 응집체를 심장 오가노이드로 분화시키는 단계는:i) 세포 응집체를 GSK-3 억제제를 포함하는 RPMI1640+B27-인슐린 배지 (B-27 Supplement, minus insulin RPMI1640)에서 배양하는 단계;ii) Wnt 저해제를 처리하고 배양하는 단계;iii) RPMI1640+B27-인슐린 배지 (B-27 Supplement, minus insulin RPMI1640)에서 배양하는 단계; 및iv) RPMI1640+B27-비타민 A 배지 (B-27 Supplement, minus vitamin A RPMI1640)에서 배양하는 단계를 포함하는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항에 있어서, 분화는 20 내지 40일 동안 수행되는, 심근병증 심장 오가노이드의 제조방법.
- 제 10항의 제조방법으로 제조된 심근병증 심장 오가노이드.
