Abstract
일 실시예에 따른 3차원 이미징 시스템은, 컴퓨터 프로그램을 수행하는 명령어를 저장하는 메모리, 상기 명령어를 수행하는 프로세서, 빛이 발생하도록 구성되는 광원, 상기 광원에서 발생되는 빛이, 형광 입자로 표지된 샘플에 조사되도록 하는 광경로를 형성하고, 상기 광경로를 통해 전달되는 빛이 상기 형광 입자로 표지된 샘플이 위치한 미리 설정된 영역에 광초점이 형성되도록 하고, 상기 광초점에 조사된 빛에 의해 상기 형광 입자로부터 방출되는 형광 신호를 측정하는 비선형 현미경 장치를 포함하되, 상기 프로세서에 의해 상기 명령어가 실행되면, 상기 프로세서는, 상기 형광 입자로부터 방출되는 형광 신호와 상기 광초점의 위치를 기반으로 3차원 영상을 생성하고, 상기 광원은 연속파동을 가지는 빛이 발생하도록 구성되는 연속파동 레이저를 출력하고, 상기 형광 입자는 상기 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 형광 입자일 수 있다.
Description
본 발명은 연속파동 레이저를 이용하는 비선형 현미경 기반의 3 차원 이미징 시스템 및 방법에 관한 것으로, 보다 상세하게 연속파동 레이저 여기(excitation)에 비선형 광학 반응(nonlinear optical response)을 나타내는 형광 입자를 이용하여 비선형 현미경을 구현하고, 이를 기반으로 3차원 영상을 생성하는 시스템 및 방법에 관한 것이다.
본 연구는 2020년도 과학기술정보통신부(정부)의 재원으로 한국연구재단의 지원을 받아 수행된 연구사업의 3차원 볼륨 동시 측정 홀로그래픽 in-vivo 이광자 현미경을 통한 뇌활성지도 작성(No. 1711170194)과 관련된다.
비선형 현미경은 샘플(시료)의 3차원 형광을 이미징하는데 있어서 공초점 현미경과 함께 생명과학 분야에서 가장 널리 사용되는 현미경이다. 비선형 현미경 기반의 3차원 이미징 기법은 형광 물질의 비선형 여기 또는 비선형 산란을 통해 여기 빛이 모이는 초점에서 유의미한 비선형 신호가 발생하는 현상을 이용한다. 따라서 비선형 현미경을 이용하는 경우, 3차원 영상을 이미징 시 공초점 현미경을 이용하는 경우와는 달리, 검출 경로에 별도의 핀홀이 필요하지 않다. 또한, 비선형 여기의 경우 선형 여기를 사용하는 현미경 기법들에 비해 동일한 형광 물질을 대상으로 할 때 긴 파장대의 여기 빛을 사용하므로 상대적으로 빛의 산란이 적다는 이점이 있다. 따라서, 산란이 심한 시료(예를 들어, 생물체의 조직 혹은 세포)의 형광을 수집하는데 있어서 비선형 현미경을 이용하면 여타 현미경(예를 들어, 공초점 현미경)을 이용하는 경우에 비해 시료의 더 깊은 곳에 있는 정보까지 수집할 수 있다. 이러한 장점으로 인해 비선형 현미경은 세포/조직의 시각화 및 분석, 그리고 뇌 과학 등 다양한 생명과학 연구에서 필수적이다.
하지만 형광 물질의 비선형 신호 여기의 흡수 단면은 굉장히 작기 때문에, 비선형 현미경으로 3차원 이미징 기법을 구현함에 있어서 매우 높은 여기 세기를 지닌 광원(102 MWcm-2 내지 GWcm-2)이 필수적이다. 일반적으로 이러한 높은 세기의 여기 빛은 연속파동 레이저로는 얻을 수 없다. 따라서, 종래의 비선형 현미경 기반 3차원 이미징에는 비선형 신호 여기에 필요한 여기 세기를 얻을 수 있는 초고속 펄스 레이저가 필수적이다. 하지만, 고품질의 초고속 펄스 레이저는 수억 원(일반적으로 100k USD 이상)을 호가하며, 추가적으로 레이저의 유지/관리에 전문적인 인력과 비용이 수반된다. 이러한 경제적인 문제로 기존의 비선형 현미경 기반 3차원 이미징 기법은 그 편리성과 탁월성에도 불구하고 널리 보급되어 있지 않다.
한편, 공초점 현미경은 비선형 현미경에 비해 이미징 성능 측면에서 그 원리상 불리한 점을 가진다. 공초점 현미경은 선형 여기에 기반하기 때문에 가시광선 대역의 형광체들을 여기시키기 위해서는 형광 대역보다 에너지가 높은 짧은 파장의 빛을 사용해야 한다. 이처럼 짧은 파장대의 여기 파장의 빛 및 검출경로 상 핀홀의 존재로 인해, 공초점 현미경으로는 시료의 깊은 곳에 있는 형광 정보를 수집하기 어려우며, 측정과 동시에 시료의 3차원 전 영역에 영구적인 피해를 준다. 그럼에도 불구하고 공초점 현미경 기반 3차원 이미징 기법은 상대적으로 저렴한 가격의 연속파동 레이저(일반적으로 약 1k USD)로 구축이 가능하기 때문에, 생명과학 연구실에 가장 많이 보급되어 있는 실정이다.
본 발명의 기술적 과제는 초고속 펄스 레이저 대비 저렴한 연속파동 레이저를 이용하여 비선형 현미경 기반의 3차원 이미징 시스템 및 방법을 구축함에 있다.
본 발명의 다른 기술적 과제는 3차원 이미징 시 연속파동 레이저를 이용하는 경우 샘플에 발생하는 손상을 줄일 수 있는 비선형 현미경 기반의 3차원 이미징 시스템 및 방법을 제공함에 있다.
본 발명의 또 다른 기술적 과제는 3차원 이미징 시 연속파동 레이저를 이용하는 경우에도 고해상도의 3차원 영상을 획득할 수 있는 비선형 현미경 기반의 3차원 이미징 시스템 및 방법을 제공함에 있다.
본 발명의 해결하고자 하는 과제는 이상에서 언급한 것으로 제한되지 않으며, 언급되지 않은 또 다른 해결하고자 하는 과제는 아래의 기재로부터 본 발명이 속하는 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명의 일 실시예에 따르면, 3차원 이미징 시스템은 컴퓨터 프로그램을 수행하는 명령어를 저장하는 메모리, 상기 명령어를 수행하는 프로세서, 빛이 발생하도록 구성되는 광원, 상기 광원에서 발생되는 빛이, 형광 입자로 표지된 샘플에 조사되도록 하는 광경로를 형성하고, 상기 광경로를 통해 전달되는 빛이 상기 형광 입자로 표지된 샘플이 위치한 미리 설정된 영역에 광초점이 형성되도록 하고, 상기 광초점에 조사된 빛에 의해 상기 형광 입자로부터 방출되는 형광 신호를 측정하는 비선형 현미경 장치를 포함하되, 상기 프로세서에 의해 상기 명령어가 실행되면, 상기 프로세서는, 상기 형광 입자로부터 방출되는 형광 신호와 상기 광초점의 위치를 기반으로 3차원 영상을 생성하고, 상기 광원은 연속파동을 가지는 빛이 발생하도록 구성되는 연속파동 레이저를 출력하고, 상기 형광 입자는 상기 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 형광 입자일 수 있다.
여기서, 상기 미리 설정된 영역은 상기 형광 입자로 입사되는 여기 빛의 세기 대비 상기 형광 입자로부터 방출되는 형광 신호의 세기를 기반으로 설정되는 영역일 수 있다.
또한, 상기 미리 설정된 영역은 상기 형광 입자로부터 방출되는 형광 신호의 세기가 상기 형광 입자로 입사되는 여기 빛의 세기와 비선형 관계를 갖는 영역일 수 있다.
또한, 상기 광원에서 발생되는 빛의 파장은 상기 형광 입자로부터 방출되는 형광 신호의 파장보다 길 수 있다.
또한, 상기 형광 입자는 란탄족 이온이 도핑된 상향변환 입자일 수 있다.
또한, 상기 비선형 현미경 장치는 상기 광경로를 조절하는 복수개의 렌즈, 상기 복수개의 렌즈를 통해 전달된 광이, 상기 미리 설정된 영역에 상기 광초점을 형성하기 위한 대물렌즈, 상기 광초점의 위치를 조절하기 위한 위치 조절부;, 상기 비선형 형광 입자에서 방출되는 형광 신호를 필터링하기 위한 형광 필터링부, 및 상기 필터링된 형광 신호를 측정하기 위한 신호 측정부를 포함할 수 있다.
여기서, 상기 위치 조절부는 적어도 하나의 거울을 포함하고, 상기 프로세서는, 상기 적어도 하나의 거울의 위치를 제어하여 상기 광초점의 수평 위치를 조절하고, 상기 대물렌즈 및 상기 샘플이 놓이는 스테이지 중 적어도 하나를 제어하여, 상기 광초점의 수직 위치를 조절할 수 있다.
또한, 상기 상기 프로세서는, 상기 광초점의 위치 별로 해당 위치에서 측정된 형광 신호를 대응시켜 상기 3차원 영상을 생성할 수 있다.
본 발명의 다른 실시예에 따르면, 3차원 이미징 시스템에 의한 3차원 이미징 방법은 미리 설정된 영역 내에서 광초점이 형성되도록 설정하여, 광원에서 발생되는 빛을 형광 입자로 표지된 샘플에 조사하는 단계, 상기 광초점에서 상기 형광 입자로부터 방출되는 형광 신호를 측정하는 단계, 및 상기 형광 입자로부터 방출되는 형광 신호의 세기 정보와 상기 광초점의 위치 정보를 제공하는 단계를 포함하되, 상기 광원은 연속파동을 가지는 빛이 발생하도록 구성되는 연속파동 레이저이고, 상기 형광 입자는 상기 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 형광 입자일 수 있다.
본 발명의 또 다른 실시예에 따르면, 컴퓨터 판독 가능 기록매체에 저장되는 컴퓨터 프로그램은, 프로세서에 의해 실행되면, 미리 설정된 영역 내에서 광초점이 형성되도록 설정하여, 광원에서 발생되는 빛을 형광 입자로 표지된 샘플에 조사하는 단계와, 상기 광초점에서 상기 형광 입자로부터 방출되는 형광 신호를 측정하는 단계와, 상기 형광 입자로부터 방출되는 형광 신호의 세기 정보와 상기 광초점의 위치 정보를 제공하는 단계를 포함하는 3차원 이미징 방법을 상기 프로세서가 수행하도록 하기 위한 명령어를 포함하되, 상기 광원은 연속파동을 가지는 빛이 발생하도록 구성되는 연속파동 레이저이고, 상기 형광 입자는 상기 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 형광 입자일 수 있다.
본 발명의 또 다른 실시예에 따르면, 컴퓨터 프로그램을 저장하고 있는 컴퓨터 판독 가능 기록매체로서, 상기 컴퓨터 프로그램은, 프로세서에 의해 실행되면, 미리 설정된 영역 내에서 광초점이 형성되도록 설정하여, 광원에서 발생되는 빛을 형광 입자로 표지된 샘플에 조사하는 단계와, 상기 광초점에서 상기 형광 입자로부터 방출되는 형광 신호를 측정하는 단계와, 상기 형광 입자로부터 방출되는 형광 신호의 세기 정보와 상기 광초점의 위치 정보를 제공하는 단계를 포함하는 3차원 이미징 방법을 상기 프로세서가 수행하도록 하기 위한 명령어를 포함하되, 상기 광원은 연속파동을 가지는 빛이 발생하도록 구성되는 연속파동 레이저이고, 상기 형광 입자는 상기 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 형광 입자일 수 있다.
본 발명의 일 실시예에 따르면, 연속파동 레이저를 이용하여 비선형 현미경 기반의 3차원 이미징 시스템을 구현할 수 있기 때문에 초고가의 펄스 레이저를 이용하는 경우에 비해 비용을 현저히 줄일 수 있다.
본 발명의 일 실시예에 따르면, 형광 입자로부터 방출되는 형광보다 긴 파장대의 여기 빛을 사용하기 때문에 생체조직 내부를 보다 깊이 영상화할 수 있으며, 비침습적으로 고해상도의 영상을 획득할 수 있다.

아래에서는 첨부한 도면을 참고하여 본 발명의 실시예에 대하여 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 상세히 설명한다. 그리고 도면에서 본 발명을 명확하게 설명하기 위해서 설명과 관계없는 부분은 생략한다.

도 1을 참조하면, 본 발명의 일 실시예에 따른 3차원 이미징 시스템은 광원(110), 현미경 장치(120) 및 이미징 장치(130)를 포함한다.
광원(110)은 형광 입자가 표지된 샘플에서 형광을 여기시키기 위한 빛(이하, 여기 빛이라 함)이 발생하도록 구성된다. 본 실시예에서 광원(110)은 연속적인 파동을 가지는 빛(또는 빔)을 방출하는 연속파동 레이저로 구현될 수 있다. 상기 연속파동 레이저는 상기 형광 입자로부터 방출되는 형광 신호의 파장보다 긴 파장의 빛을 방출하도록 구성될 수 있다.
현미경 장치(120)는 광원(110)에서 방출되는 연속파동을 가지는 빛을 형광 입자로 표지된 샘플에 조사하여 미리 설정된 영역 내에서 광초점이 형성되도록 한다. 또한 현미경 장치(120)는 광초점에서 형광 입자로부터 방출되는 형광(또는 형광 신호)을 측정한다. 본 실시예에서 현미경 장치(120)는 비선형 현미경으로 구현될 수 있다. 비선형 현미경은 다광자(multiphoton) 광학 현미경이라 불릴 수 있으며, 이광자(two-photon) 형광 현미경 및 삼광자(three-photon) 형광 현미경을 포함할 수 있다.
한편, 본 실시예에서 샘플(관찰 대상)에 표지되는 형광 입자는 광원(110)으로부터 방출되는 연속파동을 가지는 여기 빛에 의해 비선형 광학 반응을 나타내는 형광 입자일 수 있다. 이하에서, 여기 빛에 비선형 광학 반응을 보이는 형광 입자를 비선형 형광 입자라 한다. 일 예로, 비선형 형광 입자는 란탄족 이온이 도핑된 상향변환 입자를 포함할 수 있다.
현미경 장치(120)는 형광 입자로 입사되는 여기(excitation) 빛의 세기 대비 해당 형광 입자로부터 방출되는 형광의 세기를 기반으로 특정 영역을 3차원 이미징을 위한 영역으로 설정할 수 있다. 예를 들어, 현미경 장치(120)는 형광 입자로부터 방출되는 형광의 세기가 해당 형광 입자로 입사되는 여기 빛의 세기에 대해 비선형으로 형광이 방출되는 영역을 샘플에 대한 3차원 이미징을 위한 영역으로서 설정할 수 있다.
현미경 장치(120)는 상기 설정된 영역 내에서 광초점의 위치를 이동시키면서 형광 입자로부터 방출되는 형광 신호를 측정할 수 있다. 여기서 상기 설정된 영역은 평면 상의 영역뿐만 아니라 3차원 공간 상의 영역을 포함한다.
일 실시예로서, 현미경 장치(120)는 빛의 크기를 조절하기 위한 복수개의 렌즈, 미리 설정된 영역 내에서 샘플 상에 광초점을 형성하기 위한 대물렌즈, 미리 설정된 영역 내에서 광초점의 위치를 조절하기 위한 위치 조절부, 형광 입자에서 방출되는 형광 신호를 필터링하기 위한 형광 필터링부, 및 상기 필터링된 형광 신호를 측정하기 위한 신호 측정부를 포함할 수 있다.
상기 위치 조절부는 적어도 하나의 거울을 포함할 수 있다. 여기서, 거울은 갈바노(galvano) 거울을 포함할 수 있다.
또한, 상기 위치 조절부는, 적어도 하나의 거울을 고정하는 제1지지부재와, 상기 제1지지부재에 결합되어 상기 제1지지부재의 위치를 이동시키는 제1이동부재(예, 모터 액추에이터 등)와, 상기 제1이동부재의 동작을 제어하는 명령어를 수록한 메모리와, 상기 메모리에 저장된 명령어를 실행하여 제1이동부재의 동작을 제어하는 신호를 제공하는 프로세서를 포함할 수 있다.
상기 프로세서가 제공하는 제어신호를 통해 적어도 하나의 거울의 이동이 제어될 수 있으며, 이에 따라 광초점의 수평 위치, 즉, X-Y축 위치가 조절될 수 있다.
또한, 상기 위치 조절부는 대물렌즈, 샘플이 놓이는 스테이지, 튜너블(tunable) 렌즈 등을 고정하는 제2지지부재와, 상기 제2지지부재에 결합되어 상기 제2지지부재의 위치를 이동시키는 제2이동부재(예, 모터 액추에이터 등)를 포함할 수 있다. 그리고, 메모리는 제2이동부재의 동작을 제어하는 명령어를 더 포함할 수 있으며, 상기 프로세서는, 상기 메모리에 저장된 명령어를 실행하여 제2이동부재의 동작을 제어하는 신호를 제공할 수 있다. 이에 따라, 상기 프로세서가 제공하는 제어신호를 통해 대물렌즈, 샘플이 놓이는 스테이지, 튜너블(tunable) 렌즈 등의 이동이 제어될 수 있으며, 광초점의 수직 위치, 즉, Z축 위치가 조절될 수 있다.
이미징 장치(130)는 형광 입자로부터 방출되는 형광 신호와 상기 형광 신호가 측정된 광초점의 위치를 기반으로 3차원 영상을 생성할 수 있다. 예컨대, 이미징 장치(130)는, 3차원 영상을 생성하는 프로그램을 실행하는 명령어를 저장한 메모리와, 상기 메모리에 저장된 프로그램을 실행하여 형광 입자로부터 방출되는 형광 신호와 상기 형광 신호가 측정된 광초점의 위치를 기반으로 3차원 영상을 생성하는 프로세서를 포함할 수 있다.
일 실시예로서, 현미경 장치(120)는 형광 입자로부터 방출되는 형광 신호의 세기 정보와 해당 형광 신호가 측정된 광초점의 위치 정보를 이미징 장치(130)로 제공 및/또는 전달할 수 있다. 또는 다른 실시예로서, 이미징 장치(130)가 현미경 장치(120)로부터 상기 형광 신호의 세기 정보와 상기 해당 형광 신호가 측정된 광초점의 위치 정보를 획득할 수 있다.
이미징 장치(130)는 상기 형광 신호의 세기 정보와 상기 해당 형광 신호가 측정된 광초점의 위치 정보를 기반으로 광초점의 위치 별로 해당 위치에서 측정된 형광 신호를 대응시켜 3차원 영상을 생성할 수 있다.
나아가, 이미징 장치(130)에 구비된 메모리와 프로세서는, 상기 위치 조절부에 구비된 메모리와 프로세서와 동일한 개체로 구성되거나, 별도의 다른 개체로서 각각 독립적으로 구성된 메모리와 프로세서일 수 있다.
도 2는 본 발명의 일 실시예에 따른 비선형 현미경 장치의 구조를 나타내는 도면이다.
도 2를 참조하면, 본 발명의 일 실시예에 따른 비선형 현미경 장치는 복수개의 렌즈(211, 212, 213, 214, 215, 216), 갈바노 거울(220), 대물렌즈(230), 복수개의 색선별(dichroic) 거울(241, 242), 밴드패스 필터(250) 및 신호 측정부(260)를 포함한다.
비선형 현미경 장치에 구비된 제1 렌즈(211)와 제2 렌즈(212)의 입사각과 굴절각에 의해 광원(200)에서 출력되는 광의 경로가 제어되어, 광원(200)에서 출력되는 광이 갈바노 거울(220)로 전달될 수 있다. 여기서, 광원(200)은 비선형 형광 입자로부터 형광을 여기시키기 위한 것으로서 연속파동 레이저를 생성하는 장치일 수 있다.
갈바노 거울(220)을 통해 반사된 광은 비선형 현미경 장치에 구비된 대물렌즈(230)를 통해 광 경로가 제어되어, 비선형 형광 입자로 표지된 샘플(235)에 광초점이 형성된다. 이때, 비선형 현미경 장치에 구비된 갈바노 거울(220)의 위치가 조절되면, 해당 광 초점의 수평 위치를 조절 및/또는 변경될 수 있으며, 대물렌즈(230) 또는 샘플(235)이 놓이는 스테이지(도시되지 않음), 튜너블 렌즈 등의 파면 제어 장치의 위치를 제어하여 광 초점의 수직 위치를 조절 및/또는 변경할 수 있다. 이 때, 비선형 현미경 장치에 구비된 제3 렌즈(214) 및 제4 렌즈(214)의 입사각과 굴절각에 의해 갈바노 거울(220)에서 반사된 광의 경로가 제어되어, 갈바노 거울(220)에서 반사된 광이 대물렌즈(230)로 전달될 수 있다.
비선형 형광 입자에서 방출되는 비선형의 형광 신호는 제1 색선별 거울(241), 제5 렌즈(215), 제2 색선별 거울(242), 밴드패스 필터(250) 및 제6 렌즈(216)를 통과하여 신호 측정부(260)에 도달될 수 있다. 신호 측정부(260)는 일 예로, 포토다이오드(photodiode), 광전자증폭관(photomultiplier tube) 등을 포함하여 구성될 수 있으며, 포토다이오드(photodiode), 광전자증폭관(photomultiplier tube) 등을 통해 형광 신호를 검출할 수 있다.
도 3은 본 발명의 일 실시예에 따른 형광 입자의 여기 빛 세기 대비 방출되는 형광 신호의 세기를 나타내는 도면이고, 도 4는 본 발명의 일 실시예에 따른 여기 빛과 형광 신호의 공간 분포를 나타내는 도면이다.
도 3에는 연속파동 레이저 여기에 비선형 광학 반응을 보이는 형광 입자의 여기 빛 세기 대비 방출되는 형광 세기의 로그-로그 플롯(log-log plot)이 도시되어 있다. 도 3에서 Iemission은 형광 입자로부터 방출되는 형광의 세기를 의미하고, Iexcitation은 형광 입자의 여기를 위한 빛의 세기를 의미한다. 도 3에서 로그-로그 플롯의 기울기는 (Iemission = Iexcitation
s)의 방정식에 따라 s 값으로 표시된다.
도 3을 참조하면, 상대적으로 여기 세기가 낮은 영역에서는 여기 빛 세기 대비 방출되는 형광 세기를 나타내는 로그-로그 플롯의 기울기 s는 1 보다 작다. 상대적으로 여기 세기가 높은 영역에서는 로그-로그 플롯의 기울기 s는 1보다 크다. 영역 1과 영역 2의 구분은 로그-로그 플롯의 기울기 s가 1이 되는 지점의 여기 빛 세기를 기준으로 하며, 이 때의 여기 빛 세기를 A라 한다.
한편, 도 4에는 광학 시스템의 초점거리를 중심으로 하는 가우시안(Gaussian) 분포를 가진 여기 빛 세기가 점선으로 도시되어 있다. 도 3에서의 설명과 같이, 여기 빛의 세기가 A가 되는 공간상 위치를 기준으로 영역 1과 영역 2가 구분된다. 영역 1에서의 로그-로그 플롯의 기울기 s는 1보다 작다. 따라서, 영역 1의 특정 공간 위치에서 여기 빛 에너지 대비 방출되는 형광 세기의 기울기 s는 여기 빛 세기의 가우시안 분포의 기울기보다 항상 낮다. 이 때, 예상되는 형광의 분포는 가는 실선으로 도시되어 있다. 한편, 영역 2에서의 로그-로그 플롯의 기울기 s는 1보다 크다. 따라서, 영역 2의 특정 공간 위치에서 여기 빛 에너지 대비 방출되는 형광 세기의 기울기 s는 여기 빛 세기의 가우시안 분포의 기울기보다 항상 높다. 이 때, 예상되는 형광의 분포는 굵은 실선으로 도시되어 있다. 결과적으로 도 4에 도시된 것과 같이, 형광 입자에서 방출되는 형광 세기의 거리에 따른 분포(굵은 실선과 가는 실선의 합)의 폭은 여기 빛 세기의 가우시안 분포의 폭보다 좁다.
광학 시스템에서 여기 빛 세기의 분포의 폭은 여기 빛 파장의 회절 한계에 의해 결정되며, 초고해상도 영상법, 근접장 제어 등의 특별한 처리를 하지 않는 이상 물리적으로 이보다 작아질 수 없다. 그러나 도 4에 도시된 것과 같이, 비선형 광학 반응을 보이는 형광 입자에서 방출되는 형광 세기의 거리에 따른 분포의 폭은 여기 빛 파장의 회절 한계의 폭보다 작아질 수 있다. 이는 여기 빛이 모이는 광학 시스템의 초점면에서는 폭발적으로 증가하는 형광 세기를 기대할 수 있음을 의미한다. 또한, 초점면에서 먼 거리에서는 굉장히 적은 양의 형광 세기가 기대됨을 의미한다. 결과적으로, 광학 시스템의 여기 빛이 모이는 초점에서만 강한 형광이 발생하게 되며, 이는 비선형 현미경이 기반으로 하는 비선형 여기에 따른 결과와 일치한다. 따라서, 연속파동 레이저 여기 시에 도 3의 광학 반응을 보이는 형광 입자를 이용하는 경우 연속파동 레이저로 구동하는 비선형 현미경을 구축할 수 있다.
연속파동 레이저 여기 시에 도 3에 도시된 비선형 광학 반응을 보이는 형광 입자로서 란탄족 이온들이 도핑된 상향변환 입자가 존재한다. 그 종류는 호스트 결정, 도핑된 란탄족 이온의 종류, 도핑된 란탄족 이온의 농도 등에 따라 다양하며, 이에 따라 기울기 s와 여기 빛 세기 A의 설계가 가능하다. 특히 상향변환 입자를 이용하면, 형광 빛의 파장보다 긴 파장의 여기 빛을 사용할 수 있게 된다. 기존 선형 형광 입자를 활용한 영상 기술의 경우 스토크스 시프트(stokes shift)로 입사되는 여기 빛과 방출되는 형광의 파장 관계가 정의되어 형광보다 짧은 파장의 여기 빛을 사용해야 한다. 그러나 본 발명에서는 비선형 형광 입자에서 방출되는 형광보다 긴 파장대의 여기 빛을 사용함으로써 보다 깊은 생체조직 내부를 비침습적이며 고해상도로 영상화할 수 있다.
한편, 앞서 언급한 란탄족 이온들이 도핑된 상향변환 입자 이외에도, 도 3에 도시된 비선형 광학 반응을 보이는 형광 입자는 모두 연속파동 레이저 기반의 비선형 현미경을 구현하는데 사용 가능하다. 일 예로, 동일한 형광 입자라 할지라도 여기 빛의 파장 및 관찰하는 형광 빛의 파장에 따라 도 3에서의 기울기 s와 A에 해당하는 여기 빛 세기 또한 달라질 수 있다. 또한, 형광 입자의 종류 및 실험 조건에 따라, 비선형 현미경의 삼차원 이미징 성능이 달라질 수 있다. 예를 들어, 도 3의 영역 1에서 기울기 s가 낮을수록 그리고 영역 2에서 기울기 s가 높을수록 3차원 이미징 시 보다 높은 해상도를 얻을 수 있다. 더불어, A 위치의 여기 빛 세기에 따라서도 이미징 성능 또한 변화할 수 있다.
도 5는 연속파동 레이저에 의해 발생되는 일반적인 형광 신호를 나타내는 도면이고, 도 6은 본 발명의 일 실시예에 따라 발생되는 형광 신호를 나타내는 도면이다.
기존의 형광 이미징은 형광 입자에 입사되는 빛과 형광 입자로부터 방출되는 형광의 세기가 선형 관계를 갖게 된다. 형광 입자에 입사되는 여기 빛과 형광 입자로부터 방출되는 형광 세기 사이에 비선형 관계를 갖게 하기 위해서는 초고속 펄스 레이저를 이용하여 형광 입자에 높은 에너지를 순간적으로 입사시켜야 하기 때문에, 연속파동 레이저로는 비선형 형광 신호를 얻기가 거의 불가능하다. 따라서 연속 파동 레이저로 일반 형광 입자를 여기시키면 도 5에 도시된 것과 같이 레이저가 통과하는 모든 깊이면에서 형광이 발생하게 된다. 이는 해당 광원 및 형광원을 활용한 이미징 시스템은 깊이 분해능을 상실하게 됨을 의미한다.
이에 반해 본 발명에 따른 이미징 방법을 사용하면, 도 6에 도시된 것과 같이 연속파동 레이저를 이용하더라도 초점이 생기는 깊이면(600)에서만 형광이 발생하며, 이와 같은 원리를 기반으로 연속 파동 레이저를 이용한 3차원 이미징이 가능해진다. 이와 같은 장점은 비단 3차원 이미징을 가능케 할 뿐만 아니라 샘플(특히 생체조직)에 대한 손상을 줄일 수 있다. 기존의 선형 형광 이미징의 경우 여기 빛이 통과하는 샘플의 모든 깊이 영역에 대해 에너지가 흡수되어 광독성(phototoxicity) 및 광표백(photobleaching) 현상이 유도되지만, 비선형 입자를 사용할 경우 여기 빛이 통과하는 모든 깊이가 아닌 초점면에서만 에너지가 유의미하게 흡수 또는 방출되어 광독성 및 광표백을 현저히 줄일 수 있다.
도 7은 본 발명의 일 실시예에 따른 3차원 이미징 방법을 나타내는 도면이다.
도 7을 참조하면 본 발명의 일실시예에 따른 3차원 이미징 시스템의 비선형 현미경 장치는 대물렌즈 앞에 샘플이 고정되면, 미리 설정된 영역 내에서 광초점이 형성되도록 광원에서 발생되는 빛을 형광 입자로 표지된 샘플에 조사할 수 있다(S700). 여기서, 상기 광원은 연속파동을 가지는 빛을 방출하도록 구성될 수 있으며, 이를 위하여 상기 광원은 연속파동 레이저로 구현될 수 있다. 또한, 상기 연속파동 레이저는 비침습적 고해상도 이미징을 위해 형광 입자로부터 방출되는 형광 신호의 파장보다 긴 파장을 빛을 방출하도록 구성될 수 있다.
광원에서 방출된 빛은 비선형 현미경 장치의 대물렌즈를 통과한 후 샘플의 특정 위치에서 초점을 맺게 되며, 비선형 형광 입자로 표지된 샘플에서는 광초점의 위치 근방에서만 형광이 발생하게 된다. 따라서, 상기 형광 입자로서 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 비선형 형광 입자가 사용될 수 있다. 상기 비선형 형광 입자는 란탄족 이온이 도핑된 상향변환 입자를 포함할 수 있다.
광원으로부터 방출되는 빛이 상기 형광 입자로 표지된 샘플에 조사되면, 비선형 현미경 장치는 광초점에서 형광 입자로부터 방출되는 비선형의 형광 신호를 측정할 수 있다(S710). 이 때, 비선형 현미경 장치는 상기 광초점의 위치를 미리 설정된 영역 내에서 이동시키면서 3차원 공간 내에서 발생하는 비선형 형광 신호를 연속적으로 측정할 수 있다. 여기서, 상기 미리 설정된 영역은 비선형 형광 입자로 입사되는 여기 빛의 세기 대비 상기 형광 입자로부터 방출되는 형광 신호의 세기를 기반으로 3차원 공간 상에 설정될 수 있다. 일례로, 상기 미리 설정된 영역은 광초점을 3차원 공간내에서 이동시키면서 3차원 영상을 구현하고자 하는 임의의 3차원 공간을 아우를 수 있다.
예를 들어, 비선형 현미경 장치는 적어도 하나의 거울을 제어하여 광초점의 평면 상의 위치를 조절할 수 있으며, 대물렌즈, 샘플이 놓이는 스테이지, 튜너블 렌즈 등을 제어하여 광초점의 깊이 및/또는 높이를 조절할 수 있다.
이후, 비선형 현미경 장치는 상기 형광 입자로부터 방출되는 형광 신호의 세기 정보와 상기 광초점의 위치 정보를 이미징 장치로 제공할 수 있다(S720).
3차원 영상 시스템에서 이미징 장치는 비선형 현미경 장치로부터 제공받은 형광 신호의 세기 정보와 해당 세기 정보가 측정된 광초점에서의 위치 정보를 기반으로 샘플 전체의 3차원 구조를 나타내는 3차원 영상을 생성할 수 있다. 예를 들어, 상기 이미징 장치는 광초점의 위치 별로 해당 위치에서 측정된 형광 신호를 대응시켜 3차원 공간상의 복셀(voxel)에 배치함으로써 3차원 영상을 생성할 수 있다.
한편, 전술한 실시예에 따른 3차원 이미징 방법에 포함된 각각의 단계는, 이러한 단계를 프로세서가 수행하도록 하기 위한 명령어를 포함하여 기록매체에 기록된 컴퓨터 프로그램으로 구현될 수 있다.
또한, 전술한 실시예에 따른 3차원 이미징 방법에 포함된 각각의 단계는, 이러한 단계를 프로세서가 수행하도록 하기 위한 명령어를 포함하는 컴퓨터 프로그램이 기록된 컴퓨터 판독 가능한 기록매체에서 구현될 수 있다.
본 발명에 첨부된 각 흐름도의 각 단계의 조합들은 컴퓨터 프로그램 인스트럭션들에 의해 수행될 수도 있다. 이들 컴퓨터 프로그램 인스트럭션들은 범용 컴퓨터, 특수용 컴퓨터 또는 기타 프로그램 가능한 데이터 프로세싱 장비의 프로세서에 탑재될 수 있으므로, 컴퓨터 또는 기타 프로그램 가능한 데이터 프로세싱 장비의 프로세서를 통해 수행되는 그 인스트럭션들이 흐름도의 각 단계에서 설명된 기능들을 수행하는 수단을 생성하게 된다. 이들 컴퓨터 프로그램 인스트럭션들은 특정 방식으로 기능을 구현하기 위해 컴퓨터 또는 기타 프로그램 가능한 데이터 프로세싱 장비를 지향할 수 있는 컴퓨터 이용 가능 또는 컴퓨터 판독 가능 기록매체에 저장되는 것도 가능하므로, 그 컴퓨터 이용가능 또는 컴퓨터 판독 가능 기록매체에 저장된 인스트럭션들은 흐름도의 각 단계에서 설명된 기능을 수행하는 인스트럭션 수단을 내포하는 제조 품목을 생산하는 것도 가능하다. 컴퓨터 프로그램 인스트럭션들은 컴퓨터 또는 기타 프로그램 가능한 데이터 프로세싱 장비 상에 탑재되는 것도 가능하므로, 컴퓨터 또는 기타 프로그램 가능한 데이터 프로세싱 장비 상에서 일련의 동작 단계들이 수행되어 컴퓨터로 실행되는 프로세스를 생성해서 컴퓨터 또는 기타 프로그램 가능한 데이터 프로세싱 장비를 수행하는 인스트럭션들은 흐름도의 각 단계에서 설명된 기능들을 실행하기 위한 단계들을 제공하는 것도 가능하다.
또한, 각 단계는 특정된 논리적 기능(들)을 실행하기 위한 하나 이상의 실행 가능한 인스트럭션들을 포함하는 모듈, 세그먼트 또는 코드의 일부를 나타낼 수 있다. 또, 몇 가지 대체 실시예들에서는 단계들에서 언급된 기능들이 순서를 벗어나서 발생하는 것도 가능함을 주목해야 한다. 예컨대, 잇달아 도시되어 있는 두 개의 단계들은 사실 실질적으로 동시에 수행되는 것도 가능하고 또는 그 단계들이 때때로 해당하는 기능에 따라 역순으로 수행되는 것도 가능하다.
이상의 설명은 본 발명의 기술 사상을 예시적으로 설명한 것에 불과한 것으로서, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자라면 본 발명의 본질적인 품질에서 벗어나지 않는 범위에서 다양한 수정 및 변형이 가능할 것이다. 따라서, 본 발명에 개시된 실시예들은 본 발명의 기술 사상을 한정하기 위한 것이 아니라 설명하기 위한 것이고, 이러한 실시예에 의하여 본 발명의 기술 사상의 범위가 한정되는 것은 아니다. 본 발명의 보호 범위는 아래의 청구범위에 의하여 해석되어야 하며, 그와 균등한 범위 내에 있는 모든 기술사상은 본 발명의 권리범위에 포함되는 것으로 해석되어야 할 것이다.
Claims (16)
- 3차원 이미징 시스템에 있어서,컴퓨터 프로그램을 수행하는 명령어를 저장하는 메모리;상기 명령어를 수행하는 프로세서;빛이 발생하도록 구성되는 광원; 및상기 광원에서 발생되는 빛이, 형광 입자로 표지된 샘플에 조사되도록 하는 광경로를 형성하고, 상기 광경로를 통해 전달되는 빛이 상기 형광 입자로 표지된 샘플이 위치한 미리 설정된 영역에 광초점이 형성되도록 하고, 상기 광초점에 조사된 빛에 의해 상기 형광 입자로부터 방출되는 형광 신호를 측정하는 비선형 현미경 장치를 포함하되,상기 프로세서에 의해 상기 명령어가 실행되면, 상기 프로세서는,상기 형광 입자로부터 방출되는 형광 신호와 상기 광초점의 위치를 기반으로 3차원 영상을 생성하고,상기 광원은 연속파동을 가지는 빛이 발생하도록 구성되는 연속파동 레이저를 출력하고,상기 형광 입자는 상기 연속파동을 가지는 빛에 의해 비선형 광학 반응을 나타내는 형광 입자인 것을 특징으로 하는, 3차원 이미징 시스템.
- 제1항에 있어서,상기 미리 설정된 영역은,상기 형광 입자로 입사되는 여기(excitation) 빛의 세기 대비 상기 형광 입자로부터 방출되는 형광 신호의 세기를 기반으로 설정되는 영역인 것을 특징으로 하는, 3차원 이미징 시스템.
- 제2항에 있어서,상기 미리 설정된 영역은,상기 형광 입자로부터 방출되는 형광 신호의 세기가 상기 형광 입자로 입사되는 여기 빛의 세기와 비선형 관계를 갖는 영역인 것을 특징으로 하는, 3차원 이미징 시스템.

- 제6항에 있어서,상기 위치 조절부는,적어도 하나의 거울을 포함하고,상기 프로세서는,상기 적어도 하나의 거울의 위치를 제어하여 상기 광초점의 수평 위치를 조절하고,상기 대물렌즈 및 상기 샘플이 놓이는 스테이지 중 적어도 하나를 제어하여, 상기 광초점의 수직 위치를 조절하는,3차원 이미징 시스템.
- 제1항에 있어서,상기 프로세서는,상기 광초점의 위치 별로 해당 위치에서 측정된 형광 신호를 대응시켜 상기 3차원 영상을 생성하는 것을 특징으로 하는, 3차원 이미징 시스템.
