Abstract
후코이단 발효물을 포함하는 조성물 및 그의 피부 상태 개선 용도에 관한 것으로, 일 양상에 따른 조성물은 상기 락토바실러스 속 균주를 통해 후코이단을 발효하는 경우, 후코이단의 저분자를 유도할 수 있어, 후코이단의 피부 흡수력 및 용매에 대한 용해도를 개선할 수 있는 효과가 있다. 또한, 후코이단 추출물 대비 현저히 향상된 피부 개선 효과, 예를 들면, 항산화, 피부 보습 개선, 피부 장벽 개선, 피부 재생 개선, 또는 피부 미백 효과가 있다.

후코이단 발효물 및 그의 피부 상태 개선 용도에 관한 것이다.
후코이단(fucoidan)은 황 산기가 부착된 다당류로써 항암, 항고혈압, 항염증 등에 효능이 있고, 해조류에 존재하는 것으로 알려져 있다.

이에, 산이나 염기와 같은 화학적 방법을 통해 후코이단의 저분자화를 유도하는 종래 기술들이 개발되어 왔으나, 이러한 공정은 매우 복잡하고 까다로운 조건에서 진행되기 때문에 비효율적이며 비경제적이라는 단점이 있다.

이하 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 락토바실러스 루테리 KMH202의 분리 및 동정
석류로부터 락토바실러스 루테리 KMH202 균주를 분리 및 동정하기 위해 다음과 같이 수행하였다.
먼저, 껍질 채 파쇄한 석류 1 g과 MRS media 20 ml을 혼합 후 vortexing하여 시료를 현탁하였다. 그 후, 현탁된 시료를 멸균증류수를 이용하여 연속희석 하였고, MRS agar plate에 10-4 내지 10-6으로 pour plating하였다. 이 후, 37 ℃에서 1 일 이상 배양하여 균을 선별하였다.
선별한 균체 집락 중 결정된 16S rRNA부위의 염기서열을 미국 국립생물정보센터(NCBI, National Center for Biotechnology Information) 홈페이지에서 제공되는 BLAST 프로그램으로 등록된 다른 균주들과 비교 분석하였다. 비교 분석 결과 상동성 99%의 락토바실러스 루테리 KMH202를 분리하였다. 선별된 락토바실러스 루테리 KMH202 균주를 2022년 02월 25일자로 한국생물자원센터에 기탁하여 기탁번호 KCTC 14878BP를 부여받았고, 락토바실러스 루테리 KMH202 균주는 서열번호 1(complementary DNA)의 16s rRNA 유전자 서열을 갖는다.
실시예 2. 후코이단 발효물 제조
락토바실러스 루테리 KMH202로 발효한 후코이단 발효물을 제조하기 위해, 4 % 농도의 후코이단 추출물 파우더(Biocorp)를 정제수에 용해하고, 0.22 μm 필터로 여과하여 여과액을 수득하였다. 그 후, 상기 여과액을 200% 농도의 영양 배지(Glucose, yeast, 염 등을 포함)에 1 : 1 비율로 희석함으로써 2% 농도의 후코이단 추출물 용액을 수득하였다. 상기 후코이단 추출물 용액에 0.04 OD 농도의 락토바실러스 루테리 KMH202를 접종하였다.
그리고 상기 후코이단 추출물을 37 ℃, 혐기 조건 유지 하에서 17시간동안 배양하여 배양된 발효액을 수득한 후, 상기 발효액을 10 내지 20 분 동안 4000 RPM 조건으로 원심분리하여 상등액을 수득하였다. 그 후, 상기 상등액을 0.22 um 필터로 여과하여 잔여물을 제균하고, 후코이단 발효물을 제조하였다.
실험예 1. 후코이단 발효물의 항산화 효과 확인
락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 항산화 효과를 확인하기 위해 DPPH assay를 진행하였다.
보다 구체적으로, 96 well plate에 10 % 후코이단 발효물, 또는 10 % 후코이단을 각각 100 uL씩 넣어준 후, 0.5 mM DPPH 100 uL 또는 100 % 에탄올(100 uL)을 첨가하여, 25 ℃에서 30분간 반응시켰다. 그 후, 540 nm에서의 흡광도를 측정하고 그 결과를 도 1에 나타내었다. 양성대조군으로 10 ug/ml의 L-아스코르브산(ascorbic acid, 100 uL)을 사용하였다.
도 1은 락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 항산화 효과를 나타낸 그래프이다.
도 1에 나타낸 바와 같이, 후코이단 발효물에서 후코이단 추출물 대비 약 7 배 이상 증가한 항산화능 효과를 나타냄을 확인하였다.
실험예 2. 후코이단 발효물의 미백 효과 확인
락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 미백 효과를 확인하기 위해 멜라닌 세포에서 멜라닌 합성 저해 효과를 측정하였다.
보다 구체적으로, 6 well plate에 B16F1 세포를 1 X 105 cell/well의 밀도로 접종한 후, 37 ℃, 5 % CO2 조건으로 배양기에서 24 시간동안 배양하였다. 이후, 세포 배양액을 제거한 뒤 DPBS 1 mL로 세척하고, 후코이단 발효물, 후코이단, 및 알부틴(arbutin)을 각각 세포 배양 배지로 희석하여 각 2 mL/well씩 첨가하였다. 이후 37 ℃, 5 % CO2 조건의 배양기에서 72 시간동안 배양하였다. 그리고 배양액을 제거한 세포를 trypsin-EDTA로 반응시켜 세포를 수득하고, 상기 수득한 세포를 DPBS 1 mL로 1 회 세척하였다. 이후, 25 ℃, 10,000 rpm에서 10 분간 원심 분리하여 상층액을 제거한후, pellet을 70 ℃의 건조 오븐(drying oven)에서 1 시간동안 건조시켰다. 완전히 건조된 pellet에 NaOH 용액(0.4 mL)을 첨가하여 70 ℃에서 10 분동안 pellet을 녹인 후, 1,800 rpm으로 5 분간 교반하고 96 well plate에 100 uL씩 분주하여 490 nm에서의 흡광도를 측정하였다. 그리고 그 결과를 도 2에 나타내었다.
도 2는 락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 멜라닌 생성 억제 효과를 나타낸 그래프이다.
도 2에 나타낸 바와 같이, 후코이단 추출물은 약 13 %의 멜라닌 생성 억제 효과를 보인 반면, 후코이단 발효물은 약 28.3 %의 멜라닌 생성 억제 효과를 보임을 확인하였다. 이는 후코이단 발효물에서 후코이단 추출물 대비 약 20 % 이상 증가한 미백 효과를 가짐을 의미한다.
실험예 3. 후코이단 발효물의 피부 장벽 강화 효과 확인
락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 피부장벽 강화 효과를 확인하기 위해, 인간 각질형성세포(human keratinocyte) 인 HaCaT 세포주에서 필라그린(filaggrin) 유전자의 mRNA 발현량을 real-time PCR로 확인하고, 그 결과를 도 3에 나타내었다.
보다 구체적으로, HaCaT 세포를 6 well 세포배양 플레이트에 3 x 105 cells/well의 밀도로 접종한 후, 37 ℃, 5 % CO2 조건의 배양기에서 24 시간동안 배양하였다. 그 후, 후코이단 발효물, 및 후코이단을 각각 FBS를 포함하지 않는 배지(FBS free 배지)로 희석한 후, 상기 플레이트에 첨가하여 20 시간동안 배양하였다. RNA는 RNeasy Mini Kit(QIAGEN)를 사용하여 분리하였으며, 분리한 RNA는 NanoDrop Onec(Thermo Scientific)를 이용해 순도(purity)를 확인한 후 정량하여 SuperScript IV VILO Master Mix(Invitrogen)로 cDNA를 합성하였다. RT-PCR 장비로는 Step One Plus(applied biosystems)를 사용하였으며, PowerUp SYBR Green Master Mix(applied biosystems)를 합성한 cDNA 및 프라이머(filaggrin 또는 β의 프라이머)와 함께 첨가하여, 95 ℃에서 2 분간 중합효소를 활성화시킨 후, 중합반응을 진행하였다: 총 40 사이클(1 사이클당 95 ℃에서 15초, 55 ℃에서 15초, 72 ℃에서 1 분). 대조군으로 D-판테놀(D-panthenol)을 사용하였고, 사용된 프라이머의 서열은 하기 표 1과 같다.

도 3에 나타낸 바와 같이, 후코이단 발효물에서 필라그린 발현 정도가 후코이단 추출물 대비 약 1.5 배 이상 증가함을 확인하였다. 이는 후코이단 추출물보다 후코이단 발효물에서 현저한 피부 장벽 강화 효과가 나타남을 의미한다.
실험예 4. 후코이단 발효물의 피부 보습 효과 확인
락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 피부 보습 효과를 확인하기 위해, HaCaT 세포주에서 히알루론산 합성 효소인 HAS3(Hyaluronan synthase 3) 유전자의 mRNA 발현량을 real-time PCR로 확인하고, 그 결과를 도 4에 나타내었다.
보다 구체적으로, HaCaT 세포를 6 well 세포배양 플레이트에 3 x 105 cells/well의 밀도로 접종한 후, 37 ℃, 5 % CO2 조건의 배양기에서 24 시간동안 배양하였다. 그 후, 후코이단 발효물 및 후코이단을 각각 FBS free 배지로 희석한 후, 상기 플레이트에 첨가하여 20 시간동안 배양하였다. RNA는 RNeasy Mini Kit(QIAGEN)를 사용하여 분리하였으며, 분리한 RNA는 NanoDrop Onec(Thermo Scientific)를 이용해 순도를 확인한 후 정량하여 SuperScript IV VILO Master Mix(Invitrogen)로 cDNA를 합성하였다. RT-PCR 장비로는 Step One Plus(applied biosystems)를 사용하였으며, PowerUp SYBR Green Master Mix(applied biosystems)를 합성한 cDNA 및 프라이머(filaggrin 또는 β의 프라이머)함께 첨가하여, 95 ℃에서 2 분간 중합효소를 활성화시킨 후, 중합반응을 진행하였다: 총 40 사이클(1 사이클당 95 ℃에서 15초, 55 ℃에서 15초, 72 ℃에서 1 분). 대조군으로 레티노산(retinoic acid)을 사용하였고, 사용된 프라이머의 서열은 하기 표 2와 같다.

도 4에 나타낸 바와 같이, 후코이단 발효물에서 발효물의 농도에 비례하여 HAS3 mRNA의 발현 정도가 증가함을 확인하였다. 특히, 5 % 후코이단 발효물에서 HAS3 mRNA 발현 정도가 후코이단 처리군 대비 약 5 배 이상 증가함을 확인하였다. 반면, 후코이단 추출물에서는 농도에 따른 유의한 변화가 나타나지 않음을 확인하였다.
이러한 결과는 후코이단 추출물보다 후코이단 발효물에서 현저한 피부 보습 효과가 나타남을 의미한다.
실험예 5. 후코이단 발효물의 피부 재생 효과 확인
락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 피부 재생 효과를 확인하기 위해, HaCaT 세포주를 사용하여 상처 치유 분석(wound healing assay)을 수행하고 도 5 및 6에 나타내었다.
보다 구체적으로, HaCaT 세포를 96 well 세포배양 플레이트에 3 x 104 cells/well의 밀도로 접종한 후, 37 ℃, 5 % CO2 조건의 배양기에서 24 시간동안 배양하였다. 그리고 96-well wound maker(sartorius stedim biotech)를 이용하여 상처(wound scratch)를 생성한 뒤, PBS로 2번 세척하였다. 이후 FBS free 배지로 교체한 뒤, 후코이단 발효물 또는 후코이단을 1 %(v/v) 농도가 되도록 첨가한 후, 37 ℃, 5 % CO2 조건에서 12 시간동안 배양하였다. 그리고, IncuCyte(Satorius)를 사용하여 배양한 HaCaT 세포의 세포 사진을 촬영하였다. 대조군으로는 시료를 처리하지 않은 세포주를 사용하였다.
상처 컨플루언스(wound confluence)는 Incucyte Software(Scratch Wound Cell Migration and Invastion Analysis Metrics)를 사용하여 계산하였으며, 계산식은 하기 식 1과 같다:

상처 컨플루언스(%) = [(12시간 세포면적 - 0시간 세포면적)/(전체면적 - 0시간 세포면적)] x 100.
도 5 및 6은 후코이단 추출물 또는 락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 처리에 따른 상처 컨플루언스를 나타낸 그래프 및 이미지이다:
도 5는 후코이단 추출물 또는 락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 처리에 따른 상처 컨플루언스를 나타낸 그래프이고, 도 6은 후코이단 추출물 또는 락토바실러스 루테리 KMH202로 발효한 후코이단 발효물의 처리에 따른 상처 치유 효과를 나타낸 이미지이다.
도 5 및 6에 나타낸 바와 같이, 상처 컨플루언스에서 후코이단 추출물을 처리한 경우 대조군 대비 유의한 차이가 나타나지 않음을 확인하였다. 반면, 후코이단 발효물을 처리한 경우, 대조군 대비 약 1.2 배 증가, 및 후코이단 추출물 대비 약 1.4 배 증가한 상처 컨플루언스를 보임을 확인하였다. 이는, 후코이단 추출물보다 후코이단 발효물에서 현저한 피부 재생 효과가 나타남을 의미한다.

- 후코이단 추출물의 락토바실러스 루테리(Lactobacillus reuteri) 균주 발효물.
- 청구항 1에 있어서,상기 락토바실러스 루테리는 기탁번호 KCTC 14878BP로 기탁된 락토바실러스 루테리 KMH202(Lactobacillus reuteri KMH202)인 것인, 발효물.
- 청구항 1에 있어서,상기 발효물은 후코이단 추출물에 락토바실러스 루테리 균주를 접종하여 제조된 것인, 발효물.
- 청구항 3에 있어서,상기 균주의 접종 농도는 0.001 내지 0.1 OD인 것인, 발효물.
- 후코이단 추출물의 락토바실러스 루테리(Lactobacillus reuteri) 균주 발효물을 포함하는 화장료 조성물.
- 청구항 5에 있어서,상기 락토바실러스 루테리는 기탁번호 KCTC 14878BP로 기탁된 락토바실러스 루테리 KMH202(Lactobacillus reuteri KMH202)인 것인, 화장료 조성물.
