Description
본 출원은 2021년 9월 8일 출원된 대한민국 특허출원 제 10-2021-0119866호 를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다.
신규한 미생물, 그의 파쇄액, 배양액, 추출액, 이를 이용한 천연물 복합발효추출물 및 이들의 피부 상태 개선 용도에 관한 것이다.
피부의 생태계는 미생물에게 다양한 형태의 서식처를 제공하며, 광범위한 미생물들이 살고 있다. 숙주인 인간은 이들과 공생관계를 이루고 있으며, 이들은 숙주에 많은 긍정적인 영향을 미친다고 알려져 있다. 피부는 함입부, 특화 되어있는 틈새 등 다양한 형태의 서식처를 구성하고 있으며, 넓은 분포의 미생물이 자랄 수 있도록 돕는다. 기본적으로 피부는 물리적인 막을 형성하며, 외부로부터 잠재적인 위험요소 및 독성물질들로부터 방어를 하도록 도와준다. 피부는 외부환경과의 접속 지점이 되며, 다양한 미생물들(진균, 세균, 바이러스 및 작은 유충)의 집합소이기도 하다. 물리적, 화학적 기능의 선택에 맞게 미생물들은 특화된 틈새에 적응하여 서식처를 마련한다. 일반적으로 피부는 차갑고, 산성 성질을 나타내며, 건조된 상태로 유지된다. 구조적으로 표피(epidermis)는 피부 장벽을 이루고 있으며, 미생물과 독소가 침투하는 것을 차단하고, 수분을 유지하는 중요한 역할을 한다. 표피의 최상위층은 각질층(stratum corneum)으로 구성되어있다. 표피는 일명 '벽돌과 몰탈 구조'라고 불리는 형태를 띠고 있는데, 피부 조직은 계속적인 자가 회복 과정을 거치고, 분화 과정의 마지막을 거친 비늘(squames)은 끊임없이 피부 조직에서 탈락되는 과정을 반복하게 된다.
프로바이오틱스(Probiotics)는 인체에 유익한 작용을 하는 미생물을 총칭하는 말로 우리 몸에 유익(benefit)을 주는 미생물을 말한다. 현재까지 알려진 대부분의 프로바이오틱스는 유산균으로 알려져 있다. 프로바이오틱스는 인체에 여러 가지 유익 작용을 통해 효과적인 효능이 발생되는 것으로 보고되었지만, 피부상재균과 피부의 상호관계에 대한 연구는 미비한 실정이다.
이에, 피부 관련된 상태에 유용하게 사용될 수 있는 피부상재균의 개발이 필요한 실정이다.
일 양상은 신규한 비피도박테리움 애니멀리스 서브스페시스 락티스(Bifidobacterium animalis subsp. lactis) 균주를 제공하는 것이다.
다른 양상은 상기 균주의 파쇄액, 배양액, 또는 배양액의 추출물을 제공하는 것이다.
또 다른 양상은 상기 균주로 감초, 엉겅퀴 및 인동의 복합 추출물을 발효시킴으로써 수득된 복합발효추출물을 제공하는 것이다.

실시예 1. 균주의 분리 및 동정
건강한 개의 피부에서 비피도박테리움 애니멀리스 서브스페시스 락티스 균주를 분리 및 동정하였다.
구체적으로, 건강한 개의 피부를 멸균 증류수로 세척하여 확보된 샘플, 즉, 개 각질세포(canine epidermal keratinocyte)를 TSA(Tryptic Soy Agar) 배지(BD)에 접종하였다. 접종 후 48시간 동안 28℃ 인큐베이터에서 배양한 뒤 형성된 집락 100개를 순수 분리 배양하여, 48시간 동안 28℃ 인큐베이터에서 재배양 하였다.
배양이 완료된 집락은 16S rRNA 유전자 서열 동정을 실시하였다. 이때 프라이머는 박테리아에만 반응하여 증폭하도록 고안된 것을 사용하였다. PCR 증폭은 95℃ 1분, 55℃ 1분, 75℃ 1분30초씩 30 사이클로 실시 하였으며, 마지막으로 72℃에서 8분간 처리한 후 4℃에서 보관하였다. PCR 반응이 끝난 뒤 분리 배양된 종들의 DNA 서열은 ABI-3730XL(ABI, USA)를 이용하여 결정하였다.
분리 배양된 미생물 집락 중 결정된 16S rRNA 부위의 염기서열을 미국 국립생물정보센터(NCBI, National Center for Biotechnology Information) 홈페이지에서 제공되는 BLAST 프로그램을 사용하여 등록된 다른 균주들과 비교 분석 하였을 때 상동성 99% 이하의 신규 미생물 비피도박테리움 애니멀리스 서브스페시스 락티스(Bifidobacterium animalis subsp. lactis) 균주(이하 "DS008"라 함)를 선별하였다. 선별된 DS008 균주를 2021년 5월 28일자로 한국 미생물 보존센터(KCCM)에 기탁하여 기탁번호 KCCM12993P를 부여 받았다. DS008 균주는 서열번호 1(complementary DNA)의 16S rRNA 서열을 갖는다.
실시예 2. DS008 균주 배양액의 제조
실시예 1의 DS008 균주를 R2A(Reasoner's 2A) 배지(Becton Dickinson, Cockeysville, MD)에 접종하여 37℃, 혐기 조건에서 72시간 배양하였다.
실시예 3. DS008 균주를 이용한 천연물 발효액의 제조
증류수 1 L당 감초 10 g, 엉겅퀴 뿌리 50 g 및 인동 줄기 40 g를 넣고, 1 기압 이상의 압력, 112℃에서 2시간 30분 동안 중탕추출하여 천연물 복합 추출물을 얻었다. R2A 배지에 상기 천연물 복합 추출물 5%(w/w)를 첨가하고 실시예 1의 DS008 균주를 접종하여 72시간 배양하고, 상층액을 수득하고 여과하여 발효액을 제조하였다.
실험예 1. 피부 장벽 강화 및 피부 보습 활성 분석 - HAS3 및 Filaggrin 발현 증가 확인
DS008 균주 배양액 또는 이를 이용한 천연물 발효액이 피부 장벽 강화 및 피부 보습 활성에 미치는 영향을 분석하기 위해, HAS3(Hyaluronic acid synthase 3) 및 필라그린(Filaggrin)의 발현을 확인하였다.
구체적으로, 인간 각질형성 세포주(human keratinocyte)인 HaCaT 세포를 10% 우혈청(fetal bovin serum)을 포함한 DMEM 배지(Dulbecco's modified Eagle's Medium, Gibco 1210-0038)에서 배양하였고, 배양은 모두 37℃, 5% CO2 배양기에서 수행하였다. 상기 배양된 세포주에 상기 실시예 2의 배양액 1%(w/w) 또는 실시예 3의 발효액 1%(w/w)을 처리하고 24시간 동안 추가 배양하였다. 양성대조군으로는 레티노산(Retinoic acid, RA) 1 uM을 사용하였다. 피부 장벽 강화 및 피부 보습과 관련된 인자인 HAS3 및 필라그린에 대한 프라이머를 사용하여 실시간(real-time) PCR 기계에서 실시간 중합효소 연쇄반응을 수행하였다. HAS3 및 필라그린의 발현량은 β-actin 유전자에 대한 보정을 통해 최종적으로 분석하였고, 그 결과를 도 1, 2, 3 및 4에 각각 나타내었다.
도 1 및 2는 각질형성세포에서 HAS3의 상대적인 발현 수준을 나타낸 결과이다.
도 3 및 4는 각질형성세포에서 필라그린의 상대적인 발현 수준을 나타낸 결과이다.
도 1, 2, 3 및 4에 나타낸 바와 같이, 실시예 2의 균주 배양액은 HAS3의 발현을 유의적으로 증가시켰다. 또한, 실시예 3의 천연물 발효액은 HAS3 및 필라그린의 발현을 유의적으로 증가시켰다. 특히, 실시예 3의 천연물 발효액은 실시예 2의 균주 배양액 보다 HAS3 및 필라그린의 발현량이 높음을 확인할 수 있었다.
이러한 결과는 DS008 균주 및 이를 이용한 천연물 발효액이 피부 장벽 강화 및 피부 보습에 유용하게 사용될 수 있음을 의미한다.
실험예 2. 항염증, 항아토피 및 가려움증 억제 활성 분석 - 염증성 사이토카인 발현 감소 확인
DS008 균주 배양액 또는 이를 이용한 천연물 발효액이 항염증, 항아토피 및 가려움증 억제 활성이 있는지 여부를 분석하기 위해, 염증성 사이토카인 IL-1α 및 IL-6의 발현을 확인하였다.
구체적으로, 인간 각질형성 세포주(human keratinocyte)인 HaCaT 세포를 10% 우혈청(fetal bovin serum)을 포함한 DMEM 배지(Dulbecco's modified Eagle's Medium, Gibco 1210-0038)에서 배양하였고, 배양은 모두 37℃, 5% CO2 배양기에서 수행하였다. 배지는 3~4일마다 교환해 주며 세포가 과다하게 증식되었을 때는 계대배양 하였다. 이후에 상기 세포를 5x105/웰로 분주하고 배양 24시간 후, 인산 완충 식염수 용액(PBS)으로 세척하였다. FBS를 포함하지 않은 DMEM 배지가 들어있는 각 웰에 세포를 분주하고, Poly I:C 10 ㎍/㎖, 및 IL-4를 10 ng/㎖로 첨가하여 각질형성 세포주에 염증을 유도하였다. 다음으로 상기 실시예 2의 배양액 또는 실시예 3의 발효액을 1%(w/w) 농도로 처리한 후, 4시간 동안 추가 배양하였다. 양성대조군으로는 덱사메타손(dexamethasone) 1 μM을 사용하였다. IL-1α 및 IL-6에 대한 프라이머를 사용한 것만을 제외하고는 상기 실험예 1과 동일한 방법으로 발현량을 분석하였다. 유전자의 발현량은 β-actin 유전자에 대한 보정을 통해 최종적으로 분석하였고, 그 결과를 도 5 내지 도 8에 나타내었다.
도 5 및 6은 각질형성세포에서 IL-1α의 상대적인 발현 수준을 나타낸 결과이다.
도 7 및 8은 각질형성세포에서 IL-6의 상대적인 발현 수준을 나타낸 결과이다.
도 5, 6, 7 및 8에 나타낸 바와 같이, 실시예 2의 균주 배양액은 IL-1α 및 IL-6의 발현을 유의적으로 감소시켰다. 또한, 실시예 3의 천연물 발효액은 IL-1α 및 IL-6의 발현을 유의적으로 감소시켰다. 특히, 실시예 3의 천연물 발효액은 실시예 2의 균주 배양액 보다 IL-1α 및 IL-6의 발현 감소량이 크다는 것을 확인할 수 있었다.
이러한 결과는 DS008 균주 및 이를 이용한 천연물 발효액이 피부 염증성 질환의 예방, 개선, 또는 치료에 유용하게 사용될 수 있을 뿐만 아니라, 아토피의 예방, 개선, 또는 치료, 또는 가려움증(소양증)의 예방, 치료 또는 개선에도 유용하게 사용될 수 있음을 의미한다.
종합하면, 비피도박테리움 애니멀리스 서브스페시스 락티스 DS008 균주 또는 이를 이용한 천연물 발효액은 HAS3 및 필라그린의 발현을 증가시키며; IL-1α 및 IL-6의 발현을 감소시킴을 확인하였다. 따라서, DS008 균주 및 이를 이용한 천연물 발효액은 피부 장벽 강화, 피부 보습, 피부 염증 개선, 아토피 개선, 피부 가려움증 억제 효과를 가짐을 알 수 있었다.
이상의 설명은 본 발명의 기술 사상을 예시적으로 설명한 것에 불과한 것으로서, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자라면 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 다양한 수정 및 변형이 가능할 것이다.
Claims (12)
- 서열번호 1과 95% 이상의 서열 동일성을 갖는 16S rRNA를 포함하는, 비피도박테리움 애니멀리스 서브스페시스 락티스(Bifidobacterium animalis subsp. lactis) 균주.
- 청구항 1에 있어서, 기탁번호 KCCM12993P로 기탁된 것인 균주.
- 청구항 1의 균주, 그의 파쇄액, 배양액, 또는 배양액의 추출물을 포함하는 화장료 조성물.
- 청구항 3에 있어서, 피부 상태 개선용인 것인 화장료 조성물.
- 청구항 4에 있어서, 상기 피부 상태 개선은 피부 장벽 강화, 피부 보습, 피부 염증 개선, 아토피 개선, 및 가려움증 억제 중 1종 이상인 것인 화장료 조성물.
- 청구항 3에 있어서, HAS3(hyaluronic acid synthase 3) 및 필라그린(Filaggrin) 중 1종 이상의 발현을 증가시키거나; IL-1α 및 IL-6 중 1종 이상의 발현을 감소시키는 것인 화장료 조성물.
- 청구항 1의 균주, 그의 파쇄액, 배양액, 또는 배양액의 추출물을 유효성분으로 포함하는 피부 염증성 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 1의 균주, 그의 파쇄액, 배양액, 또는 배양액의 추출물을 포함하는 피부 상태 개선용 건강기능식품 조성물.
- 청구항 1의 균주로 감초, 엉겅퀴 및 인동의 복합 추출물을 발효시킴으로써 수득된 복합발효추출물을 포함하는 피부 상태 개선용 화장료 조성물.
