Abstract
肌の状態を良好に保つために有用な、皮膚常在菌叢改善剤を提供する。ソホロースを用いた皮膚常在菌叢改善剤である。ソホロースには、皮膚常在菌のうち、肌に悪い影響を及ぼす可能性を有する菌の成育を抑制しつつ、肌に良い影響を与える有益な菌の増殖を促すことができる機能性が備わる。この皮膚常在菌叢改善剤は、皮膚常在菌叢バランスを維持して、肌の状態を良好に保つ目的で好適に用いられ得る。
Description
本発明は、ソホロースを用いた皮膚常在菌叢改善剤及び皮膚外用組成物に関する。
皮膚には様々な共生微生物が集団を形成して常在しており、個人差、部位差、季節変動などがあるが、通常、ある一定のバランスが維持されており、その皮膚常在菌叢が皮膚機能に対して影響を及ぼすことが知られている。健常な人の皮膚は、この皮膚常在菌叢により、表皮のpHが弱酸性に維持されたり、保湿環境が維持されたり、病原菌に対する抵抗力が高められるなどして、肌状態が良好に維持されている。一方で、外傷、病気等による全身状態の不良、精神的ストレス、寒冷、乾燥、不適切な洗浄などによって皮膚常在菌叢のバランスが崩れると、皮膚の外界からの保護機能が低下するだけでなく、様々な皮膚症状が誘発され、疾患発現が起こることもある。
皮膚常在菌叢のバランスを保ち、皮膚機能を維持するため、その一翼を担っている常在菌としては、例えば、表皮ブドウ球菌として知られるスタフィロコッカス・エピデルミデス(Staphylococcus epidermidis)が挙げられる。この常在菌は、皮脂を分解して、皮膚を弱酸性に保つ脂肪酸や保湿成分であるグリセリンを産生したり、病原性を有する細菌の増殖を抑制する抗菌ペプチドを産生したりして、皮膚のバリア機能を高めたりしている。同じく表皮ブドウ球菌として知られるスタフィロコッカス・ホミニス(Staphylococcus hominis)も、抗菌ペプチドなどの発現を促進することで病原菌への抵抗力を高める表皮常在菌であることが知られている。
一方、皮膚常在菌のなかには過剰に増殖することで有害となる菌も知られている。例えば、アクネ菌として知られるキューティバクテリウム・アクネス(Cutibacterium acnes)は、皮膚を保護する有用性が報告されている一方で、皮脂の分泌量が増えたり毛穴が詰まったりすると、過剰に増殖して、尋常性ざ瘡(ニキビ)を生じる原因となることが知られている。
また、黄色ブドウ球菌として知られるスタフィロコッカス・アウレウス(Staphylococcus aureus)は、皮膚バリアの破壊や炎症の原因となる毒素を産生し、肌荒れ状態にある人、乾燥肌状態にある人、アトピー性皮膚炎患者の皮膚に多く存在することが知られている。
また、放線菌として知られるコリネバクテリウム・キセロシス(Corynebacterium xerosis)は、皮脂から悪臭の原因物質を産生することが知られている。同じく放線菌として知られるマイクロコッカス・ルテウス(Micrococcus luteus)は、肌荒れ状態にある人の皮膚に多く存在するほか、汗を分解して悪臭物質を産生することが知られている。
以上のようなことから、皮膚常在菌叢は、肌の状態を良好に保つうえできわめて重要であると考えられている。
皮膚常在菌叢に関し、例えば、特許文献1には、α1-2糖骨格を有するグルコオリゴ糖は、皮膚に常在する有用菌の増殖を促し、悪臭の原因菌や病原菌などの好ましくない菌の増殖を阻害する作用効果があると記載されている。また、特許文献2には、イソマルトオリゴ糖および/またはイソマルトオリゴ糖を還元して得られる糖アルコールを含有する皮膚常在菌叢改善剤の発明が開示されている。また、特許文献3には、ウロン酸残基を有するキシロオリゴ糖を含有する皮膚常在菌叢改善剤の発明が開示されている。また、特許文献4には、セロビオースやこれを主成分とするセロオリゴ糖含有組成物は、黄色ブドウ球菌や緑膿菌に対して静菌作用又は増殖抑制作用を示す一方、表皮ブドウ球菌に対しては静菌作用又は増殖抑制作用を示さないことが記載されている。また、特許文献5には、ラクトスクロースを有効成分とする皮膚常在菌叢のバランス改善剤の発明が開示されている。
従来、各種オリゴ糖等により皮膚常在菌の菌叢を改善する試みが知られているが、需要者の便宜のために新たな素材の開発が望まれている。
本発明の目的は、肌の状態を良好に保つために有用な、皮膚常在菌叢改善剤を提供することにある。
上記目的を達成するため、本発明者らは種々研究した結果、肌に良い影響を与える菌を選択的に増殖する素材としてソホロースを見出し、本発明を完成するに至った。
すなわち、本発明は、その第1の観点においては、ソホロースを有効成分とする皮膚常在菌叢改善剤を提供するものである。
本発明において、前記皮膚常在菌叢改善剤は、キューティバクテリウム・アクネス(Cutibacterium acnes)、スタフィロコッカス・アウレウス(Staphylococcus aureus)、コリネバクテリウム・キセロシス(Corynebacterium xerosis)、及びマイクロコッカス・ルテウス(Micrococcus luteus)からなる群から選ばれた1種又は2種以上を静菌しつつ、スタフィロコッカス・エピデルミデス(Staphylococcus epidermidis)及び/又はスタフィロコッカス・ホミニス(Staphylococcus hominis)の増殖を促すためのものであることが好ましい。
一方、本発明は、その第2の観点においては、上記の皮膚常在菌叢改善剤を含有する皮膚外用組成物を提供するものである。
本発明によれば、ソホロースを用いることで、皮膚常在菌のうち、肌に悪い影響を及ぼす可能性を有する菌の成育を抑制しつつ、肌に良い影響を与える有益な菌の増殖を促すことができる。
以下、本発明について更に詳細に説明する。なお、本明細書において数値範囲について「~」の記載がある場合は、とくに断りがなければ以上から以下を表し、両端の数値をいずれも含むものとする。また、上限値および下限値を適宜組み合わせることができ、それにより得られた数値範囲も本明細書の開示に含まれるものとする。
本発明にかかる皮膚常在菌叢改善剤は、その有効成分としてソホロースを用いるものである。ソホロース(英語名:Sophorose、別称:2-O-β-D-グルコピラノシル-D-グルコース)は、2分子のグルコースがβ-1,2結合した二糖類である。豆科植物であるエンジュの鞘の色素の配糖体の糖成分として最初に見出されたが、自然界に遊離の二糖の状態で存在することは稀であり、天然にはごく僅かしか存在しない糖である。僅かにデンプンの酸加水分解物中やロイヤルゼリー中に存在することが知られているにすぎない。
ソホロースの入手は、特に限定するものではないが、例えば、糖転移・縮合反応を利用して酵素的に調製することが可能である。具体的には、例えば、特許第2750374号公報や特許第3020583号公報に詳細に説明されているように、微生物起源のβ-グルコシダーゼをグルコース及び/又はβ-グルコオリゴ糖に作用させ、β-グルコシダーゼが具備する縮合・糖転移反応を利用することによりソホロースを得ることができる。また、特許第7025941号公報に詳細に説明されているように、1,2-β-オリゴグルカンホスホリラーゼをα-グルコース-1-リン酸及びグルコースに作用させる方法によれば、ソホロースをより高収率に得ることができる。
これらの方法によりソホロースを含有する組成物が得られる。得られたソホロースを含有する組成物には、必要に応じて、ろ過、脱色、脱臭、脱塩などの処理を施すことができる。また、抽出、遠心分離、晶出、微生物資化、更に活性炭、多孔質担体、疎水性樹脂、親水性樹脂、イオン交換樹脂、吸着樹脂等を利用したクロマトグラフィーによる分画、透析、限界ろ過等の膜分画などの処理を施してもよい。これらの処理により、ソホロースの純度を高めることができる。
本発明において、ソホロースは、高純度化されたものとして利用することもできるが、ソホロースを含有するものであれば利用可能であり、すなわちソホロース含有糖組成物を利用することもできる。この場合、本発明に用いるソホロース含有糖組成物における、そのソホロースの含有量としては、特に制限はないが、例えば、乾燥固形分あたり10質量%以上であることが好ましく、30質量%以上であることがより好ましく、50質量%以上であることが更に好ましく、80質量%以上であることが最も好ましい。乾燥固形分あたりのソホロースの含有量が5質量%未満では、ソホロースの機能性を十分に発揮させることができなくなる場合がある。
なお、本発明に用いる上記ソホロース含有糖組成物としては、結合様式や重合度の異なる糖類、オリゴ糖等の他の成分を含むものであってもよい。典型的に例えば、重合度が2~10のオリゴ糖であるソホロオリゴ糖(ソホロースを除く)、ゲンチオオリゴ糖、セロオリゴ糖、ラミナリオリゴ糖などの1種又は2種以上を、それぞれ下記に示す乾燥固形分換算含有量で含むものなどを例示し得る。

後述する実施例で示されるように、ソホロースには、皮膚常在菌のうち、肌に悪い影響を及ぼす可能性を有する菌の成育を抑制しつつ(「静菌」ともいう)、肌に良い影響を与える有益な菌の増殖を促すことができる機能性が備わる。具体的には、肌に良い影響を与える有益な菌としては、限定されないが、例えば、表皮ブドウ球菌として知られるスタフィロコッカス・エピデルミデス(Staphylococcus epidermidis)、表皮ブドウ球菌として知られるスタフィロコッカス・ホミニス(Staphylococcus hominis)などである。一方、肌に悪い影響を及ぼす可能性を有する菌としては、アクネ菌として知られるキューティバクテリウム・アクネス(Cutibacterium acnes)、黄色ブドウ球菌として知られるスタフィロコッカス・アウレウス(Staphylococcus aureus)、放線菌として知られるコリネバクテリウム・キセロシス(Corynebacterium xerosis)、放線菌として知られるマイクロコッカス・ルテウス(Micrococcus luteus)などである。
本発明においては、上述したような機能性が備わるソホロースを有効成分とするので、皮膚常在菌叢バランスを維持して、肌の状態を良好に保つ目的で好適に用いられ得る。
具体的には、ある態様において、本発明は、皮膚常在菌叢のバランスを改善する目的で好適に用いられ得る。皮膚常在菌叢のバランスが崩れると、常在菌の過剰増殖や他の有害菌の侵入や増殖が起こり、様々な皮膚症状が誘発されるが、本発明によればそのような状態に陥ることを防いだり、その症状の程度を軽減したり抑止したりする目的で好適に用いられ得る。この場合、そのバランスの改善は、例えば、表皮ブドウ球菌の割合を増加させたり、黄色ブドウ球菌の割合を減少させたりすることであり得る。表皮ブドウ球菌は、病原性の菌、例えば黄色ブドウ球菌に対して拮抗作用を有し、そのような有害菌の増殖を阻止しうることから、表皮ブドウ球菌の増殖を促すことにより、皮膚常在菌叢のバランス調整剤としての機能をはたすことができる。また、皮膚常在菌叢のバランスの改善は、例えば、表皮ブドウ球菌等の有益菌の生育には影響を与えず、黄色ブドウ球菌等の有害菌の生育を抑制することであってもよい。
ある態様において、本発明は、皮膚を弱酸性に保ち、他の菌の感染や乾燥を防ぐ目的で好適に用いられ得る。例えば、表皮ブドウ球菌の増殖を促すことにより、この菌が皮脂を分解して、皮膚を弱酸性に保つ脂肪酸や保湿成分であるグリセリンを産生したり、病原性を有する細菌の増殖を抑制する抗菌ペプチドを産生したりするので、上述のような目的で好適に用いられ得る。
ある態様において、本発明は、有害菌からの毒素の発生を抑制する目的で好適に用いられ得る。例えば、黄色ブドウ球菌の生育を抑制することにより、この菌が毒素を生産するのを抑制するので、上述のような目的で好適に用いられ得る。
ある態様において、本発明は、ニキビを予防したり、その症状の程度を軽減したり抑止したりする目的で好適に用いられ得る。例えば、アクネ菌の生育を抑制することにより、この菌が原因となる尋常性ざ瘡(ニキビ)の症状の発症を予防したり、症状の程度を軽減したり抑止したりすることができるので、上述のような目的で好適に用いられ得る。
ある態様において、本発明は、体臭を予防したり、その程度を軽減したり抑止したりする目的で好適に用いられ得る。例えば、放線菌として知られるコリネバクテリウム・キセロシス(Corynebacterium xerosis)やマイクロコッカス・ルテウス(Micrococcus luteus)の生育を抑制することにより、この菌が皮脂や汗を分解して悪臭の原因物質を生じさせるのを抑えることができるので、上述のような目的で好適に用いられ得る。
ある態様において、本発明は、特定の皮膚症状に向けた予防やその症状の程度の軽減や抑止に限定されない。例えば、皮膚の健康を維持したり、肌の状態を向上させたり、外部からの病原菌の侵入を防ぐバリア機能を付与したりする目的で好適に用いられ得る。

本発明により提供される皮膚常在菌叢改善剤は、化粧品、外用医薬品、医薬部外品等、各種の皮膚外用組成物の形態で利用してもよい。皮膚外用組成物としては、例えば、化粧用クリーム類、乳液、化粧水、パック剤、スキンミルク(乳剤)、ジェル剤、パウダー、リップクリーム、口紅、アンダーメークアップ、ファンデーション、サンケア、浴用剤、ヘアシャンプー、ヘアリンス、ボディシャンプー、ボディリンス、石鹸、クレンジングフォーム、軟膏、貼付剤、ゼリー剤、エアゾール剤などが挙げられる。
本発明により提供される皮膚常在菌叢改善剤が皮膚外用組成物の形態で利用される場合、上述したソホロース又はソホロース含有糖組成物は、適宜常法に従い、製剤学的に許容される適当な製剤担体・成分を用いてそれぞれの形態に調製され得る。かかる製剤担体・成分としては、例えば、グリセリン、ワセリン、尿素、ヒアルロン酸、ヘパリン等の保湿剤;PABA誘導体(パラアミノ安息香酸、エスカロール507等)、桂皮酸誘導体(ネオヘリオパン、パルソールMCX、サンガードB等)、サリチル酸誘導体(オクチルサリチレート等)、ベンゾフェノン誘導体(ASL-24、ASL-24S等)、ジベンゾイルメタン誘導体(パルソールA、パルソールDAM等)、複素環誘導体(チヌビン系等)、酸化チタン等の紫外線吸収剤・散乱剤;エデト酸二ナトリウム、エデト酸三ナトリウム、クエン酸、クエン酸ナトリウム、酒石酸、酒石酸ナトリウム、乳酸、リンゴ酸、ポリリン酸ナトリウム、メタリン酸ナトリウム、グルコン酸等の金属封鎖剤;サリチル酸、イオウ、カフェイン、タンニン等の皮脂抑制剤;塩化ベンザルコニウム、塩化ベンゼトニウム、グルコン酸クロルヘキシジン等の殺菌・消毒剤;塩酸ジフェンヒドラミン、トラネキサム酸、グアイアズレン、アズレン、アラントイン、ヒノキチオール、グリチルリチン酸及びその塩、グリチルリチン酸誘導体、グリチルレチン酸等の抗炎症剤;ビタミンA、ビタミンB群(B1,B2,B6,B12,B15)、葉酸、ニコチン酸類、パントテン酸類、ビオチン、ビタミンC、ビタミンD群(D2,D3)、ビタミンE、ユビキノン類、ビタミンK(K1,K2,K3,K4)等のビタミン類;アスパラギン酸、グルタミン酸、アラニン、リジン、グリシン、グルタミン、セリン、システイン、シスチン、チロシン、プロリン、アルギニン、ピロリドンカルボン酸等のアミノ酸及びその誘導体;レチノール、酢酸トコフェロール、アスコルビン酸リン酸マグネシウム、アスコルビン酸グルコシド、アルブチン、コウジ酸、エラグ酸、胎盤抽出液等の美白剤;ブチルヒドロキシトルエン、ブチルヒドロキシアニソール、没食子酸プロピル等の抗酸化剤;塩化亜鉛、硫酸亜鉛、石炭酸亜鉛、酸化亜鉛、硫酸アルミニウムカリウム等の収斂剤;グルコース、フルクトース、マルトース、ショ糖、トレハロース、エリスリトール、マンニトール、キシリトール、ラクチトール等の糖類;甘草、カミツレ、マロニエ、ユキノシタ、芍薬、カリン、オウゴン、オウバク、オウレン、ジュウヤク、イチョウ葉等の各種植物エキス等の他、油性成分、界面活性剤、増粘剤、アルコール類、粉末成分、色素等が挙げられる。これら製剤担体・成分は、添加しようとする製品種別、形態に応じて常法的に行われる加工(例えば、粉砕、製粉、洗浄、加水分解、醗酵、精製、圧搾、抽出、分画、ろ過、乾燥、粉末化、造粒、溶解、滅菌、pH調整、脱臭、脱色等を任意に選択、組み合わせた処理)を行い、各種の素材から任意に選択して供すればよい。
なお、各種植物からの抽出物(生薬)または動物系原料由来の添加物を、本実施形態の皮膚外用組成物に供する場合、皮膚や頭髪の保護をはじめ、保湿、感触・風合いの改善、柔軟性の付与、刺激の緩和、芳香によるストレスの緩和、細胞賦活(細胞老化防止)、炎症の抑制、肌質・髪質の改善、肌荒れ防止及びその改善、発毛、育毛、脱毛防止、光沢の付与、清浄効果、疲労の緩和、血流促進、温浴効果等の美容的効果のほか、香付け、消臭、増粘、防腐、緩衝等の効果も期待できる。
更にこの他にも、これまでに知られている各原料素材の様々な美容的、薬剤的効果を期待し、これらを組み合わせることによって、本発明の目的とする皮膚常在菌改善の機能性に加えて、多機能的な効果を期待した製品とすることも可能である。
本発明により提供される皮膚常在菌叢改善剤又は皮膚外用組成物において、ソホロースの含有量は、乾燥固形分あたり0.01質量%から100質量%の範囲であり得る。乾燥固形分あたりの含有量が0.01質量%未満では、ソホロースによる機能性が十分に発揮されない場合がある。ソホロースの含有量としては、より好ましくは0.6質量%以上であり、更により好ましくは、0.75質量%以上である。
本発明により提供される皮膚常在菌叢改善剤又は皮膚外用組成物が液体状、ジェル状、クリーム状等の水分その他の媒質を含む形態である場合、そのような形態中でのソホロースの含有量としては、適宜設定してよく特に制限はないが、典型的に例えば、0.01mg/g~400mg/g(分母は製剤の重量を示す)の範囲であってよく、0.1mg/g~100mg/gの範囲であってよく、1mg/g~50mg/gの範囲であってよい。
本発明により提供される皮膚常在菌叢改善剤又は皮膚外用組成物は、これを皮膚に適用する場合、その施与量としては、適宜設定してよく特に限定されないが、例えば、ソホロースとして0.001mg/cm2~1mg/cm2(分母は適用する皮膚の面積を示す)の範囲であってよく、0.005mg/cm2~0.5mg/cm2の範囲であってよく、0.02mg/cm2~0.05mg/cm2の範囲であってよい。
以下に実施例を挙げて本発明について更に具体的に説明するが、これらの実施例は本発明の範囲を限定するものではない。
〔1.α-グルコオリゴ糖の調製〕
75(w/w)%グルコース溶液にグルコアミラーゼ酵素製剤を対固形分1gあたり50mg添加し、pHを5.0に調整後、66℃で保持した。反応63時間後、pHを4.0に調整し80℃に昇温することで酵素を失活させ、膜分画により生成物から単糖を除去してα-グルコオリゴ糖を得た。
75(w/w)%グルコース溶液にグルコアミラーゼ酵素製剤を対固形分1gあたり50mg添加し、pHを5.0に調整後、66℃で保持した。反応63時間後、pHを4.0に調整し80℃に昇温することで酵素を失活させ、膜分画により生成物から単糖を除去してα-グルコオリゴ糖を得た。
〔2.β-グルコオリゴ糖の調製〕
60(w/w)%グルコース溶液にβ-グルコシダーゼ酵素製剤を対固形分1gあたり9.3mg添加し、pHを6.6に調整後60℃で保持した。反応72時間後、pHを4.0に調整し80℃に昇温することで酵素を失活させ、膜分画により生成物から単糖を除去してβ-グルコオリゴ糖を得た。
60(w/w)%グルコース溶液にβ-グルコシダーゼ酵素製剤を対固形分1gあたり9.3mg添加し、pHを6.6に調整後60℃で保持した。反応72時間後、pHを4.0に調整し80℃に昇温することで酵素を失活させ、膜分画により生成物から単糖を除去してβ-グルコオリゴ糖を得た。
〔3.高純度ソホロースの調製〕
1.0Mスクロースと1.0Mグルコースを100mMリン酸カリウム緩衝液(pH7.0)に溶解した。この基質1mLあたりに、19mg/mLの1,2-β-オリゴグルカンホスホリラーゼ活性を有するビフィドバクテリウム・スカルドビイ由来のタンパク質(特許第7025941号に「BSGP」として開示されたタンパク質)と、17mg/mLスクロースホスホリラーゼ(Sigma-Aldrich社)とを10μLずつ加え、30℃で反応させた。7日後、10分間煮沸することで酵素を失活させ、反応液を適宜濃縮して、活性炭カラムに供して単糖及び二糖を除去した。これに、終濃度が85mMとなるように塩酸を添加して80℃で17時間保持して酸分解を行い、分解サンプルを以下の条件でHPLCに供し、二糖画分を回収して高純度ソホロースを得た。

〔4.高純度ラミナリビオースの調製〕
カードラン(三菱商事ライフサイエンス株式会社)8.0gと超純水720mLを混合し、1M 水酸化ナトリウムを8mL加えて10分間攪拌した。1M Tris-HCl緩衝液(pH7.0)40mLと1M 塩酸8mLを添加して攪拌後、酵母細胞壁溶解酵素(「Zymolyase(登録商標)」、ナカライテスク株式会社)80mgを加えた。終濃度が10mMとなるようにシステインを加え、勢いよく攪拌しながら37℃で一晩反応させた。80℃で10分間加熱して酵素を失活させ、室温まで冷却後遠心分離して上清を回収した。これを、高純度ソホロース調製と同じ条件でHPLCに供し、二糖画分を回収して高純度ラミナリビオースを得た。

〔5.高純度ゲンチオビオースの調製〕
上記〔2.β-グルコオリゴ糖の調製〕で調製した、二糖が主成分であるβ-グルコオリゴ糖を原資として、活性炭カラムを用いた分画を行い、高純度ゲンチオビオースを得た。

〔6.糖組成分析〕
糖組成分析はHPLCを用いて行った。得られるクロマトグラムのピーク面積より各重合度成分の含有量を求めた。グルコオリゴ糖の分析はHPLC条件A、高純度サンプルの分析はHPLC条件A及びHPLC条件Bに従って実施した。

表1,2には、糖組成分析の結果を示す。
表1に示されるように、上記で調製したグルコオリゴ糖は、いずれも二糖を主成分とする糖組成を有していた。
表2に示されるように、上記で調製したソホロース、ラミナリビオース、ゲンチビオースの高純度調製物は、いずれも少なくとも90質量%以上の純度を有していた。
[試験例1]
皮膚常在菌として表3に示す各種菌について、それぞれに適した培養条件で培養する際、その培地に上記で調製したα-グルコオリゴ糖又はβ-グルコオリゴ糖を添加して培養したときの菌数変化を調べた。

具体的には、各種菌をそれぞれに適した培地で一晩培養して前培養液とし、新鮮培地3mLに対して上記で調製したα-グルコオリゴ糖又はβ-グルコオリゴ糖を終濃度0.5(w/v)%となるように添加したうえ、これに前培養液0.03mLを混合して24時間培養し、培養後の菌濁度(OD660)を測定した。また、培地に糖質としてグルコースを添加して同様に試験して、比較対照とした。結果は、糖質の代わりに滅菌超純水を添加したときの菌濁度を100%としたときの相対値(対糖無添加区%)として示した。
その結果、図1に示されるように、グルコースの添加区では、S. epidermidisとS. hominisの増殖が顕著であり、それとともにS. aureusや、C. acnes、C. xerosisの増殖も認められ、特定の菌種への選択性に乏しい結果となった。また、α-グルコオリゴ糖の添加区でも、S. epidermidisのほかに、S. aureusやC. xerosisでの増殖が認められた。一方で、β-グルコオリゴ糖の添加区では、S. epidermidisとS. hominisでの増殖が認められたが、それ以外の4菌種では増殖がほとんど認められなかった。
以上から、二糖を主成分とするβ-グルコオリゴ糖には、皮膚に有害な菌の成育は抑えつつ、皮膚に有益な表皮ブドウ球菌(S. epidermidisやS. hominis)の増殖を促す作用効果があることが明らかとなった。
[試験例2]
二糖からなるβ-グルコオリゴ糖としてソホロース、ラミナリビオース、及びゲンチオビオースについて、S. epidermidisの増殖を促す効果を評価した。具体的には、培地に添加する糖質として、上記で高純度に調製したソホロース、ラミナリビオース、又はゲンチオビオースを用い、培地への添加量を終濃度0.3(w/v)%及び0.5(w/v)%とした以外は試験例1と同様にして、S. epidermidisの増殖を促す効果を評価した。
二糖からなるβ-グルコオリゴ糖としてソホロース、ラミナリビオース、及びゲンチオビオースについて、S. epidermidisの増殖を促す効果を評価した。具体的には、培地に添加する糖質として、上記で高純度に調製したソホロース、ラミナリビオース、又はゲンチオビオースを用い、培地への添加量を終濃度0.3(w/v)%及び0.5(w/v)%とした以外は試験例1と同様にして、S. epidermidisの増殖を促す効果を評価した。
その結果、図2に示されるように、各種のβ-グルコオリゴ糖(二糖)のうちソホロースにS. epidermidisの増殖を促す効果が顕著であった。一方、ラミナリビオースとゲンチオビオースではS. epidermidisの増殖を促す効果はみられなかった。
[試験例3]
二糖からなるβ-グルコオリゴ糖として、上記で高純度に調製したソホロースを用い、培地への添加量を終濃度0.1(w/v)%又は1.0(w/v)%とした以外は試験例2と同様にして、S. epidermidisの増殖を促す効果を評価した。

その結果、図3に示されるように、ソホロースを高純度に含有する調製物を培地に終濃度0.1(w/v)%となるように添加して培養した場合も、S. epidermidisの増殖を促す効果が認められた。一方、終濃度1.0(w/v)%では、試験例2において終濃度0.5(w/v)%のときに示されたS. epidermidisの増殖を促す効果と比較して、大きな差は認められなかった。
[試験例4]
二糖からなるβ-グルコオリゴ糖として、上記で高純度に調製したソホロースを用い、培地への添加量を終濃度0.1(w/v)%、0.5(w/v)%、又は1.0(w/v)%とした以外は試験例1と同様にして、S. hominis、S. aureus、C. acnes、C. xerosis、又はM. luteusの増殖を促す効果を評価した。
二糖からなるβ-グルコオリゴ糖として、上記で高純度に調製したソホロースを用い、培地への添加量を終濃度0.1(w/v)%、0.5(w/v)%、又は1.0(w/v)%とした以外は試験例1と同様にして、S. hominis、S. aureus、C. acnes、C. xerosis、又はM. luteusの増殖を促す効果を評価した。
その結果、図4に示されるように、ソホロースを高純度に含有する調製物を終濃度0.5(w/v)%あるいは1.0(w/v)%となるように培地に添加することで、S. hominisの増殖が、糖を添加しないコントロールに対する相対値(対糖無添加区%)として、それぞれ118.0%、118.8%に増加することが明らかとなった。一方で、いずれの添加量においても、コントロールと比較してS. aureus、C. acnes、C. xerosis、又はM. luteusの増殖を促す効果は認められなかった。
以上から、試験例2,3の結果をあわせて考慮すると、二糖からなるβ-グルコオリゴ糖であるソホロースには、皮膚に有害な菌の成育は抑えつつ、皮膚に有益な表皮ブドウ球菌(S. epidermidisやS. hominis)の増殖を促す作用効果があることが明らかとなった。
[試験例5]
二糖からなるβ-グルコオリゴ糖として、上記で高純度に調製したラミナリビオース及びゲンチオビオース、ならびに市販のセロビオース(ナカライテスク株式会社)を用い、培地への添加量を終濃度0.5(w/v)%とした以外は試験例1と同様にして、S. hominis、S. aureus、C. acnes、C. xerosis、又はM. luteusの増殖を促す効果を評価した。
二糖からなるβ-グルコオリゴ糖として、上記で高純度に調製したラミナリビオース及びゲンチオビオース、ならびに市販のセロビオース(ナカライテスク株式会社)を用い、培地への添加量を終濃度0.5(w/v)%とした以外は試験例1と同様にして、S. hominis、S. aureus、C. acnes、C. xerosis、又はM. luteusの増殖を促す効果を評価した。
その結果、図5に示されるように、ラミナリビオース、ゲンチオビオース、セロビオースの添加では増殖を促進する効果は確認されなかった。また、ラミナリビオースの添加区ではS. aureusの増殖が、糖を添加しないコントロールに対する相対値(対糖無添加区%)として108.5%に増加し、ゲンチオビオース、セロビオースの添加区では添加時のM. luteusの増殖が、糖を添加しないコントロールに対する相対値(対糖無添加区%)として、それぞれ121.9%、111.7%に増加した。
以上から、試験例2の結果をあわせて考慮すると、二糖からなるβ-グルコオリゴ糖であるラミナリビオース、ゲンチオビオース、セロビオースには、皮膚に有害な菌の成育は抑えつつ、皮膚に有益な表皮ブドウ球菌(S. epidermidisやS. hominis)の増殖を促す作用効果がソホロースに比べて乏しいことが明らかとなった。
(処方例1)
本発明にかかる皮膚常在菌叢改善剤としてソホロースを化粧水に用いる処方例である。

Claims (3)
- ソホロースを有効成分とする皮膚常在菌叢改善剤。
- キューティバクテリウム・アクネス(Cutibacterium acnes)、スタフィロコッカス・アウレウス(Staphylococcus aureus)、コリネバクテリウム・キセロシス(Corynebacterium xerosis)、及びマイクロコッカス・ルテウス(Micrococcus luteus)からなる群から選ばれた1種又は2種以上を静菌しつつ、スタフィロコッカス・エピデルミデス(Staphylococcus epidermidis)及び/又はスタフィロコッカス・ホミニス(Staphylococcus hominis)の増殖を促すためのものである、請求項1記載の皮膚常在菌叢改善剤。
- 請求項1又は2記載の皮膚常在菌叢改善剤を含有する皮膚外用組成物。
