Abstract
本発明は、安全であり毎日の服用も可能で健康食品としての利用もでき、体内における変性タンパク質を低減できる変性タンパク質低減剤とその製造方法を提供することを目的とする。また、本発明は、変性タンパク質を低減するための胡麻(Sesamum indicum)の地上部、又はその抽出物の使用と、変性タンパク質を低減するための方法を提供することも目的とする。本発明に係る変性タンパク質低減剤は、胡麻(Sesamum indicum)の地上部、又はその抽出物を有効成分として含むことを特徴とする。
Description
本発明は、安全であり毎日の服用も可能で健康食品としての利用もでき、体内における変性タンパク質を低減できる変性タンパク質低減剤とその製造方法に関するものである。
加齢、過食、偏食などによって、血中のブドウ糖が過剰になり、体の各組織の構成成分の根幹をなす細胞内外のタンパク質が糖化するなどして変性タンパク質が生成蓄積する。また、特に揚げる、焼く、炒めるなどの加熱調理した動物性脂肪食品には、終末糖化産物(AGEs)が含まれる。その他、清涼飲料水や菓子類などに多く使われる人工甘味料の中には、ブドウ糖よりも迅速に終末糖化産物を生成するものがあるといわれている。これら変性タンパク質は、強い毒性を持ち、老化を進める原因物質とされている。例えば、外観上は皮膚のくすみやしわ、老人斑などの原因となるのみでなく、しなやかな動きを減少させ、各組織機能の低下や劣化の原因となる。また、終末糖化産物が血管に蓄積すると、心筋梗塞や脳梗塞、骨に蓄積すると骨粗鬆症、目に蓄積すると白内障の一因となり、全身の健康に影響を及ぼす(特許文献1~8,非特許文献1~7)。糖尿病の診断に用いられているヘモグロビンA1cも、終末糖化産物の一種である。
ヒトでの疾患と終末糖化産物との関連性、終末糖化産物の生成の抑制や、終末糖化産物を分解する天然物などは多数の報告があるにも拘わらず、ヒトが摂取して具体的な改善作用を示す安全で安価な医薬品や天然物は見当たらない。変性タンパク質は生活習慣などにより極めて微量ずつ体内に蓄積していくと考えられるので、毎日摂取可能な安全な薬剤であれば、なお望ましい。
胡麻は古くから栄養価の高い食品として知られており、生薬としても利用されている。また、その含油率の高さから、胡麻油も採取される。胡麻に含まれるセサミンは、抗酸化作用、コレステロール吸収阻害、抗がん作用など、様々な生理活性を示すといわれている。
胡麻に関しては、主に種子が食用や研究に付されており、葉など種子以外の地上部の生理活性については、ほとんど知られていない。中国最古の薬物学書である「神農本草経」には、胡麻の種子と共に葉の効能効果の記載が見られ、元の時代に出版された「食物本草」にも同様の記載が見られるが、ヒトが摂取した場合の栄養生理学や医学的評価は不明である。今でも西アフリカでは胡麻の間引き菜は食用とされており、また、若葉をスープに入れて食べるなど食材としても用いられている。
例えば、胡麻種子を発芽させてから4日経過後のスプラウトのラジカル補足作用についての報告はある(非特許文献8)。しかし、発芽から5日以上経過した地上部についての生理活性についての報告は全く見られない。

上述したように、終末糖化産物などの変性タンパク質は、体内で様々な不調や疾患などの原因になり、その低減作用を有するとされている成分は種々研究されている。
しかし、恒常的な摂取が可能なほど安全で、且つヒトへの投与で実際に体内の変性タンパク質を低減できる成分は知られていない。
そこで、本発明は、安全であり毎日の服用も可能で健康食品としての利用もでき、体内における変性タンパク質を低減できる変性タンパク質低減剤とその製造方法を提供することを目的とする。また、本発明は、変性タンパク質を低減するための胡麻(Sesamum indicum)の地上部、又はその抽出物の使用と、変性タンパク質を低減するための方法を提供することも目的とする。

本発明者らは、上記課題を解決するために鋭意研究を重ねた。その結果、食用にも付され安全な胡麻の地上部に、体内の変性タンパク質を有効に低減できる成分が含まれていることを見出して、本発明を完成した。

本発明に係る変性タンパク質低減剤の有効成分は、食用にも付される胡麻の地上部に含まれるものであるので安全であり、毎日の恒常的な摂取も可能である。また、本発明に係る変性タンパク質低減剤は、特に変性タンパク質の蓄積が問題となっている被検者への投与により、変性タンパク質を実際に低減することができる。従って本発明は、健康食品などとして人々の恒常的な健康に寄与するものとして、産業上非常に優れている。
本発明に係る変性タンパク質低減剤は、胡麻(Sesamum indicum)の地上部、又はその抽出物を有効成分として含む。本開示において有効成分とは、本発明に係る変性タンパク質低減剤に含まれる成分のうち変性タンパク質低減作用を有する成分をいい、換言すれば、本発明に係る変性タンパク質低減剤は、変性タンパク質低減効果が発揮される量の胡麻の地上部又はその抽出物を含む。具体的には、特に制限されないが、例えば、本発明に係る変性タンパク質低減剤における胡麻の地上部又はその抽出物の割合を10質量%以上、100質量%以下とすることができる。
胡麻(Sesamum indicum)は、ゴマ科ゴマ属の一年草であり、大きさや色調など種子の形状の違いに基づくと約3000種あると言われる。胡麻の種類は特に制限されないが、本発明者により、Sa Mong Nan Net Thaeとごまぞうの優れた変性タンパク質低減作用が実験的に確認されている。
Sa Mong Nan Net Thaeは、ミャンマーの特定地域で古くから栽培されているものであり、小粒の黒胡麻であり、「胡麻黒八(登録商標)」との製品名でも知られている。Sa Mong Nan Net Thaeの種子は、品種登録されているリグナン胡麻(登録番号:19697,出願番号:20790,農林水産植物の種類:Sesamum indicum L.,品種名称:ITCFA2001;登録番号:19698,出願番号:20791,農林水産植物の種類:Sesamum indicum L.,品種名称:ITCFA2002)の種子と重量や外観が非常によく似ているが、本発明者らは、ゲノム解析の結果、両者ゲノム間には大きな配列の挿入や欠失が見られるなど、進化的に異なる品種であることを明らかにした。胡麻黒八とリグナン胡麻のゲノムの差異は、PCRにより検証することが可能である。
ごまぞうは、多収の白胡麻系統Toyama016とセサミンおよびセサモリンの含有量が高い南中国原産の熱帯型遺伝資源系統H65の交配後代より選抜された、日本国内での栽培が可能な高リグナン含有品種である。
地上部とは、胡麻種子を播種して育成したとき、地中に無く、地上から見られる部分をいうものとする。胡麻の地上部は、育成した胡麻から地下部を切断除去することにより得ることができる。
胡麻の地上部は、例えば、本葉が2枚以上見られる株のものであることが好ましい。また、胡麻の地上部は、完熟種子が結実するまでに採取することが好ましい。また、草丈が50cm以上、200cm以下のものや、1以上の蕾を有するもので開花前のもの、1以上の開花が見られるまでのもの等も採取することができる。播種から採取までの期間は適宜調整すればよいが、例えば、1ヵ月以上とすることができ、2ヵ月以上または2.5ヵ月以上が好ましく、また、3.5ヵ月以下とすることができ、3ヵ月以下が好ましい。
胡麻の地上部は、そのまま用いてもよいが、例えば、水洗、乾燥、粉砕、加水分解および抽出から選択される1以上の処理を行ってもよい。加水分解処理としては、例えば、25±5℃、湿度90%以上、100%以下で、1時間以上、5時間以下程度処理したり、ドラム中で回転させた後に加熱して加水分解を停止させることが挙げられる。加水分解処理により、苦味を低減できる可能性がある。
本発明に係る変性タンパク質低減剤の有効成分の形態は特に制限されず、例えば、液状、ペースト状、及び粉末状が挙げられる。粉末には、粗粉末、マイクロパウダー、及びナノマイクロパウダーが含まれる。
本発明に係る変性タンパク質低減剤の形態は特に制限されないが、例えば、固形状、ゲル状、及び液状が挙げられる。より具体的には、散剤、細粒剤、顆粒剤、錠剤、被覆錠剤、カプセル剤、トローチ剤、及び液剤などが挙げられる。
本発明に係る変性タンパク質低減剤は、その剤形に応じて、有効成分以外の添加剤を含んでいてもよい。添加剤としては、例えば、賦形剤、結合剤、滑沢剤、崩壊剤、乳化剤、安定剤、吸収促進剤、分化剤、及び防腐剤が挙げられ、その配合量は特に制限されず、適宜調整すればよい。
本発明に係る変性タンパク質低減剤は、例えば、以下の工程を含む方法により製造することができる。
1.胡麻の播種生育工程
本工程では、胡麻の種子を播種し、生育させる。本工程は、胡麻の通常の育て方に準じて行えばよい。例えば、胡麻の種子は、最高気温が20℃を超えるようになってから、日本では5月中旬から6月中旬に播けばよい。具体的には、深さ1cm程の穴を開け、穴1つ当たり5~6粒の種を入れた後、土を被せて水を十分に与える。本葉が1~2枚のところで、1ヶ所に3本くらいになるよう、生育の悪いものを間引き、本葉が3~4枚のときに2本、本葉が5~6枚のときに1本になるよう間引く。

本工程では、生育した胡麻の地上部を採取する。具体的には、生育した胡麻の地下部から地上部を分離すればよい。採取の時期や後処理などは、上述した通りである。

3.抽出工程
本工程では、胡麻の地上部から溶媒を使って有効成分を抽出する。本工程は、実施しても実施しなくてもよいが、抽出により、有効成分の変性タンパク質低減作用をより一層高めることができる可能性がある。

本工程で抽出に用いる溶媒は、特に制限されず適宜選択すればよいが、安全性の観点から、例えば水系溶媒が挙げられる。水系溶媒は、水、水と水溶性有機溶媒との混合溶媒、又は水溶性有機溶媒をいう。混合溶媒における水溶性有機溶媒の割合は、適宜調整すればよいが、例えば、50質量%以上、98質量%以下とすることができ、60質量%以上または65質量%以上が好ましく、70質量%以上がより好ましく、95質量%以下または90質量%以下が好ましく、80質量%以下がより好ましい。水溶性有機溶媒としては、例えば、メタノール、エタノール、2-プロパノール等のアルコール系溶媒;テトラヒドロフラン等のエーテル系溶媒;ジメチルホルムアミドやジメチルアセトアミド等のアミド系溶媒:ジメチルスルホキシド等のスルホキシド系溶媒を挙げることができる。人体に害が比較的少ないという観点から、水溶性有機溶媒としてはエタノールが好適である。また、水の種類は特に制限されず、精製水、蒸留水、純水、水道水などを特に制限なく使用することができる。
本工程における抽出条件は特に制限されず、常法を用いることができる。例えば、胡麻の地上部1gあたり5mL以上、100mL以下程度の溶媒を加え、30分間以上、10時間以下程度抽出すればよい。抽出温度も特に制限されず、常温で抽出してもよいし、溶媒の沸点以下で抽出してもよいし、加熱還流してもよい。例えば溶媒として水を使う場合には、60℃以上、100℃以下で加熱してもよい。
抽出後は、一般的な後処理を行ってもよい。例えば、抽出後の混合物から濾過や遠心分離により固形分を除去してもよい。更に、得られた溶液を、蒸留したり、濃縮したり、乾燥してもよい。乾燥方法としては、例えば、加熱乾燥、減圧乾燥、凍結乾燥、スプレードライが挙げられる。また、クロマトグラフィ等により、有効成分を更に精製してもよい。
本発明に係る変性タンパク質を低減するための方法は、胡麻(Sesamum indicum)の地上部、又はその抽出物を有効成分として投与する工程を含む。
本発明に係る変性タンパク質低減剤の使用量は、予防的使用であるか治療的使用であるか、患者の重篤度、その他の状態、年齢、性別などに応じて適宜調整すべきであり、特に制限されない。例えば、本発明に係る変性タンパク質低減剤は、服用する場合には固形分換算で1日あたり10mg以上、外用の場合には100mg以上用いることができる。本発明に係る変性タンパク質低減剤は非常に安全なものであることから、使用量の上限は特に制限されないが、例えば、服用の場合には固形分換算で1日あたり2g以下、外用の場合には5g以下とすることができる。また、1日あたりの使用回数は、1回以上、5回以下程度とすることができる。
本発明に係る変性タンパク質低減剤は、ヒトに限らず、ヒト以外の動物にも投与可能である。ヒト以外の投与対象動物としては、例えば、ウマ、ウシ、ブタ、ヒツジ、ヤギ、ラクダ、ラマなどの家畜;競走馬などの競技動物;イヌ、ネコなどの愛玩動物;マウス、ラット、モルモット、ウサギなどの実験動物;ニワトリ、アヒル、七面鳥、駝鳥などの家禽;養殖魚などの魚介類などが挙げられる。
本発明に係る変性タンパク質低減剤は、極めて優れる変性タンパク質低減効果を有する。具体的には、タンパク質の糖化や多量化の抑制、いったん形成された変性タンパク質の分解、及び変性タンパク質の分解酵素の活性化といった作用を示す。よって、本発明に係る変性タンパク質低減剤は、タンパク質の変性が関与する疾患や症状の予防や治療に有効である。かかる疾患や症状としては、例えば、肌のシワ、シミ、クスミ、たるみ等;白内障、動脈硬化、腎機能障害などの糖尿病性合併症;記憶障害や認知症などの老化;がん等を挙げることができる。

以下、実施例を挙げて本発明をより具体的に説明するが、本発明はもとより下記実施例によって制限を受けるものではなく、前・後記の趣旨に適合し得る範囲で適当に変更を加えて実施することも勿論可能であり、それらはいずれも本発明の技術的範囲に包含される。
実施例1
ミャンマーの特定の地域で古くから栽培されており、現地ではSa Mong Nan Net Thae(以下、「SMNNT」と略記する)と呼ばれている小粒の黒胡麻を播種し、約3ヵ月後、蕾が見られる頃の地上部から地下部を切断除去し、37℃で72時間乾燥した。得られた乾燥SMNNT地上部をハンマーミルにより粉砕した。

実施例1の乾燥SMNNT地上部粉末2gを水40mLと混合し、80℃で1時間加熱した。次いで、濾過、遠心分離し、固形分を除いて抽出液を得た。得られた抽出液から5mLを取り、105℃で1時間加熱して溶媒を除去し、固形分量を測定した。

実施例7: ゲノム解析
SMNNTの種子は、1g中の種子数および色調から、いわゆるリグナン胡麻の種子と非常によく似ている。そこで、両者からDNAを調製し、ゲノム解析を行った。
胡麻ゲノムの塩基数は約2億7000万塩基であることがわかっており、我々のゲノム解析では、1品種当たり胡麻ゲノムの塩基数の約55倍の配列データを得たので、任意のゲノム領域を55回解析したことになる。従って、確率論的には、全てのゲノム領域が網羅されており、解析プログラムにより、読み間違いは補正できていると判断される。
ゲノム解析の結果、SMNNTとリグナン胡麻のゲノム間には大きな配列の挿入や欠失が見られるなど、SMNNTとリグナン胡麻は進化的に異なる品種であることが明らかとなった。得られた解析データより、SMNNTとリグナン胡麻のゲノムの差異は、PCRにより検証することが可能である。

試験例1: AGEs化タンパク質架橋形成抑制作用の評価
AGEs化タンパク質架橋形成抑制作用を、Perera H.K.I.ら,Asian J.Med.Sci.,6(1),28-33(2015)に記載の方法を参考にして評価した。具体的には、0.1mol/Lのリン酸緩衝液(pH7.4)、各抽出液、タンパク質としてリゾチーム、及び糖としてフルクトースを混合し、60℃で40時間インキュベートした。リゾチームとフルクトースの濃度は、それぞれ5mg/mLと0.5mol/Lに調整した。各抽出液の添加量は、各固形分量から、固形分の濃度が図1および図2に示すようになるように調整した。また、対照として、抽出液の代わりに抽出溶媒を添加し、更に陽性対照として、アミノグアニンを添加した。比較のために、フルクトースを添加せずに同様に反応液を得た。
得られたリゾチーム糖化反応液、蒸留水、及び2-メルカプトエタノール試料溶液を5:5:2(容量比)で混合して反応させた。次いで、95℃で5分間加熱することにより架橋反応を停止させ、冷却後、常法に従って、SDS-PAGE電気泳動を行った。ゲル濃度は4-20%とし、電流値は20mAとした。泳動後、リゾチームのバンドをCBBで染色し、メタノール-酢酸によって脱色した。結果を図1および図2に示す。なお、リゾチームの単量体の分子量は13.1kDa、二量体の分子量は25.8kDa、三量体の分子量は40.5kDaである。また、図1および図2中、「Fru」はフルクトースを示し、「AG」はアミノグアニンを示す。また、図1中のレーンの説明を表1に、図2中のレーンの説明を表2に示す。
AGEs化タンパク質架橋形成抑制作用を、Perera H.K.I.ら,Asian J.Med.Sci.,6(1),28-33(2015)に記載の方法を参考にして評価した。具体的には、0.1mol/Lのリン酸緩衝液(pH7.4)、各抽出液、タンパク質としてリゾチーム、及び糖としてフルクトースを混合し、60℃で40時間インキュベートした。リゾチームとフルクトースの濃度は、それぞれ5mg/mLと0.5mol/Lに調整した。各抽出液の添加量は、各固形分量から、固形分の濃度が図1および図2に示すようになるように調整した。また、対照として、抽出液の代わりに抽出溶媒を添加し、更に陽性対照として、アミノグアニンを添加した。比較のために、フルクトースを添加せずに同様に反応液を得た。
得られたリゾチーム糖化反応液、蒸留水、及び2-メルカプトエタノール試料溶液を5:5:2(容量比)で混合して反応させた。次いで、95℃で5分間加熱することにより架橋反応を停止させ、冷却後、常法に従って、SDS-PAGE電気泳動を行った。ゲル濃度は4-20%とし、電流値は20mAとした。泳動後、リゾチームのバンドをCBBで染色し、メタノール-酢酸によって脱色した。結果を図1および図2に示す。なお、リゾチームの単量体の分子量は13.1kDa、二量体の分子量は25.8kDa、三量体の分子量は40.5kDaである。また、図1および図2中、「Fru」はフルクトースを示し、「AG」はアミノグアニンを示す。また、図1中のレーンの説明を表1に、図2中のレーンの説明を表2に示す。
図1と図2に示される結果の通り、リゾチームはフルクトースにより糖化され、更に2-メルカプトエタノールにより特に二量体となり、三量体の形成も観察される。それに対して、SMNNTおよびごまぞうの地上部抽出物により、二量体の形成が抑制され、三量体はほぼ見られない。
次いで、各ゲルの撮影画像を画像処理ソフトウェア「ImageJ」によって処理し、タンパク質二量体と三量体に対応するバンドの強度を数値化し、下記式により架橋形成抑制率を算出した。結果を図3に示す。

図3に示される結果の通り、陽性対照であるアミノグアニンに比べて、本発明に係る抽出物はいずれもタンパク質の架橋を有効に抑制し、特に熱水抽出液よりも70%エタノール抽出液の方が強い架橋抑制作用を示した。また、SMNNT地上部由来の70%エタノール抽出物は、ごまぞう地上部抽出物よりも強い架橋抑制作用を示した。更に、陽性対照であるアミノグアニンよりも、SMNNT地上部由来の抽出物の架橋抑制作用は強力であった。

図3に示される結果の通り、陽性対照であるアミノグアニンに比べて、本発明に係る抽出物はいずれもタンパク質の架橋を有効に抑制し、特に熱水抽出液よりも70%エタノール抽出液の方が強い架橋抑制作用を示した。また、SMNNT地上部由来の70%エタノール抽出物は、ごまぞう地上部抽出物よりも強い架橋抑制作用を示した。更に、陽性対照であるアミノグアニンよりも、SMNNT地上部由来の抽出物の架橋抑制作用は強力であった。
試験例2: AGEs架橋切断作用の評価
いったん形成されたAGEsタンパク質間架橋のSMNNT地上部による切断作用を、Vasan Sら,Nature,382,275-278(1996)を参照して評価した。
具体的には、実施例3のSMNNT地上部抽出液、又は陽性対照としてN-フェナシルチアゾリウムブロマイド(PTB)、10mmol/L 1-フェニル-1,2-プロパンジオン(PPD)、及び0.2mol/Lリン酸緩衝液(pH7.4)を、抽出物固形分:PPD:リン酸緩衝液=5:1:4(質量比)となるよう混合し、37℃で8時間反応させた(n=3)。なお、タンパク質間架橋の切断作用を有する物質が反応系に含まれると、下記反応式の通りPPDが分解されて安息香酸が生じる。
いったん形成されたAGEsタンパク質間架橋のSMNNT地上部による切断作用を、Vasan Sら,Nature,382,275-278(1996)を参照して評価した。
具体的には、実施例3のSMNNT地上部抽出液、又は陽性対照としてN-フェナシルチアゾリウムブロマイド(PTB)、10mmol/L 1-フェニル-1,2-プロパンジオン(PPD)、及び0.2mol/Lリン酸緩衝液(pH7.4)を、抽出物固形分:PPD:リン酸緩衝液=5:1:4(質量比)となるよう混合し、37℃で8時間反応させた(n=3)。なお、タンパク質間架橋の切断作用を有する物質が反応系に含まれると、下記反応式の通りPPDが分解されて安息香酸が生じる。
次いで、0.7mol/L塩酸を加えて反応を停止させた。反応液を遠心分離した後、上清中の安息香酸量をHPLCで分析し、下記式によりAGEs架橋切断率を求めた。実験は3例ずつ行った。結果を図4に示す。

図4に示される結果の通り、SMNNTの地上部の抽出液は、陽性対照であるPPDよりも強いAGEs架橋物の切断作用を有することが示された。

試験例3: 酸化タンパク質分解酵素活性化作用の評価
糖化や酸化された損傷タンパク質は、酸化タンパク質分解酵素(以下、「OPH」と略記する)によって分解される。そこで、アシルアミノアシッドリリーシング酵素(タカラバイオ社製,以下、「AARE」と略記する)を用い、基質としてN-アセチル-L-アラニン-p-ニトロアニリド(Bachem社製,以下、「AAPA」と略記する)を用い、酸化タンパク質分解酵素活性化作用を評価した。
0.1mol/LのTris-塩酸(pH7.4)、2mmol/LのAAPA、1mU/mLのAARE、及び抽出固形分の濃度が図5に示す濃度になるように実施例2または実施例3の抽出液を混合した。得られた混合液250μLを37℃で60分間インキュベートして、遊離したp-ニトロアニリド(以下、「pNA」と略記する)の量を、分光光度計を使って405nmの吸光度を測定した。また、対照として、抽出液の代わりに蒸留水を使って測定した。得られた測定値に基づいて、OPH活性化率を下記式により算出した。結果を図5に示す。

図5に示される結果の通り、SMNNTの地上部の抽出物は、AGEsを分解する酸化タンパク質分解酵素の活性を強力に増強することが明らかとなった。
糖化や酸化された損傷タンパク質は、酸化タンパク質分解酵素(以下、「OPH」と略記する)によって分解される。そこで、アシルアミノアシッドリリーシング酵素(タカラバイオ社製,以下、「AARE」と略記する)を用い、基質としてN-アセチル-L-アラニン-p-ニトロアニリド(Bachem社製,以下、「AAPA」と略記する)を用い、酸化タンパク質分解酵素活性化作用を評価した。
0.1mol/LのTris-塩酸(pH7.4)、2mmol/LのAAPA、1mU/mLのAARE、及び抽出固形分の濃度が図5に示す濃度になるように実施例2または実施例3の抽出液を混合した。得られた混合液250μLを37℃で60分間インキュベートして、遊離したp-ニトロアニリド(以下、「pNA」と略記する)の量を、分光光度計を使って405nmの吸光度を測定した。また、対照として、抽出液の代わりに蒸留水を使って測定した。得られた測定値に基づいて、OPH活性化率を下記式により算出した。結果を図5に示す。

図5に示される結果の通り、SMNNTの地上部の抽出物は、AGEsを分解する酸化タンパク質分解酵素の活性を強力に増強することが明らかとなった。
試験例4: 臨床試験
ヒトでの皮膚への変性タンパク質蓄積に対する効果を検証するための“プラセボ対照ランダム化単盲検並行群間比較試験”を、文部科学省、厚生労働省および経済産業省告示第1号「人を対象とする生命科学・医学系研究に関する倫理指針」(令和3年3月23日発令)を遵守して行った。
男女、年齢など無作為に合計57名選択し、まず摂食前の皮膚のAGEs年齢を、AGE Scanner(Diagnoptics Technologies社製)を用いて計測した。当該日の夜から、実施例1のSMNNT地上部粉末を、1回あたり1.5g、1日2回、朝と夜に6ヶ月間摂食してもらった。計測は毎月1回行い、その他の食事内容はじめ生活習慣などに対しては特別な指示を行わずに試験を行った。プラセボ群には、SMNNT地上部粉末の代わりに、モロヘイヤ粉末を同様に摂食してもらい、計測を行った。
20歳台の被験者のAGE ScannerでのAGEs年齢は、平均して19.5±6.5歳であり、AGEsの初回測定年齢と実年齢との差は10歳以内であった。また、40歳台の被験者の場合は平均42.7±4.2歳であり、AGEsの初回測定年齢と実年齢との差はこちらも10歳以内で実年齢との差はそれほど見られなかった。しかし、50歳台になると大きなバラツキが見られ、平均は52.9±4.0であったが、AGEs初回測定年齢と実年齢では20~30歳ほどの差のある被験者も見られた。AGEsの異常蓄積には、15年とか20年など長期に及ぶ生活習慣、特に食生活の関与が推定される。
スタート時のAGEs年齢が20歳~40歳台の被験者では、変性タンパク質の皮膚への蓄積が少なく、また、プラセボ群には効果は顕著に見られなかった。しかし、AGEs Scannerでの計測で、試験開始時のAGEs年齢が70歳以上となった54~76歳までの7名は、スタート時のAGEs年齢平均78.4±3.4歳が僅か6ケ月の選抜SMNNTのUPS粉末の摂食により69.6±5.3歳となり、減少傾向が確認された。その結果を図6に示す。

- 胡麻(Sesamum indicum)の地上部、又はその抽出物を有効成分として含むことを特徴とする変性タンパク質低減剤。
- 前記変性タンパク質が終末糖化産物である請求項1に記載の変性タンパク質低減剤。
- 前記変性タンパク質がタンパク質多量体である請求項1に記載の変性タンパク質低減剤。
- 変性タンパク質を低減するための胡麻(Sesamum indicum)の地上部、又はその抽出物の使用。
- 前記変性タンパク質が終末糖化産物である請求項4に記載の使用。
- 前記変性タンパク質がタンパク質多量体である請求項4に記載の使用。
- 変性タンパク質を低減するための方法であって、
胡麻(Sesamum indicum)の地上部、又はその抽出物を有効成分として投与する工程を含むことを特徴とする方法。 - 前記変性タンパク質が終末糖化産物である請求項7に記載の方法。
- 前記変性タンパク質がタンパク質多量体である請求項7に記載の方法。
- 変性タンパク質低減剤を製造するための方法であって、
胡麻(Sesamum indicum)の種子を播種し、生育させる工程、及び、
生育した胡麻の地上部を採取する工程を含むことを特徴とする方法。 - 1以上の蕾が見られ且つ開花前の胡麻の地上部を採取する請求項10に記載の方法。
- 更に、胡麻の地上部から溶媒を使って有効成分を抽出する工程を含む請求項10に記載の方法。
- 前記溶媒としてアルコール系溶媒を用いる請求項12に記載の方法。
