Abstract
본 발명은 다양한 그람양성균으로부터 유래한 리포테이코익산이 칸디다 진균의 부착과 응집을 억제하는 것을 이용한 항진균용 조성물, 칸디다증 감염 예방 및 치료용 약학적 조성물 및 이의 용도에 관한 발명으로, 이를 이용한 경우, 칸디다 진균의 감염과 칸디다증의 대표적 독성 인자인 부착력과 응집력을 저해하는 효과를 확인하였는 바, 칸디다 진균 항진균용 및 칸디다증 감염과 관련된 예방 또는 치료용으로 이용할 수 있다.
Description
본 발명은 다양한 그람양성균으로부터 유래한 리포테이코익산이 칸디다 진균의 부착과 응집을 억제하는 것을 이용한 항진균용 조성물, 칸디다증 감염 예방 및 치료용 약학적 조성물 및 이의 용도에 관한 것이다.
칸디다 진균은 기존 생물막을 형성하는 것으로 알려진 세균과는 다른 고등생물의 진균으로, 원핵세포인 세균과는 달리 진핵세포라는 특징을 갖는다. 칸디다 진균은 환경에 따라 효모형, 가균사, 그리고 균사의 형태로 성장하며, 칸디다 진균의 이러한 자유로운 세포 형태 변환 특성을 통해 인간 조직으로의 침투 감염을 일으키고 포식세포로부터의 회피와 혈류내의 분산을 쉽게 할 수 있으며 결과적으로 사람의 조직이나 혈류 내부, 또는 인공적으로 인체 내에 삽입된 카테터와 같은 의료기구에 부착력이 강력한 생물막을 형성하게 된다. 칸디다 진균의 표면 부착은 감염으로 인한 질병에서 중요한 역할을 한다.
칸디다증(Candidiasis)은 진균의 일종인 칸디다 진균에 의해 발생한다. 신체의 다양한 부위에 감염되어 발생하는 감염 질환으로 아구창, 외음부염, 그리고 질염 등의 증상이 나타난다. 칸디다 진균 중, 칸디다 알비칸스는 건강한 사람의 점막과 위장에 상주하는 상재균이지만 면역기능에 손상이 생기거나 중증의 질병을 앓는 환자들에게서 감염을 유발시키는 기회감염균이다. 또한 칸디다 알비칸스에 의해 일어나는 흔한 질염인 칸디다성 질염은 여성 10명 중 평균 7.5명은 일생동안 한 번 이상 걸릴 정도로 발생 빈도가 높은 질병이며, 다양한 합병증을 유발하기도 한다.
칸디다 진균의 생물막은 진균의 강력한 부착력과 응집력을 통해 형성되며, 효모형 세포와 비교하였을 때 약 2000배 높은 항진균제에 대한 저항성을 가지며, 진핵생물로서 사람의 세포와 공통된 특징을 보유하기 때문에 진균 제거에 특이적인 의약품의 개발이 어렵고 몇몇 환자에게선 심한 부작용이 나타나는 만큼, 높은 치료 효과와 낮은 독성을 갖는 항진균제의 개발이 시급한 상황이다.
리포테이코익산은 그람양성균에서 유래한 물질로 세포벽의 주요 구성 성분 중 하나이며 세균의 성장과 부착, 숙주의 면역체계 활성 등의 역할을 한다.
대한민국 공개특허공보 10-2019-0017168은 리포테이코익산의 피부 재생 및 창상 치유용 약학적 조성물에 대해서 밝히고 있으나, 칸디다증을 억제한다는 정보는 개시되어 있지 않다.
이러한 배경하에서, 본 발명자들은 칸디다증을 보다 효과적으로 억제할 수 있는 조성물을 개발하고자 예의 연구 노력한 결과, 그람양성균으로부터 분리한 리포테이코익산이 칸디다 진균의 표면 부착력을 감소시키는 것을 확인함으로써, 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 i) 리포테이코익산 또는 이의 약학적으로 허용 가능한 염; 또는 ii) 리포테이코익산을 포함하는 세포벽 추출물을 포함하는 칸디다(Candida) 항진균용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 조성물을 포함하는 칸디다증 감염 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 조성물을 포함하는 코팅제, 건강기능식품, 가축사료 첨가제 및 식품 세정제를 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
이에 본 발명자들은 그람양성균 및 대표적인 유산균으로 알려진 락토바실러스로부터 리포테이코익산을 높은 순도로 분리 정제하였으며, 진균 세포에서의 다양한 효능 평가를 수행한 결과, 리포테이코익산 단독의 적은 용량으로도 칸디다 진균의 감염과 칸디다증의 대표적 독성 인자인 부착력과 응집력을 저해하는 효과를 in vitro와 ex vivo 실험모델을 통해 검증하였다. 또한, 억제 효과는 리포테이코익산이 Hsp70과 상호작용함으로써 일어난다는 작용기전을 밝힘으로써 본 발명을 완성하였다.
따라서, 본 발명은 i) 리포테이코익산 또는 이의 약학적으로 허용 가능한 염; 또는 ii) 리포테이코익산을 포함하는 세포벽 추출물을 포함하는 칸디다(Candida) 항진균용 조성물을 제공한다.

본 발명에서 상기 추출에 이용되는 용매는 통상적으로 추출 용매로 사용될 수 있는 물질이라면 제한 없이 사용할 수 있으며, 바람직하게는 메탄올(methanol), 에탄올(ethanol), 프로판올(propanol), 이소프로판올(isopropanol), 부탄올(butanol) 및 이들의 조합으로 이루어진 군에서 선택된 어느 하나이상의 용매를 사용할 수 있고, 보다 바람직하게는 부탄올을 사용할 수 있다.
상기 칸디다 항진균용 조성물에서, 칸디다는 칸디다 알비칸스(Candida albicans), 칸디다 트로피칼리스(Candida tropicalis), 칸디다 수도트로피칼리스(Candida pseudotropicalis), 칸디다 크루세이(Candida krusei), 칸디다 글라브라타(Candida glabrata) 및 이들의 혼합을 포함하는 군에서 선택된 균류(fungi)일 수 있으나, 이에 제한되지 않는다.
본 발명에서 상기 조성물은 칸디다 진균의 표면 부착력을 감소시킬 수 있다. 구체적인 일실시예에서, 도 1 내지 도 3에 나타난 바와 같이, 리포테이코익산을 포함하는 조성물을 처리하지 않은 대조군에 비하여 상기 조성물을 처리한 경우, 칸디다 진균의 표면 부착률이 유의미하게 약화되었음을 확인하였다.
또한, 칸디다 진균에 의해 발생하는 칸디다증의 치료에 현재 많은 항진균제가 사용되고 있으나, 진균에 해당하는 칸디다 곰팡이균은 사람을 포함한 고등동물의 세포와 많은 부분이 유사하여 이러한 진균에 영향을 주는 항진균제가 숙주에도 영향을 줄 수 있고, 이로 인해 강한 항진균제는 숙주에게 독성을 나타낼 가능성이 있기 때문에 항진균 약제들의 광범위한 사용에는 한계가 있었다. 또한 최근 항진균제에 대한 내성을 획득하거나 내재성 내성을 지닌 진균(특히 칸디다 아우리스)의 증가로 항진균제의 내성이 진균 감염의 치료에 중요한 문제로 대두되었다(Protein J 2013, 32:15-26). 따라서 진균 감염의 치료에 효과적이면서 부작용과 내성이 적어 장기간 사용이 가능한 항진균제의 개발이 요구되었다.
또한, 본 발명은 상기 조성물을 포함하는 칸디다증 감염 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명에서 용어 "칸디다증(candidiasis)"이란, 칸디다로 인한 진균증을 의미하며, 일례로 칸디다 알비칸스(Candida albicans)로 인한 칸디다증의 경우, 최초로 아구창 환자에게서 분리되었고 여성의 경우 질염을 일으키고 유아의 기저귀 발진을 유발한다.
본 발명에서 용어 "예방"이란, 본 발명의 약학적 조성물의 투여로 칸디다증의 발병을 억제 또는 지연시키는 모든 행위를 의미하며, "치료"란, 본 발명의 약학적 조성물의 투여로 인해 이미 유발된 칸디다증의 증세가 호전되거나 이롭게 되는 모든 행위를 의미한다.
본 발명에서 용어 “칸디다 진균에 의해 유발되는 감염증”은 본 발명의 항진균용 조성물의 항진균효과로 억제될 수 있는 감염증일 수 있으며, 이는 기존의 소독제, 항생제로 치료하던 감염증과 질병 진단시 감염증이 유발될 수 있는 경우를 포함한다.
본 발명의 약학적 조성물은 통상의 방법에 따라 적절한 담체, 부형제 또는 희석제를 추가적으로 포함할 수 있다. 화합물을 함유하는 약학적 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 약학적 조성물은 통상의 방법에 따라 산제, 환제, 과립제, 캡슐제, 현탁액, 내용액제, 유제, 시럽제, 멸균된 수용액, 비수용액제, 현탁액, 동결 건조제 및 좌제로 이루어진 군으로부터 선택되는 어느 하나의 제형으로 제제화될 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 제조될 수 있다. 당해 기술 분야에 알려진 적합한 제제는 문헌(Remington's Pharmaceutical Science, Mack Publishing Company, Easton PA)에 개시되어 있는 것을 사용하는 것이 바람직하다.
본 발명의 약학적 조성물은 약제학적으로 유효한 양으로 투여한다. 본 발명의 용어 "약제학적으로 유효한 양"이란, 칸디다 진균에 대한 항진균 효과를 나타낼 수 있을 정도의 충분한 양과 부작용이나 심각한 또는 과도한 면역반응을 일으키지 않을 정도의 양을 의미하며, 유효 용량 수준은 치료될 장애, 장애의 중증도, 특정 화합물의 활성, 투여 경로, 치료 지속 기간, 리포테이코익산을 포함하는 조성물과 조합되거나 또는 동시에 사용되는 약물, 대상체의 연령, 체중, 성별, 식습관, 일반적인 건강 상태, 및 의약 업계 및 의학 분야에 공지된 인자를 비롯한 다양한 인자들에 따라 달라질 것이다. "치료상 유효량" 결정시 고려되는 다양한 일반적인 사항들은 당업자에게 공지되어 있으며, 예를 들어 문헌 [Gilman et al., eds., Goodman And Gilman's: The Pharmacological Bases of Therapeutics, 8th ed., Pergamon Press, 1990] 및 [Remington's Pharmaceutical Sciences, 17th ed., Mack Publishing Co., Easton, Pa., 1990]에 기재되어 있다.
본 발명에서 용어 "투여"란, 어떠한 적절한 방법으로 환자에게 소정의 물질을 도입하는 것을 의미하며, 인체 또는 수의용으로 제형화되어 다양한 경로로 투여된다. 본 발명의 조성물은 비경구 경로, 예컨대 혈관내, 정맥내, 동맥내, 근육내 또는 피하 등의 경로로 투여될 수 있고, 경구, 비강, 직장, 경피 또는 에어로졸을 통한 흡입 경로로 투여될 수도 있으며, 볼루스(bolus)로 투여하거나 또는 서서히 주입할 수도 있다.
본 발명의 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 약학적 조성물에서의 바람직한 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다. 그러나 바람직한 효과를 위해서, 1일 0.01 mg/kg 내지 10 g/kg으로, 바람직하게는 1 mg/kg 내지 1 g/kg으로 투여하는 것이 좋다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수 있다. 따라서, 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 약학적 조성물은 리포테이코익산을 포함하는 조성물과 함께 칸디다증의 예방 및 치료 효과를 갖는 공지의 유효성분을 1종 이상 포함할 수 있다.
본 발명의 약학적 조성물은 전문의약품, 위생 용품 등에 이용되어 칸디다 진균에 대한 항진균 기능을 수행할 수 있다.
위생용품으로는 가글, 리스테린 등 구강 세정 용품, 치약, 구강 위생용품, 비누, 핸드솝, 질 세정제 첨가제, 전신세정제 등의 피부 세정제 조성물, 식기나 싱크대, 도마 등을 세정하기 위한 주방용품 세정제 조성물, 에센스, 크림 등의 화장료 조성물과 같은 위생용품에 유효성분으로 첨가하면, 각각의 위생용품 본래의 기능 외에 칸디다 진균에 대한 항진균의 기능을 수행할 수 있다.
본 발명에서 상기 약학적 조성물의 효과가 있는 상기 칸디다증은 외음부염, 질염, 식도염, 손발톱 주위 염, 구인두염, 소화관 칸디다증, 점막 칸디다증, 피부 칸디다증, 칸디다 진균혈증, 칸디다 내안구염, 칸디다 복막염, 칸디다 골관절염, 칸디다 척추염 및 간비장 칸디다증으로 이루어진 군에서 선택되는 어느 1종 이상일 수 있으며, 이에 제한되지 않는다.
예를 들어, "질 칸디다증"은 칸디다 알비칸스(Candida albicans), 칸디다 트로피칼리스(Candida tropicalis), 칸디다 수도트로피칼리스(Candida pseudotropicalis), 칸디다 크루세이(Candida krusei), 칸디다 글라브라타(Candida glabrata) 및 이들의 혼합을 포함하는 군에서 선택된 칸디다 속의 균류에 의해 발생한 구체적 종류의 질 진균증을 의미한다. 질 칸디다증은 또한 칸디다성 질염(candidal vaginitis)으로 불린다.
본 발명의 약학적 조성물은 칸디다 진균의 표면 단백질인 Hsp70과 상호작용함으로써, 부착력을 감소시켜 생체막 형성을 저해하고 진균 감염을 억제한다(도 3 및 도 4 참조).
또한, 본 발명은 칸디다증의 발병이 예상되거나 또는 칸디다증이 발병된 개체에게 상기 약학적 조성물을 투여하는 단계를 포함하는, 칸디다증을 예방 또는 치료하는 방법을 제공한다.
본 발명에서 용어 "개체"란, 칸디다증이 발병할 수 있는 살아있는 유기체를 의미하는데, 바람직하게는 칸디다증의 증상이 나타날 수 있는 고등 척추동물이 될 수 있고, 보다 바람직하게는 칸디다증이 발병될 수 있는 소, 돼지, 양, 염소, 개 등의 가축 및 이로부터 감염될 수 있는 사람이 될 수 있다.
본 발명의 약학적 조성물을 상기 개체에 투여하면, 칸디다증을 유발시키는 균주에 대하여 개체의 면역성을 증진시켜서, 칸디다증을 예방 또는 치료하는 효과를 나타낼 수 있다. 이러한 약학적 조성물의 투여 대상은 일차적으로는 소, 돼지, 양, 염소, 개 등의 가축이 될 수 있으나, 이에 제한되지 않고, 상기 가축에 의하여 칸디다증이 전염될 수 있는 인간의 예방 또는 치료에도 사용될 수 있다.
또한, 본 발명은 상기 조성물을 포함하는 코팅제를 제공한다.
본 발명에서 용어 "항진균 코팅제"는 진균에 의해 형성된 표면 부착력 및 생물막 형성을 방지하기 위한 코팅 제제이며, 미생물의 부착 이후 형성된 막을 제거하기 위해서는 미생물 저해제에 대한 저항성이 강한 상태이므로 고농도의 살균제나 항생제를 투여하게 되어, 주변에 심각한 손상을 줄 수 있기 때문에 표면 부착력 및 생물막 형성을 사전에 예방하는 데에 이용될 수 있다.
본 발명에서 생물막이 형성될 수 있는 위치로는, 피부, 구강, 소화관 및 질을 포함하여 점막과 피부의 습한 부위일 수 있다. 예시로서, 구내염을 일으키는 부위인 혀, 볼 안쪽, 입천장을 비롯한 구강, 구강내막, 목구멍과 식도 및 성기부위로 외음부 및 질, 음경이 포함될 수 있으며, 습한 피부나 손톱 또는 발톱, 서혜부, 겨드랑이, 손가락 사이와 발가락 사이, 유방 아래 피부 주름 부위, 복부의 피부 주름부위 또한 포함될 수 있으며, 이에 제한되지 않는다.
또한, 봉합, 적출부, 동정맥부, 공막돌융술 (scleral buckle) 부위, 콘택트 렌즈, IUD, 기관 튜브, 히크만 카테터 (Hickman catheter), 중심 정맥 카테터, 인공 심장 판막, 판막 그래프트, 정형외과 기기, 음경 보형물 등을 예로 들 수 있으며, 기타 다른 예들이 Costerton J et al 및 Costerton J and Steward (2001 Battling Biofilms, Scientific American pp 75-81)에 의한 논문에 기재되어 있고, 상기 논문은 원용에 의해 본 명세서에 포함된다. 기타, 생물막이 형성될 수 있는 위치로는, 치주농주 (gum disease) 및 강 (cavity)으로 진행될 수 있는 충치, 눈 감염으로 진행될 수 있는 콘택트 렌즈, 만성 감염이 될 수 있는 귀 및 폐렴이 될 수 있는 폐를 포함한다.
따라서, 상기 코팅제는 의료기기, 의료용 재료, 의료용 이식물, 건축 내장재, 생활용품, 산업용품, 전자제품, 의류, 인공장기에 이용될 수 있으나, 이에 제한되지 않고 칸디다 진균에 의한 오염 또는 감염될 가능성이 있는 곳이라면 사용가능하다.
이 중, 의료기기는 수술, 치료, 예방 등을 수행하기 위한 의료적 행위를 돕는 모든 기기를 의미하며, 예시적으로 혈관 스텐트, 인공 혈관, 인공 판막, 채혈기, 혈액 투석 장비 및 기기 및 전술한 카테터나 기기 등을 포함할 수 있으나, 이에 제한되지 않고, 칸디다 진균에 의한 오염 또는 감염이 될 가능성이 있는 기기를 의미한다.
또한, 본 발명은 상기 조성물을 포함하는 건강기능식품을 제공한다.
본 발명에서 "건강기능식품"은 기능성 식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
본 발명의 건강기능식품은 칸디다증 감염 예방 또는 치료를 목적으로 이용될 수 있다. 본 발명의 리포테이코익산을 포함하는 조성물을 첨가물로 사용할 경우, 상기 리포테이코익산을 포함하는 조성물을 그대로 첨가하거나 다른 식품 또는 식품성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 상기물질을 첨가할 수 있는 식품의예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
본 발명에서 유효 성분의 혼합양은 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품 또는 음료의 제조시에는 본 발명의 리포테이코익산을 포함하는 조성물은 원료에 대하여 1~20 중량%, 바람직하게는 5~10 중량%의 양으로 첨가된다. 그러나, 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다. 상기 식품의 종류에는 특별한 제한은 없다. 발명의 음료 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 ㎖당 일반적으로 약 0.01~0.04 g, 바람직하게는 약 0.02~0.03 g이다. 그 외, 본 발명의 조성물은 여러가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 그밖에 본 발명의 조성물은 천연 과일쥬스, 과일쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물 100 중량부 당 0.01~0.1 중량부의 범위에서 선택되는 것이 일반적이다.
또한, 본 발명은, 상기 조성물을 포함하는 가축사료 첨가제를 제공한다.
본 발명에서 사료 첨가제로서 이용될 경우, 상기 조성물은 20 내지 90% 고농축액이거나 분말 또는 과립 형태로 제조될 수 있다. 상기 사료첨가제는 구연산, 후말산, 아디픽산, 젖산, 사과산등의 유기산이나 인산 나트륨, 인산 칼륨, 산성 피로인산염, 폴리인산염(중합인산염) 등의 인산염이나, 폴리페놀, 카테킨, 알파-토코페롤, 로즈마리 추출물, 비타민 C, 녹차 추출물, 감초 추출물, 키토산, 탄닌산, 피틴산 등의 천연 항산화제 중 어느 하나 또는 하나 이상을 추가로 포함할 수 있다. 사료로서 이용될 경우, 상기 조성물은 통상의 사료 형태로 제제화 될 수 있으며, 통상의 사료성분을 함께 포함할 수 있다.
본 발명에서 상기 사료첨가제 및 사료는 곡물, 예를 들면 분쇄 또는 파쇄된 밀, 귀리, 보리, 옥수수 및 쌀; 식물성 단백질 사료, 예를 들면 평지, 콩, 및 해바라기를 주성분으로 하는 사료; 동물성 단백질 사료, 예를 들면 혈분, 육분, 골분 및 생선분; 당분 및 유제품, 예를 들면 각종 분유 및 유장 분말로 이루어지는 건조성분 등을 더 포함할 수 있으며, 이외에도 영양보충제, 소화 및 흡수향상제, 성장촉진제 등을 더 포함할 수 있다.
본 발명에서 상기 사료첨가제는 동물에게 단독으로 투여하거나 식용 담체 중에서 다른 사료첨가제와 조합하여 투여할 수도 있다. 또한, 상기 사료첨가제는 탑드레싱으로서 또는 이들을 동물사료에 직접 혼합하거나 또는 사료와 별도의 경구제형으로 용이하게 동물에게 투여할 수 있다. 상기 사료첨가제를 동물사료와 별도로 투여할 경우, 당해 기술분야에 잘 알려진 바와 같이 약제학적으로 허용가능한 식용 담체와 조합하여, 즉시 방출 또는 서방성 제형으로 제조할 수 있다. 이러한 식용 담체는 고체 또는 액체, 예를 들어 옥수수전분, 락토오스, 수크로오스, 콩플레이크, 땅콩유, 올리브유, 참깨유 및 프로필렌글리콜일 수 있다. 고체 담체가 사용될 경우, 사료첨가제는 정제, 캡슐제, 산제, 트로키제 또는 함당정제 또는 미분산성 형태의 탑드레싱일 수 있다. 액체 담체가 사용될 경우, 사료첨가제는 젤라틴 연질 캡슐제, 또는 시럽제나 현탁액, 에멀젼제, 또는 용액제의 제형일 수 있다.
또한, 상기 사료첨가제 및 사료는 보조제, 예를 들어 보존제, 안정화제, 습윤제 또는 유화제, 용액촉진제 등을 함유할 수 있다. 상기 사료첨가제는 침주, 분무 또는 혼합하여 동물의 사료에 첨가하여 이용될 수 있다.
본 발명에서 사료 또는 사료첨가제는 포유류, 가금 및 어류를 포함하는 다수의 동물식이에 적용할 수 있다.
본 발명에서 동물은 포유류를 포함하고 포유류로서 돼지, 소, 양, 염소, 실험용 설치동물, 및 실험용 설치 동물뿐만 아니라, 애완동물(예: 개, 고양이) 등에게 사용할 수 있으며, 상기 가금류로서 닭, 칠면조, 오리, 거위, 꿩, 및 메추라기 등에도 사용할 수 있고, 상기 어류로서 송어 등에 이용될 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 조성물을 포함하여 반려동물 간식, 구강용품, 위생용품 및 동물용 건강기능 식품으로 사용할 수 있으며, 전술한 예에 제한없이 반려동물 용품이라면 사용할 수 있다.
또한, 본 발명은 상기 조성물을 포함하는 식품 세정제를 제공한다.
본 발명에 따른 리포테이코익산을 포함하는 항진균용 조성물은 칸디다 진균의 부착력을 감소시켜 생체막 형성을 저해하고 진균 감염을 억제하는 효과를 나타내며, 생체친화적인 물질의 사용을 통해 인체 내에서 안전성이 높고 부작용을 줄일 수 있는 항진균제로서 칸디다 진균에 의한 감염 질병인 칸디다증의 효과적인 치료뿐만 아니라, 인체 내에 삽입된 의료기구를 통한 칸디다 감염 또한 제어할 수 있다.

이하, 실시예를 통하여 본 발명의 구성 및 효과를 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것일 뿐, 본 발명의 범위가 이들 실시예에 의해 한정되는 것은 아니다.
실시예 1: 락토바실러스 루테리(
Lactobacillus reuteri
) 로부터 리포테이코익산(lipoteichoic acid)을 분리 및 정제
L. reuteri로부터 리포테이코익산을 정제하기 위해 L. reuteri ATCC 23272 균주를 사용했다. L. reuteri를 de man, rogosa and sharpe broth (MRS broth) 에 접종하여 37℃에서 24시간 동안 배양한 뒤, 원심분리를 통해 균을 수확하였다. 수확한 균을 초음파 분쇄기로 균을 균질화시킨 뒤, 균질액을 유기용매인 부탄올에 녹이고 원심분리를 통해 지질층을 분리하였다. 이후 분리한 추출물을 소수성 상호작용 크로마토그래피와 음이온 교환 크로마토그래피를 순차적으로 사용하여 정제를 진행한 뒤, 정제물을 동결건조하여 최종 건조물을 얻었다.

LTA (LreLTA)에 의한 칸디다 알비칸스(
Candida albicans
)의 표면 부착력 감소 효과
LreLTA가 C. albicans 표면 부착력에 미치는 영향을 확인하기 위해 야생형 균주인 SC5314 균주를 사용했다. C. albicans를 Yeast Extract-Peptone-Dextrose (YPD) 배지에서 배양한 뒤, C. albicans를 1 x 104 CFU/ml로 준비하여 LreLTA를 3, 10, 30 μg/ml 씩 농도별로 처리하였다. 이후 37℃에서 24시간 동안 배양한 뒤 생물막 표면 부착 유지 정도를 0.1% 크리스탈바이올렛 염색을 통해 비교하였다. 실험 결과, 도 1A에 나타난 바와 같이, LreLTA가 C. albicans의 표면 부착력을 약화시키는 것을 확인하였다(도 1A).
추가적으로 야생형 균주 이외에 임상분리균주에서도 LreLTA가 같은 효과를 보이는지 확인하기 위해 C. albicans KCOM 3253, KCOM 3258과 NCCP 32701, NCCP 32702, NCCP 32703 균주를 YPD 배지에서 배양한 뒤, C. albicans를 1 x 107 CFU/ml로 준비하여 LreLTA를 30 μg/ml 단일 농도로 처리하였다. 이후 37℃에서 24시간 동안 배양한 뒤 생물막 표면부착 유지 정도를 0.1% 크리스탈바이올렛 염색을 통해 비교하였다. 실험 결과, LreLTA가 C. albicans의 임상 분리 균주들의 표면 부착력도 약화시키는 것을 확인하였다(도 1B~F).
실시예 3: LreLTA가 이미 부착되어 군집을 형성한
C. albicans
에 미치는 효과
LreLTA가 C. albicans의 부착력을 약화시키는 효능이 기형성된 C. albicans의 생물막에서도 동일하게 나타나는지 확인하기 위해, C. albicans SC5314 균주를 YPD 배지에서 배양한 뒤, 1 x 104 CFU/ml로 준비하여 37℃에서 24시간 동안 생물막을 형성시켰다. 이후 기형성된 생물막에 LreLTA를 3, 10, 30 μg/ml씩 농도별로 처리하고 24시간 동안 37℃에서 배양한 뒤 생물막 표면 부착 유지 정도를 0.1% 크리스탈바이올렛 염색을 통해 비교하였다. 실험 결과, LreLTA가 C. albicans의 기형성된 생물막에서도 표면 부착력의 감소 효과를 보이는 것을 확인하였다(도 2A).
추가적으로 임상분리균주에서도 LreLTA가 같은 효과를 보이는지 확인하기 위해 C. albicans KCOM 3253, KCOM 3258과 NCCP 32701, NCCP 32702, NCCP 32703 균주를 YPD 배지에서 배양한 뒤, 1 x 107 CFU/ml로 준비하여 37℃에서 24시간 동안 생물막을 형성시켰다. 이후 기형성된 생물막에 LreLTA를 30 μg/ml 단일 농도로 처리하고 24시간 동안 37℃에서 배양한 뒤 생물막 표면 부착 유지 정도를 0.1% 크리스탈바이올렛 염색을 통해 비교하였다. 실험 결과, LreLTA가 임상분리균주에서도 같은 효과를 보이는 것을 확인하였다(도 2B~F).
실시예 4: 여러 유산균과 그람양성균으로부터 분리 정제한 LTA에 의한 칸디다 알비칸스(
C. albicans
)의 표면 부착력 감소 효과
다른 여러 유산균과 그람양성균으로부터 분리 정제한 LTA가 C. albicans 표면 부착력에 미치는 영향을 확인하기 위해 야생형 균주인 SC5314 균주, 5 종류의 유산균과 3 종류의 그람양성균으로부터 분리 정제한 LTA를 사용하였다. C. albicans SC5314 균주를 YPD 배지에서 배양한 뒤, 세포 수를 1 x 104 CFU/ml로 준비하여 8 종류의 LTA를 30 μg/ml 의 단일 농도로 처리하였다. 이후 37℃에서 24시간 동안 배양한 뒤 생물막 표면 부착 유지 정도를 0.1% 크리스탈바이올렛 염색을 통해 비교하였다.
실험 결과, 도 3에 나타난 바와 같이, LaLTA를 제외한 7 종류의 LTA 모두 C. albicans의 표면 부착력을 약화시키는 것을 확인하였으며, 인체 유익균인 유산균으로부터 분리한 LTA 중 특히 LreLTA가 C. albicans의 표면 부착력을 효과적으로 약화시켰다(LaLTA : 락토바실러스 애시도필러스 리포테이코익산; LrLTA : 락토바실러스 람노서스GG 리포테이코익산;LpLTA : 락토바실러스 플란타럼; LcLTA : 락토바실러스 카제이 리포테이코익산; LreLTA : 락토바실러스 루테리 리포테이코익산; EfLTA : 엔테로코커스 페칼리스 리포테이코익산; SmLTA : 스트렙토코커스 뮤탄스 리포테이코익산; BsLTA : 바실러스 서브틸리스 리포테이코익산).
실시예 5: 열충격단백질70 억제제 VER-155008을 통한 LreLTA의 효능 검증
기존 연구에 따르면, Hsp70은 C. albicans의 전반적인 독성인자 조절과 약물 저항성 조절의 역할을 담당하고 있다고 알려져 있어 본 발명자들은 Hsp70 억제제인 VER-155008을 활용해 실험을 진행하였다.
VER-155008이 LreLTA의 C. albicans 표면 부착력 감소 효과에 미치는 영향을 확인하기 위해 C. albicans SC5314 균주를 YPD 배지에서 배양한 뒤, 1 x 104 CFU/ml로 준비하여 LreLTA를 30 μg/ml 단일 농도로, VER-155008을 1, 10, 50, 100 μM 농도별로 동시처리하였다. 이후 24시간 동안 배양한 뒤 생물막 표면 부착 유지 정도를 0.1% 크리스탈바이올렛 염색을 통해 비교하였다.
실험 결과, 도 4에 나타난 바와 같이, VER-155008을 고농도로 처리할수록 LreLTA의 표면 부착력 감소 효능이 줄어드는 것을 확인할 수 있었으며 결과적으로 리포테이코익산이 칸디다 알비칸스의 Hsp70과 상호작용하여 부착력과 응집력 약화 효과를 보이는 것을 확인하였다.
실시예 6:
ex vivo
모델을 통한 LreLTA의
C.albicans
부착력 감소 효과 검증
LreLTA의 C. albicans 표면 부착력 약화 효과가 생체조직에서도 동일한 현상을 보이는지, 콜로니형성단위(CFU)를 확인하기 위해 C. albicans SC5314 균주를 사용하였다. ex vivo 모델을 위한 실험동물로 C57BL/6 계통의 야생성 마우스를 두열바이오텍으로부터 구입하여 사용하였고 동물실험은 실험동물 연구 윤리규정을 철저히 준수하여 수행하였다. 마우스를 안락사 시킨 후, 혀를 적출하였다. 적출한 혀는 YPD 배지에서 배양한 C. albicans SC5314 균주 1 x 107 CFU/ml 와 함께 LreLTA를 30 μg/ml 단일 농도로 동시처리하여 1시간 동안 37℃에서 배양한 뒤, 새로운 배지에 옮겨 담은 후 24시간 동안 37℃에서 배양하였다. 이후 균과 함께 배양한 혀 조직을 균질기(homogenizer)를 이용하여 균질화한 뒤, Agar 배지에 접종하여 콜로니형성단위(Colony Forming Unit) 정도를 비교하였다.
실험 결과, 도 5에 나타난 바와 같이, LreLTA를 처리한 혀 조직은 대조군인 주사용수를 처리한 혀 조직에 비해 C. albicans의 CFU 값이 통계학적으로 유의성 있게 감소하였다. 이를 통해 LreLTA가 생체조직에서도 C. albicans의 부착력을 약화시킬 수 있음을 확인하였다.
실시예 7:
in vivo
모델을 통한 LreLTA의
C. albicans
부착력 감소 효과 검증
LreLTA의 C. albicans 표면 부착력 약화 효과가 동물 질환 모델에서도 동일한 현상을 보이는지 확인하기 위해 마우스 구강 칸디다증 모델을 사용하였다. in vivo 모델을 위한 실험동물로 C57BL/6 계통의 야생형 마우스를 두열바이오텍으로부터 구입하여 사용하였고 동물실험은 실험동물 연구 윤리규정을 철저히 준수하여 수행하였다. 마우스 구강의 설하에 YPD 배지에서 배양한 C. albicans SC5314 균주 1 x 107 CFU/ml 가 포함된 면봉을 삽입하여 감염을 진행시키고 LreLTA를 50 μg/ml 단일 농도로 동시처리하였다. 이후 매일 주사용수(대조군) 또는 LreLTA(실험군)을 마우스의 구강에 투여하였다. 5일 동안 칸디다증을 발달시킨 후, 마우스를 안락사 시키고 혀 조직을 적출하여 균질기(homogenizer)를 이용해 균질화한 뒤, Agar 배지에 접종하여 콜로니형성단위(Colony Forming Unit) 정도를 비교하였다.
실험 결과, LreLTA를 처리한 마우스의 혀 조직은 대조군인 주사용수를 처리한 혀 조직에 비해 C. albicans의 CFU 값이 통계학적으로 유의성 있게 감소하였으며, 이를 통해 LreLTA가 마우스 구강 칸디다증 질환 모델에서 효과적으로 C. albicans의 감염을 저해시킬 수 있음을 확인하였다(도 6A).
추가적으로 마우스 칸디다 질염 모델에서도 LreLTA가 같은 효과를 보이는지 확인하기 위해 C. albicans SC5314 균주를 YPD 배지에서 배양한 뒤, 세포 수를 1 x 107 CFU/ml로 준비하여 마우스의 질 내부에 주입한 뒤, LreLTA를 50 μg/ml 단일 농도로 동시처리하였다. 이후 매일 주사용수(대조군) 또는 LreLTA(실험군)을 마우스의 질 내에 주입하였다. 7일 동안 칸디다 질염을 발달시킨 후, 마우스를 안락사 시키고 질 조직을 적출하여 균질기(homogenizer)를 이용해 균질화한 뒤, Agar 배지에 접종하여 콜로니형성단위(Colony Forming Unit) 정도를 비교하였다.
실험 결과, LreLTA를 처리한 마우스의 질 조직은 대조군인 주사용수를 처리한 질 조직에 비해 C. albicans의 CFU 값이 통계학적으로 유의성 있게 감소하였으며, 이를 통해 LreLTA가 마우스 칸디다 질염 질환 모델에서 C. albicans의 부착력을 약화시키고 효과적으로 감염을 저해시킬 수 있음을 확인하였다(도 6B).
Claims (13)
- i) 리포테이코익산 또는 이의 약학적으로 허용 가능한 염; 또는ii) 리포테이코익산을 포함하는 세포벽 추출물을 포함하는 칸디다(Candida) 항진균용 조성물.
- 제1항에 있어서,상기 리포테이코익산 또는 리포테이코익산을 포함하는 세포벽 추출물은 그람양성균으로부터 유래한, 조성물.
- 제1항에 있어서,상기 리포테이코익산 또는 리포테이코익산을 포함하는 세포벽 추출물은 락토바실러스 루테리(Lactobacillus reuteri), 락토바실러스 애시도필러스(Lactobacillus acidophilus), 락토바실러스 람노서스GG(Lactobacillus rhamnosus GG), 락토바실러스 플란타럼(Lactobacillus plantarum), 락토바실러스 카제이(Lactobacillus casei), 락토바실러스 파라카제이(Lactobacillus paracasei), 락토바실러스 퍼멘텀(Lactobacillus fermentum), 락토바실러스 불가리쿠스(Lactobacillus delbrueckii subsp. bulgaricus), 락토바실러스 헬베티쿠스 (Lactobacillus helveticus), 락토바실러스 퍼멘툼 (Lactobacillus fermentum), 락토바실러스 살리바리우스(Lactobacillus salilvarius), 락토코커스 락티스(Lactococcus lactis), 엔테로코커스 페칼리스(Enterococcus faecalis), 엔테로코커스 패시움(Enterococcus faecium), 스트렙토코커스 써모필러스(Streptococcus thermophilus), 비피도박테리움 비피덤(Bifidobacterium bifidum), 비피도박테리움 브리브(Bifidobacterium breve), 비피도박테리움 롱굼(Bifidobacterium longum), 비피도박테리움 아니말리스 락티스(Bifidobacterium animalis subsp. Lactis) 및 바실러스 서브틸리스(Bacillus subtilis)로 이루어진 군으로부터 선택되는 어느 1종으로부터 유래한, 조성물.
- 제1항에 있어서,상기 세포벽 추출물은 물, C1 내지 C4의 저급 알코올 또는 이들의 혼합물을 용매로 하여 추출되는 것을 특징으로 하는, 조성물.
- 제1항에 있어서,상기 칸디다는 칸디다 알비칸스(Candida albicans), 칸디다 트로피칼리스(Candida tropicalis), 칸디다 수도트로피칼리스(Candida pseudotropicalis), 칸디다 크루세이(Candida krusei), 칸디다 글라브라타(Candida glabrata) 및 이들의 혼합을 포함하는 군에서 선택된 균류(fungi)인, 조성물.
- 제1항에 있어서,상기 조성물은 칸디다 진균의 표면 부착력을 감소시키는, 조성물.
- 제1항 내지 제6항 중 어느 한 항의 조성물을 포함하는 칸디다증 감염 예방 또는 치료용 약학적 조성물.
- 제7항에 있어서,상기 칸디다증은 외음부염, 질염, 식도염, 손발톱 주위 염, 구인두염, 소화관 칸디다증, 점막 칸디다증, 피부 칸디다증, 칸디다 진균혈증, 칸디다 내안구염, 칸디다 복막염, 칸디다 골관절염, 칸디다 척추염 및 간비장 칸디다증으로 이루어진 군에서 선택되는 어느 1종 이상인, 칸디다증 감염 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제6항 중 어느 한 항의 조성물을 포함하는 코팅제.
- 제9항에 있어서,상기 코팅제는 의료기기, 의료용 재료, 의료용 이식물, 건축 내장재, 생활용품, 산업용품, 전자제품, 의류, 인공장기에 이용되는, 코팅제.
- 제1항 내지 제6항 중 어느 한 항의 조성물을 포함하는 건강기능식품.
- 제1항 내지 제6항 중 어느 한 항의 조성물을 포함하는 가축사료 첨가제.
- 제1항 내지 제6항 중 어느 한 항의 조성물을 포함하는 식품 세정제.
