Abstract
本発明の課題は、凍結保存による細胞障害を低減する凍結保存用の組成物を提供することや、凍結保存による細胞障害を低減する凍結保存方法を提供することである。両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子を含む、細胞若しくは生体組織の凍結保存用組成物を作製する。
Description
本発明は、凍結保存用の組成物や、凍結保存方法や、凍結した細胞又は生体組織に関する。
再生医療とは、細胞等を用いて疾患や外傷等を治療する治療法である。再生医療は、近年、心筋梗塞、慢性動脈閉塞症、白血病等の様々な疾患や、脊髄損傷、外傷性軟骨欠損症等の外傷の治療法として採用されている。従来、再生医療に用いる細胞等は、輸送性、長期保存性、利便性等の観点から、凍結保存したものが用いられている。しかしながら、細胞を凍結保存すると、細胞生存率の低下、細胞の機能障害(例えば、幹細胞の場合は幹細胞の割合の低下)、増殖阻害等の現象(以下、これらの現象を総じて「細胞障害」とも称す。)が生じやすい。これは、細胞が凍結された際に細胞内に含まれる水分が結晶化して結晶により細胞壁や細胞小器官等が損傷したり、細胞が解凍された際に細胞内の水分が一部失われて細胞の形状が変化したりすることが要因として考えられる。
細胞を凍結して保存する細胞凍結保存法の一つとして、特許文献1に記載されているように、ジメチルスルホキシド(DMSO)などの低分子化合物を含む緩衝溶液中で細胞を凍結する手法が知られている。
しかし、DMSOを用いた手法は、細胞解凍後の細胞生存率が低く、改善の余地がある。本発明は、このような状況下でなされたものである。
本発明の一実施形態が解決しようとする課題は、凍結保存による細胞障害を低減する細胞又は生体組織の凍結保存用の組成物を提供することである。
本発明の他の一実施形態が解決しようとする課題は、凍結保存による細胞障害を低減する細胞又は生体組織の凍結保存方法を提供することである。
本発明の一実施形態が解決しようとする課題は、凍結保存による細胞障害を低減する細胞又は生体組織の凍結保存用の組成物を提供することである。
本発明の他の一実施形態が解決しようとする課題は、凍結保存による細胞障害を低減する細胞又は生体組織の凍結保存方法を提供することである。
前記課題を解決するための具体的手段には、下記の態様が含まれる。
<1>両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子を含む、細胞若しくは生体組織の凍結保存用組成物。
<2>前記ナノ粒子が、カチオン性ナノ粒子である、上記<1>に記載の凍結保存用組成物。
<3>前記両親媒性分子が、Gemini型界面活性剤であることを特徴とする、上記<1>又は<2>に記載の凍結保存用組成物。
<4>前記ナノ粒子のゼータ電位が、10mV以上50mV以下である、上記<1>又は<2>に記載の凍結保存用組成物。
<5>前記ナノ粒子の体積平均粒子径が、30nm以上300nm以下である、上記<1>又は<2>に記載の凍結保存用組成物。

以下、本発明を実施するための形態について説明する。これらの説明及び実施例は本発明を例示するものであり、本発明の範囲を制限するものではない。

<細胞凍結保存用組成物>
本実施形態に係る細胞若しくは生体組織の凍結保存用組成物(以下、「本件凍結保存用組成物」ともいう)は、両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子(以下、「本件ナノ粒子」ともいう)を含む。
本件凍結保存用組成物を細胞若しくは生体組織と混合すると、本件ナノ粒子が細胞若しくは生体組織を構成する細胞内に導入される。つまり、細胞内には、本件ナノ粒子が導入される。
本件ナノ粒子が細胞内に導入された状態で凍結された細胞若しくは生体組織は、本件ナノ粒子が細胞に加わる外力や温度変化に対してクッションの機能(緩衝機能)を示すことにより、細胞内に含まれる水分の結晶化及び結晶成長が抑えられ、かつ、細胞壁及び細胞小器官等が損傷されにくくなる。また、本件ナノ粒子が細胞内に導入された状態で凍結された細胞は、解凍された際にも、本件ナノ粒子の介在により細胞の形状が維持されやすくなる。その結果、凍結保存による障害が低減されると考えられる。
本実施形態に係る細胞若しくは生体組織の凍結保存用組成物(以下、「本件凍結保存用組成物」ともいう)は、両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子(以下、「本件ナノ粒子」ともいう)を含む。
本件凍結保存用組成物を細胞若しくは生体組織と混合すると、本件ナノ粒子が細胞若しくは生体組織を構成する細胞内に導入される。つまり、細胞内には、本件ナノ粒子が導入される。
本件ナノ粒子が細胞内に導入された状態で凍結された細胞若しくは生体組織は、本件ナノ粒子が細胞に加わる外力や温度変化に対してクッションの機能(緩衝機能)を示すことにより、細胞内に含まれる水分の結晶化及び結晶成長が抑えられ、かつ、細胞壁及び細胞小器官等が損傷されにくくなる。また、本件ナノ粒子が細胞内に導入された状態で凍結された細胞は、解凍された際にも、本件ナノ粒子の介在により細胞の形状が維持されやすくなる。その結果、凍結保存による障害が低減されると考えられる。
なお、従来から、リポソーム及びミセル等のナノ粒子は、ドラッグデリバリーシステムなど薬理成分を対象部位に導入するための技術として用いられている。これに対して、本開示に係る発明は、本件ナノ粒子自体の細胞に対する緩衝機能に着目したものであり、本件ナノ粒子の持つ物理的性質が、細胞の凍結保存によって引き起こされる細胞障害の低減に有効に作用するという知見に基づくものである。
[ナノ粒子]
本件凍結保存用組成物は、両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子を含む。本件凍結保存用組成物は、単層のナノ粒子だけ含んでいても、二層のナノ粒子だけ含んでいてても、単層のナノ粒子と二層のナノ粒子を両方含んでいてもよい。
本件凍結保存用組成物は、両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子を含む。本件凍結保存用組成物は、単層のナノ粒子だけ含んでいても、二層のナノ粒子だけ含んでいてても、単層のナノ粒子と二層のナノ粒子を両方含んでいてもよい。
上記単層のナノ粒子とは、両親媒性分子が単層を形成するように集まったナノ粒子を意味する。単層のナノ粒子としては、具体的にはミセルを挙げることができる。また、上記二層のナノ粒子とは、両親媒性分子が二層となり形成された二重膜を含むナノ粒子を意味する。上記二層のナノ粒子としては、具体的には(1)脂質を構成成分とし、その脂質の二重膜を含む小胞であるリポソームや、(2)脂質以外の界面活性剤を構成成分とし、その界面活性剤の二重膜を含む小胞を挙げることができる。上記二重膜を含む小胞は、二重膜を1つ有している小型一枚膜リポソーム(SUV)や大型一枚膜リポソーム(LUV)であっても、二重膜を2以上有している多重膜リポソーム(MVLs)であってもよい。
また、本件ナノ粒子にミセルを含む場合は、そのミセルは親水基が外側、疎水基が内側に集合したミセルであることが好ましい。
本件ナノ粒子の体積平均粒子径は、10nm以上300nm以下を挙げることができ、20nm以上200nm以下であっても、30nm以上150nm以下であってもよい。
本件ナノ粒子の体積平均粒子径は、小さいほど細胞の隅々に分布しやすく、細胞を凍結した際に細胞の形状がより維持されやすい。
本件ナノ粒子の体積平均粒子径が300nm以下であると、本件ナノ粒子が細胞内により効率的に導入されやすい。
本件ナノ粒子の体積平均粒子径は、小さいほど細胞の隅々に分布しやすく、細胞を凍結した際に細胞の形状がより維持されやすい。
本件ナノ粒子の体積平均粒子径が300nm以下であると、本件ナノ粒子が細胞内により効率的に導入されやすい。
本件ナノ粒子の体積平均粒子径を制御する手法は、特に制限されないが、例えば、本件ナノ粒子の製造の際に用いるフィルターの孔径で調整するサイジングなどの手法等が挙げられる。具体的には、細孔を有するフィルターに本件ナノ粒子を加圧して通過させ、物理的なせん断力により粒子径を均一化するエクストリュージョン処理を挙げることができる。
本件ナノ粒子は、カチオン性ナノ粒子であっても、中性ナノ粒子であっても、アニオン性ナノ粒子であってもよいが、カチオン性ナノ粒子であることがより好ましい。通常、細胞内は細胞外に対して負(陰性)の電位にある。そのため、本件ナノ粒子がカチオン性ナノ粒子であると、イオン間相互作用により本件ナノ粒子が細胞内の広域に効率的に導入されやすい。その結果、凍結保存による細胞障害をより低減する。
カチオン性ナノ粒子とは、ゼータ電位が10mV超のナノ粒子のことをいう。
中性ナノ粒子とは、ゼータ電位が-10mV以上10mV以下のナノ粒子のことをいう。
アニオン性ナノ粒子とは、ゼータ電位が-10mV未満のナノ粒子のことをいう。
中性ナノ粒子とは、ゼータ電位が-10mV以上10mV以下のナノ粒子のことをいう。
アニオン性ナノ粒子とは、ゼータ電位が-10mV未満のナノ粒子のことをいう。
本件ナノ粒子のゼータ電位は、10mV以上80mV以下であることが好ましく、15mV以上70mV以下であることがより好ましく、20mV以上60mV以下であることがさらに好ましく、25mV以上50mV以下であることが特に好ましい。
本件ナノ粒子のゼータ電位が10mV以上であると、本件ナノ粒子の膜が適度に正電荷を帯びる。そのため、細胞内に効率的に本件ナノ粒子がより導入されやすく、凍結保存による細胞障害をより低減する。

本件ナノ粒子のゼータ電位及び体積平均粒子径の測定は、たとえば以下のとおり行うことができる。
本件凍結保存用組成物を、リン酸緩衝生理食塩水(PBS、pH7.4)で100倍に希釈した溶液を1mL採取し、測定用1mLセルに充填する。そして、これを粒子径・ゼータ電位測定装置(ZETASIZER PRO、Malvern社製)により、室温(25℃)で測定する。
本件凍結保存用組成物を、リン酸緩衝生理食塩水(PBS、pH7.4)で100倍に希釈した溶液を1mL採取し、測定用1mLセルに充填する。そして、これを粒子径・ゼータ電位測定装置(ZETASIZER PRO、Malvern社製)により、室温(25℃)で測定する。
本件ナノ粒子の表面電荷、つまり、ゼータ電位の値は、本件ナノ粒子を構成する両親媒性分子の電荷の影響を受ける。そのため、所望のゼータ電位のナノ粒子を調製する際には、そのゼータ電位に応じて両親媒性分子を適宜選択若しくは変更して用い、本件ナノ粒子を調製する方法が挙げられる。
両親媒性分子としては、界面活性剤を挙げることができ、合成の界面活性剤でもあっても天然若しくは生体由来の界面活性剤でもよい。
合成の界面活性剤としては、Gemini型界面活性剤を挙げることができる。Gemini型界面活性剤は、1つの分子中に親水基と疎水基をもち、スペーサー基により親水基を介して互いに連結しており、2つの界面活性単位を有する二量体界面活性剤又は3つの界面活性単位を有する三量体界面活性剤である。
Gemini型界面活性剤を構成する疎水基としては、炭素数8~24、好ましくは炭素数12~18、より好ましくは炭素数12又は16の直鎖炭化水素鎖を挙げることができ、直鎖の飽和炭化水素鎖であることがナノ粒子の安定性の観点から好ましい。また、2つ又は3つの疎水基を構成する直鎖炭化水素鎖は同じでも異なってもよいが、同じであることが好ましい。
Gemini型界面活性剤を構成する親水基としては、極性基、イオン性基が挙げられる。親水基における対イオンを生じさせる元素としては、用いる水溶液に応じて適宜調整可能であるが、臭素(Br)、塩素(Cl)、窒素(N)、ナトリウム(Na)、リン(P)、ホウ素(B)、ヨウ素(I)、フッ素(F)、硫黄(S)、酸素(O)、炭素(C)、ベリリウム(Be)、鉄(Fe)、カルシウム(Ca)、マグネシウム(Mg)を挙げることができ、これらを適宜組み合わせて用いることもできる。
上記スペーサーは通常炭化水素鎖であり、疎水基が互いに独立して作用できるように十分な距離を与えるための長さを有することが好ましい。上記スペーサーの炭素数としては特に限定されないが、2~12、好ましくは2~8、更に好ましくは2~4、最も好ましくは2若しくは3を挙げることができる。
天然及び生体由来の界面活性剤としては、両親媒性の脂質を挙げることができる。両親媒性の脂質としては、リン非含有脂質でもリン脂質でもよい。また、カチオン性脂質であっても、中性脂質であっても、アニオン性脂質であってもよく、これらの組み合わせでもよい。
リン非含有脂質としては、N-(2,3-ジオレオイロキシ-1-プロピル)トリメチルアンモニウムメチルサルフェ-ト(DOTAP)、N-〔1-(2,3-オレイルオキシ)プロピル〕-N,N,N-トリメチルアンモニウムクロライド(DOTMA)、ジオクタデシルアミド-グリシルスペルミン(DOGS)等を挙げることができる。また、リン脂質としては、1,2-ジオレオイル-sn-グリセロ-3-ホスホエタノールアミン(DOPE)、1,2-ジステアロイル-sn-グリセロ-3-ホスホエタノールアミン(DSPE)、1,2-ジパルミトイル-sn-グリセロ-3-ホスホエタノールアミン(DPPE)、ジフィタノイル-sn-グリセロ-3-ホスホエタノールアミン(DPhPE)等のホスホエタノールアミン、1,2-ジパルミトイル-sn-グリセロ-3-ホスホコリン(DPPC)、1,2-ジミリストイル-sn-グリセロ-3-ホスホコリン(DMPC)、1,2-ジステアロイル-sn-グリセロ-3-ホスホコリン(DSPC)、ジフタノイル-sn-グリセロ-3-ホスホコリン(DPhPC)等のホスホコリンを挙げることができる。
両親媒性分子は標識物質(例えばクマリン、FITC(Fluorescein Isothiocyanate)等の蛍光分子、マレイミド基、PEG基など)を分子内に導入した界面活性剤で構成されていてもよい。
本件ナノ粒子は、コレステロールを含むことが好ましく、本件ナノ粒子の膜にコレステロールを含有することがより好ましい。コレステロールを膜に含むように本件ナノ粒子が構成されると、コレステロールを含まない膜に比べて、本件ナノ粒子における膜の流動性が向上する。
[その他の成分]
本実施形態に係る凍結保存用組成物は、本件ナノ粒子以外のその他の成分(以下、単に「その他の成分」とも称す。)をさらに含んでいてもよい。
その他の成分としては、例えば、本件ナノ粒子に内包される内包成分;分散媒や、本件ナノ粒子を作製する過程で単層又は二層を形成しなかった両親媒性分子若しくはそれらの凝集体などが挙げられる。
本実施形態に係る凍結保存用組成物は、本件ナノ粒子以外のその他の成分(以下、単に「その他の成分」とも称す。)をさらに含んでいてもよい。
その他の成分としては、例えば、本件ナノ粒子に内包される内包成分;分散媒や、本件ナノ粒子を作製する過程で単層又は二層を形成しなかった両親媒性分子若しくはそれらの凝集体などが挙げられる。
・内包成分
内包成分は、本件ナノ粒子の膜内や、本件ナノ粒子を構成する界面活性剤に取り込まれる成分や内水相に内包される成分のことを指す。
内包成分としては、標識物質(例えばクマリン等の蛍光物質など)、脂溶性薬物、水溶性薬物、核酸、タンパク質、抗体、結晶性薬物、生体由来成分、水溶性ビタミン、水溶性ビタミン誘導体、脂溶性ビタミン、脂溶性ビタミン誘導体等の成分が挙げられる。
具体的な内包成分としては、例えば、細胞の保護効果を有する活性型ビタミンC、活性型ビタミンA;保水成分であるフェルラ酸、ヒアルロン酸などが挙げられる。
内包成分は、本件ナノ粒子の膜内や、本件ナノ粒子を構成する界面活性剤に取り込まれる成分や内水相に内包される成分のことを指す。

本件ナノ粒子は、水溶性ビタミン、水溶性ビタミン誘導体、及びフェルラ酸からなる群から選択される少なくとも1種を内包することが好ましく、フェルラ酸及び活性型ビタミンCの少なくとも一方を内包することがより好ましい。
特に、フェルラ酸及び活性型ビタミンCは、酸化防止能、水分保持性に優れることから、本件ナノ粒子がこれらの少なくとも一方を内包することで、細胞凍結保存による細胞障害がより抑制される。

・分散媒
本件ナノ粒子は、分散媒中に分散されて存在することが好ましい。
分散媒としては、水、緩衝液、水溶性溶剤、液体培地等が挙げられる。分散媒は、1種単独の使用であっても、2種以上の併用であってもよい。

水としては、例えば、蒸留水、イオン交換水、限外濾過水、純水等が挙げられる。
水の含有量は、分散媒の全量に対して、70質量%以上であることが好ましく、80質量%以上90質量%以下がより好ましく、90質量%以上100質量%以下がさらに好ましい。水の含有量を上記数値範囲内とすることで、本件ナノ粒子がより安定に存在する。

緩衝液としては、例えば、リン酸緩衝液(PBS)、トリスヒドロキシメチルアミノメタン緩衝液(TRIS)、ヒドロキシエチルピペラジンエタンスルホン酸緩衝液(HEPES)、ホウ酸緩衝液、酢酸緩衝液等が挙げられる。
緩衝液の濃度は浸透圧値に準じて、ヒトの血漿浸透圧である285±5m0sm/Lと同程度の範囲で用いるのが好ましい。
緩衝液のpHは特に制限されないが、例えば、細胞生存性等の観点からは、pH5以上pH10以下であることが好ましく、pH6以上pH9以下であることがより好ましい。
pHは、pHメーターを用いて25℃で測定される値である(以下同様)。
緩衝液の濃度は浸透圧値に準じて、ヒトの血漿浸透圧である285±5m0sm/Lと同程度の範囲で用いるのが好ましい。

液体培地としては、例えば、幹細胞の培養に用いられる公知の培地が挙げられる。より具体的には液体培地としては、例えば、DMEM(Dulbecco's Modified Eagle's Medium)、DMEM:F-12(Dulbecco's Modified Eagle Medium: Nutrient Mixture F-12)、EMEM(Eagle's minimal essential medium)及びこれらの培地に血小板由来増殖因子(PDGF)、上皮増殖因子(EGF)、線維芽細胞増殖因子(FGF)、肝細胞増殖因子(HGF)、血管内皮増殖因子(VEGF)、インスリン様増殖因子(IGF)、神経成長因子(NGF)等の細胞増殖因子を添加した液体培地などが挙げられる。

(第1工程)
第1工程では、本件ナノ粒子の少なくとも一方を含む分散液を得る。この分散液は、本件ナノ粒子の膜を構成する成分を水溶性溶剤(例えばアルコール等)に溶解させた溶液と、水又は緩衝液と、を混合し、条件(例えばpH、温度)を制御して調製することができる。なお、本件ナノ粒子を構成する両親媒性分子については、上述の通りであるので、ここでの説明を省略する。
例えばリポソームを含む分散液は、エタノールと脂質とを混合して脂質を溶解し、pHを調整しながら水に混合、撹拌することで調製できる。

<凍結保存方法>
本件凍結保存方法は、細胞又は生体組織と本件凍結保存用組成物とを混合して本件ナノ粒子を細胞内に導入する工程(以下、「導入工程」と称す。)と、本件ナノ粒子が導入された細胞又は生体組織を凍結する工程(以下、「凍結工程」とも称す。)と、を含む。なお、生体組織を凍結する場合においては、上記「細胞内に導入」は「生体組織を構成する細胞内に導入」の意味である。
本件凍結保存方法によれば、凍結保存による細胞障害が低減される。
本件凍結保存方法は、細胞又は生体組織と本件凍結保存用組成物とを混合して本件ナノ粒子を細胞内に導入する工程(以下、「導入工程」と称す。)と、本件ナノ粒子が導入された細胞又は生体組織を凍結する工程(以下、「凍結工程」とも称す。)と、を含む。なお、生体組織を凍結する場合においては、上記「細胞内に導入」は「生体組織を構成する細胞内に導入」の意味である。

本件凍結保存方法の凍結対象となる細胞は、特に制限されず、凍結保存される公知の細胞が採用できる。例えば、細胞としては、ヒト脂肪組織由来間葉系幹細胞(hAD-MSC)、ヒト骨髄由来間葉系幹細胞(hMSC-BM)の間葉系幹細胞、神経幹細胞、皮膚幹細胞、造血幹細胞、歯髄幹細胞、肝幹細胞、筋幹細胞、脂肪幹細胞等の体性幹細胞;誘導多能性幹細胞(iPS細胞)、胚性幹細胞(ES細胞)、等の多能性幹細胞;リンパ球、好中球等の血液細胞;上皮細胞、内皮細胞、筋肉細胞、線維芽細胞(皮膚細胞等)、平滑筋細胞、メラノサイト、毛細胞、肝細胞、胃粘膜細胞、腸細胞、脾細胞、膵細胞(膵外分泌細胞等)の臓器固有の細胞;脳細胞、肺細胞、腎細胞及び脂肪細胞等の分化した細胞;胚、受精卵、精子などの生殖細胞が挙げられる。また、本件凍結保存方法の凍結対象として、オルガノイド、スフェロイド、細胞シート、上記細胞で構成される生体組織も挙げることができる。
[導入工程]
導入工程では、細胞又は生体組織と本件凍結保存用組成物とを混合して本件ナノ粒子を細胞(生体組織を凍結する場合は生体組織を構成する細胞、以下同様)内に導入する。
培地は、特に制限されず、幹細胞の培養に用いられる公知の培地が挙げられる。例えば、培地としては、先述の液体培地が挙げられる。
導入工程では、細胞又は生体組織と本件凍結保存用組成物とを混合して本件ナノ粒子を細胞(生体組織を凍結する場合は生体組織を構成する細胞、以下同様)内に導入する。
培地は、特に制限されず、幹細胞の培養に用いられる公知の培地が挙げられる。例えば、培地としては、先述の液体培地が挙げられる。
本件ナノ粒子を細胞内に導入する手段は、特に制限されず、例えば、下記(I)~(IV)等の方法が挙げられる:
(I)細胞を含む培地に本件凍結保存用組成物を添加する方法
(II)細胞を含む培地を遠心分離して上清を除いてから本件凍結保存用組成物を添加する方法
(III)培養容器から剥離した細胞を本件凍結保存用組成物に添加する方法。
(IV)エレクトロポレーションにより電圧を利用して細胞内に導入する方法。
(I)細胞を含む培地に本件凍結保存用組成物を添加する方法
(II)細胞を含む培地を遠心分離して上清を除いてから本件凍結保存用組成物を添加する方法
(III)培養容器から剥離した細胞を本件凍結保存用組成物に添加する方法。

[凍結工程]
凍結工程では、本件ナノ粒子が導入された細胞又は生体組織を凍結する。
細胞又は生体組織を凍結する手段は、特に制限されず、公知の凍結方法が採用できる。
細胞又は生体組織を凍結する温度としては、-30℃~-196℃(好ましくは-80℃~-196℃)の範囲内であることが好ましい。細胞又は生体組織を凍結する手段は、例えば、凍結保存による細胞障害をより低減する観点からは、ディープフリーザーもしくは液体窒素を用いて細胞又は生体組織を凍結することが好ましい。凍結期間は特に制限されず、0.5日以上50年以下、1日以上10年以下、1週間以上5年以下、1ヶ月以上1年以下を挙げることができる。
凍結工程では、本件ナノ粒子が導入された細胞又は生体組織を凍結する。
細胞又は生体組織を凍結する手段は、特に制限されず、公知の凍結方法が採用できる。
細胞又は生体組織を凍結する温度としては、-30℃~-196℃(好ましくは-80℃~-196℃)の範囲内であることが好ましい。細胞又は生体組織を凍結する手段は、例えば、凍結保存による細胞障害をより低減する観点からは、ディープフリーザーもしくは液体窒素を用いて細胞又は生体組織を凍結することが好ましい。凍結期間は特に制限されず、0.5日以上50年以下、1日以上10年以下、1週間以上5年以下、1ヶ月以上1年以下を挙げることができる。
[その他の工程]
本件凍結保存方法は、上記導入工程及び凍結工程以外のその他の工程をさらに含んでいてもよい。前記その他の工程としては、例えば、凍結した細胞を適切な温度下に保存する保存工程等が挙げられる。

[凍結細胞]
本実施形態にかかる凍結した細胞又は生体組織は、本件ナノ粒子を含む凍結細胞又は生体組織である。この凍結した細胞又は生体組織は、上述の本件凍結保存方法によって作製することができる。

以下に実施例を挙げて、本発明をさらに具体的に説明する。以下の実施例に示す材料、使用量、割合、処理手順等は、本発明の趣旨を逸脱しない限り適宜変更することができる。従って、本発明の範囲は以下に示す具体例により限定的に解釈されるものではない。
<実施例1>
・凍結保存用組成物の作製
以下の手順に従って実施例1としてDOTAPを構成成分とする二重膜小胞を有機溶媒注入法により作製した。
(1)ビーカーにPBS(25℃でのpH=7.4)を5.3ml入れ、25℃で攪拌した。
(2)遠心管の中に、エタノール1990μLと、コレステロール38.7mgと、両親媒性分子として脂質であるDOTAP11.1mgとを混合し、この混合液の温度を、前記脂質の相転移温度以上の75℃としコレステロールと前記脂質を完全に溶解した。なお、後述の細胞導入性の評価を行う場合には、さらに蛍光標識クマリンも加え、減衰を防ぐために遮光条件下で調整や試験を実施した。なお、クマリンは疎水性のため、主に形成されるナノ粒子における膜に取り込まれる。
(3)前記混合液をシリンジで全量取り、前記PBS中に急速に混合した。
(4)ビーカーを遮光して、25℃で1時間攪拌した。
(5)透析膜(Spectra/Por RC Biotech Dialysis Membrane MWCO:8-10)を用いて、PBS中で4時間以上、室温で透析を行った。これにより、ナノ粒子の溶液を得た。
(6)得られたナノ粒子の溶液を、Avestin社製のLF-STBリポソファストスタビライザーにセットし、使用するフィルター孔径を1000nm、800nm、400nm、200nm、100nm及び50nmのこの順で順次加圧しながら通過させることでサイジングした。得られたナノ粒子を含有する液体組成物を、実施例1の凍結保存用組成物とした。細胞凍結保存用組成物のpH(25℃条件下)は7.2であった。
・凍結保存用組成物の作製
以下の手順に従って実施例1としてDOTAPを構成成分とする二重膜小胞を有機溶媒注入法により作製した。
(1)ビーカーにPBS(25℃でのpH=7.4)を5.3ml入れ、25℃で攪拌した。
(2)遠心管の中に、エタノール1990μLと、コレステロール38.7mgと、両親媒性分子として脂質であるDOTAP11.1mgとを混合し、この混合液の温度を、前記脂質の相転移温度以上の75℃としコレステロールと前記脂質を完全に溶解した。なお、後述の細胞導入性の評価を行う場合には、さらに蛍光標識クマリンも加え、減衰を防ぐために遮光条件下で調整や試験を実施した。なお、クマリンは疎水性のため、主に形成されるナノ粒子における膜に取り込まれる。
(3)前記混合液をシリンジで全量取り、前記PBS中に急速に混合した。
(4)ビーカーを遮光して、25℃で1時間攪拌した。
(5)透析膜(Spectra/Por RC Biotech Dialysis Membrane MWCO:8-10)を用いて、PBS中で4時間以上、室温で透析を行った。これにより、ナノ粒子の溶液を得た。
(6)得られたナノ粒子の溶液を、Avestin社製のLF-STBリポソファストスタビライザーにセットし、使用するフィルター孔径を1000nm、800nm、400nm、200nm、100nm及び50nmのこの順で順次加圧しながら通過させることでサイジングした。得られたナノ粒子を含有する液体組成物を、実施例1の凍結保存用組成物とした。細胞凍結保存用組成物のpH(25℃条件下)は7.2であった。
<実施例2~5>
両親媒性分子の種類を、DOTAPから、DOPE、Gemini型、DPPC、DSPEへと変更した以外は、実施例1と同様の仕様とし、実施例2~5の凍結保存用組成物を得た。「Gemini型」は「Gemini型界面活性剤」を意味する。なお、実施例3、後述の実施例8及び13で用いたGemini型界面活性剤の分子構造を下記に示す。2つの界面活性単位を有する二量体界面活性剤であり、疎水基は炭素数12の炭化水素鎖からなり、スペーサー基の炭素数が3からなり、対イオンを生じさせる元素としては臭素及び窒素からなる。
両親媒性分子の種類を、DOTAPから、DOPE、Gemini型、DPPC、DSPEへと変更した以外は、実施例1と同様の仕様とし、実施例2~5の凍結保存用組成物を得た。「Gemini型」は「Gemini型界面活性剤」を意味する。なお、実施例3、後述の実施例8及び13で用いたGemini型界面活性剤の分子構造を下記に示す。2つの界面活性単位を有する二量体界面活性剤であり、疎水基は炭素数12の炭化水素鎖からなり、スペーサー基の炭素数が3からなり、対イオンを生じさせる元素としては臭素及び窒素からなる。
<実施例6~15>
実施例1の凍結保存用組成物を作製する操作(1)を、下記に示す操作(1’)とした以外は、実施例1~5と同じ手法により実施例6~15の凍結保存用組成物を得た。実施例6~10は順にDOTAP、DOPE、Gemini型、DPPC、DSPEを構成成分とし、フェルラ酸を含有したナノ粒子を含有する凍結保存用組成物であり、実施例11~15は順にDOTAP、DOPE、Gemini型、DPPC、DSPEを構成成分とし、活性型ビタミンCを含有したナノ粒子を含有する凍結保存用組成物である。
(1’)ビーカーにPBS(25℃でのpH=7.4)を5.3ml入れて、内包成分としてフェルラ酸又は活性型ビタミンCを10μM溶解し25℃で攪拌した。
実施例1の凍結保存用組成物を作製する操作(1)を、下記に示す操作(1’)とした以外は、実施例1~5と同じ手法により実施例6~15の凍結保存用組成物を得た。実施例6~10は順にDOTAP、DOPE、Gemini型、DPPC、DSPEを構成成分とし、フェルラ酸を含有したナノ粒子を含有する凍結保存用組成物であり、実施例11~15は順にDOTAP、DOPE、Gemini型、DPPC、DSPEを構成成分とし、活性型ビタミンCを含有したナノ粒子を含有する凍結保存用組成物である。
(1’)ビーカーにPBS(25℃でのpH=7.4)を5.3ml入れて、内包成分としてフェルラ酸又は活性型ビタミンCを10μM溶解し25℃で攪拌した。
実施例11~15にて使用した活性型ビタミンCとは、下記の分子構造を有する化合物である。
<比較例1>
比較例1では、10体積%のジメチルスルホキシド(DMSO)、40体積%ウシ胎児血清(FBS)及び50体積%幹細胞培地の混合液1mLを凍結保存用組成物とした。
比較例1では、10体積%のジメチルスルホキシド(DMSO)、40体積%ウシ胎児血清(FBS)及び50体積%幹細胞培地の混合液1mLを凍結保存用組成物とした。
<比較例2>
比較例2では、ナノ粒子を含まないZNQ社製のCELLBANKER(登録商標)1(1mL)を、凍結保存用組成物とした。
比較例2では、ナノ粒子を含まないZNQ社製のCELLBANKER(登録商標)1(1mL)を、凍結保存用組成物とした。
〔試験例1〕
<体積平均粒子径及びゼータ電位>
各実施例の凍結保存用組成物について、体積平均粒子径及びゼータ電位を測定した。具体的には、実施例1~5の凍結保存用組成物を、リン酸緩衝生理食塩水(PBS、pH7.4)で100倍に希釈した溶液を1mL採取し、測定用1mLセルに充填した。そして、これを粒子径・ゼータ電位測定装置(ZETASIZER PRO、Malvern社製)により、室温(25℃)で測定した。その結果を表1に示す。また実施例1の凍結保存用組成物に含まれるナノ粒子の粒子径の分布を図1に示す。
<体積平均粒子径及びゼータ電位>
各実施例の凍結保存用組成物について、体積平均粒子径及びゼータ電位を測定した。具体的には、実施例1~5の凍結保存用組成物を、リン酸緩衝生理食塩水(PBS、pH7.4)で100倍に希釈した溶液を1mL採取し、測定用1mLセルに充填した。そして、これを粒子径・ゼータ電位測定装置(ZETASIZER PRO、Malvern社製)により、室温(25℃)で測定した。その結果を表1に示す。また実施例1の凍結保存用組成物に含まれるナノ粒子の粒子径の分布を図1に示す。
〔試験例2〕
<細胞導入性の評価>
実施例1~5の凍結保存用組成物について、凍結保存用組成物に含まれるナノ粒子の細胞導入性を下記の手順で評価した。
(1)コラーゲンコートされた8wellチャンバースライドに凍結融解後のヒト脂肪組織由来間葉系幹細胞(hAD-MSC)を2000cells/well播種した。
(2)well内に、蛍光色素クマリンを標識した実施例1~5の凍結保存用組成物を加え、37℃で30分間インキュベートした。
(3)細胞核の染色にはDAPI溶液(4’,6-ジアミジノ-2-フェニルインドール二塩酸塩溶液)用いた。
(4)蛍光顕微鏡(KEYENCE Japan社製、BZ-9000)で20倍に拡大し、露光時間1/2秒、ブラックバランスを42としたときに観測される細胞質及び細胞核の蛍光強度をそれぞれ測定した。細胞質内であり且つ細胞核外の領域に存在するナノ粒子に内包されるクマリン蛍光強度OUTに対する、細胞核内に存在するナノ粒子に内包されるクマリン蛍光強度INの比(IN/OUT)の値を、表2における細胞導入性の評価の項目[蛍光強度比(細胞核/細胞質)]に示す。
<細胞導入性の評価>
実施例1~5の凍結保存用組成物について、凍結保存用組成物に含まれるナノ粒子の細胞導入性を下記の手順で評価した。
(1)コラーゲンコートされた8wellチャンバースライドに凍結融解後のヒト脂肪組織由来間葉系幹細胞(hAD-MSC)を2000cells/well播種した。
(2)well内に、蛍光色素クマリンを標識した実施例1~5の凍結保存用組成物を加え、37℃で30分間インキュベートした。
(3)細胞核の染色にはDAPI溶液(4’,6-ジアミジノ-2-フェニルインドール二塩酸塩溶液)用いた。
(4)蛍光顕微鏡(KEYENCE Japan社製、BZ-9000)で20倍に拡大し、露光時間1/2秒、ブラックバランスを42としたときに観測される細胞質及び細胞核の蛍光強度をそれぞれ測定した。細胞質内であり且つ細胞核外の領域に存在するナノ粒子に内包されるクマリン蛍光強度OUTに対する、細胞核内に存在するナノ粒子に内包されるクマリン蛍光強度INの比(IN/OUT)の値を、表2における細胞導入性の評価の項目[蛍光強度比(細胞核/細胞質)]に示す。
表2に示すように、蛍光強度比(細胞核/細胞質)の値が正数であることから、実施例1~5の凍結保存用組成物に含まれるナノ粒子の一部は、細胞質に留まらず細胞核内にまで導入されていることがわかった。
〔試験例3〕
<透過型電子顕微鏡>
試験例2の方法で細胞内に導入されたナノ粒子を透過型電子顕微鏡で観察した。健常者の脂肪組織由来間葉系幹細胞(hAD-MSC)は、増殖因子含有の専用培地5mLを用いて約5000cells/cm2の密度で3.5cm2シャーレに播種した。金ナノ粒子(Cytodiagnostics社)を用いて実施例3(Gemini型)の凍結保存用組成物を標識した。標識した凍結保存用組成物をADSCに30分間曝露した。透過型電子顕微鏡用のサンプル調製のため、前固定として2%グルタールアルデヒド含有リン酸緩衝液を用いてADSCを4℃で一晩固定した。固定したサンプルをリン酸緩衝液で洗浄後、後固定として2%オスミウム水溶液を用いて4℃で2時間固定した。脱水としてエタノールの濃度を30%から100%へ段階的に上昇させそれぞれ15分間処理を行った。サンプルをエポキシ樹脂に60℃で48時間かけて包埋し、ウルトラミクロトームを用いて80-90nmの超薄切片を作製し、Cu 200メッシュに積載した。透過型電子顕微鏡H-7600を用いて、100kVで観察を行った。結果を図2に示す。
<透過型電子顕微鏡>
試験例2の方法で細胞内に導入されたナノ粒子を透過型電子顕微鏡で観察した。健常者の脂肪組織由来間葉系幹細胞(hAD-MSC)は、増殖因子含有の専用培地5mLを用いて約5000cells/cm2の密度で3.5cm2シャーレに播種した。金ナノ粒子(Cytodiagnostics社)を用いて実施例3(Gemini型)の凍結保存用組成物を標識した。標識した凍結保存用組成物をADSCに30分間曝露した。透過型電子顕微鏡用のサンプル調製のため、前固定として2%グルタールアルデヒド含有リン酸緩衝液を用いてADSCを4℃で一晩固定した。固定したサンプルをリン酸緩衝液で洗浄後、後固定として2%オスミウム水溶液を用いて4℃で2時間固定した。脱水としてエタノールの濃度を30%から100%へ段階的に上昇させそれぞれ15分間処理を行った。サンプルをエポキシ樹脂に60℃で48時間かけて包埋し、ウルトラミクロトームを用いて80-90nmの超薄切片を作製し、Cu 200メッシュに積載した。透過型電子顕微鏡H-7600を用いて、100kVで観察を行った。結果を図2に示す。
図2に示すように実施例3の凍結保存用組成物に含まれるナノ粒子が細胞質内に取り込まれていた。さらに、粒子径が小さいナノ粒子の一部は核内にも取り込まれていた。
〔試験例4〕
<凍結保存用組成物を用いた細胞の凍結保存と解凍処理>
・細胞の凍結保存
(1)70%~90%コンフルエントのヒト脂肪組織由来間葉系幹細胞(hAD-MSC)をトリプシン処理し、培養容器から剥離し遠心して細胞ペレットを得た。
(2)上記の約5×106個の細胞に対し、各実施例の凍結保存用組成物を10μL加えて細胞に導入し、細胞培養液990μLを添加し細胞懸濁液を得た。比較例の場合は各実施例の凍結保存用組成物1mLを添加し細胞懸濁液を得た。
(3)上記細胞懸濁液をクライオチューブに分注し、-80℃のディープフリーザーで凍結保存した。
<凍結保存用組成物を用いた細胞の凍結保存と解凍処理>
・細胞の凍結保存
(1)70%~90%コンフルエントのヒト脂肪組織由来間葉系幹細胞(hAD-MSC)をトリプシン処理し、培養容器から剥離し遠心して細胞ペレットを得た。
(2)上記の約5×106個の細胞に対し、各実施例の凍結保存用組成物を10μL加えて細胞に導入し、細胞培養液990μLを添加し細胞懸濁液を得た。比較例の場合は各実施例の凍結保存用組成物1mLを添加し細胞懸濁液を得た。
(3)上記細胞懸濁液をクライオチューブに分注し、-80℃のディープフリーザーで凍結保存した。
・凍結保存された細胞の解凍
(1)上記凍結保存(1ヶ月以内)された細胞を含むクライオチューブを37℃の温浴で振りながら細胞を迅速に解凍した。
(2)解凍後の細胞懸濁液を、幹細胞専用培地10mLと混合し、混合液を得た。
(3)上記混合液を遠心分離(1,200rpm、5分、4℃)し、アスピレーターで上清を取り除いた後、適量の上記培地で懸濁した。
(1)上記凍結保存(1ヶ月以内)された細胞を含むクライオチューブを37℃の温浴で振りながら細胞を迅速に解凍した。
(2)解凍後の細胞懸濁液を、幹細胞専用培地10mLと混合し、混合液を得た。
(3)上記混合液を遠心分離(1,200rpm、5分、4℃)し、アスピレーターで上清を取り除いた後、適量の上記培地で懸濁した。
<細胞生存率の評価>
細胞凍結保存前後における細胞生存率は、細胞増殖アッセイキット(MTT Cell Viability Assay Kit, AR1156、BTI:コスモ・バイオ社製)を用いて、下記の手順で評価した。
(1)96wellプレートに、実施例3、6~15の凍結保存用組成物を用いて凍結保存した後に解凍処理された細胞を、2000cells/well播種した。
(2)各wellに、前記キット中のMTT標準試薬10μLを加え、37℃で4時間インキュベートした。
(3)各wellにホルマザン溶液を100μL加え、37℃で18時間インキュベートした。
(4)マイクロプレート吸光度リーダーを用いて、吸収波長570nmにおける色素の吸光度Aを測定した。
(5)上記(1)~(3)の操作を、細胞凍結する前のhMSC-ATを用いて行った後、マイクロプレート吸光度リーダーを用いて、吸収波長570nmにおける色素の吸光度Bを測定した。
(6)凍結保存前の吸光度Bに対する、上記(4)で得られた解凍後の吸光度Aの割合(=A/B×100%)を、細胞生存率とした。結果を表3、4に示す。
細胞凍結保存前後における細胞生存率は、細胞増殖アッセイキット(MTT Cell Viability Assay Kit, AR1156、BTI:コスモ・バイオ社製)を用いて、下記の手順で評価した。
(1)96wellプレートに、実施例3、6~15の凍結保存用組成物を用いて凍結保存した後に解凍処理された細胞を、2000cells/well播種した。
(2)各wellに、前記キット中のMTT標準試薬10μLを加え、37℃で4時間インキュベートした。
(3)各wellにホルマザン溶液を100μL加え、37℃で18時間インキュベートした。
(4)マイクロプレート吸光度リーダーを用いて、吸収波長570nmにおける色素の吸光度Aを測定した。
(5)上記(1)~(3)の操作を、細胞凍結する前のhMSC-ATを用いて行った後、マイクロプレート吸光度リーダーを用いて、吸収波長570nmにおける色素の吸光度Bを測定した。
(6)凍結保存前の吸光度Bに対する、上記(4)で得られた解凍後の吸光度Aの割合(=A/B×100%)を、細胞生存率とした。結果を表3、4に示す。
表3、4に示すように、実施例3の凍結保存用組成物や、フェルラ酸又は活性型ビタミンCを内包する6~15の凍結保存用組成物を用いて凍結すると、比較例の凍結保存用組成物を用いて凍結した場合に比べて、細胞生存率が高い、つまり、凍結保存による細胞障害が低減されることがわかった。
〔試験例5〕
<幹細胞の割合の評価>
実施例3、6~15の凍結保存用組成物を用いて細胞を凍結保存し、下記の手順で、Human Mesenchymal Stem Cell Multi-Color Flow Kit(FMC020、R&D systems社製)を用いて幹細胞率を評価した。当該キットは、CD90を陽性マーカーとし、CD45、CD34、CD11b、CD79A及びHLA-DRを陰性マーカーとする。
(1)実施例3、6~15の凍結保存用組成物を用いて凍結保存した後に解凍処理された5×106cellsの細胞を、上記キットに付属するフローサイトメトリー用染色緩衝液100μLと混合し、これをフローサイト用チューブにうつして、細胞懸濁液とした。
(2)上記細胞懸濁液に、上記キットに付属する2種の蛍光色素標識抗体の溶液(陽性マーカーの抗体液1種と陰性マーカーを混合した抗体液1種)を10μLずつ加え、これを室温(25℃)暗所にて45分間インキュベートした。
(3)インキュベート後の細胞懸濁液を上記フローサイトメトリー用染色緩衝液2mLで洗浄した。
(4)フローサイトメーターを用いてフローサイトメトリー分析し、細胞凍結保存後におけるCD90(+)、CD45(-)、CD34(-)、CD11b(-)、CD79A(-)及びHLA-DR(-)の形質を示す細胞を定量した。実施例3、6~15の凍結保存用組成物を用いて凍結保存した場合における当該細胞の割合を「幹細胞の割合」として表5、6に示す。
<幹細胞の割合の評価>
実施例3、6~15の凍結保存用組成物を用いて細胞を凍結保存し、下記の手順で、Human Mesenchymal Stem Cell Multi-Color Flow Kit(FMC020、R&D systems社製)を用いて幹細胞率を評価した。当該キットは、CD90を陽性マーカーとし、CD45、CD34、CD11b、CD79A及びHLA-DRを陰性マーカーとする。
(1)実施例3、6~15の凍結保存用組成物を用いて凍結保存した後に解凍処理された5×106cellsの細胞を、上記キットに付属するフローサイトメトリー用染色緩衝液100μLと混合し、これをフローサイト用チューブにうつして、細胞懸濁液とした。
(2)上記細胞懸濁液に、上記キットに付属する2種の蛍光色素標識抗体の溶液(陽性マーカーの抗体液1種と陰性マーカーを混合した抗体液1種)を10μLずつ加え、これを室温(25℃)暗所にて45分間インキュベートした。
(3)インキュベート後の細胞懸濁液を上記フローサイトメトリー用染色緩衝液2mLで洗浄した。
(4)フローサイトメーターを用いてフローサイトメトリー分析し、細胞凍結保存後におけるCD90(+)、CD45(-)、CD34(-)、CD11b(-)、CD79A(-)及びHLA-DR(-)の形質を示す細胞を定量した。実施例3、6~15の凍結保存用組成物を用いて凍結保存した場合における当該細胞の割合を「幹細胞の割合」として表5、6に示す。
表5、6より、実施例3、6~15のナノ粒子を含む凍結保存用組成物を用いて凍結保存した場合は、比較例の凍結保存用組成物を用いた場合に比べて幹細胞の割合が高い、つまり、凍結保存による幹細胞機能の保持に有用であることがわかった。
〔試験例6〕
<Gemini型界面活性剤の比較>
上記試験では実施例3、8、13の凍結保存用組成物、すなわちGemini型界面活性剤として疎水基の炭化水素鎖の炭素数が12、スペーサー基の炭素数が3からなり、対イオンを形成する元素が臭素及び窒素からなるGemini型界面活性剤を用いた。そこで、Gemini型界面活性剤として、2つのアルキル鎖の炭素数、スペーサーの炭素数、及び対イオンを形成する元素を変えて、実施例1と同様の方法で実施例16~19の凍結保存用組成物を作製して、実施例3の凍結保存用組成物と共に試験例4と同じ方法で凍結後の生存率を調べた。結果を表7に示す。表7中、12-3-12_Brは、それぞれ2つの炭化水素鎖の炭素数が12、スペーサーの炭素数3、対イオンを生じさせる元素が臭素と窒素であることを、16-3-16_Clは、それぞれ2つの炭化水素鎖の炭素数が16、スペーサーの炭素数が3、対イオンが塩素と窒素であることを意味する。
<Gemini型界面活性剤の比較>
上記試験では実施例3、8、13の凍結保存用組成物、すなわちGemini型界面活性剤として疎水基の炭化水素鎖の炭素数が12、スペーサー基の炭素数が3からなり、対イオンを形成する元素が臭素及び窒素からなるGemini型界面活性剤を用いた。そこで、Gemini型界面活性剤として、2つのアルキル鎖の炭素数、スペーサーの炭素数、及び対イオンを形成する元素を変えて、実施例1と同様の方法で実施例16~19の凍結保存用組成物を作製して、実施例3の凍結保存用組成物と共に試験例4と同じ方法で凍結後の生存率を調べた。結果を表7に示す。表7中、12-3-12_Brは、それぞれ2つの炭化水素鎖の炭素数が12、スペーサーの炭素数3、対イオンを生じさせる元素が臭素と窒素であることを、16-3-16_Clは、それぞれ2つの炭化水素鎖の炭素数が16、スペーサーの炭素数が3、対イオンが塩素と窒素であることを意味する。
表7より、炭化水素鎖の炭素数として12だけでなく16のGemini型界面活性剤で構成されるナノ粒子を含む凍結保存用組成物で凍結した場合であっても高い生存率を維持していた。また、Gemini型界面活性剤における対イオンを生じさせる元素として臭素ではなく塩素を用いても高い生存率を維持していた。さらに、Gemini型界面活性剤におけるスペーサーの炭素数も2、3いずれも高い生存率を維持していた。
〔試験例7〕
<長期の凍結保存>
実施例3の凍結保存用組成物を試験例4と同様の方法で細胞に導入して凍結処理し、3、6、12ヶ月凍結保存した場合の、解凍後の生存率を試験例4と同様の方法で調べた結果を表8に示す。

実施例3の凍結保存用組成物を試験例4と同様の方法で細胞に導入して凍結処理し、3、6、12ヶ月凍結保存した場合の、解凍後の生存率を試験例4と同様の方法で調べた結果を表8に示す。
表8より、実施例3の凍結保存用組成物を細胞に導入して12ヶ月凍結保存しても凍結解凍後の生存率は87.8%も維持していた。
〔試験例8〕
<組成物の細胞外への排出>
実施例3の凍結保存用組成物を試験例2と同様の方法で細胞に導入して凍結し、解凍後0、1、7、14日後の細胞質と核を含む蛍光強度を定量することで導入した凍結保存用組成物の細胞外への排出を調べた。結果を表9に示す。
<組成物の細胞外への排出>
実施例3の凍結保存用組成物を試験例2と同様の方法で細胞に導入して凍結し、解凍後0、1、7、14日後の細胞質と核を含む蛍光強度を定量することで導入した凍結保存用組成物の細胞外への排出を調べた。結果を表9に示す。
表9より、解凍後7日では蛍光強度が10分の1以下になり、解凍後14日ではほとんど検出されないレベルまで低下していた。したがって、実施例3の凍結保存用組成物で凍結した場合は、解凍後にその凍結保存用組成物が細胞外に排出されることが確認された。
〔試験例9〕
<解凍後の細胞の倍加時間>
実施例3の凍結保存用組成物を、試験例4と同様の方法で細胞に導入して凍結し、解凍後0、7日後の細胞が増殖して総数が2倍に増加する時間(細胞の倍加時間:PDT(Population Doubling Time))を測定した結果を表10に示す。
倍加時間は以下の式で算出した。
PDT (Population Doubling Time)の計算方式:
1)対数的増殖期にある2点において細胞の計測を行った。
2)第1点における細胞密度の測定値をN0とし、第2点における測定値をNとした。
また培養開始から第1点までの時間をt0とし、第2点までの時間をtとした。
PDT= (t-t0)log2 /( logN-logN0)
<解凍後の細胞の倍加時間>
実施例3の凍結保存用組成物を、試験例4と同様の方法で細胞に導入して凍結し、解凍後0、7日後の細胞が増殖して総数が2倍に増加する時間(細胞の倍加時間:PDT(Population Doubling Time))を測定した結果を表10に示す。
倍加時間は以下の式で算出した。
PDT (Population Doubling Time)の計算方式:
1)対数的増殖期にある2点において細胞の計測を行った。
2)第1点における細胞密度の測定値をN0とし、第2点における測定値をNとした。
また培養開始から第1点までの時間をt0とし、第2点までの時間をtとした。
PDT= (t-t0)log2 /( logN-logN0)
表10より、解凍から7日経過しても、細胞の倍加時間は維持されていた。したがって、実施例3の凍結保存用組成物で凍結しても、細胞の増殖活性は凍結保存による影響を受けていないことが確認された。
〔試験例10〕
<胚、精子、血液細胞、3Dオルガノイドの凍結後の生存率>
実施例3の凍結保存用組成物を、試験例4と同様の方法で細胞に導入した。細胞種としてヒト脂肪組織由来間葉系幹細胞と共に胚、精子、血液細胞、3Dオルガノイドを用いた。そして、試験例4と同様の方法で凍結後の生存率を調べた結果を表11に示す。
使用した細胞は下記のとおりである。
・胚(種:マウス、系統:C57BL/6J Jcl、ステージ:2cell、胚の個数:30個、購入元:国立研究開発法人 医薬基盤・健康・栄養研究所)
・精子(種:マウス、系統:C57BL/6NCr Slc、個数:1ストロー、購入元:国立研究開発法人 医薬基盤・健康・栄養研究所)
・血液細胞(種:ヒト、由来:急性単球性白血病患者末梢血、種類:単球、購入元:ATCC)
・3Dオルガノイド(種:ヒト、由来:健常脂肪組織、種類:間葉系幹細胞、Corning(登録商標) マトリゲル基底膜マトリックス オルガノイド形成用を用いたオルガノイド培養、購入元:LONZA社)
<胚、精子、血液細胞、3Dオルガノイドの凍結後の生存率>
実施例3の凍結保存用組成物を、試験例4と同様の方法で細胞に導入した。細胞種としてヒト脂肪組織由来間葉系幹細胞と共に胚、精子、血液細胞、3Dオルガノイドを用いた。そして、試験例4と同様の方法で凍結後の生存率を調べた結果を表11に示す。
使用した細胞は下記のとおりである。
・胚(種:マウス、系統:C57BL/6J Jcl、ステージ:2cell、胚の個数:30個、購入元:国立研究開発法人 医薬基盤・健康・栄養研究所)
・精子(種:マウス、系統:C57BL/6NCr Slc、個数:1ストロー、購入元:国立研究開発法人 医薬基盤・健康・栄養研究所)
・血液細胞(種:ヒト、由来:急性単球性白血病患者末梢血、種類:単球、購入元:ATCC)
・3Dオルガノイド(種:ヒト、由来:健常脂肪組織、種類:間葉系幹細胞、Corning(登録商標) マトリゲル基底膜マトリックス オルガノイド形成用を用いたオルガノイド培養、購入元:LONZA社)
表11より、実施例3の凍結保存用組成物を用いれば間葉系幹細胞だけでなく、胚、精子、血液細胞等の細胞や3Dオルガノイドを凍結した場合であっても、生存率を高く維持できることが確認された。
〔試験例11〕
<免疫不全マウスにおける移植効率の評価>
実施例8の凍結保存用組成物を、試験例4と同様の方法で細胞に導入し、凍結及び解凍した間葉系幹細胞用いて、免疫不全マウスにおける移植効率の評価を行った。
健常者脂肪組織由来間葉系幹細胞(ADSC)は、スフェロイド培養用マトリゲル及び専用培地に付属する増殖因子を添加して、移植1週間前からスフェロイド状に培養した。幹細胞スフェロイドを、移植用のマトリゲルに1対1の割合で混合した。1×107cellsのADSCを含む移植用の混合液を氷上でシリンジに充填し、免疫不全マウスの背部皮下に移植した。移植し2週間後の移植部位の縦横高さをノギスで計測し、体積(mm3)を算出した。また、ADSCを移植し3週間後に摘出した移植片の重量(mg)を精密電子天秤を用いて量った。コントロールとして、実施例8の組成物なしで凍結したADSCを用いた。結果を表12に示す。
<免疫不全マウスにおける移植効率の評価>
実施例8の凍結保存用組成物を、試験例4と同様の方法で細胞に導入し、凍結及び解凍した間葉系幹細胞用いて、免疫不全マウスにおける移植効率の評価を行った。
健常者脂肪組織由来間葉系幹細胞(ADSC)は、スフェロイド培養用マトリゲル及び専用培地に付属する増殖因子を添加して、移植1週間前からスフェロイド状に培養した。幹細胞スフェロイドを、移植用のマトリゲルに1対1の割合で混合した。1×107cellsのADSCを含む移植用の混合液を氷上でシリンジに充填し、免疫不全マウスの背部皮下に移植した。移植し2週間後の移植部位の縦横高さをノギスで計測し、体積(mm3)を算出した。また、ADSCを移植し3週間後に摘出した移植片の重量(mg)を精密電子天秤を用いて量った。コントロールとして、実施例8の組成物なしで凍結したADSCを用いた。結果を表12に示す。
表12より、実施例8の凍結保存用組成物を処置した細胞はマウスに移植した場合であっても生理活性を維持しており、移植片の増大が確認された。
〔試験例12〕
<がん原性のin vitro評価>
実施例3の凍結保存用組成物を用いて、安全性の指標としてがん原性のin vitro評価 soft-agar colony formation assayを行った。
1.8 % agar溶液をオートクレーブにて滅菌して溶解し、45℃のwater bathで保温した。
bottom mix(2×DMEM 37.5ml、血清9ml、2.8%NaHCO3 5.25ml、10×ペニシリン/ストレプトマイシン0.9ml)を作製し、45℃のwater bathで保温した。
十分に保温したbottom mixと1.8% agar溶液を滅菌した試薬ビンまたはチューブ中で1.4vol:1volの割合で2.5ml×必要枚数分+α混和し、45℃のwater bathで保温した(Final 0.75% agra)。
<がん原性のin vitro評価>
実施例3の凍結保存用組成物を用いて、安全性の指標としてがん原性のin vitro評価 soft-agar colony formation assayを行った。
1.8 % agar溶液をオートクレーブにて滅菌して溶解し、45℃のwater bathで保温した。
bottom mix(2×DMEM 37.5ml、血清9ml、2.8%NaHCO3 5.25ml、10×ペニシリン/ストレプトマイシン0.9ml)を作製し、45℃のwater bathで保温した。
十分に保温したbottom mixと1.8% agar溶液を滅菌した試薬ビンまたはチューブ中で1.4vol:1volの割合で2.5ml×必要枚数分+α混和し、45℃のwater bathで保温した(Final 0.75% agra)。
35mmディッシュ又は6ウェルプレートにディスポのピペットを用いて2.5mlずつ上記のbottom agarを注ぎ、4℃で8分間静置し固めた後インキュベーターにて保温した。
十分に保温したtop mix(上層:2×DMEM 37.5ml、血清9ml、2.8%NaHCO3 5.25ml、10×ペニシリン/ストレプトマイシン0.9ml、滅菌水19.5ml)と1.8% agar溶液を滅菌した試薬ビンまたはチューブ中で4vol:1volの割合で1.5ml×必要枚数分+α混和し、45℃のwater bathで保温した(Final 0.36%agra)。
ヒト脂肪組織由来間葉系幹細胞を通常の手順ではがした後、セルストレイナーを通すことでシングルセルとし、2×105cells/mlに調整した。
細胞懸濁液 100μlを 15 mlチューブに移し、ディスポのピペットを用いてtop agarを 3ml/tube加えて泡立たないようにピペッティングし、1.5 mlずつbottom agarの上に播いた。
4℃で8分間静置し固めた後、インキュベーターにて保温した。
2週間後にコロニーを染色し、カウントした。結果を表13に示す。
十分に保温したtop mix(上層:2×DMEM 37.5ml、血清9ml、2.8%NaHCO3 5.25ml、10×ペニシリン/ストレプトマイシン0.9ml、滅菌水19.5ml)と1.8% agar溶液を滅菌した試薬ビンまたはチューブ中で4vol:1volの割合で1.5ml×必要枚数分+α混和し、45℃のwater bathで保温した(Final 0.36%agra)。
ヒト脂肪組織由来間葉系幹細胞を通常の手順ではがした後、セルストレイナーを通すことでシングルセルとし、2×105cells/mlに調整した。
細胞懸濁液 100μlを 15 mlチューブに移し、ディスポのピペットを用いてtop agarを 3ml/tube加えて泡立たないようにピペッティングし、1.5 mlずつbottom agarの上に播いた。
4℃で8分間静置し固めた後、インキュベーターにて保温した。
2週間後にコロニーを染色し、カウントした。結果を表13に示す。
表13より、保存期間が0、3、6ヶ月のいずれもコロニーは生じず、実施例3の凍結保存用組成物を細胞に導入することによるがん原性は確認されなかった。
Claims (8)
- 両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子を含む、細胞若しくは生体組織の凍結保存用組成物。
- 前記ナノ粒子が、カチオン性ナノ粒子である、請求項1に記載の凍結保存用組成物。
- 前記両親媒性分子が、Gemini型界面活性剤であることを特徴とする、請求項1又は2に記載の凍結保存用組成物。
- 前記ナノ粒子のゼータ電位が、10mV以上50mV以下である、請求項1又は2に記載の凍結保存用組成物。
- 前記ナノ粒子の体積平均粒子径が、30nm以上300nm以下である、請求項1又は2に記載の凍結保存用組成物。
- 前記ナノ粒子が、フェルラ酸又は活性化ビタミンCを内包する、請求項1又は2に記載の凍結保存用組成物。
- 細胞若しくは生体組織と請求項1~6のいずれか1項に記載の凍結保存用組成物とを混合してナノ粒子を細胞若しくは生体組織内に導入する工程と、
前記ナノ粒子が導入された細胞又は生体組織を凍結する工程と、
を含む、細胞又は生体組織の凍結保存方法。 - 両親媒性分子を構成成分とし、前記両親媒性分子が単層又は二層のナノ粒子を含む、凍結した細胞又は生体組織。
