콜라겐을 분해하는 효소 중 하나인 콜라겐 분해 효소-1(matrix metalloproteinases-1, MMP-1)의 함량은 Human Pro-MMP-1 Quantikin ELISA kit를 사용하여 제조사의 프로토콜에 따라 측정하였다. 세포 배양이 끝난 후, 배지를 회수하여 Immuno-well plate에 항체 칵테일(antibody cocktail)과 함께 분주하고 실온에서 2시간 동안 반응하였다. 세정 버퍼(wash buffer)로 3번 세척 후, Pro-MMP-1 Conjugate를 첨가하고 실온에서 2시간 동안 반응하였다. 세정 버퍼로 3번 세척 후, 기질 용액(substrate solution)을 분주하고 빛이 차단된 조건에서 20분간 반응하였다. 20분 후, 정지액(stop solution)을 넣어 반응을 중지시켰다. VersaMaxTM tunable microplate reader를 이용하여 540 nm의 파장에서 흡광도를 측정하였으며, 결과는 도 2에 도시하였다.The content of collagen degrading enzyme-1 (matrix metalloproteinases-1, MMP-1), one of the enzymes that degrade collagen, was measured using the Human Pro-MMP-1 Quantikin ELISA kit according to the manufacturer's protocol. After the cell culture was completed, the culture medium was collected, dispensed with an antibody cocktail to an immuno-well plate, and reacted at room temperature for 2 hours. After washing three times with wash buffer, Pro-MMP-1 Conjugate was added and reacted at room temperature for 2 hours. After washing three times with a washing buffer, a substrate solution was dispensed and reacted for 20 minutes under conditions where light was blocked. After 20 minutes, stop solution was added to stop the reaction. Absorbance was measured at a wavelength of 540 nm using a VersaMaxTM tunable microplate reader, and the results are shown in FIG. 2 .
도 2에 나타난 바와 같이, 비교예에 비하여 실시예에 따른 조성물을 처리한 경우 MMP-1의 함량이 더 저하함을 확인하였다. 특히, 실시예 1 및 2는 아무것도 처리하지 않은 세포에 자외선을 조사한 경우에 비하여 MMP-1이 1/5로 크게 감소하였다.As shown in Figure 2, it was confirmed that the content of MMP-1 was further reduced when the composition according to the Example was treated compared to the Comparative Example. In particular, in Examples 1 and 2, MMP-1 was greatly reduced to 1/5 compared to the case where UV rays were irradiated to untreated cells.
상기와 같은 결과로부터 확인할 수 있는 바와 같이, 본 발명에 따른 항광노화 조성물은 콜라겐 함량 증가 및 MMP-1 억제 효과를 상승시킴이 확인된다.As can be confirmed from the above results, it is confirmed that the anti-photoaging composition according to the present invention increases the collagen content and the MMP-1 inhibitory effect.

    3) 상기 2)단계의 농축물을 건조하여 파우더를 제조하는 단계; 및3) preparing a powder by drying the concentrate of step 2); and
