Abstract
본 발명은 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 조성물을 제공한다. 본 발명의 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 조성물은 화장료 조성물 또는 약학 조성물일 수 있다. 본 발명의 조성물은 발모촉진 또는 탈모방지 효과가 뛰어나다.
Description
본 발명은 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 조성물에 관한 것이다.
또한, 본 발명은 상기 조성물을 포함하는 발모촉진 또는 탈모방지용 화장료 조성물, 및 발모촉진 또는 탈모방지용 약학 조성물에 관한 것이다.
사람의 탈모주기는 성장기(anagen), 퇴행기(catagen) 및 휴지기(telogen)로 구분된다. 퇴행기에는 모발 생성이 느려져 세포분열 및 성장이 멈춘다. 휴지기에는 모낭과 모유두가 완전히 분리되어 모낭은 위축되고 모근은 더욱 위쪽으로 올라가 머리카락이 빠지게 된다. 휴지기가 끝난 후 다시 성장기로 들어가면 새로운 모발의 모유두가 만들어지면서 휴지기에 있던 모발은 밀려나서 완전히 두피 밖으로 빠져나오게 된다.
대략 3~6년 동안 모발의 성장과 탈모가 주기적으로 발생하게 되는데, 탈모가 진행되고 있는 사람은 성장기가 짧아지고 휴지기가 긴 모발주기를 가지게 되어 전체 모발에서 성장기 모발의 비중이 감소하게 된다. 발모 및 탈모의 정확한 기전은 현재까지 완전하게 밝혀져 있지 않으나 탈모 유발 원인으로는 남성호르몬 관여에 의한 모포 기능의 저하, 모포 및 모구부의 신진대사 기능의 저하, 두피 긴장에 의한 국소 혈류 장애, 영양불량, 스트레스, 약물에 의한 부작용, 유전적 요인, 자가면역, 국소감염, 화학물질, 백혈병, 결핵 등의 질병과 모발제품의 남용 등으로 매우 다양하다.
미국 FDA(Food and Drug Administration)에서 승인된 대표적인 모발성장을 촉진하는 약물로서 미녹시딜(Minoxidil)과 피나스테리드(Finasteride)가 있다.
미녹시딜에 의한 모발성장효과는 6개월에서 12개월 이후에 나타나며 도포를 중단하면 급속하게 탈모가 진행되어 치료이전 상태로 돌아가고 장기간 사용할 경우 두피의 가려움증, 홍반, 표피 벗겨짐, 건성화를 동반한 피부염, 알레르기성 접촉성 피부염 및 지루성 피부염을 일으킬 수 있다. 또한 미녹시딜을 사용할 경우 급격한 몸무게 증가, 부종, 심장 박동수 증가, 협심증 및 혈액학적 부작용을 초래할 수 있다는 보고가 있다.
피나스테리드(Finasteride)는 FDA에서 허가받은 남성형 탈모치료제로, 테스토스테론(testosterone)이 디하이드로스테론(dihydrosterone)으로 변환할 때 필요한 환원효소인 5α-리덕테이즈 타입1(5α-reductase type1)을 억제한다. 피나스테리드는 육모의 효능은 우수하지만 모발 성장효과를 유지하기 위해서는 평생 동안 지속적으로 복용해야 하는 단점이 있고, 남성의 성기능 장애와 가임 여성의 경우 기형아 출산과 같은 부작용이 있어 안전성의 문제점이 있다. 두타스테리드(Dutasteride)는 5α-리덕테이즈 타입1(5α-reductase type1)과 5α-리덕테이즈 타입2(5α-reductase type 2)를 모두 억제하여 남성호르몬 차단에 더욱 효과적이다. 그러나, 남성호로몬 억제제의 경우, 약물 복용 후 1년 동안은 약 60% 환자에서 탈모가 개선되나 5년 이상 장기 치료시 개선율이 감소하고, 탈모 개선에 변화가 없는 환자의 비율은 30% 이상인 것으로 알려져 있다. 또한, 남성 호르몬 억제제는 성기능 장애와 자살위험도가 증가하는 부작용이 보고되고 있다.
여성형 탈모 치료제로는 스피로노락톤이 대표적이며 항안드로겐제제로 여성 탈모 환자를 대상으로 국내에서 주로 사용되고 있으나, 뚜렷한 탈모 치료 효과는 확인할 수 없고, 대표적으로 고칼슘혈증을 유발하는 부작용이 있다. 따라서 이러한 문제점을 최소화하면서도 안전하게 사용할 수 있는 새로운 탈모 방지 또는 발모 촉진 물질 개발에 대한 관심이 높아지고 있다.
최근에는 천연물을 이용한 발모 또는 탈모방지제 개발에 대한 연구가 활발하다. 이러한 천연물을 원료로 하는 발모 또는 탈모방지제의 경우, 천연 추출물 내의 유효성분 함량이 적은 관계로 발모 또는 탈모방지 효과를 얻기 위해서는 많은 양의 사용이 필요하고, 이들 중 대부분은 천연물 소재라는 점을 마케팅에 활용하고 있을 뿐 발모 또는 탈모방지의 실질적 효능에 대해서는 과학적 연구가 좀더 필요한 상황이다. 또한 식물 추출물을 유효성분으로 포함한 발모 또는 탈모방지제는 두피에 도포한 후 용액이 증발하는 과정에서 이물감이 발생할 수 있고, 효과 지속시간이 짧은 문제가 있다.
한편, 최근 세포 분비물(secretome)에 세포의 행동(behavior)을 조절하는 다양한 생체활성인자가 포함되어 있다는 연구가 보고되고 있으며, 특히 세포 분비물 내에는 세포 간 신호전달 기능을 갖는 '엑소좀(exosome)'이 포함되어 있어 그 성분과 기능에 대한 연구가 활발히 진행 중에 있다.
세포는 세포외 환경에 다양한 막(membrane) 유형의 소포체를 방출하는데, 통상 이러한 방출 소포체들을 세포외 소포체(Extracellular vesicles, EVs)라고 부르고 있다. 세포외 소포체는 세포막 유래 소포체, 엑토좀(ectosomes), 쉐딩 소포체(shedding vesicles), 마이크로파티클(microparticles), 엑소좀 등으로 불려지기도 하며, 경우에 따라서는 엑소좀과는 구별되어 사용되기도 한다.
엑소좀은 세포막의 구조와 동일한 이중인지질막을 갖는 수십 내지 수백 나노미터 크기의 소포체로서 내부에는 엑소좀 카고(cargo)라고 불리는 단백질, 핵산(mRNA, miRNA 등) 등이 포함되어 있다. 엑소좀 카고에는 광범위한 신호전달 요소들(signaling factors)이 포함되며, 이들 신호전달 요소들은 세포 타입에 특이적이고 분비세포의 환경에 따라 상이하게 조절되는 것으로 알려져 있다. 엑소좀은 세포가 분비하는 세포 간 신호전달 매개체로서 이를 통해 전달된 다양한 세포 신호는 표적 세포의 활성화, 성장, 이동, 분화, 탈분화, 사멸(apoptosis), 괴사(necrosis)를 포함한 세포 행동을 조절한다고 알려져 있다. 엑소좀은 유래된 세포의 성질 및 상태에 따라 특이적인 유전물질과 생체활성 인자들이 포함되어 있다. 증식하는 줄기세포 유래 엑소좀의 경우 세포의 이동, 증식 및 분화와 같은 세포 행동을 조절하고, 조직 재생과 관련된 줄기세포의 특성이 반영되어 있다(Nature Review Immunology 2002 (2) 569-579).
즉, 세포의 아바타라고 불리는 엑소좀은 세포와 유사하게 성장인자와 같은 생체활성 인자들이 포함되어 있는데, 생체활성 인자들을 세포와 세포 간에 실어 나르는 전달체 역할, 즉, 세포와 세포 간의 교신 역할을 한다. 엑소좀은 줄기세포, 면역세포, 섬유아세포 및 암세포 등의 동물세포로부터 방출될 뿐만 아니라 식물, 세균(bacteria), 균류(fungi), 조류(algae) 등 다양한 생물의 세포로부터도 방출되는 것으로 알려져 있다. 예를 들어, 엑소좀은 암세포, 면역세포, 중간엽 줄기세포 등의 배양액 외에도 식물세포 내지는 식물줄기세포 배양액으로부터도 분리될 수 있다.
그러나 식물세포 내지는 식물줄기세포로부터 유래된 엑소좀의 분리·정제 및 특성분석에 대한 연구는 아직 초기 단계이며, 이들 중 대부분은 단순히 식물착즙의 여과물 내에 혼합물로 혼재되어 있는 세포외 소포체를 엑소좀이라고 하면서 마케팅에 활용하고 있는 수준이다. 따라서 식물세포로부터 유래된 엑소좀에 대한 보다 면밀한 특성분석 및 기능에 대한 연구가 필요하다.
장미는 북반구의 한대, 아한대, 온대, 및 아열대 등 넓은 지역에서 다양한 품종이 재배되고 있고, 그 추출물은 향수나 화장품 등에 사용되고 있다. 그러나 현재까지의 기술은 장미의 꽃잎을 건조하여 열수 추출하거나 용매 추출하여 얻은 추출물을 향수, 방향제 등에 사용하고 있는 수준이다. 또한, 장미 캘러스 배양물에는 생장조절물질이나 캘러스 유도물질이 포함되어 있어 천연물질의 화장품 원료라고 하기 어렵고 배양물 내에 포함된 생장조절물질 등이 피부 트러블 등의 부작용을 일으킬 우려가 있다.
본 발명자는 장미 줄기세포 유래의 엑소좀이 발모촉진 또는 탈모방지 등에 효능이 있는 것을 확인하고, 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 화장료 조성물, 및 발모촉진 또는 탈모방지용 약학 조성물을 개발하였다.
한편, 상기한 배경기술로서 설명된 사항들은 본 발명의 배경에 대한 이해 증진을 위한 것일 뿐, 본 발명의 "선행 기술"로서 이용될 수 있다는 승인으로서 인용한 것은 아님을 이해하여야 한다.
본 발명의 목적은 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 조성물을 제공하는데 있다.
또한, 본 발명의 목적은 상기 조성물을 포함하는 발모촉진 또는 탈모방지용 화장료 조성물, 및 발모촉진 또는 탈모방지용 약학 조성물을 제공하는데 있다.
그러나, 전술한 바와 같은 본 발명의 과제는 예시적인 것으로, 이에 의해 본 발명의 범위가 제한되는 것은 아니다. 또한, 본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명은 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 조성물을 제공한다.
본 명세서에서 용어, "장미(Rosa spp.)"는 쌍떡잎 식물군, 장미목, 장미과 및 장미속에 속하는 식물을 총칭하며, 야생종과 개량을 가한 원예종을 모두 포함한다.
본 명세서에서 용어, "엑소좀(exosomes)"은 식물세포 또는 식물줄기세포로부터 세포외 공간으로 분비 또는 방출된 막 구조를 갖는 나노 사이즈의 소낭을 의미하며, 엑소좀-유사 베지클 또는 엑소좀-유사 파티클로도 정의된다.
본 발명의 명세서에서 사용된 "탈모"란 용어는 모발이 완전히 두피 밖으로 빠져나오는 현상을 의미한다. 또한 "발모"란 두피에서 모발이 나는 것을 의미하고, "양모"와 실질적으로 동일한 의미로 사용된다. 본 발명의 명세서에서 사용된 "모발"이란 용어는 체모(體毛)와 두발(頭髮)을 포괄하는 것으로서 헤어(hair)라는 용어와 실질적으로 동등한 의미이다. 또한 모발은 사람 및 동물의 것을 모두 포함한다.
본 명세서에서 용어, "이온토포레시스(iontophoresis)"는 유효물질이 적용된 피부에 미세전류를 흐르게 하여 전위차를 주어 피부의 전기적 환경을 변화시킴으로써 이온화된 유효성분을 전기적 반발력으로 피부를 투과하게 하는 방법을 의미한다. 본 발명의 일 구체예에 사용되는 이온토포레시스(iontophoresis)는 피부 위의 전극 패치에 외부전원으로부터의 전류가 흘러들어가 피부에 미세전류가 도입되는 방식, 전극 패치 자체에 배터리가 장착되어 피부에 미세전류가 도입되는 방식, 고농도 전해질 용액 및 저농도 전해질 용액 간의 이온 농도 차이를 통해 전류를 발생시키는 역전기투석(Reversed Electrodialysis) 수단이 장착된 패치를 통해 피부에 미세전류가 도입되는 방식 등을 포함할 수 있다. 그러나, 본 발명은 이에 제한되는 것이 아니며, 다양한 방식의 이온토포레시스가 사용될 수 있음은 물론이다.
본 발명의 일 구체예의 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 조성물은 발모촉진 또는 탈모방지 효능 중 적어도 하나의 효능을 나타낼 수 있다.
본 발명의 일 구체예의 피부 보습, 발모촉진 또는 탈모방지용 조성물에 있어서, 상기 장미 줄기세포는 장미의 잎, 줄기, 꽃잎, 뿌리 및/또는 배(胚) 유래 캘러스, 바람직하게는, 꽃잎 유래 캘러스를 유도한 후 세포 배양하여 얻은 것일 수 있다. 그러나, 본 발명은 이에 제한되는 것이 아니며, 장미의 다양한 부위에서 캘러스를 유도하여 사용할 수 있음은 물론이다.
본 발명의 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 조성물은 화장료 조성물 또는 약학 조성물일 수 있다.
본 발명은 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 화장료 조성물을 제공한다. 예를 들어, 상기 화장료 조성물은 샴푸, 비누, 린스, 계면활성제-함유 클렌징, 크림, 로션, 연고, 토닉, 트리트먼트, 컨디셔너, 현탁액, 유탁액, 페이스트, 젤, 오일, 왁스, 스프레이, 에어졸, 미스트, 또는 파우더일 수 있고, 바람직하게는 로션이나 크림일 수 있다.
또한, 본 발명은 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 발모촉진 또는 탈모방지용 약학 조성물을 제공한다.
본 발명을 한정하지 않는 예시로서, 본 발명의 일 구체예의 약학 조성물은 주사, 마이크로니들링, 이온토포레시스, 도포, 또는 이들의 조합에 의해 투여 또는 처리될 수 있다. 예를 들어, 상기 약학 조성물은 주사제형, 주입제형, 분무제형, 액상제형 또는 패취제형일 수 있다.
본 발명의 일 구체예의 조성물이 약학 조성물로 사용되는 경우 약학적으로 허용 가능한 담체, 부형제 또는 희석제 등을 포함할 수 있다. 상기 담체, 부형제 및 희석제로는 락토오스, 덱스트로오스, 트레할로오스, 수크로오스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 카보네이트, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스(microcrystalline cellulose), 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 미네랄 오일 등을 들 수 있으며, 이에 제한되지 않는다. 또한, 본 발명의 일 구체예의 약학 조성물의 유효량은 발모촉진 또는 탈모방지 효과를 기대하기 위하여 투여에 요구되는 양을 의미한다.
본 발명의 일 구체예의 약학 조성물의 배합비율은 전술한 바와 같은 추가 성분들의 종류나 양, 형태 등에 따라서 적당하게 선택할 수 있다. 예를 들어, 주사제 전량에 대해, 본 발명의 약학 조성물은 약 0.1 내지 99 중량%, 바람직하게는 약 10 내지 90 중량% 정도 포함될 수 있다. 또한, 본 발명의 일 구체예의 약학 조성물의 적합한 투여량은 질환의 경중, 제형의 종류, 제제화 방법, 환자의 연령, 성별, 체중, 건강 상태, 식이, 배설률, 투여 시간 및 투여 방법에 따라 조절될 수 있다. 예를 들어, 성인에게 본 발명의 일 구체예의 약학 조성물을 투여하는 경우, 하루에 0.001 mg/kg ~ 100 mg/kg의 용량으로 1 내지 수회에 나누어 투여할 수 있다.
한편, 본 발명의 일 구체예의 조성물이 화장료 조성물로 제조되는 경우, 본 발명의 효과를 손상하지 않는 범위내에서 통상 화장료 조성물에 이용되는 성분, 예를 들면 보습제, 산화방지제, 유성성분, 자외선 흡수제, 유화제, 계면활성제, 증점제, 알콜류, 분말성분, 색재, 수성성분, 물, 각종 피부영양제 등을 필요에 따라 적절히 배합할 수 있다.
또한, 본 발명의 일 구체예의 화장료 조성물은 장미 줄기세포 유래의 엑소좀 이외에, 그 작용(예를 들어, 발모촉진 또는 탈모방지 등)을 손상시키지 않는 한도에서 종래부터 사용된 발모촉진제, 탈모방지제 및/또는 보습제를 함께 혼합하여 사용할 수 있다.
본 발명의 일 구체예의 화장료 조성물은 예를 들면, 패취, 팩, 시트, 크림, 토닉, 연고, 현탁액, 유탁액, 페이스트, 로션, 젤, 오일, 스프레이, 에어졸, 미스트, 파운데이션, 파우더, 기름 종이 등의 다양한 형태에 적용할 수 있다.
본 발명의 일 구체예의 화장료 조성물은 발모촉진 또는 탈모방지 등의 목적으로 사용될 수 있으며, 화장품 제형은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있다. 예를 들어 패취, 팩, 시트, 유연화장수, 영양화장수, 수렴화장수, 영양크림, 마사지크림, 클렌징크림, 에센스, 클렌징로션, 클렌징폼, 클렌징워터, 선스크린, 비누, 샴푸, 계면활성제-함유 클렌징, 입욕제, 바디로션, 바디크림, 바디오일, 바디에센스, 바디세정제, 염모제, 헤어토닉 등으로 제형화할 수 있으나 이에 한정되는 것은 아니다.
본 발명의 일 구체예의 화장료 조성물은 화장료 조성물에 통상적으로 이용되는 성분들을 포함하며, 예컨대 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제 그리고 담체를 포함할 수 있다. 또한, 화장료 조성물에 대한 각각의 제형에 있어서, 다른 성분들은 화장료 조성물의 종류 또는 사용목적 등에 따라 당업자가 어려움 없이 적의 선정하여 배합할 수 있다.
본 발명의 다른 구체예는 상기 화장료 조성물을 이용하여, 치료용을 제외한 포유동물의 두피 또는 모발의 상태를 조절하는 미용방법을 제공한다. 본 발명의 미용방법에 있어서, 두피의 상태 조절이란 두피의 상태를 개선시키고/시키거나 두피의 상태를 예방적으로 조절하는 것을 의미하고, 두피의 상태 개선이란 두피 조직의 외관 및 느낌의 시각적 및/또는 촉각적으로 지각할 수 있는 긍정적인 변화를 의미한다.
예를 들어, 모발 상태의 개선이란 모발이 풍성하고 모발의 굵기가 가늘지 않으며, 모발이 부드럽고 윤기가 있는 상태일 수 있다.
본 발명의 일 구체예의 미용방법은, (a) 상기 화장료 조성물을 포유동물의 피부, 두피 또는 모발에 직접 도포하는 것, (b) 상기 화장료 조성물이 도포되거나 침적된 패취, 팩 또는 시트를 포유동물의 피부, 두피 또는 모발에 접촉 또는 부착하는 것, 또는 상기 (a) 및 (b)를 순차적으로 진행하는 것을 포함한다. 상기 (a) 단계에서는 화장료 조성물로서 로션이나 크림이 사용될 수 있다.
대안으로, 본 발명의 일 구체예의 미용방법은, (c) 상기 (b) 단계 이후에 상기 패취, 팩 또는 시트를 상기 포유동물의 피부, 두피 또는 모발로부터 제거하고, 상기 화장료 조성물을 포유동물의 피부, 두피 또는 모발에 도포하는 단계를 더 포함할 수 있다. 상기 (c) 단계에서는 화장료 조성물로서 로션이나 크림이 사용될 수 있다.
본 발명의 일 구체예의 미용방법에 있어서, 상기 포유동물은 인간, 개, 고양이, 설치류, 말, 소, 원숭이, 또는 돼지일 수 있다.
또한, 본 발명은 상기 약학 조성물의 치료학적으로 유효한 양을 포유동물에게 투여하거나, 상기 약학 조성물을 피부 또는 두피에 도포하는 단계를 포함하는, 발모촉진 또는 탈모방지를 위한 방법을 제공한다.
본 발명을 한정하지 않는 예시로서, 본 발명의 일 구체예의 발모촉진 또는 탈모방지를 위한 방법에 있어서, 상기 약학 조성물은 주사, 마이크로니들링, 이온토포레시스 또는 이들의 조합에 의해 투여될 수 있다. 예를 들어, 상기 약학 조성물은 주사제형, 주입제형, 분무제형, 액상제형 또는 패취제형일 수 있다.
본 발명의 일 구체예의 발모촉진 또는 탈모방지를 위한 방법에 있어서, 상기 포유동물은 인간, 개, 고양이, 설치류, 말, 소, 원숭이, 또는 돼지일 수 있다.
본 발명의 장미 줄기세포 유래의 엑소좀을 유효성분으로 함유하는 발모촉진 또는 탈모방지용 조성물은 종래의 장미 여과물, 단순 추출물, 캘러스 배양액에 비해 생장조절인자와 같은 불순물 함유 가능성이 낮으며, 장미 줄기세포 배양액 여과물 등에 비해 세포 독성이 적은 장점이 있다. 본 발명의 조성물은 발모촉진 또는 탈모방지 효과가 뛰어나다.
한편, 전술한 바와 같은 효과들에 의해 본 발명의 범위가 제한되는 것은 아니다.

이하 본 발명을 하기 실시예에서 보다 상세하게 기술한다. 다만, 하기 실시예는 본 발명의 내용을 예시하는 것일 뿐 본 발명의 권리범위를 제한하거나 한정하는 것이 아니다. 본 발명의 상세한 설명 및 실시예로부터 본 발명이 속하는 기술분야의 통상의 기술자가 용이하게 유추할 수 있는 것은 본 발명의 권리범위에 속하는 것으로 해석된다. 본 발명에 인용된 참고문헌들은 본 발명에 참고로서 통합된다.
명세서 전체에서, 어떤 부분이 어떤 구성 요소를 "포함"한다고 할 때, 이는 특별히 반대되는 기재가 없는 한 다른 구성 요소를 제외하는 것이 아니라 다른 구성 요소를 더 포함할 수 있는 것을 의미한다.
실시예
실시예 1: 장미 줄기세포의 제조
당업계에 알려진 식물 줄기세포 제조 및 배양 방법에 따라, 장미의 꽃잎, 잎, 줄기, 뿌리, 또는 배(胚) 유래 캘러스를 유도하고, 유도된 장미 캘러스를 세포 배양한다. 그리고, 성장상태가 좋은 캘러스를 선택하고 대량으로 배양하여 장미 줄기세포 배양액을 준비한다.
실시예 2: 장미 줄기세포 유래의 엑소좀의 제조
실시예 1과 같은 방식으로 제조된 장미 줄기세포 배양액(장미 캘러스 배양액)을 바이오스펙트럼 주식회사(대한민국 경기도에 소재하는 Damask Rose 캘러스 배양액 공급업체)에서 입수하였다. 장미 줄기세포 배양액을 0.22 ㎛ 필터로 여과하여 세포 잔해물, 노폐물 및 거대 입자 등의 불순물을 제거해 주었다. 여과된 배양액으로부터, 접선흐름여과장치(Tangential Flow Filtration; TFF)를 이용하여 장미 줄기세포 유래의 엑소좀을 분리하였다.
분리된 장미 줄기세포 유래 엑소좀의 크기와 농도는 NS300(Malvern Panalytical에서 구입)을 이용한 나노입자트래킹분석(nanoparticle tracking analysis; NTA)으로 확인하였다(도 1).
실시예 3: 장미 줄기세포 유래 엑소좀의 인체 유래 모유두세포 전달능의 확인
장미 줄기세포 유래의 엑소좀이 인체 유래 모유두세포 (Human Dermal Papilla Cells; HDPs)(대한민국 서울시 소재 주식회사 세포바이오에서 구입) 내로 전달되는지 여부를 확인하기 위하여 다음과 같은 분석을 수행하였다. 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀의 막을 형광 염색하기 위하여 PKH67(Sigma-Aldrich에서 구입) 형광 염료와 반응시켰다. 반응 후 반응액을 MiniTrap-25(Cytiva에서 구입) 컬럼으로 분획하여 엑소좀 막에 염색되지 않는 유리 PHK67(free PKH67) 형광 염료를 제거하였다. 음성대조군으로는 PKH67 형광 염료를 완충용액과 반응시킨 후 MiniTrap-25 컬럼으로 분획한 것을 사용하였다. PKH67로 염색된 엑소좀을 미리 배양하여 준비된 인체 유래 모유두세포와 함께 배양한 후 시간에 따른 엑소좀의 세포 내 전달 여부를 형광현미경을 이용하여 관찰하였다. 세포의 핵을 염색하기 위하여 훼스트(Hoechst) 형광 염료(Thermo Fisher에서 구입)를 사용하였고, 세포질을 염색하기 위하여 CellMask Orange 형광 염료(Thermo Fisher에서 구입)를 사용하였다. 엑소좀의 세포 내 전달 여부를 확인한 결과, 형광 염색된 엑소좀이 세포 내로 전달되어 녹색 형광이 세포 내부에 시간의 경과에 따라 축적되는 것을 확인하였다(도 2).
실시예 4: 모유두세포의 성장 촉진 효과의 확인
모발을 잘 자라게 하기 위해서는 모유두(papilla)의 역할이 중요하다. 모유두 세포는 발생학적으로 간엽계에서 유래된 세포로서 주위의 상피계 세포의 분열을 조절하는 능력이 있으며, 이러한 기능은 성장 인자에 의해 조절된다고 알려져 있다. 또한 모발을 구성하는 케라틴 단백질은 모근부 각질형성세포(keratinocyte)에서 생성되는데, 이러한 모근부 각질형성세포는 모유두세포로부터 분화된다. 그리고 모근부 각질형성세포는 모유두세포와 상호작용을 하며 모낭의 형성과 모낭 주기의 조절에 있어 중요한 역할을 한다. 따라서 특정 후보 물질 처리 후 모유두세포의 증식이 증가하는지 여부를 분석하면, 후보 물질의 모발 생성 촉진 효능을 평가할 수 있다.
실시예 2와 같이 준비된 장미 줄기세포 유래의 엑소좀이 인체 유래 모유두세포의 증식을 촉진하는지 여부를 평가하기 위하여 CCK-8 어세이를 시행하였다. 우태아 혈청이 포함된 DMEM 배지에 분산된 인체 유래 모유두세포(HDPs)를 96-웰 플레이트에 10,000 세포/웰의 농도로 분주한 후 24시간 배양하였다. 이후 실시예 1에서 준비한 장미 줄기세포 배양액과 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀을 각각 무혈청 배지에 희석한 후 인체 유래 모유두세포에 처리하고 24시간 배양하였다. 모유두세포를 이용한 세포 증식능을 확인하기 위해 실험군을 다음과 같이 분류하였다:
(1) 음성대조군 (Control): 무혈청 배지만 모유두세포에 처리한 실험군;
(2) 양성대조군 (Positive Control): 모유두세포 증식 촉진제인 미녹시딜(Minoxidil)(최종농도: 10μM)을 무혈청 배지에 현탁하여 모유두세포에 처리한 실험군;
(3) 장미 줄기세포 배양액 처리군 {도 3에서 "장미 캘러스 배양액 (Rose-Callus CM)"으로 표시}: 실시예 1에서 준비된 장미 줄기세포 배양액(장미 캘러스 배양액)을 무혈청 배지에 6% 또는 20%가 되도록 희석하여 모유두세포에 처리한 실험군 (처리농도 - 저농도: 웰당 배지 부피의 6%가 되도록 희석하여 처리, 1,815.5 μg/mL에 해당; 고농도: 웰당 배지 부피의 20%가 되도록 희석하여 처리, 6,051.5 μg/mL에 해당);
(4) 장미 줄기세포 유래의 엑소좀 처리군 {도 3에서 "장미 엑소좀 (Rose-Exosome)"으로 표시}: 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀을 무혈청 배지에 6% 또는 20%가 되도록 희석하여 모유두세포에 처리한 실험군 (처리농도 - 저농도: 웰당 배지 부피의 6%가 되도록 희석하여 처리, 53.6 μg/mL에 해당; 고농도: 웰당 배지 부피의 20%가 되도록 희석하여 처리, 178.7 μg/mL에 해당).
상기 각 실험군에서 실험 대상 물질을 인체 유래 모유두세포에 처리 후 24시간 배양한 후, 배양액을 제거하였다. CCK-8 어세이(Dojindo에서 구입) 시약이 10% 함유된 모유두세포 배양 배지를 인체 유래 모유두세포에 처리한 후 5% CO2, 37℃ 조건에서 2시간 동안 인큐베이션하였다. 이후 상층액을 96-웰 플레이트로 옮겨 450nm에서 흡광도를 측정하였다.
그 결과, 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀을 인체 유래 모유두세포에 처리한 경우 음성대조군과 비교하여 농도의존적으로 세포 증식이 증가하였다. 또한, 장미 줄기세포 유래의 엑소좀의 고농도 처리군(20%)에서 양성대조군인 미녹시딜과 유사한 수준의 세포 증식이 확인되었다(도 3).
따라서, 본 발명의 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 조성물은 모유두세포의 증식을 촉진시키고, 이에 따라 모발 생성을 촉진할 수 있다.
실시예 5: 인체 유래 모유두세포에서 모발 성장 인자 발현 촉진능의 확인
ALP(Alkaline Phosphatase)는 대표적인 모발 형성 마커로서, 모발 성장과 관련하여 모발의 성장과 유지에 중요한 역할을 한다. VCAN(versican)은 모낭 형성 마커로서, 모낭 형성 유도, 모발 재생 및 모발 성장 유지에 관여한다. 베타-카테닌(β-catenin)은 Wnt 신호에 따라 세포핵 내로 이동하고 모발 성장에 관여하는 다양한 유전자를 발현시킨다. 따라서 특정 후보 물질을 모유두세포에 처리 후 모유두세포에서 ALP, VCAN, 베타-카테닌(β-catenin)의 유전자 발현량이 증가하는지 여부를 분석하면, 후보 물질의 모발 생성 촉진 효능을 평가할 수 있다.
실시예 2와 같이 준비된 엑소좀이 인체 유래 모유두세포(HDPs)의 모발 성장에 미치는 영향을 확인하기 위하여 리얼타임 PCR을 이용하여 ALP, VCAN, 베타-카테닌(β-catenin)의 유전자 발현량을 확인하였다. 이를 위하여 모유두세포를 6-웰 플레이트에 웰 당 25,000 개의 세포를 분주하여 배양하였다. 이후 실시예 1에서 준비된 장미 줄기세포 배양액과 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀을 각각 우태아혈청이 1% 함유된 DMEM 배지에 희석한 후 모유두세포에 처리하고 48시간 배양하였다. ALP, VCAN, 베타-카테닌(β-catenin)의 유전자 발현량 평가를 위해 실험군을 다음과 같이 분류하였다:
(1) 음성대조군 (Control): 우태아혈청이 1% 함유된 DMEM 배지만 모유두세포에 처리한 실험군;
(2) 양성대조군 (Positive Control): 모유두세포 증식 촉진제인 미녹시딜(Minoxidil)(최종농도: 10μM)을 우태아혈청이 1% 함유된 DMEM 배지에 현탁하여 모유두세포에 처리한 실험군;
(3) 장미 줄기세포 배양액 처리군 {도 4a, 도 4b 및 도 4c에서 "장미 캘러스 배양액 (Rose-Callus CM")으로 표시}: 실시예 1에서 준비된 장미 줄기세포 배양액을 우태아혈청이 1% 함유된 DMEM 배지에 20%가 되도록 희석하여 모유두세포에 처리한 실험군 (처리농도: 웰당 배지 부피의 20%가 되도록 희석하여 처리; 6,051.5 μg/mL에 해당);
(4) 장미 줄기세포 유래의 엑소좀 처리군 {도 4a, 도 4b 및 도 4c에서 "장미 엑소좀 (Rose-Exosome)"으로 표시}: 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀을 우태아혈청이 1% 함유된 DMEM 배지에 6% 또는 20%가 되도록 희석하여 모유두세포에 처리한 실험군 (처리농도- 저농도: 웰당 배지 부피의 6%가 되도록 희석하여 처리, 53.6 μg/mL에 해당; 고농도; 웰당 배지 부피의 20%가 되도록 희석하여 처리, 178.7 μg/mL에 해당).
상기 각 실험군에서 모발 성장에 관여하는 ALP, VCAN 및 베타-카테닌의 발현을 유전자 수준에서 확인하기 위해 리얼타임 PCR을 수행하였으며, 다음과 같은 순서로 실험을 수행하였다.
48시간 배양한 모유두세포는 배양액 제거 후 DPBS로 세척하였다. 이후 RNeasy Mini kit(Qiagen에서 구입)를 이용하여 제조사의 매뉴얼에 따라 RNA를 추출한 후 마이크로플레이트 리더기를 이용하여 RNA를 정량하였다. 정량한 RNA는 PrimeScript RT Master Mix(TaKaRa에서 구입)을 이용하여 cDNA로 합성하였다. 유전자 발현을 비교 분석하기 위하여 합성한 cDNA를 템플릿트로 하고, SYBR Fast qPCR Master Mix(KAPA biosystems에서 구입)을 이용하여 리얼타임 PCR 분석을 수행하였다. 실험에 사용한 프라이머들은 주식회사 코스모진텍(대한민국 서울시 소재)에 의뢰하여 합성한 것을 사용하였다. 시료 내의 ALP, VCAN 및 베타-카테닌의 mRNA 발현량은 GAPDH로 정량하였다. 리얼타임 PCR의 반응 조건은 95℃에서 3분 동안 반응시킨 후, 1 사이클당 95℃ 3초, 60℃ 30초 및 95℃ 1초로 하여 총 40 사이클을 진행하였다. 리얼타임 PCR에 사용한 프라이머의 종류와 서열은 하기의 표 1과 같다.

도 4a에 도시된 바와 같이, 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀 6%를 모유두세포에 처리한 경우, ALP의 mRNA 발현량이 장미 줄기세포 배양액 20%를 모유두세포에 처리한 경우 보다 현저히 높았고, 양성대조군인 미녹시딜을 모유두세포에 처리한 경우의 ALP mRNA 발현량과 유사한 수준이었다. 다음으로, 도 4b에 도시된 바와 같이 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀 6% 및 20% 각각을 모유두세포에 처리한 경우 ALP의 mRNA 발현량이 음성대조군, 양성대조군 및 장미 줄기세포 배양액 20% 각각을 모유두세포에 처리한 경우의 ALP mRNA 발현량에 비해 농도의존적으로 현저히 높았다. 또한, 도 4c에 도시된 바와 같이 실시예 2에서 준비된 장미 줄기세포 유래의 엑소좀 6%를 모유두세포에 처리한 경우 ALP의 mRNA 발현량이 음성대조군, 양성대조군 및 장미 줄기세포 배양액 20% 각각을 모유두세포에 처리한 경우 보다 현저히 높았다. 따라서, 본 발명의 장미 줄기세포 유래의 엑소좀은 발모촉진 또는 탈모방지를 위한 화장료 조성물 및 약학 조성물의 유효성분으로 유용하게 사용될 수 있다.
이상, 본 발명을 상기 실시예를 들어 설명하였으나, 본 발명은 이에 제한되는 것이 아니다. 당업자라면 본 발명의 취지 및 범위를 벗어나지 않고 수정, 변경을 할 수 있으며 이러한 수정과 변경 또한 본 발명에 속하는 것임을 알 수 있을 것이다.
Claims (15)
- 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는, 발모촉진 또는 탈모방지용 화장료 조성물.
- 제1항에 있어서,샴푸, 비누, 린스, 계면활성제-함유 클렌징, 크림, 로션, 연고, 토닉, 트리트먼트, 컨디셔너, 현탁액, 유탁액, 페이스트, 젤, 오일, 왁스, 스프레이, 에어졸, 미스트, 또는 파우더인, 발모촉진 또는 탈모방지용 화장료 조성물.
- 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는, 발모촉진 또는 탈모방지용 약학 조성물.
- 제3항에 있어서,주사, 마이크로니들링, 이온토포레시스, 도포, 또는 이들의 조합에 의해 투여 또는 처리되는, 발모촉진 또는 탈모방지용 약학 조성물.
- 제3항에 있어서,주사제형, 주입제형, 분무제형, 액상제형 또는 패취제형인, 발모촉진 또는 탈모방지용 약학 조성물.
- 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 화장료 조성물을 이용하여, 치료용을 제외한 포유동물의 두피 또는 모발의 상태를 조절하는 미용방법.
- 제6항에 있어서,(a) 상기 화장료 조성물을 포유동물의 피부, 두피 또는 모발에 직접 도포하는 것, (b) 상기 화장료 조성물이 도포되거나 침적된 패취, 팩 또는 시트를 포유동물의 피부, 두피 또는 모발에 접촉 또는 부착하는 것, 또는 상기 (a) 및 (b)를 순차적으로 진행하는 것을 포함하는, 미용방법.
- 제7항에 있어서,상기 (a) 단계에서는 화장료 조성물로서 로션이나 크림이 사용되는, 미용방법.
- 제7항 또는 제8항에 있어서,(c) 상기 (b) 단계 이후에 상기 패취, 팩 또는 시트를 상기 포유동물의 피부, 두피 또는 모발로부터 제거하고, 상기 화장료 조성물을 포유동물의 피부, 두피 또는 모발에 도포하는 단계를 더 포함하는, 미용방법.
- 제9항에 있어서,상기 (c) 단계에서는 화장료 조성물로서 로션이나 크림이 사용되는, 미용방법.
- 제6항 내지 제8항 중 어느 한 항에 있어서,상기 포유동물은 인간, 개, 고양이, 설치류, 말, 소, 원숭이, 또는 돼지인, 미용방법.
- 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 약학 조성물의 치료학적으로 유효한 양을 인간을 제외한 포유동물에게 투여하는 단계, 또는 장미 줄기세포 유래의 엑소좀을 유효성분으로 포함하는 약학 조성물을 인간을 제외한 포유동물의 피부 또는 두피에 도포하는 단계를 포함하는, 발모촉진 또는 탈모방지를 위한 방법.
- 제12항에 있어서,상기 약학 조성물은 주사, 마이크로니들링, 이온토포레시스 또는 이들의 조합에 의해 투여되는, 발모촉진 또는 탈모방지를 위한 방법.
- 제12항에 있어서,상기 약학 조성물은 주사제형, 주입제형, 분무제형, 액상제형 또는 패취제형인, 발모촉진 또는 탈모방지를 위한 방법.
- 제12항 내지 제14항 중 어느 한 항에 있어서,상기 포유동물은 개, 고양이, 설치류, 말, 소, 원숭이, 또는 돼지인, 발모촉진 또는 탈모방지를 위한 방법.
