망막증(Retinophathy)은 눈(eye)의 망막에 손상에 의한 시력의 저하 및 손실을 의미한다. 망막증의 원인은 증식성 및 비증식성 혈관형성에 의하며, 기작은 다르지만 모두 망막중앙정맥의 비정상적인 혈류의 변화에 의한다. 증식성 망막증은 비정상적 혈관성장에 의한 혈관의 과도성장 및 새로운 혈관의 과도한 생성(neovascularizaton)에 의한 혈류의 변화에 의한 망막 손상이고, 비증식성 망막증은 혈관의 직접적 손상에 의한 비정상적 혈류 때문으로 여겨지고 있다. 일반적인 망막증과 연계하여, 황반변성은 퇴행성신경증의 하나로 여겨지며 완전한 실명으로 인한 정신적인 손상도 동반하여 50이상 인구층에 매우 중요한 질환으로 인식되고 있다. 황반변성증 중 습식(Wet) AMD는 비정상적인 혈관성장(abnormal boold vessel growth, choroidal neovascularization)에 의한 병증으로 VEGF에 의한 혈관생성 촉진에 의한 것으로 알려져 있다.
통상 과도한 혈관생성 및 생장에 의하여 촉발되는 황반변성은 시력의 손실을 가져오므로 예방에 주력하고 있는 실정이다. 치료의 경우, 황반변성에서 특이적 혈관생성을 유도하는 VEGF에 대한 억제제를 치료제로 사용하고 있으나, 기존의 치료제들은 여러가지 한계에 봉착해 있는 실정이다.
본 발명자는 노인황반변성(AMD)과 같은 혈관신생성 질환 관련 프로테오믹스 분석을 통해 바이오마커들의 작용 메커니즘을 규명하여 해당 질환의 치료에 효능을 가질 수 있는 새로운 타겟 단백질 물질을 스크리닝함으로써 본 발명을 완성하였다.
이에, 본 발명이 해결하고자 하는 과제는 노인황반변성과 같은 혈관신생성 질환들을 효과적으로 예방 및 치료할 수 있는 약학적 조성물을 제공하는 것이다.
본 발명이 해결하고자 하는 다른 과제는 상기 혈관신생성 질환들을 효과적으로 개선할 수 있는 건강기능식품 조성물을 제공하는 것이다.
본 발명의 과제들은 이상에서 언급한 기술적 과제로 제한되지 않으며, 언급되지 않은 또 다른 기술적 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기 과제를 해결하기 위한 본 발명의 일 실시예에 따른 혈관신생성 질환의 예방 또는 치료용 약학적 조성물은 SHC1 (Src homology 2 domain-containing-transforming protein C1) 단백질을 포함한다. SHC1 단백질은 유효성분으로서 포함될 수 있다.
SHC1 단백질은 서열번호 1의 아미노산 서열과 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 또는 99% 이상의 서열 상동성을 갖는 단백질일 수 있다. 또는, SHC1 단백질은 서열번호 1의 아미노산 서열을 포함하는 단백질일 수 있다. 바람직하게는, SHC1 단백질은 서열번호 1의 아미노산 서열과 동일한 서열을 갖는 단백질일 수 있다.
다른 예시적인 실시예에서, SHC1 단백질은 서열번호 1의 아미노산 서열을 갖는 단백질일 수 있다. 상기 서열번호 1의 아미노산 서열을 갖는 단백질이란 그 기능적 동등물도 포함한다. 상기 “기능적 동등물"은 아미노산의 부가, 치환 또는 결실의 결과, 상기 서열번호 1의 각각의 아미노산 서열과 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 또는 99% 이상의 서열 상동성을 가지는 것으로, 상기 서열번호 1의 아미노산 서열을 갖는 단백질과 실질적으로 동질의 생리활성을 가지는 것을 의미한다. 예를 들어, 상기 서열 상동성을 갖는 단백질은 상기 서열번호 1의 아미노산 서열을 갖는 단백질에서 1개 이상의 아미노산, 2개 이상의 아미노산, 3개 이상의 아미노산, 4개 이상의 아미노산, 또는 5개 이상의 아미노산이 치환(substitution)된 것일 수 있다.
상기 "실질적으로 동질의 생리활성"은 상기 단백질의 구조적, 기능적 상동성으로 인한 혈관신생성 질환의 예방 또는 치료 활성을 의미한다. 또한, 상기 단백질은 아미노산의 부가, 치환 또는 결실의 결과 물리화학적 특성을 수반하거나 수반하지 않을 수 있다. 예시적인 실시예에서, 상기 아미노산 치환은 단백질의 열안정성을 향상시키거나, 기질 특이성을 변경시키거나, 최적의 pH를 변화시킬 수 있다. 다만 이에 제한되는 것은 아니다.
상기 서열번호 1의 아미노산 서열을 갖는 단백질의 N 또는 C-말단은 아세틸기, 플루오레닐 메톡시 카르보닐기, 포르밀기, 팔미토일기, 미리스틸기, 스테아릴기 또는 폴리에틸렌글리콜(PEG)이 결합될 수 있다. 상기 단백질은 상기 변형에 의해 안정성이 현저히 개선될 수 있다. 상기 안정성은 in vivo 안정성뿐만 아니라, 저장 안정성도 포함하는 개념이다. 상기 보호기는 생체 내의 단백질 절단효소의 공격으로부터 상기 단백질을 보호할 수 있다.
혈관신생성 질환이란 혈관신생(angiogenesis)에 의해 유발되거나 진행 또는 악화되는 등 혈관신생에 직간접적인 영향을 받는 것으로 공지된 질환을 의미한다(Seminars in Nephrology, Vol 27, No 2, March 2007, pp 161-171 ; Clin Sci (Lond). 2017 Aug 1; 131(15): 1763-1780. 등). 예를 들어 노인황반변성(AMD), 당뇨병성 망막병증(DR), 미숙아 망막병증(ROP), 암(cancer), 관절염(arthritis), 건선(psoriasis), 비만(obesity), 천식(asthma), 치매(Alzheimer's disease) 및 미생물 감염성 질환(microbial infectious disease)으로 이루어진 군에서 선택되는 하나 이상을 포함할 수 있으나, 이에 제한되는 것은 아니다. 상기 AMD는 습식(wet) AMD 또는 건식(dry) AMD일 수 있다. 상기 관절염은 류마티스 관절염(rheumatoid arthritis)일 수 있다.
보다 구체적으로, 혈관신생성 질환은 혈관신생인자 중 하나인 혈관내피성장인자(vascular endothelial growth factor, VEGF)의 증가 또는 과도한 발현에 의해 유발되거나 진행 또는 악화되는 질환일 수 있다. VEGF는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, PLGF 등을 모두 포함하는 것일 수 있으나, 이에 제한되는 것은 아니다. 예시적인 실시예에서, SHC1 단백질은 혈관신생인자인 VEGF와 경쟁하여 VEGF 수용체(VEGFR)에 결합함으로써 VEGF와 VEGFR간의 결합을 방해하여 혈관생성 억제 활성을 갖는 것일 수 있다.
상기 과제를 해결하기 위한 본 발명의 다른 실시예에 따른 혈관신생성 질환의 예방 또는 치료용 약학적 조성물은 항-SHC1 항체(anti-SHC1 antibody)를 포함한다. 항-SHC1 항체는 전술한 SHC1 단백질에 결합하는 항체로서, SHC1 단백질을 항원(antigen)으로 인식하거나 SHC1 단백질 서열의 적어도 일부를 항원 결정기(epitope)로 인식하는 것일 수 있다. 항-SHC1 항체 또한, 혈관신생을 억제하여 전술한 혈관신생성 질환에 대해 예방 또는 치료 효능을 나타낼 수 있다. 즉, 항-SHC1 항체는 SHC1 단백질과 결합함으로써 VEGF 시그널을 억제하여 혈관생성 억제 활성을 갖는 것일 수 있으나, 본 발명의 일시예가 이에 제한되는 것은 아니다.
본 발명의 몇몇 실시예에서, 항-SHC1 항체는 SHC1 단백질 서열 중 적어도 일부에 대해 상보적인 서열을 포함할 수 있다.
본 발명의 예시적인 실시예에서, 항-SHC1 항체는 단클론 항체(monoclonal antibody)일 수 있다.
본 발명의 약학적 조성물은 경구 및 비경구의 여러 가지 제형으로 제조 및 투여될 수 있다. 제제화할 경우에는 통상적으로 사용되는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제를 섞어 조제된다. 또한 단순한 부형제 이외에 윤활제들도 사용될 수 있다. 경구투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 희석제, 부형제 등이 포함될 수 있다.
본 발명의 약학적 조성물은 비경구 투여할 수 있으며, 비경구 투여는 피하주사, 정맥주사, 근육 내 주사 또는 흉부 내 주사를 포함할 수 있다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제가 포함된다.
비제한적인 실시예에서, 상기 약학적 조성물은 SHC1 단백질을 예를 들어 0.1-1,000 μM으로 포함할 수 있으나, 이에 제한되는 것은 아니다.
상기 다른 과제를 해결하기 위한 본 발명의 일 실시예에 따른 혈관신생성 질환의 예방 또는 개선용 건강기능식품 조성물은 SHC1 단백질을 포함한다. SHC1 단백질은 유효성분으로서 포함될 수 있다.
SHC1 단백질은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분의 혼합량은 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 예를 들어, SHC1 단백질은 건강식품 중에 0.1 내지 90 중량부로 포함될 수 있다.
또한, 상기 조성물이 건강기능성 음료 조성물일 경우 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 나아가, 여러 가지 영양제, 비타민, 풍미제, 착색제, 중진제, 증점제, pH 조절제, 안정화제, 방부제, 탄산화제 등을 더 함유할 수 있다. 예시적인 실시예에서는, 과일 또는 야채 음료의 제조를 위한 과육을 더 함유할 수 있다.
본 발명은 다른 실시예에 따라 혈관신생성 질환의 예방 또는 치료가 필요한 환자에게 SHC1 단백질을 투여하는 단계를 포함하는 혈관신생성 질환의 예방 또는 치료 방법을 제공할 수 있다.
기타 실시예의 구체적인 사항들은 상세한 설명에 포함되어 있다.
본 발명의 실시예들에 따르면, SHC1 단백질 또는 항-SHC1 항체를 포함하는 약학적 조성물 또는 건강기능식품 조성물을 통해 노인황반변성과 같은 혈관신생성 질환들을 효과적으로 예방, 치료 및 개선할 수 있다.
본 발명의 실시예들에 따른 효과는 이상에서 예시된 내용에 의해 제한되지 않으며, 더욱 다양한 효과들이 본 명세서 내에 포함되어 있다.

실험예 1: SHC1 단백질의 확인
실험 방법
노인황반변성(Age-Related Macular Degeneration, AMD) 망막(8 mm 황반 및 주변 펀치), 망막색소상피(Retinal Pigment Epithelium, RPE) 세포 (8 mm 중앙 및 주변 펀치) 및 연령 일치 대조군 눈(n = 9, 생물학적 3중 및 기술적 3중 실험)을 라이온스 안구 은행(유타 대학교 모란 안구 센터)에서 기증받았다.
황반(I), 말초 망막(II), 중심 상피세포(III) 및 말초 상피세포(IV)의 단백질체를 연령이 일치하는 대조군 기증자 눈과 비교하여 황반변성이 진행 중인 영역별로 노화 관련 분자 메커니즘을 결정하였다.
망막 단백질은 전하 기반 스핀 컬럼 크로마토그래피로 농축하였고, 2D 겔 전기영동으로 분해하였다. 전체 용해물에서 트립신 분해된 펩티드를 Ga3+/TiO2 금속 이온 크로마토그래피를 사용하여 농축하였다. 용출된 단백질을 액체 크로마토그래피를 사용하여 분석하였다. 세린, 트레오닌 및 티로신 인산화는 포스포 웨스턴 블럿 분석을 통해 확인하였다.
분석된 내용을 바탕으로 VEGFR2(KDR)에 대한 황반변성 단백질 지도를 작성하였다. AMD 관련 단백질 타겟의 기능분석을 위해 VEGFR2 단백질 인력을 지도로 나타내었다. AMD 및 산화 스트레스의 바이오마커 상호작용 지도는 다음과 같은 바이오인포메틱스 도구를 사용하여 제작하였다. 구체적으로, STRING 11.0 (http://string-db.org/), MIPS(http://mips.helmholtz-muenchen.de/proj/ppi) 및 iHOP(http://www.ihop-net.org/UniPub/iHOP/)를 포함한 단백질-단백질 상호작용 지도 소프트웨어 및 데이터베이스를 사용하여 제작하였다. STRING 데이터베이스는 현재 2,031개 유기체의 9,643,763개 단백질을 다루고 있다. AMD 또는 산화 스트레스 조건에서 발견되는 단백질을 추가하여 AMD 상호작용체를 확립하였다.
AMD 바이오마커 상호작용체는 본 실험으로 발견한 단백질을 사용하여 제작하였다. 단백질 상호작용은 이웃(녹색), 유전자 융합(빨간색), 동시 발생(진한 파란색), 공동 발현(검정색), 결합 실험(보라색), 데이터베이스(파란색), 상동성(청록색) 및 텍스트 마이닝(라임)을 포함한 8가지 범주를 사용하여 나타냈다. 단백질 상호 작용은 게놈 컨텍스트, 높은 처리량 실험, 공동 발현 및 Pubmed의 데이터를 통해 결정하고 확인하였다. 단백질 데이터베이스 분석은 AMD 안구에서 특정 단백질의 구조 특이적 인산화를 보여주었다. AMD 프로테옴 간의 상호작용은 스트레스 조건 하에서 망막/RPE 프로테옴과 비교되었다.
선별된 후보물질들에 대해 3D 단백질-리간드의 분자 모델링 방법(3D Molecular Dymanics)을 통해 보다 안정적으로 결합한 결과를 보여 준 리간드들을 hit 초기 유효물질로 압축하였다. 3D 단백질-리간드 분자모델링은 타겟 단백질 구조는 Protein Data Bank(PDB) DB에서 입수하고 SwissDock 및 GOLD(ver.5.0)를 포함한 단백질-리간드 도킹 서버를 활용하여 리간드-단백질 상호작용을 확인하였다. VEGFR2와 결합하는 단백질의 positive control로서 VEGFA와 VEGFC를 사용하였다.
실험 결과
망막세포 사멸과 혈관생성 조절에 관여한 단백질 지도와 축적된 자체 프로테오믹스 데이터베이스를 통해 새로운 황반변성 타겟 단백질인 SHC1 (Src homology 2 domain-containing-transforming protein C1, SH2 domain protein C1)을 발견하였다. 하기 표 1은 SHC1 단백질의 VEGFR2(KDR)에 대한 결합을 VEGF와 비교하여 scoring 한 것이고, 도 1은 SHC1의 구조를 나타낸 이미지이다. 하기 표 2는 SHC1 단백질과 안토시아닌계 분자들과의 결합상수를 계산한 것이고, 도 2 및 3은 SHC1과 안토시아닌계 분자간의 결합구조를 나타낸 이미지이다.

이와 같은 SHC1 단백질에 대한 구조 분석과 문헌 조사를 바탕으로 SHC1 단백질을 신규 황반변성 단백질 타겟으로 선정하였다. SHC1 단백질의 아미노산 서열은 서열번호 1과 같은 것으로 확인되었다.SHC1 단백질은 산화 스트레스를 조절하는 기능과 혈관생성 조절기능이 있는 것으로 기대할 수 있다. 즉, 상기 실험예를 통해 확인된 SHC1 단백질의 VEGFR에 대한 결합 특성을 바탕으로, SHC1 단백질이 혈관신생인자인 VEGF와 VEGFR의 결합을 저해함으로써 황반변성 등의 혈관신생성 질환을 예방 및 치료할 것으로 기대할 수 있다.
망막세포의 산화적 손상은 황반변성의 가장 중요한 발병기전 중 하나이다. 황반변성의 첫번째 치료전략은 내인성 산화 단백질을 억제하는 것이다. 망막 세포 표면 수용체가 세포 내 단백질에 신호전달 시 도킹 도메인을 통해 신호를 증폭한다. SHC1과 같은 수용체 티로신 키나아제(Receptor Tyrosine Kinase)의 경우 자가인산화 반응은 PTB (Phospho Tyrosine-Binding) 도메인을 스캐폴드로 사용하여 하위신호 리간드와 결합한다. SHC1은 EGFR 반응 전달자로서 발암신호 또는 혈관생성신호에 가장 중요한 스위치 중 하나이다. SHC1는 상기 실험예의 프로테오믹스와 메타볼로믹스 도구를 통하여 새롭게 발견된 황반변성 타겟으로서, 산화반응이 시작인 건식-AMD와 이상 혈관생성이 주 메커니즘인 습식-AMD의 동시 타겟으로 적합하다.

Claims (5)
- 서열번호 1의 아미노산 서열과 90% 이상의 서열 상동성을 갖는 단백질을 포함하는혈관신생성 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 1에 있어서,상기 혈관신생성 질환은 노인황반변성(AMD), 당뇨병성 망막병증(DR), 미숙아 망막병증(ROP), 암(cancer), 관절염(arthritis), 건선(psoriasis), 비만(obesity), 천식(asthma), 치매(Alzheimer's disease) 및 미생물 감염성 질환(microbial infectious disease)으로 이루어진 군에서 선택되는 하나 이상을 포함하는혈관신생성 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 1에 있어서,상기 혈관신생성 질환은 노인황반변성 또는 당뇨병성 망막병증인혈관신생성 질환의 예방 또는 치료용 약학적 조성물.
