Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von Chlor- benzaldehydoximen der allgemeinen Formel (I).
Chlorbenzaldehydoxime der allgemeinen Formel (I) sind wichtige Vorstufen von agrochemischen Wirkstoffen (vgl. WO 2018/228985) und pharmazeutischen Wirkstoffen (z.B. DNA Binding Agents: Woods, Craig R. et al. Bioorganic & Medicinal Chemistry Letters, 12(18), 2647-2650, 2002).
Im Stand der Technik sind zahlreiche Chlorierungsmethoden beschrieben, beispielsweise lehrt die WO 2004/29066 sowie die Industrial Crops & Products 2019, 140, 111706 die Herstellung von Chlorbenzaldehydoximen durch die Umsetzung von Oximen mit N-Chlorsuccinimid (NCS) und anschließender wässrige Aufarbeitung (Extraktion mit EtOAc H2O). Bei dem beschriebenen Verfahren wurden jedoch nur kleinen Mengen (2,45 g) der erhaltenen Chlorbenzaldehydoxime als Feststoff isoliert. Grundsätzlich ist jedoch die Isolierung von Chlorbenzaldehydoximen im technischen Maßstab als Feststoff unerwünscht, da Chlorbenzaldehydoxime oftmals energiereiche Verbindungen sind, die eine hohe Zersetzungstendenz aufweisen. Das in der WO 2004/29066 beschriebene Verfahren verwendet Dimethylformamid (DMF) als Lösungsmittel. Allerdings ist bekannt, dass dessen Verwendung als Lösungsmittel m technischem Maßstab problematisch sein kann. Das liegt an der stark exotherm verlaufenden Reaktion zwischen DMF und dem Chlorierungsmittel, die dann möglicherweise unkontrolliert verläuft. (OPRD 2020, 24, 1586; Bull. Chem. Soc. Jpn. 1994, 67, 156).
Die Verwendung von Chlorgas zur Chlorierung von Oximen wird beispielsweise in Journal of Organic Chemistry 1971, 36, 2146 beschrieben. Dabei werden die Reaktionen ausschließlich in Chloroform oder Dichlormethan in großer Verdünnung durchgeführt, was für eine großtechnische, industrielle Synthese nicht geeignet ist und die Chlorbenzaldehydoxime in moderaten bis geringen Ausbeute von 3-80% liefert. Eine wie beschrieben bevorzugte Chlorierung unter basischen Bedingungen durch Zusatz von Triethylamin führt zu einer teilweisen Zersetzung der Chloroxim- Verbindung. Des Weiteren lehrt das Journal of Heterocyclic Chemistry; 2012, 49, 621 die Verwendung von Chlorgas in Methanol als Lösungsmittel unter leicht basischen Reaktionsbedingungen durch den Zusatz von Natriumcarbonat. Neben der Entstehung sicherheitsrelevanter Mengen gasförmigen Kohlenstoffdioxids kann es erneut zu einer Zersetzung des Produktes unter basischen Bedingungen kommen. Der bei der Chlorierung freigesetzte Chlorwasserstoff kann in Reaktion mit dem Lösungsmittel außerdem zur Bildung toxischer Verbindungen führen. Die Verwendung von Methanol als Lösungsmittel verhindert zudem einen direkten Einsatz der Produktlösungen ohne Lösungsmittelwechsel in einer Vielzahl chemischer
Reaktionen. Neben sicherheitsrelevanten Einschränkungen aufgrund der hohen Energie solcher Chlor- Verbindungen und der bevorzugten Verwendungen verdünnter Systeme, verringert der Wechsel von Lösungsmitteln auch die Effizienz sowie die Nachhaltigkeit des Verfahrens.
Das Journal of Enzyme Inhibition and Medicinal Chemistry; vol. 31; nb. 6; (2016); p. 964 - 973 lehrt die Chlorierung von Oximen mit Trichlorisocyanursäure (TCCA) mit Triethylamin als Base. Zwar wird hier auf den Einsatz von DMF als Lösungsmittel verziehet, es wurde jedoch beobachtet, dass die Chloroxime im basischen Milieu durch Bildung der Nitriloxyde zum Abbau neigen, was zu Ausbeuteverlusten führen kann (z.B. Dimerisation der Nitriloxyde zu Furoxanen: „Kinetik und Mechanismus 1.3-Dipolarer Cycloadditionen“ von Prof. Dr. R. Huisgen, Angew. Chem. 1963, 75, 742-754, Seite 751; „Fragmentation of Nitrile Oxides with Tri ethyl amine“ Tetrahedron Lett. 1983, 24, 4377-4380). Zudem fallen bei der Verwendung von Trichlorisocyanursäure (TCCA) größere Mengen der festen Isocyanursäure als Abfallprodukt an, was sich für den für den Prozess im Sinne der Nachhaltigkeit und des Abfallmanagements nachteilig! auswirkt.
Der Erfindung lag somit die Aufgabe zugrunde, ein Verfahren zur Chlorierung von Benzaldehydoximen zur Verfügung zu stellen, welches einerseits auf DMF oder chlorierte Alkanverbindungen als Lösungsmittel sowie den Einsatz von wenig atomökonmi sehen sowie abfalllastigen Chlorierungsmitteln wie Trichlorisocyanursäure oder V-Chlorsuccinimid verzichtet und andererseits, nicht die durch stärkere Basen, wie z.B. Triethylamin, verursachten Ausbeuteverluste mit sich bringt, dabei allerdings gleichzeitig kostengünstig ist und im industriellen Maßstab eingesetzt werden kann. Die Chlorbenzaldehydoxime sollten dabei in hohen Ausbeuten erhalten werden und als Lösung ohne die zwingende Notwendigkeit eines Lösungsmittelwechsels in vielseitigen chemischen Reaktionen durch geeignete Wahl des Lösungsmittels eingesetzt werden können.
Die Aufgabe wurde erfindungsgemäß gelöst durch ein Verfahren zur Herstellung von Chlor- benzaldehydoximen der allgemeinen Formel (I)

In einer bevorzugten Ausführungsform wird neben Chlorgas eine Amid-Base zum Reaktionsgemisch hinzugegeben, um die Verbindungen der Formel (II) zu Verbindungen der allgemeinen Formel (I) umzusetzen.
In einer besonders bevorzugten Ausführungsform werden neben Chlorgas katalytische Mengen einer Amid-Base zum Reaktionsgemisch hinzugegeben, um die Verbindungen der Formel (II) zu Verbindungen der allgemeinen Formel (I) umzusetzen.

Verfahren zur Herstellung von Chlorbenzaldehydoximen der Formel (I), dadurch gekennzeichnet, dass die Verbindungen der allgemeinen Formel (II) mit Hilfe von Chlorgas (Cb) zu Verbindungen der allgemeinen Formel (I) umgesetzt werden.
Bevorzugt wird neben Chlorgas eine Amid-Base zum Reaktionsgemisch hinzugegeben, um die Verbindungen der Formel (II) zu Verbindungen der allgemeinen Formel (I) umzusetzen.
Besonders bevorzugt werden neben Chlorgas katalytische Mengen einer Amid-Base zum Reaktionsgemisch hinzugegeben, um die Verbindungen der Formel (II) zu Verbindungen der allgemeinen Formel (I) umzusetzen.
Das erfmdungsgemäße Verfahren hat den Vorteil, dass der Einsatz stöchiometrischer Mengen DMF oder chlorierter Alkanverbindungen als Lösungsmittel vermieden wird. Damit wird das Risiko minimiert, dass die Reaktion stark exotherm und unkontrolliert verläuft und auf wenig nachhaltige Lösungsmittel verzichtet werden kann. Darüber hinaus kann durch die Verwendung von Chlorgas auf die Verwendung von N-Chlorsuccinimid (NCS) oder Trichlorisocyanursäure (TCCA) verzichtet und somit die Abfallmengen reduziert und die Nachhaltigkeit des Verfahrens deutlich gesteigert werden. Folglich eignet sich die Reaktion zur Durchführung in großem Maßstab.
Als Katalysator geeignete Amid-Basen sind z.B. Dimethylformamid (DMF), Dibutylformamid (DBF), Diethylformamid (DEF), oder Dimethylacetamid (DMAc), wobei Dimethylforamid oder Dibutylformamid bevorzugt sind.
Bei dem erfindungsgemäßen Verfahren werden bevorzugt keine oder 0,05 -0,3 Äquivalente einer Amid-Base eingesetzt, und zwar bezogen auf das Benzaldehydoxim (II), besonders bevorzugt 0,1 - 0,3 Äquivalente. Bevorzugt werden 0,95 - 2,0 Äquivalente Ch bezogen auf das Benzaldehydoxim (II) eingesetzt, besonders bevorzugt 1,0 - 1,5 eq.
Die Chlorierung wird gewöhnlich in einem Temperaturbereich von -10 °C bis 40 °C, vorzugsweise -5 °C bis 10 °C, besonders bevorzugt 0 °C bis 10°C durchgeführt.
Des Weiteren wird die Chlorierung in Gegenwart eines Lösungs- oder Verdünnungsmittels durchgeführt, bevorzugt sind die Lösungsmittel Tetrahydrofuran, Me-THF, Acetonitrile, N,N-Dimethylacetamid, Toluol, Xylol, Chlorbenzol, Ethylacetat, Isopropylacetat, Methyl-tert-butylether, Cyclopentylmethylether, tert- Amylmethylether oder Mischungen der genannten Lösungsmittel.
Chlor wird zu dem Benzaldehydoxim der Formel (II) als Gas eingeleitet.
Bevorzugt wird unter wasserfreien Bedingungen gearbeitet. Die Ausbeute wird dabei gesteigert.
Beispiele
Die vorliegende Erfindung wird anhand der nachfolgenden Beispiele näher erläutert, ohne die Erfindung dabei auf diese einzuschränken.
Messverfahren
Die Produkte wurden mittels ’H- und/oder 19F-NMR Spektroskopie und/oder quantitativer HPLC charakterisiert.
Die NMR-Spektren wurden mit einem Bruker Avance 400, ausgestattet mit einem Durchflussprobenkopf (60 pl Volumen), bestimmt. In Einzelfällen wurden die NMR Spektren mit einem Bruker Avance II 600 gemessen.
Die quantitativen HPLC-Bestimmungen wurden auf einer HPLC Agilent HP1260 Serie gemessen. Die Technik beruht auf HPLC mit UV-Detektion, einer Agilent XDB CI 8 Säule und mit externer Standard Auswertung (Verwendung eines externes Standards mit Referenzfaktor). Die Proben, Referenzstandard, sowie interner Standard werden in Acetonitril gelöst.
Beispiel 1 (ohne Zusatz eines Katalysators in Isopropylacetat)
In einem 0,5L Reaktor mit KPG-Rührer und Gaseinleitungs-Rohr wurden, unter Stickstoff Schutzgasatmosphäre, bei 23 °C, 25.0 g N-(3,5-Difluorbenzyliden)hydroxylamin (1,0 eq) in 225g Isopropylacetat vorgelegt. Nachdem die Lösung auf 15 °C abgekühlt worden war, wurden unter Rühren (300 rpm) innerhalb von 1h, 14,0 g CE (1.36 eq) eingeleitet. Die Temperatur wurde während der Zugabe dabei unterhalb von 14-16 °C gehalten. Nach abgeschlossener CE Dosierung wurde die Reaktionsmischung weitere 30 Minuten bei 15 °C nachgerührt. Die HPLC Analyse zeigte einen Anteil von 100% 3,5-Difluor-N-hydroxybenzencarboximidoylchlorid. Anschließend wurde die Reaktionsmischung unter Rühren auf 10 °C gekühlt und 1h bei 300mbar entgast. Danach wurden 257,3 g einer gelben Lösung von 3,5-Difluor-N-hydroxybenzencarboximidoylchlorid in Isopropylacetat erhalten (10,1 w/w%, 92,9% Ausbeute nach QHPLC). Für die Charakterisierung des Produktes mittels 1H-NMR wurde eine analytische Probe im Vakuum vollständig vom Lösungsmittel befreit.
' H-NMR (401MHz, CDCI3): 5 (ppm) = 6.84-6.89 (m, 1H), 7.37-7.45 (m, 2H), 10.86 (bs, 1H).
19F-NMR (377MHz, CDC13): 5 (ppm) = -109.3 (m, 2F).
Beispiel 2 (Ohne Zuzsatz eines Katalysators in einer Mischung aus Toluol und THF)
In einem 0,5L Reaktor mit KPG-Rührer und Gaseinleitungs-Rohr wurden, unter Stickstoff Schutzgasatmosphäre, bei 23 °C, 89.2 g einer Lösung von N-(3,5-Difluorbenzyliden)hydroxylamin (1,0 eq) in einer Mischung von Toluol und THF (25.5 w/w%, QHPLC) vorgelegt und mit weiteren 138g Toluol verdünnt. Nachdem die Lösung auf 10 °C abgekühlt worden war, wurden unter Rühren (300 rpm) innerhalb von 3h, 18,0 g CF eingeleitet. Die Temperatur wurde dabei während der Zugabe unterhalb von 9-12 °C gehalten. Nach abgeschlossener CL Dosierung wurde die Reaktionsmischung weitere 30 Minuten bei 10 °C nachgerührt. Die HPLC Analyse zeigte einen Anteil von 100% 3,5-Difluor-N- hydroxybenzencarboximidoylchlorid. Anschließend wurde die Reaktionsmischung unter Rühren auf 0 °C gekühlt und 2h bei lOOmbar entgast. Danach wurden 229,7 g einer gelben Lösung von 3,5-Difluor-N- hydroxybenzencarboximidoylchlorid in einer Mischung von Toluol und THF erhalten (9,7 w/w%, 80,5% Ausbeute nach QHPLC).
Beispiel 3 (Mit Katalysator in einer Mischung aus Toluol und THF)
In einem 1L Reaktor mit KPG-Rührer und Gaseinleitungs-Rohr wurden, unter Stickstoff Schutzgasatmosphäre, bei 23 °C, 695 g einer Lösung von N-(3,5-Difluorbenzyliden)hydroxylamin (1,0 eq) in einer Mischung von Toluol und THF (21,2 w/w%, QHPLC) vorgelegt und mit weiteren 41,7g THF verdünnt sowie 13.7 g (0.2 eq) Dimethylformamid (DMF) zugegeben. Nachdem die Lösung auf 0 °C abgekühlt worden war, wurden unter Rühren (300 rpm) innerhalb von 1,5h 77,7 g(l . 17 eq) CL eingeleitet. Die Temperatur wurde dabei während der Zugabe unterhalb von 10 °C gehalten. Nach abgeschlossener CL Dosierung wurde die Reaktionsmischung weitere 30 Minuten bei 10 °C nachgerührt. Die HPLC Analyse zeigte einen Anteil von 100% 3,5-Difluor-N-hydroxybenzencarboximidoylchlorid. Anschließend wurde die Reaktionsmischung unter Rühren auf 0°C gekühlt und 1h bei 50mbar entgast. Die Produkt- Lösung wurde im Anschluss mit 130g einer 5% wässrigen Natriumchlorid Lösung bei 0 °C gewaschen und die Phasen bei 20 °C getrennt. Die noch wasserhaltige, organische produkthaltige Phase wurde dann bei 45 °C und 85 mbar azeotrop getrocknet. Nach Zugabe von 258g Toluol, wurden 740,0 g einer schwach gelblich, klaren Lösung von 3,5-Difluor-N-hydroxybenzencarboximidoylchlorid in einer Mischung von Toluol und THF erhalten (22,3 w/w%, 91,9% Ausbeute nach QHPLC).
Beispiel 4 (Mit Katalysator in einer Mischung aus Chlorbenzol und THF)
In einem 0,5L Reaktor mit KPG-Rührer und Gaseinleitungs-Rohr wurden, unter Stickstoff Schutzgasatmosphäre, bei 23 °C, 46,1 g N-(3,5-Difluorbenzyliden)hydroxylamin (1,0 eq) in 145g Chlorbenzol und 36,0 g Tetrahydrofuran vorgelegt und im Anschluss mit 5,0g Dibutylformamid (DBF,
0,1 eq) versetzt. Nachdem die Lösung auf 0 °C abgekühlt worden war, wurden unter Rühren (300 rpm) innerhalb von 40 Minuten, 23,0 g CL (1.2 eq) eingeleitet. Die Temperatur wurde dabei während der Zugabe unterhalb von 17 °C gehalten. Nach abgeschlossener CL Dosierung wurde die Reaktionsmischung weitere 30 Minuten bei 10 °C nachgerührt. Die HPLC Analyse zeigte einen Anteil von 100% 3,5-Difluor- N-hydroxybenzencarboximidoylchlorid. Anschließend wurde die Reaktionsmischung unter Rühren auf 0°C gekühlt und 1h bei 50mbar entgast. Danach wurden 242,2 g einer gelblich, klaren Lösung von 3,5- Difluor-N-hydroxybenzencarboximidoylchlorid in einer Mischung von Chlorbenzol und THF erhalten (21,3 w/w%, 91,7% Ausbeute nach QHPLC).

Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass bei der Reaktion katalytische Mengen einer Amid-Base zum Reaktionsgemisch hinzugegeben werden. Verfahren nach Anspruch 6 oder 7, dadurch gekennzeichnet, dass die Amid-Base Dimethylformamid (DMF), Dibutylformamid (DBF), Diethylformamid (DEF) oder Dimethylacetamid (DMAc) ist. Verfahren nach Anspruch 6 oder 7, dadurch gekennzeichnet, dass die Amid-Base Dimethylformamid (DMF) oder Dibutylformamid (DBF) ist. Verfahren nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass die Reaktion bei -10°C bis 40°C durchgeführt wird. Verfahren nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass die Reaktion bei -5°C bis 10°C durchgeführt wird. Verfahren nach einem der Ansprüche 1 bis 11, dadurch gekennzeichnet, dass 0,1 - 0,3 Äquivalente Amid-Base eingesetzt, und zwar bezogen auf das Benzaldehydoxim (II). Verfahren nach einem der Ansprüche 1 bis 11, dadurch gekennzeichnet, dass 1,0 - 1,5 Äquivalente Ch eingesetzt werden, und zwar bezogen auf das Benzaldehydoxim (II).
