Abstract
본 발명은 c-Met 발현 CTC를 이용한 유방암 환자의 예후 예측 방법에 관한 것으로, 보다 상세하게는 유방암 환자의 c-Met 발현 CTC를 분리하고 이를 계수함으로써 유방암의 예후를 예측하는 방법 및 c-Met 발현 CTC에 특이적으로 결합하는 제제를 포함하는 유방암 예후 예측용 조성물에 관한 것이다. 본 발명의 방법 및 조성물은 유방암 환자의 조직검사 없이도 정확하게 유방암 환자의 예후 예측을 하는데 유용하게 이용될 수 있다.
Description
본 출원은 2022년 4월 20일에 출원된 대한민국 특허출원 제10-2022-0049175호를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다.
본 발명은 c-Met 발현 CTC (c-Met enriched circulating tumor cell)를 이용한 유방암 환자의 예후 예측 방법에 관한 것으로, 보다 상세하게는 유방암 환자의 c-Met 발현 CTC를 분리하고 이를 계수함으로써 유방암의 예후를 예측하는 방법에 관한 것이다.
유방암은 여성에 있어 가장 흔한 암이며, 두 번째로 사망률이 높은 암이다. 유방암 발명에 대한 위험 인자는 인종, 나이 및 암 억제 유전자 BRCA-1 및 BRCA-2 및 p53에서의 돌연변이 등을 포함하며, 알코올 섭취, 고지방 식이, 운동 부족, 외인성 폐경 후 호르몬 및 이온화 방사선 또한 유방암의 발병 위험을 증가시킨다. 유방암은 호르몬 수용체(에스트로겐 수용체 또는 프로게스테론 수용체)와 HER2 (human epidermal growth factor receptor 2)의 발현 상태에 따라 루미날 A형, 루미날 B형, HER2형 및 삼중음성 유방암(TNBC)의 네 종류의 아형으로 구분되고 있다.
현재의 유방암에 대한 치료 방법은 수술 이후 항암 치료, 호르몬 치료, 표적 치료 혹은 방사선 치료 등 향후 재발을 줄이기 위한 추가의 보조적인 치료가 필요한 경우가 많다. 유방암은 유방암 주요 수용체의 상태에 따라 암의 특성이 다르며, 따라서 조직검사를 통해 호르몬 수용체(ER 및 PR) 발현 유무와 과발현 여부, 전이 상태 확인 및 림프절 전이 여부(양성 또는 음성)를 고려하여 추후 치료 방법에 대한 근거를 확립하고 있다. 그러나 조직을 이용한 검사는 조직 채취 당시의 바이오마커 상태를 반영하고, 조직을 채취하기 위하여 침습적인 방법(Biopsy, Resection 등)이 사용되기 때문에 환자에게 매우 큰 위험과 부담을 초래할 가능성이 존재한다.
한편, CTC (circulating tumor cell, 순환 종양 세포)는 악성 종양 환자의 말초 혈액에서 발견되는 종양세포를 말한다. CTC는 암 전이 과정에서 중요한 역할을 하기 때문에 CTC는 암의 연구, 진단 등에 있어서 매우 중요하게 여겨지고 있으나, 말초 혈액 내에서 존재하는 순환 종양 세포의 수가 매우 적기 때문에, 수백만 개 이상의 정상 혈구 세포에 섞여 있는 수십 개 이하의 종양 세포를 검출할 수 있는 정도의 민감도가 요구되는 검출 시스템이 필요하다.
CTC를 분리하는 방법은 크게 크기, 기형, 밀도 또는 전하 등 CTC의 물리적 특성을 이용한 방법(physical isolation)과; 항체, 항체가 부착된 비드, 자성 비드(magnetic beads), 자성물질(magenetic material), 또는 압타머 등 다른 물질에 의해 검출되는 등 마커에 의존한 단백질 기반 방법인 생물학적 특성을 이용한 방법(biological isolation)으로 구분할 수 있다. 유일하게 FDA의 허가를 받은 장비는 생물학적 특성을 이용한 방법으로서 EpCAM (epithelial cell adhesion molecule) 항체로 세포를 분리하고, 면역세포염색을 통해 암세포를 확인하는 방식을 사용한다. 따라서 EpCAM enriched CTC를 이용하여 다양한 연구가 진행되었으나, 전이에 따라 EMT (epithelial mesenchymal transition)를 거치는 경우 EpCAM의 발현이 낮아, 해당 세포를 검출할 수 없다는 단점이 있다.
따라서 유방암 예후 예측을 위한 검사에 있어 환자의 위험과 부담을 줄이기 위해 CTC를 활용하면서도 조직검사를 이용한 방법의 정확도를 유지하기 위한 새로운 방법이 필요한 실정이다.

이에, 본 발명자들은 유방암 환자의 예후를 정확하게 예측할 수 있는 방법을 개발하기 위해 예의 연구를 거듭한 결과, 유방암 환자의 혈액으로부터 c-Met 발현 CTC(circulating tumor cell)를 분리하고 이를 계수함으로써, 유방암 환자의 예후를 정확하게 예측할 수 있음을 최초로 확인하고 본 발명을 완성하였다.
따라서 본 발명의 목적은 유방암 환자의 예후 예측에 필요한 정보를 제공하기 위하여, 하기 단계를 포함하는 유방암 예후 예측에 필요한 정보를 제공하는 방법을 제공하는 것이다:
(a) 유방암 환자로부터 수득된 생물학적 시료로부터 c-Met 발현 CTC (c-Met enriched circulating tumor cell)를 분리하는 단계;
(b) 상기 (a) 단계에서 분리된 CTC를 계수하는 단계; 및
(c) 상기 (b) 단계에서 계수한 값으로 유방암 예후를 예측하는 단계로서, 계수한 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계.
본 발명의 다른 목적은 유방암 환자의 예후 예측에 필요한 정보를 제공하기 위하여, 하기 단계를 포함하는 유방암 예후 예측에 필요한 정보를 제공하는 방법을 제공하는 것이다.
(a) 유방암 환자로부터 수득된 생물학적 시료로부터 cfDNA (cell-free DNA)를 분리하는 단계;

(a) 유방암 환자로부터 수득된 생물학적 시료로부터 c-Met 발현 CTC (c-Met enriched circulating tumor cell)를 분리하는 단계;
(b) 상기 (a) 단계에서 분리된 CTC를 계수하는 단계; 및
(c) 상기 (b) 단계에서 계수한 값으로 유방암 예후를 예측하는 단계로서, 계수한 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계.

이하 본 발명을 상세히 설명한다.
본 발명은 유방암 환자의 예후 예측에 필요한 정보를 제공하기 위하여, 하기 단계를 포함하는 유방암 예후 예측에 필요한 정보를 제공하는 방법을 제공한다:
(a) 유방암 환자로부터 수득된 생물학적 시료로부터 c-Met 발현 CTC (c-Met enriched circulating tumor cell)를 분리하는 단계;
(b) 상기 (a) 단계에서 분리된 CTC를 계수하는 단계; 및
(c) 상기 (b) 단계에서 계수한 값으로 유방암 예후를 예측하는 단계로서, 계수한 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계.
유방암은 유전자 및 분자생물학적 특성으로 분류하는데(표 1), 아형에 따라 치료에 따른 결과 및 예후도 다르다고 보고되며, 수술적 방법이나 항암화학요법 선택의 지표로 사용된다.

이 중, 본 발명에서의 유방암은 바람직하게는 에스트로겐 수용체(ER) 및/또는 프로게스테론 수용체(PR) 양성(HR+)이면서 HER2 음성(HER2-)인 유방암, 또는 에스트로겐 수용체(ER) 및/또는 프로게스테론 수용체(PR) 양성(HR+)이면서 HER2 양성(HER2+)인 유방암일 수 있으며, 전이성 유방암 또는 비전이성 유방암일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 "예후"란 유방암 치료 중 또는 치료 후 병의 진행 경과를 의미하는 것으로, 바람직하게는 치료 후 병의 진행 경과를 의미할 수 있으나 이에 제한되는 것은 아니다. 또한 상기 "병의 진행 경과"란 암의 완치, 재발, 전이, 전이성 재발, 무질병 생존율, 무원격전이 생존율 또는 무진행 생존율을 포함하는 개념이며, 바람직하게는 무진행 생존율을 의미하나 이에 제한되는 것은 아니다.
한편, 상기 "무진행 생존(PFS, progression-free survival)"은 암과 같은 질병을 치료 중이거나 치료 후 환자가 질병을 지닌 채 살고 있지만 악화되지는 않는 시간을 말한다. PFS는 암 치료의 효과를 평가하는데 자주 사용되는 측정 기준인데, 치료의 시작으로부터 진행 병변까지의 시간 간격은 무진행 생존의 대표적인 정의이며, 이는 치료법에 대한 임상적 이익의 척도가 된다.
이하에서는 본 발명에서 유방암 예후 예측에 필요한 정보를 제공하는 방법의 각 단계를 보다 상세히 설명한다.
(a) 유방암 환자로부터 수득된 생물학적 시료로부터 c-Met 발현 CTC (c-Met enriched circulating tumor cell)를 분리하는 단계;
본 발명에서, 생물학적 시료는 유방암 환자로부터 수득한 혈액, 혈장, 혈청, 전혈, 단리된 혈액 세포, 골수액, 림프액, 타액, 뇨, 점막액, 양수, 또는 이들의 조합일 수 있으며, 바람직하게는 유방암 환자로부터 수득한 혈액, 혈장, 혈청, 전혈 또는 이들의 조합일 수 있고, 가장 바람직하게는 유방암 환자로부터 수득한 혈액일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서, c-Met 발현 CTC (c-Met enriched circulating tumor cell) 이란, c-Met이 발현되어 세포 표면에 다수 분포하는 CTC를 말한다.
상기 "c-Met"은 세포 표면에 존재하는 대표적인 RTK (Receptor Tyrosine Kinase)를 말하는데, c-Met은 그 리간드인 HGF/SF (Hepatocyte Growth Factor/Scattering Factor)와 결합하여 세포 내 신호전달을 촉진시켜 세포의 성장을 촉진할 뿐 아니라 많은 종류의 암세포에 발현되어 암 발생, 암 전이, 암세포 이동, 암세포 침투, 신생 혈관 형성에도 광범위하게 관여한다. 또한 리간드의 이름이 의미하듯, HGF/SF를 통한 c-Met signaling은 거의 모든 종류의 epithelial tumor의 cell-cell contact를 약화시켜 scattering을 야기하는 대표적인 암 전이 초기 단계의 단백질이다.
본 발명에서, c-Met 발현 CTC를 분리하는 단계는, 당업계에 공지된 통상의 방법에 따라서 수행될 수 있다. CTC를 분리하는 방법은 크게 크기, 기형, 밀도 또는 전하 등 CTC의 물리적 특성을 이용한 방법(physical isolation)과; 항체, 항체가 부착된 비드, 자성 비드(magnetic beads), 자성물질(magenetic material), 또는 압타머 등 다른 물질에 의해 검출되는 등 마커에 의존한 단백질 기반 방법인 생물학적 특성을 이용한 방법(biological isolation)으로 구분할 수 있다.
본 발명에 따른 방법에 있어서 물리적 특성을 이용한 CTC 분리 방법을 사용하는 경우, 시료로부터 분리된 CTC로부터 c-Met 발현 CTC를 구분하기 위해 c-Met을 염색하는 과정을 수행함으로써 본 발명에 따른 방법에 필요한 c-Met 발현 CTC를 분리할 수 있다.
본 발명에 따른 방법에 있어서 생물학적 특성을 이용한 CTC 분리 방법을 사용하는 경우, 상기 항체는 바람직하게는 c-Met에 특이적으로 결합하는 항체일 수 있다. 상기 '항체가 부착되는 자성물질'은 이에 제한되지는 않으나, 자성금속 또는 자성금속 산화물을 포함하는 물질일 수 있으며, 바람직하게는 Co, Mn, Fe, Ni, Gd, MM'2O4 및 MxOy(M 또는 M'=Co, Fe, Ni, Mn, Zn, Gd 또는 Cr, x와 y는 정수를 나타낸다)일 수 있다.
본 발명에 따른 방법에 있어 상기 CTC를 분리하는 방법 중 어느 것을 사용하더라도 그 효과가 동일함은 통상의 기술자에게 있어 자명하다.
(b) 상기 (a) 단계에서 분리된 CTC를 계수하는 단계;
본 발명에서, 상기 CTC를 계수하는 단계는 당업계에 공지된 방법으로 계수할 수 있다. CTC를 계수하는 방법으로는 이에 제한되는 것은 아니나, 고전적인 면역세포화학법, 레이저 주사 세포분석법; 역전사정량 중합효소 연쇄반응, 다중 역전사 중합효소 연쇄반응 등 핵산 분석법을 기반으로 한 분자분석 방법; EPISPOT assay (Epithelail Immuno-SPOT assay)와 같은 단백질 기반 분석 방법에 의할 수 있다.
(c) 상기 (b) 단계에서 계수한 값으로 유방암 예후를 예측하는 단계로서, 계수한 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계;
본 발명의 일 실시예에 따르면, c-Met 발현 CTC의 계수 결과는 유방암 환자의 예후와 큰 연관성이 있음이 확인되었다. 따라서 c-Met 발현 CTC를 계수한 결과 그 값이 낮으면 낮을수록 유방암 환자의 예후는 좋은 것으로 판단할 수 있으며, 이와 반대로 c-Met 발현 CTC를 계수한 결과 그 값이 높으면 높을수록 유방암 환자의 예후는 나쁜 것으로 판단할 수 있다.
본 발명에 있어서, 상기 c-Met 발현 CTC를 계수한 결과 그 값이 높다 혹은 낮다고 함은, 정상 대조군 시료에서 수득한 생물학적 시료로부터 분리한 c-Met 발현 CTC를 계수한 결과와 비교하였을 때, 유방암 환자로부터 수득한 생물학적 시료로부터 c-Met 발현 CTC를 계수한 값이 더 큰 경우에 높은 것으로, 더 작은 경우에 낮은 것으로 판단할 수 있다. 또는, 유방암 환자로부터 수득한 생물학적 시료로부터 c-Met 발현 CTC를 계수한 결과, 그 값이 통계적인 방법을 이용하여 산출된 일정 cut-off 이하/미만인 경우에는 좋은 예후를 갖는 것으로, 일정 cut-off 이상/초과인 경우에는 나쁜 예후를 갖는 것으로 판단할 수 있다. 상기 cut-off는 통상의 기술자가 당 업계에 널리 알려진 다양한 cut-off 산출 방법, 즉 maxstat package, Youden Index 등을 이용하여 용이하게 산출할 수 있으며, 통상의 기술자는 이렇게 산출된 적절한 값을 기준으로 삼고 cut-off를 설정하여 본 발명에 따른 예후 예측 방법에 용이하게 적용할 수 있다.
본 발명에 있어, 상기 "좋은 예후"란 재발, 전이 또는 전이성 재발의 확률이 낮고, 무질병 생존율, 무원격전이 생존율 또는 무진행 생존율이 높은 저위험군을 의미하며, 바람직하게는 무진행 생존율이 높은 것을 의미한다. "나쁜 예후"란 재발, 전이 또는 전이성 재발의 확률이 높고, 무질병 생존율, 무원격전이 생존율 또는 무진행 생존율이 낮은 고위험군을 의미하며, 바람직하게는 무진행 생존율이 낮은 것을 의미한다.
또한 본 발명은 본 발명은 유방암 환자의 예후 예측에 필요한 정보를 제공하기 위하여, 하기 단계를 포함하는 유방암 예후 예측에 필요한 정보를 제공하는 방법을 제공한다:
(a) 유방암 환자로부터 수득된 생물학적 시료로부터 cfDNA (cell-free DNA)를 분리하는 단계;
(b) 상기 (a) 단계에서 분리된 cfDNA로부터 cfDNA의 농도(concentration) 또는 cfDNA의 ESR1 유전자 돌연변이 지수(mutation index)를 계산하는 단계; 및
(c) 상기 (b) 단계에서 계산된 값으로 유방암 예후를 예측하는 단계로서, 계산된 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계.
본 발명에 따른 상기 방법에 있어서 사용된 용어는 상술한 바와 같으며, 이하에서는 상기 방법에서만 사용되는 용어에 대하여 설명한다.
본 발명에 있어서 상기 cfDNA (cell-free DNA)는 세포핵 안에 존재하지 않고 혈액에 떠돌아다니는 DNA 조각을 의미한다. 혈장에 존재하는 세포에서 유래한 DNA를 의미한다. 상기 cfDNA는 통상 이중나선구조를 가질 뿐 아니라, 코일드코일 구조를 가지는 경우가 많다. 상기 cfDNA는 종양 세포에서 유래된 것일 수 있다. 또한, 암 환자로부터 수득된 혈액, 혈장 또는 소변 등의 생물학적 시료에서는 종양 세포에서 유래된 cfDNA를 발견할 수 있다. 본 발명에서 cfDNA를 분리하는 단계는, 당 업계에 공지된 통상의 방법에 따라서 수행될 수 있으며, cfDNA를 분리하는 공지된 방법 중 어느 것을 사용하더라도 효과가 동일함은 통상의 기술자에게 있어 자명하다.
본 발명에 따른 방법에 있어서, cfDNA의 농도는 다음의 공식에 의하여 계산될 수 있다.
본 발명에 있어서 에스트로겐 수용체(Estrogen Receptor 1, ESR1)은 리간드 활성 전사(ligand-activated transcription)를 시작하며, 사이클린 패밀리의 일종인 사이클린 D1(CCND1) 및 CDK-활성 키나아제(CDK-activating kinase 7, CDK7)는 세포주기 진행과 세포 발달의 핵심 조절자이다. 정지세포(quiescent cell)가 세포주기로 진입할 때, CCND1의 발현이 에스트로겐과 에스트로겐 수용체(estrogen receptor, ER)와의 결합에 의하여 유도되며, CCND1이 CDK4 및 CDK6와 결합하여 세포 핵으로 유입된 후 CDK7에 의하여 인산화됨으로서 단백질 기질이 인산화된다. CDK7은 사이클린 H와 복합체를 이루어 CDK 활성 키나아제(CDK-activating kinase, CAK)로서 기능하며, 이들은 다양한 CDK의 활성 인산화를 유도하여 세포주기를 조절하게 된다.
본 발명에 있어서 상기 ESR1 유전자의 돌연변이는 ESR1 핫스팟 돌연변이(hot-spot mutation)을 말하며, E380, S463, V534, L536, Y537 및 D538으로 이루어진 군에서 선택된 어느 하나의 아미노산에서 일어난 돌연변이를 말한다.
본 발명에 있어서, 상기 ESR1 유전자의 돌연변이 지수(mutation index)는 다음의 공식에 의하여 계산될 수 있다.
본 발명의 일 실시예에 따르면, cfDNA의 농도(concentration) 또는 cfDNA의 ESR1 유전자의 돌연변이 지수가 유방암 환자의 예후와 큰 연관성이 있음이 확인되었다. 따라서 cfDNA의 농도가 낮거나 ESR1 유전자의 돌연변이 지수가 낮을수록 유방암 환자의 예후는 좋은 것으로 판단할 수 있으며, 이와 반대로 cfDNA의 농도가 높거나 cfDNA의 ESR1 유전자 돌연변이 지수가 높을수록 유방암 환자의 예후는 나쁜 것으로 판단할 수 있다.
본 발명에 있어서, 상기 cfDNA의 농도 또는 cfDNA의 ESR1 유전자 돌연변이 지수가 높다 혹은 낮다고 함은, 정상 대조군 시료에서 수득한 생물학적 시료로부터 분리한 cfDNA의 농도 또는 cfDNA의 ESR1 유전자 돌연변이 지수와 비교하였을 때, 유방암 환자로부터 수득한 생물학적 시료로부터 분리한 cfDNA의 농도 또는 cfDNA의 ESR1 유전자 돌연변이 지수가 더 큰 경우에 높은 것으로, 더 작은 경우에 낮은 것으로 판단할 수 있다. 또는, 유방암 환자로부터 수득한 생물학적 시료로부터 분리한 cfDNA의 농도 또는 cfDNA의 ESR1 유전자 돌연변이 지수를 통계적인 방법을 이용하여 산출된 일정 cut-off 이하/미만인 경우에는 좋은 예후를 갖는 것으로, 일정 cut-off 이상/초과인 경우에는 나쁜 예후를 갖는 것으로 판단할 수 있다. 상기 cut-off는 통상의 기술자가 당업계에 널리 알려진 다양한 cut-off 산출 방법, 즉 maxstat package, Youden Index 등을 이용하여 용이하게 산출할 수 있으며, 통상의 기술자는 이렇게 산출된 적절한 값을 기준으로 삼고 cut-off를 설정하여 본 발명에 따른 예후 예측 방법에 용이하게 적용할 수 있다.
또한 본 발명은 c-Met 발현 CTC (c-Met enriched circulating tumor cell)에 특이적으로 결합하는 제제를 포함하는 유방암 예후 예측용 조성물을 제공한다.
본 발명에서, c-Met 발현 CTC (c-Met enriched circulating tumor cell)에 특이적으로 결합하는 제제는 항체, 압타머, DNA, RNA, 단백질, 폴리펩티드로 이루어진 군에서 선택될 수 있으며, 바람직하게는 항체 또는 압타머, 가장 바람직하게는 항체일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시 양태에서, 상기 항체는 서열번호 1로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L1, 서열번호 2로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L2 및 서열번호 3으로 표시되는 아미노산 서열을 포함하는 상보성 결정 부위(CDR) L3를 포함하는 항체 경쇄가변영역(VL) 및 서열번호 4로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H1, 서열번호 5로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H2 및 서열번호 6으로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H3를 포함하는 항체 중쇄가변영역(VH)을 포함하는 c-Met 단백질에 특이적으로 결합하는 항체 또는 그 단편일 수 있다.
한편, 본 발명의 다른 일 실시 양태에서, 상기 항체는 서열번호 7로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L1, 서열번호 8로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L2 및 서열번호 9으로 표시되는 아미노산 서열을 포함하는 상보성 결정 부위(CDR) L3를 포함하는 항체 경쇄가변영역(VL) 및 서열번호 10로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H1, 서열번호 11로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H2 및 서열번호 12으로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H3를 포함하는 항체 중쇄가변영역(VH)을 포함하는 c-Met 단백질에 특이적으로 결합하는 항체 또는 그 단편일 수 있다.
본 발명에서, '항체', '항 c-Met 항체', '인간화 항 c-Met 항체' 및 '변형 인간화 항 c-Met 항체', 'anti-c-Met antibody'는 본 발명에서 가장 광의의 의미로 사용되며, 구체적으로 단일클론 항체(모노클로날 항체, 완전 길이 단일클론항체 포함), 다클론 항체(폴리클로날 항체), 다중 특이 항체(예를 들어, 이중 특이 항체), 및 항체 단편(예를 들어, 가변 영역 및 목적하는 생물 활성(예를 들어, c-Met과의 결합)을 나타내는 항체의 다른 부분)을 포함한다.
본 발명의 항체는 c-Met과 선택적으로 결합할 수 있도록 특정 아미노산 서열이 경쇄 및 중쇄 CDR에 포함되어 있는 항체로 모노클로날 항체 및 폴리클로날 항체를 모두 포함하며, 바람직하게는 모노클로날 항체일 수 있다. 또한 본 발명의 항체는 키메라 항체, 인간화된 항체, 인간항체를 모두 포함하며 바람직하게는 인간항체 일 수 있다.
본 발명의 모노클로날 항체는 실질적으로 동질 항체의 집단으로부터 수득된 항체를 나타내며, 즉, 집단을 구성하는 개개의 항체는 소량으로 존재할 수 있는 가능한 천연적으로 존재하는 돌연변이를 제외하고는 동일하다. 모노클로날 항체는 단일 항원 에피토프에 매우 특이적으로 결합한다.
본 발명에서 '모노클로날'이란 항체가 실질적인 상동성 집단으로부터 수득되는 것과 항체의 특성을 나타내는 말이며, 반드시 항체를 특정 방법에 의해 생산해야 한다는 것은 아니다. 예를 들어, 본 발명의 모노클로날 항체는 당업계에 최초로 공지된 하이브리도마 방법에 의해 제조할 수 있거나, 또는 재조합 DNA 방법(참조: 미국 특허 제4,816,567호)에 의해 제조할 수 있다. 또한, 당업계에 기술된 기술을 사용하여 파아지 항체 라이브러리로부터 분리할 수 있다.
본 발명의 항체는 구체적으로 키메라 항체를 포함하며, 이 경우 중쇄 및/또는 경쇄의 일부는 특정 종으로부터 기원하거나 또는 특정 항체의 상응하는 서열과 동일하거나 상동성을 보이지만, 나머지 부분은 본 발명의 항체가 바람직한 생물학적 활성(예를 들어 NRS와의 선택적 결합)을 나타내는 한, 다른 종으로부터 기원하거나 또는 다른 항체의 상응하는 서열과 동일하거나 상동성을 나타내는 것이어도 무방하다(미국 특허 제4,816,567호).
인간화된 항체는 인간 및 비-인간(예: 쥐, 랫트) 항체의 서열을 모두 포함하는 항체로 일반적으로, 에피토프와 결합하는 부위(CDR)를 제외한 나머지 부분은 인간 항체의 것이며, 에피토프와 결합하는 부위(CDR)는 비-인간 유래의 서열을 포함할 수 있다. 완전한 인간항체는 사람 면역글로불린 단백질 서열만을 포함하는 항체를 말하며, 마우스, 마우스 세포, 또는 마우스 세포로부터 기원한 하이브리도마에서 생산하거나, 파지 디스플레이 방법으로 생산할 수 있다.
생체에서 생산되는 천연 항체는 통상적으로, 2개의 동일한 경쇄(L)와 2개의 동일한 중쇄(H)로 구성된, 약 150,000 달톤의 이종-사량체성 당단백질이다. 각 경쇄는 1개의 공유 디설파이드 결합에 의해 중쇄와 연결되지만, 디설파이드 연쇄수는 상이한 면역글로불린 이소형의 중쇄들 간에 다양하다. 각 중쇄 및 경쇄는 규칙적으로 이격된 쇄내 디설파이드 브릿지를 또한 갖고 있다. 각 중쇄는 한 말단에 가변 도메인(VH)에 이어 수많은 불변 도메인을 갖는다. 각 경쇄는 한 말단에 가변 도메인(VL)을 갖고, 그의 다른 말단에 불변 도메인을 갖는데; 경쇄의 불변 도메인은 중쇄의 제1 불변 도메인과 정렬되고, 경쇄 가변 도메인은 중쇄의 가변 도메인과 정렬된다. 특별한 아미노산 잔기가 경쇄 가변 도메인과 중쇄 가변 도메인 간에 계면을 형성하는 것으로 여겨진다. 항체의 "가변 영역" 또는 "가변 도메인"은 항체의 중쇄 또는 경쇄의 아미노-말단 도메인을 지칭한다. 중쇄의 가변 영역은 "VH"로 기재하며, 경쇄의 가변 영역은 "VL"로 기재한다. 이들 도메인은 일반적으로, 항체의 가장 가변 부분이고, 항원 결합 부위를 포함한다.
본 발명에서 '초가변성(hypervariable)'은 상기 가변 영역 내의 몇몇 서열들이 항체들간 서열에 있어서 광범위하게 상이하며 그의 특이적인 항원 결정인자들에 대한 각각의 특정 항체의 결합 및 특이성에 직접적으로 관련되는 잔기들을 포함한다는 사실을 지칭한다. 경쇄 및 중쇄 가변 영역 모두에 있어서 초가변성은 상보성 결정부위(CDR) 또는 초가변성 루프(HVL)로서 공지된 3 개의 분절들에 집중된다. CDR은 당업계에 공지된 서열 비교에 의해 한정되는 반면, HVL은 당업계에 공지된 바와 같이, 상기 가변 영역의 3차원 구조에 따라 구조적으로 한정된다.
상기 중쇄 및 경쇄 각각 내의 3 개의 CDR들은 틀 부위(FR)에 의해 분리되며, 상기 부위는 덜 가변적인 경향이 있는 서열들을 포함한다. 상기 중쇄 및 경쇄 가변 영역의 아미노 말단에서부터 카복시 말단까지, 상기 FR 및 CDR은 하기의 순서로 배열된다: FR1, CDR1, FR2, CDR2, FR3, CDR3 및 FR4. 상기 FR의 큰 β 시트 배치는 상기 각각의 쇄 내부의 CDR을 서로뿐만 아니라 다른 쇄로부터의 CDR에 가깝게 한다. 생성되는 형태는 항원 결합 부위에 기여하지만, 모든 CDR 잔기들이 항원 결합에 직접 관여할 필요는 없다.
본 발명의 상기 단편은 디아바디, Fab, Fab', F(ab)2, F(ab')2, Fv 및 scFv로 이루어진 군에서 선택되는 단편인 것을 특징으로 한다.
본 발명에서 항체의 단편은 전체 항체의 항원 특이적 결합력을 유지하고 있는 항체의 단편을 의미하며, 바람직하게 상기 단편은 모항체의 인간 유래 c-Met 단백질 친화도의 적어도 20%, 50%, 70%, 80%, 90%, 95% 또는 100% 또는 그 이상을 보유한다. 구체적으로는 Fab, F(ab)2, Fab', F(ab')2, Fv, 디아바디(diabody), scFv 등의 형태일 수 있다.
Fab (fragment antigen-binding)는 항체의 항원 결합 단편으로, 중쇄와 경쇄 각각의 하나의 가변 도메인과 불변 도메인으로 구성되어 있다. F(ab')2는 항체를 펩신으로 가수분해시켜서 생성되는 단편으로, 두 개의 Fab가 중쇄 경첩(hinge)에서 이황결합(disulfide bond)으로 연결된 형태를 하고 있다. F(ab')는 F(ab')2 단편의 이황결합을 환원하여 분리시킨 Fab에 중쇄 경첩이 부가된 형태의 단량체 항체 단편이다. Fv (variable fragment)는 중쇄와 경쇄 각각의 가변영역으로만 구성된 항체 단편이다. scFv (single chain variable fragment)는 중쇄가변영역(VH)과 경쇄가변영역(VL)이 유연한 펩티드 링커로 연결되어 있는 재조합 항체 단편이다. 디아바디(diabody)는 scFv의 VH와 VL가 매우 짧은 링커로 연결되어 서로 결합하지 못하고, 동일한 형태의 다른 scFv의 VL와 VH와 각각 결합하여 이량체를 형성하고 있는 형태의 단편을 의미한다.
본 발명의 목적상 항체의 단편은 인간 유래 c-Met 단백질에 대한 결합 특이성을 유지하고 있는 것이라면 구조나 형태의 제한을 받지 않지만, 바람직하게 scFv일 수 있다. 본 발명에 따른 scFv는 상기한 인간 유래 c-Met 단백질에 특이적인 CDR 구성, 또는 VH와 VL의 구성을 갖는 것으로서 VH의 C-말단과 VL의 N-말단이 링커를 통해 연결된 것이라면 그 서열이 특별히 제한되지 않는다. 상기 링커는 당 업계에 scFv에 적용되는 링커로서 알려진 것이라면 그 종류가 특별히 제한되지 않는다.
본 발명의 항체 또는 그 단편은 이의 생물학적 활성을 실질적으로 변경하지 않는 보존적 아미노산 치환(항체의 보존적 변이체라고 함)을 포함할 수 있다.
또한 전술한 본 발명의 항체 또는 그 단편은 효소, 형광 물질, 방사선 물질 및 단백질 등과 접합된 것일 수 있으나, 이에 한정되지는 않는다. 또한, 항체에 상기 물질을 접합하는 방법은 당업계에 잘 알려져 있다.
본 발명의 항체는 인간을 포함하는 포유동물, 조류 등을 포함한 임의의 동물로부터 유래한 것일 수 있다. 바람직하게는, 상기 항체는 인간, 생쥐, 당나귀, 양, 토끼, 염소, 기니피그, 낙타, 말 또는 닭의 항체일 수 있으며, 가장 바람직하게는 인간 또는 생쥐일 수 있다.
인간 항체는 인간 면역글로불린의 아미노산 서열을 가진 항체로서, 인간 면역글로불린 라이브러리로부터 분리된 항체 또는 하나 이상의 인간 면역글로불린에 대하여 형질 이식되고 내재적 면역글로불린은 발현하지 않는 동물로부터 분리된 항체가 포함된다(미국특허 제 5,939,598호 참조).
본 발명의 항체는 효소, 형광 물질, 방사선 물질 및 단백질 등과 접합된 것일 수 있으나, 이에 한정되지 않는다. 또한, 항체에 상기 물질을 접합하는 방법은 당업계에 잘 알려져 있다.
본 발명은 유방암 예후 예측용 조성물을 제조하기 위한 c-Met 발현 CTC (c-Met enriched circulating tumor cell)에 특이적으로 결합하는 제제의 용도를 제공한다.
본 명세서에서 용어 "을 포함하는(comprising)"이란 "함유하는(including)" 또는 "특징으로 하는(characterized by)"과 동일한 의미로 사용되며, 본 발명에 따른 조성물 또는 방법에 있어서, 구체적으로 언급되지 않은 추가적인 구성 성분 또는 방법의 단계 등을 배제하지 않는다. 또한 용어 "로 이루어지는(consisting of)"이란 별도로 기재되지 않은 추가적인 요소, 단계 또는 성분 등을 제외하는 것을 의미한다. 용어 "필수적으로 이루어지는(essentially consisting of)"이란 조성물 또는 방법의 범위에 있어서, 기재된 물질 또는 단계와 더불어 이의 기본적인 특성에 실질적으로 영향을 미치지 않는 물질 또는 단계 등을 포함할 수 있는 것을 의미한다.
따라서, 본 발명은 c-Met 발현 CTC를 이용한 유방암의 예후 예측 방법 및 c-Met 발현 CTC에 특이적으로 결합하는 제제를 포함하는 유방암 예후 예측용 조성물을 제공한다. 본 발명의 방법 및 조성물을 이용하면 유방암 환자의 조직검사 없이도 정확하게 유방암 환자의 예후 예측이 가능하다.

이하 본 발명을 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
실시예 1. 유방암 환자의 CTC 분리 및 계수
1-1. 유방암 환자의 CTC 분리
치료를 받는 HR+ 전이성 유방암 환자 중 연구에 동의한 환자 97명을 대상으로 전혈 10 mL를 채혈하였다. 이 중 RBC 오염이 있었던 4명을 제외한 93명을 대상으로 분석하였다. 검체는 수집 순서대로 번호를 부여하여 모두 익명화하였다. 채혈한 혈액은 cell-free DNA blood collection tube (treck, La Viasta, NE, USA)에 곧바로 넣은 후, 튜브를 위아래로 뒤집어 항응고제와 충분히 혼합한다. 검체는 CTC (circulating tumor cell, 순환 종양 세포) 분리 전까지 상온에 보관하였다. 검체는 미세유체와 자성을 통해 특정 단백질을 특이적으로 분리하는 GenoCTC V2 및 분리 키트 (Genobio Corp, Seoul, South Korea)를 사용하여 EpCAM 발현 CTC와 c-MET 발현 CTC를 다음의 과정을 거쳐 분리하였다.
10 mL 혈액을 나누어 4 mL 전혈에는 항-c-MET 항체와 자성 비드가 결합된 물질을, 4 mL 전혈에는 항-EpCAM 항체와 자성 비드가 결합된 물질을 넣고 상온에서 30분간 반응시켰다. 자성 비드가 결합된 항체와의 반응이 끝나면, GenoCTC 장비에 검체를 넣고 제조사에서 제공하는 프로토콜에 따라 순환 종양 세포를 분리하였다. 분리가 완료된 후 원하는 물질이 분리되는 central line에서 수집된 용액에 최종적으로 0.1% BSA가 되도록 2% BSA를 첨가하였다. 그 후 100g에서 5분간 원심분리를 수행하였다. 파이펫으로 상등액을 제거한 후, 잔여량은 잘 섞어서 슬라이드 글라스에 올린 다음 heat block을 40℃로 설정하고 슬라이드 글라스를 얹어서 용액을 증발시켰다.
말초혈액단핵구는 Ficoll-paque를 사용하여 매뉴얼에서 제시하는 방법에 따라 분리하였다. PBS와 전혈을 1:1로 섞은 후, Ficoll-paque를 담은 conical tube에 층이 유지되도록 조심히 넣고, 400g에서 30분간 원심분리를 수행하여 말초혈액단핵구를 얻은 다음, PBS를 넣고 100g에서 5분간 원심분리를 수행하여 세척하였다. 분리한 말초혈액단핵구도 40℃로 설정한 heat block에 올린 후 용액을 증발시켰다.
1-2. 분리한 CTC의 염색
용액이 증발된 후 GenoCTC profiling kit (Genobio corp.)를 사용하여 면역세포 화학염색을 진행하였다. 슬라이드 위에 부착된 세포를 고정하고 투과(permeabilization) 하기 위한 용액을 로딩 한 다음, 상온에서 10분간 반응시켰다. 그 후 PBS로 세척한 뒤 2% BSA로 상온에서 10분간 blocking을 진행하였다. 이후 형광이 부착된 항-CK18 항체와 항-CD45 항체를 넣고 상온에서 1시간 반응시켜 염색을 수행하였다. 형광이 부착된 항체이므로 은박지 등을 활용하여 빛을 차단한 뒤 반응시켰다. 반응이 완료되면 PBS로 세척하여 부착되지 않은 잔여 항체를 제거하고, DAPI로 핵을 염색한 뒤 커버슬라이드를 덮어 슬라이드를 고정하였다.
실시예 2. CTC의 계수에 따른 무진행 생존기간 및 cox 위험 비례모형 분석
형광 현미경을 사용하여 앞선 실시예 1에서 슬라이드에 부착된 세포를 계수하였다. HR+ 전이성 유방암 환자 97명 중 CTC 분리 도중에 RBC 오염이 있었던 4 case는 제외하고, 총 93명의 데이터로 결과를 분석하였다. 순환 종양 세포는 CK18+, CD45-, DAPI+인 세포를 순환 종양 세포로 정의한다. CD45 형광의 감도는 PBMC를 부착한 슬라이드를 기준으로 조정한 후, 순환 종양 세포를 분리한 슬라이드에도 적용하였다. 순환 종양 세포는 사진을 촬영하고 계수 결과를 기재하였다.
환자 모집이 완료된 후, 임상병리학적 정보와 무진행 생존기간 등에 대한 정보를 독립적으로 수집하였다. 무진행 생존기간은 RECIST v1.1에 따라 영상의학적 판독을 기반으로 정의하며, 채혈일로부터 진행과 사망 중 먼저 발생한 사건의 발생 시점을 기준으로 계산하였다. 통계학적 분석은 R programming environment (ver. 1.4.1103)을 활용하였으며, cut-off의 기준은 standardized log-rank statistics를 기반으로 cut-off를 산정하는 maxstat R package를 통해 계산하였다. 생존곡선 분석 후 cox 위험 비례모형을 통해 c-Met 발현 CTC 및 EpCAM 발현 CTC와 다른 임상병리학적 정보와 진행(progression)과의 연관성을 분석하였다. 통계학적으로 유의미한 기준은 p-value < 0.05로 설정하여 분석하였다. 이에 대한 결과를 도 1a 및 b 및 표 2에 나타냈다.
그 결과, 도 1a 및 b에서 보는 바와 같이, HR+ 전이성 유방암 환자에서 c-Met 발현 CTC 계수는 무진행 생존기간과 유의미한 연관성이 인정되는 인자임을 확인할 수 있었다(도1a). 이와 달리 EpCAM 과발현 CTC의 경우 무진행 생존기간과 통계적으로 유의한 연관성을 보이지 않았다(도 1b).
나아가 도 2 a 및 b 및 도 3 a 및 b에서 보는 바와 같이, HR+/HER2- 전이성 유방암 환자에서는 c-Met 발현 CTC 계수가 무진행 생존기간과 현저하게 유의미한 연관성이 있음을(도 2a), HR+/HER2+ 전이성 유방암 환자에서도 c-Met 발현 CTC 계수가 무진행 생존기간과 유의미한 연관성이 있음을 확인할 수 있었다(도 3a).
또한 Cox 위험 비례모형에 따라 임상병리학적 정보와 CTC 계수 결과의 단변량(Univariate) 및 다변량(Multivariate) 분석을 수행한 결과, cMet 발현 CTC는 통계적으로 유의하게 HR+ 유방암 환자의 무진행 생존에 대한 예측인자임을 확인할 수 있었다(표 2 참조).
실시예 3. cfDNA 농도에 따른 생존분석
상기 1-1과 동일한 환자를 대상으로 채혈 및 분석 대상을 정하였다. 혈액은 cell-free DNA blood collection tube (Streck, La Viasta, NE, USA)을 이용하여 채취하였으며, 환자 특성 및 종양 조직학 세부 정보는 삼성서울병원(SMC)(서울, 대한민국) 병리과의 병리 보고서에서 추출하였다. 질병 진행은 고형 종양의 반응 평가 기준(RECIST, 버전 1.1) 가이드라인에 따라 방사선 이미지를 사용하여 평가하였다. 무진행 생존기간(PFS)은 채혈 시점에서 방사선학적인 질병 진행 또는 모든 원인에 의한 사망까지의 기간으로 정의하였다.
혈액 샘플을 4℃에서 15분간 3000x g로 원심분리하여 혈장을 분리한 다음, QIAamp Circulating Nucleic Acid Kit (Qiagen, Hilden, Germany)를 이용하여 제조사에서 제공하는 프로토콜에 따라 4~6 mL 혈장 샘플에서 cfDNA를 추출하였다. cfDNA는 100 μL elution buffer에서 용출되었다. Droplet PIK3CA 및 Droplex ESR1 Mutation Test Kits (Gencurix Inc., 서울, 대한민국)을 이용하여 제조사에서 제공한 프로토콜에 따라 cfDNA 샘플의 ESR1 및 PIK3CA 돌연변이를 분석하였다. ddPCR 시약을 용출된 12.9 μL의 cfDNA/well과 8-strip PCR 튜브에서 혼합하였다. 프라이머와 프로브 세트는 PIK3CA 핫스팟 돌연변이(R88Q, N345K, E542, E545, Q546, E726, H1047, M1048, G1049) 및 ESR1 핫스팟 돌연변이(E380, S463, V534, L536, Y537, D538)를 검출하도록 설계되었다. 양성 및 음성 대조군을 반응 혼합물과 혼합하여 8-strip PCR 튜브에 넣은 다음 이 혼합물을 QX200TM Droplet Generator (Bio-Rad, Hercules, CA, USA)에 넣고 droplet으로 전환하였다. 이 droplet에 대하여 96 well plate에서 PCR을 실시하였다. 증폭이 완료된 후, QX200TM Droplet Reader (Bio-Rad)를 이용하여 droplet을 계수하였다. 대조군(internal control)은 PIK3CA 또는 ESR1을 검출하도록 설계되었으며, cfDNA 농도(cfDNA concentration) 및 돌연변이 지수(mutation index)의 지표로서 사용되었다.
cfDNA 농도(cfDNA concentration) 및 돌연변이 지수(mutation index)는 다음 식에 의해 계산하였다:
cfDNA 농도는 Droplex ESR1 Mutation Test Kits의 IC 카피를 이용하여 계산되었다. 환자들은 HR+ mBC의 PFS에서 최대 통계적 유의성이 있는 것으로 확인된 혈장 1490 카피/mL의 컷오프 값을 사용하여 cfDNA 농도가 높거나 낮은 것으로 분류되었으며, 그 결과를 도 4a 내지 f 및 도 5a 내지 d에 도시하였다.
도 4 a 내지 f는 cfDNA 농도가 높은 군과 낮은 군을 나누어 도시한 결과이며, 그 결과 HR+/HER2- mBC 환자 군에서 cfDNA 농도가 높은 그룹의 환자의 예후가 좋지 않음을 확인할 수 있었다(도 4c 참조).
또한, 도 5a 내지 d는 HR+/HER2- mBC 환자군에서 cMet 발현 CTC 및 cfDNA 농도에 따라 그룹을 나누어 도시한 결과이며, 그 결과 cMet 발현 CTC와 cfDNA 농도가 모두 낮은 환자와 그렇지 않은 환자에서 통계적으로 유의하게 예후에 있어서 차이가 있음을 확인하였다(도 5b 참조). 이는 EpCAM 발현 CTC 및 cfDNA 농도에 따라 그룹을 나누어 분석한 결과도 동일하였다(도 5d 참조).
상기 결과를 바탕으로 Cox 회귀 다변량 분석(multivariate analysis)를 수행한 결과, HR+/HER2- mBC 환자군에서 c-Met 발현 CTC, cfDNA 농도, EpCAM 발현 CTC는 유방암의 예후 예측 판단에 있어서 독립적인 인자임을 확인할 수 있었다(도 6 참조).
이상 살펴본 바와 같이 본 발명은 c-Met 발현 CTC를 이용한 유방암의 예후 예측 방법 및 c-Met 발현 CTC에 특이적으로 결합하는 제제를 포함하는 유방암 예후 예측용 조성물을 제공한다. 본 발명의 방법 및 조성물은 유방암 환자의 조직검사 없이도 정확하게 유방암 환자의 예후 예측을 하는데 유용하게 이용될 수 있다.
Claims (12)
- 유방암 환자의 예후 예측에 필요한 정보를 제공하기 위하여, 하기 단계를 포함하는 유방암 예후 예측에 필요한 정보를 제공하는 방법:(a) 유방암 환자로부터 수득된 생물학적 시료로부터 c-Met 발현 CTC (c-Met enriched circulating tumor cell)를 분리하는 단계;(b) 상기 (a) 단계에서 분리된 CTC를 계수하는 단계; 및(c) 상기 (b) 단계에서 계수한 값으로 유방암 예후를 예측하는 단계로서, 계수한 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계.
- 제1항에 있어서, 상기 유방암은 HR+/HER2- 또는 HR+/HER2+ 유방암인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 유방암 예후는 무질병 생존율, 무원격전이 생존율 또는 무진행 생존율인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 생물학적 시료는 혈액, 혈장, 혈청, 전혈, 단리된 혈액 세포, 골수액, 림프액, 타액, 뇨, 점막액, 양수, 또는 이들의 조합인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 c-Met 발현 CTC를 분리하는 단계는 상기 CTC (circulating tumor cell)에 특이적으로 결합하는 항체 및 상기 항체에 결합되는 자성 비드로 분리하는 것을 특징으로 하는 방법.
- 유방암 환자의 예후 예측에 필요한 정보를 제공하기 위하여, 하기 단계를 포함하는 유방암 예후 예측에 필요한 정보를 제공하는 방법:(a) 유방암 환자로부터 수득된 생물학적 시료로부터 cfDNA (cell-free DNA)를 분리하는 단계;(b) 상기 (a) 단계에서 분리된 cfDNA로부터 cfDNA의 농도(concentration) 또는 cfDNA의 ESR1 유전자 돌연변이 지수(mutation index)를 계산하는 단계; 및(c) 상기 (b) 단계에서 계산된 값으로 유방암 예후를 예측하는 단계로서, 계산된 값이 낮으면 좋은 예후인 것으로 유방암 예후 예측에 필요한 정보를 제공하는 단계.
- c-Met 발현 CTC (c-Met enriched circulating tumor cell)에 특이적으로 결합하는 제제를 포함하는 유방암 예후 예측용 조성물.
- 제6항에 있어서, 상기 제제는 항체, 압타머, DNA, RNA, 단백질, 폴리펩티드로 이루어진 군에서 선택되는 적어도 하나인, 유방암 예후 예측용 조성물.
- 제7항에 있어서, 상기 항체는 서열번호 1로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L1, 서열번호 2로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L2 및 서열번호 3으로 표시되는 아미노산 서열을 포함하는 상보성 결정 부위(CDR) L3를 포함하는 항체 경쇄가변영역(VL) 및 서열번호 4로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H1, 서열번호 5로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H2 및 서열번호 6으로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H3를 포함하는 항체 중쇄가변영역(VH)을 포함하는 c-Met 단백질에 특이적으로 결합하는 항체 또는 그 단편인 것을 특징으로 하는 유방암 예후 예측용 조성물.
- 제7항에 있어서, 상기 항체는 서열번호 7로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L1, 서열번호 8로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) L2 및 서열번호 9으로 표시되는 아미노산 서열을 포함하는 상보성 결정 부위(CDR) L3를 포함하는 항체 경쇄가변영역(VL) 및 서열번호 10로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H1, 서열번호 11로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H2 및 서열번호 12으로 표시되는 아미노산 서열을 포함하는 상보성 결정부위(CDR) H3를 포함하는 항체 중쇄가변영역(VH)을 포함하는 c-Met 단백질에 특이적으로 결합하는 항체 또는 그 단편인 것을 특징으로 하는 유방암 예후 예측용 조성물.
- 제7항에 또는 제8항에 있어서, 상기 단편은 디아바디, Fab, Fab', F(ab)2, F(ab')2, Fv 및 scFv로 이루어진 군에서 선택되는 단편인 것을 특징으로 하는 유방암 예후 예측용 조성물.
- 유방암 예후 예측용 조성물을 제조하기 위한 c-Met 발현 CTC (c-Met enriched circulating tumor cell)에 특이적으로 결합하는 제제의 용도.
