Abstract
本発明は、簡便なプロセスで医療デバイスの表面に優れた生体適合性を付与すると共に、その生体適合性を長期間に亘って維持することが可能な、共重合体およびその製造方法等を提供することを目的とする。本発明は、下記(A)、(B)および(C)をモノマー成分として含む、共重合体を提供する。 (A)カルボキシベタイン基および/またはスルホベタイン基を有する化合物 (B)(メタ)アクリル酸 (C)アクリルアミド誘導体
Description
本発明は、共重合体、医療用組成物および被覆医療デバイス並びにそれらの製造方法に関する。
コンタクトレンズやステントのような生体に直接接触する医療デバイスは生体適合性を有する必要がある。コンタクトレンズは、安全性の確保や装用感向上のために、その表面が水濡れ性、易滑性および防汚性といった生体適合性を有する必要がある。ステントは、体内に留置可能なインプラント医療デバイスであり、血栓、血小板、粘液などの付着に起因した合併症の発生が大きな課題とされている。特に呼吸用のステントにおいて、粘液付着を抑制し、かつ生体適合性を改善した、長期間使用可能なステントは大きな臨床ニーズがある。医療デバイスの表面に優れた生体適合性を付与するための代表的な手法としては、医療デバイスの表面処理が知られている。
医療デバイスの表面処理方法としては、互いに性質の異なるポリマーを積層してコーティングする方法(特許文献1)、両性電解質化合物を含むポリマーをコーティングする方法(特許文献2)、あるいは、N-(メタ)アクリロイルオキシエチル-N,N-ジメチルアンモニウム-α-N-メチルカルボキシベタインとブチルメタクリレートとのランダム共重合体をコーティングする方法(特許文献3)等が知られている。
しかしながら、従来の医療デバイスの表面処理方法は煩雑な操作を含むものが多く、さらには表面処理により得られた生体適合性の耐久性が十分ではないのが現状であった。
そこで本発明は、簡便なプロセスで医療デバイスの表面に優れた生体適合性を付与すると共に、摩擦刺激にさらされてもその生体適合性を維持することが可能な、共重合体およびその製造方法等を提供することを目的とする。
上記の課題を達成するために、本発明は下記(A)、(B)および(C)をモノマー成分として含む、共重合体を提供する:
(A)カルボキシベタイン基および/またはスルホベタイン基を有する化合物;
(B)(メタ)アクリル酸;
(C)アクリルアミド誘導体。

(C)アクリルアミド誘導体。
本発明によれば、煩雑な操作を伴うことなく医療デバイスの表面に優れた生体適合性を付与することができ、かつ、摩擦刺激にさらされてもその生体適合性を維持することが可能な共重合体を提供することができる。
本発明の共重合体は、(A)カルボキシベタイン基および/またはスルホベタイン基を有する化合物、(B)(メタ)アクリル酸、および、(C)アクリルアミド誘導体をモノマー成分として含む多元共重合体(以下、「共重合体T」)である。
本発明の共重合体は非架橋型であり、線状であっても分枝状であっても構わないが、製造の容易さの観点からは、線状が好ましい。本発明の共重合体は、ランダム共重合体、交互共重合体、ブロック共重合体またはグラフト共重合体のいずれであっても構わない。製造の容易さの観点からは、ランダム共重合体が好ましい。
本発明の共重合体の原料となる「モノマー成分」とは、光重合開始剤および/または熱重合開始剤を用いて重合および/または架橋させることができる、低分子量化合物をいう。また、共重合体がモノマー成分として前記(A)、(B)および(C)を含むとは、共重合体が、前記モノマー成分(A)、(B)および(C)に由来する構造を含むことを言う。
モノマー成分の一つである(A)「カルボキシベタイン基および/またはスルホベタイン基を有する化合物」(以下、「モノマー成分(A)」)としては、例えば、2-[[2-(メタクリロイルオキシ)エチル]ジメチルアンモニオ]酢酸、3-[[2-(メタクリロイルオキシ)エチル]ジメチルアンモニオ]プロパン酸(以下、「CBMA」)、3-[(3-アクリルアミドプロピル)ジメチルアンモニオ]プロパン酸(以下、「CBAA」)、3-[[2-(メタクリロイルオキシ)エチル]ジメチルアンモニオ]プロパン-1-スルホン酸(以下、「SBMA」)、4-[[2-(メタクリロイルオキシ)エチル]ジメチルアンモニオ]ブタン-1-スルホン酸、3-[[2-(アクリロイルオキシ)エチル]ジメチルアンモニオ]プロパン-1-スルホン酸(SPDA)、ビス[2-(メタクリロイルオキシ)エチル](メチル)アンモニオ]プロパン-1-スルホン酸、3-[(3-メタクリルアミドプロピル)ジメチルアンモニオ]プロパン-1-スルホン酸、4-[(3-メタクリルアミドプロピル)ジメチルアンモニオ]ブタン-1-スルホン酸または3-[(3-アクリルアミドプロピル)ジメチルアンモニオ]プロパン-1-スルホン酸が挙げられる。タンパク質の非吸着特性の観点から、CBMAまたはSBMAが好ましい。
モノマー成分の一つである(B)「(メタ)アクリル酸」(以下、「モノマー成分(B)」)とは、メタクリル酸および/またはアクリル酸(以下、「AA」)をいう。重合安定性の観点から、アクリル酸が好ましい。
モノマー成分の一つである(C)「アクリルアミド誘導体」(以下、「モノマー成分(C)」)としては、例えば、アクリルアミド、N-イソプロピルアクリルアミド、N,N-ジメチルアクリルアミド(以下、「DMAA」)、N,N-ジエチルアクリルアミド、N-メチル-N-エチルアクリルアミドまたはN,N-ジ(ヒドロキシエチル)アクリルアミドが挙げられる。重合安定性の観点から、DMAAが好ましい。
本発明の共重合体に占めるモノマー成分(A)、モノマー成分(B)、および、モノマー成分(C)の割合は、モノマー成分(A)が1~40モル%であることが好ましく、モノマー成分(B)が1~35モル%であることが好ましく、モノマー成分(C)が25~98モル%であることが好ましい。
本発明の共重合体に占めるモノマー成分(A)の割合は、1~30モル%であることがより好ましく、5~25モル%であることがさらに好ましい。
本発明の共重合体に占めるモノマー成分(B)の割合は、1~25モル%であることがより好ましく、5~15モル%であることがさらに好ましい。
本発明の共重合体に占めるモノマー成分(C)の割合は、45~98モル%であることがより好ましく、60~90モル%であることがさらに好ましい。
本発明の共重合体の製造方法は、(A)カルボキシベタイン基および/またはスルホベタイン基を有する化合物、(B)(メタ)アクリル酸、並びに、(C)アクリルアミド誘導体、を含むモノマーを重合して、共重合体を得る、重合工程を備える。
上記重合工程において各モノマー成分を重合および/または架橋させるために、熱重合開始剤や光重合開始剤等の重合開始剤を用いることができる。
熱重合開始剤としては、例えば、過酸化物またはアゾ化合物が挙げられる。熱重合開始剤は所望の反応温度に対して最適な分解特性を有するものを選択すればよいが、一般的には、10時間半減期温度が40~120℃のアゾ化合物または過酸化物が好ましい。
より具体的には、例えば、2,2’-アゾビス(イソブチロニトリル)(AIBN)、2,2’-アゾビス(2-メチルブチロニトリル)、2,2’-アゾビス(2,4-ジメチルバレロニトリル)、1,1’-アゾビス(シクロヘキサン-1-カルボニトリル)、2,2’-アゾビス[2-(2-イミダゾリン-2-イル)プロパン]、2,2’-アゾビス(2-メチルプロピオンアミジン)二塩酸塩(以下、「V-50」)、2,2’-アゾビス[N-(2-カルボキシエチル)-2-メチルプロピオンアミジン]、2,2’-アゾビス[2-メチル-N-(2-ヒドロキシエチル)プロピオンアミド]、2,2’-アゾビス(N-ブチル-2-メチルプロピオンアミド)、2,2’-アゾビス(2-メチルプロピオン酸)ジメチル、4,4’-アゾビス(4-シアノ吉草酸)、tert-ブチルヒドロペルオキシド、クメンヒドロペルオキシド、ジ-tert-ブチルペルオキシドまたは過酸化ベンゾイルが挙げられる。
光重合開始剤としては、例えば、芳香族α-ヒドロキシケトン、アルコキシオキシベンゾイン、アセトフェノン、アシルホスフィンオキシド若しくはビスアシルホスフィンオキシド等のカルボニル系化合物第三級アミン+ジケトン、過酸化物、アゾ化合物、硫黄化合物、ハロゲン化合物または金属塩が挙げられる。
より具体的には、例えば、1-ヒドロキシシクロヘキシルフェニルケトン、2-ヒドロキシ-2-メチル-1-フェニル-プロパン-1-オン、ビス(2,6-ジメトキシベンゾイル)-2,4,4-トリメチルペンチルホスフィンオキシド(DMBAPO)、ビス(2,4,6-トリメチルベンゾイル)フェニルホスフィンオキシド(“イルガキュア”(登録商標)819)、2,4,6-トリメチルベンジルジフェニルホスフィンオキシドおよび2,4,6-トリメチルベンゾイルジフェニルホスフィンオキシド、ベンゾインメチルエーテルまたはカンファーキノンとエチル4-(N,N-ジメチルアミノ)ベンゾエートとの組み合わせが挙げられる。
これらの重合開始剤は単独で用いても混合して用いても構わないが、モノマー成分の重合や架橋を適度に促進させつつ、所望の分子量の共重合体を得るため、重合混合物に占める重合開始剤の割合は、5質量%以下が好ましい。
ここで「重合混合物」とは、共重合体を得るための各モノマー成分、重合開始剤、および、必要に応じて添加されても構わない連鎖移動剤を溶媒に溶解させた混合物をいう。
重合混合物を得るための溶媒としては、例えば、水、メタノール、エタノール、プロパノール、2-プロパノール、ブタノール、2-ブタノール、tert-ブタノール、tert-アミルアルコール、3,7-ジメチル-3-オクタノール、エチレングリコール、ジエチレングリコール、トリエチレングリコール、テトラエチレングリコール若しくはポリエチレングリコール等のアルコール系溶媒;ベンゼン、トルエン若しくはキシレン等の芳香族炭化水素系溶媒;ヘキサン、ヘプタン、オクタン、デカン、石油エーテル、ケロシン、リグロイン若しくはパラフィン等の脂肪族炭化水素系溶媒;シクロペンタン、シクロヘキサン若しくはエチルシクロヘキサン等の脂環族炭化水素系溶媒;アセトン、メチルエチルケトン若しくはメチルイソブチルケトン等のケトン系溶媒;酢酸エチル、酢酸ブチル、酢酸アミル、乳酸エチル、安息香酸メチル若しくは二酢酸エチレングリコール等のエステル系溶媒;または、ジエチルエーテル、tert-ブチルメチルエーテル、テトラヒドロフラン、ジオキサン、エチレングリコールジメチルエーテル、ジエチレングリコールジメチルエーテル、トリエチレングリコールジメチルエーテル、テトラエチレングリコールジメチルエーテル若しくはポリエチレングリコールジメチルエーテル等のエーテル系溶媒が挙げられる。これらの溶媒は単独で用いても混合して用いても構わない。ラジカル重合を阻害しにくいという観点から、溶媒としては、水、メタノール、tert-ブタノール、tert-アミルアルコール、3,7-ジメチル-3-オクタノール若しくはテトラヒドロフランまたはそれらの混合溶媒が好ましく、水がより好ましい。
重合混合物に占めるモノマー成分の総割合は、重合熱による暴走を適度に抑制しつつ、所望の分子量の共重合体を得るため、10~80質量%が好ましく、15~65質量%がより好ましい。
重合工程においては、前記重合混合物を加熱することによって、重合反応を進行させ、所望の共重合体を得る。
重合工程における反応温度は、重合開始剤の分解温度や溶媒及びモノマーの沸点、反応時間等に応じて選択すればよいが、所望の共重合体を収率よく得るため、20℃~180℃が好ましく、50℃~150℃がより好ましく、60~120℃がさらに好ましい。
重合工程における反応時間は、使用する重合開始剤及びモノマーの種類・濃度や反応温度等に応じて選択すればよいが、所望の共重合体を収率よく得るため、1~48時間が好ましく、2~24時間がより好ましく、3~12時間がさらに好ましい。
本発明の共重合体の重量平均分子量(以下、「Mw」)は、溶液の粘度(操作性)を適度なものとしつつ好適な易滑性を得るため、1,000~5,000,000が好ましく、5,000~3,000,000がより好ましく、10,000~1,000,000がさらに好ましい。
本発明の被覆医療デバイスは、本発明の共重合体によって、医療デバイスの表面の少なくとも一部が被覆されたものである。
医療デバイスの材質としては、(シリコーンハイドロゲルを含む)ハイドロゲル等の含水性基材、または、ポリメチルメタアクリレート等のアクリル樹脂、シロキサン結合を有するシリコーン基材、アルミ等の金属若しくはガラス等の非含水性基材のいずれであっても構わない。
医療デバイスとしては、例えば、眼用レンズ、皮膚用被覆材、創傷被覆材、皮膚用保護材、膚用薬剤担体、輸液用チューブ、気体輸送用チューブ、排液用チューブ、血液回路、被覆用チューブ、カテーテル、ステント、シースバイオセンサーチップ、人工心肺または内視鏡用被覆材が挙げられる。これらの中でも本発明の効果に対する要求が高いことから、眼用レンズおよびステントから選ばれる医療デバイスが好ましい。
眼用レンズとしては、ソフトコンタクトレンズ、ハードコンタクトレンズ、ハイブリッドコンタクトレンズ等のコンタクトレンズ、強膜レンズ、眼内レンズ、人工角膜、角膜インレイ、角膜オンレイまたはメガネレンズが挙げられる。なかでも、コンタクトレンズが好ましい。
ステントとしては、呼吸器用のステントが好ましい。本発明における呼吸器とは、呼吸に関する器官の総称であり、例えば、気道、口腔、鼻腔、喉頭、気管、気管支、細気管支、肺等が挙げられる。本発明の実施形態に係るステントは、呼吸器用のステントであり、気道、気管、気管支、または肺用のステントであることが好ましい。
本発明の医療用組成物は、本発明の共重合体および溶媒を含む。本発明の医療用組成物は、医療デバイスの表面の少なくとも一部を本発明の共重合体によって被覆して、本発明の被覆医療デバイスを得るために使用する。
本発明の医療用組成物の製造方法は、本発明の共重合体と溶媒とを混合して、医療用組成物を得る、調合工程を備える。
上記調合工程で用いる溶媒としては、上記「重合混合物」を得るための溶媒と同様のものが挙げられる。
本発明の医療用組成物に占める本発明の共重合体の割合は、被覆層の厚さを適度なものとしつつ、溶液の粘度(操作性)を高めるため、0.0001~30.0000質量%が好ましく、0.0010~20.0000質量%がより好ましく、0.0050~15.0000質量%がさらに好ましい。
本発明の医療用組成物を、医療デバイスの表面に物理的に接触させることで、医療デバイスの表面を本発明の共重合体で被覆することができる。すなわち被覆医療デバイスの製造方法は、本発明の医療用組成物を医療デバイスの表面に接触させ、被覆医療デバイスを得る、被覆工程を備える。例えば、医療デバイスが眼用レンズである場合を例に説明すると、容器の中に本発明の医療用組成物と眼用レンズとを封入して接触させ、加熱処理すること(被覆工程)により、眼用レンズの表面を本発明の共重合体でコーティングすることができる。
上記加熱操作の手法としては、例えば、高圧蒸気法、可視光、赤外線若しくはマイクロ波等の電磁波照射、電熱法、電磁誘導加熱法、乾熱法または火炎法が挙げられるが、水濡れ性や易滑性を高め、かつ製造工程を短縮する観点から、オートクレーブを用いた高圧蒸気法が好ましい。
上記加熱操作における加熱温度は、医療デバイス自体の強度を維持しつつ、良好な水濡れ性および易滑性を示す被覆医療デバイス表面を得るため、60~200℃が好ましく、80~180℃がより好ましく、100~170℃がさらに好ましく、110~150℃が特に好ましい。
上記加熱操作における加熱時間は、医療デバイス自体の強度を維持しつつ、良好な水濡れ性および易滑性を示す被覆医療デバイス表面を得るため、5~600分が好ましく、10~400分がより好ましく、15~300分がさらに好ましい。
本発明の医療用組成物の初期pHとしては、組成物に濁りが生じず、透明性や表面の水濡れ性および易滑性が良好な医療デバイスが得られることから、2.0~6.0が好ましい。初期pHは、2.2~5.0がより好ましく、2.4~4.5がさらに好ましく、2.5~4.0が特に好ましい。
本発明の医療用組成物のpHは、例えば、酸性物質を添加することにより調整することができる。酸性物質としては、例えば、酢酸、クエン酸、ギ酸、アスコルビン酸、トリフルオロメタンスルホン酸、メタンスルホン酸、硝酸、硫酸、リン酸または塩酸が挙げられる。揮発性低く生体に対する安全性が高い、クエン酸、アスコルビン酸または硫酸が好ましい。またpHの微調整を容易にするために、本発明の医療用組成物に緩衝剤を添加しても構わない。
緩衝剤としては、例えば、ホウ酸、ホウ酸ナトリウム等のホウ酸塩類;クエン酸、クエン酸カリウム等のクエン酸塩類;重炭酸ナトリウム等の重炭酸塩;Na2HPO4、NaH2PO4、若しくはKH2PO4等のリン酸塩;TRIS(トリス(ヒドロキシメチル)アミノメタン)、2-ビス(2-ヒドロキシエチル)アミノ-2-(ヒドロキシメチル)-1,3-プロパンジオール、ビス-アミノポリオール、トリエタノールアミン、ACES(N-(2-アセトアミド)-2-アミノエタンスルホン酸)、BES(N,N-ビス(2-ヒドロキシエチル)-2-アミノエタンスルホン酸)、HEPES(4-(2-ヒドロキシエチル)-1-ピペラジンエタンスルホン酸)、MES(2-(N-モルホリノ)エタンスルホン酸)、MOPS(3-[N-モルホリノ]-プロパンスルホン酸)、PIPES(ピペラジン-N,N’-ビス(2-エタンスルホン酸)またはTES(N-[トリス(ヒドロキシメチル)メチル]-2-アミノエタンスルホン酸)あるいはそれらの塩が挙げられる。本発明の医療用組成物に占める緩衝剤の割合は、所望のpHを達成するため、0.001~2.000質量%が好ましく、0.010~1.000質量%がより好ましく、0.050~0.300質量%がさらに好ましい。
なお本発明の医療用組成物のpHは、加熱操作等の被覆工程の際に変化し得る。例えば加熱操作を行った直後(pH調整前)の医療用組成物のpHは、得られる被覆医療デバイスの物性を好適化するため、2.0~6.5が好ましく、2.2~5.9がより好ましく、2.3~5.5がさらに好ましく、2.4~5.0が特に好ましい。
上記の被覆工程により得られた被覆医療デバイスに、さらに他の処理を施しても構わない。ここで他の処理としては、前記と同様の被覆工程を再度行う処理、本発明の共重合体を含まない溶媒または緩衝剤溶液等を被覆医療デバイスの表面に接触させて同様の加熱操作を行う処理、放射線照射、反対の荷電を有するポリマー材料を1層ずつ交互にコーティングするLbL処理(Layer by Layer処理)、金属イオンによる架橋処理、または、化学架橋処理が挙げられる。
なお加熱処理に用いる緩衝剤溶液の緩衝剤溶液のpHは、生理学的に許容できる6.3~7.8が好ましく、6.5~7.6以上がより好ましく、6.8~7.4がさらに好ましい。
以下、実施例等により本発明を具体的に説明するが、本発明はこれらによって限定されるものではない。先ず実施例等における分析方法および評価方法を説明する。
なお用いたリン酸緩衝液の組成は以下の通りとした。KCl:0.2g/L、KH2PO4:0.2g/L、NaCl:8.0g/L、Na2HPO4:1.2g/L、EDTA:0.5g/L。
(1)ポリマーのMwおよび数平均分子量(Mn)の測定
Prominence GPCシステム(株式会社島津製作所製)を用いてポリマーの分子量を測定した。装置構成は以下の通りとした。
ポンプ:LC-20AD、オートサンプラ:SIL-20AHT、カラムオーブン:CTO-20A、検出器:RID-10A、カラム:GMPWXL(東ソー株式会社製;内径7.8mm×30cm、粒子径13μm)。溶出溶媒として水/メタノール=1/1(0.1N硝酸リチウム添加)を用い、流速0.5mL/分、測定時間30分で測定した。なおサンプル濃度は0.1質量%とし、サンプル注入量を20μLとした。検量線はPEG/PEO標準サンプル(Agilent社製;0.1~1500k)を用いて作成した。

(2)水濡れ性(液膜保持時間)
被覆医療デバイスまたは医療デバイス(以下、「被験サンプル」)を、室温でリン酸緩衝液を用いて洗浄した後、リン酸緩衝液中に室温で24時間以上浸漬し静置した。この被験サンプル(以下、「被験サンプルS」)をリン酸緩衝液から引き上げ、その長手方向が重力方向になるように空中にて保持した。被験サンプルがレンズ形状の場合は、レンズの縁部が形成する円の直径方向が重力方向になるように空中にて保持した。被験サンプルSの空中での保持を開始した時点から、被験サンプルSの表面を覆う液膜の一部が切れるまでの時間を目視観察して、N=3の平均値を下記基準で判定した。なお「液膜の一部が切れる」とは、被験サンプルSの表面の液膜が形状を保つことができなくなり、被験サンプルS表面の一部が液膜に覆われなくなる状態を指す。
A:表面の液膜が20秒以上保持される。
B:表面の液膜が15秒以上20秒未満で切れる。
C:表面の液膜が5秒以上15秒未満で切れる。
D:表面の液膜が1秒以上5秒未満で切れる。
E:表面の液膜が瞬時に切れる(1秒未満)。
被覆医療デバイスまたは医療デバイス(以下、「被験サンプル」)を、室温でリン酸緩衝液を用いて洗浄した後、リン酸緩衝液中に室温で24時間以上浸漬し静置した。この被験サンプル(以下、「被験サンプルS」)をリン酸緩衝液から引き上げ、その長手方向が重力方向になるように空中にて保持した。被験サンプルがレンズ形状の場合は、レンズの縁部が形成する円の直径方向が重力方向になるように空中にて保持した。被験サンプルSの空中での保持を開始した時点から、被験サンプルSの表面を覆う液膜の一部が切れるまでの時間を目視観察して、N=3の平均値を下記基準で判定した。なお「液膜の一部が切れる」とは、被験サンプルSの表面の液膜が形状を保つことができなくなり、被験サンプルS表面の一部が液膜に覆われなくなる状態を指す。
A:表面の液膜が20秒以上保持される。
B:表面の液膜が15秒以上20秒未満で切れる。
C:表面の液膜が5秒以上15秒未満で切れる。
D:表面の液膜が1秒以上5秒未満で切れる。
E:表面の液膜が瞬時に切れる(1秒未満)。
(3)易滑性
リン酸緩衝液から引き上げた被験サンプルSを人指で5回擦った時の感応評価をN=1で行い、下記基準で判定した。
A:非常に優れた易滑性がある(被験サンプルSの表面を流れるように指が滑り、抵抗を全く感じない)。
B:やや優れた易滑性がある(AとCとの中間程度)。
C:中程度の易滑性がある(被験サンプルSの表面を指が滑り、抵抗をほとんど感じない)。
D:易滑性がほとんど無い(CとEとの中間程度)。
E:易滑性が無い(被験サンプルSの表面を指が容易に滑らず、大きな抵抗を感じる)。
リン酸緩衝液から引き上げた被験サンプルSを人指で5回擦った時の感応評価をN=1で行い、下記基準で判定した。
A:非常に優れた易滑性がある(被験サンプルSの表面を流れるように指が滑り、抵抗を全く感じない)。
B:やや優れた易滑性がある(AとCとの中間程度)。
C:中程度の易滑性がある(被験サンプルSの表面を指が滑り、抵抗をほとんど感じない)。
D:易滑性がほとんど無い(CとEとの中間程度)。
E:易滑性が無い(被験サンプルSの表面を指が容易に滑らず、大きな抵抗を感じる)。
(4)含水率
リン酸緩衝液から引き上げた被験サンプルSの表面をワイピングクロス“キムワイプ”(登録商標)(日本製紙クレシア製)で拭き取った後、基材の質量(Ww)を測定した。その後、真空乾燥器で被験サンプルSを60℃で2時間乾燥した後、その質量(Wd)を測定した。これらの質量から、下式(1)により被験サンプルの含水率を算出した。得られた値が1%未満の場合は測定限界以下と判断し、「1%未満」と表記した。N=3の平均値を含水率とした。
被験サンプルの含水率(%)=100×(Ww-Wd)/Ww ・・・ 式(1)。
リン酸緩衝液から引き上げた被験サンプルSの表面をワイピングクロス“キムワイプ”(登録商標)(日本製紙クレシア製)で拭き取った後、基材の質量(Ww)を測定した。その後、真空乾燥器で被験サンプルSを60℃で2時間乾燥した後、その質量(Wd)を測定した。これらの質量から、下式(1)により被験サンプルの含水率を算出した。得られた値が1%未満の場合は測定限界以下と判断し、「1%未満」と表記した。N=3の平均値を含水率とした。

(5)摩擦係数
リン酸緩衝液で濡れた状態の被験サンプルSの表面の摩擦係数を下記の条件で測定し、N=5の平均値を摩擦係数とした。

測定SPEED:2×1mm/秒
摩擦荷重:44g。
リン酸緩衝液で濡れた状態の被験サンプルSの表面の摩擦係数を下記の条件で測定し、N=5の平均値を摩擦係数とした。
装置:摩擦感テスターKES-SE(カトーテック株式会社製)
摩擦SENS:H
測定SPEED:2×1mm/秒
摩擦荷重:44g。
(6)脂質付着量
20ccのスクリュー管にパルミチン酸メチル0.03g、純水10gおよび被験サンプルを入れた。37℃、165rpmの条件下3時間スクリュー管を振とうさせた。振とう後、スクリュー管内の被験サンプルを40℃の水道水と家庭用液体洗剤“ママレモン”(登録商標)(ライオン株式会社製)を用いて擦り洗いした。洗浄後の被験サンプルをリン酸緩衝液の入ったスクリュー管内に入れ、4℃の冷蔵庫内で1時間保管した。その後、被験サンプルを目視観察し、白濁した部分があればパルミチン酸メチルが付着していると判定して、サンプルの表面全体に対するパルミチン酸メチルが付着した部分の面積を観察した。
20ccのスクリュー管にパルミチン酸メチル0.03g、純水10gおよび被験サンプルを入れた。37℃、165rpmの条件下3時間スクリュー管を振とうさせた。振とう後、スクリュー管内の被験サンプルを40℃の水道水と家庭用液体洗剤“ママレモン”(登録商標)(ライオン株式会社製)を用いて擦り洗いした。洗浄後の被験サンプルをリン酸緩衝液の入ったスクリュー管内に入れ、4℃の冷蔵庫内で1時間保管した。その後、被験サンプルを目視観察し、白濁した部分があればパルミチン酸メチルが付着していると判定して、サンプルの表面全体に対するパルミチン酸メチルが付着した部分の面積を観察した。
(7)ムチン付着量
被験サンプルがコンタクトレンズの場合、打抜型を用いて幅(最小部分)5mm、長さ14mmの試験片を切り出した(以下、「サンプルP」)。被験サンプルがステントの場合、打抜型を用いて直径4mmの試験片を切り出した(以下、「サンプルK」)。ムチンとしてMucin, Bovine Submaxillary Gland(Millipore社製;カタログ番号499643)を用いて、濃度0.1質量%のムチン水溶液を調製し、サンプルPまたはサンプルKを22時間37℃の条件で浸漬した。その後BCA(ビシンコニン酸)プロテインアッセイ法によってサンプルPまたはサンプルKに付着したムチンの量を定量し、N=3の平均値をムチン付着量とした。
被験サンプルがコンタクトレンズの場合、打抜型を用いて幅(最小部分)5mm、長さ14mmの試験片を切り出した(以下、「サンプルP」)。被験サンプルがステントの場合、打抜型を用いて直径4mmの試験片を切り出した(以下、「サンプルK」)。ムチンとしてMucin, Bovine Submaxillary Gland(Millipore社製;カタログ番号499643)を用いて、濃度0.1質量%のムチン水溶液を調製し、サンプルPまたはサンプルKを22時間37℃の条件で浸漬した。その後BCA(ビシンコニン酸)プロテインアッセイ法によってサンプルPまたはサンプルKに付着したムチンの量を定量し、N=3の平均値をムチン付着量とした。
(8)リゾチーム付着量
リゾチームとして卵白由来のリゾチーム(富士フイルム和光純薬株式会社製;カタログ番号127-06724)を用いて、0.1質量%のリゾチーム水溶液を調製し、前記サンプルPを22時間37℃の条件で浸漬させた。その後BCAプロテインアッセイ法によってサンプルPに付着したリゾチームの量を定量し、N=3の平均値をリゾチーム付着量とした。
リゾチームとして卵白由来のリゾチーム(富士フイルム和光純薬株式会社製;カタログ番号127-06724)を用いて、0.1質量%のリゾチーム水溶液を調製し、前記サンプルPを22時間37℃の条件で浸漬させた。その後BCAプロテインアッセイ法によってサンプルPに付着したリゾチームの量を定量し、N=3の平均値をリゾチーム付着量とした。
[実施例1]
500mL四ツ口フラスコにCBMA(東京化成工業製)2.29g(10mmol)、AA(富士フイルム和光純薬製)0.72g(10mmol)、DMAA(富士フイルム和光純薬製)7.93g(80mmol)、重合開始剤としてV-50(富士フイルム和光純薬製)6.8mg(0.025mmol)、および精製水(古河薬品工業株式会社製)98.49gを加え、デジタル温度計、三方コックを取り付けたジムロート冷却管、撹拌羽付きシーラーを装着した。超音波照射下、四ツ口フラスコ内を圧力10mmHgまで吸引した後、窒素フラッシュするというサイクルを5回繰り返して、混合溶液内の溶存酸素を除去した。続いて、オイルバス上90℃で撹拌させながら4時間反応させた後、四ツ口フラスコをオイルバスから引き上げて空冷し、共重合体T1を109.44g得た。得られた共重合体T1の性状は表1に示す通りであった。
500mL四ツ口フラスコにCBMA(東京化成工業製)2.29g(10mmol)、AA(富士フイルム和光純薬製)0.72g(10mmol)、DMAA(富士フイルム和光純薬製)7.93g(80mmol)、重合開始剤としてV-50(富士フイルム和光純薬製)6.8mg(0.025mmol)、および精製水(古河薬品工業株式会社製)98.49gを加え、デジタル温度計、三方コックを取り付けたジムロート冷却管、撹拌羽付きシーラーを装着した。超音波照射下、四ツ口フラスコ内を圧力10mmHgまで吸引した後、窒素フラッシュするというサイクルを5回繰り返して、混合溶液内の溶存酸素を除去した。続いて、オイルバス上90℃で撹拌させながら4時間反応させた後、四ツ口フラスコをオイルバスから引き上げて空冷し、共重合体T1を109.44g得た。得られた共重合体T1の性状は表1に示す通りであった。
[実施例2]
CBMAに代えてSBMA(東京化成工業製)2.79g(10mmol)を用いた以外は、実施例1と同様の操作をして、共重合体T2を114.45g得た。得られた共重合体T2の性状は表1に示す通りであった。

[比較例1]
CBMAに代えてリン酸2-(メタクリロイルオキシ)エチル2-(トリメチルアンモニオ)エチル(以下、「MPC」;東京化成工業製)2.95g(10mmol)を用いた以外は、実施例1と同様の操作をして、共重合体Tである共重合体TC1を116.04g得た。得られた共重合体TC1の性状は表1に示す通りであった。
CBMAに代えてリン酸2-(メタクリロイルオキシ)エチル2-(トリメチルアンモニオ)エチル(以下、「MPC」;東京化成工業製)2.95g(10mmol)を用いた以外は、実施例1と同様の操作をして、共重合体Tである共重合体TC1を116.04g得た。得られた共重合体TC1の性状は表1に示す通りであった。
[比較例2]
CBMAに代えてN-ビニルピロリドン(以下、「NVP」;東京化成工業製、1.11g、10mmol)を用いた以外は、実施例1と同様の操作をして、共重合体TC2を97.63g得た。得られた共重合体TC2の性状は、表1に示す通りであった。

医療デバイスAとして、ポリビニルピロリドンおよびシリコーンを主成分とする市販シリコーンハイドロゲルレンズ“Acuvue Oasys”(登録商標;Johnson&Johnson社製)を用いた。共重合体T1を0.3質量%の濃度になるようにリン酸緩衝液に添加し、クエン酸を用いてpH3.0に調整した。調製した溶液をガラスバイアル瓶に3mL移し、医療デバイスAを浸漬させて121℃で30分間オートクレーブ中にて加熱した。加熱後の医療デバイスをリン酸緩衝液で30秒洗浄後、新たなリン酸緩衝液に入れ、さらに121℃で30分間オートクレーブ中にて加熱し、被覆医療デバイスを得た。
医療デバイスAとして、ポリビニルピロリドンおよびシリコーンを主成分とする市販シリコーンハイドロゲルレンズ“Acuvue Oasys”(登録商標;Johnson&Johnson社製)を用いた。共重合体T1を0.3質量%の濃度になるようにリン酸緩衝液に添加し、クエン酸を用いてpH3.0に調整した。調製した溶液をガラスバイアル瓶に3mL移し、医療デバイスAを浸漬させて121℃で30分間オートクレーブ中にて加熱した。加熱後の医療デバイスをリン酸緩衝液で30秒洗浄後、新たなリン酸緩衝液に入れ、さらに121℃で30分間オートクレーブ中にて加熱し、被覆医療デバイスを得た。
[実施例4]
共重合体T1に代えて共重合体T2を用いた以外は、実施例3と同様の操作をして、被覆医療デバイスを得た。

AA2.16g(30mmol)、DMAA6.94g(70mmol)および精製水(61.91g)を用いた以外は、実施例1と同様の操作をして、共重合体TC4を91.02g得た。得られた共重合体TC4の性状は表4に示す通りであった。

共重合体T1に代えて表5に示す共重合体Tを用いた以外は、実施例3と同様の操作をして、被覆医療デバイスを得た。

Claims (10)
- 下記(A)、(B)および(C)をモノマー成分として含む、共重合体:
(A)カルボキシベタイン基および/またはスルホベタイン基を有する化合物;
(B)(メタ)アクリル酸;
(C)アクリルアミド誘導体。 - 前記モノマー成分(A)の含有率が1~40モル%である、請求項1記載の共重合体。
- 前記モノマー成分(B)の含有率が1~35モル%である、請求項1または2記載の共重合体。
- 請求項1記載の共重合体および溶媒を含む、医療用組成物。
- 請求項1記載の共重合体が医療デバイスの表面に被覆された、被覆医療デバイス。
- (A)カルボキシベタイン基および/またはスルホベタイン基を有する化合物、(B)(メタ)アクリル酸、並びに、(C)アクリルアミド誘導体、を含むモノマーを重合して、共重合体を得る、重合工程を備える、共重合体の製造方法。
- 請求項6記載の方法により得られた共重合体と溶媒とを混合して、医療用組成物を得る、調合工程を備える、医療用組成物の製造方法。
- 請求項7記載の方法により得られた医療用組成物を医療デバイスの表面に接触させ、被覆医療デバイスを得る、被覆工程を備える、被覆医療デバイスの製造方法。
- 前記医療デバイスが、眼用レンズ、皮膚用被覆材、創傷被覆材、皮膚用保護材、皮膚用薬剤担体、輸液用チューブ、気体輸送用チューブ、排液用チューブ、血液回路、被覆用チューブ、カテーテル、ステント、シースバイオセンサーチップ、人工心肺および内視鏡用被覆材からなる群から選ばれる、請求項8記載の被覆医療デバイスの製造方法。
- 前記眼用レンズが、コンタクトレンズである、請求項9記載の被覆医療デバイスの製造方法。
