Abstract
本発明は、リパスジルを含有する、防腐効果に優れた眼科用水性組成物を提供することを課題とする。本発明によれば、リパスジル若しくはその塩又はそれらの溶媒和物と、緩衝剤(ただし、(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有するものと、(b)pKaを3つ以上有し、そのpKaがすべて7より小さいものと、を除く。)と、を含有し、防腐剤を含有しない、眼科用水性組成物が提供される。
Description
本発明は、眼科用水性組成物及び防腐効力を付与する方法に関する。
リパスジル(4-フルオロ-5-{[(2S)-2-メチル-1,4-ジアゼパン-1-イル]スルホニル}イソキノリン)は、緑内障治療薬として用いられている。例えば、特許文献1には、リパスジル及びベンザルコニウム塩化物を含有する水性組成物が開示されている。特許文献2には、リパスジル及びトロメタモールを含有する水性組成物が開示されている。
グラナテック(登録商標)点眼液0.4%医薬品インタビューフォーム
点眼剤等の眼科用水性組成物には一般に十分な防腐性能を有することが必要とされる。例えば、特許文献1では防腐剤としてよく使われているベンザルコニウム塩化物が防腐剤として用いられている。また、特許文献2のリパスジル及びトロメタモールを含有する水性組成物においても、ベンザルコニウム塩化物のような防腐剤が用いられている。
現在、日本で上市されているグラナテック(登録商標)点眼液0.4%は、リパスジル塩酸塩水和物を有効成分とする点眼液であり、防腐効果を付与する目的で、ベンザルコニウム塩化物が用いられている(非特許文献1)。
このように、特許文献1及び2に記載の水性組成物やグラナテック(登録商標)点眼液0.4%では、防腐剤を用いることで防腐性能を付与させているが、ベンザルコニウム塩化物のような点眼剤に使用される防腐剤は角膜上皮障害を引き起こす可能性が増大させる。

そこで、本発明は、リパスジル若しくはその塩又はそれらの溶媒和物を含有し、防腐剤を配合しなくても防腐効果に優れた眼科用水性組成物を提供することを目的とする。
本発明の一実施形態に係る眼科用水性組成物は、リパスジル若しくはその塩又はそれらの溶媒和物を含有する眼科用水性組成物であって、特定の成分組成とすることにより、防腐剤を含有せずに防腐効果を有する。
すなわち、本発明は以下に関する。
[1]リパスジル若しくはその塩又はそれらの溶媒和物と、緩衝剤(ただし、(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有するものと、(b)pKaを3つ以上有し、そのpKaがすべて7より小さいものと、を除く。)と、を含有し、防腐剤を含有しない、眼科用水性組成物。
[2]緩衝剤が、(i)及び(ii)から選択される1以上を含む、[1]に記載の組成物。
(i)ホウ酸、ホウ酸塩、ホウ酸又はホウ酸塩の水和物、及び、ホウ酸又はホウ酸塩の無水物からなる群から選択される1以上
(ii)ε-アミノカプロン酸及びε-アミノカプロン酸塩から選択される1以上

前記眼科用水性組成物は防腐剤を含有しない、方法。
本発明によれば、リパスジル若しくはその塩又はそれらの溶媒和物を含有し、防腐剤を含有しなくても優れた防腐効果を有する眼科用水性組成物を提供することができる。
以下、実施形態を詳しく説明する。なお、以下の実施形態は特許請求の範囲に係る発明を限定するものではなく、また実施形態で説明されている特徴の組み合わせの全てが発明に必須のものとは限らない。実施形態で説明されている複数の特徴のうち二つ以上の特徴は任意に組み合わされてもよい。
本発明の一実施形態に係る眼科用水性組成物は、リパスジル若しくはその塩又はそれらの溶媒和物を含有し、防腐剤を含有しない。本願発明者は、眼科用水性組成物を特定の組成とすることにより、防腐剤を含有しなくても、眼科用水性組成物が防腐効果を示すことを見出した。本明細書において、防腐効果、防腐性能等という場合には、細菌、真菌等に対する殺菌作用ないし静菌作用を意味する。
本発明において、眼科用水性組成物とは、対象の眼に局所的に投与される目的を有し、例えば、点眼、眼局所注射等により投与される水性組成物をいう。また、水性組成物とは、少なくとも水を含有する組成物をいい、その性状は特に限定されない。水性組成物は、例えば、水を基剤とする、水溶液、水性懸濁液、エマルション等であってよい。本発明の好ましい態様においては、眼科用水性組成物は、水性点眼液である。
水性組成物に含まれる水の含有量は特に限定されないが、5w/v%以上が好ましく、20w/v%以上がより好ましく、50w/v%以上がさらに好ましく、90w/v%以上が更により好ましく、90~99.8w/v%が特に好ましい。
眼科用水性組成物が含有するリパスジルの塩の種類は特に限定されないが、例えばリパスジル塩酸塩であってもよい。本明細書においてリパスジル若しくはその塩又はそれらの溶媒和物は、リパスジル又はその塩の水和物やアルコール和物等の溶媒和物も含んでいる。
眼科用水性組成物が含有するリパスジルの濃度は特に限定されないが、例えば0.05w/v%以上であってもよく、0.1w/v%以上、又は0.4w/v%以上とすることもでき、その上限は例えば、5.0w/v%である。また、前記濃度は、1.0w/v%以下であってもよく、特に好ましくは、0.1~0.5w/v%であり、その下限は、0.1w/v%、0.2w/v%、0.3w/v%、0.36w/v%、0.4w/v%又は0.5w/v%であることがさらにより好ましい。本明細書において、リパスジル若しくはその塩又はそれらの溶媒和物の濃度は、リパスジルとしての濃度で表される。
上記のとおり、本発明の一実施形態に係る眼科用水性組成物は防腐剤を含有しない。眼科用水性組成物は、ベンザルコニウム塩化物、ベンゼトニウム塩化物、又は臭化ベンゾドデシニウムのような、陽イオン界面活性剤系の防腐剤を含有しない。また、眼科用水性組成物は、ポリヘキサニド(ポリヘキサメチレンビグアニド)、クロルヘキシジン又はその塩(例えばクロルヘキシジングルコン酸塩、クロルヘキシジン塩酸又はクロルヘキシジン酢酸)のような、ビグアナイド骨格(N-C(=N)-NC(=N)-N)を有するビグアナイド系の防腐剤を含有しない。また、眼科用水性組成物は、クロロブタノール又はパラオキシ安息香酸エステル(例えばパラオキシ安息香酸メチル、パラオキシ安息香酸エチル、パラオキシ安息香酸ブチル又はパラオキシ安息香酸プロピル)のような、水酸基を有するアルコール系の防腐剤を含有しない。また、眼科用水性組成物は、亜塩素酸塩(例えば亜塩素酸ナトリウム)のような塩素系の防腐剤を含有しない。また、眼科用水性組成物は、ポリドロニウムのような陽イオンポリマー系の防腐剤を含有しない。また、眼科用水性組成物は、ソルビン酸、又はソルビン酸ナトリウム若しくはソルビン酸カリウムのようなソルビン酸塩も含有しない。本明細書において、防腐剤を含有しないとは、防腐作用を発揮する量の防腐剤を含有しない、又は防腐剤を全く含有しないことを意味する。
本発明の一実施形態に係る眼科用水性組成物は、緩衝剤を含有する。緩衝剤は、(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有するものと、(b)pKaを3つ以上有し、そのpKaがすべて7より小さいものと、を除くものとすることができる。pKa(酸解離定数)は、酸におけるプロトンの解離のしやすさを表す指標であり、その値は化合物に固有のものである。本明細書において、pKaは、任意の温度における値を用いることができ、例えば20~25℃における値であってよい。pKaを測定する方法は、当業者に公知の方法であってよく、例えば、中和滴定、吸光光度法、キャピラリー電気泳動等であってよい。
(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有する緩衝剤の例としては、リン酸(pKa1:2.16、pKa2:7.13、pKa3:12.32(25℃におけるpKa))、グルタミン酸(pKa1:2.10、pKa2:4.07、pKa3:9.47(25℃におけるpka))等が挙げられる。
(b)pKaを3つ以上有し、そのpKaがすべて7より小さい緩衝剤の例としては、クエン酸(pKa1:2.90、pKa2:4.34、pKa3:5.66(25℃におけるpKa))等が挙げられる。
(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有する緩衝剤の例としては、リン酸(pKa1:2.16、pKa2:7.13、pKa3:12.32(25℃におけるpKa))、グルタミン酸(pKa1:2.10、pKa2:4.07、pKa3:9.47(25℃におけるpka))等が挙げられる。
(b)pKaを3つ以上有し、そのpKaがすべて7より小さい緩衝剤の例としては、クエン酸(pKa1:2.90、pKa2:4.34、pKa3:5.66(25℃におけるpKa))等が挙げられる。
本発明の眼科用水性組成物が含有する緩衝剤としては、pKaを1つ又は2つ有する緩衝剤、及び、pKaを3つ以上有し、そのpKaがすべて7より大きい緩衝剤が挙げられる。
pKaを1つ又は2つ有する緩衝剤としては、例えば、酢酸(pKa:4.756(25℃におけるpKa))、ε-アミノカプロン酸(pKa1:4.43、pKa2:10.75(25℃におけるpKa))、炭酸(pKa1:6.46、pKa2:10.36(25℃におけるpKa))等が挙げられる。pKaを3つ以上有し、そのpKaがすべて7より大きい緩衝剤としては、例えば、ホウ酸(pKa1:9.14、pKa2:12.74、pKa3:13.80(20℃におけるpKa))等が挙げられる。
pKaを1つ又は2つ有する緩衝剤としては、例えば、酢酸(pKa:4.756(25℃におけるpKa))、ε-アミノカプロン酸(pKa1:4.43、pKa2:10.75(25℃におけるpKa))、炭酸(pKa1:6.46、pKa2:10.36(25℃におけるpKa))等が挙げられる。pKaを3つ以上有し、そのpKaがすべて7より大きい緩衝剤としては、例えば、ホウ酸(pKa1:9.14、pKa2:12.74、pKa3:13.80(20℃におけるpKa))等が挙げられる。
防腐剤を含有しない本発明の一実施形態に係る眼科用水性組成物は、ホウ酸若しくはホウ砂のようなホウ酸塩、ホウ酸若しくはホウ酸塩の水和物、ホウ酸若しくはホウ酸塩の無水物、ε-アミノカプロン酸若しくはその塩、酢酸若しくはその塩、酢酸若しくは酢酸塩の水和物、酢酸若しくは酢酸塩の無水物からなる群から選択される1以上を含有する。本明細書において、塩、水和物、無水物(例えばホウ酸塩、ホウ酸水和物、無水ホウ酸等)を用いる場合、その濃度は対応する母体化合物(例えばホウ酸)の濃度で表すものとする。
好ましくは、前記組成物は、エチレンジアミン四酢酸(エデト酸)若しくはその塩、エデト酸若しくはエデト酸塩の水和物、エデト酸若しくはエデト酸塩の無水物、D-ソルビトール若しくはその塩を含有していてもよい。これらを含有する場合、眼科用水性組成物の防腐性を向上させることができる。なお、これらの成分が、防腐性能を保つ機能に加えて安定化剤としての機能を有していてもよい。
例えば、ホウ酸を用いる場合、組成物全体に対するホウ酸の濃度は0.001w/v%以上、0.005w/v%以上、0.01w/v%以上、0.05w/v%以上、0.1w/v%以上、0.2w/v%以上、0.3w/v%以上、0.4w/v%以上、0.41w/v%以上、0.5w/v%以上、0.6w/v%以上、0.7w/v%以上、0.8w/v%以上、0.9w/v%以上、1w/v%以上、1.1w/v%以上、1.2w/v%以上、1.27w/v%以上、又は1.3w/v%以上でありうる。また、一実施形態において、組成物全体に対するホウ酸の濃度は10w/v%以下、5w/v%以下、又は3w/v%以下でありうる。このような濃度のホウ酸を含有することにより、眼科用水性組成物の防腐性を効果的に向上させることができる。
本発明の一実施形態に係る眼科用水性組成物がリパスジル及びホウ酸を含有する場合、組成物中のリパスジルの含有量に対するホウ酸の含有量の比(組成物中のホウ酸の含有量(g)/組成物中のリパスジルの含有量(g))は、例えば、0.1~20であってよく、好ましくは1~15であってよく、より好ましくは1.025~5である。眼科用水性組成物中のリパスジルの含有量に対するホウ酸の含有量の比が、このような範囲である場合においては、眼科用水性組成物の防腐性を効果的に向上させることができる。
例えば、ε-アミノカプロン酸を用いる場合、組成物全体に対するε-アミノカプロン酸の濃度は0.001w/v%以上、0.005w/v%以上、0.01w/v%以上、0.05w/v%以上、0.1w/v%以上、0.2w/v%以上、0.3w/v%以上、0.4w/v%以上、又は0.5w/v%以上でありうる。また、一実施形態において、組成物全体に対するε-アミノカプロン酸の濃度は10w/v%以下、5w/v%以下、又は3w/v%以下でありうる。このような濃度のε-アミノカプロン酸を含有することにより、眼科用水性組成物の防腐性を効果的に向上させることができる。
本発明の一実施形態に係る眼科用水性組成物がリパスジル及びε-アミノカプロン酸を含有する場合、組成物中のリパスジルの含有量に対するε-アミノカプロン酸の含有量の比(組成物中のε-アミノカプロン酸の含有量(g)/組成物中のリパスジルの含有量(g))は、例えば、0.1~10であってよく、好ましくは1~7.5であってよく、より好ましくは1.25~5である。眼科用水性組成物中のリパスジルの含有量に対するε-アミノカプロン酸の含有量の比が、このような範囲である場合においては、眼科用水性組成物の防腐性を効果的に向上させることができる。
本発明の一実施形態に係る眼科用水性組成物がエデト酸を更に含有することにより、眼科用水性組成物の防腐性及び安定性を効果的に向上させることができる。
エデト酸を用いる場合、組成物全体に対するエデト酸の濃度は、例えば、0.001w/v%以上、0.005w/v%以上、0.01w/v%以上0.05w/v%以上、又は0.1w/v%以上でありうる。また、一実施形態において、組成物全体に対するエデト酸の濃度は10w/v%以下、5w/v%以下、又は3w/v%以下でありうる。このような濃度でエデト酸を含有することにより、眼科用水性組成物の防腐性及び安定性を効果的に向上させることができる。
エデト酸を用いる場合、組成物全体に対するエデト酸の濃度は、例えば、0.001w/v%以上、0.005w/v%以上、0.01w/v%以上0.05w/v%以上、又は0.1w/v%以上でありうる。また、一実施形態において、組成物全体に対するエデト酸の濃度は10w/v%以下、5w/v%以下、又は3w/v%以下でありうる。このような濃度でエデト酸を含有することにより、眼科用水性組成物の防腐性及び安定性を効果的に向上させることができる。
本発明の一実施形態に係る眼科用水性組成物がリパスジル及びエデト酸を含有する場合、組成物中のリパスジルの含有量に対するエデト酸の含有量の比(組成物中のエデト酸の含有量(g)/組成物中のリパスジルの含有量(g))は、例えば、0.01~1であってよく、好ましくは0.1~0.75であってよく、より好ましくは0.1~0.5である。
眼科用水性組成物が含む溶媒は特に限定されないが、少なくとも水を含有する。眼科用組成物中に含まれる水は、特に限定されないが、好ましくは精製水、滅菌精製水、注射用水等である。
眼科用水性組成物は、その他の添加剤を含んでいてもよい。その他の添加剤の例としては、安定化剤、等張化剤、pH調節剤、粘稠化剤、界面活性剤等が挙げられる。また、眼科用水性組成物が、リパスジル又はその塩以外の有効成分を含んでいてもよい。
安定化剤の例としては、アスコルビン酸、トコフェロール、ジブチルヒドロキシトルエン、ポビドン、亜硫酸塩、モノエタノールアミン、シクロデキストリン、デキストラン、及びタウリン等が挙げられる。これらの安定化剤は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。
等張化剤の例としては、塩化ナトリウム、塩化カリウム、塩化カルシウム、塩化マグネシウム、グリセリン、プロピレングリコール、ポリエチレングリコール、グルコース、ソルビトール、マンニトール、トレハロース、マルトース、スクロース、及びブチレングリコール等が挙げられる。これらの等張化剤は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。等張化剤の好ましい例としては、グリセリン、プロピレングリコール、及び塩化ナトリウムが挙げられる。
pH調節剤の例としては、塩酸、クエン酸、水酸化ナトリウム、水酸化カリウム、炭酸ナトリウム、及び炭酸水素ナトリウム等が挙げられる。これらのpH調節剤は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。
粘稠化剤の例としては、カルボキシビニルポリマー、ポリビニルピロリドン、ポリエチレングリコール、ポリビニルアルコール、キサンタンガム、コンドロイチン硫酸ナトリウム、ヒアルロン酸ナトリウム、ヒドロキシエチルセルロース、メチルセルロース、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロースナトリウム等の水溶性高分子等が挙げられる。これらの粘稠化剤は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。
界面活性剤の例としては、ポリオキシエチレン硬化ヒマシ油、モノステアリン酸ポリオキシエチレングリコール、ポリソルベート、ポリオキシエチレンポリオキシプロピレングリコール、ショ糖脂肪酸エステル、及びチロキサポールなどが挙げられる。これらの界面活性剤は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。
眼科用水性組成物のpHは点眼可能であれば特に制限されないが、眼への刺激を低減する観点から、例えば4.5以上又は5以上であってもよく、9以下、8以下、7以下、又は6以下であってもよい。
一実施形態に係る眼科用水性組成物の浸透圧比は、特に限定されないが、眼への刺激を低減する観点から、0.5以上であってもよく、2以下であってもよい。浸透圧比とは、生理食塩水に対する浸透圧比である。
一実施形態に係る眼科用水性組成物は、点眼液であり、患者への点眼用に用いることができる。
一実施形態に係る眼科用水性組成物は、容器に収容することができる。容器の種類は特に限定されないが、ガラス容器又は樹脂製容器であってもよい。樹脂製容器の材質は、例えば、ポリオレフィン、ポリエステル、ポリスルホン、ポリアミド、又はポリ塩化ビニルでありうる。
一実施形態において、変色抑制作用の観点から、眼科用水性組成物はポリオレフィン系樹脂製容器に収容される。ポリオレフィン系樹脂としては、例えば、低密度ポリエチレン(直鎖状低密度ポリエチレンを含む)、高密度ポリエチレン、中密度ポリエチレン、ポリプロピレン、又は環状ポリオレフィンを用いることができる。ポリオレフィン系樹脂製容器は紫外線吸収剤及び紫外線散乱剤等の紫外線の透過を妨げる物質を有していても良い。
本明細書において「ポリオレフィン系樹脂製容器」とは、容器のうち少なくとも水性組成物と接する部分が「ポリオレフィン系樹脂製」である容器を意味する。したがって、例えば、水性組成物と接する内層にポリオレフィン層を設け、その外側に他の材質の樹脂等を積層等させてなる容器も、「ポリオレフィン系樹脂製容器」に該当する。ポリオレフィン系樹脂は特に限定されず、単一種のモノマーの重合体(ホモポリマー)であっても、複数種のモノマーの共重合体(コポリマー)であってもよい。また、コポリマーである場合においては、その重合様式は特に限定されず、ランダム重合でもブロック重合でもよく、さらにその立体規則性(タクティシティー)は特に限定されない。
一実施形態に係る眼科用水性組成物はマルチドーズ型容器に収容される。マルチドーズ型容器は、複数回分の使用量の眼科用水性組成物を保持し、繰り返し使用可能な容器のことをいう。もっとも、本実施形態における眼科用水性組成物は、ユニットドーズ型容器に収容されていてもよい。ユニットドーズ型容器は、単回分の使用量の眼科用水性組成物を保持し、1回の点眼で使用済みとなる容器のことをいう。
また、眼科用水性組成物は、エチレンオキサイドガス又は過酸化水素等のガスで滅菌処理された容器に収容されてもよい。ガス滅菌処理で使用されるガスの種類については、特に制限されないが、例えば、エチレンオキサイドガス、過酸化水素ガス、及びこれらと二酸化炭素等との混合ガス等が挙げられる。眼科用水性組成物は、ガンマ線照射又は電子線照射等の放射線で滅菌処理された容器に収容されていてもよい。
眼科用水性組成物の製造方法は特に限定されない。リパスジル又はその塩と、必要に応じて有効成分以外とを、精製水に溶解することにより、眼科用水性組成物を製造することができる。
本実施形態で得られた眼科用水性組成物は、フィルターを通過させる工程を経て患部へ提供することができる。ここで「フィルター」とは、眼科用水性組成物を通過させるが、細菌及び真菌を通過させない多孔性膜を指す。フィルターの細孔径は、通常5μm以下であり、好ましくは0.1~2.5μm、更に好ましくは0.1~1μmである。本実施形態において、眼科用水性組成物は、フィルター付き容器に収容され、使用時に容器のフィルターを通過させて容器外に注出される眼科用水性組成物であってもよい。また、眼科用水性組成物の製造時のろ過滅菌工程において、眼科用水性組成物にフィルターを通過させてもよい。フィルターの素材は、特に制限されないが、例えば、ポリエーテルスルホン、ポリフッ化ビニリデン、ポリカーボネート、ポリテトラフルオロエチレン、セルロース混合エステル、ナイロン、又はポリアミド等が挙げられる。
一実施形態に係る眼科用水性組成物は、十分な防腐効力を有している。一実施形態において、眼科用水性組成物は日本薬局方(第18改正)に規定する保存効力試験に適合する。同保存効力試験法は、多回投与容器中に充填された製剤自体又は製剤に添加された防腐剤の効力を微生物学的に評価する方法である。本明細書において、防腐効果に優れる、等という場合、特に限定されないが、同保存効力試験に適合することにより判断することができる。
日本薬局方(第18改正)に規定する保存効力試験法は、以下のように例示されるが、日本薬局方(第18改正)の規定に従う範囲においては、以下の方法と同一である必要はない。
Escherichia coli、Pseudomonas aeruginosa、Staphylococcus aureus、Candida albicans及びAspergillus brasiliensisの5種の菌株をそれぞれカンテン培養又は液体培養し、接種菌液を調製する。試験対象の製剤を含む容器5個のそれぞれの中に、調製した接種菌液をそれぞれ単独に無菌的に注入し、均一に混合する。ここで、混合する接種菌液の量は、製剤の0.5~1.0%とし、通常、製剤1mL又は1g当たり1×105~1×106CFUの生菌数になるように菌液を接種、混合する。これらの容器を遮光下で20~25℃に保存し、接種後0、7、14及び28日目に混合試料中の生菌数を測定する。生菌数の経時的な変化は、接種菌数(CFU/mL又はg)からの対数減少値で表す。
点眼剤においては、日本薬局方(第18改正)の保存効力試験法に規定されるカテゴリーIAの判定基準に従い、保存効力が判定される。同カテゴリーIAの判定基準に適合するには、細菌について、接種7日後に、接種菌数に比べて1.0log以上の減少があり、接種14日後に、接種菌数に比べて3.0log以上の減少があり、かつ、接種28日後には、接種14日後の菌数から増加しないことが要求され、さらに真菌について、接種7日後、14日後及び28日後に、接種菌数から増加していないことが要求される。

点眼剤においては、日本薬局方(第18改正)の保存効力試験法に規定されるカテゴリーIAの判定基準に従い、保存効力が判定される。同カテゴリーIAの判定基準に適合するには、細菌について、接種7日後に、接種菌数に比べて1.0log以上の減少があり、接種14日後に、接種菌数に比べて3.0log以上の減少があり、かつ、接種28日後には、接種14日後の菌数から増加しないことが要求され、さらに真菌について、接種7日後、14日後及び28日後に、接種菌数から増加していないことが要求される。
本発明の一実施形態に係る方法は、防腐剤を添加することなく、リパスジル若しくはその塩又はそれらの溶媒和物を含有する眼科用水性組成物に防腐効力を付与する方法である。この方法により防腐効力が付与された眼科用水性組成物は、防腐剤を含有していない。この方法は、リパスジル若しくはその塩又はそれらの溶媒和物を含有する眼科用水性組成物に、緩衝剤(ただし、(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有するものと、(b)pKaを3つ以上有し、そのpKaがすべて7より小さいものと、を除く。)を添加することを含んでいてよい。これにより、眼科用水性組成物に防腐効力を付与することができる。
本発明の防腐効力を付与する方法は、リパスジル若しくはその塩又はそれらの溶媒和物を含有する眼科用水性組成物にエデト酸を添加することをさらに含んでいてもよい。エデト酸を添加した眼科用水性組成物においても、良好な防腐効果が得られる。
(実施例1)
リパスジル(0.4g)、ホウ酸(1.27g)、及びグリセリン(0.65g)を精製水に溶解させ、pH調節剤を加えてpHを6.0に調整することにより、100mLの点眼液を調製した。なお、ホウ酸のpKa(例えば20℃におけるpKa)は、pKa1:9.14、pKa2:12.74、pKa3:13.80である。
リパスジル(0.4g)、ホウ酸(1.27g)、及びグリセリン(0.65g)を精製水に溶解させ、pH調節剤を加えてpHを6.0に調整することにより、100mLの点眼液を調製した。なお、ホウ酸のpKa(例えば20℃におけるpKa)は、pKa1:9.14、pKa2:12.74、pKa3:13.80である。
次に、得られた点眼液に対して保存効力試験を行った。保存効力試験は、日本薬局方(第18改正)に従って行った。具体的には、Escherichia coli、Pseudomonas aeruginosa、Staphylococcus aureus、Candida albicans、及びAspergillus brasiliensisの各菌種を用いて、カテゴリーIAの判定基準を充たすかどうかを確認した。具体的な判定基準は、細菌については接種7日後に接種菌数に比べて1.0log以上の減少があり、接種14日後に接種菌数に比べて3.0log以上の減少があり、接種28日後には14日後の菌数から増加していないこと、真菌については接種7日後、14日後及び28日後に接種菌数が増加していないこと、である。
その結果、実施例1は、防腐剤を含有していないにもかかわらず、保存効力試験に適合することが確認された。
(比較例1)
リパスジル(0.4g)、無水リン酸二水素ナトリウム(0.4g)、及びグリセリン(2.316g)を精製水に溶解させ、pH調整剤を加えてpHを6.0に調整することにより、100mLの点眼液を調製し(表1)、実施例1と同様に保存効力試験を行った。その結果、比較例1の点眼液は保存効力試験に適合しないことが確認された。この結果は、特許文献1の結果と同様の結果であった。なお、リン酸のpKa(例えば25℃におけるpKa)は、pKa1:2.16、pKa2:7.13、pKa3:12.32である。
リパスジル(0.4g)、無水リン酸二水素ナトリウム(0.4g)、及びグリセリン(2.316g)を精製水に溶解させ、pH調整剤を加えてpHを6.0に調整することにより、100mLの点眼液を調製し(表1)、実施例1と同様に保存効力試験を行った。その結果、比較例1の点眼液は保存効力試験に適合しないことが確認された。この結果は、特許文献1の結果と同様の結果であった。なお、リン酸のpKa(例えば25℃におけるpKa)は、pKa1:2.16、pKa2:7.13、pKa3:12.32である。
(実施例2~6)
下表1に記載する組成となるように、実施例2~6の各点眼液を調製した。実施例2~6の各点眼液は、組成以外は、実施例1と同じ方法により調製した。

得られた点眼液に対して保存効力試験を行った結果を下表2に示す。
表2に示す通り、実施例1~6は全て保存効力試験に適合することが確認された。
(実施例7~15)
下表3に記載する組成となるように、実施例7~15の各点眼液を調製した。実施例7~15の各点眼液は、組成以外は、実施例1と同じ方法により調製した。

得られた点眼液に対して保存効力試験を行った結果を下表4に示す。
表4に示す通り、実施例7~15は全て保存効力試験に適合することが確認された。実施例7~8に示す通り、等張化剤がプロピレングリコール及び塩化ナトリウムである場合においても保存効力試験に適合することが確認された。実施例9に示す通り、安定化剤がD-ソルビトールである場合においても保存効力試験に適合することが確認された。実施例10に示す通り、緩衝剤がε-アミノカプロン酸である場合においても保存効力試験に適合することが確認された。また、実施例11~15に示す通り、リパスジル濃度が、0.36%及び0.1%である場合においても保存効力試験に適合することが確認された。特許文献1及び特許文献2には、リパスジル単独でも、十分ではないが防腐効果が確認されるとの記載があるが、ホウ酸を配合することにより、防腐効果があるとされるリパスジルの濃度が減少しても(実施例11~15)、十分な保存効力が奏されることが確認された。
(製剤例1)
リパスジル(0.4g)及びホウ酸(0.6g)を精製水に溶解させ、さらに等張化のために適切な量のグリセリンを添加し、pH調節剤を加えてpHを6.0に調整することにより、100mLの点眼液を調製する。

製剤例1の組成に更にエデト酸を0.1g用いて製剤例1と同様に点眼液を調製する。

それぞれエデト酸を0.05g、0.02g、及び0.01g用いたことを除き、製剤例2と同様に点眼液を調製する

(製剤例6~10)
それぞれホウ酸を0.2g、0.1g、0.05g、0.02g、及び0.01g用いたことを除き、製剤例1と同様に点眼液を調製する。

(製剤例11~13)
それぞれホウ酸の代わりに酢酸ナトリウム水和物を0.2g、0.1g、及び0.05g用いたことを除き、製剤例1と同様に点眼液を調製する。

(製剤例14~18)
それぞれリパスジルを0.8g、0.2g、0.1g、0.08g及び0.05g用いたことを除き、製剤例1と同様に点眼液を調製する。

各製剤例によれば、防腐性能を有する点眼剤を得ることができる。
発明は上記の実施形態に制限されるものではなく、発明の要旨の範囲内で、種々の変形・変更が可能である。
Claims (10)
- リパスジル若しくはその塩又はそれらの溶媒和物と、緩衝剤(ただし、(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有するものと、(b)pKaを3つ以上有し、そのpKaがすべて7より小さいものと、を除く。)と、を含有し、防腐剤を含有しない、眼科用水性組成物。
- 緩衝剤が、(i)及び(ii)から選択される1以上を含む、請求項1に記載の組成物。
(i)ホウ酸、ホウ酸塩、ホウ酸又はホウ酸塩の水和物、及び、ホウ酸又はホウ酸塩の無水物からなる群から選択される1以上
(ii)ε-アミノカプロン酸及びε-アミノカプロン酸塩から選択される1以上 - 防腐剤が、ベンザルコニウム塩化物、ベンゼトニウム塩化物、臭化ベンゾドデシニウム、ポリヘキサメチレンビグアニド、クロルヘキシジン又はその塩、クロロブタノール、パラオキシ安息香酸エステル、亜塩素酸塩、塩化ポリドロニウム、及び、ソルビン酸又はその塩からなる群から選択される1以上である、請求項1に記載の組成物。
- エデト酸、エデト酸塩、エデト酸又はエデト酸塩の無水物、及び、エデト酸又はエデト酸塩の水和物からなる群から選択される1以上を含有する、請求項1に記載の組成物。
- pHが5以上9以下である、請求項1に記載の組成物。
- 浸透圧比が0.5以上2以下である、請求項1に記載の組成物。
- ポリオレフィン系樹脂製容器に収容されている、請求項1から6のいずれか1項に記載の組成物。
- マルチドーズ型容器に収容されている、請求項1から6のいずれか1項に記載の組成物。
- 日本薬局方(第18改正)に規定する保存効力試験に適合する、請求項1から6のいずれか1項に記載の組成物。
- リパスジル若しくはその塩又はそれらの溶媒和物を含有する眼科用水性組成物に防腐効力を付与する方法であって、緩衝剤(ただし、(a)pKaを3つ以上有し、かつ7より大きいpKa及び7より小さいpKaを有するものと、(b)pKaを3つ以上有し、そのpKaがすべて7より小さいものと、を除く)を前記眼科用水性組成物に添加することを含み、
前記眼科用水性組成物は防腐剤を含有しない、方法。
